O carcinoma espinocelular de cabeça e pescoço (CECP) é um problema de saúde pública que apresenta alta taxa de mortalidade, frequentemente relacionado à presença de recorrências locais e metástases. A descoberta de um pequeno subconjunto de células tumorais com características semelhantes às células-tronco, conhecidas como células-tronco tumorais (CTTs), tem sido relatadas como as principais responsáveis pelo início, progressão e recidiva do CECP. O processo de metástase nestas neoplasias é bastante complexo e envolve o desprendimento de células epiteliais tumorais do local de aparecimento primário devido à subexpressão ou superexpressão de algumas proteínas específicas nestas células, caracterizando um processo conhecido como transição epitélio-mesenquimal (EMT). A compreensão dos mecanismos envolvidos no processo de EMT têm sido investigados para o desenvolvimento de terapias específicas. O ácido retinoico (AR) vem sendo empregado em diversas terapias devido a sua capacidade de controlar a proliferação e promover diferenciação celular, entretanto, anormalidades na expressão ou função de seus receptores são relatadas em muitos tipos de células do câncer. Este estudo tem por objetivo correlacionar a expressão de marcadores do processo de EMT, marcador de célula tronco tumoral (ALDH1) e receptores do ácido retinoico e de retinoide X (isoformas α e β) em amostras teciduais provenientes de portadores de CECP primários, além, de correlacionar os resultados obtidos com os parâmetros clínicos, características histopatológicas e prognóstico destes pacientes em um período de acompanhamento de 7 anos. / The head and neck squamous cell carcinoma (HNSCC) is a public health problem that presents high mortality rates in relation to the presence of local recurrences and metastases. A finding of a small subset of tumor cells with stem like-cells characteristics, known as cancer stem cells (CTTs), has been reported as being primarily responsible for the onset, progression and recurrence of CECP. The metastasis process in these neoplasms is quite complex and involves the tumor epithelial cells detachment from the primary site of appearance due to underexpression or overexpression of some specific proteins in these cells, characterizing the epithelial-mesenchymal transition (EMT) process. An understanding of the mechanisms involved in the EMT process has been investigated for the development of specific therapies. Retinoic acid (AR) has been used in several therapies because its ability to control the proliferation and promote cell differentiation, however, abnormalities in the expression and function of its receptors are reported in many types of cancer cells. The aim of this study was to correlate the expression EMT process markers, tumor stem cell marker (ALDH1), retinoic acid and retinoic acid X receptors (α and β isoforms) in tissue samples from primary CECP, in addition, to correlate the results with the clinical parameters, histopathological and prognostic characteristics of these patients in a 7 years follow-up.

On The Side of the Forest Owner: : A Discourse Analysis of the Swedish Reception of the EU ForestStrategy for 2030

The new EU Forest Strategy for 2030, one of the flagship initiatives of the European Green Deal, has put EU forest policy in a new expansionary phase with ambitious goals for future forest management in Member States. This thesis analyzes contestation over the EU Forest Strategy for 2030 in Sweden to understand how discursive representations of forests are interlinked with justice perceptions. By utilizing poststructuralist discourse theory and the logics framework, and analyzing EU forest policy documents, Swedish parliamentary debates, and interviews with Swedish forest stakeholders, this thesis shows how sustainable forest management functions as a floating signifier and how different discursive understandings of this concept leads to different understandings of justice in forest management and thus how a just transition in forestry should look like. In the Swedish reception of the new Strategy, a commodity discourse and a conservation discourse could be identified where both highlighted the importance of the private forest owner in relation to justice perceptions in Sweden. The results show how the discourses attempt to invest the role of private forest owners with different meanings. Thus, this thesis contributes to the literature on just transitions by showing how justice perceptions are discursively interlinked with understandings of the role of forests, and how political contestation over this takes place.

