Non-coding RNA in T cell activation and function

For a long time research has focused on the protein-coding mRNA, but there is a complex world of non-coding RNAs regulating the human body that we yet know very little about. Non-coding RNAs (ncRNAs) are involved in modulation of different cell processes including proliferation, differentiation and apoptosis. In the current study the role of ncRNAs in T cell activation and function was investigated. T cells are important mediators of immune responses, for example upon viral infections. The T helper cells (TH or CD4+ cells) are involved in orchestrating immune processes like aiding the activation of macrophages and enhancement of B cell function. The TH1 cell subtype is generally pro-inflammatory and IFNγ-secreting. There are regulatory T (Treg) cells that are involved in downregulation of TH1 cells, to decrease or terminate the immune response. It has been shown that upon repeated stimulation TH1 cells can switch into a Treg-like IL10-secreting anti-inflammatory phenotype. In the IL10-secreting Treg-like cells the microRNA 150 (miR-150) was found upregulated compared to IFNγ-secreting TH1 cells. Thus, miR-150 was believed to be a candidate in key regulation of the switch between the two phenotypes. Predicted target genes of miR-150 were identified using mRNA arrays investigating down-regulated genes in the IL10-secreting Treg-like subpopulation. In this thesis predicted targets of miR-150 were investigated using luciferase assays. Unfortunately no targets were identified. Upon isolation of IFNγ-secreting TH1 cells and Treg-like IL10-secreting cells, it was found that the ncRNA 886 (nc886) was upregulated in these activated cells, compared to resting TH cells. This indicates that nc886 has an important role in T cell activation. Nc886 has been shown to inhibit PKR activation in other cell types. The effect of nc886 on protein kinase R (PKR) was therefore investigated. PKR shuts down translation upon activation in response to viral double-stranded RNA or cellular stress. We showed that in an activated T cell phenotype nc886 is affecting PKR upon activation by dsRNA from HIV or synthetic origin. The PKR activation pattern is reversed in a resting T cell phenotype.

The absence of borated fertilization reduces the levels of leaf N in peanut / O objetivo do trabalho foi avaliar a produtividade, os componentes de produção e a nutrição do amendoim em razão da dose, época de fornecimento e parcelamento das doses de adubação boratada foliar na cultura do amendoim. O experimento foi conduzido entre os meses de novembro de 2010 a março de 2011 em um Latossolo Vermelho distroférrico, em Rubiácea, SP. Foram estudadas quatro doses de boro, com aplicações via foliar entre 30 e 75 dias após emergência (DAE), utilizando como fonte o ácido bórico (17% de B). A ausência de adubação boratada reduz os teores de N foliares no amendoim. A ausência de adubação boratada ou a aplicação de 0,5 kg ha-1 aplicada aos 30 DAE promovem os menores teores de B foliar no amendoim. Já a dose de 2,0 kg ha-1 aplicada integralmente aos 75 DAE ou 50% aos 30 DAE e 50% aos 75 DAE promovem os maiores teores de B foliar.

