F.Macbride, uma árvore com grande valor na indústria madeireira, é uma leguminosa (Fabaceae) encontrada na Caatinga. Lectinas são proteínas capazes de reconhecer e se ligar a carboidratos e glicoconjugados de acordo com sua especificidade e, por isso, têm diversas aplicações bioquímicas e biotecnológicas. Sementes de leguminosas são reconhecidas por serem boas fontes de lectinas. Este trabalho descreve a purificação, caracterização e a atividade antimicrobiana deApulSL (Apuleia leiocarpa seeds lectin). ApulSL foi extraída misturando a farinha das sementes em solução salina (NaCl 150 mM), gerando o extrato bruto (EB) que foi aplicado em cromatografia emcoluna de quitina eluida com ácido acético 1M, obtendo-se um único pico ativo. A ApulSL é termorresistente (100°C/2h) e íon dependente (Mn2+), tem caráter ácido e afinidade por N-acetilglicosamina, D(-)-arabinose e azocaseína. Trata-se de uma proteína com estrutura conformacional desordenada, com resíduos de tirosina no cerne hidrofóbicoe um peso molecular nativo de 55,8 kDa, mas que em condições desnaturantes e redutorasse desdobra emtrês a quatro oligopeptídeos, sendo portanto, uma proteína oligomérica. A análise emMALDI TOF/TOF detectou vários peptídeos, mas não houve homologia suficiente (< 30%) entre os peptídeos detectados e o banco de dados de proteínas para a elucidação da sequência primária. A ApulSLapresentouatividade inibitória e bactericida contra três variedades de Xanthomonas campestris, como também inibiu outras cepas gram-negativas e gram-positivas. Em conclusão, a ApulSL é uma lectina sem registros anteriores na literatura e que apresentou potencial antimicrobiano contra a espécie Xanthomonas campestris

Deficiência de ferro em grápia: efeito da adubação fosfatada e potássica / Iron deficiency in grápia: effects of the phosphorus and potassium fertilization

Macbride) showed that the increase in the P fertilization induced visual symptoms of Fe-deficiency in the younger leaves. At cellular level, potassium may contribute to balance charges between the cytoplasm and apoplasm, hence allowing the uptake of iron by plant cells. The aim of this study was to evaluate the effects of phosphorus and potassium fertilization in relation to Fedeficiency in grapia seedlings cultivated in a Paleudalf soil. Twenty seven fertilization treatments were used, represented by the combination of three levels of P (80, 150 e 220 mg P kg-1 of the soil), three levels of K (70, 110 e 150 mg kg-1 of the soil) and three levels of Fe as Fe-EDTA (0, 7,5 e 15 mg Fe kg-1 of soil), under unclimatized greenhouse conditions. The plant growth in number of leaves, number of fallen leaves, height and stem diameter was analyzed monthly. Until 150 days after fertilization (DAF), leaves with visual symptons of Fe deficiency was counted monthly. At 150 DAF δ-Aminolevulinic acid dehydratase activity was determined. Dry matter of leaves, stem, shoot, roots and whole plant, root/shoot dry matter ratio, chemistry features of the soil, as well as P, K, Ca, Mg, Fe, Zn and Cu concentration and content from leaves of grápia were analyzed at 180 DAF. The high availability of P in the soil gave rise to chlorosis in the younger leaves similar to that of Fe deficiency. The P fertilization increased the P/Fe, P/Zn and P/Cu ratios in the leaves, whereas K and Fe fertilization decreased those ratios. Nevertheless, the potassium fertilization did not influence the porphobilinogen formation, a precursor molecule of chlorophyll. Therefore, the increased K availability was not sufficient to resolve the nutritional unbalance induced by phosphorus fertilization on grápia. / Em estudos prévios realizados com a grápia (Apuleia leiocarpa (Vog.) Macbride) observouse que o aumento da adubação de fósforo induziu sintomas visuais de deficiência de ferro nas folhas. O potássio, em nível celular, pode contribuir para o balanceamento de cargas entre o citoplasma e o apoplasto, permitindo que a célula absorva quantidade de ferro necessária para suprimir tal deficiência. O objetivo do presente trabalho foi avaliar os efeitos da adubação fosfatada e potássica em relação à deficiência de ferro em plantas jovens de grápia, cultivadas num Argissolo Vermelho Distrófico arênico. Foram testados três níveis de adubação de P (80, 150 e 220 mg kg-1 de solo), três níveis de K (70, 110 e 150 mg kg-1 de solo) e três níveis de Fe, na forma de Fe-EDTA (0, 7,5 e 15 mg kg-1 de solo), totalizando 27 tratamentos, sob condições de casa de vegetação. Mensalmente, a partir dos 30 dias após a adubação (DAA) até o término do experimento, 180 DAA, foram avaliados o número de folhas, número de folhas caídas, altura da planta e o diâmetro do caule. Também, mensalmente, até os 150 DAA efetuou-se a contagem das folhas com sintomas visuais típicos de deficiência de ferro. Aos 150 DAA foi determinada a atividade da enzima citoplasmática δ-aminolevulinato desidratase. Aos 180 DAA avaliaram-se ainda a produção de matéria seca das folhas, do caule, da parte aérea, das raízes e do total da planta; a relação entre a matéria seca do sistema radicular e matéria seca da parte aérea, as características químicas do solo, assim como o teor e a quantidade total de P, K, Ca, Mg, Fe, Zn e Cu nas folhas da grápia. Semelhante ao registrado anteriormente para a grápia, constatou-se também no presente trabalho que a alta disponibilidade de P no solo induziu aparecimento de sintomas visuais de deficiência de Fe nas folhas. As relações entre os teores de P/Fe, P/Zn e P/Cu nas folhas aumentaram com a adubação fosfatada, enquanto que adubações de K e Fe diminuíram tais relações. Apesar disso, a adubação potássica não influenciou na formação do porfobilinogênio, molécula precursora da clorofila. Portanto, o aumento da disponibilidade do K não foi o suficiente para sanar o desbalanço nutricional causado pela adubação fosfatada.

