A fim de analisar o fluxo de informa??o em oper?rias de Dinoponera quadriceps investigamos duas col?nias com base na quantidade de oper?rias e larvas no ninho, n?mero de intera??es individuais, n?mero de sa?das, ?for?a? e ?alcance? de cada intera??o, al?m do fluxo de informa??o em diferentes hor?rios. O estudo foi realizado no Laborat?rio de Biologia Comportamental da UFRN. Foram observadas duas col?nias com diferentes densidades de oper?rias por 16h/col?nia, totalizando 8h/dia atrav?s de grava??o com c?mera digital. As col?nias de D. quadriceps n?o apresentaram correla??o entre o n?mero total de intera??es com o n?mero total de oper?rias e nem com as sa?das do ninho. As taxas de intera??es per capita apresentaram padr?o semelhante para as duas col?nias, sendo maiores no hor?rio com disponibilidade de alimento. A col?nia com menor densidade de oper?rias apresentou mais intera??es por individuo, principalmente no dia e hor?rio com alimento, e estas intera??es obtiveram maior ?for?a? e ?alcance?. A din?mica de redes sociais tamb?m apresentou padr?o semelhante entre as col?nias, apresentando maiores fluxos de informa??o no hor?rio com alimento. Com base no exposto conclu?mos que as oper?rias de D. quadriceps regulam a atividade de forrageamento atrav?s de autoestimula??o, como tamb?m suas taxas de contato, aumentando o n?mero de intera??es em dia e hor?rio com alimento na col?nia, modificando o fluxo de informa??o e aumentando a dissemina??o da informa??o. / Social interactions between workers in a colony do not happen randomly, since workers decide how often they will interact and who they will interact with. Interactions rate in the colony are regulated by the density of individuals it contains and through which can quickly transfer information in the colony, causing changes in network dynamics. In order to analyze the flow of information among workers of Dinoponera quadriceps we investigated two colonies based on the number of workers and larvae in the nest, number of individual interactions, number of outputs, 'strength' and 'reach of each interaction, and also the information flow at different times. The study was conducted in the Behavioral Biology Laboratory at UFRN. Two colonies with different densities of workers were observed for 16h/colony, totaling 8 hours/day by recording using a digital camera. Colonies of D. quadriceps showed no correlation between the total number of interactions with the total number of workers nor to the number of nest exits. The per capita rates of interactions showed a similar pattern in both colonies, being higher during the food availability periods. The colony with lower density of workers had more interactions per individual, especially on the day and period with food, and these interactions got stronger and greater range. The dynamics of social networks also showed a similar pattern among colonies, with greater flow of information during the feeding time. Based on these results we conclude that workers of D. quadriceps regulate foraging activity through self-stimulation, as well as with their contact rates by increasing the number of interactions when food was offered to the colony, changing the flow of information and increasing dissemination of information.

Adaptações neuromusculares de extensores de joelho ao treinamento excêntrico em dinamômetro isocinético

