DEVELOPMENT OF EARLY POSTMORTEM TUMBLING METHODS TO IMPROVE TENDERNESS AND PROTEOLYSIS OF FRESH BEEF LOINS

<p>Historically, the meat industry has struggled to provide consumers with consistent beef tenderness. Various post-harvest technologies have been used in industry; however, there is still a need to develop a natural and safe post-harvest processing system that can be used to create consistently tender products for consumers. In addition to postmortem aging being a time-consuming process, literature has suggested that it is not a sufficient method to achieve tenderization in certain cull cow muscles. This has resulted in the large supply of cull cow beef to be underutilized due to its inferior quality, specifically tenderness. Applying a combination of mechanical tenderization with additional postmortem aging may be an effective strategy to overcome deficiencies in beef tenderness. Recent studies have found that tumbling without brine addition can be successful at improving instrumental tenderness and consumer liking of tenderness of fresh beef loin. The physical disruptions of muscles, which likely occur during tumbling, may enhance activity of proteolytic enzymes and thus induce more tenderization. The overall objective of this thesis was to investigate the effects of fresh beef tumbling methods and postmortem aging times on the tenderness and proteolysis of loin muscles from both A maturity cattle and cull cows.</p> <p>The first chapter of this thesis is a literature review that will address the factors affecting tenderness and the methods used by the industry to improve tenderness, specifically focusing on meat tumbling and cull cow beef. The second chapter is a study that investigated the effects of fresh beef tumbling at different postmortem times on meat quality attributes and proteolytic features of loins. The results from this study suggest that early postmortem tumbling coupled with aging can synergistically impact the improvements of beef loin tenderness and proteolysis, shortening the necessary aging period. The third and final chapter of this thesis is a study that aimed to determine the effect of fresh beef tumbling and postmortem aging on the quality and proteolysis of loins from cull cows. The results from this study indicate that aging would be effective at improving the quality and palatability of cull cow beef loins, although tumbling could improve consumer liking of tenderness at earlier postmortem times.</p>

Combined Tumbling and Postmortem Aging to Improve Fresh Beef Quality, Palatability, and Proteolysis

