The aim of this work is to revitalize the public space of náměstí Míru on Kraví hora. The work tries to find further functional use of the, which would include rebuilding the currently unused former military premises at Lerchova street while respecting the current state of the surrounding buildings and greenery. The proposed solution of a new square design signifies urban space as the center of the Masaryk's Quarter. The design takes into account the current location of the tramway balloon loop, which is located in the middle of the area and shapes the unique character of the location. This element of transport is thus left at the heart of the entire urban design. It forms an islet made up of designed buildings with a square created inside. The intention is to build a lively core of the district attracting attention not only from a local populace.

Efekt tlumiče na limity vozidla / Effect of Shock Absorber on Vehicle Limits

The aim of this master’s thesis is to evaluate the effect of shock absorber on vehicle limits. At the beginning of the thesis, shock absorber properties were described. Then computational model was created and manoeuvres for shock absorber behaviour were defined. Created mathematical model is based on quarter model of a car and excitation in form of road with a random profile is an essential part of the model. This model was used for evaluation of heave. After heave analysis, shock absorber behaviour during drive was investigated. Drive conditions were defined as set of handling manoeuvres. For the drive investigation, complete multibody virtual model of racing car was used. Based on drive investigation analysis, optimal damping characteristics for each manoeuvre were found. Furthermore, each optimal characteristic was compared for different manoeuvres. Obtained results were compared. As a conclusion, compromise damping characteristic was suggested with the aim to fit the combination of all defined drive conditions. Final part of the thesis was aimed at validation of the computational model. Data measured during real drive were used as an input for this validation.

Furthermore, a quarter model of the car was created in the Simulink software, where a procedure for evaluating the condition of the shock absorber while driving the vehicle was created. This procedure was subsequently validated on real measured data from the Hummer H3 car.

Third section introduced two structural modification of rectangular patch antenna and measurements of return loss |S11|.

The Unwanted Horse Population: A Survey

Unwanted horses are a growing problem in the United States, and through the use of surveys, researchers can understand public opinion and experiences related to this problem. This study administered surveys specific to horse and non-horse owners through face-toace delivery and to rescue/retirement and auction facilities through the mail. Data was analyzed using a CRD, GLM and Fisher’s Protected LSD with á of 0.05. Comparisons were completed between horse and non-horse owners, horse owners and non-horse owners with equine experience, non-horse owners with and without equine experience, Quarter horse and other breeds owners, and auction and rescue/retirement facilities. Similarities were seen between groups for questions associated with participant location, understanding of processing/harvesting regulations and procedures, processing/harvesting facility restrictions preferences, and knowledge of management cutbacks and rescue facility locations. The only question that was significantly different for all comparisons was concerning the support towards the reopening of equine processing/harvesting facilities.

High efficiency is an essential requirement for any system, where the energy can be saved with full retention of system performance. The power amplifier in modern mobile communications system consumes most of the supplied power through the dissipated power and the required cooling systems. However, as new services were added as features for the developed mobile generations, the required data rate has increased to fulfil the new requirements. In this case, the data should be sent with the allocated bandwidth, so complex modulation schemes are used to utilise the available bandwidth efficiently. Nevertheless, the modulated signal will have a Peak to Average Power Ratio (PAPR) which increases as the modulation complexity is increasing. In this case, the power amplifier should be backed off and designed to provide good linearity and efficiency over high PAPR. Among the efficiency enhancement techniques, the Doherty technique (Load modulation technique) is the simplest one, where no additional circuity nor signal processing is required. In this work, the theory of load modulation amplifiers is investigated through two asymmetrical Doherty Power Amplifiers (DPA) targeting 3.3-3.5 GHz were designed and fabricated using two transistors (25 W and 45 W). In addition, more compact load modulation amplifiers targeting sub 6-GHz bandwidth of 5G specifically 3.4-3.8 GHz is discussed including the theory of implementing these amplifiers, where different amplifier capabilities are explored. Each amplifier design was discussed in detail, in which the input and output matching networks were designed and tested in addition to the design of the stability circuit to make sure that the amplifier is stable and working according to the specified requirements. The fabricated circuits were evaluated practically using the available instrument test, whereas Microwave Office software was used for the simulation purpose, each amplifier was designed separately, where all the designed amplifiers were able to provide the targeted efficiency at different back-off power points. A Novel Method for Vibration Analysis of the Tire-Vehicle System via Frequency Based Substructuring

