This thesis focuses on research of classifier verbs in Czech sign language. The aim of the thesis is to describe different types of classifier verbs in Czech sign language based on language material collected from deaf users of Czech sign language by means of the Verb of motion production. The thesis defines classifier verbs of motion, location, visual and geometric description, and handling and also describes various types of classifier handshapes that form them for a semantic perspective. Syftet med den här studien var att undersöka och beskriva kuratorers arbete med döva och hörselskadade klienter. Frågeställningar som undersöktes var:-Hur beskriver kuratorerna sitt arbete med döva och hörselskadade klienter?-Vilka arbetsmetoder används av kuratorerna i arbetet med döva och hörselskadade klienter?-Vad innebär en kulturkompetens och en bikulturell kompetens enligt kuratorerna?Författaren ville belysa ämnet utifrån ett perspektiv där den professionella är teckenspråkig då hon anser att detta perspektiv saknas i litteraturen. Semistrukturerade intervjuer genomfördes med tre kuratorer som är teckenspråkiga och verksamma inom dövområdet. Resultatet visar att kuratorernas arbete består av praktiska åtgärder och stödsamtal. De har ett eklektiskt förhållningssätt i tillämpningen av arbetsmetoder, t.ex. att arbetet kan vara systematiskt med kognitiva inslag. Kuratorerna ansåg att det var viktigt att vara teckenspråkig och ha särskilda kunskaper om döva (kulturkompetens) för att kunna bemöta och bedriva behandlingsarbete med döva klienter. De ansåg också att kunskaper om den hörande världen och det svenska språket var betydelsefullt för att kunna ge information till hörande och döva om varandra (bikulturell kompetens). Resultatet visar också att kuratorerna uppfattade att annan typ av kommunikation än den teckenspråkiga var viktig att vara medveten om och reflektera över, så som den icke verbala kommunikationen. / The purpose of this study was to explore and describe the counselors’ work with deaf and hard of hearing clients. The issues examined were: -How do the counselors describe their work with deaf and hard of hearing clients?-What are the working methods used by the counselors at work with deaf and hard of hearing clients? -What does a cultural competence and a bicultural competence means according to the counselors? The author wanted to illustrate the subject form a perspective in which the professional can communicate in sign language since she believes that this perspective is lacking in the literature. Semi-structured interviews were conducted with three counselors who know sign language and work in the deaf area. The results show that the counselors’ work consists of practical measures and counseling. They have an eclectic approach in the application of the methods, for example that the work can be systematically with cognitive elements. The counselors thought it was important to know sign language and have specific knowledge about the deaf (cultural competence) in order to meet and engage in treatment with deaf clients. They also thought knowledge of the hearing world and the Swedish language was important in order to provide information to the hearing and deaf about one another (bicultural competence). The results also show that the counselors understood that other types of communication than the sign language communication was important to be aware of and to consider, as the non-verbal communication.

The Process by which Physicians Extract Information from Electronic Progress Notes During Handoffs

A handoff requires that the responsibility for patient patient’s care is transferred from one healthcare professional to another. The goals of this research were to identify, evaluate, and use analytical methods to describe how physicians (n=10) extracted information from electronic progress notes, one important source of information used during handoffs. Participants also verbally summarized the notes as they would during handoffs. Six methods were used to analyze how participants read progress notes, each uniquely contributing to our understanding of physicians’ visual attention patterns during this process. The participants focused their visual attention on the Impression and Plan section of the progress notes in that over 60% of the participants’ total time was spent reading that section. Physicians could miss an error or critical piece of information if the information is not located in the Impression and Plan. The importance given by the participants to the Impression and Plan section was confirmed in that the majority of participants’ verbal handoff content focused primarily on information that could be found in the Impression and Plan. Participants relied on the Medication Profile section quite heavily if it was present in the progress note. We determined that if the participant was currently reading in one section (s)he most likely would transition his/her visual attention to the physically closest section in the note, meaning the format of progress notes may dictate how notes are read. We determined what the most likely paths were through the progress notes, which could be a first step in reordering of the progress note for evaluation in future studies. Participants’ responses to debriefing questions suggested that they were aware of their reliance on the Impression and Plan, but that they thought the way they read notes is context-specific, depending on factors such as their use of the note and the reputation of the author of the note. These findings suggest a need for more research that evaluates how different note structures and content affect how physicians and other health providers extract and use information in varied clinical contexts.

SigN z Bacillus subtilis: Funkční charakterizace. / SigN from Bacillus subtilis: Functional characterization.

Bacillus subtilis strain 3610 is an ancestral undomesticated strain. It diers from the laboratory strain 168 in many aspects. One dierence in strain 3610 is the presence of plasmid pBS32 encoding the sigma factor N (σN). This σ factor is activated when DNA damage occurs and induces the bacteria's cell death. The aim of the Thesis was a systematic characterisation of σN-dependent transcription. First, I showed that plasmid-borne but not chromosome-borne predicted σN-dependent promoters were ac- tive in transcription in vitro. Next, the anities of RNAP with σN for DNA, initiating NTP (iNTP) were determined for both relaxed and supercoiled DNA templates. Sur- prisingly, the activity of RNAP on relaxed σN-dependent promoters was higher than on their supercoiled versions, an opposite trend than displayed by RNAP associated with other σ factors. This property of σN-dependent promoters was not encoded by the core promoter sequence. In summary, this Thesis contributed to our understanding of the bacterial transcription apparatus. 1

The dissertation deals with visuality, its perception by pupils of the secondary school. The theoretical part of the work defines the concept of image, visuality, their contextual frameworks. It takes into account the discursivity of these concepts. The aim of the research is to determine students' preferences in the concept of visuality. It seeks theoretical foundation in the areas of: visual sphere - artistic production, new media; art history; spheres of humanities - philosophy, pedagogy, psychology. It also deals with communication and linguistics. It applies knowledge of a semiotic nature. Research design is applied action research of a qualitative nature. It uses the methodology of Grounded Theory and triangulation. The research part of the thesis contains evaluated questionnaire surveys, reflecting selected artifacts. Interpreted student outcomes. These are evaluated using the obtained data to answer research questions. The thesis brings a new perspective on visual information, from the perspective of the pupil. Browser-Based Trust Negotiation

Trust negotiation allows two parties on the Internet to establish trust in each other according to the digital credentials thateach other possesses. Traditionally, trust negotiation uses certificates as digital credentials. However, certificates make trust negotiation difficult to use since people rarely have certificates available to them, and they must physically possess and secure all needed certificates in order to negotiate. To avoid these problems, this thesis proposes that credential authorities negotiate on behalf of the user. This thesis defines BrowserBased Trust Negotiation (BBTN) as a method for negotiating with credential authorities using the Secure Assertion Markup Language (SAML).