Systematics and floral evolution of the Dialiinae (Caesalpinioideae), a diverse lineage of tropical legumes

Le clade Dialiinae représente l’une des premières lignées de la sous-famille Caesalpinioideae des Leguminosae. Il se compose de 17 genres (environ 90 espèces), avec des taxons qui sont répandus dans toutes les régions tropicales du monde. Morphologiquement, le groupe comprend un assemblage divers de taxons qui peut représenter une «phase expérimentale» dans l’évolution florale des légumineuses. Différents représentants du clade présentent de la poly-, mono-, et asymétrie, et semblent avoir subi un haut degré de perte d’organe, produisant, dans certains cas, des fleurs extrêmement réduites qui sont à peine reconnaissables comme appartenant à la famille des légumineuses. Afin d’obtenir une image plus claire de l’évolution florale du clade Dialiinae, une phylogénie bien résolue et bien soutenue est nécessaire. Dans le but de créer une telle phylogénie, un total de 37 échantillons d’ADN des Dialiinae a été séquencé pour deux régions chloroplastiques, soit rps16 et trnL. De plus, une étude morphologique complète a été réalisée. Un total de 135 caractères végétatifs et reproductifs a été évalué pour 79 espèces de Dialiinae et pour quatre groupes externes. Les analyses phylogénétiques ont d’abord été effectuées sur un groupe restreint de taxons pour lesquels les trois types de données étaient disponibles. Les nœuds fortement soutenus de cette phylogénie ont ensuite été utilisés comme contrainte pour une seconde analyse de parcimonie avec les données morphologiques d’un ensemble plus important de taxons. Les caractères morphologiques ont été optimisés sur l’un des arbres les plus parcimonieux de cette seconde analyse. Un certain nombre de nouvelles relations au niveau de l’espèce ont été résolues, créant une image plus claire quant à l’évolution de la forme florale dans le temps, particulièrement pour les genres Labichea et Dialium. En plus de leur morphologie florale mature diverse, les Dialiinae sont également très variables dans leur ontogénèse florale, affichant à la fois la perte et la suppression des organes, et présentant une variété de modes d’initiation d’organes. Afin de construire une image plus complète du développement floral et de l’évolution dans ce clade, l’ontogénèse florale de plusieurs espèces non documentées à ce jour a été étudiée. La série complète du développement a été compilée pour six espèces de Dialiinae; quatre de Dialium, ainsi que Poeppigia procera et Mendoravia dumaziana. Le mode et le moment de l’initiation des organes étaient pour la plupart uniforme pour toutes les espèces de Dialium étudiés. Tant pour ce qui est des gains ou des pertes d’organes chez Dialium, une tendance est apparente – l’absence d’organe abaxial. Que ce soit pour les sépales ou les étamines, les gains se produisent toujours en position médiane adaxiale, tandis que les étamines et les pétales perdus sont toujours les organes les plus ventraux. Les taxons étudiés ici illustrent le manque apparent de canalisation du développement observé chez les Caesalpinioideae. Cette plasticité ontogénétique est le reflet de la diversité morphologique au niveau des fleurs tel qu’observée dans l’ensemble de la sous-famille. Une des espèces de Dialiinae, Apuleia leiocarpa, produit une inflorescence andromonoïque, une caractéristique qui est unique en son clade et rare dans les légumineuses dans son ensemble. La microscopie optique et électronique ont été utilisées pour entreprendre une étude détaillée de la morphologie florale de ce taxon. On a constaté que tandis que les fleurs hermaphrodites produisent un seul carpelle et deux étamines, les fleurs staminées produisent trois étamines sans toutefois montrer signe de développement du carpelle. Les inflorescences semblent produire près de quatre fois plus de fleurs staminées que de fleurs hermaphrodites, lesquelles occupent toujours la position centrale de l’inflorescence cymeuse. Ce