Saponinas dos frutos de ilex paraguariensis A. St. Hil. (mate) : desenvolvimento de metodologia analítica, estudos físico-químico e biológico

O presente trabalho teve por objetivo a avaliação físico-química e biológica de uma fração de saponinas enriquecida, obtida a partir de frutos verdes de Ilex paraguariensis A. St. Hil (mate). A espécie apresenta grande importância econômica para vários países sul-americanos, dentre eles o Brasil. Os frutos verdes da espécie acumulam significativa quantidade de saponinas, entretanto, são considerados subproduto da indústria do beneficiamento das folhas de mate. O extrato liofilizado dos frutos (EX40) foi obtido por turboextração na proporção planta-solvente de 1:10 utilizando como solvente solução hidroalcoólica a 40%. Para análise do extrato bruto foi desenvolvido e validado um método analítico por CLAE-UV, que permitiu: 1) quantificar o marcador químico ilexosídeo II em EX40; 2) estimar o conteúdo em saponinas totais do extrato, tendo sido obtidos valores de 8,2 g% e 47,6 g% respectivamente. Uma fração enriquecida em saponinas, denominada MSF, foi obtida mediante fracionamento em fase sólida a partir de EX40, utilizando resina hidrofóbica poliaromática e gradiente metanol:água de polaridade decrescente. Os maiores teores de saponinas foram obtidos para as frações contendo 70 e 90 % de metanol. A reprodutibilidade do processo foi avaliada por CLAE-UV, considerando o desvio padrão relativo (DPR) entre a área dos dois picos majoritários de MSF produzidas em seis lotes distintos. Valores de DPR de 8,17% para o pico I (ilexosídeo II) e 5,96 % para o pico II (majoritário) foram obtidos. A toxicidade da fração foi avaliada pelo potencial hemolítico in vitro e citotoxicidade sobre cultura de células de mamífero (MDBK ATCC CCL-22). Para atividade hemolítica foi determinado um valor de IC50 de 0,2 g/L (0,22 mM, expresso em ilexosídeo II). O valor de IC50 para citotoxicidade foi 3,8 g/L (4,04 mM, expresso em ilexosídeo II), caracterizando um produto de baixa toxicidade. Dentre as várias atividades biológicas descritas para saponinas, selecionouse avaliar MSF quanto ao seu potencial como imunoadjuvante, assim como a atividade tricomonicida. Para o primeiro caso, MSF foi testada em uma vacina contendo herpesvírus bovino tipo 5 como antígeno viral, nas doses de 100 e 500 μg. Entretanto, o incremento no título de IgG totais, avaliado por ELISA, não foi significativo. A atividade tricomonicida foi comparada aos tensoativos polissorbato 80 e tiloxapol e a uma fração enriquecida em saponinas de Quillaja saponaria. Após 24 h de incubação foi observado para MSF um efeito dose-dependente, atingindo letalidade total dos trofozoítos na concentração de 0,6 g/L. Esse efeito foi superior ao observado para polissorbato 80 e tiloxapol e similar ao de quillaia. O tratamento com metronidazol, administrado tanto após tratamento prévio dos trofozoítos com MSF quanto em associação com essa fração, não revelou qualquer alteração significativa no efeito de ambos os produtos, o que sugere ausência de interação entre MSF e MTZ e a possível existência de mecanismos de ação distintos. A caracterização físico-química enfatizou o estudo do tamanho e forma das micelas, seu comportamento reológico e capacidade de solubilizar compostos hidrofóbicos. A concentração micelar crítica foi determinada em 0,41 g/L utilizando um corante hidrofóbico como marcador. A análise por microscopia eletrônica de transmissão (MET) e CRIO-MET revelou a presença simultânea de micelas alongadas, com tamanho superior a 500 nm, e micelas esféricas de tamanho menor. Para a fração de micelas esféricas, a análise por espalhamento de nêutrons a baixo ângulo permitiu estabelecer diâmetro de 17,9 Å, perfazendo 23,1 e 32,6 % das micelas observadas em soluções de MSF a 0,5 e 1,0 % m/v, respectivamente. O comportamento reológico de MSF nas concentrações de 0,25 a 4,0 %, determinado por reometria dinâmica com controle de deformação, foi do tipo viscoelástico para todas as concentrações. Esse fato corrobora a existência de agregados não-esféricos, filiformes, semelhantes a micelas tipo wormlike. A solubilização micelar de fármacos mediante MSF foi avaliada utilizando como substâncias-modelos os fármacos flurbiprofeno – FLB (ácido fraco) e carbamazepina - CBZ (base fraca), ambos pouco solúveis, e saponinas de quillaia como produto de comparação. Nas concentrações de 0,13 a 1,5 % as saponinas de quillaia foram mais eficientes frente