Obtenção e avaliação de mutantes exoZ- e phbAB- envolvidos no metabolismo de polímeros de carbono em Rhizobium tropici SEMIA 4080 com potencial biotecnológico

A diversidade de estruturas e composição química apresentadas pelas moléculas de EPS é refletida pela diversidade de enzimas responsáveis pela sua síntese. Neste trabalho, buscou-se a inativação através da mutagênese insercional por transposição in vitro dos genes exoZ da estirpe selvagem Rhizobium tropici SEMIA 4080 envolvidas na síntese das subunidades repetitivas do EPS e no tanden phbAB, responsáveis pela síntese do PHB, com o intuito de obter microrganismos com aspecto altamente mucoso, produtor superior de EPS em relação a estirpe selvagem. Os mutantes obtidos apresentaram colônias mucosas quando cultivados nos meios de culturas PSYA, demonstrando que estirpes de rizóbio mutantes nos genes exoZ e phbAB são capazes de formar biofilme “in vitro” e, ao avaliar a eficiência relativa na produção de EPS, o mutante 4080 Z03 apresentou o melhor resultado. Para as três amostras de EPS foi possível observar um comportamento de fluxo de líquidos pseudoplástico e, também, a influência da concentração de EPS sobre a viscosidade aparente das soluções aquosas. A estirpe selvagem e todos os mutantes induziram a formação de nódulos em feijoeiro (fenótipo Fix+), sugerindo que os genes exoZ e phbAB não estão envolvidos nos processos de infecção e formação nodular mas, sendo necessários para a fixação biológica de nitrogênio / Rhizobium tropici and other bacteria belonging to the order Rhizobiales are extracellular polysaccharides (EPS) producers, which possess the function of the receptor molecule to function as micro-symbiont, making an interaction cell / cell and triggering the nodulation process. The diversity of structures and chemical composition of EPS presented by molecules is reflected by the diversity of enzymes responsible for its synthesis. In this study, we sought to inactivation by insertional mutagenesis by “in vitro” transposition of genes from wild type exoZ Rhizobium tropici SEMIA 4080 involved in the synthesis of the EPS repeating subunits and phbAB tanden, responsible for synthesis of PHB, in order to obtain microrganisms with high mucosal aspect producer of EPS compared to wild type. The mutants showed mucous colonies when grown in culture media PsyA, demonstrating that mutant strains in genes exoZ and phbAB from rhizobia are able to form biofilm “in vitro”, and to evaluate the relative efficiency in the production of EPS, the mutant 4080 Z03 showed the best result. For the three samples of EPS was possible to observe a flow behavior of pseudoplastic fluids, and also the influence of EPS concentration on the apparent viscosity of aqueous solutions. The wild type and mutants induced the formation of nodules in common bean (Fix+ phenotype), suggesting that genes exoZ and phbAB are not involved in the processes of infection and nodule formation, but are necessary for nitrogen fixation

O vínculo entre reologia, mecanismos de deformação que atuaram na formação de várias microestruturas e texturas ou orientações cristalográficas preferenciais (OCP), fornecem resultados que podem ser aplicados para uma explotação estratégica dos diferentes tipos de minérios, implicando na melhoria de áreas alvos. Este estudo foi focado na avaliação da correlação de microestruturas/texturas com parâmetros de flotação, visando otimizar o processo de flotação para aumentar a recuperação dos minérios pobres (ricos em sílica), que comumente contêm menos de 64% de Fe. Uma série de técnicas, incluindo difração por nêutrons, raios-X e elétrons retroespalhados (elétron backscattering difraction), bem como análises granulométricas e de imagens dos concentrados de flotação foram aplicados neste trabalho. Os resultados obtidos por meio das análises microestruturais e texturais, permitiram enquadrar os diferentes minérios no seu contexto tectônico, dentro do Quadrilátero Ferrífero. Adicionalmente, a diversidade reológica, obtida por várias técnicas resultou na definição de parâmetros de correlação com a flotação, que podem ser aplicados na indústria de beneficiamento, contribuindo para aumentar a recuperação do minério de ferro. / The wide range of minerals, microstructures and textures encountered in iron ores of the Quadrilátero Ferrífero region in Brazil, generates a diversity of physical properties with implications to its processing. The linking between rheology and deformation mechanisms which acted in microstructure and texture or crystallographic preferred orientations (CPO), formation provide results witch can be applied in strategic exploitation of different ore types, leading to the improvement of the selected targets. This study was focused in the evaluation and correlation of textures, microstructures with flotation parameters, aiming to optimize the flotation processing to increase the recovery of low-grade iron ores, which commonly contain less than 64% Fe. A series of techniques, including neutron diffraction, X-ray diffraction, electron backscattering diffraction as well as granulometric and imaging analysis of flotation concentrates were applied in this study. The results obtained through the textural and microstructural studies allow fitting different ores in your tectonic setting, into Quadrilátero Ferrífero. Additionally, the diversity of rheologic features defined by the different techniques led to define new correlation parameters with selectivity indices of flotation that can be applied to the iron ore processing industry, contributing to increase iron ore recovery.

