Caracterização do perfil sorológico de nulíparas suínas e da progênie, frente ao parvovírus suíno / Serological characterization of gilts and progeny, under Porcine Parvovirus

O parvovírus suíno (PPV) apresenta grande importância, principalmente em fêmeas nãoimunes, por causar perdas reprodutivas significativas. O primeiro trabalho foi desenvolvido sobre forma de revisão, e serviu como base para realização dos estudos seguintes. O segundo trabalho foi conduzido para determinar a resposta de anticorpos para PPV em 127 leitoas após a vacinação, avaliar a transferência de imunidade passiva e estimar a queda de anticorpos colostrais para PPV na leitegada. Foi realizada coleta de sangue nas leitoas em: (A) antes da primeira vacinação para PPV, (B) após a segunda dose; (C) no parto e (D) durante a segunda gestação. Além disto, colostro também foi coletado (E). Três leitões de cada fêmea foram selecionados e amostras de sangue foram coletadas: antes de mamar o colostro, 7, 21, 57, 87 e 128 dias de idade, a fim de verificar o declínio da imunidade passiva e estimar a meia-vida de anticorpos para PPV. O número de fetos mumificados, natimortos, nascidos vivos e nascidos totais do primeiro e segundo parto foram analizados. Os anticorpos para PPV foram testados por inibição da hemaglutinação (HI) e enzymelinked immunosorbent assay (ELISA), a fim de verificar a concordância entre estes dois métodos. A possível associação entre os títulos de anticorpos das fêmeas e dos leitões no soro e no colostro com os dados reprodutivos também foi investigada. A maioria das fêmeas (85,83%) tiveram anticorpos para PPV antes da vacinação, mas depois da vacina, todas as fêmeas soroconverteram. Aos sete dias de idade a maioria dos leitões apresentaram anticorpos para PPV e em torno dos 57 dias de idade somente 35,29% dos leitões eram positivos, alcançando a nulidade de anticorpos para PPV aos 87 dias de idade. A meia-vida estimada dos anticorpos colostrais foi 29,80 dias. A correlação entre o soro dos leitões e da fêmea no momento do parto foi r=0,77 (P<0,001) e com o colostro o valor de r foi 0,72 (P<0.001). A concordância entre os testes de ELISA e HI foi moderada (Spearman’s ρ= 0,89 e R2= 0.67). Houve diferença somente no número de mumificados entre o primeiro e segundo parto (P<0,001). O terceiro trabalho objetivou avaliar o perfil de anticorpos para PPV em diferentes sistemas de reposição de leitoas, correlacionando com dados reprodutivos. Cento e cinquenta 11 nulíparas com duas doses de vacina para PPV foram selecionadas de três sistemas de reposição diferentes: quarto sítio - A (n=36), granja receptora do quarto sítio - B (n=57) e granja multiplicadora - C (n=57). Os anticorpos para PPV foram medidos utilizando um teste de ELISA. Houve diferença entre as três granjas com relação ao título de anticorpos (P<0,05). Ao comparar os dados reprodutivos entre as granjas, houve diferença entre elas no número de nascidos totais e nascidos vivos, mas não foi observada diferença no percentual de natimortos e de mumificados (P>0,05). A correta preparação da leitoa, objetivando a proteção no momento da cobertura é fundamental para alcançar bom desempenho reprodutivo, independente do sistema de reposição utilizado. / Porcine parvovirus (PPV) has a great importance because causes significantly reproductive losses, mainly in non-immune gilts. The first study was developmented as a review, and served as a basis to carry out the following studies. The second study was conducted to determine the antibody response for PPV of 127 gilts in field conditions after vaccination, to evaluate the transfer of passive immunity and to estimate the decay of acquired colostral antibodies to PPV in the littermate. Gilts were bled at: (A) before the first vaccination to PPV, (B) after the second dose; (C) at farrowing and (D) during the second pregnancy. Added to these, colostrum was also collected (E). Three piglets of each gilt were selected and blood samples were collected: prior to initial colostrum intake, 7, 21, 57, 87 and 128 day-old, in order to verify the decrease of passive immunity and estimate the half-life of PPV antibodies. The number of mummified fetus, stillbirths, born alive and total born were analyzed from first and second parturition. The PPV antibodies were tested both with haemagglutination inhibition (HI) and enzyme-linked immunosorbent assay (ELISA) in order to study the agreement between these methods. The possible association between gilts and piglets antibody titers in serum and colostrum with reproductive data was also investigated. Most gilts (85.83%) had antibodies to PPV before vaccination, but after vaccine, all gilts seroconverted. At 7 day-old most part of piglets had PPV antibodies and around 57 days-old only 35.29% of piglets were positive, reaching the PPV antibodies nullity at 87 days-old. The estimated average half-life of acquired colostral antibodies was 29.80 days. The correlation between piglets serum with gilt serum at farrowing time was r=0.77 (P<0.001) and with colostrum the r value was 0.72 (P<0.001). The agreement between ELISA and HI tests was moderate (Spearman’s ρ= 0.89 and R2= 0.67). The only difference between first and second parturition was observed on mummified fetuses (P<0.001). The objective of the third study was to evaluate the PPV antibodies profile in different gilts replacement systems, correlating with reproductive data. A hundred and fifty gilts with two doses of 13 PPV vaccine were selected from three different gilts replacement systems: Fourth site - A (n=36), fourth site receiver herd - B (n=57) and a farm producing dam lines - C (n=57). The PPV antibodies were measured by an ELISA test. There were a difference on antibody titers among the three herds (P<0.05). When we compared the reproductive data among herds, there were difference on total born and born alive, but this difference was not observed on the percentual of stillbirths and mummified (P>0.05). The correct gilt preparation, aiming the protection on mating time is fundamental to reach a great reproductive performance, independent of the replacement gilt system used.

