De nouveaux nano-objets anisotropes fluorescents sont obtenus par l’assemblage d’acènes spécifiquement conçus. Dans des cristaux, nano-rubans et nanoparticules anisotropes de 2,3-dialkyldiphenylanthracènes, les efficacités et la polarisation de l’émission bleue sont remarquables. La couleur de l’émission est accordée par le dopage avec des émetteurs verts et oranges (di- et tétra-phényltétracènes). La microscopie confocale de fluorescence permet d’étudier les cinétiques des états excités et des transferts d’énergie photo-induits, ainsi que la dispersion et les orientations des émetteurs. Pour la première fois, l’influence de la largeur de nano-rubans sur la cinétique d’annihilations triplet-triplet de tétracènes est mise en évidence. La microscopie révèle également le polymorphisme inhabituel d’un dérivé diéthynylphényl-anthracène. Ce travail ouvre des perspectives pour le développement et l’étude de processus fondamentaux de nano-matériaux luminescents. / New fluorescent anisotropic nano-objects are obtained by the assembly of specifically designed acenes. In crystals, nano-ribbons and anisotropic nanoparticles of 2,3-dialkyldiphenylanthracenes, the efficiencies and the polarization of the blue emission is remarkable. The color of the emission is tuned by doping with green and orange emitters (di-and tetra-phenyltetracenes). Confocal fluorescence microscopy is used to study the kinetics of excited states and photo-induced energy transfers, as well as the dispersion and orientation of the emitters. For the first time, the influence of the width of the nano-ribbons on the kinetics of tetracene triplet-triplet annihilations is highlighted. Microscopy also reveals the unusual polymorphism of a diethynylphenyl anthracene derivative. This work opens perspectives for the development and study of fundamental processes of luminescent nano-materials.

O estudo do processo da peroxidação de lipídios tem aumentado nos últimos anos, principalmente devido à implicação dos hidroperóxidos de lipídios (LOOH) em diversos processos patológicos. A decomposição destes LOOH é capaz de gerar subprodutos capazes de promover danos em biomoléculas, incluindo proteínas e DNA. No presente trabalho, utilizando hidroperóxidos de ácido linoléico isotopicamente marcado com átomo de oxigênio-18 (LA18O18OH), fomos capazes de demonstrar que estas moléculas gerararam oxigênio singlete marcado [18(1O2)] em células em cultura. A detecção de tal espécie foi possível através da utilização de uma nova metodologia utilizando um derivado antracenico. Para este propósito foi utilizado o derivado de antraceno 3,3\'-(9,10-antracenodiil) bisacrilato (DADB), cujo produto especifico da reação com o 1O2 (o endoperóxido do DADB DADBO2) do pode ser facilmente detectado por HPLC-MS/MS. De forma a expandir a compreensão dos efeitos tóxicos desses LOOH, investigamos o efeito destes compostos gerados intracelularmente. Para tal, foi utilizado o Rosa bengala (RB), um fotosensibilizador que tem afinidade por espaços apolares como membranas e lisossomos. A fotosenssibilização deste composto foi capaz de induzir a morte celular, e esta morte estaria relacionada a uma maior formação de 1O2 e a um maior acumulo de peróxidos. Nestes estudos foi possível demonstrar que carotenóides e sistemas antioxidantes dependentes de glutationa foram capazes de proteger contra os efeitos tóxicos da fotosensibilização na presença de RB. Adicionalmente foram avaliados os efeitos da hemoglobina (Hb) e do hidroperóxido do ácido linoléico (LAOOH) em uma série de parâmetros toxicológicos, como citotoxicidade, estado redox, a peroxidação lipídica e dano ao DNA. Nós demonstramos que a pré-incubação das células com Hb e sua posterior exposição à LAOOH (Hb + LAOOH) levou a um aumento na morte celular, a oxidação do DCFH, formação de malonaldeído e fragmentação do DNA e que esses efeitos estavam relacionados com o grupo peróxido e ao heme presentes na Hb. Foi demonstrado que as células incubadas com LAOOH e Hb apresentaram um nível maior das lesões de DNA; 8-oxo-7,8-diidro-2 \'desoxiguanosina e 1,N2-etheno-2\'-desoxiguanosina. Além disso, as incubações com Hb levaram a um aumento nos níveis de ferro intracelular, e este alto nível de ferro correlacionada com a oxidação do DNA, avaliadas através da medida de sitios EndoIII e Fpg sensíveis. Nossos resultados comprovam que os LAOOHs apresentaram efeito citotóxico e genotóxico, mesmo em concentrações muito baixas, podendo contribuir para o desencadeamento de processos patologicos como o câncer e doenças cardiovasculares e neurodegenerativas. / The study of the process of lipid peroxidation has increased in recent years, mainly due to the involvement of lipid hydroperoxide (LOOH) in a series of pathological processes. The decomposition of LOOH is able to generate products that can promote damage to biomolecules, including proteins and DNA. In the present work, using linoleic acid hydroperoxide isotopically labeled with 18O2 (LA18O18OH), we demonstrate that these molecules were able to generate