Sylvie Stretti The dissertation submitted is dealing with the relationship between the subjects of the death of a close person and spirituality, especially personal concept of spirituality. Death of a close person is a crucial turning point in the life of a human being. The turning point affecting the next life in all aspects of the human life, in all its directions - biological-, psychological-, and sociological-spiritual. The complete care about a human being is more and more applied in the context of health or disciplines working with relations or connections; it is thus relevant to be aware of this fact of multidisciplinary cooperation and enrichment. I introduced the status of current approaches to subjects of the death of a close person and spirituality and introduction to the problematic in the beginning of my work. The Cardiovascular diseases may evocated the high percentual risk of sudden cardiac death in whole world. In several western countries is the number of death higher then number of cancer death. In this time is used a lot of methods for prediction of sudden cardiac death with focus on ECG T-wave alternance. The aim of the theses was to do stronger relation and cooperation with Internal Cardiac Clinic of Faculty Hospital Brno Bohunice on the risk analysis of sudden cardiac death. Secondly, we met the methods used for detection and quantification of simulated TWA. Last but not least was necessary to find TWA detection methods improvement and process the data on real signals obtained from Faculty Hospital Brno Bohunice. First part of the Thesis is focused on summary of pathologic artifacts in ECG signal, which are important for sudden cardiac risk stratification. There are described further known detection and quantification methods for TWA analysis. An interesting part for clinical practice is analysis of TWA trend in time and looking for the best method, which is able to catch and track the short TWA trend changes. Second part describes the new methods improvements, which were tested with interesting outputs. Further, there was developed method for TWA presence statement probability evaluation.

Sesqiterpene lactones, a group of plant secondary metabolites which include Cnicin from Cnicus benedictus plant, have an anti-proliferative and anti-tumor effect on mammalian cells by activating specific signaling pathways while also generating oxidative stress. These factors combined drive tumor cell apoptosis. A few of these compounds have reached clinical trials and seem to be a promising chemotherapeutics. The focus of this work is to elucidate the effect of cnicin on C-MYC transcription factor and oncoprotein which is overexpressed in majority of tumor tissues, the effect of cnicin on DEAD-box RNAhelicase DDX3 and on the expression levels of several metabolic genes is also studied. Through the use of western blotting, immunodetection and qPCR it was found out, that cnicin is regulating the expression of C-MYC oncoprotein on both transcriptional and translational levels, while also lowering C-MYC protein stability probably through the effect on PIM-2 kinase. Cnicin is not affecting the total amount of DDX3 protein in cells, but it seems it is lowering its degradation rate. The possible transcriptional regulation of DDX3 by cnicin is still not clear and requires further research. Organ transplantation is a method of treatment of damaged or missing organ by replacement by a new one that is removed from a donor. Donors may be not only living people but also dead people after cardiac arrest or people with brain death. The aim of the study was to assess the attitude of laymen to organ transplantation. The quantitative research used questionnaires which were distributed since 25th October - 31st December 2017 to non-medical adults older than 18 years living in the Czech Republic. The results demonstrated that the laymen have basic knowledge which organs and tissues can be used for transplantation in humans. Total 110 respondents answered that organs can be removed from people with diagnosed brain death. Knowledge of legislation was a surprise. Over one half of respondents knew about implied consent with organ removal after death and knew at least one possibility how to deny post-mortem organ harvesting. Generally, the public lacks interest in organ donation. People dislike registration unless necessary and dislike discussion concerning death which is a symbol of pain and suffering. Majority of public agree that there is need for more education in the field of organ transplantation and donation. They would prefer a specialised lecture or discussion with medical professionals....

The aim of the dissertation is to present the complex system of Buddhist rituals of death. Being the first lengthy study on the topic written outside Korea, this work introduces these rituals in the context of the rich Korean religious scene and indicates its connection to other religious and philosophical ideas present in the Sinitic cultural area. The dissertation is structured into five sections, each of which approaches the rituals from a different perspective. The "Introduction," presents the methodology, including the theoretical framework; textual research of ritual texts and fieldwork are the two key methodological approaches used to study the topic at hand. Chapter 2, "The Ritual Constituents," is an attempt to define an apparatus for analysis of the rituals. The two-axis scheme is an attempt to provide a tool, which will help us grasp not only the relative "richness" of the ritual but also its variability. Chapter 3, "The Structure and Typology of the Rituals," presents an overall analysis of the ritual process from the deathbed, to rebirth, and beyond. Its structure is based on contemporary rituals. Předkládaná práce je nesena snahou o zodpovězení otázky: V čem spočívá nesoulad filosofie? Nemůže být Lévinasova etika chápána jako doplnění Heideggerovy ontologie? Na zmíněné otázky je hledána odpověď prostřednictvím vpravení se do možného "dialogu" mezi Heideggerem a Lévinasem. Odlišné pojímání bytí u obou filosofů se odhalí nejenom jako možná příčina sporu, ale i jako důvod, kvůli kterému by nemohla být Lévinasova a Heideggerova filosofie nikdy komplementární. Lévinas na rozdíl od Heideggera, který je neustále na cestě bytí, bude rozvíjet veškerá svá témata na rovině "autrement qu'être", hranicemi bytí.