<p>  </p> <p>Tenderness is a key sensory trait influencing beef palatability. Tumbling is a value-adding process that has been extensively applied and studied within the realm of processed meats. Various post-harvest strategies to ensure fresh beef reaches acceptable levels of tenderness have been employed, often with the aim of physically disrupting myofibrillar structure or enhancing the rate and extent of postmortem proteolysis. One such method would be the application of postmortem aging; however, the effectiveness of aging on tenderization is well-known to differ throughout individual muscles of the beef carcass. For inherently tough cuts, physical interventions such as mechanical tenderization are often used, although several detriments to quality attributes may be induced. Further, some modern consumers prefer meat products with no added non-meat ingredients. An alternative method of applying tumbling in the absence of a brine solution followed by additional postmortem aging could be a practical means to facilitate tenderization while potentially minimizing detriments to other eating quality attributes.</p> <p>To evaluate the efficacy of tumbling without brine a method of beef tenderization, the process was first assessed in the <em>longissimus lumborum </em>muscle (n=9). In this study, muscles were allocated among 0, 60, and 90 minutes of tumbling, after which aging for 0, 7, and 14 days was conducted. Immediately after the application of the tumbling process, steaks from muscles that had been tumbled were considerably more tender (24.7 N and 21.6 N for 60 and 90 minutes, respectively) than non-tumbled controls (34.8 N). Steaks from the tumbled groups maintained greater instrumental tenderness throughout the course of the aging period. These results were supported by increases in myofibril fragmentation index, as well as increased troponin-T degradation during aging. However, cooking loss was increased in tumbled steaks, which could have implications for sensory juiciness. Considering this study demonstrated that tumbling without brine inclusion followed by postmortem aging resulted in profound changes to sensory traits, further study regarding its impacts on sensory attributes and proteolysis among different beef muscles was warranted.</p> <p>The following study evaluated the combined tumbling and aging process on the quality, palatability, and proteolytic attributes of beef <em>longissimus lumborum </em>and <em>semitendinosus </em>muscles (n=16). Muscle sections were allocated among 0, 40, 80, and 120 minutes of tumbling, as well as 0 or 10 days of subsequent aging. Regardless of aging duration, tumbling for any duration increased instrumental tenderness of the <em>longissimus lumborum</em> but not <em>semitendinosus</em> muscle. Similar to the previous study, increased cooking loss was induced through tumbling. In both muscles, obvious fragmentation of the myofibrillar structure with tumbling was observed through increases in myofibril fragmentation index and transmission electron microscopy. Tumbling with aging favored the degradation of myofibrillar proteins including troponin-T and desmin; however, calpain-1 autolysis appeared mostly unchanged. Neither tumbling nor aging influenced the amount and properties of collagen, which may indicate why the process did not influence instrumental tenderness of the <em>semitendinosus </em>despite myofibrillar fragmentation and degradation. <em>Longissimus lumborum </em>muscles tumbled for any durations were rated by consumers (n=120) to be more tender with greater overall liking than control steaks. <em>Semitendinosus </em>steaks that were tumbled for 120 minutes and further aged had improved liking of tenderness with similar juiciness and flavor to control steaks at the same postmortem timepoint. These results indicated that tumbling without brine would result in myofibrillar fragmentation and favor the degradation of myofibrillar proteins during aging, while impacts on connective tissues would be minimal. Consequently, muscles without a high extent of background toughness would be effectively tenderized through tumbling, while the results would be more limited in inherently tough cuts.</p> <p>Considering these results, the process was then applied to muscles of intermediate tenderness from the sirloin, specifically the <em>gluteus medius, biceps femoris, </em>and <em>tensor fasciae latae </em>muscles (n=16). Muscles were tumbled for 0 or 120 minutes, then aged for 0 or 10 additional days. Tumbling increased the instrumental tenderness of the <em>gluteus medius </em>and <em>tensor fasciae latae </em>but not the <em>biceps femoris</em>, regardless of aging time. Cooking loss was increased with tumbling in all muscles. Similarly, myofibrillar fragmentation was also increased in all muscles, and there was some evidence to suggest that tumbling with subsequent aging would aid in the degradation of troponin-T in the <em>biceps femoris</em>. To further understand how tumbling might affect specific descriptive sensory attributes, a trained panel (n=8) was conducted on aged samples. Tumbled <em>gluteus medius </em>steaks had greater myofibrillar tenderness than non-tumbled controls; however, tenderness scores of other muscles were not affected. There was some evidence that tumbling with aging could induce the generation of off-flavors in the <em>gluteus medius </em>and <em>tensor fasciae latae</em>, as well as decrease juiciness of the <em>biceps femoris</em>.</p> <p>Taken together, these results support that tumbling without brine inclusion would be an effective strategy to improve beef tenderness and palatability, dependent on the traits of the individual cut. Improved tenderness would be primarily attributed to the fragmentation and degradation of myofibrillar structure. However, the results indicate that tenderization would be limited in cuts with a high extent of background toughness, which tumbling alone would be largely unable to disrupt. Future studies should focus on the effects of tumbling without brine inclusion with aging on oxidative stability and the potential introduction of hazards prior to industry application. Further elucidation of how the process could be optimized to maximize tenderization while minimizing potential negative impacts to flavor and juiciness would be beneficial to improving overall palatability.</p>

