Role of nuclear rotation in H[subscript]2[superscript]+ dissociation by ultra short laser pulses

Esry / The nuclear rotational period of the simplest molecule H[subscript]2[superscript]+ is about 550 fs, which is more than 35 times longer than its vibrational period of 15 fs. The rotational time scale is also much longer than widely available ultra short laser pulses which have 10 fs or less duration. The large difference in rotational period and ultra short laser pulse duration raises questions about the importance of nuclear rotation in theoretical studies of H[subscript]2[superscript]+ dissociation by these pulses. In most studies, reduced-dimensionality calculations are performed by freezing the molecular axis in one direction, referred to as the aligned model. We have systematically compared the aligned model with our full-dimensionality results for total dissociation probability and field-free dynamics of the dissociating fragments. The agreement between the two is only qualitative even for ultra short 10 fs pulses. Post-pulse dynamics of the bound wave function show rotational revivals. Significant alignment of H[subscript]2[superscript]+ occurs at these revivals. Our theoretical formulation to solve the time-dependent Schrodinger equation is an important step forward to make quantitative comparison between theory and experiment. We accurately calculate observables such as kinetic energy, angular, and momentum distributions. Reduced-dimensionality calculations cannot predict momentum distributions. Our theoretical approach presents the first momentum distribution of H[subscript]2[superscript]+ dissociation by few cycle laser pulses. These observables can be directly compared to the experiment. After taking into account averaging steps over the experimental conditions, we find remarkable agreement between the theory and experiment. Thus, our theoretical formulation can make predictions. In H[subscript]2[superscript]+ dissociation by pulses less than 10 fs, an asymmetry in the momentum distribution occurs by the interference of different pathways contributing to the same energy. The asymmetry, however, becomes negligible after averaging over experimental conditions. In a proposed pump-probe scheme, we predict an order of magnitude enhancement in the asymmetry and are optimistic that it can be observed.

Regulation of human endocardial endothelial cells' secretion of endothelin-1 by neuropeptide Y

Endocardial endothelial cells (EECs) can exert a significant influence on cardiac function by releasing various factors such as nitric oxide (NO), prostanoids, endothelin-1 (ET-1) and angiotensin II (Ang II). Recently, results obtained in our laboratory demonstrated the presence of NPY and its receptors, Y[subscript 1] and Y[subscript 2], as well as ET-1 and its receptors, ET[subscript A] and ET[subscript B], at the level of endocardial endothelial cells (EECs). We have also shown that NPY induces a sustained rise in the intracellular calcium level of these cells, and that only right ventricular EECs have the capacity of secreting NPY. Moreover, the evidence in the literature has become plentiful about complex interactions existing between ET-1 and other cardioactive mediators, such as NO and Ang II. Based on the above-mentioned data, the objective of this study was to investigate if a dialogue equally exists between the systems of NPY and ET-1 at the level of human right (hREECs) and left (hLEECs) ventricular EECs. Using the technique of indirect immunofluorescence coupled to 3-D confocal microscopy, as well as ELISA, our results show that increasing concentrations of NPY (10[superscript -15], 10[superscript -10] and 10[superscript -5]M) induce the release of ET-1 from REECs and LEECs in a time- and dose-dependent fashion. However, right ventricular EECs seem to have a higher ET-1 secretory capacity as compared to their left counterparts. Upon the use of selective antagonists for the NPY receptors, Y[subscript 1], Y[subscript 2] and Y[subscript 5], and the ET-1 receptors, ET[subscript A] and ET[subscript B], our results demonstrated that in REECs the NPY-induced release of ET-1 seems to be primarily due to Y[subscript 2] receptor activation, with the subsequent activation of the ET[subscript A] and ET[subscript B] receptors by the released ET-1. On the other hand, in LEECs, the NPY-evoked secretion of ET-1 seems to be mainly the result of Y[subscript 5] receptor activation by NPY. Unlike REECs, the ET-1 released by NPY in this type of cells does not seem to be contributing further to its own release by activation of its ET[subscript A] and ET[subscript B] receptors. Therefore, our results suggest that NPY is a regulator of ET-I secretion at the level of human EECs, and that this secretory process of ET-1 is different between the right and left ventricular cells. Moreover, these results serve to highlight and endorse the important sensory and tuning roles that right and left ventricular EECs possess, respectively. The ability of EECs to contribute to the local as well as systemic release of factors, such as NPY and ET-1, can affect not only the excitation-secretion coupling of EECs and the excitation-contraction coupling of cardiomyocytes, but also the physiological and pathophysiological state of the underlying, heart muscle.

