L’angiogenèse, indispensable à la mise en place d’un réseau vasculaire fonctionnel, est au cœur des stratégies thérapeutiques des pathologies ischémiques. L’hypoxie, caractérisant ces tissus ischémiques, est un stimulus majeur de l’angiogenèse, en induisant l’expression de facteurs de croissance tels que le VEGF et de protéines de la matrice extracellulaire endothéliale. Nous avons identifié la protéine ANGPTL4, comme une cible majeure de l’hypoxie et ayant des effets opposés au VEGF sur la perméabilité vasculaire. Le but de cette thèse a consisté en l’analyse du rôle d’ANGPTL4 sur la formation de capillaire et l’organisation des jonctions adhérentes dans un contexte dépendant du VEGF. J’ai démontré que le VEGF stimule la formation d’un dense réseau capillaire 3D alors qu’ANGPTL4 induit la formation de capillaires étroits et peu ramifiés. ANGPTL4 réduit la taille du réseau de capillaire induit par le VEGF en limitant le nombre de bourgeons, de branchements et la largeur des capillaires. ANGPTL4 renforce l’intégrité des capillaires formés en présence de VEGF en préservant des jonctions adhérentes stables. J’ai démontré qu’ANGPTL4 limite les processus de migration 3D et de prolifération induits par le VEGF. L’analyse de la voie de signalisation VEGF/ANGPTL4 a montré une potentialisation par ANGPTL4 de la phosphorylation Y1175 du VEGFR2, impliqué dans l’internalisation de VEGFR2. En conclusion, ce modèle révèle un effet d’ANGPTL4 dépendant du contexte 3D, qui stimule les processus d’angiogenèse en absence de VEGF et qui contrecarre la morphogenèse induite par le VEGF en renforçant l’intégrité des jonctions adhérentes et en régulant la signalisation en aval du VEGFR2. / Angiogenesis, by promoting new functional capillaries, is a main target of therapeutic strategies of ischemic pathologies. Ischemic tissues are characterized by hypoxic environment, which stimulates angiogenesis by inducing expression and secretion of growth factors such as VEGF and by remodeling endothelial extracellular matrix. Our team identified ANGPTL4 as a hypoxia-induced target and characterized its counteracting effect on VEGF-induced vascular permeability. This PhD study therefore aimed to decipher the role of ANGPTL4 on angiogenesis, capillary architecture and adherens junction (VE-cadherin) organization in a VEGF-dependent context. I demonstrated that VEGF induced formation of branched capillaries forming a dense 3D network while ANGPTL4 enhanced the formation of unbranched and tight capillaries. Remarkably, ANGPTL4 reduces VEGF-induced angiogenesis, by limiting branching and widening of the capillaries. Furthermore, ANGPTL4 regulates the local VE-cadherin patterning during the sprouting process by maintaining lateral linear structures and limiting the VEGF-induced formations involved in the migratory capacities. I demonstrated that ANGPTL4 limited VEGF-induced 3D endothelial cell migration and proliferation. Analysis of VEGF/ANGPTL4 signaling pathway pointed out that ANGPTL4 enhanced phosphorylation of Y1175 VEGFR2, known to enhance internalization of VEGFR2. In conclusion, this study modeled the 3D context-dependent effect of ANGPTL4 that stimulates angiogenesis in absence of VEGF whereas it counteracts VEGF-induced endothelial morphogenesis by regulating VEGFR2 trafficking and strengthening adherens junctions.

The context of my dissertation is an art concept of a period short story. It includes a theoretical, as well as a practical, part. The theoretical part concerns two movies and a TV series from world cinematography, as well as an example of a Czech movie; it evaluates and appraises the artistic aspect of each chosen example. The practical part includes my own art proposal for a short story Osudna vejce. I comment on my proposal, using images and providing examples of precedents. I describe the plot and show the sequence of scenes based on the text of the story.

My thesis is treated using descripvive and analytical method. The aim of my thesis is analyzing the investment climate in Vietnam completely. Its benefits lie on the evaluation of the efficiency of the vietnamese investment climate.