ao flurbiprofeno, enquanto MSF foi capaz de aumentar significativamente a solubilidade da carbamazepina. O incremento na solubilização gerado por MSF foi de 0,0145 e 0,0129 g/L para CBZ e FLB, respectivamente. / The present work aimed the physicochemical and biological evaluation of a saponin enriched fraction from the green fruits of Ilex paraguariensis A. St. Hil. (mate), a species of great economical importance in several Southern American countries, including Brazil. Despite possessing high amounts of saponins, mate green fruits have no commercial use to date and are considered a waste product from the mate leaves processing companies. Mate fruits lyophilized extract (EX40) was produced using turbo-extraction having a 40:60 mixture of water and ethanol as solvent and a 1:10 drug/solvent proportion. An HPLC-UV method was developed and validated aiming at: I) Quantify the EX40 chemical marker, ilexoside II; II) Determine the EX40 total saponin content. The values obtained were of 8.20 wt% and 47.60 wt%, respectively. An enriched saponin fraction (MSF) was obtained from EX40 through a solid phase extraction process within a polyaromatic resin and a decreasing solvent polarity gradient of methanol and water. The higher recovery of mate saponins was achieved with the 70 and 90 % methanol-containing fractions. The process reproducibility was assessed by HPLC-UV through the analysis of the relative standard deviation (RSD) of the two major MSF saponins peak areas obtained in six different batches. RSD values of 8.17 % for peak I (ilexoside II) and 5.96 % for peak II (major peak) were obtained. The MSF toxicity was evaluated according to its haemolytical activity and in vitro cytotoxicity against a mammalian cell culture lineage (MDBK ATCC CCL-22). Values of IC50 of 0.2 g/L (0.22 mM expressed as ilexoside II) and 3.8 g/L (4.04 mM as Ilexoside II) were found for the haemolysis and cytotoxicity respectively. Therefore we could classify MSF as a low toxicity product. Among the several biological activities ascribed to saponins, this work focused on the investigation of MSF immune enhancer potential and its anti-trichomonads vaginalis activity. For the first evaluation, MSF wad added to a bovine herpesvirus 5 vaccine, at 100 and 500 μg doses. However total IGg titres determined by ELISA were not statistically significant. The antitrichomonads activity of MSF was compared a saponin enriched fraction from the species Quillaja saponaria (Quillaja) and to tyloxapol and polysorbate 80. After 24h of incubation MSF presented a dose-dependent activity with total trophozoites lethality at a concentration of 0.6 g/L. The activity was higher than the synthetic surfactants and similar to Quillaja. The treatment with metronidazole (MTZ), peformed in association and after exposing the trophozoites with MSF did not show any synergistic effect of MZT and MSF which suggest an absence of interaction between the compounds and also that they probably have distinct mechanisms of action. The physicochemical evaluation of MSF focused the determination of MSF micelles size and shape, their rheological behaviour and ability to increase the solubility of hydrophobic compounds. MSF critical micellar concentration was determined as 0.41 g/L using a hydrophobic dye as a probe. The transmission electron microscopy (TEM) and CRYO-TEM analysis revealed the occurrence of filiform micelles higher than 500 nm and smaller sizes spherical micelles simultaneously. The spherical micelles radius of 17.9 Å were determined using small angle neutron scattering.and they corresponded to 23.1 and 32.6 % of the total micelles in MSF solutions of 0.5 and 1.0 % w/v, respectively. The rheological behavior of MSF ranging in concentration from 0.25 to 4.0% was determined on a dynamic strain control rheometer and a viscoelastic behavior was observed for all concentrations. These findings corroborate the occurrence of nonspherical micelles very long in size (wormlike). MSF ability to improve the solubilisation of poor aqueous soluble drugs was assessed using carbamazepine (CBZ) and flurbiprofen (FLB) as a model of amphiphilic basic and acidic coumpounds. Saponin enriched fractions were evaluated in concentrations ranging from 0.13 to 1.5 %. Quillaja was more efficient toward the weak acid while MSF was able to increase significantly the CBZ solubility. The increase on CBZ and FLB solubility after MSF addition was of 0.0145 and 0.0129 g/L, respectively.

