Rhythmic Growth And Vascular Development In Brachypodium Distachyon

Plants reduce inorganic carbon to synthesize biomass that is comprised of mostly polysaccharides and lignin. Growth is intricately regulated by external cues such as light, temperature, and water availability and internal cues including those generated by the circadian clock. While many aspects of polymer biosynthesis are known, their regulation and distribution within the stem are poorly understood. Plant biomass is perhaps the most abundant organic substance on Earth and can be used as feedstock for energy production. Various grass species are under development as energy crops yet several of their attributes make them challenging research subjects. Brachypodium distachyon has emerged as a grass model for food and energy crop research. I studied rhythmic growth, a phenomenon important to understanding how plant biomass accumulates through time, and vascular system development, which has biofuel feedstock conversion efficiency and yield. Growth rate changes within the course of a day in a sinusoidal fashion with a period of approximately 24 hours, a phenomenon known as rhythmic growth. Light and temperature cycles, and the circadian clock determine growth rate and the timing of rate changes. I examined the influences of these factors on growth patterns in B. distachyon using time-lapse photography. Temperature and, to a lesser extent, light influenced growth rate while the circadian clock had no noticeable effect. The vascular system transports important materials throughout the plant and consists of phloem, which conducts photosynthates, and xylem, which conducts water and nutrients. The cell walls of xylem elements and ground tissue sclerenchyma fibers are comprised of cellulose, hemicelluloses, and lignin. These components are important to alternative energy research since cellulose and hemicellulose can be converted to liquid fuel, but lignin is a significant inhibitor of this process. I investigated vascular development of B. distachyon by applying various histological stains to stems from three key developmental. My results described in detail internal stem anatomy and demonstrated that lignification continues after crystalline cellulose deposition ceases. A better understanding of growth cues and various anatomical and cell wall construction features of B. distachyon will further our understanding of plant biomass accumulation processes.

of disertation Rhythmic gymnastics (RG) is the only Olympia sport of girls and women and it is characterized by precision, fluency and elegance of movement that emphasizes feminine beauty and grace. At present, there is no coherent concept of long-term sports training of talented gymnasts in Czech RG. The outflow of athletes is evident especially during prepubescence. Basic RG preparation aims gradual development of motor abilities, practice of sports (gymnastics) skills and the beginning of specialized techniques based on the principle of comprehensive movement base. This period (in rhythmic gymnastics between 7-9 years of gymnasts) for further performance growth is very substantial, in terms of content and in terms of training forms and influence of trainers (coaches). Defining the content of movement and gymnastic activities determines the path of development athletes and also defines (for caoches) performance criteria in each age category. The aim of disertation was to verify correspondence between published scientific theories and trainers experience of coaching using the Repertory Grid Technique. Patterns of trees’ endogenous rhythmic growth (ERG) and paralleled C allocation shift between root and shoot systems have been studied, but there is still a need to understand their impact in shaping soil microbiomes. Moreover, the impact of plants on soil microbial communities can be modulated or overweighed by time-induced plant and/or seasonal changes. Thus, we intended to analyze the structure of soil microbiomes as response to simultaneous alternated host tree root and shoot flushes and time-induced changes within one vegetation period at two sites in Central Germany. In this study, we utilized oak phytometers (Quercus robur L., clone DF159) as host trees, and made use of their ERG, whereby consecutive root and shoot flushes make a complete growth cycle. We studied two complete growth cycles during the same vegetation period, performed a non-destructive soil sampling and applied high-throughput amplicon sequencing of the bacterial 16S gene and the fungal ITS2 region. As C allocation shifts between the tree root and shoot, released root exudates and consequently the nutrient availability alternate for soil microorganisms. We therefore anticipated different microbial communities in the host tree root zone along the growth cycles until autumnal leaf senescence. In our results, the bacterial community exhibited a directional change over time along the vegetation period. In contrast, the fungal community appeared sample specific. Our findings enlarge the current understanding of the temporal microbial assembly in the host tree root zone.