Determinação de espectros de relaxação e distribuição de massa molar de polímeros lineares por reometria

A distribuição de massa molar (DMM) e seus parâmetros são de fundamental importância na caracterização dos polímeros. Por este motivo, o desenvolvimento de técnicas que permitam a determinação da DMM de forma mais rápida e a menor custo é de grande importância prática. Os principais objetivos deste trabalho foram a implementação de alguns dos modelos baseados da teoria da reptação dupla propostos na literatura para descrever o mecanismo de relaxação das cadeias poliméricas, a avaliação dessas implementações e a análise de dois passos fundamentais na obtenção da DMM a partir de dados reológicos que são a metodologia de cálculo do espectro de relaxação baseado no modelo de Maxwell e a estratégia para a avaliação numérica das integrais que aparecem nos modelos de relaxação. Foi resolvido o problema denominado problema inverso, ou seja, a determinação da DMM a partir de dados reológicos usando um modelo de relaxação especificado e uma função de distribuição imposta. Foi usada a função Exponencial Generalizada (GEX) para representar a probabilidade de distribuição, sendo consideradas duas abordagens: i) cálculo explícito do espectro de relaxação e ii) aproximações paramétricas de Schwarzl, que evitam a necessidade do cálculo explícito do espectro de relaxação. A metodologia de determinação da DMM foi aplicada para amostras de polietileno e foram estimadas distribuições com boa representação dos dados experimentais do GPC, ao considerarem-se amostras com polidispersões inferiores a 10. Com relação a metodologia de cálculo do espectro de relaxação, foi realizado um estudo comparativo da aplicação de espectros de relaxação discreto e contínuo, com o objetivo de estabelecer critérios para especificação do número ótimo de modos de Maxwell a serem considerados. Ao efetuar-se a comparação entre as técnicas, verificou-se o espectro discreto apresenta como um sistema melhor condicionado, permitindo assim obter maior confiabilidade dos parâmetros estimados. Também é proposta uma modificação da metodologia de determinação da DMM, em que é aplicada a quadratura de Gauss-Hermite para a resolução numérica da integral dos modelos de relaxação. / The molecular weight distribution (MWD) and its parameters are of the fundamental importance in the characterization of polymers. Therefore, the development of techniques for faster and less time consuming determination of the MWD is of great practical relevance. The goals of this work were the implementation of some of the relaxation models from double reptation theory proposed in the literature, the evaluation of these implementations and the analysis of two key points in the recovery of the MWD from rheological data which are the methodology for calculation of the relaxation spectrum based on the Maxwell model and the numeric strategy for the evaluation of the integrals appearing in the relaxation models. The inverse problem, i.e., the determination of the MWD from rheological data using a specified relaxation model and an imposed distribution function, was solved. In the analysis of the inverse problem, the Generalized Exponential (GEX) was used as distribution function and two approaches were considered: i) explicit calculation of the relaxation spectrum and ii) use of the parametric method proposed by Schwarzl to avoid the explicit calculation of the relaxation spectrum. In the test of commercial samples of polyethylene with polidispersity less than 10, the application of this methodology led to MWD curves which provided good fit of the experimental SEC data. Regarding the methodology for calculation of the relaxation spectrum, a comparison between the performance of discrete and continuous relaxation spectrum was performed and some possible a criteria to determine the appropriate number of relaxation modes of Maxwell to be used were evaluated. It was found that the technique of discrete spectrum leads to better conditioned systems and, consequently, greater reliability of the estimated parameters. With relation to the numeric strategy for the evaluation of the integrals appearing in the relaxation models, the use of Gauss-Hermite quadrature using a new change of variables was proposed.