O exercício excêntrico é utilizado como uma estratégia de treinamento de força em atletas e sujeitos saudáveis de diferentes faixas etárias. Esse tipo de exercício também é largamente difundido na área da reabilitação musculoesquelética, sendo o quadríceps femoral o músculo mais frequentemente investigado devido à sua relevância clínica. O tema dessa tese de Doutorado é a adaptação do sistema neuromuscular ao treinamento excêntrico de extensores de joelho. No Capítulo I, um estudo de revisão procurou descrever os diferentes métodos empregados pelos cientistas para o treinamento excêntrico de extensores de joelho, assim como avaliar as adaptações neurais, morfológicas e na produção de força muscular de sujeitos saudáveis. Após busca sistematizada nas bases de dados, 26 estudos foram incorporados ao nosso estudo de revisão. Os programas de treinamento excêntrico realizados em dinamômetro isocinético e em equipamentos isotônicos (leg-press e cadeira extensora de joelho) foram descritos e discutidos. Os resultados desses estudos indicam um efeito positivo do treinamento excêntrico sobre a força e a ativação muscular, especialmente em testes excêntricos, assim como uma significativa resposta hipertrófica. Porém, resultados conflitantes e lacunas identificadas na literatura incentivaram a realização de dois estudos originais para verificar: (1) o comportamento temporal das adaptações neurais e morfológicas ao treinamento excêntrico de extensores de joelho e suas contribuições para os ganhos de força (Capítulo II); e (2) as adaptações específicas de músculos sinergistas da extensão de joelho: reto femoral (RF) e vasto lateral (VL) (Capítulo III). Vinte homens saudáveis realizaram um período controle de quatro semanas sem treinamento, seguido por um programa de 12 semanas de treinamento excêntrico em dinamômetro isocinético. Avaliações de torque, eletromiografia e ultrassonografia de extensores de joelho foram realizadas a cada quatro semanas. Um maior ganho de força foi verificado nos testes excêntricos, seguido pelos testes isométricos e concêntricos. A ativação muscular nos testes excêntricos e isométricos aumentou após quatro e oito semanas de treinamento, respectivamente, ao passo que os testes concêntricos não apresentaram alteração da atividade eletromiográfica. Os valores de espessura muscular, área de secção transversa anatômica e comprimento de fascículo aumentaram após quatro e oito semanas de treinamento, mas não modificaram entre a oitava e a 12ª semana de intervenção. As adaptações na geometria fascicular dos músculos RF e VL foram semelhantes ao longo do programa de treinamento. Nossos achados sugerem que: (1) existe um forte efeito da especificidade do exercício excêntrico sobre os aumentos de força e ativação muscular; (2) os ganhos de força excêntrica e isométrica até a oitava semana de treinamento estão relacionados a adaptações neurais e morfológicas mensuráveis, enquanto outros mecanismos podem estar relacionados aos ganhos nas quatro últimas semanas de intervenção; (3) o ganho de força concêntrica não é afetado por adaptações neurais; (4) o treinamento excêntrico aumenta o comprimento, mas não o ângulo de penação fascicular; (5) as adaptações na arquitetura muscular podem ocorrer nas primeiras semanas de treinamento; e (6) apesar das diferenças estruturais e funcionais, RF e VL apresentam índices semelhantes de adaptação. / Eccentric exercise is used as a strategy for strength training in athletes and healthy subjects with different ages. This type of exercise is also frequently used in musculoskeletal rehabilitation, and the quadriceps femoris is the most frequently investigated muscle due its clinical relevance. The theme of this PhD thesis is the adaptation of the neuromuscular system to knee extensor eccentric training. In Chapter I, a review study aimed at describing the different methods employed by scientists for knee extensor eccentric training, as well as for evaluating the neural, morphological and muscular strength adaptations in healthy subjects. After a systematized search in related data bases, 26 studies were incorporated to our review study. Eccentric training programs performed in isokinetic dynamometers and isotonic devices (leg-press and knee extension chair) were described and discussed. Results from these studies indicate a positive effect of eccentric training on muscle strength and activation, especially in eccentric tests, as well as a significant hypertrophic response. However, conflicting results and gaps observed in the literature encouraged us to perform two original studies to verify: (1) the time course of neural and morphological adaptations to knee extensor eccentric training and their contribution to strength gains (Chapter II); and (2) the specific adaptive responses from synergistic knee extensor muscles: rectus femoris (RF) and vastus lateralis (VL) (Chapter III). Twenty healthy men performed a four-week control period without training, followed by a 12-week eccentric training on an isokinetic dynamometer. Knee extensor evaluations of torque, electromyography and ultrasonography were made every four weeks. A higher strength increase was observed in eccentric tests, followed by isometric and concentric tests. Muscle activation in eccentric and isometric tests increased after four and eight weeks of training, respectively, while the concentric tests presented no change in the electromyographic activity. Muscle thickness, anatomical cross-sectional area and fascicle length values increased after four and eight weeks of training, but did not change between the eighth and twelfth intervention week. Adaptations in RF and VL fascicular geometry were similar throughout the training program. Our findings suggest that: (1) there is a strong specificity effect of eccentric exercise on increases in muscle strength and activation; (2) eccentric and isometric strength gains up to the eighth week of training are related to measurable neural and morphological adaptations, while other mechanisms may be related to strength increase in the last four weeks of intervention; (3) concentric strength gain is not affected by neural adaptations; (4) eccentric training increases fascicle length, but not pennation angle; (5) muscle architecture adaptations may occur in the first weeks of training; and (6) despite the structural and functional differences, RF and VL present similar adaptation levels.

