Atividade antimutagênica e citotóxica da β-glicana de Agaricus brasiliensis dependente da MAPK p38

Muitos estudos têm demonstrado que extratos de Agaricus brasiliensis têm atividade citotóxica e antimutagênica, estes efeitos foram relacionados à β-glicana, um polissacarídeo constituinte de parede celular presente em um grande numero de organismos. No entanto, os mecanismos pelos quais este composto atua ainda não foram elucidados. O propósito deste trabalho foi identificar as possíveis vias de sinalização celular pelas quais a β-glicana induz os seus efeitos. Nós empregamos o inibidor da Proteína Cinase Ativada por Mitógeno p38 (MAPK p38) para avaliar a possível participação desta proteína de sinalização celular nas atividades citotóxica e antimutagênica da β-glicana. Os efeitos da β-glicana sobre a citotoxicidade (Sulforhodamine B – SRB – e thiazolyl blue tetrazolium bromide – MTT), antimutagênese (Micronúcleos), ciclo celular (Citometria de fluxo) e indução de apoptose (Vybrant® Apoptosis Assay Kit) foram verificados na linhagem celular H460 (Adenocarcinoma de efusões pleurais). Citotoxicidade foi observada com altas concentrações do polissacarídeo, porém estas concentrações não induziram a apoptose pelo ensaio Vybrant®. No entanto, as mesmas concentrações levaram a um grande acumulo de células em G2/M, indicando que a citotoxicidade da β-glicana resulta de sua capacidade de induzir a paradas de G2/M que podem inibir a proliferação celular. Por outro lado, concentrações não citotóxicas exibiram efeito antimutagênico, reduzindo o dano ao DNA induzido pelo MMS e permitindo a progressão do ciclo celular. Além disso, tanto a antimutagênese quanto a citotoxicidade da β-glicana foram inibidas pelo uso do inibidor da MAPK p38, demonstrando que estas duas diferentes atividades são dependentes de uma mesma via de sinalização celular. / Many studies have demonstrated that Agaricus brasiliensis has cytotoxic and antimutagenic activities; these effects were related to ß-glucan, a polysaccharide constituent of the cell wall present in a large number of organisms. Cytotoxicity was only observed at high concentrations of the polysaccharide. These concentrations did not induce a significant induction of apoptosis. However, these same concentrations trigger a high G2/M arrest, indicating that cytotoxicity of ß-glucan in H460 results from its capacity to induce cell cycle arrest that could prevent cell proliferation. On the other hand, low concentrations showed antimutagenesis effect, reducing MMS-induced DNA damage and allowing cell cycle progression. Besides, both antimutagenesis and cytotoxicity were greatly abrogated by the use of p38MAPK inhibitor, showing that these different activities are dependent on the same signaling pathway.

The main bioactive substances of this mushroom are polysaccharides obtained from the fruiting bodies, being their immunomodulatory activities attributed mainly to -glucans. In this paper we aimed to study the role of a polysaccharide–rich fraction (ATF) of this mushroom on innate immunity receptors expression such as -glucan and mannose receptors (GR and MR), toll-like receptors (TLRs:TLR2 and TLR4) , phagocytosis of Candida albicans, cytokine production (TNF-, IL-1, IL-12 and IL-10) and H2O2 and NO release by human monocytes. ATF significantly increased Candida adherence/ phagocytosis by modulating TLR2 and TLR4 expression, since this polysaccharide had no effect on GR and MR as well as on H2O2 and NO production. Moreover, this polysaccharide increased IL-1 and TNF- production, being this effect also related to its capacity to increase TLR2 and TLR4 respectively. IL-10 levels were also increased. However an association between this effect and toll like receptors expression was not detected. In summary, our results provide evidence about the role of this extract on host resistance against some infectious agents through modulation of some phagocytic cells activities, including those of human monocytes