ratio élevé mâle/bisexuel et la détermination précoce du sexe chez Apuleia sont rares chez les Caesalpinioideae, ce qui suggère que l’andromonoecie se développe dans ce genre comme un moyen d’accroître la dispersion du pollen plutôt qu’en réponse à des limitations de ressources. / The Dialiinae clade represents one of the early-diverging lineages of the legume subfamily Caesalpinioideae; it consists of 17 genera (circa 90 species), and is pantropically distributed. Morphologically, the group comprises a diverse assemblage of taxa that may represent a so-called “experimental phase” in legume floral evolution. Different members of the clade exhibit poly-, mono-, and asymmetry, as well as having undergone a high degree of organ loss, producing, in some cases, extremely reduced flowers which are barely recognisable as belonging to the legume family. In order to obtain a clearer picture of floral evolution in the Dialiinae, a well resolved and well supported phylogeny is needed onto which morphological characters may be optimised. With the goal of creating such a phylogeny, a total of 37 Dialiinae DNA samples were sequenced for two plastid genes, rpS16 and trnL. Additionally, a comprehensive morphological study was carried out. A total of 135 vegetative and reproductive characters were scored for 79 ingroup and four outgroup taxa. Phylogenetic analyses were carried out first on a restricted group of taxa for which all three data sets were available. The highly supported nodes of this phylogeny were then used as a constraint for a second parsimony analysis of morphological data from a much larger taxon set. Morphological characters were then mapped onto one of 20,000 most parsimonious trees from this second analysis. A number of novel species-level relationships were resolved, creating a clearer picture of changes in floral form over time, particularly in the genera Labichea and Dialium. In addition to their diverse mature floral morphology, the Dialiinae are also widely variable in their floral ontogeny, displaying both organ loss and suppression, and exhibiting a wide variety of organ initiation modes. In order to build a more complete picture of floral development and evolution in this clade, the floral ontogeny of several previously undocumented species was investigated. Complete developmental series were compiled for six species of the Dialiinae; four from Dialium, as well as Poeppigia procera and Mendoravia dumaziana. Mode and timing of organ initiation were mostly consistent across the Dialium species studied. In the instances of both gains and losses of floral organs in Dialium, one trend is apparent — an absence of abaxial organs. Gains in both sepals and stamens occur in the adaxial median position, while stamens and petals that are lost are always the ventral-most organs. The taxa examined here exemplify the apparent lack of developmental canalisation seen in caesalpinioid legumes. This ontogenetic evolvability is reflective of the morphological diversity shown by flowers across the subfamily. One of the species of the Dialiinae, Apuleia leiocarpa, produces an andromonoecious inflorescence, a feature that is unique in its clade and rare in the Leguminosae as a whole. Light and electron microscopy were used to undertake a detailed study of the floral morphology of this taxon. It was found that while hermaphrodite flowers produced a single carpel and two stamens, staminate flowers developed three stamens but showed no sign of carpel development. Inflorescences also appear to produce approximately four times as many staminate as hermaphrodite flowers, with hermaphroditic flowers consistently occupying the central position in cymose inflorescences. Both this high male-to-bisexual ratio and the early determination of gender seen in Apuleia are rare in the Caesalpinioideae and suggest that andromonoecy developed in this genus as a means to increase pollen dispersal rather than in response to resource limitations.

