At this temperature the first wax crystals are formed by the cooling systems paraffin / solvents. Paraffins are composed of a mixture of saturated hydrocarbons of high molecular weight. The removal of petroleum from wells and the production lines means a surcharge on produced oil, thus solubilize these deposits formed due to modifications of thermodynamics has been a constant challenge for companies of oil exploration. This study combines the paraffin solubilization by microemulsion systems, the determination of WAT systems paraffin / solvent and performance of surfactant in reducing the crystallization. We used the methods: rheological and the photoelectric signal, validating the latter which was developed to optimize the data obtained due to sensitivity of the equipment used. Methods developed for description of wax precipitation are often in poor agreement with the experimental data, they tend to underestimate the amount of wax at temperatures below the turbidity point. The Won method and the Ideal solution method were applied to the WAT data obtained in solvent systems, best represented by the second interaction of Won method using the solvents naphtha, hexane and LCO. It was observed that the results obtained by WAT photoelectric signal when compared with the viscosity occur in advance, demonstrating the greatest sensitivity of the method developed. The ionic surfactant reduced the viscosity of the solvent systems as it acted modifying the crystalline structure and, consequently, the pour point. The curves show that the WAT experimental data is, in general, closer to the modeling performed by the method of Won than to the one performed by the ideal solution method, because this method underestimates the curve predicting the onset of paraffin hydrocarbons crystallization temperature. This occurs because the actual temperature measured was the crystallization temperature and the method proposes the fusion temperature measurement. / A TIAC ? a temperatura de in?cio do aparecimento dos cristais de parafina. Nesta temperatura os primeiros cristais de parafina s?o formados, pelo resfriamento de sistemas parafina/solventes. As parafinas s?o compostas de uma mistura de hidrocarbonetos saturados de alto peso molecular. A remo??o de parafinas dos po?os e das linhas de produ??o significa um custo adicional ao petr?leo produzido, portanto solubilizar estes dep?sitos formados decorrentes das modifica??es termodin?micas tem sido um desafio constante das empresas exploradoras de petr?leo. Foram aplicados os m?todos de Won e o da solu??o ideal aos dados de TIAC da parafina obtidos em sistemas com solventes, sendo melhor representado pela segunda intera??o do m?todo de Won com os solventes: nafta, hexano e LCO. Foi observado que os resultados de TIAC obtidos pelo sinal fotoel?trico quando comparados com a viscosidade, ocorrem antecipadamente, demonstrando a maior sensibilidade do m?todo desenvolvido. O tensoativo i?nico reduziu a viscosidade dos sistemas paraf?nicos, pois ele atuou modificando a estrutura cristalina, conseq?entemente o ponto de fluidez. Estudo sequencial do perfil de expressão gênica em biópsias endomiocárdicas parafinadas: associação com rejeição humoral e vasculopatia do aloenxerto cardíaco / Sequential study of gene expression profiles in paraffin embedded endomyocardial biopsies: association with humoral rejection and cardiac allograft vasculopathy