O amendoim (Arachis hypogaea L.) é uma das oleaginosas mais consumidas no mundo, e o Brasil apresenta potencial para despontar como grande produtor e exportador. Diferentes cultivares de amendoim foram desenvolvidas visando melhorar a produtividade, a qualidade nutricional e a vida de prateleira dos grãos e seus derivados. A bibliografia indica diferença na composição química e nutricional de cultivares obtidas por melhoramento genético. Este projeto teve como objetivo identificar cultivares de amendoim em produção e sua composição nutricional com vistas à alimentação humana, perfil nutricional e programas governamentais para incentivo ao consumo. As cultivares selecionados foram Granoleico (origem Argentina), IAC OL3, IAC 886, IAC 505 e IAC Tatu Vermelho (desenvolvidas no Brasil). Amostras dos cinco cultivares foram coletadas em cooperativas das principais regiões produtoras do estado de São Paulo, localizadas nos municípios de Tupã e Marília, representantes da região da Alta Paulista com solos mais claros e Jaboticabal e Sertãozinho, correspondentes à região da Alta Mogiana, com solos mais avermelhados. Estas cinco cultivares, produzidas no Estado de São Paulo, atingem 83% de área de cultivo nacional, que é responsável por 92% da produção do Brasil. As amostras foram analisadas quanto à composição centesimal, a presença de fatores antinutricionais (ácido fítico, taninos condensados, saponinas e atividade da urease), minerais (ferro, zinco, fósforo, cálcio, manganês e magnésio), perfil de ácidos graxos, tocoferóis totais e β-caroteno. A análise estatística foi realizada por Análise de Variância - ANOVA e aplicação do Teste de Tukey para comparação das médias. Os resultados observados revelaram variações de 18% de carboidratos, 9,7% de proteína, 28% de fibras e 8,2% de lipídios, com a cultivar IAC 505 apresentando as maiores porcentagens de lipídios e proteínas. Os minerais variaram 45% para Fe, 40% para Ca, 12% para Mg, 39% para Mn, 41% para P e 30% para Zn. Foram observadas diferenças nos teores de antinutricionais, constatando-se cultivares com teores de saponinas (Tatu Vermelho), ácido fítico (IAC OL3), taninos condensados (IAC 886) e atividade da urease (IAC 886), superiores às demais. Os níveis de tocoferóis totais e β-caroteno foram superiores nas cultivares Tatu Vermelho e Granoleico, respectivamente. Do ponto de vista de qualidade do perfil de ácidos graxos, a cultivar IAC 505 se destacou com 42% da composição de ácidos graxos monoinsaturados, 1,4 % de poli-insaturados, 6% de saturados e razão oleico/linoléico de 26. Foram elaboradas tabelas de comparação entre os resultados obtidos e as recomendações de ingestão dietética para verificação do suprimento de nutrientes. Com os resultados obtidos, pode se concluir que a composição química e nutricional de cultivares desenvolvidas pode ser afetada pelo processo de melhoramento genético, mas estudos complementares devem ser realizados. Há cultivares com qualidade nutricional superior às demais que suprem necessidades orgânicas e são mais interessantes para consumo de acordo com o objetivo nutricional. Estas deveriam ser inseridas em programas de incentivo à produção agrícola e o consumo incentivado devido a impacto benéfico no perfil nutricional de populações. / Peanut (Arachis hypogaea L.) is one of the most consumed oil in the world and Brazil has the potential to emerge as a major producer and exporter. Peanut different cultivars have been developed to improve the productivity, nutritional quality and shelf life of grain and its derivatives. The literature indicates difference in the chemical and nutritional composition of cultivars obtained by genetic improvement. This project aimed to identify peanut cultivars in production and its nutritional composition with a view to food, nutritional profile and government programs to boost consumption. The selected cultivars were Granoleico (Argentine origin), OL3 IAC, IAC 886, IAC 505 and IAC Red Tatu (developed in Brazil). Samples of five cultivars were collected in cooperatives of the main producing regions of São Paulo, in the cities of Tupa and Marilia, representatives of Alta Paulista region with light soils and Jaboticabal and Sertaozinho, corresponding to the region of Upper Mogiana with red soil. These five cultivars produced in the State of São Paulo, reaching 83% of cultivation area, which is responsible for 92% of production in Brazil. The samples were analyzed for chemical composition, the presence of anti-nutritional factors (phytic acid, tannins, saponins and activity of urease), minerals (iron, zinc, phosphorus, calcium, manganese and magnesium), fatty acid profile, total tocopherols and β-carotene. Statistical analysis was performed by analysis of variance - ANOVA and application of the Tukey test for comparison of means. The observed results showed variations of 18% carbohydrate, 9.7% protein, 28% fiber and 8.2% lipids, with IAC 505 having the highest percentage of lipids and proteins. Mineral varied to 45% Fe, 40% Ca, 12% Mg, 39% for Mn, 41% to 30% for P and Zn. Differences were observed in anti-nutritional content, confirming if cultivars with saponins content (Red Tatu), phytic acid (IAC OL3), condensed tannins (IAC 886) and urease activity (IAC 886), higher than the others. The levels of total tocopherols and β-carotene were higher in cultivars Tatu Vermelho and Granoleico respectively. The quality viewpoint of the fatty acid profile, the IAC 505 is highlighted with the composition 42% monounsaturated fatty acids, 1.4% from polyunsaturated, saturated and 6% oleic reason / linoleic 26. Were elaborated comparison tables between the results obtained and the dietary intake recommendations to check the supply of nutrients. With the results, it can be concluded that the chemical and nutritional composition of developed cultivars may be affected by genetic improvement, but further studies should be conducted. There are varieties with higher nutritional quality to other supplying organic needs and are more interesting for consumption according to the nutritional purpose. These should be included in the incentive programs for agricultural production and encouraged consumption due to beneficial impact on the nutritional profile of populations.

The reign of Leo VI (886-912) : personal relationships and political ideologies

Leo VI (886-912) is an emperor who has suffered from a hostile and inadequate press. He has been portrayed as a weak and careless emperor, known mainly for his dubious parentage and marital exploits. This thesis questions these popular perceptions of Leo, and attempts to present a more realistic account of the emperor and the politics of his age. The aspects of the reign tackled focus on essential elements of Leo's life and rule, presented in a rough chronological framework, and the themes of personal relationships and political ideologies are recurrent. Chapter One examines Leo's relationship with Basil I and his attitude to his Macedonian heritage. Chapter Two considers the fate of the monumental figure of Photios at the emperor's hands. Chapter Three deals with the position and role of the 'all powerful' Stylianos Zaoutzes during the first half of the reign. Chapter Four ponders the origin and meaning of Leo's 'wise' epithet. Chapter Five focuses on the emperor's four marriages. Chapter Six turns to the course of foreign affairs during the reign, concentrating on Bulgaria and the Arab navy, and considers the emperor's attitude towards these military problems. Chapter Seven examines the emperor's relationship with his senatorial officials, focusing on two distinct groups, eunuchs and the generals who originated from families of the eastern frontier. Finally Chapter Eight addresses the tense relationship that existed between Leo and his brother and co-emperor Alexander. What emerges from a consideration of these aspects of Leo and his reign is that this is an emperor who does not deserve the popular perceptions that still persist about him. He was an emperor who forged a 'new' and distinctive imperial style, a style that should not deceive us; he may have been literate, sedentary and city-based, but he was also forceful, strong-willed and conscientious.