Associação de fungos ectomicorrízicos com espécies florestais nativas do estado do Rio Grande do Sul / Association of ectomycorrhizal fungi with native forestry species of state of Rio Grande do Sul

Conselho Nacional de Desenvolvimento Científico e Tecnológico / The work was development in Forestry Fepagro, Santa Maria, RS and in Laboratory of Biology and Microbiology of Soil and Environment Prof. Marcos Rubens Fries, from Department of Soil of Federal University of Santa Maria. First, the objective was to identify mycorrhizal associations in six native forestry species of Rio Grande do Sul State. This research was conducted in tree seasons of the year, in six native forestry species of State of Rio Grande do Sul. The roots were processed and analyzed according to mycorrhizal colonization type. The sporocarps found of native ectomycorrhizal fungi were identified, isolated and multiplied. The studies species were Araucaria angustifolia (Bertol.) Kuntze), Enterolobium contortisiliquum (Vell.) Morong.), Peltophorum dubium (Spreng) Taub.), Tabebuia chrysotricha (Mart. ex DC.) Standl.), Tabebuia impetiginosa (Mart.) Standl.), Apuleia leiocarpa (Vogel) J.F. Macbride). The forestry species didn t show ectomycorrhizal colonization in native conditions, although, it was found in some sporocarps near to some plants. It was observed the presence of associations with endomycorrhizal fungi in all species. The following study had the objective to test the possibility to form associations with ectomycorrhizal fungi and Apuleia leiocarpa and, in the fungi isolated UFSM RA 2.8 and UFSM RA 3.6 from Fepagro Forestry, respectively. These fungi were classified as Suillus sp. and Scleroderma sp. respectively. The fungi UFSC Pt 116 (Pisolithus microcarpus) and UFSC Pt 24 (Pisolithus sp.), were from of Federal University of Florianópolis, including the treatment without fungi. In Apuleia leiocarpa the fungi UFSM RA 3.6 was substituted by UFSC Sc 124 (Scleroderma sp.) fungi. It was analyzed seedling length, brash mass of aerial part and root, dry mass of part aerial, length and root and mycorrhizal colonization. It was observed the formation of ectomycorrhizal association in little plants of Apuleia leiocarpa and in Peltophorum dubium when inoculated with the ectomycorrhizal fungi Suillus sp.(UFSM RA 2.8). Furthermore, the results showed the best development in Apuleia leiocarpa in high plants, brash mass of aerial part and root, dry mass of part aerial. The Apuleia leiocarpa showed ectomycorrhizal association with isolated UFSM RA 2.8, in vitro . The Peltophorum dubium showed evidence that is possible have ectomycorrhizal associations with the isolate UFSM 2.8 in laboratory conditions. / O trabalho foi desenvolvido na Fepagro Florestas, Santa Maria RS e no Laboratório de Microbiologia e Biologia do Solo e do Ambiente Prof. Marcos Rubens Fries, do Departamento de Solos da Universidade Federal de Santa Maria. Primeiramente, o objetivo foi realizar um levantamento da ocorrência bem como a identificação das associações micorrízicas em seis espécies florestais nativas presentes no Estado do Rio Grande do Sul. O estudo foi conduzido em 3 épocas do ano com a coleta de amostras de solo, raízes e esporocarpos dos fungos. As raízes foram processadas e analisadas quanto ao tipo de colonização micorrízica. Os esporocarpos fungos ectomicorrízicos nativos encontrados foram identificados, isolados e multiplicados. As espécies estudadas foram o pinheiro-do-paraná (Araucaria angustifolia (Bertol.) Kuntze), timbaúva (Enterolobium contortisiliquum (Vell.) Morong.), canafístula (Peltophorum dubium (Spreng.) Taub.), ipê-amarelo (Tabebuia chrysotricha (Mart. ex DC.) Standl.), ipê-roxo (Tabebuia impetiginosa (Mart.) Standl.), grápia (Apuleia leiocarpa (Vogel) J.F. Macbride). As espécies florestais estudadas não apresentaram colonização ectomicorrízica a campo, entretanto, foram encontrados esporocarpos próximos de algumas plantas. Observou-se a presença de associações com fungos endomicorrízicos em todas as espécies. Posteriormente, estudou-se a possibilidade de formar associações com fungos ectomicorrízicos, grápia e canafístula. Foram utilizados os seguintes isolados: na grápia os fungos utilizados foram os isolados UFSM RA 2.8 e UFSM RA 3.6 oriundos da Fepagro Florestas, Santa Maria-RS, classificados como Suillus sp. e Scleroderma sp., respectivamente e os isolados UFSC Pt 116 (Pisolithus microcarpus) e UFSC Pt 24 (Pisolithus sp.), oriundos da Universidade Federal de Florianópolis, mais a testemunha sem fungo. Na canafístula o fungo UFSM RA 3.6 foi substituído pelo fungo UFSC Sc 124 (Scleroderma sp.). As avaliações foram: altura de planta, massa verde da parte aérea, massa verde radicular, massa seca da parte aérea e colonização micorrízica. Ocorreu a formação de associação ectomicorrízica nas plântulas de grápia e indícios desta formação com a canafístula quando inoculadas com o fungo ectomicorrízico Suillus sp. UFSM RA 2.8. Além disso, os resultados mostram que ocorreu melhor desenvolvimento nas plântulas de grápia quanto a altura de plantas, massa verde radicular e da parte aérea, e massa seca da parte aérea. A grápia apresentou associações ectomicorrízicas com o isolado UFSM RA 2.8, in vitro . A canafístula apresentou características morfológicas evidenciando uma possível associação ectomicorrízicas com o inóculo UFSM RA 2.8 em condições de laboratório.