L’ischémie des membres inférieurs chez les diabétiques : le rôle du récepteur AT2 / Lower limb ischemia in diabetic patients : the AT2 receptor

Résumé : Les patients diabétiques ont plus de risques d’être amputés d’une jambe en raison d’une plus faible néovascularisation suite à une ischémie. Nous avons montré une association entre une plus faible réponse angiogénique du VEGF chez les souris diabétiques (DM) et une augmentation de l’expression de SHP-1, pouvant être activée par les récepteurs (AT[indice inférieur 1]/AT[indice inférieur 2]). La délétion du récepteur AT[indice inférieur 2] chez des souris favorise l’angiogenèse dans le muscle ischémique, mais son rôle en condition diabétique demeure inconnu. Notre objectif est de vérifier si la délétion du récepteur AT[indice inférieur 2] chez des souris DM favorise l’angiogenèse suivant l’induction d’une ischémie. Des souris DM de type 1 déficientes (KO) ou non pour le récepteur AT[indice inférieur 2] ont été utilisées. L’ischémie a été induite par la ligature de l'artère fémorale. La perfusion sanguine a été mesurée pendant 2 ou 4 semaines avant la récolte des tissus. Les effets de l’ischémie sur l’expression des récepteurs AT[indice inférieur 1] et AT[indice inférieur 2], des phosphatases SHP-1, SHP-2 et PTP1B, ainsi que l’état de la voie de signalisation du VEGF ont été mesurés. Un essai phosphatase a aussi été effectué suite à l’immunoprécipitation de SHP-1 chez des BAECs stimulés au CGP42112A. Quatre semaines après la chirurgie, le flot sanguin dans le muscle ischémique des souris DM AT[indice inférieur 2]KO s’est rétabli plus rapidement (80%) comparativement à une récupération de 47% chez les souris DM contrôles. L’expression des facteurs pro-angiogéniques (HIF-1α et VEGF) était similaire dans tous les groupes après 2 semaines d’ischémie, mais diminuée chez les DM et retournait à un niveau basal chez les DM-AT[indice inférieur 2]KO après 4 semaines, suggérant un reperfusion plus rapide chez ces souris. La phosphorylation de Akt était aussi plus faible chez les souris DM contrôles mais était rétablie chez les souris AT[indice inférieur 2]KO après 4 semaines d’ischémie. L'expression de SHP-1 était doublée dans le muscle ischémique des souris DM, en comparaison aux souris non DM, un effet absent chez les souris DM AT[indice inférieur 2]KO. L’expression de SHP-2 et PTP1B ne variait pas chez les souris DM sauvages et AT[indice inférieur 2]KO. De plus, l’expression des récepteurs AT[indice inférieur 1] et AT[indice inférieur 2] est augmentée chez les souris DM sauvages en comparaison aux souris NDM. La stimulation du récepteur AT[indice inférieur 2] chez les BAECs a permis d’augmenter l’activité phosphatase de SHP-1. Nos résultats suggèrent que l’expression élevée d’AT[indice inférieur 2] chez les souris DM mène à la surexpression et/ou l’activation de SHP-1, inhibant le signal angiogénique issu du VEGF et empêchant la reperfusion sanguine suite à l’ischémie. / Abstract : Ischemia due to narrowing of femoral artery and distal vessels is a major cause of peripheral arterial disease and morbidity affecting patients with diabetes. We have previously reported that the inhibition of the angiogenic response to PDGF and VEGF in diabetic mice (DM) is associated with the increase expression of SHP-1, a protein that can be activated by the AT[subscript 2] receptor. It has been shown that the deletion of AT[subscript 2] receptor in mice promotes angiogenesis within the ischemic muscle, but its role in diabetic condition remains unknown. Our hypothesis is that AT[subscript 2] receptor induced SHP-1 which contributed to inhibition of pro-angiogenic factor actions in DM mice during ischemia. Non-DM and DM AT[subscript 2] null mice underwent femoral artery ligation. Blood perfusion was measured every week up to 4 weeks post-surgery. Expression of AT[subscript 1] and AT[subscript 2] receptors, SHP-1, SHP-2 and PTP1B, and VEGF pathway was evaluated. A phosphatase assay was executed to assess SHP-1 activity in bovine aortic endothelial cells (BAECs) following CGP42112A stimulation. Blood flow in the ischemic muscle of DM AT[subscript 2]KO mice recovered faster and up to 80% four weeks following the surgery, compared to a 47% recovery in DM mice. After four weeks, the expression of pro-angiogenic factors (HIF-1α and VEGF) was similar in each group after two weeks but diminished in the DM and remained at a basal state in the DM-AT[subscript 2]KO after four weeks suggesting a faster recovery process in these mice. Akt phosphorylation was also diminished in the DM mice but restored in DM AT[subscript 2]KO after four weeks of ischemia. SHP-1 expression was two times more elevated in DM compared to NDM mice, an effect that was not seen in AT[subscript 2]KO mice. SHP-2 and PTP1B expression did not varied in DM wildtype and AT[subscript 2]KO mice. Also, expression of AT[subscript 1] and AT[subscript 2] receptor was upregulated by diabetes in wildtype mice. Stimulation of AT[subscript 2] receptor in BAECs caused increased SHP-1 activity. Our results suggest that elevated expression of the AT[subscript 2] receptor in DM mice lead to up-regulation/activation of SHP-1, inhibiting the VEGF angiogenic signal causing diminished blood flow reperfusion following ischemia.