Performance of No Vibration/No Admixture Masonry Grout Containing High Replacement of Portland Cement with Fly Ash and Ground Granulated Blast Furnace Slag

When hollow concrete masonry is used for construction in high seismic regions, structural designs typically require fully grouted walls. The grouting process is labor-intensive, time-consuming and has a high energy demand due to requirements of consolidation in each and subsequent grout lifts. Self-consolidating grout with admixtures has been successfully used without segregation in walls of up to 12.67 ft. in height. Investigation of self-consolidating grout mixes without admixtures has potential for sustainability improvement. This thesis reports on the compression strength and consolidation observations of self-consolidating characteristics of no vibration/no admixture grout made by substituting various proportions of Portland cement with Type F fly ash and/or ground granulated blast furnace slag (GGBFS). The percentages of Portland cement replacement evaluated were 0%, 50%, 60%, and 70% for Type F fly ash. The percentages of Portland cement replacement evaluated were 0%, 60%, 70% and 80% for Type F fly ash and GGBFS. Grout compressive strengths were evaluated from individually filled grout specimens constructed in concrete masonry hollow core units, dry cured, and tested after 7, 14, 28, 42, 56, and 130 days. Also, hollow concrete masonry walls were built 12.67 ft. tall and grouted. The relative performance was assessed by comparing to conventional grouted masonry and evaluating consolidation characteristics around mortar fins and reinforcement; compressive strength tests after 130 days of curing, and rebar pull-out tests were taken from various wall heights. All experimental grouts had acceptable consolidation characteristics but fly ash replacement grouts did not meet the compressive strength requirements.