Observou-se que a taxa de crescimento micelial dos isolados AB51, LF e SO em meio BDA e em composto comercial foi semelhante. Quando esses isolados foram cultivados em meio de cultura líquido, à base de batata, acrescido de glicose, também não foi observada diferença entre o crescimento dos isolados. Porém, quando esse meio foi acrescido de sacarose (açúcar comercial não refinado), o isolado SO foi o que produziu maior massa micelial seca. Ao cultivar o isolado LF em meios de cultura à base de farelos de arroz, farinha de mandioca e,ou fubá, observou-se que nos meios que continham o farelo de arroz, houve maior produção de massa micelial seca. O inóculo líquido, preparado com a mistura de 2% de farelo de arroz e 2% de farinha de mandioca, foi muito eficiente na produção de semente-inóculo à base de grãos de arroz, permitindo uma colonização uniforme do substrato, quando comparado ao inóculo sólido, devido a uma melhor distribuição do micélio fúngico. Entretanto, ao utilizar o inóculo líquido para a inoculação do composto comercial pasteurizado ou esterilizado, mesmo nos casos em que o fungo foi cultivado em meio contendo extrato do composto, não houve crescimento micelial. Pode-se concluir que a produção de pré-inóculo utilizando meio líquido enriquecido com farelos de arroz e,ou farinha de mandioca é uma técnica viável e promissora para a produção de inóculo-semente, por favorecer o crescimento micelial e também pelo baixo custo desses substratos. / The objective of this work was to evaluate the viability of mycelial production of Agaricus blazei in liquid culture with different compositions to be used both as pre- inoculum and as inoculum for compost. The mycelial growth rate of isolates AB51, LF, and SO in PDA and in commercial compost was similar. When these isolates were cultivated in potato dextrose broth (PD), no difference was observed among isolates. However, when commercial sugar (sucrose) was added to the medium, isolate SO produced the highest mycelial dry mass. When isolate LF was grown in culture media based on rice bran, cassava flour and,or corn bran, the highest mycelial dry mass was observed when rice bran was present. The liquid inoculum prepared with a mixture of 2% rice bran and 2% cassava flour was more efficient than solid culture for the production of inoculum/spawn based on rice grains, allowing xa uniform colonization of substrate due to a better distribution of the fungal mycelium through the pores spaces between the grains. However, when the liquid inoculum was used to inoculate commercial compost, either pasteurized or sterilized, no mycelial growth was observed, even in cases when the mycelium was cultivated in medium containing compost extract. The production of pre-inoculum using liquid medium, enriched with rice bran and,or cassava flour, is a viable and promising technique for inoculum/spawn production, not only by improving mycelial growth but also because of the low cost of substrates.

Destes fármacos, a DDS é a principal responsável por reações adversas, como a metemoglobinemia e anemia hemolítica. Estas reações estão relacionadas ao metabólito da DDS, a dapsona-hidroxilamina (DDS-NOH). Na tentativa de promover a redução de efeitos hematotóxicos, são estudadas alternativas terapêuticas com os antioxidantes. Assim, este trabalho teve como objetivo avaliar o efeito protetor da N-acetilcisteína (NAC), A. brasiliensis e Glutationa-etil-éster (GSH-EE) na metemoglobina (MetHb) e no dano em DNA induzidos pela DDS-NOH in vitro, correlacionando aos parâmetros do estresse oxidativo. Para isto, as suspenções de eritrócitos humanos a 50% foram pré e pós-tratadas com NAC, A. brasiliensis e GSH-EE em diferentes concentrações, sendo os grupos expostos a DDS-NOH para induzir a formação de MetHb. Além disso, avaliou-se também a atividade das enzimas SOD e CAT, bem como os níveis de GSH, TEAC e MDA. Em leucócitos avaliou-se a indução de EROs no meio intracelular utilizando o DCFH-DA e o dano em DNA através do ensaio cometa. Os resultados mostraram que o metabólito DDS-NOH foi capaz