Time Dependent Rheological Response of Composite Binders

abstract: The need for sustainability in construction has encouraged scientists to develop novel environmentally friendly materials. The use of supplementary cementitious materials was one such initiative which aided in enhancing the fresh and hardened concrete properties. This thesis aims to explore the understanding of the early age rheological properties of such cementitious systems. The first phase of the work investigates the influence of supplementary cementitious materials (SCM) in combination with ordinary Portland cement (OPC) on the rheological properties of fresh paste with and without the effect of superplasticizers. Yield stress, plastic viscosity and storage modulus are the rheological parameters which were evaluated for all the design mixtures to fundamentally understand the synergistic effects of the SCM. A time-dependent study was conducted on these blends to explore the structure formation at various time intervals which explains the effect of hydration in conjecture to its physical stiffening. The second phase focuses on the rheological characterization of novel iron powder based binder system. The results of this work indicate that the rheological characteristics of cementitious suspensions are complex, and strongly dependent on several key parameters including: the solid loading, inter-particle forces, shape of the particle, particle size distribution of the particles, and rheological nature of the media in which the particles are suspended. Chemical composition and reactivity of the material play an important role in the time-dependent rheological study. A stress plateau method is utilized for the determination of rheological properties of concentrated suspensions, as it better predicts the apparent yield stress and is shown to correlate well with other viscoelastic properties of the suspensions. Plastic viscosity is obtained by calculating the slope of the stress-strain rate curve of ramp down values of shear rates. In oscillatory stress measurements the plateau obtained within the linear visco-elastic region was considered to be the value for storage modulus. Between the different types of fly ash, class F fly ash indicated a reduction in the rheological parameters as opposed to class C fly ash that is attributable to the enhanced ettringite formation in the latter. Use of superplasticizer led to a huge influence on yield stress and storage modulus of the paste due to the steric hindrance effect. In the study of iron based binder systems, metakaolin had comparatively higher influence than fly ash on the rheology due to its tendency to agglomerate as opposed to the ball bearing effect observed in the latter. Iron increment above 60% resulted in a decrease in all the parameters of rheology discussed in this thesis. Rhéologie et mécanique des fluides du conditionnement de produits semi-solides / Rheology and fluid mechanics of semi-solid products packaging

Le conditionnement, opération unitaire clé du processus industriel de fabrication des produits fromagers, consiste à remplir par un jet de façon homogène des emballages de formes variées par des produits à propriétés rhéologiques complexes tout en assurant des cadences optimisées. L’objectif de cette étude est de prédire le remplissage d’un emballage, à partir de la connaissance de la rhéologie des produits et des paramètres opératoires pour le dimensionnement de nouveaux procédés de conditionnement ou l’optimisation de procédés existants.Nécessaire à la poursuite de l’étude, le comportement rhéologique des produits fromagers dans les domaines de sollicitation du procédé à 80°C a été caractérisé comme rhéofluidifiant, à seuil de contrainte, élastique en-dessous et au-dessus du seuil d’écoulement.L’hydrodynamique des jets et du remplissage lors du conditionnement a été étudiée avec trois types de produits grâce au pilote à l’échelle laboratoire du procédé, dimensionné pour reproduire les conditions opératoires industrielles. Après avoir vérifié la cohérence des résultats obtenus avec le dispositif expérimental avec ceux de la littérature pour un fluide newtonien, l’étude s’est portée sur des fluides modèles viscoplastiques, gels de Carbopol®, et sur des produits fromagers.Cinq régimes d’écoulement ont été observés et caractérisés à partir des évolutions du diamètre de jet et de la vitesse d’atteinte des bords pendant le remplissage et de la hauteur de motte finale du dépôt après le remplissage, paramètre décisif de la qualité du conditionnement. Des diagrammes de fonctionnement adimensionnels d’observation des régimes ont été déterminés à partir des forces en jeu dans le système. Ainsi, le régime d’écoulement peut être prédit par ces diagrammes à partir des propriétés rhéologiques du produit et des paramètres géométriques et opératoires du procédé. / Packaging is a key step of cheese products industrial manufacturing process. It consists in filling containers of various shapes with non-Newtonian products while ensuring optimized rates. The aim of the present work is to describe the influence of cheese rheological properties as well as process parameters onto packaging, thereby contributing towards its optimization.Necessary for the study, the rheological behavior of cheese products on the process stress range at 80°C has been identified as shear-thinning with a yield stress, with elasticity both below and above the yield stress.Jets and filling hydrodynamics during packaging have been studied with three types of products by the means of the laboratory-scale process pilot, which is able to reproduce the industrial process conditions. After checking for consistency between experimental and litterature results for Newtonian products, the study focused on viscoplastic model fluids, Carbopol® hydrogels, and on cheeses.Five flow patterns have been observed and characterized through jet diameter, spread velocity during filling and final mound height after filling, which is a key factor of the packaging quality. Flow patterns dimensionless distributions have been determined from the forces involved in the system. Thus, the flow pattern can be predicted thanks to theses distributions from product rheological properties and geometrical and process parameters.