labeled singlet oxygen [18(1O2)] in cultured cells. The detection of such species was possible using a new methodology using an anthracene derivative .For this purpose we used the anthracene derivative of 3,3\'-(9,10-antracendiil) bisacrilate (DADB), whose specific reaction product with 1O2 (DADB endoperoxide DADBO2) can be easily detected by HPLC-MS/MS. In order to expand the understanding of the toxic effects of LOOH, we investigated the effect of these compounds generated intracellularly. For this porpoise, we used Rose Bengal (RB), a photosensitizer that has affinity for apolar spaces such as membranes and lysosomes. The photosensitization of this compound was able to induce cell death, and this death was related to increased formation of 1O2 and a higher accumulation of peroxides. In these studies we have shown that carotenoids and glutathione-dependent antioxidant systems were capable of protecting against the toxic effects of photosensitization in the presence of RB. Additionally, we evaluated the effects of hemoglobin (Hb) and linoleic acid hydroperoxide (LAOOH) in a series of toxicological endpoints such as cytotoxicity, redox status, lipid peroxidation and DNA damage. We demonstrated that preincubation of cells with Hb and its subsequent exposure to LAOOH (Hb + LAOOH) led to an increase in cell death, DCFH oxidation, formation of malonaldehyde and DNA fragmentation, and that these effects were related to the peroxide and the heme group. It was demonstrated that cells incubated with LAOOH and Hb showed a higher level of the DNA lesions, 8-oxo-7,8-dihydro-2\'deoxyguanosine and 1,N2-etheno-2\'-deoxyguanosine. Furthermore, incubations with Hb led to an increase in intracellular iron levels, and this high level of iron correlates with the oxidation of DNA, measured as EndoIII and Fpg-sensitive sites. Our results show that the LOOHs showed cytotoxic and genotoxic, even at very low concentrations and may contribute to the onset of chronic malignancies like cancer, cardiovascular and neurodegenerative diseases.

Neste trabalho avaliamos a formação de oxigênio singlete in vitro em fagócitos, (células mononucleares e neutrófilos) isolados de sangue periférico humano, e eosinófilos, de lavado bronco alveolar de camundongos balb/c, ativados por estímulo partículado: zimosan opsonizado contendo o 9,10difenilantraceno (DPA) adsorvido como sonda captadora de 1O2. Por este método, a formação do 1O2 pode ser verificada pela formação do 9,10-difenilantraceno endoperóxido (DPAO2), que é detectado por HPLC. Observamos, que os fagócitos formam 1O2 e que esta formação parece ocorrer de forma diferenciada para os dois tipos celulares (neutrófilos e células mononucleares). Visando ampliar os estudos anteriores sobre o papel da melatonina (MLT) no processo inflamatório, foi testado seu efeito em fagócitos e a relação na produção de 1O2 destas células. Observamos que MLT inibe a formação de 1O2 totalmente no caso de neutrófilos e parcialmente no caso de células mononucleares e eosinófilos. Paralelamente, foi desenvolvida a síntese de um novo captador químico de 1O2, o éster 9,10-antracenil-3-bispropionato de etila (ABPE), cuja finalidade principal é o acúmulo no interior da célula, depois de sofrer hidrólise enzimática. Esta sonda, terá facil acesso ao interior das células em sua forma ester. Este novo captador de 1O2 foi testado em células mononucleares e neutrófilos estimulados de formas diferentes: via receptor independente e dependente. Os resultados demonstraram produção equivalente de 1O2 nestes fagócitos. / In this study, we evaluated the singlet oxygen (1O2) formation in vitro from phagocytes (neutrophils and mononuclear cells) isolated from human blood cells and eosinophils isolated from bronchoalveolar lavage fluid of mice balb/c activated, by opsonized zymosan. To determine whether singlet oxygen is produced by phagocytes, zymosan particles were coated with a specific chemical trap for 1O2, 9,10-diphenylanthracene (DPA). The production of 1O2 was followed using HPLC, to measure its product, 9,10-diphenylanthracene endoperoxide (DPAO2). We also noticed that the 1O2 production occurs at different levels of for two cell types, neutrophils and mononuclear cells. In order to broaden previous studies on the role of melatonin (MLT) in inflammatory processes, its effect was tested in phagocytes was tested in relation to 1O2 formation by these cells. We observed that MLT inhibits the 1O2 formation totallymt neutrophils and partiallym mononuclear cells and eosinophils. At the some time, it was also developed the synthesis of a new probe for 1O2, the 9,10-anthracene-bis-3-ethyl-propionate (ABEP), with the purpose to accumulate inside the cells, after its enzymatic hydrolysis. This probe presents easy acess to the inferior of the cells in its ester form. This new probe for trapping 1O2 was tested in mononuclear cells and neutrophils stimulated in two ways: via independent and dependent receptor. The results showed equivalent production of 1O2 for both cell types.