O transplante cardíaco é a última opção terapêutica para pacientes portadores de insuficiência cardíaca grave. Apesar dos avanços na terapia imunossupressora, a rejeição continua sendo o principal obstáculo para o sucesso do transplante. No presente estudo, propõe-se avaliar o perfil de expressão gênica no tecido cardíaco. Com isso, espera-se contribuir para o melhor entendimento do processo de rejeição a nível molecular. Foram analisadas as amostras sequenciais (1, 3 e 6 meses, 1 e 2 anos pós-transplante) de biópsias endomiocárdicas em parafina de 63 indivíduos transplantados cardíacos. O diagnóstico de rejeição humoral foi realizado pela detecção de C3d e C4d do complemento na reação de imuno-histoquímica, e as expressões dos genes protetores (ADIPOR1, ADIPOR2, HMOXO-1, BCL2L1 e VEGF) e genes associados à inflamação (IL-6, TNF?, IFN?, TGF-?, AIF-1, NOS2, ICAM, VCAM e MCP-1); foram avaliadas pela reação em cadeia da polimerase quantitativa em tempo real (qPCR). As frequências de indivíduos positivos para C4d (28,6%) e vasculite (20,0%) foram significantemente maiores em relação ao teste de reatividade de anticorpos realizado nos receptores antes do transplante (6,3%). Houve mudança no perfil de expressão gênica no tecido cardíaco após o transplante, com aumento da expressão dos genes inflamatórios (AIF-1, TNF?, IL-6, NOS2 e VCAM) e diminuição da expressão dos genes protetores (ADIPOR1, ADIPOR2, BCL2L1 e VEGF). Além disso, as expressões dos genes ADIPOR1 e ADIPOR2 foram significantemente menores nos indivíduos positivos para C4d (p<0,001) e gene VEGF (p<0,001) no grupo com vasculite. Houve também uma correlação positiva entre a expressão do gene VEGF e ADIPOR1 (r=0,5688) e ADIPOR2 (r=0,5191). Por outro lado, a expressão aumentada do gene VCAM (p<0,001) foi detectada em todos os tipos de rejeição. Conclui-se que, depois do transplante, o sistema imune do receptor passou a reconhecer os antígenos do órgão transplantado. Com isso, ocorreu uma mudança no perfil de expressão gênica no enxerto cardíaco, caracterizada pela expressão aumentada dos genes inflamatórios e diminuição dos genes protetores. A expressão aumentada do gene VCAM associada à baixa expressão dos genes protetores: ADIPOR1, ADIPOR2, BCL2L1 e VEGF resultaram em maior gravidade da rejeição celular, humoral e vasculopatia do aloenxerto cardíaco. / Heart transplantation is the ultimate treatment for patients with severe heart failure. Despite advances in immunosuppressive therapy, rejection still remains the main obstacle to successful transplant. The purpose of this study is to evaluate the gene expression profile in cardiac tissue. With this we hope to contribute to a better understanding of the rejection process at the molecular level. Sequential samples (1, 3, and 6 months, 1 to 2 years post-transplant) endomyocardial biopsies paraffin of 63 heart transplant subjects were analyzed. The diagnosis of humoral rejection was performed by detection of C3d and C4d complement in immunohistochemistry and the expression of protective genes (ADIPOR1, ADIPOR2, HMOXO-1, and VEGF BCL2L1) and genes associated with inflammation (IL-6,TNF?, IFN?, TGF-?, AIF-1, NOS2, ICAM, VCAM and MCP-1); were evaluated by quantitative real-time polymerase chain reaction (qPCR). The frequency of individuals positive for C4d (28.6%) and vasculitis (20.0%) were significantly higher compared to antibodies reactivity test conducted in the recipients before transplantation (6.3%). There was a change in the gene expression profile in cardiac tissue after transplantation, with increased expression of inflammatory genes (AIF-1,TNF?, IL-6, NOS2, and VCAM) and a decreased expression of protective genes (ADIPOR1, ADIPOR2, BCL2L1, and VEGF). Furthermore, the expressions of ADIPOR1 and ADIPOR2 genes were significantly lower in C4d positive individuals (p<0,001), and VEGF (p<0,001) in the group with vasculitis. There was also a positive correlation between VEGF expression and ADIPOR1 (r=0.5688) and DIPOR2 (r=0.5191). Moreover, increased expression of VCAM (p<0,001) was detected in all types of rejection. We conclude that, after transplantation, the recipient\'s immune system began to recognize the antigens of the transplanted organ. Thus, a change in gene expression profile in cardiac graft is characterized by increased inflammatory genes and decreased expression of protective genes. Increased VCAM gene associated with lower expression of protective genes expression: ADIPOR1, ADIPOR2, BCL2L1 and VEGF resulted in increased severity of cellular and humoral rejection and cardiac allograft vasculopathy.