Noise and vibration transmitted through the tire and suspension system are strong indicators of overall vehicle ride quality. Often, during the tire design process, target specifications are used to achieve the desired ride performance. To validate the design, subjective evaluations are performed by expert drivers. These evaluations are usually done on a test track and are both quite expensive and time consuming due to the several experimental sets of tires that must be manufactured, installed, and then tested on the target vehicle. In order to evaluate the performance, expert drivers tune themselves to the frequency response of the tire/vehicle combination. Provided the right models exist, this evaluation can also be achieved in a laboratory. The research presented here is a method which utilizes the principles of frequency based substructuring (FBS) to separate or combine frequency response data for the tire and suspension. This method allows for the possibility of combining high fidelity tire models with analytical or experimental suspension data in order to obtain an overall response of the combined system without requiring an experimental setup or comprehensive simulations. Though high fidelity models are not combined with experimental data in the present work, these coupling/decoupling techniques are applied independently to several quarter car models of varying complexity and to experimental data. These models range from a simplified spring-mass model to a generalized 3D model including rotation. Further, decoupling techniques were applied to simulations of a rigid ring tire model, which allows for inclusion of nonlinearities present in the tire subsystem and provides meaningful information for a loaded tire. By reducing the need for time consuming simulations and experiments, this research has the potential to significantly reduce the time and cost associated with tire design for ride performance. In order to validate the process experimentally, a small-scale quarter car test rig was developed. This novel setup was specifically designed for the challenges associated with the testing necessary to apply FBS techniques to the tire and suspension systems. The small-scale quarter car system was then used to validate both the models and the testing processes unique to this application. By validating the coupling/decoupling process for the first time on the tire/vehicle system with experimental data, this research can potentially improve the current process of tire design for ride performance. / Ph. Several analytical tire and suspension models were developed for this work and the process of separating/combining the frequency data was performed. Then, a small scale test system was developed and used to establish experimental procedures to collect the data necessary to carry out the Dynamic Substructuring techniques. Finally, the process was validated by repeating the process of separating/combing the frequency properties of the experimental data.

An Assessment Of The Accuracy Of The Euler-Bernoulli Beam Theory For Calculating Strain and Deflection in Composite Sandwich Beams

This study focuses on assessing the accuracy of the Euler-Bernoulli beam theory as computational bases to calculate strain and deflection of composite sandwich beam subjected to three-point and four-point bending. Two groups of composite sandwich beams tests results will be used for comparison purposes. Mechanical properties for the laminated skin are provided by researchers from University of Mississippi (Ellen Lackey et al., 2000). Mechanical properties for the balsa wood core are provided by Alcan Baltek Corporation. Appropriate material properties and test geometries are then used in the Euler-Bernoulli-based algorithm in order to generate analytical data for comparison to experimental data provided by researchers from University of New Orleans (UNO, 2005). The resulting single material cross section is then analyzed in the traditional manner using the Euler-Bernoulli beam theory. In general, the Euler-Bernoulli beam theory provides an appropriate analytical approach in predicting flexural behavior of composite sandwich beams.

Esta dissertação trata dos conflitos entre distintos agentes sociais pelo espaço urbano na metrópole paulistana. Para tanto, descreve e analisa dois eventos ocorridos na região sudoeste: a compra de setenta barracos por parte de uma construtora na favela Jardim Panorama e uma reintegração de posse na favela Real Parque. Depreendem-se da descrição e da análise de ambos os eventos basicamente três fenômenos: o aprofundamento do processo de privatização da gestão urbana já observado em outros estudos, sobretudo na região sudoeste da metrópole; a reconfiguração do campo de conflito expresso na relativa diminuição da presença dos movimentos sociais urbanos e do poder público como agente mediador dos eventos em questão, que foram protagonizados por empresas, ONGs, advogados e movimento hip-hop; por fim, apresentam-se mecanismos de deslegitimação da população pobre como interlocutora. Para esta dissertação, esses conflitos constituem processos de segregação sócioespacial. / This dissertation deals with the conflicts between distinct social agents for the urban space in the São Paulo metropolis. In such a way it describes and analyzes two events occurred in the southwestern region: the purchase of seventy barracos (precarious dwelling) by a constructor in the slum quarter Jardim Panorama and a reintegration of ownership in the Real Parque slum quarter. From the description and the analysis of both events, we basically infer three phenomena: the deepening of the process of privatization of the observed urban management, already observed in other studies, specially in the southwestern region of the metropolis; the reconfiguration of the field of express conflict in the relative reduction of the presence of the urban social movements and the public power as mediating agent of the events in question, that had been carried out by private companies, NGOs, lawyers and hip-hop movement; and finally, mechanisms of disqualification of the poor population as interlocutor are presented. For this dissertation, these conflicts constitute processes of social-spatial segregation.

