A presente dissertação aborda considerações e experimentações sobre a filosofia de Friedrich Nietzsche e a literatura de Franz Kafka. Objetiva revalorizar o corpo como protagonista da existência, especificamente a ressignificação do aspecto fisiológico tão denegrido e subestimado pela tradição metafísica. O trabalho parte da experimentação de diferentes textos dos autores estudados e de comentadores, procurando acompanhar o ato de escrever como expressão da vida e uma forma de expressar imageticamente a superação da condição homem. Através da vitalidade que a escrita pode mostrar (sobre o autor/para o leitor), a dissertação empenha-se em percorrer os caminhos trilhados pela decadência, apatia, sofrimento, ressentimento e pela formação da memória no corpo submetido aos desmandos da razão, para finalmente, apresentar uma alternativa a todo o apequenamento diante da vida. Tal alternativa, a metamorfose, é o projeto de superação das estruturas habituais do homem, que evidencia a insurgência do super-homem nietzschiano (Übermensch), assim como corrobora o protagonismo do corpo e do homem diante de sua própria existência. / The present dissertation broaches considerations and experiments on the Friedrich Nietzsches philosophy and Franz Kafkas literature. Through the vitality that writing can show (about the author/for the reader), this dissertation strives to walk the paths trodden by decadence, apathy, suffering, resentment and the memory formation in the body subjected to the excesses of the reason to finally present an alternative to any diminution towards life. Such an alternative, the metamorphosis, is the project of overcoming the habitual structures of man, which highlights the insurgency Nietzschean overman (Übermensch), and confirms the role of the body and the man in front of his own existence.

Em um trabalho desenvolvido como pesquisa de IC intitulado “Poética e Figuratividade: uma análise de ‘Io’ (Ovídio, Metamorfoses, I, 583-747)”, procurou-se concentrar na primeira etapa dos processos de figuratividade, isto é, na figuração do discurso, quando um tema é revestido por figuras semióticas. Tomando os efeitos de sentido captados pela percepção e apreendidos por meio da leitura como dados de base, pretende-se investigar no corpus o arranjo particular da linguagem. Como resultado dessa investigação produziu-se um discurso metalinguístico a fim de reconhecer os recursos da figuratividade poética determinantes da expressão. Ainda, como base para o desenvolvimento do trabalho, será produzida uma tradução de estudo (literal) acompanhada de notas de referência, com comentários concernentes a dados gerais de cultura (mitologia, história, geografia, filosofia, etc.). / This research is a proposal to investigate the figurative poetics in the Latin text, drawing on the theoretical tools provided by Poetics and Literary Semiotics, with the corpus of the episode "Cinyras and Myrrha", integrating part of the Metamorphoses (Book X, 298-502), by Ovid (43 BC - 17 AD), regarded as one of the greatest poets of Ancient Rome. The Metamorphoses is a long poem in hexameter verses, separated into fifteen books. It depicts the creation of elements of the world and the transmutations of several mythological beings, with a narration that takes place in a continuous form. The selected passage for analysis recounts the transformation of a beautiful young woman into the myrrh tree, after committing incest with her own father, Cinyras, the king of Cyprus. In an already developed undergraduation research entitled “Poetics and Figurativity: an analysis of ‘Io’ (Ovid, Metamorphoses, I, 583-747), we have focused on the first step of the figuration process, that being the figuration of speech, when a subject is covered with semiotic figures. Considering the effects of meaning captured by perception and seized by careful reading as database, we intend to investigate in the corpus particular arrangements of language. The result of this research has produced a metalinguistic discourse to recognize the features of poetical figurativity that are determinants to the expression. Furthermore, as a basis for the development of this work, we will produce a literal study translation, accompanied by background notes, and comments concerning general data culture, such as mythology, history, geography, philosophy, etc.

Transcriptomic analysis of thyroid hormone effects on Rana [Lithobates] catesbeiana tadpole tissues with special emphasis on the innate immune system

