Mechanistic Studies and Inhibition of N-hydroxylating Monooxygenases

N-hydroxylating monooxygenases (NMO) are members the class B flavoprotein monooxygenases. They catalyze the N-hydroxylation of lysine and ornithine and play and essential role in the biosynthesis of hydroxamate containing siderophores. Siderophores are high affinity iron-chelators composed of catechol and hydroxamate functional groups that are synthesized and secreted by several microorganisms and plants. It has been showed that many NMOs are essential for virulence in many opportunistic pathogens such as Aspergillus fumigatus and Pseudomonas aeruginosa. The focus of my research is on the N-hydroxylating enzymes: Siderophore A (SidA) from Aspergillus fumigatus and Amycolatoposis alba monooxygenase (AMO). One of my projects is focusing on identifying inhibitors of SidA that will ultimately block the siderophore biosynthesis in A. fumigatus. Out of 973 compounds screened using an activity high-throughput assays two compounds were identified. These were, wortmannin a steroid metabolite and ebselen a benzoselenazole as SidA inhibitors with IC50 values of 369 µM and 11 µM respectively. A second part of this works investigates the hydroxamate formation of the siderophore albachelin in Amycolatoposis alba with the purpose of better understanding this class of enzymes and their catalytic mechanism. The enzyme was purified and characterized in its holo (FAD-bound) and apo (unbound) forms. Pre-steady and steady state kinetics shows that the two forms have different coenzyme preference; apo-AMO prefers NADH while holo-AMO has a higher affinity to NADPH. / Master of Science in Life Sciences

The black box of human resources: The effect of high performance work systems on organizational commitment, work engagement and intention to leave

Employee attrition is a critical issue for organizations as retaining the best professional talent and controlling the costs linked to losing and employing new employees is a costly process. This is especially true for small and medium-sized enterprises that have limited resources and where every individual employee is a big part of the collective effort of the company. It has been argued that in most organizations the utility of human resources is not maximized and that investments in the right human resource practices would provide returns that exceed the costs. The unclear relationship between human resource practices and performance is commonly referred to as ‘the black box' of human resource management. Human resource practices are conceptualized differently in the literature and one of the most popular views states that practices should support and synergize with each other and that a combined implementation of practices leads to better results. This concept is often referred to as high performance work systems (HPWS) which is designed to enhance employee effectiveness. Based on the research of HPWS, the AMO-framework was introduced to better explain the linkage between human resources management and organizational performance. The framework is based on the idea that employees will perform better when they are able to do so (Ability), when they possess the motivation to do so (Motivation), and when their work environment provides the necessary support (Opportunity). Due to the lack of research of the AMO-framework in Scandinavian countries, the purpose of this quantitative study is to tests the relationship between the AMO-framework, intention to leave and the two constructs of organizational commitment and work engagement. Based on the purpose, a research question for this study is addressed as follows: What is the impact of Ability, Motivation and Opportunity HR-practices on organizational commitment, work engagement and employees' intention to leave? The sample consisted of a total of 95 employees from various small and medium-sized enterprises in Sweden. The empirical findings suggest that even though the AMO-framework does not impact employees' intention to leave directly, the individual dimensions of Ability and Motivation might do so indirectly through the constructs of organizational commitment and work engagement. These findings highlight the importance of the impact of the AMO framework on the above-mentioned constructs. Furthermore, this study contributes to the studies on human resource management in SMEs by providing more insight for the owner-managers on how to wisely invest in human resources in order to decrease employee attrition.