Suspension rheology and extrusion : a discrete element method study

A suspension is a fully saturated mixture of discrete solid particles and interstitial liquid. Examples of suspensions include pastes, slurries, cement, food-spreads, drilling fluids and some geophysical flows. The present work focusses on granular (as opposed to colloidal) suspensions, which we define as those for which the thermal motion of the solid particles is negligible. Despite such ubiquity in industry and nature, our understanding of the mechanical properties of suspensions lags behind that of their constituent solid and liquids. In this thesis, the discrete element method is used to simulate suspension flow in shear, capillary and constriction geometries, mapping and characterising the fundamental flow, or rheological, regimes. As a starting point (Chapter 2), we consider an established regime map for dry granular materials, appropriate for flows of sand, grains and dry debris. Taking guidance from shear flow simulations that consider the lubricating effect of an interstitial liquid, we recast the regime map for a general suspension, elucidating flows comparable to the dry material or to a viscous liquid, dependent on the shear rate, liquid viscosity and particle stiffness. We give an account of the microstructural traits associated with each regime. Motivated by recent groundbreaking theoretical, computational and experimental work, we incorporate the emerging picture of frictional shear thickening into our regime map (Chapter 3). Our shear flow simulations capture the shear thickening behaviour and demonstrate that it may, in principle, occur in any of the identified flow regimes. Our simulations of time-dependent shear flows (Chapter 4), specifically flow reversal, provide a detailed micro-mechanical explanation of a longstanding and previously unexplained experimental finding, guiding future experimentalists in decomposing the particle and liquid contributions to the viscosity of any suspension. Indeed, the findings have already been exploited in the devising of an experimental protocol that has successfully proven the dominance of particle contacts in driving shear thickening. We next consider suspension flow in a microchannel (Chapter 5), finding that the identified shear flow regimes are locally applicable to flows in complex geometries under inhomogeneous stress conditions only when the local mean shear rate exceeds temporal velocity fluctuations. A more comprehensive description is therefore required to fully characterise the flow behaviour in this geometry. Finally (Chapter 6), we simulate pressure driven suspension flow through a constriction geometry, observing highly inhomogeneous stress distributions and velocity profiles. The roles of particle and fluid properties are considered in the context of an industrial paste extrusion process.

