Změny kvality cukrovky během vegetace a možnosti jejich ovlivnění

One of the aims of the dissertation thesis was to evaluate the dynamics of changes in the sugar beet quality during the growth and development with respect to different agro-ecological conditions on small-plot experiments in the years of 2010 and 2011. The possibilities of foliar nutrition application were proved on yield and technological parameters of sugar beet (digestion, soluble ash, alpha-amino nitrogen). Another objective was to assess the dynamics of yield and changes in the technological quality of sugar beet during the growing season in Central Moravia carried out in the cooperation with the sugar factory called Vrbátky. In the monitoring, the data were evaluated from the years of 2005 - 2011. The monitoring was carried out in the second half of sugar beet growth, as sugar factories normally prove in the country. The conditions for the growth and development of sugar beet and its technological quality were better in the year of 2010 than in 2011. The highest weight of tops was reached with the use of spray fertilizer called Magnitra L for the variants with within small plot experiments. The highest root weight showed sugar beet harvested after the application of Carbonbor K. The highest sugar content proved the tubers after the fertilization with Bortrac. The content of soluble ash was observed after the application of Magnitra L. Dangerous nitrogen content was low in all variants. Sugar beet was mature. In 2011, the highest leaf area of sugar beet was found out using the spray called Carbonbor K. The highest root weight was determined in the variant with Carbonbor K spray. The highest sugar content proved the tubers after the fertilization of Carbonbor K and Carbonbor Na. The content of soluble ash and harmful nitrogen was higher than in 2011. In the monitoring, the following dependences were proved: Weight tops decreased during the vegetation. On the average of the years, a weight lowered from 980 g/plant to 450 g/plant. The root reached from 640 to 900 g /plant as well as sugar content increased from 14, 26 to 15.8 %. Significant differences were found out between the years.

Hur påverkas en fonds prestation av dess förmögenhet? : en studie av svenska fonder under perioden 2003-2008

<p>Uppsatsen undersöker om fonders prestation beror på dess förmögenhet. Syftet är att se om små fonder presterar bättre än stora (sett till riskjusterad prestation) och således är att föredra. Undersökningen innehåller 21 stycken svenska fonder med inriktning på nationella placeringar över perioden 2003-2008. Som mått på prestation har den historiska sharpekvoten och Jensens alfa använts. Undersökning har gjorts på paneldata, tidseriedata och tvärsnittsdata över olika tidsperioder och tidsperspektiv. I resultaten påträffades ett signifikant negativt samband mellan prestation och fondförmögenhet, dvs. fonder presterade sämre ju större de blev. Det påträffades dock inget samband i skillnaden i hur små fonder presterat gentemot större fonder. En förklaring till detta kan vara att stora fonder gynnas av att ha bättre förvaltare medan små fonder gynnas av sin storlek och att dessa fördelar tar ut varandra.</p>

Numerical modeling of bronze coating formation using concentration dependent diffusivity

The engineering of a component’s surface is of remarkable importance, it allows applications that surpass what is achievable by the substrate alone by improving surface properties such as hardness, wear and corrosion resistance and even aesthetic value. This project focuses on obtaining bronze coatings through the diffusion of electrodeposited tin on copper via heat treatment. Specifically, the single phase α is targeted due to its attractive properties. Determining the correct heat treatment parameters is crucial since only a specific range of compositions lead to the formation of the desired microstructure and, consequently, properties. The diffusion of a thin coating of tin onto copper often leads to the formation of undesirable features: small craters containing different phases than the one targeted. In the present work, the diffusion behavior of tin-copper couple was investigated; a numerical model was developed to predict the composition profile after heat treatment, which agrees with experimental observations; and the mechanism for the formation of undesirable surface craters during heat treatment was studied and the probable solution proposed. / February 2017