Development of an Exercise Test to Predict VO2max in Children and Adolescents

The purpose of this study was to evaluate the use of nonexercise (N-EX) data, specifically, the Perceived Functional Ability (PFA) and Physical Activity Rating (PA-R) questionnaires, with the treadmill walk-jog-run protocol to estimate VO2max in 12 to 17 year old boys and girls. Ninety-one participants (49 males and 42 females) took part in this study. Data were collected via PFA and PA-R questionnaires, a walk-jog-run submaximal treadmill test, and a maximal graded exercise test (GXT). Data collected included gender, age, height, weight, PFA and PA-R scores, heart rate (HR), treadmill speed, maximal treadmill grade, respiratory exchange ratio (RER), rating of perceived exertion (RPE), and VO2max. Regression analysis resulted in the development of two valid and reliable models to predict VO2max. Nonexercise and submaximal exercise test data were used to build the following model: VO2max (mL∙kg-¹∙min-¹) = 11.201 + (6.877 x Gender; 0 = female; 1 = male) + (3.573 x treadmill speed; mph) – (0.174 x kg) + (0.405 x PFA score) + (0.653 x PA-R score) + (1.019 x age). The model resulted in an R2 = 0.69 and a SEE = 5.16 mL∙kg-¹∙min-¹. Maximal exercise test data were used to build the following model: VO2max (mL∙kg-¹∙min-¹) = -3.264 + (3.359 x Gender; 0 = female; 1 = male) – (0.082 x kg) + (7.351 x treadmill speed; mph) + (1.750 x maximal treadmill grade). The model resulted in an R2 = 0.88 and a SEE = 3.16 mL∙ kg-¹∙min-¹. The cross-validation PRESS statistics for both models demonstrated minimal shrinkage in the accuracy of the regression model. The results of this study demonstrate, for the first time, that N-EX data can be used to accurately predict VO2max in youth. The submaximal and maximal exercise tests validated in this study can be used to assess cardiorespiratory fitness of youth having a wide range on interests and fitness levels. In addition, the use of PFA and PA-R questionnaires enforces initiatives to increase physical activity among youth. Both exercise tests use a self-selected treadmill speed that elicits a steady-state HR of 70% of the participants age-predicted maximal HR. The use of a self-selected walking, jogging, or running speed accommodates youth with different levels of physical fitness, motivation, and interests. The exercise test protocol presented in this study is practical for use in schools, athletic facilities, and community fitness centers. The equipment required to administer the exercise test presented in this study is limited to a treadmill and a HR monitor. Together with the use of PFA and PA-R questionnaires, the submaximal and maximal exercise tests are efficacious to coaches, fitness professionals, and physical educators in a variety of settings.