Olivier Messiaen is regarded as one of the most significant composers of the 20th century. His compositions are performed regularly and his teachings have influenced many well-known composers like Boulez and Stockhausen. This study focussed on his use of rhythm in his piano compositions. I supplied a short biography of the composer along with a brief discussion of his compositional techniques. Thereafter his rhythmic techniques were examined through relevant music examples from his piano repertoire. Particular attention was given to works from the Vingt Regards sur L’Enfant-Jésus along with Cantéyodjayâ, Mode de valeurs et d’intensités and Neumes rythmiques from his experimental period (1949-1951). In Mode de valeurs et d’intensités he revolutionized the serial treatment of duration, pitch, intensity and attack. 本文旨從語用和語音方面,來探討中文「敘述性」言談(narrative discourse)對話中,說話者自然產生的節拍手勢。在語用方面,主要是觀察說話者使用節拍手勢表達前後景(Grounding)和新/舊訊息(Information state)的關係。研究結果發現,說話者較常使用節拍手勢來傳達前景(foreground)的新訊息,以提示聽話者注意。此外,說話者在敘述的過程中,常使用節拍手勢來預示前後景訊息的轉換。在語音方面,主要探討說話者使用節拍手勢時,其所對應的語句之音調高低及說話能量強度的關係。研究發現,語句的音高和強度無法預測這類手勢的產生;但是,當傳達前景的新訊息時,這類手勢的出現常伴隨著較強的語氣。此外,研究結果也顯示,說話者使用節拍手勢通常是有規律性的。 / This thesis investigates gestural beats in Chinese narrative discourse at both pragmatic and acoustic levels. The relationship among grounding, information state and beat gestures is examined at the pragmatic level, and at the acoustic level, the relationship among pitch, intensity, and gestural beats is analyzed. Moreover, whether there is a rhythmic pattern for beat gestures is further studied. The subjects were four undergraduates who were asked to view an animated color cartoon episode and then to immediately recount the events in it to a listener. The subjects were audio-video taped. They were not informed of the particular interest of the study. 291 beats were transcribed and analyzed. The audio data was analyzed on Praat and a Kay’s Model 4100. The continuous beats were further analyzed to investigate the rhythmic patterns and were annotated in Anvil, a video annotation tool. Gestural beats, narrative data, and the annotations were statistically analyzed and five phenomena for beat gestures were explored. First, gestural beats can appear in both foregrounded and backgrounded clauses. Second, the occurrence of beat gestures signals shifts between levels of the narrative structure. Third, speakers usually incline to utter new information in foregrounded clauses in Chinese narrative discourse with accompanying beats. Fourth, neither intensity nor pitch changes can predict and correlate with the occurrence of gestural beats in the relationship between beats and two acoustic factors. However, when grounding and the information state of the associated referents are considered, beats are produced for new information in foregrounded clauses with greater intensity. Finally, there is a rhythmic pattern in the production of continuous beats and the regularity of the patterns correlates with the clause boundary. A close relationship is revealed between speech and gestures in this thesis. The findings provide some linguistic details that may help explore the performance of gestural beats and speech in Chinese narrative discourse.

The distribution and physiological roles of nitric oxide in the locomotor circuitry of the mammalian spinal cord

