Design och Kalibrering av mätrigg för Poly-V remmar / Design and calibration of a measuring rig for poly-v belts

Scania är en av världens största tillverkare av lastbilar, bussar och marina motorer och examensarbetet har utförts i syfte att utveckla en ny remmätrigg för att bättre kunna verifiera slitage, livslängd och töjning efter olika testcykler på Poly-V remmar. Riggen skall utföra mätningar enligt standard SAE-J1459 och klara att mäta remlängder mellan 1000-3300 mm. Därtill måste konstruktionen vara utformad enligt gällande säkerhetsföreskrifter och ergonomiska aspekter beaktas. Mätningar av Poly-V remmar sker efter principen att remmen spänns in mellan två remskivor med en kraft på 100 N per ribba. Vid examensarbetets start fanns en äldre mätrigg att tillgå, vilken inte längre är i bruk p.g.a. att den inte uppfyller alla krav för mätprocessen. Den har dock använts i syfte för att identifiera ergonomi- och säkerhetsrisker. Påvisade risker har sedan beaktats vid utformningen av koncept, för att begränsa dessa vid konstruktion av den nya mätriggen. Konstruktionen av den nya remmätriggen är framtagen med hjälp av erkänd produktutvecklingsmetodik, med arbete från idé till färdig produkt. Allt i enlighet med en förstudie i konstruktionsmetodik, en konceptstudie som behandlar ett flertal koncept, konceptval och analys av teknisk utrustning som används i den nya konstruktionen samt konstruktionsarbete och utvärdering. Vid examensarbetets slut har en färdigmonterad remmätrigg levererats klar för användning. Konstruktionen klarar att mäta remlängder mellan 1000-3300 mm och upp till tolv ribbor. Mätprocessen enligt SAE-J1459 kan utföras samtidigt som riggen är utformat med ergonomi och säkerhet i åtanke. Tillhörande remmätriggen finns en användarmanual som beskriver rutiner kring användning av utrustningen, risker och själva mätprocessen steg för steg. I rapporten bilagas en förstudie inom ergonomi och säkerhet, som har utförts vid examensarbetets första fas med syfte att användas som grund vid utformningen av den nya riggen. Det för garantera att hänsyn är tagen till gällande ergonomi- & arbetsmiljö-föreskrifter. / Scania is one of the World greatest producers of trucks, busses and marine engines. The purpose of the master thesis has been to develop a new measuring rig for Poly-V belts, to better be able to verify wear, duration and elongation at different testing cycles. The rig shall do measurements following the SAE-J1459 standard and be able to perform on belts with length between 1000-3300 mm. In addition, the design must be made in accordance with safety regulations and ergonomic aspects. Measurement of Poly-V belts are carried out by straining the belt between to pulleys with a force of 100 N per crossbar. At the project start access was granted to an old measuring rig, which is no longer in use due to not fulfilling the requirements for the measuring procedure. However, it has been used to identify ergonomic and safety gaps. The development of new concepts has then been carried out with those gaps in mind. The design of the new measuring rig is developed by using proven product development methodology, which covers work from idée to completed product. That includes accordance with a pre-study in design methodology, a concept study which deals with a number of concepts, the selection of concepts and an analysis of the technical equipment used in the new design during the product design. The result of the thesis is a completed rig, delivered and ready to use. The rig is capable of measuring ribbed belt length between 1000-3300 mm and up to twelve ribs. Measurement process according to SAE-J1459 can be performed meanwhile the rig is designed with ergonomics and safety aspects. A user manual describing the use of equipment, risks and the actual measurement process step by step is also included with the rig. The report annexes a pre-study of ergonomic and safety, which has been written in the initial phase of the thesis. The pre-study has been used as a guide for the design of the new rig, taking into consideration the existing regulations from different acquis.

