Synthesis of Insecticidal Mono- and Diacylhydrazines for Disruption of K+ Voltage-Gated Channels, and Elucidation of Regiochemistry and Conformational Isomerism by NMR Spectroscopy and Computation

Based on the success of diacyl-tert-butylhydrazines RH-5849 and RH-1266 in controlling agricultural crop pests, we endeavored to synthesize our own diacylbenzyl- and arylhydrazine derivatives for use against the malaria vector Anopheles gambiae. In the process of producing a library of compounds for assay against An. gambiae, it became clear that employing regioselective acylation techniques (in molecules that feature two nucleophilic, acyclic nitrogen atoms α to one another) would be imperative. Synthesis of the library derivatives proceeded rapidly and after topical assay, we found three compounds that were more toxic than the RH-series leads. One of the three displayed an LD50 value of half that of RH-1266, though patch clamp assay concluded that toxicity was not necessarily linked to inhibition of mosquito K+ channel Kv2.1. The acylation of monoarylhydrazines appears simple, but its regioselectivity is poorly understood when assumed as a function of basicity correlating to nucleophilic strength. We determined the ratio of the rate constants for distal to proximal N-acylation using 19F NMR spectroscopic analysis of reactions of 4-fluorophenylhydrazine with limiting (0.2 equiv) acylating agent in the presence of various bases. Acid anhydrides gave consistent preference for distal acylation. The selectivity of acylation by acyl chlorides when using pyridine gives strong distal preference, whereas use of triethylamine or aqueous base in conjunction with aroyl chlorides showed a moderate preference for proximal acylation. This observation yielded a convenient one-step method to synthesize proximal aroylarylhydrazines in yields comparable or superior to that provided by the standard three-step literature approach. Combined with NMR evidence of the distal nitrogen as the unambigiously stronger base of the two nitrogens, we propose a single electron transfer mechanism that predicts the regiochemistry of arylhydrazines toward acylating agents better than the nucleophilicity model based on pKa values. While synthesizing the acylhydrazine library for assay against An. gambiae, NMR spectroscopy revealed rotational isomerisms of two types: chiral helicity (M)/(P) and acyl (E)/(Z)-isomerism due to hindered rotation. Variable temperature NMR allowed the measurement of N-N bond rotational barriers, as well as estimate the barrier of (E)/(Z) interconversion. We obtained the X-ray crystal structures of four diacylhydrazines to test this hypothesis and revealed both the twist conformation around the N-N bond axis and (E)/(Z)-isomerism around the proximal acyl group. Computation (which agreed with the crystal structures) allowed us to estimate which (E)/(Z)-isomers were most likely being observed in solution at room temperature by NMR spectroscopy. In addition, we were able to calculate transition structures corresponding to N-N bond rotational barriers of (E,Z)- and (Z,Z)-isomers of model molecules and rationalize the difference in coalescence temperatures between (E,Z)- and (Z,Z)-isomers. / Ph. Synthèse et caractérisation de composés fluorés pour le piégeage de fluorures gazeux / Synthesis and characterization of nanofluorides for the reactivity of gaseous fluorides

Ce travail porte sur la synthèse et la caractérisation de fluorures à base d’alcalins, d’alcalinoterreuxet de terres rares et sur la réactivité de ces composés vis-à-vis de ReF6 pour lapurification de UF6. L’étude traite dans un premier temps de la synthèse de fluorures tels queKMgF3, MgF2 et CaF2 en couplant une voie solvothermale assistée par micro-ondes et uneétape de fluoration à partir de fluor élémentaire (F2). Les techniques de caractérisationutilisées mettent notamment en avant le caractère basique au sens de Lewis de ces composéset la possibilité de contrôler leur surface spécifique et leur taux d’oxygène, à l’origine dessites basiques, via la température de fluoration (F2) en phase gaz. Dans un second temps, lasolution solide Ce1-xZrxF4 à base de Ce4+ et Zr4+ a été obtenue par fluoration directe d’oxydesmixtes à partir de fluor élémentaire dilué. L’étude par DRX et RMN du 19F montre laprésence d’une solution solide pour des compositions voisines de x=0,5. Enfin, la réactivité deReF6 et UF6 sur KMgF3, MgF2 et CaF2 montre que KMgF3 est un très bon candidat pour lapurification de UF6 vis-à-vis de ReF6, qui dépend à la fois de paramètres intrinsèques auxfluorures divisés (surface spécifique, taux d’oxygène, basicité, cations mis en jeu…) maisaussi du procédé de purification (température, temps de contact). / This work deals with the synthesis and characterization of alkali, alkali-earth and rare earthbased fluorides and the reactivity of the latter with ReF6 for UF6 purification. In a first part,we focused our attention on the synthesis of KMgF3, MgF2 and CaF2 high surface area metalfluorides coupling both microwave assisted solvothermal process and a fluorination step withelemental fluorine (F2). The higher the surface area, the higher the oxygen rate. Thesenanofluorides exhibit Lewis basic character. In a second part, several compositions of theCe1-xZrxF4 solid solution were synthesized by direct fluorination of mixed oxides usingelemental fluorine (F2). XRD an 19F NMR characterizations show the occurence of a newsolid solution for compositions close to x=0.5. Finally, the reactivity between ReF6/UF6 andKMgF3, MgF2 and CaF2 leads to conclude that KMgF3 is the best candidate for thepurification of UF6. This study indicates that the purification depends on several parameters :intrinsic parameters of the divided fluorides (surface area, oxygen rate, basicity, cations…)and parameters directly related to the process (temperature, contact time).