Les activités industrielles sont à l’origine d’un grand nombre de sites et sols pollués en France. Parmi les polluants retrouvés, les hydrocarbures aromatiques polycycliques (HAP), notamment ceux de haut poids moléculaire, posent un réel problème environnemental et de santé publique en raison de leur toxicité et de leur persistance dans les sols. Les techniques biologiques de remédiation qui stimulent la dégradation microbienne des HAP en utilisant des plantes (phyto/rhizoremédiation), des nutriments (biostimulation), ou encore des microorganismes (bioaugmentation), apparaissent comme des stratégies intéressantes car elles sont plus respectueuses de l’environnement et moins onéreuses que les méthodes physico-chimiques. Dans ce contexte, la capacité de la plante Miscanthus x giganteus (MxG) à améliorer la dissipation du benzo(a)anthracène (BaA), un HAP de haut poids moléculaire peu étudié en remédiation, a été évaluée par une approche pluridisciplinaire en réalisant deux types d’expérimentations à partir d’un sol artificiellement contaminé. La première a consisté à analyser l’influence rhizosphérique de la plante (phyto/rhizoremédiation) au cours d’une période de 12 mois. Les résultats ont montré que la présence de MxG conduisait en fin d’exposition à une réduction 3 fois plus importante de la teneur en BaA dans le sol grâce à une augmentation de la biodisponibilité du contaminant et à un effet positif sur les communautés microbiennes via une modification de leur diversité et une augmentation de leur densité de gènes ARNr 16S et PAH-RHDα GP. Afin d’améliorer la compréhension de ce phénomène, une deuxième expérimentation a été mise en place sur une durée de 105 jours dans le but d’étudier spécifiquement l’influence de certaines molécules exsudées au niveau du système racinaire de la plante (biostimulation) et de microorganismes spécifiques, associés à sa rhizosphère, ayant la capacité de dégrader le BaA (bioaugmentation). Ces essais ont témoigné de la complexité des mécanismes rhizosphériques et de la nécessité d’approfondir ce type d’analyses afin d’améliorer la compréhension des processus de biodégradation dans la rhizosphère de la plante. L’ensemble de ces résultats a alors montré que MxG était une candidate idéale pour la phyto/rhizoremédiation de sols contaminés par des HAP de haut poids moléculaires mais que des études supplémentaires étaient encore nécessaires pour comprendre les processus intervenant dans sa rhizosphère / Industrial activities result of a large number of contaminated sites and soils in France. Among the pollutants found, polycyclic aromatic hydrocarbons (PAHs), and especially those of high molecular weight, are a real environmental and public health problem because of their toxicity and persistence in soils. Biological remediation techniques which stimulate microbial degradation of PAHs using plants (phyto/rhizoremediation), nutrients (biostimulation), or microorganisms (bioaugmentation), appear as interesting strategies because they are more environmental friendly and less expensive than physico-chemical methods. In this context, the ability of the plant Miscanthus x giganteus (MxG) to improve benzo(a)anthracene (BaA) dissipation, a high molecular weight PAH few studied in remediation, was assessed with a multidisciplinary approach through two types of experimentations using an artificially contaminated soil. The first one was to analyze the rhizospheric influence of the plant (phyto/rhizoremediation) over a period of 12 months. Results showed that the presence of MxG led to a 3-fold reduction in BaA concentration in soil at the end of exposure thanks to an increase in pollutant bioavailability, and a positive effect on microbial communities due to a modification of their diversity and an increase in their 16S rRNA and PAH-RHDα GP genes density. In order to improve the understanding of this phenomenon, a second experiment was carried out for 105 days to study specifically the influence of certain molecules exuded in the plant’s root system (biostimulation), and specific microorganisms associated with its rhizosphere having the capacity to degrade BaA (bioaugmentation). These tests demonstrated the complexity of rhizospheric mechanisms, and the necessity to explore this type of analysis to improve the understanding of biodegradation processes in plant rhizosphere. All these results showed that MxG was an ideal candidate for phyto/rhizoremediation of soils contaminated with high molecular weight PAH, but that further studies were still needed to understand processes involved in its rhizosphere