FundaÃÃo Cearense de Apoio ao Desenvolvimento Cientifico e TecnolÃgico / O presente trabalho relata o estudo quÃmico do caule de Cestrum laevigatum Schlecht (Solanaceae), coletado no municÃpio de Pacoti-CE. A partir da investigaÃÃo do extrato etanÃlico utilizando tÃcnicas clÃssicas de cromatografia de adsorÃÃo (gel de sÃlica e sÃlica flash) e exclusÃo (Sephadex LH-20) foi possÃvel o isolamento de esterÃides e um triterpeno. Os esterÃides de esqueleto espirostano foram caracterizados como uma mistura de sapogeninas 25(S)-5-espirostano-2,3-diol e 25(R)-5-espirostano-2,3-diol (gitogenina) (CL-1), a mistura de epÃmeros (25R)-5-espirostano-2,3-diol-3-O--D-galactopiranosÃdeo e (25S)-5-espirostano-2,3-diol-3-O--D-galactopiranosÃdeo (CL-2), e a mistura epimÃrica da saponina de carÃter inÃdito na literatura (25R,S)-5-espirostan-2,3-diol-3-O--D-galactopiranosil(1→4)- -D-galactopiranosil(1→4)--D-glicopiranosil (CL-3). AlÃm destes compostos, foram isolados ainda a mistura dos esterÃides -sitosterol e estigmasterol (CL-5) e seus derivados glicosilados (CL-6), alÃm do triterpeno pentacÃclico de esqueleto do tipo lupano β-lup-20(29)-en-3-ol (lupeol) (CL-4). A caracterizaÃÃo estrutural dos compostos isolados foi possÃvel atravÃs do uso de tÃcnicas espectroscÃpicas, principalmente RMN, incluindo seqÃÃncias de pulso uni e bidimensionais, e comparaÃÃo com dados relatados na literatura. Das saponinas investigadas quanto ao potencial antimicrobiano, apenas CL-3 mostrou-se ativa contra Bacillos subtilis, Pseudomonas aeruginosa e Enterobacter aerogenes. A atividade leishmanicida foi realizada com CL-1, CL-2 e CL-3, porÃm, apenas CL-3 mostrou-se ativa contra Leishmania amazonensis. Em relaÃÃo à atividade antifÃngica, CL-1 mostrou-se ativa apenas contra Microsporum canis. CL-2 apresentou excelentes resultados contra Candida albicans, Trichophyton rubrum e Trichophyton mentagrophytes, e uma atividade relativamente boa contra Microsporum canis. Jà CL-3 aparece revelou uma excelente atividade antifÃngica contra todas as cepas testadas. Os resultados promissores obtidos atà o momento a partir do extrato etanÃlico do caule de C. laevigatum confirmam o grande potencial quÃmico/farmacolÃgico das espÃcies deste gÃnero, e justificam a importÃncia do trabalho realizado / This work reports the chemical study of the stem of Cestrum laevigatum Schlecht (Solanaceae), collected at the Pacoti-CE. The investigation of the ethanol extract using conventional techniques of adsorption chromatography (silica gel and silica flash) and exclusion (Sephadex LH-20) was possible the isolation of steroids and triterpene. The spirostan steroids have been characterized as the mixture at the sapogenins 25(S)-5-espirostan-2,3-diol and 25(R)-5-espirostan-2,3-diol (gitogenin) (CL-1), a mixture of epimers (25R)-5-espirostan-2,3-diol-3-O--Dgalactopyranoside and (25S)-5-espirostan-2,3-diol-3-O--D-galactoyiranoside (CL-2), and the mixture of new epimer (25R,S)-5-espirostan-2,3-diol-3-O--Dgalactopyranosil(1→4)--D-galactopyranosil(1→4)--D-glicopyranosil (CL-3). Besides these compounds, were also isolated the mixture of steroids β-sitosterol and stigmasterol (CL-5) and its glycosides (CL-6), and the pentacyclic triterpene β-lup-20(29)-en-3-ol (lupeol) (CL-4). The structural characterization of the isolated compounds was possible through the use of spectroscopic techniques, mainly NMR, including sequences of uni and bidimensional pulse, and comparison with literature data. CL-3 was shown to be active against Bacille subtilis, Pseudomonas aeruginosa and Enterobacter aerogenes. The leishmanicide activity was performed with CL-1, CL-2 and CL-3, however, only CL-3 was shown to be active against Leishmania amazonensis. For the antifungical activity, CL-1 was shown to be active only against Microsporum canis. CL-2 showed excellent results against Candida albicans, Trichophyton rubrum and Trichophyton mentagrophytes, and a relatively good activity against Microsporum canis. CL-3 appears with excellent activity antifungical against all strains tested. The promising results obtained so far from the ethanol extract of the stem of C. laevigatum confirm the great potential chemical / pharmacological species of this genus, and justify the importance of work

A concern about the useful of these natural products is the quality control pharmacobotanical, phytochemical and purity tests, which make up the technical specifications of a particular plant species. Moreover, pharmacological and toxicological studies must be conducted in order to effectiveness and safety. This study aimed to carry out the assessment pharmacognostic, acute toxicity and evaluation of antimicrobial and leishmanicidal of the ethanol extract of the leaves, fruit, stem bark and root bark of R. elaeocarpum Reissek. Samples of R. elaeocarpum were collected in the municipality of Campestre-GO, a description of the macroscopic aspects of the leaves, stem bark and root bark was carried to the naked eye by observing the macroscopic general of vegetal drugs; the microscopical analysis was performed by observing the histological stained. The qualitative analysis of the major classes of secondary metabolites present in the sprayed sample. The tests for water content, total ashes and acid insoluble ashes had been carried through according to the brazilian pharmacopoeia IV. The rude ethanol extract was obtained by maceration and the fractions