The •OH scavenging effect of bromide ions on the yield of H[subscript 2]O[subscript 2] in the radiolysis of water by [superscript 60]Co γ-rays and tritium β-particles at room temperature : a Monte Carlo simulation study / Effet de capture des radicaux •OH par les ions bromure Br- sur le rendement de H[indice inférieur 2]O[indice inférieur 2] dans la radiolyse de l'eau par les rayons γ de [indice supérieur 60]Co et les électrons β du tritium à la température ambiante: une étude par simulation Monte Carlo

Abstract: Monte Carlo simulations were used here to compare the radiation chemistry of pure water and aqueous bromide solutions after irradiation with two different types of radiation, namely, tritium β-electrons (~7.8 keV) and [superscript 60]Co γ-rays/fast electron (~1 MeV) or high energy protons. Bromide ions (Br-) are known to be selective scavengers of hydroxyl radicals •OH precursors of hydrogen peroxide H[subscript 2]O[subscript 2]. These simulations thus allowed us to determine the yields (or G-values) of H[subscript 2]O[subscript 2] in the radiolysis of dilute aqueous bromide solutions by the two types of radiations studied, the first with low linear energy transfer (LET) (~0.3 keV/μm) and the second with high LET (~6 keV/μm) at 25 °C. This study was carried out under a wide range of Br- concentrations both in the presence and the absence of oxygen. Simulations clearly showed that irradiation by tritium β-electrons favored a clear increase in G(H[subscript 2]O[subscript 2]) compared to [superscript 60]Co γ-rays. We found that these changes could be related to differences in the initial spatial distributions of radiolytic species (i.e., the structure of the electron tracks, the low-energy β-electrons of tritium depositing their energy as cylindrical “short tracks” and the energetic Compton electrons produced by γ-radiolysis forming mainly spherical “spurs”). Moreover, simulations also showed that the presence of oxygen, a very good scavenger of hydrated electrons (e-[subscript aq]) and H• atoms on the 10[superscript-7] s time scale (i.e., before the end of spur expansion), protected H[subscript 2]O[subscript 2] from further reactions with these species in the homogeneous stage of radiolysis. This protection against e-[subscript aq] and H• atoms therefore led to an increase in the H[subscript 2]O[subscript 2] yields at long times, as seen experimentally. Finally, for both deaerated and aerated solutions, the H[subscript 2]O[subscript 2] yield in tritium β-radiolysis was found to be more easily suppressed than in the case of cobalt-60 γ-radiolysis, and interpreted by the quantitatively different chemistry between short tracks and spurs. These differences in the scavengeability of H[subscript 2]O[subscript 2] precursors in passing from low-LET [superscript 60]Co γ-ray to high-LET tritium β-electron irradiation were in good agreement with experimental data, thereby lending strong support to the picture of tritium-β radiolysis in terms of short tracks of high local LET. / Résumé: Les simulations Monte Carlo constituent une approche théorique efficace pour étudier la chimie sous rayonnement de l'eau et des solutions aqueuses. Dans ce travail, nous avons utilisé ces simulations pour comparer l’action de deux types de rayonnement, à savoir, le rayonnement γ de [indice supérieur 60]Co (électrons de Compton ~1 Me V) et les électrons β du tritium (~ 7,8 keV), sur la radiolyse de l’eau et des solutions aqueuses diluées de bromure. Les ions Br- sont connus comme d’excellents capteurs des radicaux hydroxyles •OH, précurseurs du peroxyde d’hydrogène H[indice inférieur 2]O[indice inférieur 2]. Les simulations Monte Carlo nous ont donc permis de déterminer les rendements (ou valeurs G) de H[indice inférieur 2]O[indice inférieur 2] à 25 °C pour les deux types de rayonnements étudiés, le premier à faible transfert d'énergie linéaire (TEL) (~0,3 keV/μm) et le second à haut TEL (~6 keV/μm). L’étude a été menée pour différentes concentrations d’ions Br-, à la fois en présence et en absence d'oxygène. Les simulations ont montré que l’irradiation par les électrons β du tritium favorisait nettement la formation de H[indice inférieur 2]O[indice inférieur 2] comparativement aux rayons γ du cobalt. Ces changements ont pu être reliés aux différences qui existent dans les distributions spatiales initiales des espèces radiolytiques (i.e., la structure des trajectoires d'électrons, les électrons β du tritium déposant leur énergie sous forme de «trajectoires courtes» de nature cylindrique, et les électrons Compton produits par la radiolyse γ formant principalement des «grappes» de géométrie plus ou moins sphérique). Les simulations ont montré également que la présence d'oxygène, capteur d’électrons hydratés et d’atomes H• sur l'échelle de temps de ~10[indice supérieur -7] s (i.e., avant la fin des grappes), protégeait H[indice inférieur 2]O[indice inférieur 2] d’éventuelles réactions subséquentes avec ces espèces. Une telle «protection» conduit ainsi à une augmentation de G(H[indice inférieur 2]O[indice inférieur 2]) à temps longs. Enfin, en milieu tant désaéré qu’aéré, les rendements en H[indice inférieur 2]O[indice inférieur 2] obtenus lors de la radiolyse par les électrons β du tritium ont été trouvés plus facilement supprimés que lors de la radiolyse γ. Ces différences dans l’efficacité de capture des précurseurs de H[indice inférieur 2]O[indice inférieur 2] ont été interprétées par les différences quantitatives dans la chimie intervenant dans les trajectoires courtes et les grappes. Un excellent accord a été obtenu avec les données expérimentales existantes.