Mise en évidcence de la dégradation du liant ionomère dans les électrodes de pile à combustible / Evidence for degradation of ionomer binder in electrodes of fuel cell

Ce travail de thèse a pour but de suivre le comportement du liant ionomère après vieillissement en condition réelle d'utilisation de la pile. Dans un premier temps, diverses techniques de caractérisation en phase solide ont été utilisées afin d'étudier le ionomère présent dans les électrodes. La majeure partie de ces techniques se sont avérées insatisfaisantes pour le suivi du vieillissement du ionomère. Seule l'analyse par diffraction rayon X (DRX) a mis en avant un changement d'organisation structurale du ionomère dans les électrodes. Par la suite, nous avons choisi de caractériser le ionomère en solution après extraction par l'eau. Préalablement, un protocole de quantification en 2 à 3 étapes, dans lequel intervient une quantification via le rapport signal-sur-bruit (S/N), a été mis en place afin de quantifier le Nafion® et autres petites molécules fluorés. L'extraction Soxhlet a ensuite été réalisée sur les électrodes permettant de révéler la présence d'un produit de dégradation hydrosoluble après fonctionnement en pile, à la fois en cathode et en anode. Enfin, la macération des électrodes dans le diméthylacétamide (DMAc) a permis d'extraire le polymère Nafion® ainsi que deux acides : l'acide trifluoroacétique (TFA) et l'acide triflique (TFI). En conclusion, la corrélation de l'ensemble des observations nous a permis de proposer un mécanisme de dégradation du liant ionomère présent dans les électrodes. / The purpose of this study was to follow the behavior of ionomer binder after fuel cell operation. First, a series of techniques were used to investigate to characterize ionomer in electrode at solid state. Most of them were inefficient to study ionomer degradation. Only X-Ray Diffraction (XRD) pointed out a structural change of the binder in electrodes. Second, it has been decided to characterize ionomer in liquid state after water extraction. But first of all, a quantitative 19F NMR protocol composed of two to three steps, with a first step using a quantification through signal-to-noise ratio (S/N), was establish in order to quantify Nafion® and degradation products. Soxhlet extraction performed on electrodes allowed to detect a degradation product water-soluble. Finally, extraction with organic solvent (Dimethylacetamide) allowed to extract Nafion® and two acid: trifluoroacetic acid (TFA) and triflic acid (TFI) from electrodes. In conclusion, correlation between observation and literature allowed us to propose a degradation mechanism of ionomer in electrodes.

Glycoconjugates : synthesis and investigation of carbohydrate-protein interactions

To study the functions of glycoconjugates in biological systems reliable and efficient protocols for glycoconjugate synthesis are needed. To reach this goal we have developed methods for solid-phase synthesis of glycoconjugates that can be monitored with gel-phase 19F spectroscopy using fluorinated linkers, building blocks, and protecting groups. We have developed a new fluorine containing linker suitable for solid-phase synthesis of glycoconjugates. The linker was more acid-labile than similar linkers in order to enable cleavage under mild conditions of the target compound from the linker resin.  A carbamate-based strategy has been applied to attach a spacer carrying an amino group to a fluorinated Wang linker for synthesis of amino-functionalized glycoconjugates using thioglycoside donors with fluorinated protective groups. Cleavage from the solid support was performed with trifluoroacetic acid and subsequent protecting group removal gave the target compound. The terminal amine was conjugated with didecyl squarate and this derivative can be attached to various proteins and solid surfaces carrying primary or secondary amines. To evaluate this methodology we have immobilized glycoconjugates in amino-functionalized microtiter plates and successfully probed them with lectin. In addition, a novel fluorine containing protecting group has been designed, synthesized and evaluated. The protecting group was used for protection of the unreactive 4-OH in a galactose building block that was applied in the synthesis of 6-aminohexyl galabioside and was removed with TBAF in THF. Adenovirus serotype 8 (Ad8), Ad19, and Ad37 cause the severe ocular infection, epidemic keratoconjunctivities (EKC). During infection, the adenoviruses interact with sialic acid containing glycoconjugates on the epithelial cells via fiber structures extending from the viral particles. The virus particle most likely binds to the host cell in a multivalent way by simultaneously using multiple fiber proteins and binding sites. Multivalent sialic acid containing conjugates could efficiently inhibit Ad37 cell attachment and subsequent infection of human corneal epithelial (HCE) cells. Three compact tri- and tetravalent sialic acid conjugates were prepared and evaluated as inhibitors of adenoviral host cell attachment and subsequent infection and all conjugates were potent as anti-adenoviral agents. The conjugates can readily be synthesized from accessible starting materials. A crystal structure of the Ad37 fiber knob protein and the trivalent sialic acid conjugate showed that the three binding sites were all occupied by one sialic acid residue each.