Para distingui-los dessa suposta natureza e caracterizá- los como objeto da Ciência, separou língua e fala. Desde Saussure, então, a ideia de estrutura passa a influenciar diversos pensadores, em especial a Lévi-Strauss que, em As Estruturas Elementares do Parentesco (1949), afirma a existência dos sistemas de signos como objeto da Antropologia. Essa temática, qual seja, a do signo, bem como suas implicações sobre o Direito, são objeto da primeita parte da dissertação. A ênfase dada pelo Antropólogo à função do significante sobre o significado e o conceito de mitema, tomado como extensão ao de mito, mostraram-se fundamentais às construções teóricas de Lacan, notadamente quanto à função simbólica (nomes-do-pai ou lei) como constitutiva do sujeito e ao conceito de significante (como o que representa o sujeito para outro significante). Desse arcabouço, aliado ao retorno a Freud e aos dados colhidos na clínica, concebe o sujeito enredado ou atado a um discurso estruturado pelas categorias real-simbólico-imaginário. Assim, em seminário proferido na Faculdade de Direito, já que, no seu próprio dizer, os lugares falam, interpreta o direito como o discurso que, por excelência, estrutura o real. Todavia, não como qualquer discurso ou discurso diferenciado por seus enunciados, mas direito como discurso do mestre, avesso ao discurso do analista, pois, se as relações fundamentais são a de um significante com outro significante (S1 a S2) de onde resulta o sujeito ($), no direito, se trata de uma série discursiva caracterizada como efeito de verdade; quer dizer, o mestre, assim como Édipo, enuncia a verdade na ignorância de que ela só pode ser um semidizer. Para o mestre, tudo o que existe deve submissão à Lei (no lugar do desejo), já que ele mesmo se sujeitou à castração (renunciou ao “gozo absoluto”). Legendre descobre esse discurso no Ocidente como estrutura vazia (sem corpo e sem fala) e, ao mesmo tempo, voraz (preenchida com textos). Uma ficção, porém não menos atuante e eficaz que outras ficções. Para o Jurista, a lei equivale à instituição, e instituir é igual a escrever. Assim, nas instituições, a verdade está instituída em um texto. E, para que um texto funcione, basta acreditar nele. Portanto, no sistema jurídico (a instituição), além da obediência à norma, que no imaginário opera a crença de amor ou lógica de submissão, joga-se, também, com dois princípios: o da autoridade e o da razão. Esse último aparece miticamente confundido com o primeiro, isto é, com o Outro. Dessa forma, a mudança (na instituição) consiste na mudança de nada. Consiste apenas em deslocar ou agregar (textos), nunca substituir. Essa é a dimensão dogmática: “Evitar el cambio”. Assim, a teoria Lacaniana, bem como suas possíveis implicações no campo do direito, são o objeto da segunda parte desta dissertação. Ao final, desde o caminho percorrido pelo significante (De Saussure a Lacan), conclui-se, diferentemente do suposto pelo título do trabalho, que direito é ficção: primeiro, porque se estrutura como discurso e todo o discurso é morte da coisa, embora não tudo nas palavras; segundo, porque se organiza sobre a questão da verdade, e não há verdade externa à ficção; terceiro, porque produz o efeito-ficção, ou seja, se constitui como um Texto sem sujeito, que mesmo assim fala; quarto, porque nele a linguagem cumpre plenamente o seu papel de equívoco; quinto, porque está no lugar de um grande Outro no qual o homem crê, pois, simplesmente, a condição humana supõe uma entrada imaginária na vida; e, por fim, porque nele se confundem o princípio de autoridade com o princípio de racionalidade. / Saussure propuso como objeto de la Lingüística el resultante de un proceso teórico que funcionaria como un modelo capaz de dar cuenta del sistema de significación- estructura, en la cual, la alteración de uno de los elementos implica el cambio de la totalidad - subyacente a la apariencia empírica de los signos. Legendre descubre ese discurso en Occidente como estructura vacía (sin cuerpo y sin palabra) y, al mismo tiempo, voraz (llena de textos). Así, en las Instituciones, la verdad está instituida en un texto. Y, para que un texto funcione, basta creer en él. Por lo tanto, en el sistema jurídico (la institución), más allá de la obediencia a la norma, que en el imaginario opera la creencia del amor o la lógica de la sumisión, se juega también, con dos principios: el de autoridad y el de la razón. Ese último aparece míticamente confundido con el primero, es decir, con el Otro. De esta forma, la transformación (en la institución) consiste en la transformación de la nada. Consiste apenas en desplazar o agregar (textos), nunca substituir. Esa es la dimensión dogmática: “Evitar el cambio”. Así, la teoría Lacaniana, tanto como sus posibles implicaciones en el campo del derecho, son el objeto de la segunda parte de esta disertación. Al final, desde el camino recorrido por el significante (De Saussure a Lacan), se puede concluir, diferente del supuesto por el título de este trabajo, que Derecho es ficción: primero, porque se estructura como discurso y todo el discurso es muerte de la cosa; sin embargo, no todo está en las palabras; Segundo, porque se organiza sobre a la cuestión de la verdad, y no hay verdad externa a la ficción. Y tercero, porque produce el efecto-ficción, o sea, se constituye como un Texto sin sujeto, que además habla; cuarto, porque en él el lenguaje cumple plenamente su papel de equívoco; quinto, porque está en el lugar del Otro con mayúscula, en el cual el hombre cree, porque simplemente la condición humana supone una entrada imaginaria en la vida; y por final, porque en él se confunden el principio de autoridad con el principio de racionalidad.