Punção ecoendoscópica de massas sólidas pancreáticas por técnica de pressão negativa versus capilaridade: estudo prospectivo e randomizado / Negative pressure versus capillarity for EUS-FNA of pancreatic solid mass: a prospective and randomized trial

INTRODUÇÃO: A punção aspirativa por agulha fina guiada por ecoendoscopia (PAAF-EE) é método consagrado para o diagnóstico de lesões sólidas de pâncreas. A aplicação de pressão negativa (PN) durante a PAAF-EE tem objetivo de manter o tecido junto à ponta cortante da agulha durante sua passagem através da lesão, resultando no desprendimento e retenção de células. A capilaridade (CP) é outra técnica que utiliza a retirada gradual do estilete durante a PAAF-EE e parece ser igualmente eficaz, além de evitar aspiração indesejada de hemácias e coágulos. OBJETIVO: Analisar o rendimento da ecopunção de lesões sólidas pancreáticas utilizando PN versus (vs) CP e os aspectos citológicos qualitativos e semiquantitativos da celularidade e contaminação por sangue dos grupos estudados. MÉTODO: No estudo, foram randomizados 50 pacientes com lesões sólidas de pâncreas para realização de PAAF-EE. Cada paciente foi submetido a quatro punções, intercalando as duas técnicas. A ordem das punções foi randomizada por computador. O grupo A (PN, CP, PN, CP) e o grupo B (CP, PN, CP, PN). RESULTADOS: As sensibilidade, especificidade e acurácia da PN vs CP foram 95,2% vs 92,3%; 100% vs 100%; 95,7% vs 93%, respectivamente. Quando foi considerada a associação das duas técnicas (PN+CP), os resultados da sensibilidade, especificidade e acurácia foram 95,6%, 100% e 96%. O valor preditivo positivo (VPP) foi alto para todas técnicas (100%) e os valores preditivos negativos (VPN) para PN, CP e associação dos métodos foram de 66,7% vs 57,1% vs 66,7%, respectivamente. Não houve diferença estatística no rendimento diagnóstico entre a PN e CP: 88% vs 80% (p=0,344), entretanto as técnicas somadas (PN+CP) foi superior à CP isolada: 94% vs 80% (p=0,016). A avaliação citológica qualitativa e semi-quantitativa de celularidade e contaminação por sangue no esfregaço e emblocado pelas técnicas de PN e CP não apresentou diferença estatística (p>0,05). CONCLUSÃO: O rendimento diagnóstico da PAAF-EE pela técnica de PN versus CP de massas sólidas de pâncreas não apresenta diferença estatística, entretanto a utilização das técnicas combinadas é superior, particularmente em relação à CP isolada. A celularidade e contaminação por sangue é semelhante independente da técnica utilizada (PN ou CP) ou do tipo de preparação citológica (esfregaço ou emblocado) / BACKGROUND: Endoscopic ultrasound fine needle aspiration (EUS-FNA) is an established diagnostic method for solid pancreatic masses. Application of negative pressure (NP) during EUS-FNA has the purpose of holding tissue against the cutting edge of the needle as it is moved through the lesion leading to cell detachment and drawing them up. Capillary (CP) sampling is another technique that removes the needle stylet gradually and continuously during the EUS-FNA and seems to have comparable results with less blood and clot aspiration. AIM: Analyze diagnostic yield of EUS-FNA of pancreatic solid mass using NP versus CP and evaluate cytological qualitative and semi-quantitative features of cellularity and blood contamination of each group. METHOD: A total of 50 patients were randomized for NP or CP EUSFNA with a standard 22G needle. Each patient had four passes using NP and CP in an alternate fashion. Computer randomization generated two groups. Group A (NP, CP, NP, CP) and group B (CP, NP, CP, NP). RESULTS: Diagnostic yield of NP versus CP had no statistical difference (p=0.344). Both techniques (NP+CP) was superior to CP alone (p=0.016), but not to NP alone (p=0.25). Sensitivity, specificity and accuracy of NP vs CP were 95.2% vs 92.3%; 100% vs 100%; 95.7% vs 93%, respectively. Results for sensitivity, specificity and accuracy of both NP+CP were 95.6%, 100% and 96%. Positive predictive value was high for all techniques (100%) and Negative predictive value for NP, CP and both techniques were 66.7%, 57.1% and 66.7%, respectively. NP and CP smears and cell blocks had no statistical differences in qualitative and semi-quantitative citological evaluation for cellularity and bloodiness (p > 0.05). CONCLUSIONS: NP versus CP in EUS-FNA of solid pancreatic mass yielded similar overall outcomes in terms of diagnostic performance, however the combination of both techniques is superior, particularly compared to CP alone. Cellularity and bloodiness are equivalent no matter which technique (NP or CP) or type of cytological preparation (smear or cell block) is used