de induzir MetHb in vitro, sendo este efeito dose-dependente. Em relação ao pré-tratamento, todos os antioxidantes preveniram a formação de MetHb induzida pela DDS-NOH, assim como no pós-tratamento. Em relação a SOD, apenas o pré-tratamento com NAC e A. brasiliensis reduziram a atividade de SOD. Já no pós-tratamento, houve aumento quando tratados com os antioxidantes. O pré-tratamento com NAC e GSH-EE aumentou a atividade de CAT, por outro lado o A. brasiliensis reduziu, assim como no pós-tratamento com os antioxidantes. Quanto aos níveis de GSH, o pré-tratamento com NAC e A. brasiliensis aumentaram GSH, por outro lado, não alteraram no pós-tratamento. Em relação ao TEAC, não houve alteração. Com relação aos danos oxidativos no pré-tratamento, o A. brasiliensis e a GSH-EE reduziram o MDA. Já no pós-tratamento, houve aumento no grupo A. brasiliensis e redução no grupo GSH-EE. Apenas a NAC mostrou ser eficiente na remoção de EROs do meio intracelular induzido por DDS-NOH em leucócitos. Enquanto que em eritrócitos, a NAC e o A. brasiliensis foram capazes de reduzir tal efeito. No estudo do ensaio cometa, a DDS-NOH foi capaz de induzir danos no DNA em leucócitos do sangue periférico, entretanto este danos foram reduzidos quando tratadas com NAC e A. brasiliensis. Pode-se concluir com estes dados que os compostos antioxidantes avaliados apresentam potenciais terapêuticos na prevenção da MetHb e no dano em DNA induzido por DDS-NOH in vitro, sendo a NAC mais eficaz contra estes efeitos. / Dapsone (DDS) is one of the drugs used in polychemotherapy of leprosy associated with rifampicin and clofazimine. Of these drugs, DDS is primarily responsible for adverse reactions, such as methemoglobinemia and hemolytic anemia. These reactions are related to the DDS metabolite, dapsone hydroxylamine (DDS-NOH). In an attempt to promote the reduction of toxic effects are studied alternative therapies with antioxidants. This work aimed to evaluate the protective effect of N-acetylcysteine (NAC), A. brasiliensis and Glutathione ethyl ester (GSH-EE) in methemoglobin (MetHb) and DNA damage induced by DDS-NOH in vitro , correlating to oxidative stress parameters. For this, suspensions of human erythrocytes to 50% were pre- and post-treated with NAC, A. brasiliensis and GSH-EE in different concentrations, being exposed groups DDS-NOH to induce the formation MetHb. It also assessed whether the activity of enzymes-SOD and CAT and GSH levels, TEAC and MDA. In leukocytes evaluated the induction of ROS intracellularly using DCFH-DA and the damage to DNA by comet assay. The results showed that the DDS-NOH metabolite was capable of inducing MetHb in vitro, this dose-dependent effect. Regarding the pre-treatment, all the antioxidants prevented MetHb formation induced by DDS-NOH, as well as post-treatment. For SOD, only the pre-treatment with NAC and A. brasiliensis reduced SOD activity. In the post-treatment, there was increased when treated with antioxidants. Pre-treatment with NAC and GSH-EE increased CAT activity, moreover A. brasiliensis reduced, as after treatment with antioxidants. As for GSH levels, pre-treatment with NAC and GSH increased A. brasiliensis, on the other hand, did not alter after treatment. Regarding TEAC did not change. With respect to oxidative damage in the pre-treatment A. brasiliensis and GSH-EE reduced MDA. In the post-treatment, there was an increase in group A. brasiliensis and reduced GSH-EE group. Only the NAC was shown to be effective in removing the intracellular ROS induced by DDS-NOH in leukocytes. While in erythrocytes, a NAC and A. brasiliensis were able to reduce this effect. In the study of the comet assay, the DDS-NOH was able to induce DNA damage in peripheral blood leukocytes, however this damage was reduced when treated with NAC and A. brasiliensis. It can be concluded from our data that the evaluated antioxidants have therapeutic potential in the prevention of MetHb and DNA damage induced by DDS-NOH in vitro, more effective NAC against these effects.