Amphibian metamorphosis is facilitated solely by thyroid hormones (THs), L-thyroxine (T4) and 3,5,3’-triiodothyronine (T3). TH modulates the remodeling of many different organs and systems in the body of developing tadpoles, including the immune system. Previous research found evidence of T4 action on direct-response genes in outer ring deiodinase-poor premetamorphic tadpole tail fin and liver without the required conversion to T3 described by current TH dogma. The mechanisms of environmental endocrine disrupting chemicals (EDCs) may be better understood by expanding our understanding of the transcriptomic effects of both forms of THs and how they relate to estrogen signaling. Furthermore, analysis of TH-modulation of the immune system may enable a greater understanding of the devastating effects of amphibian pathogens such as Ranavirus. Premetamorphic Rana (Lithobates) catesbeiana tadpoles were exposed to physiological concentrations of T4, T3, or 17-beta-estradiol (E2) through water bath immersion. qPCR analysis was performed to assess the response of canonical TH-responsive genes thra, thrb, and thibz to these hormones in the liver and tail fin tissues of bullfrog tadpoles. E2 treatment did not elicit a response in these gene transcripts in either tissue. T3 treatment in the tail fin elicited an overall stronger response than T4, while T4 treatment in the liver recapitulated results consistent with non-genomic mechanisms of T4 signaling for thrb and thibz transcripts. Illumina Hiseq2500 was used to sequence RNA isolated from hormone-treated premetamorphic tadpole liver and tail fin tissues to assess differential transcriptomic responses and identify TH-responsive immune system-associated transcripts. The impact of TH-treatment on the general immune system in the liver and tail fin transcriptomes was also analyzed using RNA-seq data. We found that E2 modulates at least some shared TH pathways in the liver, but none in the tail fin and that the tail fin transcriptome is more affected by T3, while the liver transcriptome is more affected by T4. Additionally, evidence of immune system modulation by both THs was found in both the liver and tail fin transcriptomes. Antimicrobial peptides (AMPs) are an important component of the amphibian immune response. Details regarding the regulation, synthesis, and expression of AMPs remain obscure, although evidence of TH-modulation of specific AMPs has been identified, as well as evidence of increased expression of AMPs throughout metamorphosis. Frog skin is a prolific source of AMPs that may prove useful in the quest for alternative antimicrobial agents in the face of antibiotic resistance. Identification of new AMPs is hindered by the practical limitations of classical protein-based discovery approaches. By using known AMP characteristics and common ¬AMP properties, we developed a high throughput bioinformatics approach predicated on the use of R. catesbeiana genome resources. We mined these resources and identified novel and known AMPs that exhibited verified antimicrobial activity against various bacterial organisms. This thesis sought to elucidate the differential and modulatory effects of both forms of TH on a transcriptomic level and in the context of immunity, and to examine the utility of the bullfrog transcriptome and genomics resources in identifying and characterizing novel bullfrog-derived AMPs and elucidating aspects of AMP expression. / Graduate / 2018-12-08

Make Minuscule Monsters

This essay follows closely the process of making the work for my degree project. The aim of the text is to reflect as much as possible the entire journey of my thinking and doing, demonstrating how one of the main concepts of the work, metamorphosis, served the final purpose of the project, to build a world for bizarre creatures made of colorful piles of clothes. The methods used were looking for inspiration material online i.e. a performance by Ingri Fiksdal, several drag performers, or a music video by the band Primus, bringing in Posthuman theory and decolonial thinking as well as readings on the grotesque, including writers like Rosi Braidotti, María Lugones, and Sara Cohen Shabot respectively, taking an excessive amount of notes on diary form, and playing dress-up in the studio. The information that surfaced while all the above was combined, is presented in quite a raw form here, not as one smooth text, but as chunks that mirror where my process was at each moment. More so than anything else, this essay demonstrates the messiness of a/my artistic process and acts as a mind map of interests as they get closer to some kind of crystallization. / <p>This master work includes both a performing and a written part.</p>

Ultrastructural Studies of Spermatogenesis in Anthocerotophyta v. Nuclear Metamorphosis and the Posterior Mitochondrion of Notothylas Orbicularis and Phaeoceros laevis

Ultrastructural observations reveal that the spermatozoids of the hornworts Notothylas and Phaeoceros contain two mitochondria and not one as described previously. Mitochondrial ontogeny and nuclear metamorphosis during spermiogenesis in these plants differ from all other archegoniates. The discovery that the posterior region of the coiled nucleus (when viewed from the anterior aspect) lies to the left of the anterior, in striking contrast to the dextral coiling of the nucleus of spermatozoids of other embryophytes, underlines the isolated nature of the hornworts among land plants. As the blepharoplast develops, the numerous ovoid mitochondria initially present in the nascent spermatid fuse to form a single elongated organelle which is positioned subjacent to the MLS and extends down between the nucleus and plastid. At the onset of nuclear metamorphosis, the solitary mitochondrion has separated into a larger anterior mitochondrion (AM) associated with the MLS and a much smaller posterior mitochondrion (PM) adjacent to the plastid. The PM retains its association with the plastid and both organelles migrate around the periphery of the cell as the spline MTs elongate. By contrast, in moss spermatids, where mitochondria undergo similar fusion and division, the AM is approximately the same size as the PM and the latter is never associated with the spline. As in other archegoniates, except mosses, spline elongation precedes nuclear metamorphosis in hornworts. Irregular strands of condensed chromatin compact basipetally to produce an elongated cylindrical nucleus which is narrower in its mid-region. During this process excess nucleoplasm moves rearward. It eventually overarches the inner surface of the plastid and entirely covers the PM.