Ab initio-Untersuchung von 29 SI- und 17 O-NMR-chemischen Verschiebungen in Zeolithen und dichten SiO 2 -Modifikationen

In der vorliegenden Arbeit sollte untersucht werden, inwieweit man quantenchemische Methoden zur Berechnung von NMR-chemischen Verschiebungen auf Festkoerper anwenden kann. Es wurden die 29Si- und 17O-NMR-chemischen Verschiebungen von Zeolithen und dichten SiO2-Modifikationen berechnet. Dabei wurden zahlreiche Untersuchungen zur Abhaengigkeit der chemischen Verschiebung von der Festkoerperstruktur und von verschiedenen Parametern bei der Berechnung (Basissatz, Methode) durchgefuehrt. Die Qualitaet der Simulationen wurde an experimentellen Daten gemessen. Es konnte gezeigt werden, dass mit einer geeigneten Kombination von Rechenparametern eine Simulation von NMR-Spektren moeglich ist, die das Experimente sehr genau wiedergibt. / The following work studies, how far quantum chemical methods to compute NMR chemical shifts are applicable to solids. The 29Si and 17O NMR spectra of several zeolites and silicates were investigated. Numerous investigations were carried out to examine the dependence of the chemic al shift on the used zeolite structures and computing methods. The quality of the results was judged on experimental data. It could be shown, that a calculation of NMR spectra with a suitable combination of simulation parameters leads to results which reproduce very well the experimental results.

Ab initio free energies of adsorption from anharmonic vibrations

Die Thermodynamik von Adsorptionsvorgängen wurde mittels quantenchemischer Methoden und der statistischen Thermodynamik untersucht. Eine neue rechentechnische Methode wird vorgestellt, die den Mangel an Genauigkeit vorhandener Methoden zur Untersuchung periodischer Systeme, wie z. B. der Dichte-Funktional-Theorie, behebt, und es ermöglicht thermodynamische Funktionen mit chemischer Genauigkeit zu bestimmen. Das im Rahmen dieser Arbeit entwickelte Protokoll besteht aus verschiedenen rechentechnischen Schritten, als da wären, eine Strukturoptimierung in Normalkoordinaten anstatt in kartesischen Koordinaten, eine numerische Berechnung der harmonischen Frequenzen durch Abtasten der Potentialenergieoberfläche entlang der Normalkoordinaten und anschließender anharmonischer Korrektur. Die Normalkoordinatenoptimierung garantiert eine korrekte Relaxation der Struktur mit ausschließlich reellen harmonischen Frequenzen. Die anharmonischen Korrekturen ermöglichen eine gute Beschreibung der Schwingungsstrukturen von Systemen die durch eine besonders Flache PES gekennzeichnet sind. Gleichzeitig, wird durch die Verwendung eines QM:QM-Hybridverfahrens zur Bestimmung des elektronischen Anteils der Adsorptionsenergie eine höhere Genauigkeit in der Bestimmung der Korrelationsenergie im Vergleich zu DFT garantiert. Der elektronische Anteil der Adsorptionsenergie, sowie der Schwingungsthermische Anteil werden abschließend kombiniert um einen genauen Wert der thermodynamischen Funktionen zu erhalten. / The thermodynamic of adsorption is investigated from the vibrational point of view using quantum chemical methods via statistical mechanics. Due to the lack of accuracy of the present available methods for investigating periodic systems, such as plane-wave density functional theory (DFT), a novel computational strategy is presented to overcome these limitations and bring the estimate of the thermodynamic functions within chemical accuracy limits. The protocol presented in this work consists of different computational steps, namely a structure optimization using normal mode coordinates instead of Cartesians, a numerical harmonic frequency calculation via sampling of the potential energy surface along the normal mode coordinates and the inclusion of anharmonic correction to the latter. The normal mode coordinate optimization ensures a proper relaxation of the structure and a reliable set of real harmonic frequencies while the anharmonic corrections account for a proper description of the vibrational structure of a system characterized by a very flat potential energy surface. Parallel to these calculations the electronic part of the adsorption energy is corrected using a hybrid QM:QM scheme to account the electronic correlations effects more accurately than DFT. The hybrid electronic adsorption energy and the vibrational thermal contributions obtained using anharmonic corrections are finally combined to get accurate estimate of the adsorption thermodynamic functions.