Des matériaux ayant des propriétés émissives spécifiques peuvent être obtenus par l'organisation contrôlée de fluorophores aux échelles moléculaire, nano- et micro-métrique. Dans ce travail, l'émission de lumière bleue polarisée est obtenue par l'auto-assemblage hautement anisotrope de n-acènes alcoxylés en nano-rubans. Des techniques de microscopie de fluorescence ont été utilisées pour déterminer le mécanisme de leur croissance et ont été combinées à la cristallographie aux rayons X pour déterminer l'empilement moléculaire dans les nano-objets. L'étude a révélé que la formation des nano-rubans est induite non seulement par le mécanisme de maturation d'Ostwald très commun, mais aussi par une croissance par attachement orienté rarement démontré dans des systèmes organiques. En plus des techniques plus courantes, la microscopie en polarisation de fluorescence de molécules uniques a contribué à caraxctériser l'emplilement moléculaire, bien que les nano-objets à haute densité en chromophore constituent des échantillons très difficiles à étudier. Dans ce travail, les propriétés des nano-rubans ont été contrôlées au niveau microscopique par les conditions de croissance, ainsi que par l'addition de dopants. Ainsi, en combinant différentes molécules et une réaction photochimique sous microscopie, des rubans à motifs colorés sub-micrométriques ont été obtenus. Par ailleurs, l'assemblage orthogonal a été exploité pour développer des réseaux interpénétrés. Ces derniers se distinguent par une émission à double couleur, un transfert d'énergie entre objets et une électroluminescence aux jonctions. / Materials with specific emissive properties can be obtained by the controlled organization of fluorophores at the molecular, nano- and microscales. In this work, polarized blue light emission is achieved by the highly anisotropic self-assembly of alkoxylated n-acenes into nano-ribbons. Fluorescence microscopy techniques were used to determine the growth mechanism and were combined to X-ray crystallography to determine the molecular packing in the nano-objects. The study revealed that the formation of the nano-ribbons is induced not only by the very common Ostwald ripening mechanism but also by an oriented attachnment growth, rarely observed with such evidence in organic systems. Besides more common techniques, single molecule fluorescence polarization microscopy contributed to characterize the molecular packing, although the nano-objects with high chromophore density represent very challenging samples. In this work, the properties of the nano-ribbons have been controlled at the microscopic level by the growth conditions, as well as by the addition of dopants Thereby, combining different molecules and photochemistry at the sub-micrometer scale under the microscope, colorful patterned ribbons could be obtained. In addition, orthogonal assembly was exploited to grow interpenetrated networks. The latter demonstrated dual color-emission, as well as inter-object energy transfer and electroluminescence at junctions.

Paralelamente, foi desenvolvida a síntese de um novo captador químico de 1O2, o éster 9,10-antracenil-3-bispropionato de etila (ABPE), cuja finalidade principal é o acúmulo no interior da célula, depois de sofrer hidrólise enzimática. Esta sonda, terá facil acesso ao interior das células em sua forma ester. Este novo captador de 1O2 foi testado em células mononucleares e neutrófilos estimulados de formas diferentes: via receptor independente e dependente. Os resultados demonstraram produção equivalente de 1O2 nestes fagócitos. / In this study, we evaluated the singlet oxygen (1O2) formation in vitro from phagocytes (neutrophils and mononuclear cells) isolated from human blood cells and eosinophils isolated from bronchoalveolar lavage fluid of mice balb/c activated, by opsonized zymosan. To determine whether singlet oxygen is produced by phagocytes, zymosan particles were coated with a specific chemical trap for 1O2, 9,10-diphenylanthracene (DPA). The production of 1O2 was followed using HPLC, to measure its product, 9,10-diphenylanthracene endoperoxide (DPAO2). We also noticed that the 1O2 production occurs at different levels of for two cell types, neutrophils and mononuclear cells. In order to broaden previous studies on the role of melatonin (MLT) in inflammatory processes, its effect was tested in phagocytes was tested in relation to 1O2 formation by these cells. We observed that MLT inhibits the 1O2 formation totallymt neutrophils and partiallym mononuclear cells and eosinophils. At the some time, it was also developed the synthesis of a new probe for 1O2, the 9,10-anthracene-bis-3-ethyl-propionate (ABEP), with the purpose to accumulate inside the cells, after its enzymatic hydrolysis. This probe presents easy acess to the inferior of the cells in its ester form. This new probe for trapping 1O2 was tested in mononuclear cells and neutrophils stimulated in two ways: via independent and dependent receptor. The results showed equivalent production of 1O2 for both cell types.