Separação e pré-concentração de metais : uma transferência do meio aquoso para fase sólida ou líquidos orgânicos

A utilização de cera de parafina derretida como um extrator químico permitiu agregar as principais vantagens da extração em fase líquida (homogeneidade do extrato, curto tempo de extração e simplicidade de operação) na extração em fase sólida, delineando um procedimento inovador. O papel matte foi outro material estudado nesse trabalho e mostrou-se interessante como extrator químico, especialmente pelo baixo custo dessa fase sólida. Como prova de conceito, o íon metálico cobre (II) em solução aquosa foi utilizado para o desenvolvimento da metodologia. O extrato resultante de ambos os procedimentos de extração foi analisado diretamente por Espectrometria de Emissão Óptica com Plasma Induzido por LASER (LIBS). Os fatores que afetam a reação de complexação, a extração e a detecção do analito na fase sólida foram otimizados num estudo univariado. Nas melhores condições encontradas, foram construídas curvas de calibração numa faixa linear de 0,50 até 10,00 mg L-1 e alcançados limites inferiores de detecção de 0,12 mg L-1 e 0,08 mg L-1 de cobre (II) em solução aquosa, para os procedimentos com cera de parafina e com o papel matte, respectivamente. A precisão de ambos os métodos propostos mostrou-se inferior a 5% e a exatidão foi avaliada por: (i) análise de material de referência certificado de soro sanguíneo humano; (ii) ensaios de adição e recuperação do analito em amostras líquidas de composição química distintas (água mineral, água do mar, água de torneira, cachaça, refrigerante e urina humana) e (iii) comparação dos resultados obtidos com os fornecidos pelo método de Espectrometria de Absorção Atômica com Forno de Grafite. Ambos os procedimentos se destacaram pela: (i) elevada eficiência no processo de extração (>94%); (ii) facilidade de execução; (iii) baixo custo e inocuidade dos extratores; (iv) microhomogeneidade das fases sólidas obtidas; (v) possibilidade de préconcentração de íons metálicos e (vi) superação das dificuldades intrínsecas na análise de líquido por LIBS. / Two new extraction procedures that use non-conventional extractant materials are described in this thesis. The use of melted paraffin wax as a chemical extractant allowed to aggregate the main advantages of the liquid phase extraction (homogeneity of the extract, short extraction time and simplicity of operation) in solid phase extraction, delineating an innovative procedure. The matte paper was another material studied in this work and was interesting as a chemical extractant, especially for the low cost of this solid phase. As proof of concept, the copper (II) ion in aqueous solution was applied for the development of the methodology. The extract obtained from both extraction procedures was analyzed directly by LASER-Induced Breakdown Spectroscopy (LIBS). The parameters that affecting the complexation reaction, extraction and analyte detection in the solid phase were optimized in a univariate approach. Under the best conditions, calibration curves were constructed in a linear range of 0.50 to 10.00 mg L-1 and lower detection limits of 0.12 mg L-1 and 0.08 mg L-1 of copper (II) in aqueous solution were obtained, for the procedures with paraffin wax and matte paper, respectively. The precision of both methods was less than 5% and accuracy was assessed by: (i) analysis of certified reference material of human blood serum; (ii) spike and recovery analyte in liquid samples of different chemical compositions (seawater, tap water, mineral water, cachaça, soft drink and human urine) and (iii) comparison of the results obtained with those provided by the Graphite Furnace Atomic Absorption Spectrometry. Both procedures were highlighted by: (i) high efficiency in the extraction process (> 94%); (ii) ease of execution; (iii) low cost and safety of extractants; (iv) microhomogeneity of the solid phases obtained; (v) the ability of preconcentration metal ions and (vi) overcoming the intrinsic drawbacks in liquid analysis by LIBS.