Den ökade efterfrågan på pellets har fört med sig högre krav på pelletkvalité och detta har lett till att forskningen i Sverige har övergått från att studera möjliga bränslen till att studera pelletkvalité, ofta med fokus på mekanisk hållfasthet. För att pröva den mekaniska hållfastheten så finns olika standardmetoder såsom tumling enligt ISO 17831-1:2015, som är europeisk standard, och lignotest. Standardmetoderna kräver en större mängd pellets och kan inte användas för att pröva pellets från en singelpelletpress, vilket är ett mycket vanligt sätt att producera nya pelletsammansättningar. En singelpelletpress tillverkar en pellet åt gången och för att prediktera mekaniska hållfastheten finns många olika metoder, till exempel dropp- och hårdhetstest. Det är idag oklart hur väl metoderna för att pröva pellets från en singelpelletpress predikterar mekaniska hållfastheten och det finns en begränsad mängd forskning som undersöker korrelationen med standardmetoderna. Äldre forskning visar på en positiv linjär korrelation medan nyare forskning visar att korrelationen även beror på pelletens sammansättning. Det finns också studier som använder nya sätt att pröva enstaka pellets. En ny studie tumlar pellets tillsammans med plastpellets, men inga jämförelser med andra metoder utförs. En annan studie prövade tumling samt lignotest för en pellet åt gången och jämförde med tumling enligt ISO 17831-1:2015. Den studien visade ett R2 på 0.83 respektive 0.94 för pellets med en mekanisk hållfasthet över 90%.  En metod som korrelerar med standardmetoderna skulle mycket tydligare indikera hur en pellet från en singelpelletpress skulle prestera i större produktionsskala. Om metoden även utnyttjar redan tillgänglig teknik finns möjligheten för en bred användning, likt en standardmetod, som skulle underlätta jämförelser mellan olika studier och samarbeten mellan forskningsinstitut.  I detta arbete undersöks validiteten av att tumla enstaka pellets tillsammans med plastpellets. Plast- och singelpelletsmixen som innehöll 3, 10 och 20 pellets, resterande del (upp till 500g ±10g) plastpellets, tumlades för 6 olika sorters pellets. Standardavvikelse, relativ standardavvikelse samt medelvärdet av mekaniska hållfastheten bestämdes och den linjära regressionen med tumling enligt ISO 17831-1:2015 undersöktes. För samma pelletsorter genomfördes dropptest och hårdhetstest där den linjära regressionen med tumling enligt ISO 17831-1:2015 även gjordes för hårdhetstestet. Resultaten visar att tumling med plast- och singelpelletmix fungerar som alternativ metod till ISO 17831-1:2015 för pellets med en mekanisk hållfasthet över 92,45%, där mycket tillförlitligt resultat fås för 3 eller fler tumlade pellets över 96,62% mekanisk hållfasthet där 8 av 9 uppmätta standardavvikelser var under 1 %. För hela det prövade kvalitetsintervallet visades en korrelation (R2) med standardmetoden på 0,88-0,98 och ett R2 nära 1 för pellets med en mekanisk hållfasthet över 95 %. Dropptestet gav ingen information om pelletens mekaniska hållfasthet och bedömdes inte som en lämplig metod. Hårdhetstestet visade en hög relativ standardavvikelse mellan 19-40 % och en korrelation (R2) med standardmetoden på 0,78. Tumling med plast- och singelpelletmix hade både lägre standardavvikelse och högre korrelation med standardmetoden som visar att det är en bättre metod för att prediktera mekaniska hållfastheten för enstaka pellets än hårdhetstest. / The increased demand of pellets has led to higher demand on pellet quality causing a shift in Swedish research from studying possible fuels to studying pellet quality, often with a focus on mechanical durability. To test the mechanical durability there are various standard methods such as tumbling according to ISO 17831-1:2015, which is the European standard, and lignotest. The standard methods require a large amount of pellets and can´t be used to test pellets from a single-pellets press, a common way to produce new pellet compositions. A single-pellet press produces one pellet at a time and to predict the mechanical durability there are different methods, such as drop and hardness tests. Today it´s unclear how well the methods for testing pellets from a single-pellets press predicts the mechanical durability and there are a limited amount of research that examines the correlation with the standard methods. Earlier research shows a positive linear correlation while a recent publish study showed that the correlation also depends on the composition of the pellet. However, also new ways to test individual pellets has recently bin published, where single pellets were tumbled together with plastic pellets, but no comparisons with other methods were made. Another study tried tumbling and lignotest for one pellet at a time and compared with ISO 17831-1:2015. The study showed an R2 of 0,83 and 0,94 respectively for pellets with a mechanical durability above 90% A method that correlates with the standard methods would indicate how a pellet from a single-pellet press would preform on a larger production scale. If the method also uses already known technology there is the possibility of a broad use, similar to a standard method, wich would facilitate comparisons between different studies and collaborations between research institutes.   In this study, the validity of tumbling individual pellets together with plastic pellets is investigated. The plastic and single-pellet mix containing 3, 10 and 20 pellets, the remaining part (up to 500 ±10g) plastic pellets, was tumbled with 6 different types of pellets. Standard deviation, relative standard deviation and the mean value of the mechanical durability were determined and the linear regression with tumbling according to ISO 17831-1:2015 was examined. For the same pellet varieties, drop and hardness tests where preformed, where the linear regression with ISO 17831-1:2015 was also examined for the hardness test.   The results show that tumbling with plastic and single-pellet mix works as an alternative method to ISO 17831-1:2015 for pellets with a mechanical durability above 92,45 %, where very reliable results was obtained for 3 or more tumbled pellets above 96.62 % mechanical durability where 8 out of 9 standard deviations measured below 1%. For the entire tested quality range, a correlation (R2) was shown with ISO 17831-1:2015 of 0.88-0.98 and an R2 close to 1 for pellets with a mechanical durability over 95 %. The drop test did not provide any information on the mechanical durability of the pellet and was not considered an appropriate method. The hardness test showed a high relative standard deviation between 19-40 % and a correlation (R2) with ISO 17831-1:2015 of 0.78. Tumbling plastic and single-pellet mix had both lower standard deviation and higher correlation with the standard method ISO 17831-1:2015 which shows that it is the superior method for predicting the mechanical durability of individual pellets over hardness test.