were obtained by partition, liquid-liquid, successive with hexane, dichloromethane and ethyl acetate. Minimum inhibitory concentration (MIC) was determined by agar dilution. The procedure was performed according to NCCLS standards 2004 (M7-A7). The evaluation of acute toxicity of ethanol extracts had reference to the Guidelines of 423 Organization for Economic Co-Operation and Development (OECD). The leishmanicidal activity was tested for the promastigotes forms of Leishmania (L) chagasi. Most of the morphological and anatomical characteristics observed are consistent with the typical family. In phytochemical screening of leaf, root bark, stem bark and fruit of R. elaeocarpum verified the presence of flavonoids and alkaloids. It was also the presence of heterosides saponins and tannins in leaf, root bark, stem bark and is absent in the fruit. The extracts and fractions that had obtained the lowest value of minimum inhibitory concentration (0,390 mg.mL-1) against microorganisms tested showed moderate inhibitory activity. None of the extracts and fractions showed good inhibitory activity. As is said about the toxicological research, throughout the period of observation of animals during the evaluation of acute toxicity was not death. Through behavioral observations made through the hippocratic screening is assigned a zero score for all evaluated items. The necropsy of the animals did not evidence any macroscopic alterations to justify the histopathology of the organs (kidney, spleen and liver) selected for analysis. This study demonstrated that the use of rude extract and fractions of leaves of R. elaeocarpum induced death of promastigotes forms of L. (L) chagasi in vitro. The presented results open perspectives for the future isolation, purification and identification of bioactive substances of R. elaeocarpum / Nos últimos anos o tratamento com medicamentos à base de plantas medicinais tem aumentado. Uma preocupação em relação à utilização desses produtos naturais consiste no controle de qualidade farmacobotânico, fitoquímico e ensaios de pureza, que compõem as especificações técnicas de uma determinada espécie vegetal. Além disso, estudos farmacológicos e toxicológicos devem ser realizados, visando a sua eficácia e segurança. Este trabalho teve por objetivo realizar a avaliação farmacognóstica, toxicidade aguda e avaliação das atividades antimicrobiana e leishmanicida do extrato etanólico das folhas, frutos, cascas do caule e cascas da raiz de R. elaeocarpum Reissek. Amostras da R. elaeocarpum foram coletadas no município de Campestre-GO, a descrição dos aspectos macroscópicos das folhas, casca do caule e casca da raiz foi realizada à vista desarmada observando as características macroscópicas gerais de drogas vegetais; a análise microscópica foi realizada através da observação de cortes histológicos submetidos à coloração. A análise qualitativa das principais classes de metabólitos secundários presentes foi realizada na amostra pulverizada. Os ensaios para determinação do teor de água, cinzas totais e cinzas insolúveis em ácido foram realizados segundo a Farmacopéia Brasileira IV. O extrato etanólico bruto foi obtido por maceração e as frações foram obtidas por partições, líquido-líquido, sucessivas com hexano, diclorometano e acetato de etila. A Concentração Inibitória Mínima (CIM) foi determinada pelo método de diluição em ágar. O procedimento foi realizado segundo normas do NCCLS 2004 (M7-A7). A avaliação da toxicidade aguda dos extratos etanólico brutos teve como referência as Diretrizes 423 da Organisation for Economic Co-Operation and Development (OECD). A atividade leishmanicida foi testada para as formas promastigotas de Leishmania (L) chagasi. A maior parte das características morfoanatômicas observadas está de acordo com o padrão típico da família. Na prospecção fitoquímica da folha, cascas da raiz, cascas do caule e fruto de R. elaeocarpum verificou-se a presença de flavonóides e alcalóides. Verificou-se, também, a presença de heterosídeos saponínicos e taninos na folha, cascas da raiz, cascas do caule sendo ausente no fruto. Os extratos e as frações que obtiveram o menor valor de Concentração Inibitória Mínima (0,390 mg.mL-1) frente aos microrganismos testados apresentaram atividade inibitória moderada. Nenhum dos extratos e frações apresentou boa atividade inibitória. No que se diz a respeito à investigação toxicológica, durante todo o período de observação dos animais durante a avaliação de toxicidade aguda não ocorreu morte. Através das observações comportamentais feitas por meio do screening hipocrático atribuiu-se pontuação zero para todos os itens avaliados. A necropsia dos animais não evidenciou nenhuma alteração macroscópica que justificasse o estudo histopatológico dos órgãos (rins, baço e fígado) selecionados para análise. Neste estudo foi demonstrado ainda que a utilização do extrato bruto e as frações de folhas de R. elaeocarpum induziu a morte das formas promastigotas de L. (L) chagasi in vitro. Os resultados apresentados abrem perspectivas para o futuro isolamento, purificação e identificação das substâncias bioativas de R. elaeocarpum

Chá mate (Ilex paraguariensis): compostos bioativos e relação com atividade biológica / Mate tea (Ilex paraguariensis): bioactive compounds and relationship with biological activity