Petra Matoušková, Ph.D. Title of diploma thesis: Effect of alpha-humulene on adhesion of breast cancer cell line MDA-MB-231 α-humulene is a sesquiterpene contained in the essential oil of Chinese Bayberry (Myrica rubra), which has various biological effects. The aim of this thesis was to study the cell adhesion of tumor cell line MDA-MB-231, and the effects of α-humulene on expression of cell adhesion molecules. Furthermore, the cytotoxic effect of α-humulene on this cell line was verified. Effect of α-humulene on cell proliferation was evaluated by neutral red uptake test (NRU test) and by xCelligence system. Cell adhesion was also continuously monitored by xCelligence. Expression ganges of cell adhesion molecules upon α-humulene treatment were determined by Western blotting and by quantitative polymerase chain reaction on the protein and mRNA levels, respectively. The proliferation of the cells was significantly affected by α-humulene after 24 hour treatment (IC50 13.6 µg/ml). α-humulene alone caused slightly increased cell adhesion. Foi produzida a proteína recombinante do domínio extracelular da subunidade 2 do receptor do IFN-alfa em forma de proteína de fusão com a glutatione-S-transferase (GST-IFNAR2cEC) em E. coli, com o objetivo de avaliar seu efeito bloqueador sobre a ação do IFN-alfa. A GST-IFNAR2cEC de forma solúvel, com peso molecular 54 kDa, foi obtida quando a proteína recombinante foi expressa a 25 graus C por indução de IPTG e foi lisada por French Press. A avaliação do efeito bloqueador da GST-IFNAR2cEC foi feita utilizando-se 3 métodos: 1) Ensaio antiproliferativo com células Daudi, 2) ensaio antiviral com células Hep 2/c e vírus encefalomiocarditis, e 3) expressão gênica semi-quantitativa do STAT 1 (transdutor de sinal e ativador de transcrição) e da 2',5'-oligoadenilato sintetase (OAS) pela RT-PCR duplex nas células Daudi e Hep 2/c. A GST-IFNAR2cEC, mesmo a 420 nM, não inibiu as atividades antiproliferativa e antiviral de 24,5 pM e 49 pM de rIFN-alfa2 provavelmente devido à sua ineficiência na competição com o IFNAR da superfície celular durante o longo período de incubação nos ensaios realizados. A expressão gênica do STAT 1 não foi inibida pela GST-IFNAR2cEC nas células Daudi e Hep 2/c, porém inibiu-se a da 2’, 5’-OAS. Estes resultados sugerem que a expressão gênica do STAT 1 é independente da ligação do IFN-alfa com a IFNAR 2c, e que a da 2’, 5’-OAS é dependente dessa ligação nessas células

Hypophosphastasia (HPP): pathogenesis and current treatment options

Hypophosphatasia (HPP) is a rare, inherited, metabolic bone disease characterized by the abnormal development of bones and teeth, due to defective mineralization (1,2). HPP is caused by inactivating mutation(s) within the gene that encodes the tissue-nonspecific isozyme of alkaline phosphatase (TNSALP) (1). These mutations lead to impaired bone mineralization leading to various skeletal abnormalities such as rickets and fractures, as well as other systemic complications including respiratory manifestations, seizures, dental anomalies, pain, and nephrocalcinosis (1–4). Historically, management of HPP focused solely on supportive therapy in an attempt to minimize disease-related symptoms (1,2). However, in 2015, the Food and Drug Administration (FDA) approved Strensiq® (asfotase alfa), an enzyme replacement therapy approved for the treatment of patients with perinatal/infantile- and juvenile-onset HPP (3,5). This paper will provide an overview of HPP and current and past treatment options for HPP, specially analyzing the effectiveness of the newest enzyme-replacement therapy, Strensiq®, which will be more effective than previously used therapies in preventing the adverse clinical outcomes of HPP.

Impacto do tratamento do Helicobacter pylori na ansiedade e depressão em pacientes dispépticos funcionais

Background – A associação da infecção pela bactéria Helicobacter pylori com algumas doenças gástricas, como por exemplo, a úlcera gástrica e o câncer gástrico, é bem definida. No entanto, a associação da bactéria com outras doenças, especialmente extragástricas não está estabelecida. Métodos – Foi realizada avaliação da ansiedade e depressão e pesquisa do Helicobacter pylori com endoscopia em participantes de um estudo que avaliou os efeitos da erradicação do Helicobacter pylori sobre a dispepsia funcional. Pacientes com infecção por Helicobacter pylori dispepsia funcional foram randomizados para receber omeprazol, amoxicilina e claritromicina (grupo antibiótico) ou omeprazol mais placebo (grupo controle) por 10 dias. Os pacientes foram avaliados na baseline e 12 meses após. O principal desfecho do estudo foi avaliar a mudança no status de ansiedade e depressão após 12 meses nos dois grupos. Resultados – Foram incluídos 234 pacientes, 115 no grupo antibiótico e 119 no grupo controle. As informações do Hospital Anxiety and Depression Scale e pesquisa da bactéria foram obtidas de 213 pacientes (91,03%) na visita de 12 meses. Com respeito à avaliação da ansiedade após 12 meses os resultados foram: no grupo antibiótico 66,3%(69/104) não modificaram o status da ansiedade, 13,5% (14/104) que não apresentavam passaram a apresentar ansiedade, 20,2%(21/104) que apresentavam ansiedade passaram a não apresentar; no grupo controle 70,6% (77/109), 7,3% (8/109) e 22,0% (24/109) respectivamente. Resultando em um valor de P de 0,340. Quanto a depressão após 12 meses os resultados foram: no grupo antibiótico 71,2%(74/104) não modificaram o status da depressão, 18,3% (19/104) que não apresentavam passaram a apresentar depressão, 10,6%(11/104) que apresentavam depressão passaram a não apresentar; no grupo controle 69,7% (76/109), 10,1% (11/109) e 20,2% (22/109) respectivamente. Resultando em um valor de P de 0,057. Conclusão – Não foi observada relação estatística significativa entre o tratamento do Helicobacter pylori e a mudança de status de ansiedade e depressão nos sujeitos. / Background – The association of infection due to Helicobacter pylori bacteria with some gastric diseases, such as gastric ulcer and gastric cancer, is well defined. However, the association of the bacterium with other diseases, especially extra gastric ones, is not. Methods – We evaluated anxiety and depression and the presence of Helicobacter pylori through endoscopy, in subjects who participated in a study assessing the effects of Helicobacter pylori eradication, in functional dyspepsia. Subjects with Helicobacter pylori infection, suffering functional dyspepsia, were randomized to receive omeprazol, amoxicillin and claritromicin (antibiotic group) or omeprazol plus placebos (control group) during 10 days. Subjects were evaluated at baseline and after 12 months. The main outcome of the study was to assess the anxiety and depression change of status, after 12 months, in both groups. Results – We included 234 subjects: 115 in the antibiotic group and 119 in the control group. We obtained HADS information and bacteria research from 213 subjects (91.03%) at the 12 months visit. Regarding the 12-month anxiety assessment the results were: 66.3% (69/104) did not modify the anxiety status, 13.5% (14/104) who did not present previous signs of anxiety began showing signs of anxiety and 20.2% (21/104) who presented anxiety, ceased to present the condition in the antibiotic group; in the control group the results was 70.6% (77/109), 7.3% (8/109) and 22.0% (24/109) respectively, resulting in a P value of 0,340. As for depression, after 12 months, the results were: 71.2% (74/104) did not modify the depression status, 18.3% (19/104) who did not present previous signs of anxiety began showing signs of depression and 10.6% (11/104) who presented depression, ceased to present the condition in the antibiotic group; in the control group the results was 69.7% (76/109), 10.1% (11/109) and 20.2% (22/109) respectively, resulting in a P value of 0,057. Conclusion – No significant statistical value was observed between the treatment for Helicobacter pylori and the changing status of anxiety and depression in the subjects.