Les particules soumises à un écoulement peuvent manifester des orientations préférées et une variété de déformations en fonction de leur géométrie et élasticité et du champ de vitesse de l'écoulement. A l’inverse, les flux peuvent être modifiés lorsque les contraintes des particules sont non négligeables. Cette thèse présente des résultats théoriques et numériques sur cette relation bilatérale en deux parties. La Ière partie commence par une analyse de stabilité et des simulations numériques qui montrent une particule brownienne semi-flexible dans un écoulement élongationnel effectuant un retournement, un phénomène associé aux flux de cisaillement. Le Chap. 2 étend les outils analytiques dédiés aux modèles simples ou aux flux indépendante du temps pour les modèles perle-barre-bond généraux dans les flux aléatoires. En partant des résultats des chapitres précédents, Le Chap. 3 aboutit à l'étude d'un degré de liberté lagrangien inexploré dans un écoulement turbulent : la flexion. Une particule semi-flexible courbe différemment dans les flux aléatoires bidimensionnels et tridimensionnels. La IIème partie concerne la turbulence élastique, un régime chaotique créé dans un écoulement de faibles forces inertielles par l'ajout de polymères élastiques. Le Chap. 4 estime le nombre de degrés de liberté d'une solution de ce régime via la dimension Lyapunov de l'attracteur du modèle Oldroyd-B bidimensionnel, un modèle connu de reproduire la turbulence élastique dans les simulations numériques. Le Chap. 5 pose des questions sur la nécessité d'élasticité pour produire un régime chaotique et conclut qu'une solution de polymère de barres peut créer un régime comparable à la turbulence élastique. / Particles, when subjected into a flow, may display preferred orientations and a variety of deformations depending on their geometry and elasticity and the flow velocity field. Flows can conversely be modified when the particle stresses are sufficiently large. This thesis presents theoretical and numerical results on this two-way relationship between particles and flows in two parts. Part I starts with a stability analysis and numerical simulations that show a simple semiflexible Brownian particle in an extensional flow undergo tumbling, a phenomenon normally associated to shear flows. Chapter 2 extends analytical tools available only for elementary polymer models or for steady flows to general bead-rod-spring models in random flows. By building on the results from the previous chapters, Chap. 3 culminates with the study of an unexplored Lagrangian degree of freedom in a turbulent flow: bending. A semiflexible particle is shown to display different bending behaviours in two- and three-dimensional random flows. This prediction is confirmed via direct numerical simulations of the particle in a turbulent flow. Part II concerns “elastic turbulence", a chaotic regime created in a flow with low inertial forces by the addition of elastic polymers. Chapter 4 provides an estimate for the number of degrees of freedom of a solution of this chaotic system via the Lyapunov dimension of the attractor of the two-dimensional Oldroyd-B model, a model known to reproduce elastic turbulence in numerical simulations. Chapter 5 questions the necessity of elasticity in producing a chaotic regime and concludes that a rodlike polymer solution can create a regime similar to elastic turbulence.

Influence of Post-treatments on Corrosion Properties of 3D printed Metal Components for Bioprocessing Applications