Quantum Control and Quantum Chaos in Atomic Spin Systems

Laser-cooled atoms offer an excellent platform for testing new ideas of quantum control and measurement. I will discuss experiments where we use light and magnetic fields to drive and monitor non-trivial quantum dynamics of a large spin-angular momentum associated with an atomic hyperfine ground state. We can design Hamiltonians to generate arbitrary spin states and perform a full quantum state reconstruction of the results. We have implemented and verified time optimal controls to generate a broad variety of spin states, including spin-squeezed states useful for metrology. Yields achieved are of the range 0.8-0.9.We present a first experimental demonstration of the quantum kicked top, a popular paradigm for quantum and classical chaos. We make `movies' of the evolving quantum state which provides a direct observation of phase space dynamics of this system. The spin dynamics seen in the experiment includes dynamical tunneling between regular islands, rapid spreading of states throughout the chaotic sea, and surprisingly robust signatures of classical phase space structures. Our data show differences between regular and chaotic dynamics in the sensitivity to perturbations of the quantum kicked top Hamiltonian and in the average electron-nuclear spin entanglement during the first 40 kicks. The difference, while clear, is modest due to the small size of the spin.

Springboard, Parachute, and Sprint : How Emerging Market Multinational Enterprise Can Manage Cultural Distance and Recruit Top Talent in Advanced Markets

The purpose of this thesis is to analyze how emerging market multinational enterprises (EMNEs) can effectively bridge the cultural gap between their home market operations and internationalized operations in advanced markets like Sweden, particularly with regards to adapting recruitment strategies to secure top talent in the new business context. The authors aim to expand the existing ‘springboard framework’ for internationalization by multinational enterprises originating in emerging market countries which are increasingly reshaping the global competitive landscape. The study interviewed seven managers with extensive experience working in both advanced and emerging markets, and five top talent from two of the highest ranked Swedish business schools. The study found that for EMNEs cultural distance is a secondary consideration when expanding to advanced markets and their main focus is showcasing their value proposition to clients, customers, and top talent alike. From the experience of the executives interviewed for this thesis, top talent prioritizes organizational and managerial values that a company imbibes and the career progression that it provides over remunerations. This was confirmed by the top talent we interviewed who repeatedly spoke about how important career opportunities and organizational values were for them. The findings from our thesis contribute to expanding the field of cross- cultural management theory by supplying a qualitative study of EMNE adaptation for internationalization. They also contribute to the recruitment literature by demonstrating how EMNEs have to adapt their HRM systems to the local environments in order to gain the attention of and acquire the local top talent.

Examining the Impact of Human Resource Management (HRM) on Telework Participation Among US Federal Employees

This dissertation investigates telework participation within the US federal government, structured through three interconnected articles from a human resource management perspective. It sheds light on the factors critical for the successful implementation of telework including human resource management (HRM) practices, differences among generational cohorts, and the role of line managers. Chapter 1 presents the motivation for this dissertation, theoretical foundations, an overview of telework within the US federal government, and a summary of the three research studies conducted. Chapter 2 introduces the first manuscript, which examines the impact of HRM practices on telework participation among US federal employees using the Ability, Motivation, and Opportunity (AMO) framework. This study analyzes data from the 2015 Federal Employee Viewpoint Survey (FEVS) through multinomial logistic regression. The results highlight that skills enhancement, promotional opportunities, and cooperative work environments significantly correlate with the likelihood of telework participation. Chapter 3, the second manuscript, applies generational cohort theory to investigate differences in telework participation among generational groups. Utilizing multinomial logistic regression analysis on FEVS data from 2013, 2014, and 2015, the study finds varied preferences for telework across Millennials, Generation X, Late Baby Boomers, and Early Baby Boomers. Generation X shows the most likelihood of telework participation compared to Millennials and Baby Boomers, with Early Baby Boomers displaying the least inclination. Chapter 4, the third manuscript, assesses the role of line managers as HRM partners and facilitators in telework participation. Applying Leader Member Exchange (LMX) theory and 2015 FEVS data, the study finds that the quality of relationships between line managers and employees -marked by mutual trust, support, and respect – enhances the likelihood of telework participation. Chapter 5 concludes this dissertation with summaries of research findings, theoretical and practical implications, research limitations, and avenues for future research directions. / Doctor of Philosophy / This dissertation investigates telework challenges within the US federal government, focusing on key factors that contribute to successful telework implementation. It consists of three interconnected studies, each addressing different aspects of telework from a human resource management perspective. The first study examines how human resource management (HRM) practices influence telework participation among federal employees. It found that skills enhancement, fair promotion opportunities, and cooperative work environments significantly increased the likelihood of teleworking. These findings suggest that HRM systems need to be adaptive and supportive through the alignment of practices with telework goals to ensure effective implementation. The second study explores generational differences in telework participation. The research showed that Generation X employees are the most likely to participate in telework, followed by Millennials and Late Baby Boomers, while Early Baby Boomers are the least likely. These findings underscore the importance of tailoring telework policies to the preferences of different generational groups. The third study focuses on the role of line managers in supporting telework. It demonstrated that strong relationships between employees and their supervisors, characterized by mutual trust, support, and respect, significantly enhance telework participation. This highlights the critical role of managerial support in successful telework programs. The dissertation concludes with a summary of research findings, practical implications for managing telework in the federal government, and recommendations for future research. These insights can help public sector organizations develop effective telework programs that meet the diverse needs of their employees.