Snižování pracovních teplot při výrobě a pokládce asfaltových směsí / Reducing the operating temperature of the production and laying of asphalt mixtures

The subject of this thesis is the issue of reduction of the operating temperatures in the production and laying asphalt mixtures with a more specific focus on influencing the functional properties of these mixtures, especially low-temperature parameters. The thesis is notionally divided into two parts. The first part refers to the low-temperature asphalt mixtures and the second part pays the attention to asphalt mixtures with an added R-material. For production of asphalt mixtures in the first part of thesis were used five additives and for the second part were used a total of four different asphalt mixtures which differs in the participation of added R-material. Both parts are divided into a theoretical part, which discusses the existing experience, the use of these mixtures and the practical part, which analyses in detail the tested mixture ( the composition and preparation of tested specimens ) and deals with the result of laboratory test. In particular, it is a test of asphalt mixture resistance against the formation of frost cracks according to ČSN EN 12697-46 and by comparing the individual mixtures in comparison with the reference sample and among each other.

Možnosti aplikace systémů s akumulací tepla v jaderné energetice / Application possibilities of systems with heat accumulation in nuclear power

This dissertation covers the application of heat accumulation systems in nuclear power engineering, namely in nuclear power plants. It is mainly a case of passive emergency systems, whose task is to accumulate the heat produced in the reactor’s active zone and in spent fuel pools during DBA (design-basis accidents) or beyond DBA. A particular example of heat accumulation is steam condensation after LOCA (loss of coolant accident). The primary circuit steam leakage increases containment pressure and has to be decreased by the steam condensation. This thesis deals with a theoretical substitute for ice condensers, which are used as a passive safety measure in some nuclear power plants. The substitute involves a choice of an alternative material, whose melting temperature (for heat accumulation) is closer to nuclear power plant operating temperatures. The other part of the dissertation discusses heat accumulation in spent fuel pools in case of all cooling systems failure.