Triumfetta cordifolia et Bridelia thermifolia sont deux arbustes de la région tropicale d'Afrique dont les gommes extraites des écorces fraîches ou séchées sont confinées aux utilisations traditionnelles, notamment dans la cuisine (propriétés d'épaississement et d'aromatisation des sauces, d'amélioration de la texture des pâtisseries) et dans le procédé de décantation des bières traditionnelles de sorgho (bili-bili). Dans le but d'identifier la nature des gommes responsables de ces propriétés et d'évaluer leur potentiel de valorisation, la composition chimique (protéines, polysaccharides, fibres et minéraux) des écorces a été déterminée. Les gommes ont été extraites, caractérisées sur le plan chimique (composition en monosaccharides et en minéraux) et physique (comportement rhéologiques des extraits, poids moléculaire, viscosité intrinsèque et rayon de giration) et l'effet combiné des facteurs du milieu (température, pH, et concentration saline (NaCl, KCl, CaCl2 et MgCl2)) influençant la viscosité a été analysé selon un plan composite centré. Enfin, un essai d'application de ces gommes dans la préparation des beignets de maïs et de sorgho a été réalisé. Les résultats des analyses chimiques montrent une prédominance des polysaccharides dans les écorces (52 et 55% respectivement pour B. thermifolia et T. cordifolia) et dans les extraits (76 et 80% respectivement pour B. thermifolia et T. cordifolia), avec des teneurs 5 à 8 fois supérieures à celles des protéines. Ce qui permet de considérer que ces polysaccharides sont responsables des propriétés physiques d'épaississement exprimées par les extraits. Ces propriétés ne sont aucunement modifiées par le séchage des écorces. Les polysaccharides des extraits sont constitués principalement de rhamnose (24,51%), de galactose (15,23%), d'acides glucuronique (25,51%) et galacturonique (20,23%). La présence significative d'acides uroniques justifie la persistance des minéraux dans les extraits purifiés. Les masses moléculaires moyennes des polysaccharides sont de 6,14 x106 et 3,58 x106Da respectivement pour T. cordifolia et B. thermifolia. Sur le plan physique, leurs viscosités intrinsèques et leur rayon de giration sont relativement élevés (18,33dl/g et 269,6nm pour T. cordifolia; 17,98dl/g et 217,8nm pour B. thermifolia). La cinétique de séchage des écorces de T. cordifolia et B. thermifolia se déroule en deux périodes cinétiques représentées par deux constantes de séchage k1 et k2 qui augmentent avec la température de séchage. La viscosité des extraits diminue pour des températures de séchage des écorces au-délà de 70°C. Cette baisse de viscosité des extraits serait plus le résultat d'un changement de conformation des polysaccharides que de leur dégradation. L'étude du comportement rhéologique de ces polysaccharides traduit un comportement rhéofluidifiant, viscoélastique, peu thixotrope et avec des contraintes seuil d'écoulement comprises entre 0,2 et 5Pa et entre 0,5 et 1Pa respectivement pour T. cordifolia et B. thermifolia à des concentrations comprises entre 0,52 et 0,82g/l. La température et les cations monovalents (K+ et Na+) ont un effet dépresseur sur la viscosité des gommes tandis que les pHs faiblement acide et alcalin et les cations divalents (Ca2+ et Mg2+) l'augmentent. L'effet combiné de ces facteurs sur la viscosité des gommes se décrit de façon satisfaisante par un modèle de second degré. L'analyse des interactions entre les facteurs de variation (définit dans le modèle) met en évidence, pour chaque facteur, une plage de valeurs pour lesquelles il contribue à l'obtention d'une zone de viscosités optimales et offre sur le plan pratique une gamme de choix des niveaux de variables en fonction du niveau de viscosité voulue. L'incorporation d'extraits de T. cordifolia (3,5 g/l) et B. thermifolia (11 g/l) dans la farine de maïs ou de sorgho, contribue de façon significative à l'amélioration du gonflemen / Triumfetta cordifolia and Bridelia thermifolia are two shrubs of tropical Africa whose gums extracted from fresh or dried barks are confined to traditional uses, particularly in the kitchen (thickening and flavoring properties in sauces and improvement of the texture of bakery ptoducts) and in the decantation process of the sorghum based traditionnal beer (bili-bili). In order to identify the nature of the gums responsible for these properties and assess their potentials for valorization, the chemical composition (proteins, polysaccharides, minerals and fiber) of barks was determined. The gums were extracted, and characterized for their chemical (monosaccharide composition and minerals) and physical (rheological behavior of extracts, molecular weight, intrinsic viscosity and radius of gyration) properties. Then, the combined effects of environmental factors (temperature, pH and salt concentration [NaCl, KCl, CaCl2 and MgCl2]) influencing the viscosity was analyzed using a central composite design. Finally application test of these gums in preparing doughnuts from maize and sorghum flour was carried out. The results of chemical analyzes showed that polysaccharides dominated in the barks (52 and 55% respectively for B.thermifolia and T. cordifolia) and in extracts (76 and 80% respectively for B.thermifolia and T. cordifolia), with levels 5-8 times higher than those of proteins. This allowed to hypothesize that these polysaccharides are responsible for the thickening properties demonstrated by the extracts. These properties are not be affected by the drying of the bark. The polysaccharides of the extracts are mainly composed of rhamnose (24.51%), galactose (15.23%), glucuronic acid (25.51%) and galacturonic acid (20.23%). The significant presence of uronic acids justifies the persistence of minerals in the purified extracts. The molecular weights of polysaccharides are 6.14 x106 and 3.58x106 Da, respectively for T. cordifolia and B. thermifolia. On the physical level, their intrinsic viscosities and their gyration radius are relatively high (18.33 dl/g and 269.6 nm for T. cordifolia; 17.98 dl/g and 217.8 nm for B. thermifolia). The drying kinetics for barks of T. cordifolia and B. thermifolia presents two falling rate periods represented by two kinetic constants k1 and k2, which increase with increasing drying temperature. Viscosity of extracts decreases when barks are dried beyond 70°C. This may be probably the consequence of a conformational change of polysaccharides than the result of their degradation. The study of the rheological behavior of these polysaccharides indicates a shear thinning, viscoelastic and weak thixotropic behavior, with flowing pressures yield point between 0.2 and 5Pa and between 0.5 and 1Pa, respectively for T. cordifolia and B. thermifolia, at concentrations between 0.52 and 0.82 g/l. Temperature and monovalent cations (K+ and Na+) have a depressant effect on viscosity of gums, while weakly acidic and alkaline pH and divalent cations (Ca2+ and Mg2+) increase it. The combined effect of these factors on the viscosity of gums is satisfactorily described by a second degree model. Analysis of interactions between factors of variation shows, for each factor, a range of values for which it contributes to the achievement of a zone of optimal viscosity. This zone offers a practical range for the choice of variable levels, depending on the desired viscosity. The incorporation of extracts of T. cordifolia (3.5 g/l) and B. thermifolia (11 g/l) in the maize or sorghum flour, improves significantly the swelling of the dough. The doughnuts obtained after baking are, from organoleptic point of view, comparable to wheat doughnuts.