Safety Management Systems (SMS) for aircraft manufacturers and maintainers?

There is much dialogue in the global aviation industry about Safety Management Systems (SMS) and how it should be integrated across all domains of the industry including aircraft design, production, flight operations, overhaul and maintenance, suppliers, service providers, airports, and so forth (Johnson, 2012). Regulators have made significant progress in recent years to implement ICAO’s SMS into airlines, albeit as a required or recommended practice. More recently the regulators are seeking to implement SMS into the aircraft manufacturing and aircraft maintenance domains. This research reviewed regulatory publications from multiple countries to assess the technical makeup of SMS, and understand what regulators are requiring, or recommending, and when. It was found that global regulators accept the ICAO published definition of SMS, but different regulators have varying approaches regarding implementation. However, they are consistent in initially targeting airlines for SMS implementation. SMS comments range from “The best thing since sliced bread” to “Worst thing since the creation of the FAA; I don’t need anyone telling me what’s safe when I already know it; waste of time and money”. This investigation experimented with field tests to connect the engineering, production and airline domains into one ICAO SMS model. Results indicate that because the different domains are risk-specific, the application of one safety risk management model to all domains is not viable. The SMS model applies to airlines because airlines’ primary risk is about operational safety. Aircraft production and maintenance is about production risk – therefore the risk model must be centric to process risk. Field test 3 tailored the ICAO SMS risk architecture to assess and mitigate process risk as applicable to the aircraft manufacturing and maintenance. Although the SMS architecture was usable, the content and focus was significantly adjusted to be production process-risk centric, to the point where the term “SMS’ was deemed out of place. The resulting model was therefore named Production Risk Management System (PRMS). Following the emergence of PRMS from field tests, this investigation reviewed industry, research and regulatory arguments for and against SMS in the airline industry, and correlated those arguments with the benefits and non-benefits of PRMS for the manufacturing and aircraft maintenance domains. The researcher advocates PRMS as a viable model that meets ICAO SMS-like architecture for aircraft production and maintenance. Methods were identified for developing and implementing PRMS, and for evaluating its ROI. If and when “SMS” is truly mandated in these domains, the researcher proposes PRMS as a viable model that should be considered. Furthermore, the researcher proposes that PRMS can be an effective production risk management system that can enhance the organization’s existing QMS, regardless of “SMS” regulations.

Progress towards a new parity non-conservation measurement in cesium-133

Atomic parity violation measurements provide a way to probe physics beyond the Standard Model. They can provide constraints on conjectures of a massive Z′ bosonor a light boson, or searches of dark energy. Using the two-pathway coherent control technique, our group plans to make a new measurement of the weak interaction induced parity non-conservation (PNC) transition moment (<i>E<sub>PNC</sub></i>) on the cesium 6S→7S transition. We will coherently interfere a 2-photon transition with the Stark and PNC transitions to amplify and extract the PNC amplitude. Previously, our lab has measured the magnetic dipole transition moment on the same 6S→7S transition to about 0.4% uncertainty using this technique. In this dissertation, I discuss improvements made to the system, and review what future upgrades are needed for a new<i> E</i><sub><i>PNC </i></sub>measurement. Key systematics are also described. For an accurate determination of <i>E<sub>PNC</sub></i>, properties of cesium such as the scalar (<i>α</i>) and vector (<i>β</i>) transition polarizabilities are needed. I present improved determinations of keyelectric dipole matrix elements, and calculate new high precision determinations of <i>α</i> and <i>β</i>. Finally, using <i>β</i> and the previously measured value of <i>E<sub>PNC</sub>/β</i>, I calculate new values for the weak charge of the cesium nucleus Q<sub>w</sub>.<br>