Static and Dynamic Aspects of the Roman Family Abstract: The thesis deals with Roman family law, the core of the research being the Roman family in the Republican era and the beginning of the Principate. However, the archaic rules of regal period on one hand, and of the period of Dominate or even of the times of Justinian on the other, can also be included to present the overall picture of the development of a certain institution. Three main research goals are outlined in the introduction of the work to be reached throughout the following three chapters. The first goal is to define the term of the Roman family; the second is to connect theory and practice, i.e. so-called law in books and law in action. Therefore, we shall first analyze the legal rules and then compare these theoretical findings with their practical application. The practice shall be ascertained mainly from the non-legal sources of literal or epigraphic character. The third goal is to utilize an interdisciplinary approach, i.e. to use the outcomes, methods, and procedures from the research fields other than legal sciences to deepen our knowledge of the Roman family. On the factors affecting the ductile-brittle transition in as-quenched fully and partially martensitic low-carbon steels

Abstract From the largest discontinuities to the smallest of the elements, various factors can threaten structural integrity. Susceptibility to these factors elevates with higher yield strengths. As-quenched low-carbon steels with a martensitic or martensitic-bainitic microstructure are modern ultra-high-strength structural steels. They can possess sufficient toughness, formability, and weldability, and are typically used in weight-critical and high-performance structures. Common problems with as-quenched steels with a yield strength of 900 MPa or more are that they do not obey the conventional correlation between the fracture toughness reference temperature T₀ and the impact toughness transition temperature T₂₈ᴊ used in many standards and structural integrity assessment procedures, and a lack of design rules in general. This thesis studies the relationship between the T₀ and T₂₈ᴊ to provide additional knowledge for future standardisation, the microstructural features governing the toughness at these temperatures on both global and local scale, and whether hydrogen embrittlement is present at subzero temperatures. It uses steels produced via laboratory rolling and quenching as well as from pilot-scale and full-scale industrial production, studying them with standardised toughness tests, microstructural characterisation, fractography, and cohesive zone modelling. As-quenched steels have a distinct correlation between T₀ and T₂₈ᴊ. An improved general T₀ – T₂₈ᴊ correlation applies to a wide range of steels. T₂₈ᴊ correlates closely with a dynamic reference toughness, which can be used together with the fraction of detrimental {100} cleavage planes near the main fracture plain to effectively estimate the transition temperatures. On a local scale, centreline segregation decreases the effective coarse grain size, which more than compensates for the harmful effects associated with the higher hardness and inclusion content of the centreline, resulting in increased fracture toughness. Hydrogen embrittlement causes a decrease in fracture toughness and local deformability, thereby increasing T₀ while leaving T₂₈ᴊ unaffected. Overall, the results show that high toughness demands good control of effective coarse grain size and hydrogen content. / Tiivistelmä Tekijät suurimmista epäjatkuvuuskohdista aina pienimpään alkuaineeseen voivat uhata rakenteellista eheyttä, minkä lisäksi alttius näille kasvaa materiaalin myötölujuuden kasvaessa. Modernit karkaistun tilan ultralujat matalahiiliset rakenneteräkset voivat silti omata riittävän sitkeyden, muovattavuuden ja hitsattavuuden. Tyypillisiä käyttökohteita näille ovat painon suhteen kriittiset ja korkean suorituskyvyn rakenteet. Yleinen ongelma myötölujuudeltaan noin ja yli 900 MPa karkaistun tilan teräksillä on se, että ne eivät noudata perinteistä murtumissitkeyden referenssilämpötilan T₀ ja iskusitkeyden transitiolämpötilan T₂₈ᴊ välistä korrelaatiota, jota käytetään useissa standardeissa ja suunnitteluohjeissa, jotka eivät myöskään vielä salli näin lujien terästen käyttöä. Tämä väitöstyö tutkii transitiolämpötilojen T₀ ja T₂₈ᴊ välistä suhdetta edistääkseen näiden terästen sisällyttämistä standardeihin, haurasmurtuma-sitkeyteen vaikuttavia mikrorakenteellisia tekijöitä sekä yleisellä että paikallisella tasolla, ja vetyhaurautta matalissa lämpötiloissa. Koeteräkset ovat laboratoriovalmisteisia, tuotantokokeita ja tuotantolaatuja. Niitä tutkitaan standardisoiduilla sitkeyskokeilla, mikrorakenteen karakterisoinnilla, fraktografialla ja koheesiovyöhykettä hyödyntävällä mallinnuksella. Tulokset osoittavat karkaistun tilan terästen omaavan erityisen korrelaation T₀ ja T₂₈ᴊ välillä. Muokattu, ultralujat teräkset huomioiva yleinen T₀ – T₂₈ᴊ -korrelaatio soveltuu laajalti eri terästyypeille. T₂₈ᴊ korreloi läheisesti dynaamisen referenssisitkeyden kanssa, jonka avulla yhdessä haitallisten {100} lohkomurtumatasojen osuuden kanssa voidaan estimoida joukko transitiolämpötiloja. Paikallisella tasolla keskilinjasuotauma pienentää efektiivistä karkeiden rakeiden kokoa, mikä suotauman suurista sulkeumista ja kovuudesta huolimatta parantaa murtumissitkeyttä. Vetyhauraus taas huonontaa sitkeyttä ja paikallista muodonmuutoskykyä myös matalissa lämpötiloissa nostaen T₀ lämpötiloja. Kokonaisuutena erinomainen transitiolämpötilasitkeys vaatii efektiivisen karkearaekoon ja vetypitoisuuden minimointia.