Avaliação do tendão quadríceps e ligamento patelar pela ultrassonografia e ressonância magnética em jogadores de futebol assintomáticos / Ultrasonography and magnetic resonance imaging for assessment of the quadriceps tendon and patellar ligament in asymptomatic soccer players

INTRODUÇÃO: A ultrassonografia (US) e a ressonância magnética (RM) representam os principais métodos de diagnósticos por imagem na avaliação do mecanismo extensor do joelho. Há carência de trabalhos que avaliem por US e RM a presença de alterações no mecanismo extensor de jogadores de futebol assintomáticos. OBJETIVO: Avaliar, por meio de RM e US, o tendão quadríceps e o ligamento patelar de jogadores de futebol assintomáticos e dos participantes do grupo-controle. Identificar se há alterações de imagens ou lesões. Determinar se essas sofrem influências pelos seguintes critérios: canhoto ou destro, quanto à dominância ao chute, posição em campo, idade do atleta e tempo de prática esportiva. MÉTODO: Foram avaliados um total de 248 joelhos: 112 eram de 56 de jogadores de futebol assintomáticos e 136 eram do grupo-controle. O grupo composto por jogadores de futebol apresentava idades entre 14 e 34 anos. Já o grupo-controle era composto por indivíduos com idade também entre 14 e 34 anos, principalmente, universitários e funcionários do hospital, com gênero igual aos dos jogadores selecionados, mas sem atividades físicas de impacto, como vôlei, futebol e salto, por exemplo. RESULTADO: Constatou-se a presença de alterações de imagens ou de lesões em nove ligamentos patelares e apenas duas nos tendões quadríceps de jogadores de futebol assintomáticos, tanto pela US quanto pela RM, e nenhum caso alterado no grupo-controle. Houve concordância significativa entre RM e US quanto aos resultados obtidos. Além disso, os jogadores de futebol apresentaram anormalidades (alterações de imagens ou lesões) no tendão quadríceps ou no ligamento patelar significativamente (p<0,05) maior do que no grupo-controle; a idade média dos jogadores com alguma anormalidade é significativamente menor do que a dos jogadores sem lesão (p<0,003); o tempo médio de prática desportiva dos jogadores que apresentam anormalidade é menor do que o tempo médio dos jogadores que não têm anormalidade (p<0,001); os jogadores destros apresentaram anormalidades, enquanto que os jogadores canhotos não (p<0,05). Dentre os jogadores, há correlação entre mais alterações de imagem ou lesão de joelho e menor tempo como jogador e menor idade (R² de Nagelkerke = 0,700). Destros têm risco aumentado de ter anormalidade nas referidas estruturas (Razão de Chances = 15,204) quanto menor for o tempo como jogador de futebol. Os jogadores da defesa têm 4,76 vezes mais chance de desenvolver anormalidade do que atacantes e goleiros (p<0,04). CONCLUSÃO: Os jogadores de futebol assintomáticos apresentam anormalidades no tendão do quadríceps e ligamento patelar detectadas pela RM e US. As alterações estão relacionadas a menor idade, menor tempo de prática desportiva, ser destro e ser jogador de defesa / INTRODUCTION: Ultrasonography (US) and magnetic resonance imaging (MRI) are the main diagnostic imaging methods used in evaluation of the extensor mechanism of the knee. Theres a scarcity of works assessing alterations on the extensor mechanism of the knee of asymptomatic soccer players using US and MRI. OBJECTIVE: To assess, by means of MRI and US imaging, the quadriceps tendon and patellar ligament of asymptomatic soccer players and healthy control group; identify abnormal imaging findings or injuries; ascertain whether these are affected by leg dominance (left or right), field position, age, and time spent practicing the sport. METHOD: A total of 248 knees were evaluated: 112 of 56 asymptomatic soccer players and 136 of healthy controls. The players ages ranged from 14 to 34 years. The control group was composed mostly of college students and hospital staff members, also with ages between 14 and 34, matched for gender to the selected soccer players, and who did not engage in high-impact activities such as volleyball, soccer, or jumping. RESULTS: Abnormal imaging findings or injuries were detected, by US and MRI alike, in nine patellar ligaments and two quadriceps tendons of asymptomatic soccer players. No abnormal findings were detected in the control group. CONCLUSION: There was significant consistency between MRI and US findings. Soccer players were much more likely (p<0.05) to have abnormal imaging findings or injuries of the quadriceps tendon and patellar ligament as compared with the control group; the average age of players presenting some abnormality is significantly lower than of those without injuries (p<0.003); the average time spent practicing the sport for players presenting abnormalities is lower than those who dont present injuries (p<0.001); a correlation between the dominant leg and the affected knee was observed, with the players with a dominant right leg presenting injuries while the ones with a dominant left leg not showing abnormalities (p<0.05). Amongst the players there is a correlation between more abnormal imaging findings or injuries on the knee and less professional practicing time and age (Nagelkerke R² = 0.700).Players with a dominant right leg have an increased chance of presenting abnormalities on the quadriceps tendon and patellar ligament (Odds Ratio = 15.204) with a lower average time spent practicing the sport. Defense players have 4.76 times more chance to develop abnormalities than forward players and goalkeepers (p<0.04). CONCLUSION: The asymptomatic soccer players present abnormalities on the quadriceps tendon and patellar ligament identified using ultrasonography (US) and magnetic resonance imaging (MRI). The alterations are related with less age, less time of professional practice, right leg dominance and defense field position