Comportement d'une discontinuité dans un géomatériau sous sollicitation chemo-mécanique : expérimentations et modélisations

Résumé : Ces travaux de thèse s'intéressent à l'étude du comportement d'une discontinuité dans un géomatériau sous sollicitations chemo-mécaniques à l'échelle du laboratoire. Des essais de traction-compression cyclique étudient la refermeture d'une fissure. Ils indiquent que les déformations inélastiques seraient gouvernées en partie par les frottements générés lors du ré-emboîtement des lèvres de la discontinuité, non correspondantes du fait des contraintes internes. Des joints rocheux altérés chimiquement sous sollicitation tangentielle sont étudiés au travers d'essais de cisaillement direct : le comportement des joints dégradés est profondément modifié du fait de la diminution des propriétés mécaniques du matériau de part et d'autre de la discontinuité et de l'accentuation de la non-correspondance des profils rugueux. Le comportement d'une discontinuité est modélisée par le couplage d'un modèle élasto-plastique endommageable continu avec une résolution discrète du problème de contact/frottement (code calcul aux éléments finis Cast3M). Les résultats numériques confirment les phénomènes constatés expérimentalement.//Abstract: To probate the technology of CO[indice inférieur 2] geological storage, the integrity of the site must be assure over time. This industruial problematic involves the study of the mechanical properties alteration of geomaterials in the presence of CO[indice inférieur 2]. The scenario at the origin of this thesis illustrates the possibility of a CO[indice inférieur 2] leakage on a fault located in the caprock. This geological problem is complicated by the many parameters to consider: in situ temperature and pressure, scale effect, heterogeneities of the geomaterial, geofluide composition, chemical reactions ... These works focus on the behavior of a discontinuity in a geomaterial solicited chemomechanically at the laboratory scale. They were realised in cotutelle between SIAME laboratory at the University of Pau and Pays de l'Adour (France) and the laboratory of rock mechanics and engineering geology from the University of Sherbrooke (Quebec, Canada). The first part of the experimental program was defined to characterize the reclosing of a crack under cyclic uniaxial stress. The second experimental campaign has studied the shear behavior of a rock joint chemically degraded. The data obtained were used to model the behavior of a discontinuity by the finite element method. The mechanical behavior of a crack under normal stress is assessed with cyclic tension-compression tests. Stress curve showed hysteresis during opening and closing cycles of a discontinuity in concrete, it indicated inelastic deformations The analysis of displacement field by image correlation indicated that theses deformations were partially governed by the friction generated during the closing of the discontinuity lips. Frictional phenomena are due to asperities mismatching induced by the internal stresses in the concrete. The shear behavior of a rock joint chemically damaged was studied through direct shear tests. Rough surfaces were immersed in acid solution during 6 hours at constant pH. Digitalization of these surfaces befor and after immesion, with a lase profilometer, indicates little modifications of the geometry induced by dissolution of material. Results of tests pointed out significant modifications for altered joints illustred by a of the peak shear strength and an increased of contractancy. They are induced by: 1) the mismatch enhancement of the rough profiles of the discontinuity and, 2) the degradation of the mechanical properties of the material on both sides of the discontinuity due to the chemical attack. Numerical contribution of the thesis lies in modeling the behavior of a discontinuity by the coupling of an continuous elastic-plastic damaged model with a discrete resolution of the contact/friction problem. The model is developed with the finite element code Cast3M. Geometries lips discontinuities are modeled directly from the roughness profiles from experimental scans. The numerical results correctly represent the friction phenomena observed experimentally. Finally, a model of the shear test altered joints is performed by coupling the mechanical model with chemical damage model.