Avaliação das características organolépticas de grãos e qualidade fisiológica de sementes em função do tempo de armazenamento em amendoim /

O presente trabalho foi realizado com o objetivo de avaliar as características organolépticas e qualidade fisiológica das sementes de três cultivares de amendoim IAC Tatu (60/70 - Peneira 19), Runner IAC 886, IAC -Tatu ST (70/80 - Peneira 17), nos períodos de 1, 6 e 12 meses de armazenamento. As avaliações para verificar a qualidade das sementes foram realizadas no Laboratório de Análise e Tecnologia de Sementes da UNESP - Ilha Solteira, através do teste de germinação, primeira contagem, envelhecimento acelerado, deterioração controlada, extração de proteína e óleo e as características organolépticas dos grãos. Os três genótipos foram armazenados em condições ambientais (±30°C e 70% UR) e em câmara seca (20°C/ 50% UR). Utilizou-se o programa SISVAR para realização das análises de variância, sendo que o delineamento utilizado foi o inteiramente casualizado com 4 repetições. Com base nos resultados foi possível interpretar que os cultivares não apresentaram diferenças na qualidade organoléptica dos grãos nos tempos 1, 6 e 12 meses de armazenamento em câmara seca a 20ºC / 50% UR, mas apresentaram diferenças na qualidade fisiológica dos grãos, concluindo assim que as sementes nestes tempos de armazenamento podem ser comercializadas para uso na alimentação mas não para a semeadura / Abstract: Deteriorations besides reducing the physiological quality of the seeds, it can also cause some alterations in the organoleptical and sanitary aspects. This way the aim of this research was to investigate the storage time influence over three varieties of peanut grain, IAC Tatu, IAC Runner 886, IAC-Tatu ST in a dry chamber (20°C/50%/UR). All analysis, such as germination test, first counting, accelerated aging, controlled deterioration and protein and oil extraction, were performed on the Laboratory of the Análise e Tecnologia de Sementes UNESP-Ilha Solteira, in three different time, 1, 6 and 12 months. SISVAR program was used in the accomplishment of variance analysis, it was used a experimental design interlay randomized with four repetitions. Through the analysis of obtained results was possible to infer that the cultivars showed no difference on grain organoleptical quality in all periods under investigation but showed differences on seed physiological quality. This way, was possible to conclude that seeds in all storage time investigated can be commercialized by its use in alimentation but can not be applied for the seeding / Mestre

Violences et souffrances en milieu hospitalier : le cas des infirmiers du gouvernorat de Tunis / Violence and suffering in hospitals : the case of the nurses of the governorate of Tunis

Des études récentes confirment que la violence au travail dans le secteur de la santé est universelle, malgré certaines différences locales, et qu’elle impacte aussi bien les hommes que les femmes, (Di Martino, 2002). Cette recherche, s’attache à décrire les perceptions de la violence et de la souffrance chez ces professionnels de santé, à travers l’évaluation de la perception des risques psychosociaux, par les infirmiers et ce, dans les services des urgences, et d’hospitalisation à Tunis.Il s’agit de souffrances délétères multiples, qui se teintent du stress, et du burn out, s’alimentent de l’anxiété, s’amplifient par un système organisationnel défaillant et une qualité relationnelle inconsistante Il s’agit d’une étude, descriptive, transversale, prospective, portant sur une population d’infirmiers, exerçant dans trois hôpitaux répartis sur des services d’urgence et des services d’hospitalisation. L’étude est divisée, en parties ; exploratoire, quantitative et qualitative. Le cadre de référence général, étant l’approche systémique, notamment par Brondenfenbrenner (1979), ainsi que ; dans le domaine de l’organisation, par Mintzberg et en communication par l’école Palo Alto…, associée, essentiellement, à L’approche transactionnelle, sur le stress et le coping ; ainsi que l’approche humaniste en sciences infirmières (en matière de la relation soignant-soigné). / Recent studies, confirm that workplace violence in the health sector is universal, despite some local differences. It affects both men and women. (Di Martino, 2002). This research attempts to describe the perceptions of violence and suffering within these health professionals through the evaluation of perception of psychosocial risks by nurses in the emergency and hospitalization departments in the governorate of Tunis. It is about multiple deleterious sufferings, which are tinged with stress, and burn out. Fed with anxiety, and amplified by a faulty organizational system, and an inconsistent relational quality. It is a descriptive cross-sectional prospective study of a nursing population working in three hospitals distributed over emergency and hospitalization departments. The study isdivided into exploratory, quantitative and qualitative parts. The general frame of reference, being the systemic approach, notably by Brondenfenbrenner (1979), as well as; in the field of organization, by Mintzberg, and in communication, by the school Palo Alto ..., associated essentially with the transactional approach, on stress and coping as well as the humanistic approach to nursing (Healer-healed relationship).