Detecção e contagem de Staphylococcus aureus causador da mastite bovina em amostras de leite pelo método de quantificação da reação em cadeia da polimerase em tempo real / Detection and counting of bovine mastitis causative Staphylococcus aureus in milk samples by real-time polymerase chain reaction method

Os objetivos do presente estudo foram os de verificar a validade do método de reação em cadeia da polimerase em tempo real (qPCR) para detectar e quantificar o Staphylococcus aureus em amostras de leite conservadas com bronopol oriundas de quartos mamários bovinos subclinicamente infectados, e de avaliar os efeitos da presença e da quantidade de células da bactéria sobre a contagem de células somáticas (CCS), a composição do leite (lactose, gordura, proteína bruta, proteína verdadeira e caseína), e a produção de leite de quartos mamários bovinos subclinicamente infectados pelo patógeno. Para a quantificação do S. aureus e das células somáticas bovinas por meio do qPCR, foi utilizado leite cru bovino para o preparo dos padrões como meio de diluição da inoculação seriada de células somáticas e do S. aureus ATCC 29213, e construídas as equações log10UFC = 37,86 23,54 log10CtSAU e log10CCS = 49,3 - 34,0 log10CtBMCB, com base nos resultados obtidos pelas metodologias de referência para cada procedimento. Para testar a equivalência dessas equações aos respectivos métodos de referência, determinar a sensibilidade e especificidade analíticas e a repetibilidade do método proposto, foram coletadas amostras de leite dos quartos mamários de 60 animais de 2 rebanhos leiteiros da região de Pirassununga dos quais se determinou previamente a ocorrência de casos subclínicos de mastite por S. aureus. Dos quartos mamários também foram mensuradas as produções e coletadas amostras de leite para análise de composição, diagnóstico da mastite, e determinação da sensibilidade e especificidade diagnósticas do procedimento de qPCR estabelecido no estudo. Cada amostra foi submetida à análise de composição, CCS, cultura microbiológica, contagem em placas do S. aureus, processadas para a extração do DNA genômico bovino e do S. aureus, e submetida à reação de qPCR. Para análise da concordância entre os resultados obtidos pelos métodos de referência para o diagnóstico da mastite por S. aureus e o de qPCR foi utilizado o teste Kappa. Para avaliação da equivalência das contagens obtidas pelos métodos de referência do S. aureus e de células somáticas bovinas, foi utilizado o teste das diferenças de Bland-Altman. Para a identificação do efeito da infecção subclínica pelo S. aureus sobre a composição e produção de leite do quarto mamário afetado foi utilizada a análise da variância num delineamento em parcelas subdivididas em faixas. Para estimar o grau de relação entre as contagens de S. aureus, a CCS, produção e composição do leite produzido pelo quarto mamário afetado foi utilizado o coeficiente de correlação de Pearson. A correlação entre os resultados de contagem de células somáticas bovinas determinados pelos métodos de rotina e de qPCR para a quantificação de células somáticas apresentou coeficiente r = - 0,978 (P < 0,001). A correlação entre os resultados da contagem do S. aureus ATCC 29213 determinados pelos métodos de rotina e de qPCR para a quantificação do patógeno apresentou coeficiente r = - 0,989 (P < 0,001). A especificidade analítica do qPCR para a detecção do S. aureus em amostras de leite frente a Escherichia coli, Enterococcus sp., Pseudomonas aeruginosa, Streptococcus agalactiae, Streptococcus dysgalactiae, Streptococcus uberis, os estafilococos coagulase-negativa, e as espécies coagulase-positiva Staphylococcus hyicus e Staphylococcus intermedius foi de 100%. O método de qPCR aplicado à detecção de Staphylococcus aureus ATCC 29213 em amostras de leite é replicável e apresentou sensibilidade analítica com limite de detecção para a faixa de 10 UFC/mL à 4,2 x 106 UFC/mL. Em amostras de leite conservadas com bronopol provenientes de quartos mamários subclinicamente infectados, o S. aureus pôde ser detectado, mas não pôde ser quantificado pelo método de qPCR. Nessas amostras, a CCS pôde ser determinada de forma equivalente ao método de rotina. A CCS independe da contagem de S. aureus viáveis, mas foi observada correlação linear e negativa entre