Monte Carlo simulation of the radiolysis of water by fast neutrons at elevated temperatures up to 350°C / Simulation Monte Carlo de la radiolyse de l'eau par des neutrons rapides à températures élevées allant jusqu'à 350°c

Résumé : Le contrôle de la chimie de l'eau dans un réacteur nucléaire refroidi à l'eau nécessite une compréhension détaillée des effets de la radiolysede l'eau afin de limiter la corrosion et la dégradation des matériaux par oxydation générée par les produits de cette radiolyse. Toutefois, la mesure directe de la chimie dans le cœur des réacteurs est extrêmement difficile, sinon impossible, en raison des conditions extrêmes de haute température et haute pression, et les champs d’irradiation mixtes neutrons/γ, qui ne sont pas compatibles avec l'instrumentation chimique normale. Pour ces raisons,des modèles théoriques et des simulations sur ordinateur sont essentielles pour la prédiction de la chimie sous rayonnement de l'eau de refroidissement dans le cœur et son impact sur les matériaux. Dans ce travail, des simulations Monte Carlo ont été utilisées pour calculer les rendements des principales espèces (e[indice supérieur -][indice inférieur aq], H[indice supérieur •], H[indice inférieur 2], [indice supérieur •]OH et H[indice inférieur 2]O[indice inférieur 2]) formées lors de la radiolyse de l’eau liquide neutre par des neutrons mono-énergétiques de 2 MeV à des températures entre 25 et 350 °C. Le choix des neutrons de 2 MeV comme énergie d'intérêt est représentatif du flux de neutrons rapides dans un réacteur. Pour l'eau légère, la contribution la plus significative à la radiolyse vient des quatre premières collisions des neutrons qui produisent, dans la majorité des cas, des protons avec des énergies de recul de ~1.264, 0.465, 0.171 et 0.063 MeV et des transferts d’énergie linéique (TEL) moyens respectivement de ~22, 43, 69et 76 keV/[micro]m. Par ailleurs, nous avons négligé les effets des radiations dus aux ions de recul de l’oxygène. Les rendements moyens finaux peuvent alors être estimés comme étant la somme des rendements résultant de l’action de ces protons après pondérations en fonction de l’énergie déposée. Les rendements ont été calculés à 10[indice supérieur -7], 10[indice supérieur -6] et 10[indice supérieur -5] s. Les valeurs obtenues sont en accord avec les données expérimentales disponibles. En comparant nos résultats avec les données obtenues pour les rayonnements à faible TEL (rayons γ de [indice supérieur 60]Co ou électrons rapides), nos rendements calculés pour les neutrons rapides ont montré une dépendance en température essentiellement similaire, mais avec des valeurs plus faibles pour les rendements en radicaux libres et des valeurs plus élevées pour les rendements moléculaires. Nous avons également utilisé les simulations Monte Carlo pour étudier l'existence de la chute rapide de la constante de vitesse de réaction de l'électron hydraté (e[indice supérieur -][indice inférieur aq]) sur lui-même – l’une des principales sources de formation de H[indice inférieur 2] – au-dessus de 150 °C. Cette dépendance en température a été observée expérimentalement en milieu alcalin par divers auteurs, mais jamais en milieu neutre. Lorsque cette baisse de la constante de vitesse d’auto-réaction de e[indice supérieur -][indice inférieur aq] est incluse dans nos codes de simulation, tant pour des rayonnements de bas TEL (grappes isolés) que de haut TEL (trajectoires cylindriques), g(H[indice inférieur 2]) montre une discontinuité marquée à la baisse à ~150°C, ce qui n'est pas observée expérimentalement. Les conséquences de la présence de cette discontinuité dans le rendement en H[indice inférieur 2] pour les rayonnements à bas et haut TEL sont discutées. Enfin, nous avons tenté d’expliquer l'augmentation – considérée