The effect of sacroiliac joint adjustment in conjunction with myofascial dry needling of the rectus femoris muscle and myofascial dry needling on its own on quadriceps femoris muscle strength

M.Tech. / This study was conducted to determine the effect of Sacroiliac joint adjustment to the restricted Sacroiliac joint in combination with myofascial dry needling to the active/latent Rectus Femoris muscle trigger point and Rectus Femoris myofascial dry needling on its own on Quadriceps Femoris muscle strength. Thirty participants between the ages of 18 and 40 years were recruited through the use of advertisements placed in and around the University of Johannesburg’s Chiropractic Day Clinic. Participants were assessed for exclusion criteria by performing a Full Case History, Pertinent Physical Examination, Lumbar Spine and Pelvis Regional Examination and S.O.A.P note. The participants who conformed to the specific inclusion criteria were accepted for this study. These participants were randomly placed into two groups of sixteen participants each. Group One receive Sacroiliac joint adjustment to the restricted Sacroiliac joint and ipsilateral myofascial dry needling of the Rectus Femoris muscle active/latent myofascial trigger points. Group two received myofascial dry needling of the Rectus Femoris muscle active/latent myofascial trigger point on the same side as the Sacroiliac joint restriction. Each participant received five treatments over a three week period, i.e. two treatments for the first two weeks and one follow-up treatment in the third week. The subjective data of the patients lower back pain was assessed using the Numerical Pain Rating Scale. The objective data was obtained from the Isometric Dynamometer measuring the Quadriceps Femoris muscle strength before and after each treatment. In addition the Algometer was used for measuring the pressure pain threshold of the Rectus Femoris trigger points. An analysis was performed using Repeated Measures Analysis, t-Test, Frequencis, Descriptives and Friedman’s Tests. The results of this study demonstrate that Sacroiliac joint adjustment to a restricted Sacroiliac joint with ipsilateral myofascial dry needling of the Rectus Femoris muscle showed a statistically significant, but temporary increase in Quadriceps Femoris muscle strength. Myofascial dry needling of the Rectus Femoris muscle on its own was found to be statistically insignificant for the increase in Quadriceps Femoris muscle strength.

EFFECT OF A 12-WEEK HOME-BASED NEUROMUSCULAR ELECTRICAL STIMULATION TREATMENT ON CLINICAL OUTCOMES FOLLOWING ARTICULAR CARTILAGE KNEE SURGERY