The mammalian spinal cord contains the neuronal circuitry necessary to generate rhythmic locomotor activity in the absence of inputs from the higher brain centre or sensory system. This circuitry is regulated by local neuromodulatory inputs, which can adjust the strength and timing of locomotor output. The free radical gas nitric oxide has been shown to act as an important neuromodulator of spinal circuits, which control locomotion in other vertebrate models such as the tadpole and lamprey. Despite this, the involvement of the NO-mediated soluble guanylate cyclase/cyclic guanosine monophosphate secondary messenger-signalling pathway (NO/sGC/cGMP) in mammalian locomotion has largely been under-investigated. The NADPH diaphorase histochemical reaction was used to identify sources of NO in the lumbar spinal cord. The largest population NADPH diaphorase reactive neurons were located in the dorsal horn, followed by the laminae of the ventral horn, particularly around the central canal (lamina X) and lamina VII. NADPH diaphorase reactive neurons were found along a rostrocaudal gradient between lumbar segments L1 to L5. These results show that that discrete neuronal sources of NO are present in the developing mouse spinal cord, and that these cells increase in number during the developmental period postnatal day P1 – P12. NADPH diaphorase was subsequently used to identify NADPH diaphorase reactive neurons at P12 in the mouse model of ALS using the SODG93A transgenic mouse. Physiological recordings of ventral root output were made to assess the contribution of NO to the regulation induced rhythmic fictive locomotion in the in vitro isolated spinal cord preparation. Exogenous NO inhibits central pattern generator (CPG) output while facilitating and inhibiting motor neuron output at low and high concentrations respectively. Removal of endogenous NO increases CPG output while decreasing motor neuron output and these effects are mediated by cGMP. These data suggest that an endogenous tone of NO is involved in the regulation of fictive locomotion and that this involves the NO/sGC/cGMP pathway. Intracellular recordings from presumed motor neurons and a heterogeneous, unidentified sample of interneurons shows that NO modulates the intrinsic properties of spinal neurons. These data suggest that the net effect of NO appears to be a reduction in motor neuron excitability.

In Folge eines Schlaganfalls leiden 90% aller Patienten an einer Handparese, die sich in 30-40% als chronisch manifestiert. Derzeit wächst seitens der Neurologie und Technologie das Forschungsinteresse an der Effektivität robotergestützter Therapieansätze, welche für schwer betroffene Patienten als besonders vielversprechend eingestuft werden. Die hierfür verwendeten Therapieroboter setzen sich aus einem mechanischen Teil und einer softwaregestützten virtuellen Umgebung zusammen, welche neben dem graphischen Interface, audio-visuelles Feedback sowie Musik beinhaltet. Bisher wurden Effekte der klanglichen Anteile dieses Szenarios noch nicht hinsichtlich möglicher Einflüsse auf Motivation, Bewegungsdurchführung, motorisches Lernen und den gesamten Rehabilitationsprozess untersucht. Die vorliegende Arbeit untersucht die Rolle von Sound in robotergestütztem Handfunktionstraining. Die Hauptziele im Rahmen dessen sind es, 1) Potentiale von Sound/ Musik für den Kontext robotergestützten Handfunktionstrainings zu explorieren, 2) spezifizierte klangliche Umgebungen zu entwickeln, 3) zu untersuchen, ob Schlaganfallpatienten von diesen spezifizierten Soundanwendungen profitieren, 4) ein besseres Verständnis über Wirkmechanismen von Sound und Musik mit Potential für robotergestützte Therapie darzulegen, und 5) Folgetechnologien über eine effektive Applikation von Sound/ Musik in robotergestützter Therapie zu informieren. / 90% of all stroke survivors suffer from a hand paresis which remains chronic in 30-40% of all cases. Currently, there is an increasing research interest in neurology and technology on the effectiveness of robot-assisted therapies. Robotic training is considered as especially promising for patients suffering from severe limitations. Commonly, rehabilitation robots consist of a mechanical part and a virtual training environment with a graphical user interface, audio-visual feedback, sound, and music. So far, the effects of sound and music that are embedded within these scenarios have never been evaluated in particular while taking into account that it might influence motivation, motor execution, motor learning and the whole recovery process. This thesis investigates the role of sound in robot-assisted hand function training post-stroke. The main goals of this work are 1) to explore potentials of sound/ music for robotic hand function training post-stroke, 2) to develop specified sound-/ music-applications for this context, 3) to examine whether stroke patients benefit from these specified sound/ music-application, 4) to gain a better understanding of sound-/ music-induced mechanisms with therapeutic potentials for robotic therapy, and 5) to inform further arising treatment approaches about effective applications of sound or music in robotic post-stroke motor training.