Les glissements de terrain argileux affectent de nombreux versants à travers le monde et menacent régulièrement les activités humaines dans les zones urbanisées montagneuses. Ces glissements sont caractérisés par des cinématiques souvent lentes mais ils peuvent brutalement se liquéfier et accélérer de manière imprévisible. Cette transition solide-liquide a été étudiée sur les argiles de la région du Trièves (Alpes Françaises) à l'aide d'études rhéologiques. Elles ont montré le caractère de fluide à seuil thixotrope avec une bifurcation de viscosité importante lors de la fluidification pouvant expliquer le caractère catastrophique de l'accélération observée sur le terrain. Cette perte de rigidité du matériau peut être observée par une chute de la vitesse des ondes de cisaillement (Vs). Des études réalisées en parallèle à la fois sur un modèle analogique de plan incliné et sur le terrain (glissement de Pont-Bourquin, Suisse) ont permis d'observer une chute de Vs précédent à cette fluidification montrant ainsi que Vs pourrait être un bon proxy pour la surveillance des instabilités de terrain argileux. / Landslides affect many clay slopes in the world and regularly threaten people in urban areas mountainous. These landslides are characterized by a slow velocity but they may suddenly liquefy and accelerate unexpectedly. The solid-liquid transition on the clay has been studied of Trièves region (French Alps) using rheological experiments. They have shown the yield stress thixotropic behavior with a viscosity bifurcation which can explain the catastrophic fluidization observed in the field. This loss of material stiffness can be followed by a drop in the shear wave velocity (Vs). Inclined plane test and field experiments (Pont-Bourquin landslides in Switzerland) have both shown a precursor drop of Vs indicating that it could be a good proxy for monitoring unstable clay slope.