Articular cartilage defects in the knee are common, and can result in pain, decreased function and decreased quality of life. Untreated defects are considered to be a risk factor for developing osteoarthritis, a progressive degenerative joint disease with minimal treatment options. To address these issues, various surgical procedures are available to treat articular cartilage defects in the knee. While these procedures overall have positive results, after surgery patients experience large and persistent deficits in quadriceps strength. A contributing factor to this post-surgical weakness is believed to be the extended post-operative non-weight bearing period, with full weight bearing not initiated until approximately 4 – 6 weeks after surgery. During this non-weight bearing period a minimal amount of demand is placed upon the muscle. Subsequently, the quadriceps muscle undergoes a large degree of atrophy with a significant decrease in muscle strength. Muscular strength deficits reduce the knee joint stability, also increasing the risk of osteoarthritis development. Interventions that can be used to facilitate quadriceps strength while protecting the articular cartilage repair are needed. Neuromuscular electrical stimulation (NMES) is an effective post-knee surgery rehabilitation technique to regain quadriceps musculature. In recent years manufactures have been developing knee sleeve garments integrated with NMES allowing for portability of the NMES treatment. The primary aim of this study was to evaluate the effectiveness of a 12-week home-based neuromuscular electrical stimulation treatment on post-surgical clinical outcomes (quadriceps strength, lower extremity function, and patient reported outcomes) after articular cartilage knee surgery. Patients were randomized between a standard of care home-treatment group and a NMES home-treatment group. Patients completed isometric quadriceps strength testing, the Y-balance test, and the Knee Injury and Osteoarthritis Outcome Score (KOOS) before surgery and at 3-months after surgery. The secondary aims of this study were to determine the most effective NMES parameters for post-surgical quadriceps strength; and to develop a framework to identify factors that may influence a patient’s adherence to a prescribed therapy program. From our results we can make several conclusions. First, we found only a small number of studies utilize similar parameters for post-surgical quadriceps strength treatments. The majority of the parameters reported in the literature were highly variable between studies. Second, clinicians can utilize the expanded Health Belief Model to identify situational and personal factors unique to a patient that may impact adherence to a prescribed treatment. Clinicians can then implement the proposed interventional strategies to address the identified situational and personal factors. Finally, there was no difference in quadriceps strength, lower extremity function, or self-reported scores at 3-month between a home-based NMES treatment and a standard of care home-based treatment. Patients’ adherence to the treatment protocols may have been a major factor contributing to these results. Utilizing a model, such as the proposed expanded Health Belief Model, may assist clinicians in improving a patients’ adherence to future prescribed home-treatment programs.

Optimizing strength training for hypertrophy : A periodization of classic resistance training and blood-flow restriction training

Aim The main aim of this study was to investigate if a combination of classic resistance training and blood flow restricted resistance exercise (BFRE) training would result in greater increases in quadriceps muscle growth compared with other strength training studies. The second aim was to investigate if there would be any difference in muscle hypertrophy between men and women after the training intervention.   Method Twenty untrained subjects (10 males and 10 female) were recruited to participate in a 10-week unilateral resistance training intervention. Sixteen subjects completed the training intervention. After two familiarization sessions subjects performed three sessions per week in leg press and leg extension, except for week 4 and 8 were subjects performed five BFRE training sessions Monday to Friday. All subjects performed a one repetition maximum test in leg press and leg extension pre and post the training intervention. Ultrasound screening was performed pre and post training intervention to measure muscle thickness in m. vastus lateralis (VL).   Results The 10-week intervention resulted in a significant increase of VL muscle thickness by 15,1 % ± 7,6 (p ≤ 0,01). Both men and women increased in VL muscle thickness, men (n=7) by 15,4 % ± 9,3 (p ≤ 0,01) and women (n=9) by 14,8 % ± 6,0 (p ≤ 0,01), with no difference between genders. Maximal strength increased for the entire group in the leg press by 59,1 % ± 27,4 (p ≤ 0,01) and in the leg extension by 19,8 % ± 13,1 (p ≤ 0,01). Men had an increase of 58,1 % ± 18,0 (p ≤ 0,01) and women with 60,3 % ± 32,8 (p ≤ 0,01) in the leg press. In the leg extension women and men increased their maximal strength by 23,3 % ± 7,4 (p ≤ 0,01) respectively 17,0 % ± 14,4 (p = 0,051).   Conclusions Our unique training protocol resulted in a superior increase in muscle growth in comparison with most other strength training studies. Our result can be converted to an increase of 17,3 % (0,25 % per day) in VL muscle CSA, which is much greater than the mean increase of 0,11 % per day reported in a large meta-analysis (Wernbom, Augustsson &amp; Thomeé 2007).