comme anormale – du rendement en H[indice inférieur 2] en fonction de la température au-dessus de 200 °C par l’intervention de la réaction des atomes H[indice supérieur •] avec l'eau, préalablement proposée par Swiatła-Wojcik et Buxton en 2005. La constante de vitesse de cette réaction est toujours controversée. // Abstract : Controlling the water chemistry in a water-cooled nuclear power reactor requires understanding and mitigating the effects of water radiolysis to limit the corrosion and degradation of materials by oxidizing radiolysis products. However, direct measurement of the chemistry in reactor cores is extremely difficult due to the extreme conditions of high temperature, pressure, and mixed neutron/γ-radiation fields, which are not compatible with normal chemical instrumentation. For these reasons, theoretical models and computer simulations are essential for predicting the detailed radiation chemistry of the cooling water in the core and the impact on materials. Monte Carlo simulations were used to calculate the yields for the primary species (e[superscript -][subscript aq], H[superscript •], H[subscript 2], [superscript •]OH, and H[subscript 2]O[subscript 2]) formed from the radiolysis of neutral liquid water by mono-energetic 2-MeV neutrons and the mechanisms involved at temperatures between 25 and 350 °C. In this work, we chose 2-MeV neutron as our energy of interest since it is known as representative of a fast neutron flux in a nuclear reactor. For light water, for that chosen energy, the most significant contribution to the radiolysis comes from the first four neutron collisions that generate mostly ejected protons with energies of ~1.264, 0.465, 0.171, and 0.063 MeV, which had, at 25 °C, mean linear energy transfers (LETs) of ~22, 43, 69, and 76 keV/[micro]m, respectively. In this work, we simply neglected the radiation effects due to oxygen ion recoils. The average final fast neutron yields could be estimated as the sum of the yields for these protons after allowance was made for the appropriate weightings (by using the Eq (2) in Chapter 4) according to their deposited energy. Yields were calculated at 10[superscript -7], 10[superscript -6] and 10[superscript -5] s. Our computed yield agreed reasonably well with the available experimental data. By comparing our results with data obtained for low-LET radiation ([superscript 60]Co γ-rays or fast electrons), our computed yields for fast neutron radiation showed essentially similar temperature dependences over the range of temperature studied, but with lower values for yields of free radicals and higher values for molecular yields. In this work, we also used our Monte Carlo simulation to investigate the existence of drop of hydrated electron (e[superscript -][subscript aq]) self-reaction rate constant at 150 °C. One of the main sources of H[subscript 2] formation is the self-reaction of hydrated electrons. The temperature dependence of the rate constant of this reaction (k[subscript 1]), measured under alkaline conditions, reveals that the rate constant drops abruptly above ~150 °C. However, when this drop in the e[superscript -][subscript aq] self-reaction rate constant is included in our code for low (isolated spurs) and high (cylindrical tracks) linear energy transfer (LET), g(H[subscript 2]) shows a marked downward discontinuity at ~150 °C which is not observed experimentally. The consequences of the presence of this discontinuity in H[subscript 2] yield for both low and high LET radiation are discussed. Another reaction that might explain the anomalous increasing of H[subscript 2] yield with temperature is the reaction of H[superscript •] atoms with water previously proposed by Swiatla-Wojcik and Buxton (2005) whose rate constant is still in controversial.