Design and Implementation of a Test Rig for a Gyro Stabilized Camera System

PolyTech AB in Malmköping manufactures gyro stabilized camera systems or helicopter applications. In this Master´s Thesis a shaker test rig for vibration testing of these systems is designed, implemented and evaluated. The shaker is required to have an adjustable frequency and displacement and different shakers that meet these requirements are treated in a literature study. The shaker chosen in the test rig is based on a mechanical solution that is described in detail. Additionally all components used in the test rig are described and modelled. The test rig is identified and evaluated from different experiments carried out at PolyTech, where the major part of the identification is based on data collected from accelerometers. The test rig model is used to develop a controller that controls the frequency and the displacement of the shaker. A three-phase motor is used to control the frequency of the shaker and a linear actuator with a servo is used to control the displacement. The servo controller is designed using observer and state feedback techniques. Additionally, the mount in which the camera system is hanging is modelled and identified, where the identification method is based on nonlinear least squares (NLS) curve fitting technique.

This result can be accomplished with Web 2.0 technologies by creating a repository of skeletons and animations that any animator may use for free. While free Maya™ skeletons currently exist on the Internet, the websites housing them have only brief features and functions for browsing and interacting with these files. None of these websites contain downloadable animations for the provided skeletons. The Chimaera Project improves the field of Web 2.0 sites offering free rigs by offering many new features and freedoms to the animation community. Users may upload and download Maya™ skeletons, share comments and tips with each other, upload animations associated with the skeletons, and search or browse the skeletons in a variety of ways. The skeletons include descriptions and information provided by the creator and are categorized by class, order, and species. Users may access a freely provided script called “zooXferAnim” to import and export animations into text files to be uploaded and downloaded on the website. Many animations per skeleton may be uploaded. The Chimaera Project extends the Web 2.0 community by creating an interactive resource for animators to contribute and share content in a better, more organized format than previously seen on the Internet.

High temperature, permanent magnet biased, homopolar magnetic bearing actuator

The EEC (Electron Energy Corporation) in conjunction with the National Aeronautics and Space Administration is researching the magnetic bearings for an alternative to conventional journal or ball bearings. The purpose of this research was to design and develop a high-temperature (1000ºF) hybrid Magnetic Bearing using High Temperature Permanent Magnets (HTPM), developed by the EEC for high performance jet engines at high speeds that supply loads of 500 lbf. Another objective is to design and build a test rig fixture to measure the load capacity of the designed bearing. The permanent magnet bias of the Homopolar radial magnetic bearing reduces the amount of current required for magnetic bearing operation. This reduces the power loss due to the coil current resistance and improves the system efficiency because the magnetic field of the HTPM can suspend the major portion of the static load on bearing. A high temperature radial magnetic bearing was designed via an iterative search employing 3D finite element based electromagnetic field simulations. The bearing was designed to produce 500 lbf of force at 1000ºF and the design weight is 48 lbs. The bias flux of the Homopolar radial bearing is produced by EEC HTPM to reduce the related ohmic losses of an electromagnetic circuit significantly. An experimental procedure was developed to measure actual load capacity of the designed bearing at the test rig. All the results obtained from the experiment were compiled and analyzed to determine the relation between bearing force, applied current and temperature.

Étude de la régulation du facteur de transcription NF-kB dans l'infection par le RSV