Thermomechanical and rheological properties of investment casting patterns

(Department of Chemical Engineering, Faculty of Engineering and Technology), Vaal University of Technology. / Investment casting process is the most suitable technique for producing high quality castings which are dimensionally accurate with excellent surface finish and complex in nature. Recently with the ever-changing manufacturing landscape, the process has been increasingly used to produce components for the medical, aerospace and sports industry. The present study looked at three investigative scenarios in the development of a pattern material for investment casting process: (i) the development of wax/ethyl vinyl acetate (EVA) and wax/linear low-density polyethylene (LLDPE) blends as the carrier vehicle materials for the development of pattern material for investment casting; (ii) the development of wax/EVA/polymethyl methacrylate (PMMA) based investment casting pattern and lastly (iii) the development of wax/LLDPE/PMMA based investment casting pattern material. The first part of the studies elucidates the effects in terms of the thermal, mechanical, surface and rheological properties when paraffin wax in blended with poly EVA and LLDPE. The developments involved the extrusion of seven formulations for EVA and also LLDPE using a twin-screw extrusion compounder. The paraffin wax weight percent investigated ranged from 33% to 87% thus encompassing both low and high wax loading ratios. The thermal properties of the developed binary blends were characterized via thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC). The mechanical properties were characterized using three-point bending test. The thermo-mechanical and rheological properties were determined using thermomechanical analysis (TMA) and a rheometer respectively. A scanning electron microscope (SEM) was used to study the surface texture of the extruded blends. The thermal properties indicated that the thermal stability of paraffin wax is improved when it is blended with both EVA and LLDPE. DSC curves showed two endothermic melting peaks and two exothermic crystallisation peaks. In the case of wax/EVA blends, there was no distinct peak showing the independent melting of neat wax and EVA. The peak at a temperature of 50 – 72 °C corresponds to the melting of the wax/EVA blend. In the case of wax/LLDPE blends, the peak at 50 -66 °C corresponds to the melting of wax whereas the large peak at 112 - 125°C corresponds to the melting of the LLDPE. Wax/EVA and wax/LLDPE had improved mechanical properties as compared to that of neat wax. The rheological properties of both the EVA based and LLDPE based blends indicated that the viscosity of the blends increased as compared to that of neat wax. SEM confirmed that EVA alters the wax crystal habit at higher concentrations. In the case of wax/LLDPE blends, at 20-30 % wax content, a heterogeneous surface was observed, indicating the immiscibility of the paraffin wax within the LLDPE matrix. At a high wax content, there was agglomeration of wax. LLDPE allows amorphous structure of wax to disperse easily between the chains. The second part of the studies focussed on the wax/EVA filled with poly (methyl methacrylate) (PMMA) microbeads. TGA behaviour on the pyrolysis of wax/EVA/PMMA showed that the compounds volatilise readily with virtually no residue remaining above 500 °C. The DSC curves indicated that, the incorporation of PMMA reduced the crystallinity of wax/EVA blend. A distinct endothermic peak and another small peak was observed in all the formulations. The mechanical properties of wax/EVA/PMMA improved significantly. The methylene group present in both wax and EVA combined to form a blend with enhanced mechanical properties whereas the PMMA microbeads improved the needle penetration hardness. The melt viscosity of wax/EVA/PMMA increased as the EVA and/or the PMMA content is increased. The rheological experimental data fitted with the data predicted using the modified Krieger and Dougherty expression. The maximum attainable volume fraction of suspended PMMA particles was at max = 0.81. The SEM micrograph of wax/EVA/PMMA revealed a near perfect spherical nature for the filler particles in the wax/EVA polymer matrix. It further shows that the PMMA microbeads were weakly bonded and well distributed in the wax/EVA matrix. The third part of the studies focussed on the wax/LLDPE filled with Poly (methyl methacrylate) (PMMA) microbeads. The incorporation of LLDPE and PMMA into paraffin wax had a strong influence on the thermal properties, tensile properties, flow properties and its morphology. The TGA analysis showed that there was a slight observable decrease in the melting onset temperatures when the wax content was increased. From the DSC curves, the corresponding values of onset temperatures observed are between melting and crystallization temperature of neat paraffin wax and neat LLDPE. The short chains of the paraffin wax and the fragments formed by scission of wax chain have sufficient energy to escape from the matrix at lower temperatures. The slight decrease in peak temperatures associated with melting and crystallization could be attributed to the decrease in the average lamellar thickness of the blends. The tensile properties by three-point bending tests indicated an increase in the stress with an increase in the LLDPE content. This can be attributed to the formation of paraffin wax crystals in the amorphous phase of the blend which may influence the chain mobility. Since the paraffin wax used for this study had a low viscosity as compared to LLDPE, both LLDPE or PMMA had an influence on the viscosities of the blends. The data obtained from the experiments fitted with the data predicted obtained from the modified Krieger and Dougherty expression. The maximum attainable volume fraction of suspended PMMA particles was at max = 0.74. Similar observation with that of wax/EVA/PMMA was made in terms of the morphology of the wax/LLDPE/PMMA blends. The excellent thermal stabilities, the superior mechanical strength of wax/EVA/PMMA and wax/LLDPE/PMMA and the flow properties with relatively high EVA and also with high PMMA loadings, open new opportunities for EVA and LLDPE based pattern material for in investment casting process. It is worth pursuing further comprehensive studies since it offers a strong potential for realizing further technological improvement in the field of investment casting and rapid prototyping technologies.

Laboratory Evaluation of Water-Repellent Soils for Water Harvesting

From the Proceedings of the 1974 Meetings of the Arizona Section - American Water Resources Assn. and the Hydrology Section - Arizona Academy of Science - April 19-20, 1974, Flagstaff, Arizona / Reported are laboratory evaluations to screen water-repellent materials and treatments before testing them in the field. Water repellency tests were conducted on paraffin wax, a wax emulsion and silicon, lard, and a liquid dust suppressant. Six water repellency tests showed that the high rates of paraffin wax and all rates of the dust suppressant produced highly water-repellent soil surfaces. The six water repellency tests were: (1) the aqueous-alcohol drop test for determination of the 90 degree surface tension for a porous solid, (2) the water drop penetration time test, (3) the relative height of a large sessile water drop resting on the smoothed, treated soil surface, (4) and (5) the presence and persistence of air bubbles trapped between the soil-water interface, and test (6) was made to note whether the large sessile water drop from test (3) would infiltrate the soil or evaporate. Tests (3), (4), and (5) proved the most useful of the six methods for measuring water repellency. Soil type had no significant influence on degree of water repellency as measured in the laboratory by these six tests.
