Stálost objektu jako metoda pro výzkum vyšších kognitivních funkcí primátů / Object permanence as a method to study higher cognitive functions of primates

Object permanence is a cognitive ability, which allows individual to realize the existence of an object even it is not directly accessible to its senses. This ability is essential for successful using of complex cognitive operations. Object permanence is qualitatively and gradually change throughout the development of a child. Congruently, it is not developed to the same level in various species of animals. The aim of this study is to study object permanence in naive rhesus monkeys (Macaca mulatta), because there is still some uncertainty about the development of this ability in macaques. Our results show that the naive subjects do not have the highest stage of object permanence (and they do not use representative strategy to solve the tasks), however, other results of our team suggest that more experienced individuals are able to achieve the highest stage under certain circumstances. We show that experimental design used to test object permanence can be modified and used also for studying of other cognitive abilities. We test the preferences of macaque monkeys toward novel non-food stimuli. The reactions of different species of animals can vary. Numerical competences include the number of abilities as representation of the quantity and transformations or operations with quantity. Many species of animals can use these abilities for example during searching for food, sexual partners and also for detection of number of competitors or predators. Numerical abilities such as counting or estimating quantity can increase the chance of survival and reproduction. In this work, I summarize some previous studies of numerical competences in primates and humans and I also mention experiments in other species. I divide the competences into chapters: estimating of the numbers (relative and absolute), counting, summation, conservation of quantity, proportion, ordinality and transitivity. The aim of this study was to study the ability of macaques (Macaca mulatta) to distinguish between the quantities of two sets. In the following part of experiment I observed the preference of monkeys for size or the number of stimuli. This preference was studied also in children. The results confirm the ability of relative numerousness judgment of monkeys. The results demonstrate that with increasing difference of quantity between the two sets the ability of distinguish between the two quantities also increase. Habituation and Desensitization as Methods for Reducing Fearful Behavior in Singly-Housed Rhesus Macaques

Operant conditioning using positive reinforcement techniques has been used extensively in the management of nonhuman primates in both zoological and laboratory settings. Based on a large body of previous research that demonstrates the utility of such techniques in reducing stress, abnormal behavior, and aggression, this research project was intended to develop and test the usefulness of habituation and counter-conditioning techniques in reducing the fear-responses of singly-housed male rhesus macaques living in the laboratory environment. Additionally, we investigated the variable of temperament as it relates to the reduction of fear-responsivity and overall training success. Based on a Wilcoxon Matched-Pairs Sign Test, we found that animals exposed to desensitization training were significantly likely to show a reduction in the rate at which they engaged in cringing toward humans (exact significance = .016, one-tailed, N ties = 6), cringing in general (exact significance = .016, one-tailed, N ties = 6), and in stress-related behaviors (exact significance = .016, one-tailed, N ties = 6). Animals exposed to basic husbandry training or exposed to no training at all were not significantly likely to show a reduction in the rates of these behaviors. When these same behaviors were analyzed in terms of duration of behavior, desensitization-exposed animals were significantly likely to show reduction in the amount of time spent cringing toward humans (exact significance = .016, one-tailed, N ties = 6), but not in cringing behaviors in general or in stress-related behaviors. Neither the husbandry-exposed group nor the group exposed to no training showed a significant number of subjects exhibiting a reduction in duration of any of these behaviors. Additionally, initial temperament assessments were found to significantly predict the relative ability of subjects exposed to training to acquire trained behaviors such that animals generally ranked as more inhibited in terms of temperament also ranked as slower learners based on a Wilcoxon Matched-Pairs Signed-Ranks test, z = -.316, p = .752 (two-tailed). Results of this study could enhance both laboratory animal welfare and laboratory animal research, and could be a first step in developing techniques for reducing fearful behavior in rhesus monkeys in the laboratory environment.

Analysis of a quantitative behavioral assessment program to identify and treat abnormal behaviors in captive primates

Facilities housing non-human primates are required to make provisions for their psychological wellbeing, which may include monitoring animals for signs of decreased wellbeing such as the presence of abnormal behaviors or alopecia. By analyzing archival behavioral data collected by the Behavior Management Unit at the Yerkes National Primate Research Center (YNPRC), I aimed to identify behavioral predictors of self-wounding and alopecia and to evaluate the effectiveness of current treatments in reducing abnormal behavior and alopecia in rhesus macaques (Macaca mulatta). The behaviors of self-biting and hair plucking (conditional logistic regression, p < .05) as well as floating limb and self-oral behaviors (Mantel-Haenszel chi-square tests, p < .05) were identified as behavioral predictors of self-wounding. Fear behaviors were associated with an increased risk of developing alopecia (Mantel-Haenszel chi square, p < .05). An inverse relationship was found between alopecia and stereotypic locomotor behaviors such as pacing, with animals who displayed these behaviors being less likely to develop significant hair loss (conditional logistic regression, p < .05). Overall, the type of treatment provided (e.g., additional foraging opportunities, the provision of toys, or the provision of visual barriers) did not predict improvement in levels of abnormal behavior or alopecia (logistic regression, p > .05). The results of these analyses add to the literature on self-wounding and alopecia and will allow refinement of the quantitative behavioral monitoring system at YNPRC such that more at-risk animals can be identified and treated prior to the development of abnormal or harmful behaviors.