Introdução: A erva mate (Ilex paraguariensis St. Hilaire) é uma espécie nativa do Brasil, Paraguai e Argentina e tem importante apelo social, econômico e cultural. É utilizada na preparação de vários tipos de bebidas, como chimarrão, tererê e chá-mate, consumidas em toda a América Latina. O chá mate, assim como o chimarrão, apresenta potencial protetor à saúde devido à atividade antioxidante, ente outras, conforme demonstrado em diversos estudos. A alta concentração de compostos bioativos, como as saponinas, os compostos fenólicos e as metilxantinas respondem, em conjunto, pela sua atividade biológica. Objetivos: a) validar metodologia para isolar e quantificar as principais classes de compostos bioativos do chá mate (compostos fenólicos, xantinas, sapogeninas e melaninas); b) avaliar se há diferenças nos teores desses compostos nas diferentes marcas deste produto comercializado em São Paulo; c) avaliar a atividade biológica de cada classe por diferentes métodos in vitro. Metodologia: Foram analisados três lotes das três marcas de chá mate 100 por cento presentes em hipermercados no município de São Paulo. A validação da metodologia para quantificação, empregando CLAE, consistiu no cálculo da exatidão (recuperação), repetibilidade e sensibilidade para os compostos: ácido caféico, ácido-5-cafeoilquínico, cafeína, teobromina, acido ursólico e ácido oleanólico. Foram determinados os teores de fenólicos totais por método espectrofotométrico com o reagente Folin-Ciocalteu. As frações de fenólicos e xantinas, sapogeninas e melaninas foram isoladas e testadas quanto à atividade antioxidante pelo: sistema de oxidação -caroteno e ácido linoléico, teste da capacidade de absorbância de oxigênio radical ORAC (Oxygen Radical Absorbance Capacity), capacidade de seqüestrar o radical DPPH, e atividade ligante de ácidos biliares (ácidos glicocólico, cólico, deoxicólico e taurocólico). Resultados: Os coeficientes de determinação das curvas de calibração foram maiores que 0,99. A recuperação dos compostos variou de 84,2 a 115,88por cento e o coeficiente de variação médio foi 5 por cento. Uma xícara de chá mate (182 mL) apresentou, em média, 161,96 mg de fenólicos totais; 14,71 mg de ácido clorogênico (5CQA); 3,13 mg de ácido caféico ; 2,74 mg de teobromina e 15,16 mg de cafeína. Os teores de ácido ursólico e ácido oleanólico no chá mate (sachês), foram, em média 82,60 g/g e 54,08 g/g, respectivamente. As concentrações dos compostos variaram significativamente entre as marcas analisadas. A atividade antioxidante do chá mate (69,2 por cento de inibição da oxidação pelo sistema -caroteno e ácido linoléico; DPPH (IC50) = 0,014 mgSS/mL; ORAC = 9997molTrolox/xícara de chá), diferiu significativamente (p<0,05) entre as marcas apenas para o IC50. Para as frações houve diferença estatística apenas para a porcentagem de inibição da oxidação do ácido linoléico para o extrato melânico. Verificou-se correlação linear (p<0.05) positiva ou negativa entre a atividade antioxidante e a concentração de alguns compostos bioativos. A capacidade ligante de ácidos biliares foi semelhante ao controle positivo colestiramina para o chá mate, FS e FFX. O EM demonstrou baixa ou nenhuma capacidade de si ligar aos ácidos biliares. Conclusões: O chá mate é uma excelente fonte de compostos bioativos com atividade biológica. Ocorreu variação da concentração de compostos bioativos em função da marca analisada. Essa variação, no entanto, não influenciou na atividade antioxidante da bebida, conforme avaliada pelos métodos ora utilizados. / Introduction: Yerba maté (Ilex paraguariensis St. Hilaire) is a spontaneous species that grows in Brazil, Argentina and Paraguay and several beverages (chimarrão, terere, maté tea) are produced from its leaves. Maté tea, one of I. paraguariensis beverages present antioxidant activity and other biological effects. Compounds such as aponins, polyphenols and methylxanthines, which are present in considerable amounts, are responsible for the biological effects. Objectives: The aims of this work were: a) to validate analytical methodologies to evaluate the main bioactive compounds from maté tea (phenolic compounds, xanthines, sapogenins and melanins); b) to evaluate if there are differences on these compounds contents among maté tea brands commercialized in São Paulo; c) to evaluate biological activity of each class of compounds by different in vitro methodologies. Methods: Three lots from three commercial brands of maté tea present in 100 per cent of the main supermarkets in São Paulo city were analysed. HPLC methodology validation was assessed by determining accuracy (recovery), repeatability and sensibility (linearity, limits of detection and quantitation) for caffeic acid, 5- caffeoylquinic acid, caffeine, theobromine, ursolic and oleanolic acids. Total phenolic content was assessed with the Folin-Ciolcalteu´s reagent. Maté tea phenolic and methylxanthines (FFX), sapogenins (FS) and melanins (EM) fractions were isolated and tested for theirs in vitro antioxidantactivity by the following methods: -carotene and linoleic acid system, ORAC (Oxygen Radical Absorbance Capacity), radical scavenging capacity (DPPH). The ability of binding bile acids (glycocholic, cholic, taurocholic and deoxycholic acids) was also assayed. Results: Calibration curves determination coefficient (r 2) were higher than 0,99 for all compounds. Recovery ranged from 84,2 to 115,88per cent and repeatability average was 5per cent. One maté tea cup (182 