A resistência elástica é aplicada amplamente em programas de fortalecimento muscular nos processos de reabilitação física. No entanto, as conseqüências biomecânicas para o músculo exercitado com esta forma de sobrecarga são pouco estudadas. A ausência de fundamentação científica no que tange a aplicação da resistência elástica fez com que surgissem algumas suposições a respeito de sua caracterização biomecânica. Uma delas sugere que o exercício com resistência elástica pode ser caracterizado como sendo isocinético. Neste contexto, o objetivo global deste trabalho foi o de caracterizar o comportamento biomecânico do músculo quadríceps femoral frente ao exercício com resistência elástica. Tal caracterização foi realizada através da utilização de um modelo biomecânico bidimensional da articulação do joelho. O seu comportamento mioelétrico também foi avaliado. Conclui-se de uma forma geral, que o desenvolvimento deste estudo permitiu um entendimento a respeito da interação biomecânica entre o músculo e a resistência elástica durante o exercício. Permitindo dessa forma elucidar algumas questões ligadas a aplicação clínica desta modalidade de exercício / The elastic resistance is applied in programs of muscular fortification in processes of physical rehabilitation. However, the biomechanicals consequences for the muscle exercised with this resistance form aren’t very studied. The low scientific fundamentation for the application of the elastic resistance, results in some suppositions about its biomechanic characterization. One of them suggests that the exercise with elastic resistance can be characterized as isokinetic. In this context, the global objective of this work was characterizing the biomechanic behavior of the quadriceps muscle front to the exercise with elastic resistance. Such characterization was accomplished through the use of a bidimensional biomechanic model of the knee joint. The electromiographic behavior was also evaluated. In a general conclusion, the development of this study allowed an understanding of the biomechanic interaction between the muscle and the elastic resistance during the exercise. Allowing to elucidate some subjects of the clinical application of this exercise modality

Belastad knäböj: Skillnader i muskelaktivering mellan utförande i fri bana och Smith-maskin samt mellan personer med olika anteversionsvinklar / The loaded barbell squat: Muscle activation with the barbell in a free compared to a fixed vertical movement path and in individuals with different hip anteversion angles

Syfte: Att undersöka om det finns skillnader i muskelaktivering i lår- och sätesmuskulatur under belastad knäböj då den utförs med fri skivstång på axlarna respektive i Smith-maskin. Ett andra syfte var att undersöka om anteversionsvinklar kan påverka muskelaktiveringen.   Metod: Sexton deltagare utförde fem repetitioner av belastade (motsvarande 100% kroppsvikt) knäböj i två omgångar. En omgång i Smith-maskin och en i en fri bana. Tiden för varje knäböj standardiserades till fyra sekunder. Likaså standardiserades djup och fotposition. Ordningen mellan de två olika konditionerna slumpades. Muskelaktiviteten mättes genom att studiepersonerna bar shorts med insydda elektroder som mätte elektromyografisk aktivitet. Craig’s test användes före utförandet för att mäta anteversionsvinklarna hos deltagarna.  Resultat: När knäböjen utfördes i en Smith-maskin var det en lägre muskelaktivitet i mm. quadriceps jämfört med utförande i en fri bana. Tydligast var skillnaden under knäböjens första tre sekunder. Under andra och tredje sekunden var det även lägre aktivitet i hamstrings. Det var ingen skillnad i aktivitet mellan konditioner i m. gluteus maximus eller mellan studiedeltagare med femoral retroversion och neutral höftledsvinkel i någon av konditionerna.  Slutsats:  Resultaten indikerar att unga vältränade individer har en högre aktivitet i mm. Quadriceps under utförande av en belastad knäböj då den utförs i en fri bana jämfört med Smith-maskin. Även mm. hamstrings tycks aktiveras mer men endast i knäledens mest flekterade läge. Inga slutsatser kunde dras gällande skillnader mellan personer med femoral anteversion och femoral retroversion. Mer forskning behövs i området gällande anteversionsvinklar och dess påverkan på muskelaktivitet.