When and where will a target go? A behavioural and electrophysiological study of expectation in primates

In a rapidly changing visual environment, the delay between perception and action might impair the probability of survival of a prey or the efficiency of a predator. In order to compensate for delays associated with sensory-motor processing, primates often make predictions about future events and initiate anticipatory movements. To prepare an anticipatory movement, an estimation of when and where to a target is likely to move is necessary. Such an internal representation is often termed 'expectation'. The aim of this thesis was to investigate the gradual changes of a subject's expectation at the behavioral and electrophysiological levels. Anticipatory smooth pursuit was used in order to study temporal and directional changes in expectation. We found that temporal uncertainty strongly modulated the latency and the velocity of anticipatory movements suggesting that monkeys could estimate the hazard rate of target motion onset in order to decide when to initiate an anticipatory movement. In addition, we have shown that monkeys could use prior directional information in order to voluntarily initiate anticipatory responses in the direction of expected target motion. This prior directional information significantly affected the latency and velocity of these movements. Finally, we have shown that the majority of recorded supplementary eye field (SEF) neurons encoded expected target motion direction. The presence of a directional cue induced an increase of activity in the preferred direction of the neuron. Moreover, a large sub-population of neurons encoded the direction of future anticipatory movement. These results suggest that the SEF could be involved in the cognitive control of anticipatory pursuit eye movements when prior temporal and directional information is provided.

Decision-Making in the Primate Brain

<p>Making decisions is fundamental to everything we do, yet it can be impaired in various disorders and conditions. While research into the neural basis of decision-making has flourished in recent years, many questions remain about how decisions are instantiated in the brain. Here we explored how primates make abstract decisions and decisions in social contexts, as well as one way to non-invasively modulate the brain circuits underlying decision-making. We used rhesus macaques as our model organism. First we probed numerical decision-making, a form of abstract decision-making. We demonstrated that monkeys are able to compare discrete ratios, choosing an array with a greater ratio of positive to negative stimuli, even when this array does not have a greater absolute number of positive stimuli. Monkeys’ performance in this task adhered to Weber’s law, indicating that monkeys—like humans—treat proportions as analog magnitudes. Next we showed that monkeys’ ordinal decisions are influenced by spatial associations; when trained to select the fourth stimulus from the bottom in a vertical array, they subsequently selected the fourth stimulus from the left—and not from the right—in a horizontal array. In other words, they begin enumerating from one side of space and not the other, mirroring the human tendency to associate numbers with space. These and other studies confirmed that monkeys’ numerical decision-making follows similar patterns to that of humans, making them a good model for investigations of the neurobiological basis of numerical decision-making. </p><p>We sought to develop a system for exploring the neuronal basis of the cognitive and behavioral effects observed following transcranial magnetic stimulation, a relatively new, non-invasive method of brain stimulation that may be used to treat clinical disorders. We completed a set of pilot studies applying offline low-frequency repetitive transcranial magnetic stimulation to the macaque posterior parietal cortex, which has been implicated in numerical processing, while subjects performed a numerical comparison and control color comparison task, and while electrophysiological activity was recorded from the stimulated region of cortex. We found tentative evidence in one paradigm that stimulation did selectively impair performance in the number task, causally implicating the posterior parietal cortex in numerical decisions. In another paradigm, however, we manipulated the subject’s reaching behavior but not her number or color comparison performance. We also found that stimulation produced variable changes in neuronal firing and local field potentials. Together these findings lay the groundwork for detailed investigations into how different parameters of transcranial magnetic stimulation can interact with cortical architecture to produce various cognitive and behavioral changes.</p><p>Finally, we explored how monkeys decide how to behave in competitive social interactions. In a zero-sum computer game in which two monkeys played as a shooter or a goalie during a hockey-like “penalty shot” scenario, we found that shooters developed complex movement trajectories so as to conceal their intentions from the goalies. Additionally, we found that neurons in the dorsolateral and dorsomedial prefrontal cortex played a role in generating this “deceptive” behavior. We conclude that these regions of prefrontal cortex form part of a circuit that guides decisions to make an individual less predictable to an opponent.</p> / Dissertation