Électrolytes solides fluorés pour batteries tout solide à ions F- / Fluoride solid electrolytes for Fluoride Ion Battery

Ce travail porte sur la synthèse, la mise en forme et la caractérisation desolutions solides de type tysonite RE1-xMxF3-x (RE = La, Sm, Ce et M = Ba, Ca, Sr). Dans unpremier temps, une démarche d’étude rigoureuse est mise en place pour la solution solide ditede référence, La1-xBaxF3-x. Les synthèses menées à l’état solide aboutissent à une maîtrise dela composition chimique et à l'établissement de lois de variations des paramètres structuraux.Une meilleure compréhension de l’influence de la structure sur la mobilité des ions F- estégalement acquise. L’influence du frittage dans l’obtention de bonnes valeurs de conductivitéionique est également à souligner. Dans un second temps, les effets de la nanostructurationpar mécanobroyage sur les propriétés de conductivité sont évalués. L’utilisation de laméthodologie des plans d’expériences mène à la mise au point des réglages optimums debroyage. Il apparaît alors que la synthèse des électrolytes peut être accélérée et mise àl’échelle tout en gardant des valeurs optimales de conductivité. Enfin, la démarche déterminéeest appliquée à d'autres solutions solides de type tysonite et à la recherche du conducteurionique le plus performant. Si les composés issus de la substitution Ce/Sr ou encore Sm/Casemblent les plus prometteurs, la plus grande stabilité chimique de la solution solide La1-xSrxF3-x est le compromis idéal pour l'utiliser comme électrolyte solide lors des mesuresélectrochimiques des batteries. / This work deals with the synthesis, shaping and characterization of RE1-xMxF3-x (RE = La, Sm, Ce et M = Ba, Ca, Sr) tysonite-type solid solutions. In a first part, onemeticulous approach has been set up for La1-xBaxF3-x solid solution, chosen as a reference.The solid-state synthesis of these materials led to a better knowledge of their chemicalcomposition (Vegard’s laws) and of the structure-ionic mobility correlations. The impact ofthe sintering process on the ionic conductivity is also highlighted. In a second part, the effectsof the nanostructuration conducted by ball-milling of the microcrystalline samples areevaluated. The use of the Design of Experiments methodology led to identify the optimummilling conditions. It appears that the synthesis of electrolytes can be sped- and scaled-up,while keeping high ionic conductivity properties. At last, this approach is applied on othertysonite-type solid solutions, to look for the best electrolyte. The Ce/Sr and Sm/Casubstitutions generate very promising ionic conductors but not really (electro)chemicallystable compounds. A compromise has been found with the choice of the La1-xSrxF3-x solidsolution as the FIB electrolyte for the electrochemical performances tests, regarding its higherchemical stability.

Une nouvelle famille de complexes de difluorure de bore photosensibles est développée. Elle est basée sur des structures moléculaires contenant une unité β-dicétone à conjugaison électronique π.La grande variété de groupements aromatiques et la nature donneur ou accepteur d'électrons des différents substituants permet l'élaboration de systèmes électroniques donneur-accepteur-donneur d'électrons (D1-A-D1) et donneur-accepteur (D2-A).L'absorption électronique de cette famille de molécule se situe dans la partie visible du spectre électromagnétique et une partie du spectre ultraviolet, et est caractérisée par une bande d'absorption π-π* intense avec des coefficients d'absorption molaire supérieurs à 50 000 M-1cm-1.L'émission de fluorescence couvre une plage spectrale qui va du visible au proche infrarouge avec des rendements quantiques de fluorescence en solution relativement élevés pouvant atteindre 62 %.En fin, cette famille de molécule est photochimiquement stable et est, contrairement à d'autres familles de complexes de difluorure de bore, chimiquement très stable en solution.Mots-clés : Difluorure de bore, β-dicétone, matériaux π-conjugués, luminescence, fluorescence stationnaire et résolue dans le temps (TRES), synthèse organique, RMN-19F dynamique, complexes & colorants fluorescents, curcumine & curcuminoide, complexe BF2, photophysique. / A new photosensitive family of boron difluoride complex is developed. It is based on π-conjugated molecular structures containing β-diketonates unit.The wide variety of aromatic groups and the nature of donor or electron acceptor of the different substituents allow the development of electron donor-acceptor-donor (DAD) and donor-acceptor (DA) electronic systems.The electronic absorption of this family of molecules is in the visible part of the electromagnetic spectrum and a portion of the ultraviolet spectrum, and is characterized by an intense π-π* absorption band with molar absorption coefficient greater than 50 000 M-1.cm-1.The fluorescence emission covers a spectral range going from visible to near infrared, with relatively high fluorescence quantum yields of up to 62 % in solution.This new material family is photochemically stable and, unlike some other families of boron difluoride complexes, chemically very stable in solution.