mL) contains in average 161,96 mg5CQA of total phenolic compounds, 14,71 mg of chlorogenic acid (5CQA), 3,13 mg of caffeic acid, 2,74 mg of theobromine and 15,16 mg of caffeine. The roasted leaves contains 82,60 and 54,08 g/g of ursolic and oleanolic acids in average, respectively. Compounds contents varied significantly (p<0,05) among brands. The antioxidant activity assays showed no significant difference (p>0,05) among brands to maté tea, except for DPPH (IC50), and the average results for the maté tea are the following: 69,2 per cent in -carotene and linoleic acid system, DPPH (IC50) = 0,014mgSS/mL; ORAC = 9997,7molTrolox/tea cup. Antioxidant activity of the isolated fractions showed significant difference only in -carotene and linoleic acid system to melanic extract. A positive or negative correlations between the antioxidant activity and the bioactive compounds contents was observed (p<0.05). Maté tea, FS and FFX bile acid binding capacity were similar to the positive control cholestyramine. EM showed very little or no bile acid binding capacity. Conclusions: Maté tea is a source of bioactive compounds. Although concentration can vary significantly from brand to brand, this fact did not affect maté tea antioxidant activity evaluated by the methods used in this study.

This diploma thesis focuses on a research of the foam stability of the beer. We specifically observed the influence of ethanol and surface tension on the foam stability, and influence of ethanol and surface tension on the height of the foam. It was observed the influence of the hop acids in non-alcoholic beers. Furthermore, different biosurfactants were added into to the beer to observe better stability of the foam. The theoretical part of this work describes the foam from the physical aspect. There are described fourth key events involved in foam formation. Then it focuses on the beer foam – its structure, substances that influence its behavior and measurement of the foam stability. In this part there are described biosurfactants as well – their characteristics, qualities, distribution and applications. The experimental part is dedicated to the influence of ethanol, surface tension, the amount of iso- acids on foam stability in beer. There is observed the influence of biosurfactants of the foam stability and surface tension, which are added into to the beer. For the measurement of the foam stability was used the Rudin method. Liquid chromatography with DAD detector was used for the measurement the amount of the hop acids. The surface tension was determined by tensiometer. According to the results addition of ethanol changed the characteristics of the foam stability, but it’s not easy to find correlation between the increasing amount of ethanol and its stability. It was found out, that there is connection between the amount of ethanol and surface tension. On the other hand, there is not much connection between the surface tension and foam height. Within the framework of addition of biosurfactants was observed, that these foaming agents interacted the surface tension of the beer (lowering), however these agents accelerated the fall of the beer foam.

Saponin Removal from Quinoa by Abrasion Processing

Quinoa is coated with a thin layer of saponins, glycosylated triterpenoids, that produce a bitter flavor when consumed. The average saponin content in commercial varietals from Bolivia average around 2.7% saponins and organoleptic testing shows the threshold for noticing a bitter flavor is below 0.12% (Medina-Meza et al., 2016). Current industrial processing methods use a combination of abrasion and turbulent water flow to remove saponin. This study will address the following research question: How will grain-to-grain and grain-to-surface abrasion affect the processing time to remove saponin from quinoa? In particular, can effective saponin removal be achieved in less than 10 minutes without washing with water? Three different laboratory scale systems for generating grain-to-grain abrasion alone and in combination with grain-to-surface abrasion were constructed. Preliminary studies using mass balance and visual observation found the tubular system removed 4.45% of the quinoa mass in saponin containing fractions compared to the conical system (1.33%) and fluidized bed (0.62%). After preliminary studies, a saponin estimation method was adapted and the conditions of the tubular system were determined. A randomized experiment was carried out in triplicate at three levels of time (5, 10, and 15 minutes) and mass (200, 300, and 400 grams). The effect of surface roughness in the tubular system was also evaluated. The samples were collected and saponin content in each sample was quantified using the adapted method. The saponin levels were compared to commercially processed quinoa in the market. The processing conditions of 15 minutes at an input mass of 300 grams yielded the lowest saponin level of 0.19%. The four processing conditions of (10 minutes/200 grams, 10 minutes/400 grams, 15 minutes/200 grams and 15 minutes/300 grams) were statistically different (p The tubular system showed promise for reducing process time and saponin content in commercially processed quinoa without washing. However, further experimentation is needed for industrial implementation. The system would provide the industry with a sustainable process with better saponin removal capacity.