Migration and adhesion associated molecules in lymphoma biology and their potential roles as biomarkers

Abstract Lymphomas are a heterogeneous group of malignancies that arise from lymphatic tissues. Diffuse large B-cell lymphoma (DLBCL) is the most common lymphoma sub-type. It is an aggressive malignancy with an increasing incidence. The prognosis of DLBCL has improved significantly, but problems also remain. The clinical significance of central nervous system (CNS) relapses has become increasingly important. As secondary CNSL (sCNSL) and primary CNS lymphoma (PCNSL) are known to have poor prognoses; the prevention of sCNSL is of crucial importance. Peripheral T-cell lymphomas (PTCL) are rare neoplasms and include several lymphoma subtypes that possess complex and also overlapping morphological and immunophenotypic characteristics. The identification of different entities has improved, but the biological knowledge remains scarce when compared to DLBCL. The optimal treatment schemas for PTCLs are still lacking and they have long been treated with the same therapies as B-cell lymphomas, mainly with suboptimal treatment results. The aim of this study was to identify poor prognostic markers in DLBCL and PTCLs and potential biological markers for the prediction of DLBCL CNS relapse. The study material included patients with systemic DLBCL without CNS affision (sDLBCL), sCNSL, PCNSL and PTCLs. The expression of epithelial-mesenchymal transition (EMT) transcription factors (TFs), chemokines and their receptors and adhesion-, migration- and inflammatory responses-associated molecules were studied by means of immunohistochemistry. IEM was used to verify the specific subcellular location of the studied molecules. GEP was performed on 12 PTCL samples in order to compare the poor prognosis group with the good prognosis group and on one sDLBCL and one sCNSL sample from the time of primary diagnosis. The EMT TFs were found to be expressed in both DLBCL and PTCLs, where they ultimately proved to have prognostic relevance as well. In PTCLs, these TFs were able to delineate a disease group with a specific gene-expression profile. CXCR4, CXCR5, ITGA10, PTEN and CD44 were found to be differently expressed between DLBCL cases with CNS affision when compared to those without CNS disease. These molecules seem to play a role in the development of CNS relapse and hopefully, if further verified, will lead towards the identification of biological markers for CNS relapse prediction. / Tiivistelmä Lymfoomat ovat heterogeeninen ryhmä imukudossyöpiä, joista diffuusi suurisoluinen B-solulymfooma (DLBCL) on yleisin alatyyppi. Se on aggressiivinen maligniteetti, jonka insidenssi on noussut viime vuosina. DLBCL potilaiden ennuste on parantunut merkittävästi, mutta yhä osa potilaista menehtyy tautiinsa. DLBCL:n keskushermostorelapsin kliininen merkitys on tänä päivänä aiempaa suurempi. Sekundaarisen keskushermostolymfooman (sCNSL) ja primaarin aivolymfooman (PCNSL) ennusteet ovat nykyhoidoilla huonoja, joten keskushermostorelapsin ennaltaehkäiseminen on tärkeää. Perifeeriset T-solulymfoomat (PTCLs) ovat ryhmä harvinaisia neoplasioita, joka sisältää useita eri alatyyppejä, joiden morfologiset ja immunofenotyyppiset ominaisuudet ovat monimuotoisia ja osin päällekkäisiä. Eri entiteettien indentifiointi on parantunut, mutta PTCL:ien biologinen tietämys on yhä DLBCL:aa heikompaa. PTCL:ien optimaalinen hoito ei ole selvillä ja tätä tautiryhmää on pitkään hoidettu samoilla hoidoilla kuin DLBCL:aa, mutta huonommilla hoitotuloksilla. Tutkimuksen tavoitteena oli löytää huonon ennusteen markkereita, joilla