Na abordagem terapêutica inicial da sepse são utilizados antimicrobianos de amplo espectro, que não é suficiente para o controle da infecção, sendo necessário associações com outras terapias. Dessa forma, o Ilex paraguariensis por ser um potente antimicrobiano, antioxidante, e Agaricus brasiliensis possui propriedades imunomoduladora poderiam ser uma nova fonte de terapia. Assim, o presente estudo visou avaliar o efeito antimicrobiano, antioxidante e imunomodulador in vitro e in vivo dos extratos. Para tanto, neste estudo, foi avaliada a ação antimicrobiana in vitro dos extratos aquosos de A.brasiliensis e I. paraguariensis frente à Staphylococcus aureus e Escherichia coli pelo método da microdiluição e espectrofotmétrico para determinação de concentração inibitória Mínima e técnica de cultivo em placa de petri para a concentração bactericida mínima. Foi também avaliada a citotoxicidade em macrófagos, produção de óxido nítrico (NO), proliferação, fagocitose, capacidade antioxidante equivalente ao Trolox (TEAC) e determinação da atividade antioxidante total pela captura do radical livre e produção de espécies reativas de oxigênio (ERO). In vivo, os camundongos swiss com sepse induzida foram pré-tratados com os extratos aquosos de A.brasiliensis e I. paraguariensis e após 12 e 24 horas suas amostras coletadas,e avaliado a sobrevida, migração leucocitária, hemograma, carga bacteriana, produção de NO, níveis de malonaldeído, TEAC em ex vivo avaliou-se capacidade fagocítica e liberação de EROs. O A.brasiliensis não apresentou atividade antimicrobiana in vitro, manteve as células viáveis, reduziu a capacidade fagocítica, aumentou NO, mas na presença de LPS reduziu o NO, apresentou efeito proliferativo, mas na presença do mitógeno teve efeito antiproliferativo e possui uma forte atividade antioxidante e capacidade de sequestro de radicais in vitro. O I.paraguariensis apresentou atividade antimicrobiana assim como efeito citotóxico, induziu à capacidade fagocítica dos leucócitos, aumentou NO, mas na presença de LPS reduziu, teve efeito proliferativo e atividade antioxidante com capacidade de sequestro de radicais in vitro. Em modelo de sepse in vivo, ambos aumentaram da sobrevida dos animais. O A.brasiliensis reduziu o influxo de leucócitos enquanto que o I. paraguariensis aumentou. Somente A.brasiliensis teve o hemograma semelhante ao sham, ambos extratos reduziram a carga bacteriana e diminuíram os níveis de NO, MDA e aumentaram os níveis de antioxidante nos tecidos, além disso, ambos diminuíram a produção de ERO. Apesar de ambos extratos apresentarem excelente resultados in vitro, o extrato aquoso de A.brasiliensis mostrou ser mais promissor que o extrato de I.paraguariensis como uma terapia adjuvante na sepse, devido sua elevada atividade antimicrobiana, antioxidante e anti-inflamatória in vivo. / Sepsis is an organ dysfunction caused by a dysregulated immune response to an infection, the initial therapeutic approach to sepsis are broad spectrum antimicrobials, which is not sufficient for control of infection, requiring association with other therapies. Thus, Ilex paraguariensis where to be a potent antimicrobial, antioxidant and Agaricus brasiliensis has immunomodulatory properties could be a new source of therapy. Thus, this study aimed to evaluate the antimicrobial effect, antioxidant and immunomodulatory in vitro and in vivo of the extracts. Therefore, in this study, we evaluated the in vitro antimicrobial activity of aqueous extracts from A.brasiliensis and I. paraguariensis against Staphylococcus aureus and Escherichia coli by the method of microdilution and by spectophotometry for determination of minimum inhibitory concentration and cultivation technique on petri dish for minimum bactericidal concentration, cytotoxicity in macrophages was evaluated, nitric oxide (NO) production, proliferation, phagocytosis, equivalent antioxidant capacity to Trolox (TEAC) and determination of the total antioxidant activity by capturing the free radical and reactive species production oxygen (ROS). In the swiss mice with induced sepsis were pretreated with aqueous extracts of A.brasiliensis and I. paraguariensis, after 12 and 24 hours collected their samples and evaluated the survival, leukocyte migration, hemogram, bacterial load, NO production, malondialdehyde (MDA), TEAC and ex vivo we evaluated phagocytic capacity and release of ROS. The A.brasiliensis showed no antimicrobial activity in vitro, remained viable cells reduced the phagocytic capacity, increased NO, but in the presence of LPS reduced, showed proliferative effect, but in the presence of mitogen had antiproliferative effect and has a strong antioxidant activity and capacity to sequester radicals in vitro. I.paraguariensis presented antimicrobial activity as well as cytotoxic effect induced phagocytic capacity increased NO, but in the presence of LPS reduced, had proliferative effect, antioxidant activity and capacity to sequester radicals in vitro. In vivo model of sepsis, both increased the survival of animals, A.brasiliensis reduced leukocyte influx while Ilex increased, only A.brasiliensis had hemogram similar to sham, both extracts reduced bacterial load and levels decreased NO, MDA and increased antioxidant levels in the tissues, in addition, both reduced the production of both extracts present ERO. Although excellent in vitro results, the aqueous extract from A.brasiliensis was found to be most promising as an adjuvant therapy in sepsis that I. paraguariensis due to its high antimicrobial activity, antioxidant and anti-inflammatory in vivo.