Composition et atteintes musculaires du quadriceps des patients atteints de la maladie pulmonaire obstructive chronique

Introduction : Les patients atteints de la maladie pulmonaire obstructive chronique (MPOC) ont une dysfonction importante au niveau des muscles locomoteurs caractérisés, entre autres, par de la faiblesse, de l’atrophie et des dommages oxydatifs. Certains auteurs ont également suggéré que la fonction mitochondriale soit altérée dans la MPOC, possiblement en lien avec le tabagisme. Objectifs : Évaluer la fonction et le nombre de mitochondries, ainsi que les dommages à l’ADN chez les sujets atteints de la MPOC et déterminer les effets du tabagisme sur les mitochondries et la qualité du muscle, indépendamment de la maladie. Méthodes/Résultats : Nous avons recruté 22 patients atteints de la MPOC et 23 sujets témoins. Tous sont ex-fumeurs et sédentaires. Une biopsie du quadriceps a été effectuée. Nous avons démontré que les sujets atteints de la MPOC ont une plus grande proportion de fibres dont le complexe IV de la chaine de transport des électrons ne fonctionne pas comparativement aux sujets témoins (p=0,032). Ils ont également une diminution de l’expression de la protéine SDHB (succinate dehydrogenase complexe iron sulfur subunit B) (complexe II) comparativement aux sujets témoins (p=0,002), mais celle de la protéine COX4 (cytochrome c oxidase subunit 4) (complexe IV) est similaire entre les deux groupes (p=0,076). Il n’y a pas de différence entre les deux groupes pour la présence de la délétion commune, la quantité de mitochondries, l’expression protéique de MFN1 (mitofusin-1), MFN2 (mitofusin-2), TFAM (mitochondrial transcription factor A) et Mn-SOD (manganese-dependent superoxide dismutase) et la quantité de 8-OHdG (8-hydroxy-2’- deoxyguanosine) présente dans l’ADN. Il y a une corrélation entre l’exposition cumulative au tabac (nombre de paquets-année) et la quantité de 8-OHdG (p=0,026), la proportion de fibres déficientes en COX (p=0,002), l’atténuation musculaire moyenne (p=0,041) et l’atténuation du muscle normal (p=0,020) lorsque tous les sujets (témoins et MPOC) sont regroupés. Conclusion : Nous avons démontré qu’il y a des anomalies de la fonction mitochondriale dans la MPOC. Il est intéressant de noter que le tabac affecte la fonction des mitochondries et la qualité du muscle de façon indépendante à la MPOC. / Rationale : Patients with chronic obstructive pulmonary disease (COPD) demonstrate a locomotor muscle dysfunction characterized by weakness, atrophy and poor oxidative capacity. Some authors also suggested that the mitochondrial function might be altered in this disease, possibly linked with smoking. Objectives : To evaluate the function and the number of mitochondria, and DNA damage in patients with COPD and to determine the effects of smoking on the mitochondria and the quality of the muscle, independently of the disease. Methods/Results : We recruited 22 patients with COPD and 23 healthy subjects. All were exsmokers and sedentary. A biopsy of the quadriceps was performed. We demonstrated that patients with COPD had a larger proportion of COX (cytochrome c oxidase)-deficient fibers than healthy subjects (p=0.032). They also had a decreased expression of the protein SDHB (succinate dehydrogenase complex iron sulfur subunit B) (complex II) compared to controls (p=0.002), but no change in the expression of the protein COX4 (cytochrome c oxidase subunit 4) (complex IV) (p=0.076). There was also no difference between the two groups for the common deletion, the quantity of mitochondria, the protein expression of MFN1 (mitofusin-1), MFN2 (mitofusin-2), TFAM (mitochondrial transcription factor A) and Mn-SOD (manganese-dependent superoxide dismutase) and the quantity of 8-OHdG (8- hydroxy-2’-deoxyguanosine) in the DNA. There was a correlation between the cumulative smoking exposure (pack-year) and the quantity of 8-OHdG (p=0.026), proportion of COX-deficient fibers (p=0.002), mean muscle attenuation (p=0.041) and attenuation of the normal muscle (p=0.020) when all subjects (healthy and COPD) were regrouped. Conclusion : We demonstrated evidences of mitochondrial dysfunction in COPD. Interestingly, there was an effect of smoking on the mitochondria and the quality of the muscle, independently of COPD.