Additive manufacturing (AM) has the potential to overtake the conventional production of pipe systems of stainless steel grade 316L intended for bioprocessing applications by utilising powder bed fusion laser beam (PBF-LB) technologies. However, challenges such as high surface roughness need to be overcome and corrosion properties improved to meet industry standards. This can be done by applying surface post-treatments to the printed parts. Therefore, the first aim of this thesis was to investigate the ability of different post-treatments of PBF-LB 316L samples, including tumbling, electropolishing, chemical milling, Hirtisation®, and DryLyte®, to reduce the surface roughness. One set of samples was left untreated, as-built set, and used as reference. The second aim was to try to correlate the surface roughness with the corrosion properties of the various post-treated samples. Analytical techniques used included needle profilometry, light optical microscopy (LOM), critical pitting temperature (CPT), cyclic potentiodynamic polarisation (CPDP) and immersion in 1M NaCl and 1M NaOH for two different durations for each solution. While some results were expected, others were surprising. Even though tumbled samples resulted in the lowest surface roughness, the results for immersion and CPDP curves tests indicated the lowest corrosion resistance compared to the other post-treatments. On the other hand, electropolishing resulted in the lowest surface roughness reduction among the post-treated samples. However, these samples had comparatively high corrosion resistance in the CPT and CPDP tests. The DryLyte® samples were tested using two DryLyte® medias where one of them resulted in the second lowest surface roughness and showed highest corrosion resistance in all three corrosion tests even though the process parameters are still to be developed. The results for samples post-treated by Hirtisation® and chemical milling were inconsistent and need further investigations. The reference as-built set showed high corrosion resistance in its untreated state, indicating post-treatments having minor impact on its performance from a corrosion perspective. / Additiv tillverkning (AM) har möjlighet att konkurrera ut konventionell produktion av rörsystem i rostfritt stål typ 316L avsett för bioprocessapplikationer genom att använda pulverbäddsinfusionslaserstråleteknik (PBF-LB). För tekniken kvarstår dock utmaningar för att uppfylla branschstandarder då tillverkningen medför material med hög ytråhet och sämre korrosionsegenskaper än traditionella metoder. Detta kan åstadkommas genom olika typer av efterbearbetning av de printade ytorna. Ett syfte med denna studie var att utvärdera hur olika typer av efterbehandling av PBF-LB 316L (trumling, elektropolering, kemisk etsning, Hirtisation® och DryLyte®) påverkar ytråheten. Som referensmaterial användes obearbetade printade ytor. Studiens andra huvudmål var att undersöka möjligheten för korrelation mellan ytråhet efter behandling och korrosionsegenskaperna. Analyser gjordes med hjälp av nålprofilometri, ljusoptisk mikroskopi (LOM), bestämning av kritisk gropfrätningstemperatur (CPT), framtagning av cykliska potentiodynamiska polarisationskurvor (CPDP) samt nedsänkning i 1M NaCl och 1M NaOH under två olika tidsperioder per lösning. Resultaten var delvis väntade och delvis överraskande. Trots att de trumlade proverna resulterade i lägst ytråhet, visade de elektrokemiska studierna (CPDP) i de olika lösningarna på att lägre korrosionsbeständighet jämfört med övriga efterbehandlingar. Å andra sidan resulterade elektropolering resulterade i den lägsta minskningen i ytråhet bland de efterbehandlade proverna. Dessa prover visade dock på en jämförelsevis hög korrosionsbeständighet i både CPT och CPDP testerna. DryLyte®-proverna studerades med två olika DryLyte®-medier, varav en av dem gav näst lägst ytråhet samt visade högst korrosionsbeständighet i samtliga korrosionstester trots att processparametrarna inte ännu är optimerade för denna nya teknik. Resultaten för prover som efterbehandlats med Hirtisation® och kemisk etsning var tvetydiga och behöver studeras vidare. De obehandlade referensproverna indikerade hög korrosionsbeständighet, vilket tyder på att efterbehandlingar hade minimal påverkan ur ett korrosionsperspektiv.

Longitudinal handling characteristics of a tailless gull-wing aircraft

A handling quality investigation was performed on the swept gull-wing configuration. The swept gull-wing configuration is tailless and has a wing with a transition in the sweep and dihedral angle. An example of this type of aircraft is the Exulans. This aircraft is currently under development at the University of Pretoria. The handling quality study was focussed on pitch axis dynamics. The Exulans is a research testbed that will be used to investigate the swept gull-wing configuration and its special controls by means of full-scale flight testing. Variable wing sweep, twisting elevons and winglets will be investigated as means of control. These control devices are configured in such a way as to have minimum impact on the performance of the aircraft. The handling qualities of the swept gull-wing configuration have to be acceptable while using these different control strategies. The study was launched to investigate whether a gull-wing configuration aircraft will have satisfactory handling qualities at CG positions associated with the most favourable aerodynamic performance. There is an aerodynamic performance gain in designing an aircraft so that the CG falls on the so-called `E-point'. The E-point is the centre of pressure for an elliptical circulation distribution. An elliptical circulation distribution is associated with the highest Oswald efficiency for an aircraft. Time domain simulation techniques and frequency domain analysis techniques were used to analyse the handling qualities of the gull-wing configuration. The C-star criterion was used to analyse handling qualities with time domain simulation data as input. Comparative time domain simulations were performed between the Exulans and other aircraft to compare handling qualities. Eigenvalue analysis was used together with the thumbprint criterion to investigate inherent gull-wing airframe dynamics. The Shomber-Gertsen and Military Specification 8785 criteria were also used for the same purpose. The Neal-Smith method was used to investigate the effect of control authority on handling qualities and the effect of a pilot. The Monnich and Dalldorff criterion was used to evaluate gust handling qualities. An analysis chart by Fremaux and Vairo was used to evaluate the tumbling susceptibility of the gull-wing configuration. The pitch handling quality investigation shows sufficient promise that the swept gull-wing configuration will have acceptable handling qualities with the CG placed at positions associated with optimised aerodynamic performance. Analysis showed that the swept gull-wing configuration is potentially prone to tumbling. With low static margins, the configuration should exhibit improved handling qualities in gusty conditions when compared to existing tailless aircraft. It is recommended that a lateral handling quality study be performed before full scale flight testing commences on the Exulans.