L’infection par le Virus Respiratoire Syncytial cause des affections pulmonaires aiguës en pédiatrie caractérisée par une réponse inflammatoire excessive médiée par la production de cytokines par les cellules épithéliales des voies aériennes. Les gènes codant pour ces cytokines sont régulés par le facteur de transcription NF-κB (p50/p65) dont l’activation est classiquement induite par la phosphorylation de son inhibiteur IκBα, ce qui permet l’accumulation de l’hétérodimère au noyau. Par contre, nous avons récemment identifié la phosphorylation en sérine 536 de la sous-unité p65 comme une autre étape essentielle à son activation lors de l’infection des AEC par RSV. Le travail présenté dans ce mémoire a permis de démontrer que l’inhibition de l’expression de RIG-I, de Cardif ou de TRAF6, 3 protéines impliquées dans la reconnaissance cellulaire des virus, conduit à l’inhibition de cette phosphorylation en réponse à RSV. Nous avons également établi à l’aide d’inhibiteurs pharmacologiques et d’ARNi que, parmi les diverses kinases connues pour phosphoryler p65 en réponse à divers stimulus, IKKα/β sont essentielles à cette phosphorylation lors d’une stimulation par RSV. Puisque TRAF6 est bien connu dans la littérature pour activer le complexe IKK, nous proposons que TRAF6, après reconnaissance de l’ARN viral de RSV par RIG-I, active le complexe IKK qui induit la phosphorylation de la sousunité p65 de NF-κB, permettant l’expression de gènes cibles. D’autre part, nous avions précédemment démontré que Nox2, un isoforme de NADPH oxydase, contrôle l’activation de NF-κB en régulant les phosphorylations de IκBα et p65. Nous montrons ici que l’inhibition de Nox2 réduit fortement l’activité du complexe kinase IKK. De plus, la présence au niveau basal de Nox2 est critique pour le niveau d’ARN messager de Cardif. Nous proposons donc que la régulation de la phosphorylation de p65 en ser536 par Nox2 soit via son effet sur Cardif en permettant la fonctionnalité de la voie RIG-I. / The infection by the Respiratory Syncytial Virus causes acute respiratory tract affections among children characterized by an excessive inflammatory response mediated by airway epithelial cells production of cytokines. The genes coding for theses cytokines are regulated by the transcription factor NF-κB (p50/p65) which is classically activated by phosphorylation of its inhibitor IκBα, permitting the nuclear accumulation of the heterodimer. The work presented in this master’s thesis allowed demonstrating that the inhibition of either RIG-I, Cardif or TRAF6, 3 proteins implicated in the cellular recognition of virus, leads to the inhibition of this phosphorylation in response to RSV. Moreover, we established with pharmacological inhibitors and siRNA that, among all kinases known to phosphorylate p65 in response to various stimulus, IKKα/β are essential to this phosphorylation in RSV stimulation. Since TRAF6 is a well-known IKK complex activator in the literature, we propose that TRAF6, after the recognition of the RSV viral RNA by RIG-I, activates the IKK complex witch induces the phosphorylation of the NF-κB subunit p65, allowing the expression of targets genes. Furthermore, we had previously demonstrated that Nox2, a NADPH oxydase isoform, controls the activation of NF-κB by regulating the phosphorylation of IκBα and p65. We show here that the inhibition of Nox2 by siRNA reduces strongly the activity of the IKK complex. Moreover, the basal level presence of Nox2 is critic for the messenger RNA level of Cardif. Thus, we propose that the Nox2 regulation of p65 ser536 is done by its effect on Cardif, which allows the integrity of the RIG-I pathway.

Development of a rig and testing procedures for the experimental investigation of horizontal axis kinetic turbines