Contribution à l'étude phytochimique de Solidago virgaurea : application dans le domaine bucco-dentaire et étude de la variabilité phytochimique pour la création d'une filière / Contribution to the phytochemical study of Solidago virgaurea

Dans le but de valoriser la biodiversité végétale des Alpes-Maritimes, une plante commune dans cette région, Solidago virgaurea, a été sélectionnée pour son potentiel inhibiteur de la conversion levure-hyphe de Candida albicans, micro-organisme responsable d’infections bucco-dentaires de type candidose. Le fractionnement bioguidé de l’extrait aqueux a conduit à l’identification d’une famille de saponines particulièrement active. Parmi les onze saponines majoritaires caractérisées par RMN et HRMS, cinq se sont révélées être de nouvelles structures. Les tests biologiques ont néanmoins montré qu’elles n’étaient pas toutes actives contre la forme filamenteuse de C. albicans. Ces résultats ont conduit à une étude de la variabilité de la composition en saponines de plusieurs populations alpines de S. virgaurea. Trois méthodes de dosage des saponines ont été développées par HPLC et HPTLC. Les résultats ont démontré l'influence de différents facteurs sur la composition en saponines. Enfin, la composition globale de différents extraits de S. virgaurea a été étudiée dans le but d'identifier des activités biologiques complémentaires. Parmi les composés identifiés, trois nouveaux acides octulosoniques ont été caractérisés, aux côtés de trois composés phénoliques identifiés pour la première fois chez S. virgaurea. Les tests biologiques sur les extraits et fractions ont par ailleurs mis en évidence des activités antioxydante, anti-tyrosinase et inhibitrice de cellules cancéreuses in vitro. Ces tests devront être approfondis ultérieurement. / Toward the promotion of plant diversity of Maritime Alps, a common plant of the alpine area, Solidago virgaurea, was chosen to its inhibiting activity of Candida albicans yeast-hyphal conversion, a causal agent of opportunistic oral infections named candidiasis. In a first step, an aqueous extract of S. virgaurea was submitted to bioassay guided fractionation. This led to an active saponin-containing fraction from which eleven saponins were characterized by carrying out NMR experiments along with HRMS analyses. Five out of these were identified for the first time and bioassays showed that saponins activity varied according to the molecular structure of the compound. In a second step, the saponins composition of various S. virgaurea populations was studied qualitatively and quantitatively, using HPLC and HPTLC. Results demonstrated that saponins composition depends on various factors. Finally, the overall chemical composition of different S. virgaurea extracts was investigated searching for additional bioactivities. Among all the identified compounds, three new octulosonic acids were characterized and three phenolic compounds were found for the first time in S. virgaurea. Moreover, bioassays on extracts and fractions showed antioxidant, anti-tyrosinase activity and inhibition of cancer cell lines in vitro. Further bioassays have now to be completed. As a conclusion, this work was the starting point of an oral care product development and the setting-up of an innovative sector.

L’apparition de plusieurs maladies, telles que le cancer, le diabète, l'hypertension artérielle et la propagation d'infections de type virus mutagènes peuvent être liées à la qualité et au mode de vie que nous menons aujourd’hui. En effet, plusieurs études sur les facteurs déclenchant ces maladies dites "morbides" à long ou à court terme, sont liées au stress et à la qualité des aliments consommés, qu'ils soient d'origine végétale ou animale. Ces maladies deviennent un phénomène courant, elles touchent différentes races et toutes les catégories de la société. D'après les recherches ethnobotaniques, les substances d’origine naturelle, ont permis à des civilisations de survivre à des maladies mortelles. A titre d'exemple, on retrouve ainsi des références à des périodes de fièvre paludique en Chine et à des symptômes de cette maladie dans le «Huangdi Neijiang» Le Canon de Médecine datant des environs du premier siècle avant notre ère, plus de 2000 ans, qui relate de l'emploi de plantes médicinales, pour soulager les fièvres (Desgrouas et al., 2014).Vers 186 avant J.-C. apparaît, dans certaines régions de Chine, l'utilisation en tisane, du Qing hao su, appelé plus tard artémisinine en Occident et extrait d'une plante médicinale utilisée comme antipyrétique appelée "Qing hao", Artemisia annua ou Armoise annuelle. L'artemisinine bloque une enzyme qui permet au parasite de pomper le calcium et l'empêche ainsi de se développer. Au jour d'aujourd'hui l’Artemisinin-based combination therapy, en français Thérapie combinée à base d'artémisinine et en sigle ACT, est une thérapie et une prévention tertiaire dans les cas de paludisme simple.Dans cette optique notre étude vient s'ajouter à une longue série d'études menées sur les plantes médicinales et les substances naturelles extraites. Elle a pour objectif de révéler de nouvelles biomolécules, de mettre en évidence leurs activités biologiques grâce à des techniques de biotechnologies d'une part. D'autre part ces investigations permettront de valoriser les ressources naturelles qui se distinguent par leur endémicité.Pour se faire, notre choix s'est porté sur deux plantes