Analysis of hindfoot alignment for total ankle arthroplasties

Total ankle arthroplasties (TAAs) are mechanical devices used to replace the articular surfaces of the ankle joint in order to relieve pain for patients with osteoarthritis. Since most osteoarthritis is post-traumatic, and due to the highly variable individual foot geometry, TAAs are rarely inserted into normal geometry. This leads to serious problems with stresses and contact pressures in TAA components. This study uses finite element (FE) modeling to determine how hindfoot alignment, or how far in varus or valgus the most distal part of the calcaneus is perpendicularly from the axis of the tibia, affects the stresses and contact pressures in the articulating surfaces of two different TAA models. To investigate the effects of foot alignment on hardware stresses after TAA, FE models were generated. Models of the mobile bearing, three component Scandinavian Total Ankle Replacement (STAR) and the fixed bearing, two component Zimmer Trabecular Metal Total Ankle (Zimmer) were generated from laser scans of the hardware and virtually implanted into 3D models of the tibia and talus. Ligaments were modeled as linear springs to impart physiologically realistic flexibility in the model. The stance phase of a walking gait cycle was applied and stresses and contact pressures at the articulation between model components were recorded for various degrees of hindfoot alignment [1]. Data analyzed shows that both models have areas of high concentrations of stress and contact pressure. The Zimmer TAA seems to favor a valgus alignment due to the lower stresses and contact pressures in valgus alignments compared to varus. Though the STAR does not generally favor one alignment over the other, it does have significantly lower stresses and contact pressures than the Zimmer. These differences may be due to the geometric congruency of the STAR versus the anatomical articulation of the Zimmer.

Revenue Recovery Through Meter Replacement

Previous studies have identified water meter inaccuracy at low flow rates as a significant source of non-revenue flow for water systems; however a lack of available data makes it difficult to include low flow accuracy degradation in meter replacement plans. This thesis examines results from an extensive accuracy test program carried out at the Utah Water Research Laboratory on small water meters over a wide range of flow rates and at various levels of throughput. The study compares expected apparent losses of different types of water meters based on a flow profile and expected daily use for the State of California. By including an average composite charging rate, use of the method developed in this study can determine the meter replacement payback period for different meter types. The analysis contained in this document is intended as a guide to assist utility managers when developing meter replacement plans.

Experimental study of zeotropic refrigerant mixture HFC-407C as a replacement for HCFC-22 in refrigeration and air-conditioning systems

HCFC-22 is the world�s most widely used refrigerant. It serves in both residential and commercial applications, from small window units to large water chillers, and everything in between. Its particular combination of efficiency, capacity and pressure has made it a popular choice for equipment designers. Nevertheless, it does have some ODP, so international law set forth in the Montreal Protocol and its Copenhagen and Vienna amendments have put HCFC-22 on a phase out schedule. In developed countries, production of HCFC-22 will end no later than the year 2030. Zeotropic blend HFC-407C has been established as a drop-in alternative for HCFC-22 in the industry due to their zero Ozone Depletion Potential (ODP) and similarities in thermodynamic properties and performance. However, when a system is charged with a zeotropic mixture, it raises concerns about temperature glide at two-phase state, differential oil solubility and internal composition shift. Not enough research has been done to cover all aspects of alternative refrigerants applications in the systems. This research intended to explore behavior of this alternative refrigerants compare to HCFC-22 and challenges facing the industry in design, operation service and maintenance of these equipments. The purpose of this research is to investigate behavior of R407C refrigerant in chiller systems. This includes performance and efficiency variations when it replaces R22 in an existing system as well as challenges involved maintaining the system charged with R407C. It is a common practice in the industry these days to evacuate and completely recharge when part of the new refrigerant blend was leaked from the system. This has proved to be extremely costly exercise with grave environmental ramifications. This research is intended to address challenges faced in the real world and practical terms. Theoretical and experimental approaches used as a methodology in this work. The system mathematically modeled to predict detailed system performance and effect of the leak at various conditions. To make this feasible and accurate enough, two separate approaches made, first system performance for pure R22 and R407C, and second system subjected to range of leak fractions. The earlier model was relatively straight forward when compared to the latter. Modeling a system charged with R407C ternary mixture and subjected to range of leaks posed enormous challenges. A sophisticated experimental test apparatus was also designed and built. Comprehensive and detailed tests at various conditions were conducted with special attention on instrumental accuracy and correct methodology. The first part has been successfully modeled and predicted all the factors and performance with excellent accuracy when compared to the test results. In these approaches pure refrigerants R22 and R407C were used and simulated the system behavior at range of conditions. However, the second part was the most challenging ever. Comprehensive leak process simulations produced trends of R32/R125/R134a composition change as function of rate of leak. Starting from this point, equations have been created to represent the composition change as function of percentage of the leak. The system thermodynamic cycle was also modeled to calculate capacity, power input and COP at the range of the conditions. Despite many affecting parameters and complexity of the model, the mathematical model successfully predicted the test outcome with a very reasonable accuracy, averaging around 3% with some times reaching to 5 to 6%. On the experimental stage the system charged with the new HFC-407C was deliberately subjected to refrigerant leak at various leak stages. The aim was to objectively determine to what extend the gas leak can be still acceptable without going through the expensive complete gas charge. The effect of leak was tested and verified at 10% steps, from 10% up to 50% mass fraction for the total charge. It has been observed that at the leaks beyond 30%, the adverse effect on the capacity becomes more significant, from 8 to about 15% decrease. While the power input decreased at slower pace, from 3% up to about 8% depending on the test conditions. This translated to COP decrease ranging from 4 to about 7%. This capacity loss and efficiency decrease are significant figures which suggests that the system, here chiller, can not be allowed to degrade the performance to that extend and still continue operating.