Viscoélasticité du sang et du caillot / Blood and blood clot viscoelasticity

Le sang est un fluide complexe mis en écoulement par la pompe très peu puissante qu'est le cœur (environ 1 W), dans un réseau branché de plusieurs milliers de kilomètres de vaisseaux. Pour que cela soit réalisable, il se peut que les propriétés mécaniques du sang contribuent à l'entretien de l'écoulement. Malgré le nombre important d'études sur la rhéologie du sang, sa viscoélasticité n'a jamais été caractérisée en cisaillement simple. Le rôle physiologique du caillot est, lui, d'éviter un épanchement excessif de sang en présence d'une brèche vasculaire. Une de ses fonctions principales est donc de résister aux contraintes générées par l'écoulement sanguin, c'est-à-dire d'avoir une résistance mécanique appropriée. Que ce soit pour la caractérisation mécanique du sang ou du caillot, le principal verrou est l'absence de méthode de mesure adaptée à un matériau peu consistant, et dont les propriétés mécaniques sont en évolution rapide. Il est donc nécessaire de produire une méthode de mesure adéquate, couplée à un système de mesure assez sensible. Dans ce travail, nous présentons la méthode de rhéométrie que nous avons développée dans ce but, baptisée Optimal Fourier Rheometry (OFR). Cette technique a été validée avec succès sur différents matériaux modèles de plus en plus complexes : une huile newtonienne, une gomme viscoélastique (PDMS), une suspension de micelles vermiformes (CpCl Nasal) et enfin un alginate dentaire tout au long de sa gélification. Nous montrons ainsi que l'OFR est une technique de mesure fonctionnelle, fiable et optimale temporellement. Elle permet le suivi de grandeurs mécaniques dont le temps caractéristique de mutation est très inférieur à la minute. En raison de la sédimentation des globules rouges, le sang est un fluide évoluant dans le temps. Par conséquent l'OFR est bien adaptée pour la mesure de ses propriétés viscoélastiques. Pour nous affranchir de la variabilité très importante du sang de témoins, nous avons balayé de façon systématique la concentration en les composants sanguins les plus abondants sur des suspensions de globules rouges lavés. De façon a priori surprenante, nous montrons qu'en présence de fibrinogène, le sang présente une élasticité importante, du même ordre de grandeur, voire plus grande que sa viscosité. Cette élasticité augmente avec la concentration en fibrinogène et l'hématocrite et provient du réseau percolé de globules rouges agrégés de dimension fractale 2.08 qui existe dans la suspension lorsqu'elle est peu cisaillée. L'OFR a également été appliquée au suivi de la coagulation activée par voie intrinsèque et extrinsèque. Cela a permis de montrer que le procédé d'activation n'avait d'effet que sur la cinétique de la réaction, mais que cela ne changeait pas les étapes mécaniques observées. L'OFR permet grâce à sa résolution fréquentielle élevée et son temps de mesure minimal, d'affirmer que le processus de coagulation du sang n'est pas une transition sol-gel. / Blood is a complex fluid set into flow by the heart, which is a very low power pump (approximately 1 W), in a connected network consisting of several thousand kilometers of vessels. To do so, it seems reasonable that the mechanical properties of blood contribute to the maintenance of the flow. In spite of the important number of studies on blood rheology, the viscoelasticity of blood has never been characterized in simple shear. The physiological role of the blood clot is to avoid an excessive effusion of blood in the presence of a vascular breach. Thus, it has to resist to the stress induced by the blood flow. So, one of its essential functions is this mechanical resistance. Whether it is for the mechanical characterization of the blood or the clot, the main obstacle is the absence of viscoelasticity measurement techniques adapted to a low viscosity material evolving rapidly in time. So, it is necessary to provide an adapted measurement method, coupled with a sensitive enough measurement. In this work, we present the new rheometry method we developed, named Optimal Fourier Rheometry (OFR) as it is optimal both in duration and signal to noise ratio This method was successfully validated on materials of increasing complexity: a Newtonian oil, a viscoelastic gum (PDMS), a suspension of wormlike micelles (CpCl Nasal) and a dental alginate during its gelation. Because of the sedimentation of red blood cells, the mechanical properties of blood are evolving in time. Consequently the use of the OFR is well suited for the measurement of its viscoelastic properties. A systematic scanning of the concentrations in the most abundant blood components added to washed blood allowed to highlight the most important parameters. Our results show that blood has a surprisingly large elasticity, which is of the same order of magnitude as the viscosity of the material. This elasticity increases with fibrinogen concentration and hematocrit. When these two parameters are in the physiological range, a percolated network of aggregated red blood cells exists in the suspension of fractal dimension 2.08. The, OFR was applied to the monitoring of blood clot formation. The activation by intrinsic and extrinsic pathway was used on whole blood. It showed that the process of activation affects only the kinetics of the reaction, but does not change the observed mechanical s. Due to its high frequency resolution and minimal measurement time, OFR shows that coagulation is not a gelation process.