Dentre as formas de tratamento da obesidade, a reeducação alimentar, atividade f´ısica regular e psicoterapia são as mais recomendadas. O presente projeto de pesquisa tem como objetivo o planejamento, síntese e caracterização de novos candidatos a fárrmacos com padrão estrutural ftalimídico para o tratamento da obesidade, por meio da inibição da enzima sn-1-Diacilglicerol Lipase α (DAGL-α), esta enzima é responsável pela produção do sinalizador lipídico 2-araquidonoilglicerol (2-AG), que é diretamente responsável pela modulação de receptores canabinoides do subtipo CB1. Nota-se em pacientes obesos elevada quantidade de 2-AG, dado que sugere uma possével explicação para o descontrole da saciedade alimentar, além disso, observa-se que modulações geradas por antagonistas ao receptor CB1 foram capazes de reduzir a resistência à insulina e a obesidade, todavia, efeitos colaterais como: depressão profunda e descontrole emocional foram relatados. Nesta perspectiva, a estratégia adotada neste trabalho consistiu em inibir a enzima que catalisa a síntese de 2-AG, para isso, foi utilizado o planejamento racional de fármacos adotando-se o ligante sintetizado pelo grupo de pesquisa da universidade de Leiden, ao qual foi relatada atividade biológica promissora frente à enzima DAGL-α. No presente projeto de pesquisa foi possível desenvolver uma rota sintética de quatro etapas, e com isso, sintetizar 12 derivados ftalimídicos que foram enviados para a universidade de Leiden em colaboração científica com o professor Dr. Mário Van der Stelt para execução dos testes de inibição enzimática. Dentro do contexto do CIBFar foram realizados testes biológicos paralelos perante as doenças negligenciadas e o câncer. Os resultados para o derivado CYOJ4 foram promissores para a malária e câncer de mama, proporcionando novas perspectivas para futuros trabalhos. / Obesity has been highlighted as an inflammatory process, this association has been evidenced by visceral adipose tissue. The physiologic response of the inflammation promotes problems in the homeostatic control of food intake. In addition, obesity and metabolic syndrome are capable of triggering a group of disease. Here, we promote the planning, synthesis, and characterization of a novels drug candidates with a phthalimide structural standard for the treatment of obesity, by inhibiting the enzyme sn-1-Diacilglicerol Lipase α (DAGL-α). This protein is responsible for the production of the 2-arachidonoylglycerol a lipid marker (2-AG), which is directly associated with the modulation of cannabinoid receptors called CB1. In obese patients there is a high plasma level of 2-AG, it suggests a possible explanation for high food intake, in addition, has been observed that modulations generated by antagonists to the CB1 receptor were able to reduce insulin resistance and obesity, however, side effects such as profound depression and change in emotional responses were reported. In this perspective, the strategy used in this project was to inhibit the enzyme that catalyzes the synthesis of 2-AG, for this, the rational drug planning has adopted with the ligand synthesized by the research group of the University of Leiden, which show a biological activity promising against the DAGL-α enzyme. In this research project has been possible to develop a synthetic route of four steps, and synthesize 12 phthalimide derivatives which were sent to the university of Leiden in scientific collaboration with the professor Dr. Mario Van Der Stelt to carry out the tests of enzymatic inhibition. Within the context of CIBFar, parallel biological tests have been carried out in the tropical diseases and cancer. The results were promising for malaria and breast cancer, providing novel perspectives for future projects. / CNPq: 132099/2015-2