Energy Balanced Sensor Node Organisation For Maximising Network Lifetime

Recent advances in Micro-Electro-Mechanical Systems (MEMS) and low-power short-range radios have enabled rapid development of wireless sensor networks. Future sensor networks are anticipated to include hundreds or thousands of these devices in many applications, such as capturing multimedia content for surveillance, structural health monitoring, tracking of accidental chemical leaks, machine failures, earthquakes and intrusion detection. With the increase of sensor applications, a number of challenging problems related to the network protocol design has emerged - the most important ones relating to energy efficiency and lifetime maximisation. Techniques devised for sensor networks should deal with a large number of sensors distributed in the field. Wireless sensor nodes are deployed with limited energy reserves, so the networks should operate with minimum energy overhead. In fact, the network should take into account not only individual node's energy efficiency but also consider the global picture, because surviving nodes' energy reserves in a failed network are wasted energy. This thesis examines a node organisation technique to deal with the above challenges. The focus is on improving network lifetime via organising the nodes in a distributed and energy efficient manner. The main goal is lowering wasted energy via energy balancing and exploiting node redundancy in case of node failure. In particular, this thesis proposes Energy Balanced Clustering (EBC) method for node self-organisation where network tasks (such as data aggregation and data forwarding) are shifted to high-energy neighbours to reduce the energy consumption of low energy nodes. After showing how to extend network lifetime by energy balanced node organisation, the effect of redundant node deployments on network lifetime is addressed. Redundant nodes consume energy by performing unnecessary tasks so a method called Self-Calculated Redundancy Check (SCRC) is proposed to deactivate redundant nodes. A deactivated redundant node can be used as a replacement for a failed node. The Asynchronous Failed Sensor node Detection (AFSD) proposed in this thesis uses the data packets exchanged between neighbours to identify failed neighbours. To restore coverage for network holes caused by failed nodes, policies are given for re-activating redundant nodes. Detailed analytical analysis and simulation of the proposed methods demonstrate that by taking into account energy balancing, eliminating redundant tasks and replacing failed nodes sensor network lifetime can significantly be improved.

Risk Talk : On Communicating Benefits and Harms in Health Care

One of the most critical elements in empowering the patient, and ensuring concordance, is communication of the possible benefits and harms of different actions in health care. Risk assessment is a complex task due both to the different interpretations of the concept of risk, and the common lack of hard facts. Hormone, or hormone replacement, therapy (HT) is used by many women in, and after, the menopause. The benefits and possible harms associated with short and long term treatment with HT have been extensively discussed the last decade and the use of HT has decreased dramatically internationally the last few years. The aims of this thesis were to study the interaction between patient and physician when discussing risks and benefits of different treatment alternatives, and to suggest strategies to improve risk communication in clinical practice. The studies have focused on how risks and benefits with HT were communicated between women and physicians during firsttime consultations in 1999- 2000 on this subject (20 women, 5 gynaecologists), and through questionnaires how attitudes towards HT have changed between 1999 (n=1,760) and 2003 (n=1,733) among women entering the menopause (53-54 years). Through a qualitative analysis of the risk communication in the consultations a system was constructed to classify how risk is communicated in relation to benefits. This was used to assess and present differences in risk communication in the consultations. Different rhetorical strategies by the physicians were identified and the dominating tendency was a move from the woman’s current problems to the long-term effects of HT. The questionnaires showed a marked difference in attitudes towards HT between the years. In 2003 women perceived HT to be associated with higher risk and less benefits than in 1999. This correlated to a drastic reduction in the use of HT over the same period. Media was the most frequent source of information about HT during the last twelve months before the questionnaire in 2003. Possible explanations for the different attitudes towards HT between women entering the menopause and gynaecologist; how this difference might have influenced the results; and how they may have implications for future communication strategies are discussed. This thesis illustrates the importance of a deeper understanding in health care of the concept of risk in order to achieve an adequate communication of risk. Total knee arthroplasty : aspects on improved fixation in the younger patient