myös pystyttäisiin ennustamaan DLBCL:n keskushermostorelapsia. Aineisto koostui DLBCL, sCNSL, PCNSL ja PTCL näytteistä. Immunohistokemiallisilla värjäyksillä tutkittiin epiteliaalis-mesenkymaalisen transition (EMT) transkriptiotekijöitä (TF), kemokiinireseptoreita sekä adheesioon-, migraatioon ja inflammaatioon assosioituja molekyylejä. Immunoelektronimikroskopialla varmennettiin molekyylien lokalisaatio soluissa. Geeniekspressioprofiloinnilla (GEP) verrattiin kahdentoista hyvän ja huonon ennusteen ryhmään kuuluvan PTCL näytteen välisiä geeniekspressioeroja sekä kahden DLBCL potilaan näytteitä, joista toiselle kehittyi keskushermostorelapsi. EMT TF:ien ekspressiota nähtiin DLBCL ja PTCL näytteissä, joissa niillä myös todettiin olevan ennusteellista merkitystä. PTCL:ssa TF:t pystyivät erottelemaan tautiryhmän, jolla oli oma spesifinen geeniekspressioprofiilinsa. CXCR4, CXCR5, ITGA10, PTEN ja CD44 ekspressio oli erilaista systeemisissä DLBCL tapauksissa verrattuna sCNSL tapauksiin. Edellä mainituilla molekyyleillä näyttää olevan oma roolinsa keskushermostotaudin kehittymisessä ja jos nämä tulokset pystytään vahvistamaan tulevissa tutkimuksissa, johtavat ne toivottavasti kohti keskushermostorelapsiriskin tarkempaa tunnistamista.

Lapsen koulunaloittaminen ekologisena siirtymänä:vanhemmat informantteina lapsen siirtymisessä esiopetuksen kasvuympäristöistä perusopetuksen kasvuympäristöön

Abstract In this study I examine how the parents describe the child's starting school as an ecological transition from pre-school to school growth environments. This concept is based on Bronfenbrenner's ecological theory. In terms of time-frames starting school is understood as a transition process which begins in the pre-school year, continues during the first school year and includes changes in the child's growth environment as well as in the child's role. The data derives from interviews with 21 parents of children from diverse educational contexts and the journals and interviews of parents from an additional three families. In this study, a child's growth environment changed during the transition process from a child-centred, play- and learning environment to a goal-oriented learning and teaching environment directed by the teacher; from a social growth environment to a more individual working environment; and from a preparatory working culture (directed towards school-attendance) to an educative working culture aimed at citizenship. The child's role changed in the process from that of a pre-schooler to a school beginner, to a school child. This study brings in the school beginner's role. The transition process was most flexible for pre-schoolers from the combined class, secure and multi-phased for pre-schoolers from the pre-school and school co-operation unit. For the Montessori child the transition was natural and quick, for pre-schoolers from the day-care unit, it was longest and most problematic. In conclusion, my research suggests that, although our school system aims to be equal for all, this is not the reality during the pre-school and school starting phases. However, by the end of the first school year all the children in this study had adjusted to the school context. They had fulfilled the criteria and expectations set for the role of the school child: such as becoming an academic learner; a performer of tasks, an object of evaluation; a responsible, concerned and well-behaved pupil, and a school companion. In summary, the school working-culture had a powerful homogenizing effect, adjusting the child to the school and to the role of school child. / Tiivistelmä