Molecular relaxation dynamics of Anthracene cations studied in an electrostatic storage ring / Dynamique de relaxation de cations d'Anthracène étudiée dans un anneau de stockage électrostatiqueJi, Ming Chao 28 April 2015 (has links)
Les molécules hydrocarbures aromatiques polycycliques (HAP) sont à l'heure actuelle considérées comme probablement responsables des bandes d'émission infrarouge non identifiées du milieu interstellaire (MIS). La dynamique de refroidissement des molécules HAP est essentielle pour estimer leur photo-stabilité, leur durée de vie et les distributions de taille dans le MIS. Au cours des dernières années, les expériences s'appuyant sur le stockage électrostatique d'ions moléculaires ou d'agrégats sont devenus des outils puissants pour étudier leur refroidissement dans une large gamme de temps allant de la microseconde à quelques secondes. En général, l'étude des courbes de déclin associées aux processus de dissociation dans le cas des cations ou bien de détachement d'électrons dans le cas des anions fournit des informations sur l'évolution de l'énergie interne des ions stockés. Dans ce travail de thèse, le refroidissement de cations d'anthracène a été étudié dans un anneau de stockage électrostatique compact, le Mini-Ring, jusqu'à 8 ms. Les courbes de déclin spontané provenant de la dissociation par émission de fragment C2H2 ou H neutres montrent trois régions distinctives. Ces trois régions indiquent différents régimes de refroidissement en fonction du temps de stockage, la dissociation domine pour les temps inférieurs à 1 ms, l'effet de l'émission radiative entre alors en compétition avec la dissociation puis domine au-delà de 3 ms / The polycyclic aromatic hydrocarbon (PAH) molecules have been considered as possible carrier of the unidentified infrared emission bands from the interstellar medium (ISM) for about thirty years. The cooling dynamics of the PAH molecules which is essential to estimate their photostability and therefore their lifetime and size distributions in the ISM, has attracted numerous theoretical and experimental studies. In recent years, electrostatic storage devices (ESD) became powerful tool to investigate the cooling regime of molecules and clusters in a large time range from microseconds to seconds. Generally speaking, the decay of the emitted neutral yields due to dissociation of molecular cations or electron detachment of anions in such experiments carries information on the internal energy of the stored molecular ions. In this thesis work, the cooling regimes of anthracene cations are studied by following the time evolution of the internal energy distribution (IED) of the stored anthracene cations. A spontaneous neutral yield curve obtained from the stored molecular ions as a function of the storage time shows three distinguishable regions. The three regions indicate different cooling regimes at corresponding storage time range, i.e., the dissociation mechanism of the molecule dominates at storage time t < 1 ms, quenching of the dissociation by radiative cooling processes occurs during 1 < t < 3 ms and radiative cooling governs at t > 3 ms

Propriétés magnétiques et structurales de complexes moléculaires supportés par des ligands de type acyle-hydrazone / Magnetic and structural properties of molecular complexes bearing acyl-hydrazone ligandsSpecklin, David 12 November 2014 (has links)
Ces travaux consistent en l’étude selon trois axes de la chimie de coordination des ligands de type acyle-hydrazone, faisant suite à l’isolation d’un complexe binucléaire de manganèse (III) portant un tel ligand, et montrant un fort couplage ferromagnétique. Le développement de leur chimie de coordination avec des éléments de la première série de transition a permis la synthèse de plusieurs complexes binucléaires, permettant d’étudier la relation entre les structures moléculaires de ces systèmes, et leurs propriétés magnétiques. En parallèle, une série de polymères de coordination de métaux alcalins et alcalino-terreux portant des ligands acyle-hydrazone fonctionnalisés par un groupement sulfonate a été étudiée. Enfin, la synthèse de deux ligands dérivés d’un anthracène est présentée, celle-ci est accompagnée d’une étude de leurs propriétés de coordination. / This work consists in the study of the coordination chemistry of acyl-hydrazone ligands, following the discovery of a manganese (III) dinuclear complex showing a strong ferromagnetic coupling. The development of the acyl-hydrazone coordination to first-row transitional metals resulted in several dinuclear complexes allowing the study of the relationship between their structural and magnetic properties. In a second part the study of several coordination polymers of alkali and alkali-earth metals bearing acyl-hydrazone ligands functionalized with a sulfonate group is presented. Finally the synthesis of two anthracene-based ligands is presented along a study of their coordination properties.