The results of total knee arthroplasty are inferior in younger patients. The challenge today is therefore to develop designs and concepts that will last at least 25 years. This thesis has evaluated the fixation to bone of modern designs of knee prostheses uring RSA analysis. Coating implant surfaces with hydroxy-apatite have proven to enhance fixation to bone. Addition of screws for fixation of the tibial component enhances the fixation, but has negative side effects such as osteolysis around the screws, in turn leading to a higher risk of component loosening. The magnitude and pattern of migration was studied in a randomized study of uncemented tibial implants coated with hydroxy-apatite with and without additional screw fixation in patients younger than 65 years. The uncemented implants migrated initially more than the cemented implants that constituted the control group. Both uncemented groups stabilized at 3 monthes with no further migration, while the cemented implants showed a continuous migration up to the 2 year follow-up, indicating continuous bone resorption at the implant-bone interface, a fact that might lead to an increased risk of late implant loosening. This may not be a problem in older patients, but may have consequences for long-term fixation in younger patients. There was no difference between the two uncemented groups indicating that screws do not improve fixation. Hydroxy-apatite coated knee implants might be well suited for younger patients. Mobile bearing total knee arthroplasty theoretically uncouples the forces at the implant-bone interface, thus improving fixation of the implant to bone. The magnitude and pattern of migration of a cemented mobile bearing knee arthroplasty and a fixed bearing total knee arthroplasty was compared in a randomized study. The results showed that mobile bearings did not improve fixation. Trabecular metal, a new material recently introduced for total knee arthroplasty, has several theoretical advantages. Trabecular metal tibial implants were evaluated in a randomized study in patients younger than 60 years. The implants displayed the typical migration pattern for uncemented implants with greater migration initially followed by early stabilization. The majority of the trabecular metal implants subsided into the bone with no lift-off. Lift-off has the potential of exposing the interface to joint fluid with the potential risk of bone resorption and late loosening, and is commonly seen in metal-backed implants. The finding of absence of lift-off is regarded beneficial for uncemented fixation. Trabecular metal tibial implants might be suited for younger patients. The optimal mode of fixation of the femoral component is yet to be established. Comparing cemented femoral components with uncemented femoral components in a randomized study in patients younger than 60 years revealed no differences of the magnitude or the pattern of migration. Uncemented femoral component seems equally as good as cemented components in younger patients.