Mineração de estruturas musicais e composição automática utilizando redes complexas / Musical structures mining and composition using complex networks

A teoria das redes complexas tem se tornado cada vez mais em uma poderosa teoria computacional capaz de representar, caracterizar e examinar sistemas com estrutura não trivial, revelando características intrínsecas locais e globais que facilitam a compreensão do comportamento e da dinâmica de tais sistemas. Nesta tese são exploradas as vantagens das redes complexas na resolução de problemas relacionados com tarefas do âmbito musical, especificamente, são estudadas três abordagens: reconhecimento de padrões, mineração e síntese de músicas. A primeira abordagem é desempenhada através do desenvolvimento de um método para a extração do padrão rítmico de uma peça musical de caráter popular. Nesse tipo de peças coexistem diferentes espécies de padrões rítmicos, os quais configuram uma hierarquia que é determinada por aspectos funcionais dentro da base rítmica. Os padrões rítmicos principais são caracterizados por sua maior incidência dentro do discurso musical, propriedade que é refletida na formação de comunidades dentro da rede. Técnicas de detecção de comunidades são aplicadas na extração dos padrões rítmicos, e uma medida para diferenciar os padrões principais dos secundários é proposta. Os resultados mostram que a qualidade da extração é sensível ao algoritmo de detecção, ao modo de representação do ritmo e ao tratamento dado às linhas de percussão na hora de gerar a rede. Uma fase de mineração foi desempenhada usando medidas topológicas sobre a rede obtida após a remoção dos padrões secundários. Técnicas de aprendizado supervisionado e não-supervisionado foram aplicadas para discriminar o gênero musical segundo os atributos calculados na fase de mineração. Os resultados revelam a eficiência da metodologia proposta, a qual foi constatada através de um teste de significância estatística. A última abordagem foi tratada mediante o desenvolvimento de modelos para a composição de melodias através de duas perspectivas, na primeira perspectiva é usada uma caminhada controlada por critérios sobre redes complexas predefinidas e na segunda redes neurais recorrentes e sistemas dinâmicos caóticos. Nesta última perspectiva, o modelo é treinado para compor uma melodia com um valor preestabelecido de alguma característica tonal subjetiva através de uma estratégia de controle proporcional que modifica a complexidade de uma melodia caótica, melodia que atua como entrada de inspiração da rede. / The theory of complex networks has become increasingly a powerful computational tool capable of representing, characterizing and examining systems with non-trivial structure, revealing both local and global intrinsic structures that facilitate the understanding of the behavior and dynamics of such systems. In this thesis, the virtues of complex networks in solving problems related to tasks within the musical scope are explored. Specifically, three approaches are studied: pattern recognition, data mining, and synthesis. The first perspective is addressed by developing a method for extracting the rhythmic pattern of a piece of popular music. In that type of musical pieces, there coexist different types of rhythm patterns which constitute a hierarchy determined by functional aspects within the basic rhythm. The main rhythmic patterns are characterized by a higher incidence within the musical discourse and this factor is reflected in the formation of communities within the network constructed from the music piece. Community detection techniques are applied in the extraction of rhythmic patterns, and a measure to distinguish the main patterns of the secondary is proposed. The results showed that the quality of extraction is sensitive to the detection algorithm, the method of representing rhythm, and treatment of percussion lines when generating the network. Data mining is performed using topological measures over the network obtained after the removal of secondary patterns. Techniques of supervised and unsupervised learning are applied to discriminate the musical genre according to the attributes calculated in the data mining phase. The quantitative results show the efficiency of the proposed methodology, which is confirmed by a test of statistical significance. Regarding the melody generation, an algorithm using a walk controlled by criteria on predefined complex networks has been developed, as well as the development of melody composition models using recurrent neural networks and chaotic dynamical systems. In the last approach, the model is trained to compose a melody with a subjective characteristic melodic value pre-established by a proportional control strategy that acts on the parameters of a chaotic melody as input inspiration.