Notopleural Mutations Enhance Defects In Imaginal Disc Epithelial Morphogenesis And Macrochete Elongation Associated With Mutations in the Stubble-Stubbloid Locus

The Stubble-stubbloid locus encodes a transmembrane serine protease (Stubble) necessary for the proper formation of sensory bristles, and the morphogenesis of leg and wing epithelia. Genetic and cell biological analysis indicate a role for Stubble in actin cytoskeletal dynamics and cell shape changes in developing epithelia and bristles. Previously reported genetic interactions between Stubble and the Rho1 signaling pathway suggest Stubble influences actin cytoskeleton dynamics in developing imaginal discs through interactions with the Rho1 pathway. This work will discuss a genetic screen conducted to further investigate the role of Stubble in bristle and imaginal disc morphogenesis. From 50,000 EMS-mutagenized chromosomes 12 enhancers of the recessive sbd201 allele were identified, including 6 new sbd alleles. Consistent with the current understanding of genetic interactions regulating imaginal disc morphogenesis, mutations in two Rho1 pathway genes, zipper (2 alleles) and Rho1, were isolated. Additionally, three new mutant enhancers of sbd201 were isolated, one of which has been identified as an allele of the cadherin gene Dacshous, another as an allele of the muscle myosin heavy chain gene, and the last as an allele of Notopleural (Np). Dominant and recessive mutations in the Stubble locus interact with the Np allele identified in this screen, in regards to both limb and bristle development, respectively. Mutations in the Np locus were first identified in 1936, but this locus remains poorly characterized and has never been cloned The genetic and phenotypic characterization of Np will be discussed along with experiments that have mapped the position of the Np locus to a 50kb region at the border of the 44F12, 45A1 cytological regions.

Oxygen uptake and thyroid activity during amphibian metamorphosis

Oxygen consumption can most readily be related to wet weight. However, the weight of a tadpole increases so long as it is feeding, then decreases during final non-feeding stages. Oxygen uptake may also be related to dry weight, but dry weight increases significantly from early tadpole stages to adult (Funkhouser and Mills, in press). There is change in one factor at the same time the change in the the other factor is in the opposite direction. Total nitrogen content may be used as an index of the metabolizing tissue, but is subject to the same criticism as dry weight (Moore, 1964). Ideally, the animal's respiration should be related to as many criteria as possible. The pattern of thyroid activity during amphibian metamorphosis has been described by a number of investigators using a variety of methods. Histological criteria for the functional activity of the thyroid glad include epithelial cell height, the number of epithelial cells in serial sections of gland, "vacuolization" of the colloid, the number of secretion droplets present in the follicle, the number of mitochondria, and changes in the size of the Golgi apparatus. Not all of these criteria are considered to be of equal validity (Etkin, 1930; Gorbman and Bern, 1962). The amount of iodine accumulate in the follicle has been used as an index of the activity of the gland. Kaye (1961) using radioactive iodine (I131) and radioautography found a 12% increase in I131 accumulation during early metamorphic stages, and a 43% increase during or near the later stages of metamorphosis. The rate of development is significantly increased in normal tadpoles immersed or injected with thyroxine. Kollros (1961) was able to reproduce the normal metamorphic pattern in thyroidectomized Rana pipiens with gradually increasing concentrations of T4. All of these studies indicate that during normal metamorphosis there is a gradual increase in the concentration of thyroid hormone reaching a climax in the last stages and dropping metamorphosis has been assumed to increase because the increase in thyroxine levels accompanying metamorphosis in amphibia would be sufficient to cause a marked increase in metabolic rate in mammals. The latent period between exposure to thyroxine and maximum calorigenic effect has been described for many animals, In man this period is about two weeks (Guyton, 1964). The present study was designed to show that the small increase in oxygen uptake observed in a number of amphibians during metamorphic climax (Table II) is the result of the increase in thyroxine levels in the immediately preceding stages.