Variabilidade do sistema de monções de verão durante os últimos 1500 anos na região de Bonito-MS com base em registros paleoclimáticos de espeleotemas

Utilizando registros isotópicos (\'delta\' \'POT.18\'O e \'delta\'\'POT.13\'C) de alta resolução, razões de elementos-traço em relação ao cálcio e taxas de crescimento de espeleotemas, precisamente datados pelo método U-Th, foi feita uma reconstrução da intensidade do Sistema de Monções Sul-Americano (SMSA) nos últimos 1500 anos na região do município de Bonito, Mato Grosso do Sul. O estudo do sinal climático, interpretado por meio de variações geoquímicas dos espeleotemas, foi realizado com base em estudos e comparações com dados instr umentais de uma estação meteorológica de Bonito, com dados de monitoramentos do IAEA - GNIP de Campo Grande e Cuiabá e com informações de modelos computacionais que simulam a variação de \'delta\'\'POT.18\'O em relação à intensidade das monções na América do Sul, o qual e stá relacionado com variação regional de pluviosidade. Apesar destes dados mostrarem que o amount effect é um dos fatores determinantes na variação das razões isotópicas dos espeleotemas, outros fatores como o degree of rainout upstream e a fonte de umidade (Amazônia x oceano Atlântico sul) também devem ser considerados importantes. Assim, a interpretação das razões de \'delta\'\'POT.18\'O é atribuída à intensidade do sistema de monções e, desta forma, à atuação da Zona de Convergência do Atlântico Sul (ZACS), principalmente associada à propagação de chuvas da região Amazônica para o centro-oeste e sudeste brasileiro. O registro paleoclimático de Bonito mostrou excelente coerência com variados tipos de registros de outros locais da América do Sul, e também com dados do Hemisfério Norte, evidenciando a influência de eventos de escala secular como MCA e LIA na intensidade das monções na América do Sul. A boa correlação entre algumas razões de elementos-traço com a precipitação mostra que o \"amount effect\" não controla totalmente os registros isotópicos. Análises de estatísticas de séries temporais aplicadas aos registros isotópicos e aos registros de elementos-traço permitiram a observação de vários eventos de freqüêcia decadal a multidecadal influenciando as monções da América do Sul. Os principais destes eventos são a NAO (North Atlantic Oscillation) e a AMO (Atlantic Multidecadal Oscillation), os quais, pelas flutuações de TSM no Atlântico Norte, desencadeiam teleconexões que têm resultados no posicionamento da Zona de Convergência Intertropical (Intertropical Convergence Zone - ITCZ) e da ZACS. / High-resolution isotope records (\'delta\'\'POT.18\'O e \'delta\'\'POT.13\'C), growth rates, elemental ratios in precisely dated speleothems by U/Th method are used to reconstruct the intensity of the South American Monsoon System (SAMS) during the 1500 years in Bonito Region, Mato Grosso do Sul. The interpretations of geochemical variations in speleothems are based on comparative studies with instrumental climatic data and at isotope measurements in rain water at IAEA - GNIP stations in Campo Grande and Cuiabá, as well as from climate model studies using oxygen isotope ratios as tracers of monsoon rainfall intensity. Despite of these data suggest that the \"amount effect\" is a dominant factor controlling the isotope variations in preci pitation, the \"degree of rainout upstream\" is considered also important because the distal influence of SAMS and South Atlantic Convergence Zone (SACZ) along the moisture transport tra jectory from Amazon to SE Brazilian Coast. The other significant factor to isotope composition to precipitation, therefore to speleothems, is the difference in moisture source area from Amazon Region (summer monsoonal precipitation) and Atlantic Ocean (extratropical regime). Bonito\'s paleoclimate Record indicate very good coherence with other records in South America and Northern Hemisphere, particularly in the response to the global scale climate events such as Medieval Climate Anomaly (MCA) e Little Ice Age (LIA) on summer precipitation. In these events were evidenced dryer and wetter conditions, respectively, not just from the stable isotope ratios but also from Sr/Ca and Ba/Ca ratios. Time series statistical analysis in the isotope and trace -element records showed that the SAMS is largely influenced by decadal to multidecadal events. In this regard, the most influent climate modes are the NAO (North Atlantic Oscillation) and the AMO (Atlantic multi Decadal Oscillation), with significant influence on the precipitation variability. In both cases these changes in precipitation are resulted from teleconection with Atlantic Ocean, which impacts the location of ITCZ and consequently the SACZ activity in Central-West Brazil.