Analýza příčin deficitu ve veřejném zdravotním pojištění v České republice / The causes of the deficit in the statutory health insurance system in the Czech Republic

Diplomová práce se zabývá deficitem ve veřejném zdravotním pojištění se základním cílem analyzovat příčiny vzniku deficitu. Úvodní kapitoly vysvětlují teoretická východiska a obecné principy zdravotního pojištění v návaznosti na deficit v systému, dále vývoj a současný stav veřejného zdravotního pojištění v ČR. Deficit je způsoben nesouladem mezi tokem příjmů a nákladů v systému, a proto jsou jednotlivé příčiny v analytické části zkoumány z těchto dvou hledisek. Příjmová strana se zabývá vlivem makroekonomických indikátorů, vlivy na straně pojištěnců, vlivem platby za státní pojištěnce, vlivem pohledávek pojišťoven a vlivy přerozdělení pojistného. Nákladová strana se zabývá jednotlivými faktory, které způsobují růst nákladů na zdravotní péči. V další části práce jsou zkoumány souvislosti mezi migrací pojištěnců, příjmy a náklady systému. Poslední kapitola zkoumá dopady deficitu ve veřejném zdravotním pojištění.

Title: Evaluation of the most frequent injuries in competition of league MOL and the 2nd league in women handbal in Czech republic. Objectives: The main objective in this work is evaluation of the most frequent injuries in women handball players, who play in season 2017/2018 the highest competition in Czech Republic - league MOL and the 3rd highest competition in Czech Republic - the 2nd league in women handball. The next objective is find out, if the teams have in their training unit the regenerative and compensatory practise, also there is physiotherapist in team or no. Methods: As the method for data pooling, I choose the form of a non-standardized questionnaire. The questionnaire was send between all women handball players in both competition, which are written in objective. Overall is analysed 112 questionnaires. Results: From the results, we can see, that for the most common injuries belongs to ankle. Overall 56,3 % suffered the injury of ankle, then injury of hands (50,9 %) and knees (46,4 %). Smaller group of injuries belongs to the of face, head, shoulders and elbows. Wings and lineplayers has the most common injuries of ankle, on the other hand the middle lineplayers, backs and goalkeepers has the most common injuries of hands and knees. The thesis is focused on comparison of several ways of the calculation of thermal and moisture fields in wood containing bound water during drying and their comparison with commonly used standards for wood drying processes. Each of the four models presents different way of the unsteady-state calculation of the coupled thermal and moisture field in wood. The first model describes linear simulation, other three models present nonlinear simulation using material coefficients dependent on temperature and moisture content, differing in the method of the thermodiffusion (Soret effect) coefficient settings. The aim of the thesis is to establish the influence of Soret effect on the wood drying process and to describe the difference between commonly used presumption of this process and its presumption given by the unsteady-state computation of the coupled thermal and moisture fields in wood.

Phosphorylation sur tyrosine de la VE-cadhérine en réponse au VEGF : mécanismes moléculaires et implications physiopathologiques

L'intégrité de l'endothélium vasculaire est dépendante de l'organisation des jonctions adhérentes entre les cellules endothéliales. La VE (Vascular Endothelial)-cadhérine un constituant essentiel de ces jonctions et sa phosphorylation sur tyrosine pourrait être un moyen de régulation de la perméabilité endothéliale. Ce travail de thèse a permis de valider l'existence in vivo de la phosphorylation sur tyrosine de la VE-cadhérine. Nous montrons en effet la très forte induction de cette phosphorylation au cours de l'angiogenèse physiologique dans l'ovaire et l'utérus, par rapport aux tissus quiescents. Pour la première fois, nous rapportons l'association constante de la tyrosine kinase Src à la VE-cadhérine, à la fois in vivo et in vitro. De plus, en accord avec les données de la littérature in vitro, nous confirmons in vivo l'association transitoire du VEGFR2 à la VE-cadhérine dans un contexte angiogénique. Par différentes approches, nous apportons la preuve que la VE-cadhérine est un substrat direct de Src, et que la tyrosine 685 du domaine cytoplasmique est la cible unique de la kinase en réponse au VEGF. Grâce à un test de blessure d'HUVECs, nous montrons l'importance de la phosphorylation par Src de la tyrosine 685 dans la migration des cellules endothéliales induite par le VEGF. Enfin, l'étude de carcinomes mammaires nous as permis de démontrer l'existence d'une forme phosphorylée sur tyrosine de la VE-cadhérine également dans l'angiogenèse tumorale, et son association avec une activité kinase de type Src et PI3K. L'ensemble de ce travail permet de conclure à la phosphorylation par Src de la VEcadhérine sur la Y685 en réponse au VEGF, et suggère que cette phosphorylation représente in vivo la signature d'un endothélium activé.