The research detailed in this thesis was focused on developing an experimental testing system to characterize the non-dimensional performance coefficients of horizontal axis kinetic turbines, including both wind turbines and tidal turbines. The testing rig was designed for use in a water tunnel with Particle Image Velocimetry (PIV) wake survey equipment to quantify the wake structures. Precision rotor torque measurement and speed control was included, along with the ability to yaw the rotor. The scale of the rotors were purposefully small, to enable rapid-prototyping techniques to be used to produce many different test rotors at low cost to furnish a large experimental dataset. The first part of this work introduces the mechanical design of the testing rig developed for measuring the output power of the scaled rotor models with consideration for the requirements imposed by the PIV wake measurements. The task was to design a rig to fit into an existing water tunnel facility with a cross sectional area of 45 by 45 cm, with a rotor support structure to minimize the flow disturbance while allowing for yawed inflow conditions. A rig with a nominal rotor diameter of 15 cm was designed and built. The size of the rotor was determined by studying the fluid similarities between wind and tidal turbines, and choosing the tip speed ratio as a scaling parameter. In order to maximize the local blade Reynolds number, and to obtain different tip speed ratios, the rig allows a rotational speed in the range of 500 to 1500 RPM with accurate rotor angular position measurements. Rotor torque measurements enable rotor mechanical power to be calculated from simulation results. Additionally, it is included in this section a description of the instrumentation for measurement and the data acquisition system. It was known from the outset that measurements obtained in the experiments would be subject to error due to blockage effects inherent to bounded testing facilities. Thus, the second part of this work was dedicated to developing a novel Computational Fluid Dynamics (CFD) methodology to post-process the experimental data acquired. This approach utilizes the velocity field data at the rotor plane obtained from the water tunnel PIV test data, and CFD simulations based on the actuator disk concept to account for blockage without the requirement for thrust data which would have been unreliable at the low forces encountered in the tests. Finally, the third part of this work describes the practical aspects of the laboratory project, including a description of the operational conditions for turbine testing. A set of preliminary measurements and results are presented, followed by conclusions and recommendations for future work. Unfortunately, the water tunnel PIV system was broken and thus unavailable for more than a year, so only mechanical measurements were possible with the rig during the course of this thesis work. / Graduate

Analysis of heat dissipation from railway and automotive friction brakes

The thesis presents research into the understanding and improvement of heat dissipation from friction brakes. The investigations involved two brake types, considered to be the most thermally loaded and therefore most challenging; axle mounted high speed railway and commercial vehicle disc brakes. All three modes of heat transfer (conduction, convection and radiation) and airflow characteristics have been analysed experimentally and theoretically in order to increase the understanding of heat dissipation. Despite the very practical aspects of this research, a 'generic heat transfer approach' was applied, enabling wider engineering applications of the results. Experimental analyses conducted on a specially developed Spin Rig allowed measurements of cooling and airflow characteristics for different designs. Methodologies have been developed to determine thermal contact resistance, heat transfer coefficients, emissivity and aerodynamic (pumping) losses. Established values and relationships compared very favourably with theoretical work. Analytical, FE and CFD analyses were employed to further investigate design variations and perform sensitivity studies. Inertia dynamometer route simulations provided disc temperatures for validation of the overall work. Recommendations have been made for optimising heat dissipation, by proposing practically acceptable and economically viable design solutions. A proposed ventilated disc design efficiency ratio allows large, high speed ventilated disc designs, to be efficiently and accurately evaluated and compared, providing a valuable disc design optimisation tool. The determination of the methodologies, parameters and functions defining cooling characteristics, enable heat dissipation to be predicted confidently and accurately for brakes and other engineering assemblies at early design stages.

L’étude de l’impact des protéines non structurales NS1 et NS2 du virus respiratoire syncitial sur la réponse immunitaire innée