Esta dissertação apresenta um estudo seguido de tradução da tragédia Reso, tradicionalmente atribuída a Eurípides. No estudo, busca-se comparar o tratamento dado na peça ao mito de Reso e ao tema da astúcia (mêtis) com aquele presente em Ilíada X, tomando-se Odisseu como figura central: a partir dele, são abordados, por semelhança e dessemelhança, as demais personagens da peça e a visão de mêtis que cada uma tem ao longo da trama, além da estrutura da tragédia e as relações existentes entre os planos humano e divino. O objetivo principal é mostrar que os dois textos estão bem inseridos nos seus respectivos períodos e ilustram certa transformação no status do conceito estudado. / This research presents a study followed by a translation of Rhesus, a tragedy traditionally attributed to Euripides. In the study, we aim at comparing the treatment the play gives to Rhesuss myth and the theme of cunning intelligence (mêtis) with that presented by Iliad X. Taking Odysseus as the central figure, we make an approach (through similitude and dissimilitude) of the other characters of the play, as well as of the view concerning mêtis that each of them expresses in the action, the structure of the play and the relationship between men and gods. Our main purpose is to show that both texts are well inserted in their respective periods and illustrate some change in the status of the concept of mêtis.

The Rhesus Macaque Corticospinal Connectome

The corticospinal tract (CST), which carries commands from the cerebral cortex to the spinal cord, is vital to fine motor control. Spinal cord injury (SCI) often damages CST axons, causing loss of motor function, most notably in the hands and legs. Our preliminary work in rats suggests that CST circuitry is complex: neurons whose axons project to the lower cervical spinal cord, which directly controls hand function, also send axon collaterals to other locations in the nervous system and may engage parallel motor systems. To inform research into repair of SCI, we therefore aimed to map the entire projection pattern, or “connectome,” of such cervically-projecting CST axons. In this study, we mapped the corticospinal connectome of the Rhesus macaque - an animal model more similar to humans, and therefore more clinically relevant for examining SCI. Comparison of the Rhesus macaque and rat CST connectome, and extrapolation to the human CST connectome, may improve targeting of treatments and rehabilitation after human SCI. To selectively trace cervically-projecting CST motor axons, a virus encoding a Cre-recombinase-dependent tracer (AAV-DIO-gCOMET) was injected into the hand motor cortex, and a virus encoding Cre-recombinase (AAV-Cre) was injected into the C8 level of the spinal cord. In this intersectional approach, the gCOMET virus infects many neurons in the cortex, but gCOMET expression is not turned on unless the nucleus also contains Cre-recombinase, which must be retrogradely transported from axon terminals in the C8 spinal cord. Thus, gCOMET is only expressed in neurons that project to the C8 spinal cord, and it proceeds to fill the entire neuron, including all axon collaterals. Any gCOMET-labeled axon segments observed in other regions of the nervous system are therefore collaterals of cervically-projecting axons. gCOMET-positive axons were immunohistochemically labeled, and axon density was quantified using a fluorescence microscope and Fiji/ImageJ software. Specific regions of interest were chosen for analysis because of their known relevance in motor function in humans, and for comparison to results of a similar study in rats. Results in the first monkey have revealed both similarities and differences between the monkey and rodent CST connectome. Analyses of additional monkeys are ongoing. The final results will provide detailed information about differences between rodent and primate CST, will serve as a baseline for examining changes in the CST connectome after SCI, and will provide guidance for studies targeting treatment and functional recovery after SCI.

Behavioral and neurophysiological investigations of short-term memory in primates