médicinales endémiques à l'Algérie Cyclamen africanum Boiss. & Reuter et Zygophyllum cornutum Coss. , après une recherche ethnobotanique sur la pharmacopée traditionnelle du Nord de l'Afrique, et qui a révélé l’efficacité de ces plantes dans les problèmes inflammatoires minimes chez les autochtones, nous avons entrepris des investigations pharmaco- biochimiques.Ces dernières nous ont permis d'isoler : cinq composés à partir de l'extrait méthanolique des racines de l'espèce Cyclamen africanum Boiss. & Reuter, deux nouvelles saponines triterpéniques de type Oleanane, Afrocyclamin A et B (1, 2), ainsi que trois saponines triterpénoïdes connus sous le nom de lysikokianoside (3), deglucocyclamin I (4) et de son dérivé d'acide dicrotalique (5); et Sept saponosides connus à partir de l'extrait méthanolique de la plante entière de Zygophyllum cornutum Coss., ces saponosides sont de type ursane, ce type de triterpène est rapporté dans cette espèce pour la première fois et peuvent être considérés comme un marqueur chimio-taxonomique (chemotype) du genre Zygophyllum. Les structures ont été élucidées, sur la base de l'analyse des spectres de l'expérience RMN-1D et RMN- 2D (COSY, TOCSY, NOESY, HMBC et HSQC) et spectrométrie de masse en source FAB mode ion négatif. Des activités biologiques, des fractions saponosidiques Fr.1 et Fr.2, ont été testées sur des lignées de Rats mâles et femelles, de la race Winstar pour évaluer l'activité anti inflammatoire.La fraction saponosidique Fr.1 de Cyclamen africanum à la dose 5 mg, a montré un effet significatif sur l'inflammation causé par la carragénine, en réduisant l'oedème et la réponse immunitaire, qui s'est traduite par la concentration des protéines de la réponse inflammatoire (PRI) à travers leurs action sur les pro-médiateurs de l'inflammation (COX-2, PGE2, TNF -α, iNOS). / The appearance of several diseases, such as cancer, diabetes, high blood pressure and spread of infections mutagenic virus type can be linked to the quality and lifestyle that we lead today. Indeed, several studies on the factors triggering these so-called "morbid" long-or short-term illnesses are related to stress and quality of food consumed, whether of plant and animal origin. These diseases are becoming a common occurrence, they affect different races and all classes of society. According ethnobotanical research, naturally occurring substances, allowed civilizations to survive deadly diseases. For example, we thus find references to periods of malarial fever in China and one of the symptoms of this disease in the "Huangdi Neijiang" The Canon of Medicine dating from around the first century BC, more than 2000 years, which relates to the use of herbal medicines to relieve fevers (Desgrouas et al., 2014).Around 186 BC appears, in some parts of China, the use in herbal tea, Qing hao su, later known as artemisinin in the West and extracted from a medicinal plant used as antipyretic called "Qing hao" Artemisia annua or annual wormwood. Artemisinin blocks an enzyme which enables the parasite to pump calcium and prevents it from developing. As of today the Artemisinin-based combination therapy in French Combination therapy of artemisinin and ACT acronym, is a therapy and tertiary prevention in cases of uncomplicated malaria.From this perspective our study adds to a long series of studies on medicinal plants and natural substances extracted. It aims to reveal new biomolecules, highlighting their biological activities through techniques of biotechnology on the one hand. Moreover, these investigations will develop natural resources that are characterized by endemic.To do this, our choice is focused on two endemic medicinal plants in Algeria Cyclamen africanum Boiss. & Reuter and Zygophyllum cornutum Coss. After an ethnobotanical research on traditional medicine in Northern Africa, which showed the effectiveness of these plants in minimal inflammatory problems among Aboriginal, we undertook biochemical pharmacological investigations.The latter allowed us to isolate, five compounds from the methanol extract of the roots of the species Cyclamen africanum Boiss. Reuter & two new oleanane triterpene saponins type, Afrocyclamin A and B (1, 2) and three triterpenoid saponins known lysikokianoside of (3), deglucocyclamin I (4) and its derivative dicrotalique acid (5) September and known from the methanol extract saponins from the whole plant of Zygophyllum cornutum Coss. these saponins are ursane type, type triterpenes are reported in this species for the first time and can be considered a chemotherapy marker Taxonomic (chemotype) of Zygophyllum kind. The structures were elucidated on the basis of the analysis of NMR spectra of the experience-1D and 2D-NMR (COSY, TOCSY, NOESY, HSQC and HMBC) and mass spectrometry method negative ion FAB source. The biological activities of saponosidiques FR.1 and Fr.2 fractions were tested on lines of male and female rats of the Winstar rats to evaluate the anti-inflammatory activity. The saponosidique fraction FR.1 Cyclamen africanum the 5 mg dose, showed a significant effect on inflammation caused by carrageenan, reducing edema and immune response, which resulted in the concentration of protein the inflammatory response (PRI) through their action on the pro-inflammatory mediators (COX-2, PGE2, TNF -α, iNOS). The fraction of Fr.2 saponosidique Zygophyllum dose 20 mg did not show a significant effect on inflammation in general.