o número total de células do patógeno e a CCS. A mastite subclínica pelo S. aureus aumentou a CCS nos quartos mamários, mas não alterou a composição do leite. A doença diminuiu a produção de leite e de gordura dos quartos mamários anteriores acometidos pela infeção, mas não se observou efeito da interação entre o posicionamento da glândula e a infecção sobre a produção de leite. Houve correlação entre as concentrações de lactose (r = 0,42; P = 0,0051), de gordura (r = 0,46; P = 0,0016), de produção de gordura (r = 0,49; P = 0,001), e de leite com produção ajustada para o teor de 3,5% de gordura (r = 0,41; P = 0,006), e o número de S. aureus presentes na amostra de leite. / The objectives of this study were to verify the validity of the real-time polymerase chain reaction (qPCR) to detect and quantify Staphylococcus aureus in bronopol-preserved milk samples from subclinically infected mammary quarters, and to assess the effects of the presence and amount of the pathogen on the somatic cell count (SCC), the composition of milk and milk yield of bovine mammary quarters subclinically infected by the pathogen. In order to quantify S. aureus and bovine somatic cells through qPCR, raw bovine milk was used as a means of serial inoculation media of somatic cells and S. aureus ATCC 29213. From that, equations based on the reference methods for each procedure were built, log10UFC = 37,86 23,54 log10CtSAU and log10CCS = 49,3 - 34,0 log10CtBMCB, respectively. To test their equivalence with the reference methods, determine the analytical sensitivity and specificity, and repeatability of the proposed method, milk was sampled from quarters of 60 animals from two dairy herds in Pirassununga, where subclinical S. aureus mastitis cases had been previously diagnosed. Also, quarter milk yield had been measured and samples collected for milk composition analysis, diagnosis of mastitis, sensitivity and specificity of the procedure established in the study had been determined. Each sample was subjected to composition analysis, SCC, microbiological culture, plate counting of S. aureus, DNA extraction, and subjected to qPCR reaction. Agreement between results from reference methods and qPCR for the diagnosis of mastitis by S. aureus was assessed by Kappa test. Equivalence between S. aureus, SCC scores obtained by reference and qPCR was assessed with Bland-Altman procedures. The effect of S. aureus subclinical infection on milk composition and milk yield of affected quarters was measured using a strip plot design. To estimate the degree of relationship between the counts of S. aureus, SCC, yield and composition of the milk from affected quarters was assessed by the Pearson Correlation. Correlation between SCC determined by routine methods and qPCR was r = - 0.978 (P <0.001). Correlation between S. aureus ATCC 29213 determined by routine methods and qPCR was r = - 0.989 (P <0.001). Analytical specificity of qPCR to detect S. aureus in milk samples against Escherichia coli, Enterococcus sp., Pseudomonas aeruginosa, Streptococcus agalactiae, Streptococcus dysgalactiae, Streptococcus uberis, coagulase-negative staphylococci and coagulase-positive species, Staphylococcus hyicus and Staphylococcus intermedius was 100%. The use of the qPCR to detect S. aureus ATCC 29213 in milk samples is replicable. Analytical sensitivity detection limit of the method ranged from 10 CFU/mL to 4.2 x 106 CFU/mL. S. aureus could be detected, but not quantified by qPCR in bronopol-conserved milk samples from subclinically infected quarters. In these samples, SCC could be determined by qPCR as it had been done by routine method. SCC was not dependent on S. aureus viable cells, but a negative linear correlation between the total number of cells of the pathogen and SCC was observed. S. aureus subclinical mastitis increased quarters SCC, but did not change milk composition. The disease decreased quarter milk and fat yield, but no interaction effect was observed between the gland positioning and S. aureus subclinical infection on milk production. Correlations between lactose (r = 0.42, P = 0.0051), fat (r = 0.46, P = 0.0016), fat yield (r = 0.49, P = 0.001), and 3.5% fat adjusted milk yield (r = 0.41, P = 0.006), and the number of S. aureus present in the milk sample were observed.