The role of P2Y[subscript]2 nucleotide receptor in lipoprotein receptor-related protein 1 expression and aggregated low density lipoprotein uptake in vascular smooth muscle cells

Medeiros / Laman Mamedova / The internalization of aggregated low-­density lipoprotein (agLDL) may involve the actin cytoskeleton in ways that differ from the endocytosis of soluble LDL. Based on previous findings the P2Y[subscript]2 receptor (P2Y[subscript]2R) mediates these effects through interaction with filamin‐A (FLN‐A), an actin binding protein. Our findings also showed that uridine 5’‐ triphosphate (UTP), a preferential agonist of the P2Y[subscript]2R, stimulates the uptake of agLDL, and increases expression of low‐density lipoprotein receptor related protein 1 (LRP 1) in cultured mouse vascular smooth muscle cells (SMCs). The strategy of this research was to define novel mechanisms of LDL uptake through the modulation of the actin cytoskeleton in order to identify molecular targets involved in foam cell formation in vascular SMCs. For this project, we isolated aortic SMCs from wild type (WT) and P2Y[subscript]2R‐/‐ mice to investigate whether UTP and the P2Y[subscript]2R modulate expression of LRP 1 and low‐density lipoprotein receptor (LDLR). We also investigated the effects of UTP on uptake of DiI‐labeled agLDL in WT and P2Y[subscript]2R‐/‐ vascular SMCs. For LRP1 expression, cells were stimulated in the presence or absence of 10 [mu]M UTP. To determine LDLR mRNA expression, and for agLDL uptake, cells were transiently transfected for 24 h with cDNA encoding hemagglutinin-­tagged (HA-­tagged) WT P2Y[subscript]2R or a mutant P2Y[subscript]2R that does not bind FLN‐A, and afterwards treated with 10 [mu]M UTP. Total RNA was isolated, reversed transcribed to cDNA, and mRNA relative abundance determined by RT-­PCR using the delta-­delta Ct method with GAPDH as control gene. Results show SMCs expressing the mutant P2Y[subscript]2R that lacks the FLN‐A binding domain exhibit 3‐fold lower LDLR expression than SMCs expressing the WT P2Y[subscript]2R. There was also decrease in LRP1 mRNA expression in response to UTP in P2Y[subscript]2R‐/‐ SMCs compared to WT. Actinomycin‐D (20 [mu]g/ml) significantly reduced UTP-­induced LRP1 mRNA expression in P2Y[subscript]2R‐/‐ SMCs (P < 0.05). Compared to cells transfected with mutant P2Y[subscript]2R, cells transfected with WT P2Y[subscript]2R showed greater agLDL uptake in both WT VSMC and P2Y[subscript]2R-­/-­ cells. Together these results show that both LRP 1 and LDLR expressions are dependent on an intact P2Y[subscript]2R, and P2Y[subscript]2R/ FLN‐ A interaction is necessary for agLDL uptake.

Les r??sultats num??riques confirment les ph??nom??nes constat??s exp??rimentalement.//Abstract: To probate the technology of CO[indice inf??rieur 2] geological storage, the integrity of the site must be assure over time. This industruial problematic involves the study of the mechanical properties alteration of geomaterials in the presence of CO[indice inf??rieur 2]. The scenario at the origin of this thesis illustrates the possibility of a CO[indice inf??rieur 2] leakage on a fault located in the caprock. This geological problem is complicated by the many parameters to consider: in situ temperature and pressure, scale effect, heterogeneities of the geomaterial, geofluide composition, chemical reactions ... These works focus on the behavior of a discontinuity in a geomaterial solicited chemomechanically at the laboratory scale. They were realised in cotutelle between SIAME laboratory at the University of Pau and Pays de l'Adour (France) and the laboratory of rock mechanics and engineering geology from the University of Sherbrooke (Quebec, Canada). The first part of the experimental program was defined to characterize the reclosing of a crack under cyclic uniaxial stress. The second experimental campaign has studied the shear behavior of a rock joint chemically degraded. The data obtained were used to model the behavior of a discontinuity by the finite element method. The mechanical behavior of a crack under normal stress is assessed with cyclic tension-compression tests. Stress curve showed hysteresis during opening and closing cycles of a discontinuity in concrete, it indicated inelastic deformations The analysis of displacement field by image correlation indicated that theses deformations were partially governed by the friction generated during the closing of the discontinuity lips. Frictional phenomena are due to asperities mismatching induced by the internal stresses in the concrete. The shear behavior of a rock joint chemically damaged was studied through direct shear tests. Rough surfaces were immersed in acid solution during 6 hours at constant pH. Digitalization of these surfaces befor and after immesion, with a lase profilometer, indicates little modifications of the geometry induced by dissolution of material. Results of tests pointed out significant modifications for altered joints illustred by a of the peak shear strength and an increased of contractancy. They are induced by: 1) the mismatch enhancement of the rough profiles of the discontinuity and, 2) the degradation of the mechanical properties of the material on both sides of the discontinuity due to the chemical attack. Numerical contribution of the thesis lies in modeling the behavior of a discontinuity by the coupling of an continuous elastic-plastic damaged model with a discrete resolution of the contact/friction problem. The model is developed with the finite element code Cast3M. Geometries lips discontinuities are modeled directly from the roughness profiles from experimental scans. The numerical results correctly represent the friction phenomena observed experimentally. Finally, a model of the shear test altered joints is performed by coupling the mechanical model with chemical damage model.