Le virus respiratoire syncytial (RSV) est un virus à ARN de polarité négative. Les études démontrent que toute la population sera infectée par ce virus au moins deux fois avant l’âge de 3 ans. Le RSV peut provoquer plusieurs pathologies respiratoires telles que la bronchiolite aiguë et la pneumonie. Les infections sévères corrèlent avec le développement de l’asthme. Lors d’une infection virale, les particules du RSV sont détectées par le senseur RIG-I qui induit l’activation des facteurs de transcription NF-κB et IRF-3. Respectivement, les facteurs de transcription activeront les réponses inflammatoire et antivirale. Au coeur des pathologies induites par le RSV se trouve une réponse immunitaire mal adaptée. Plus précisément, par l’entremise de NF-κB, le RSV provoque une production exagérée de cytokines et chimiokines qui induisent une réponse inflammatoire démesurée provoquant du dommage tissulaire. Paradoxalement, le RSV est capable d’échapper à la réponse antivirale. Ces deux phénomènes sont contrôlés par l’entremise des protéines non structurales NS1 et NS2. Le mécanisme délimitant le mode d’action de NS1 et NS2 sur la réponse antivirale reste à être déterminé. Avec pour objectif d’élucider comment NS1 et NS2 inhibent la réponse antivirale, nous avons investigué le mécanisme de reconnaissance de l’hôte vis-à-vis de RSV. Nous démontrerons, pour la première fois, que le senseur cytosolique MDA5 est impliqué dans la réponse antivirale contre le RSV. Nous présenterons des résultats préliminaires qui suggèrent que le rôle de MDA5 est non redondant à RIG-I. À l’aide d’ARN interférant dirigé contre RIG-I et de transfection de MDA5, nous démontrerons que MDA5 ne contribue pas à la phosphorylation d’IRF-3, mais plutôt qu’elle régit la stabilité du facteur de transcription. Nous démontrerons aussi que, contrairement à l’hypothèse actuelle sur le fonctionnement de NS1 et NS2, l’inhibition de ces derniers ne provoque pas une augmentation de la cytokine antivirale IFN−β. Cependant, l’expression ectopique de NS1 et NS2 réduit l’activité du promoteur de l’IFN-β et de la protéine cytoplasmic antivirale ISG56 lorsqu’elle est mesurée par essai luciférase. / Respiratory Syncytial Virus (RSV) is a RNA virus with negative polarity. RSV infections are the most common cause of hospitalization among infants. Among populations at risk, infection of RSV can be quite severe. RSV infections can cause bronchiolitis, pneumonia, while severe infections are linked to the development of asthma. Early in the infectious cycle of RSV, the cytosolic sensor RIG-I captures viral particles, and activates the immune response by engaging the transcription factors IRF-3 and NF-κB. At the heart of RSV mediated pathologies is a skewed immune response. More precisely, RSV over stimulates the release of proinflammatory chemokines and cytokines. Intriguingly, while RSV is able to stimulate the production of proinflammatory cytokines and chemokines, RSV under stimulates the antiviral response. The ability of RSV to evade the antiviral response is thought to be mediated by its non-structural proteins: NS1 and NS2. However, the mechanism by which NS1 and NS2 enable RSV to evade the antiviral response remains to be determined. In this memoir we investigated, how RSV is recognized by the innate immune response in airway epithelial cells. With this information we hope to improve our understanding of how NS1 and NS2 allow RSV to circumvent the antiviral response. We show for the first time that cytosolic sensor MDA5 plays a role in the recognition of RSV particles. Using a combination of interfering RNA directed against RIG-I, and transfection of MDA5, we show that MDA5 does not contribute to the phosphorylation of IRF-3. According to the data presented, we suggest that MDA5’s role in the immune response is to prevent the degradation of IRF-3. Contrary to previous research, we show that the inhibition of the nonstructural protein does not increase the production of the antiviral cytokine IFN-β. However, the ectopic expression of NS1 and NS2 does lead to a reduction of the promoter activity of IFN-β and the antiviral protein ISG56 when measured by luciferase assay. This research highlights the importance of MDA5 as a potential therapeutic target in the development of a cure for RSV.

Fuel injector spray diagnostic development

New technologies are constantly developing towards the goal of increasing the performance of gas turbine engines while reducing pollutant emissions. The design of the combustion system is vital in the drive to reduce pollutants in order to meet legislative targets. As part of this, the fuel injector is crucial in preparing the fuel for combustion through atomization and correct mixing with the air flow. Thus, it is desirable to develop techniques to allow the analysis of performance in these key criteria and improve the understanding of both fuel injector aerodynamics and fuel atomisation. Particle Image Velocimetry (PIV) allows for spatially resolved velocity data of flow fields to be recorded and therefore enables the inspection of flow behaviour.