Tutkimuksessa lähestyn lapsen koulunaloittamista ekologisena siirtymänä esiopetuksen kasvuympäristöstä perusopetuksen kasvuympäristöön ja tarkastelen millaiseksi vanhemmat kuvaavat tämän siirtymän. Koulunaloittaminen ekologisena siirtymänä perustuu Bronfenbrennerin ekologiseen teoriaan. Ajallisesti koulunaloittaminen ymmärretään siirtymäprosessina, joka alkaa esiopetusvuotena ja jatkuu ensimmäisen kouluvuoden aikana. Siirtymäprosessi sisältää lapsen kasvuympäristön ja lapsen roolin muutoksen. Erityisesti tarkastelen millaiseksi 21 oululaista vanhempaa kuvaavat kasvuympäristön kulttuurisen muutoksen esikoulun toimintakulttuurista koulun toimintakulttuuriksi ja lapsen roolin muutoksen esikoululaisesta koululaiseksi. Tutkimus on lähestymistavaltaan ekologinen, ja tarkastelun kohteena ovat arkielämän kontekstissa tapahtuvat yksilön ja kasvuympäristön vastavuoroiset vuorovaikutus- ja muutosprosessit. Koulunaloittaminen siirtymäprosessina on lapsen ja kasvuympäristön keskinäinen muutos- ja sopeutumisprosessi. Kasvuympäristön kulttuurisen muutoksen tarkastelu muodostaa tutkimukseni taustan ja lähtökohdan. Roolimuutosta tutkin kunnallisen päiväkodin, esi- ja alkuopetuksen yhteistoimintayksikön, montessoripainotteisen päiväkodin ja esi- ja alkuopetuksen yhdysluokan esiopetuskontekstissa sekä koulukontekstissa. Tutkimusaineisto koostuu 13 perheen vanhempien haastatteluista ja kolmen perheen vanhempien kirjoittamista päiväkirjadokumenteista ja heidän haastatteluistaan. Vanhempien mukaan lapsen kasvuympäristö muuttui esikoulusta kouluun siirryttäessä lapsikeskeisestä, leikki- ja oppimisympäristöstä opettajajohtoiseksi, tavoitteelliseksi oppimis- ja opetusympäristöksi, sosiaalisesta kasvuympäristöstä enemmän yksin työskentely -ympäristöksi ja kouluun valmentavasta toimintakulttuurista kansalaiskasvatuksen toimintakulttuuriksi. Tämän muutoksen rinnalla vastaavasti lapsen rooli muuttui prosessinluonteisesti esikoululaisesta koulunaloittajaksi ja koululaiseksi. Koulunaloittajan roolin löytyminen oli yksi tutkimukseni päätulos. Roolimuutos kaikkinensa oli vähäisin esi- ja alkuopetuksen yhdysluokan esikoululaisilla, jotka olivat harjoitelleet koululaisen roolia koulukontekstissa jo esiopetusvuoden aikana. Suurin ja jyrkin roolimuutos oli kunnallisesta päiväkodista sekä montessoripainotteisesta päiväkodista siirtyneillä esikoululaisilla, joille koulukonteksti ja koululaisen rooli olivat etukäteen vieraita. Yksilöllinen ja vaihteleva roolimuutos oli esi- ja alkuopetuksen yhteistoimintayksiköstä siirtyneillä esikoululaisilla, jotka olivat etukäteen tutustuneet koulun sosiaaliseen ja akateemiseen ympäristöön. Kouluun siirtyminen lapsen ja kasvuympäristön keskinäisenä muutos- ja sopeutumisprosessina oli joustavin, helpoin ja yksilöllisin esi- ja alkuopetuksen yhdysluokan esikoululaisilla, turvallinen ja monivaiheinen esi- ja alkuopetuksen yhteistoimintayksikön esikoululaisilla, luonteva ja nopea montessorilapsella ja pitkäkestoisin ja ongelmallisin päiväkodin esikoululaisilla. Tulosten johtopäätöksenä voidaan todeta, että vaikka koulutusjärjestelmämme pyrkii olemaan kaikille yhdenvertainen, se ei ole sitä esiopetus- ja koulunaloitusvaiheessa. Kuitenkin ensimmäisen kouluvuoden päättyessä kaikki tutkimuksen lapset olivat sopeutuneet koulukontekstiin ja täyttivät koululaisen roolille asetetut kriteerit ja odotukset akateemisena oppijana, tehtävien suorittajana, arvioitavana, vastuullisena, huolehtivana ja hyvin käyttäytyvänä koulunkävijänä ja koulukaverina. Koulun toimintakulttuurilla oli vahva sopeuttava ja yhdenmukaistava merkitys lapsen kouluun sopeutumisessa ja koululaiseksi kasvussa.