Hormone replacement therapy : benefits and adverse effects

Background: Numerous studies have shown that estrogen replacement therapy (ERT) is an effective treatment for vasomotor symptoms, insomnia and vaginal dryness. Beneficial effects have also been shown on lipid patterns and on the incidence of osteoporotic fractures. As ERT increases the risk of endometrial adenocarcinoma, combinations with various progestogens have been developed in order to protect the endometrium. However, the addition of progestogens tends to reduce the beneficial effects of estrogens on mood, cognition and lipid metabolism. The added progestogen often causes side effects such as irritability and depression. There is evidence that the effect on wellbeing varies between women and with the type of progestogen used. Women who prefer to avoid withdrawal bleedings can be given continuous combined hormone replacement therapy (HRT). Unfortunately, irregular bleedings are common at the beginning of treatment and reduces compliance. Recently, several studies have reported an increased risk of breast cancer and venous thrombosis, and therefore long-term treatment with HRT for women without climacteric symptoms is no longer recommended. The ongoing debate has, for the time being, resulted in a recommendation that improving quality of life (QoL) by treatment of climacteric symptoms should be the only indication for prescribing HRT. Aims and methods: The aims of the study were to investigate bleeding patterns, changes in wellbeing at onset and during long-term treatment, and lipid and lipoprotein profiles with two different types of continuous combined HRT. In addition, women starting, and women switching from mainly sequential HRT were compared. The design was a randomised, double-blind, one year, prospective, multicentre study including 249 healthy postmenopausal women who were given continuous daily oral treatment with either combined 0.625mg conjugated estrogen (CE) and 5mg medroxyprogesterone acetate (MPA) or combined 2mg 17β - estradiol (E2) and 1mg norethisterone acetate (NETA). Bleedings, if any, were recorded daily throughout the study. The main outcome measures (changes in wellbeing and climacteric symptoms) consisted of daily ratings of 12 items on a validated symptom scale. Serum concentrations of lipids and lipoproteins were measured at baseline and after one year of treatment. Results and conclusions: The majority of drop-outs were confined to the first three months, and the main reasons were bleedings and/or decreased wellbeing. Drop-outs were three times more common in the E2/NETA group. During the first month, 67% of the women reported irregular bleedings. The number of bleeding days decreased on both treatments during the first four months. Treatment with CE/MPA resulted in less irregular bleedings and a shorter time to amenorrhoea compared to E2/NETA. As expected, "starters" experienced more sweats than "switchers" at the onset of treatment, but both groups improved significantly. Side effects such as breast tenderness, swelling, depression and irritability appeared during the first treatment week in both groups. The side effects of HRT appeared much more quickly than the benefits and were more frequent in women with a history of premenstrual syndrome (PMS). Breast tenderness was more common in the E2/NETA group throughout the whole study period. Apart from that, there were no differences between the two treatment regimens as regards effects on well-being at the end of the study. Lipoprotein(a) levels, an important risk factor for cardiovascular disease, decreased in both treatment groups. Triglyceride levels increased in women treated with CE/MPA, and levels of total cholesterol, high density lipoprotein and low density lipoprotein fell in the E2/NETA group. In conclusion, treatment with E2/NETA caused more bleeding problems than treatment with CE/MPA. CE/MPA was better tolerated than E2/NETA at the beginning of the study, but among the women remaining in the study there was no difference in QoL between the two treatment groups. HRT counselling should take into account that a history of PMS increases the likelihood of side effects and that these may precede any beneficial effects. Both treatments produced beneficial effects on lipid and lipoprotein levels, and neither of the regimens was superior in this respect.

Near-surface study of structure-property relationships in electrochemically fabricated multi-component catalysts

This work outlines a series of developments and discoveries related to surface chemistry of controlled near-surface architectures. Through a combination of various X-ray spectroscopy techniques and innovative electrochemical fabrication techniques, valuable knowledge has been added to the fields of electrochemical fabrication, electrocatalysis, and fundamental surface chemistry. Described here is a specific new development in the technique of surface limited redox replacement (SLRR). This work, along with an accompanying journal publication1, reports the first-ever use of nickel as an intermediary for SLRR. In addition, this work identifies specific deviations from the nominal reaction stoichiometry for SLRR-grown films. This led to the proposal of a new reaction mechanism for the initial stages of the SLRR process, which will assist future fabrication attempts in this field. This work also discovered fundamental changes in Pt overlayer systems as the thickness of the overlayer on a gold support is increased from less than a single atomic monolayer to multilayer thicknesses. It was found that Pt overlayers below a certain threshold thickness exhibited increased affinity for hydroxyl groups, along with an increased propensity for formation of oxide and chloride species. These films were also studied for methanol, carbon monoxide, and ethylene glycol electro-oxidation. Finally, this work reports controlled surface architectures of Pt and Cu deposits on application-oriented TiO₂ nanotube arrays and Au-carbon supports.