Detecting and interpreting sensory events, and remembering those events in in the service of future actions, forms the foundation of all behavior. Each of these pillars of the so-called "perception-action cycle" have been topics of extensive inquiry throughout recorded history, with philosophical foundations provided by early BCE and CE periods (especially during the Classic and Renaissance eras) leading to intensive empirical study in the twentieth and twenty-first centuries. Such experiments have described detailed (but incomplete) behavioral functions reflecting perception and memory, and have begun to unravel the extraordinarily complex substrates of these functions in the nervous system. The current dissertation was motivated by these findings, with the goal of meaningfully extending our understanding of such processes through a multi-experiment approach spanning the behavioral and neurophysiological levels. The focus of these experiments is on short-term memory (STM), though as we shall see, STM is ultimately inseparable from sensory perception and is directly or indirectly associated with guidance of motor responses. It thus provides a nexus between the sensory inputs and motor outputs that describe interactions between the organism and environment. In Chapter 2, previous findings from nonhuman primate literature describing relatively poor performance for auditory compared to visual or tactile STM inspired similar comparisons among modalities in humans. In both STM and recognition memory paradigms, accuracy is shown to be lowest for the auditory modality, suggesting commonalities among primate species. Chapters 3-5 examined STM processing in nonhuman primates at the behavioral and neurophysiological levels. In Chapter 3, a systematic investigation of memory errors produced by recycling memoranda across trials (proactive interference) is provided for the understudied auditory modality in monkeys. Such errors were ameliorated (but not completely eliminated) by increasing the proportions of unique memoranda presented within a session, and by separating successive trials by greater time intervals. In Chapter 4, previous results revealing a human memory advantage for audiovisual events (compared to unimodal auditory or visual events) inspired a similar comparison in monkeys using a concurrent auditory, visual, and audiovisual STM task. Here, the primary results conformed to a priori expectations, with superior performance observed on audiovisual trials compared to either unimodal trial type. Surprisingly, two of three subjects exhibited superior unimodal performance on auditory trials. This result contrasts with previous results in nonhuman primates, but can be interpreted in light of these subjects' extensive prior experience with unimodal auditory STM tasks. In Chapter 5, the same subjects performed the concurrent audiovisual STM task while activity of single cells and local cell populations was recorded within prefrontal cortex (PFC), a region known to exhibit multisensory integrative and memory functions. The results indicate that both of these functions converge within PFC, down to the level of individual cells, as evidenced by audiovisual integrative responses within mnemonic processes such as delay-related changes in activity and detection of repeated versus different sensory cues. Further, a disproportionate number of the recorded units exhibited such mnemonic processes on audiovisual trials, a finding that corresponds to the superior behavioral performance on these trials. Taken together, these findings reinforce the important role of PFC in STM and multisensory integration. They further strengthen the evidence that "memory" is not a unitary phenomenon, but can be seen as the outcome of processing within and among multiple subsystems, with substantial areas of overlap and separation across modalities. Finally, cross-species comparisons reveal substantial similarities in memory processing between humans and nonhuman primates, suggesting shared evolutionary heritage of systems underlying the perception-action cycle.

Evaluation of sperm functionality in non-human primates, focussing on sperm capacitation

Magister Scientiae (Medical Bioscience) - MSc(MBS) / The incidence of male infertility is increasing, with up to 50% of infertile males having “unexplained” (idiopathic) infertility. Newly developed molecular techniques have great value in detecting subtle causes of male infertility, as compared to idiopathic infertility which may be explained by standardizing and optimizing sperm functional and structural tests in non-human primate (NHP) sperm. The aim of the study was to evaluate sperm functionality utilizing the sperm of two NHP species, i.e.1) the rhesus monkey (Macaca mulatta) and 2) the vervet monkey (Chlorocebus aethiops), and further evaluate the effect of physiological media (including commonly used, and newly formulated sperm wash and sperm capacitating media) on NHP sperm functionality. Sperm functionality was evaluated by investigating the following sperm functions i.e.: sperm motility, vitality, acrosome reaction (AR), hyperactivation, and mitochondrial membrane potential (MMP). Sperm functional tests included computer-aided semen analysis (CASA), motility analysis, BrightVit staining for sperm vitality, flourescenin isothiocyanate (FITC)- conjugated peanut agglutinin (PNA) staining for sperm acrosome integrity, induction of hyperactivation by stimulants (sperm preparation media containing capacitating ingredients), and mitochondrial inhibitor (Oligomycin-A) for testing MMP. All functional and structural tests were investigated in both species, except for acrosome integrity, mitochondrial inhibition and functional tests compared over time that could not be successfully completed and investigated in the rhesus species. Motility analysis tests proved that within the vervet species, the use of different physiological media results in statistically significant differences in motility and kinematic parameters over a 1 hour time period. Hyperactivation tests proved that capacitating physiological media produced significantly higher percentages hyperactivation when compared to sperm wash media within the vervet species over a 1 hour time period. Furthermore, within both NHP species, sperm structural analysis (vitality and acrosome integrity) results showed that no significant differences are present when making use of different physiological media over a period of 1 hour incubation. The incubation of vervet sperm with different concentrations of mitochondrial inhibitor, Oligomycin-A (0 μM, 5 μM, and 25 μM), resulted in motility inhibition over a 1 hour incubation period. By the evaluation of these tests it was found that the use of different sperm wash [Human tubal fluid (HTF), Ham‟s F-10® and HD Sperm Wash Plus (HDSWP)] and sperm capacitation media [Human tubal fluid with added caffeine (HTFC) and HD Sperm Capacitating Plus (HDSCP)] resulted in significantly different results within sperm functional tests as compared to sperm structural tests. The study indicates that the composition of media, varying from simple to more complex, used for semen preparation plays an important role in determining NHP sperm functionality. Based on these findings further investigation in larger NHP sample groups and human sperm are required to evaluate the role of certain ingredients in the development of more cost-effective media producing satisfactory results in terms of sperm functionality for artificial reproductive technologies (ART).
