Sherlock Holmes har adapterats till film i över hundra år. Syftet med den här uppsatsen är att genomföra en studie om adaptioner av Sherlock Holmes romanen The Hound of the Baskerville har anpassats för sin samtid mellan 1939 och 2012. Analysen utgår därmed ifrån Sidney Lanfields adaption ifrån 1939 med Basil Rathbone, Terence Fishers adaption ifrån 1959 med Peter Cushing. Slutligen TV-serien Sherlocks adaption ifrån 2012 med Benedict Cumberbatch i rollen som Holmes. Den grundläggande teorin för uppsatsen är adaptionsteori, för att få fram hur Sir Arthur Conan Doyles roman har ändrats och anpassats för att bli lämplig för sin samtida publik. Analysen är uppdelad i tre kapitel, i vilka olika delar av det som har adapterats analyseras. De olika kapitlen handlar om filmskaparna har omarbetat och tolkat Doyles roman för sin samtid? Har de tolkat och omarbetat de kvinnliga karaktärerna för sin samtids publik? Har Sherlock själv utvecklats mellan de tre adaptionerna? Slutsats omfattar sedan en diskussion om uppsatsens resultat, baserad på Linda Hutcheons teori om adaption. / Sherlock Holmes have been adapted to film for over a hundred years. The purpose with this essay is to conduct a study of how adaptations of the Sherlock Holmes novel The Hound of the Baskervilles have been adjusted between 1939 and 2012, to make the story more appropriate for their contemporary audience. The analysis is based on Sidney Lanfields 1939 adaptation with Basil Rathbone, the 1959 Terence Fisher adaptation with Peter Cushing and the 2012 adaptation for the TV series Sherlock. The Essay is based in adaption theory, to determine how Sir Arthur Conan Doyle’s novel has been changed and adjusted to make the story more appropriate for the contemporary audience of the adaptation in question. The analysis is divided into three chapters, which examines different aspects of what has been adapted. The different chapters analyze how the filmmakers have reworked and interpreted Doyle’s novel for their time, how they have interpreted and reworked the female characters and how Sherlock himself has evolved between the three adaptations.

A repeti??o com diferen?a de livros e obras p?e em questionamento imagens arraigadas, abrindo espa?o para o ?flagrante delito? da fabula??o de mundos, que se constituem na superf?cie da escrita e que n?o se reduzem a sedimenta??es. / This thesis discusses and analyzes as the parodic metafiction, namely, the process that makes fiction visible by repetition with difference, enhances the creation of a different detective narrative in the novel A Samba for Sherlock (1995), by J? Soares. The research is established by readings which take into account mainly the parodic metafiction, the autopoietic writing, the humor as the art of surfaces, as well as the fabulation of worlds to come, making use of concepts of the philosophy of difference in Deleuze and Guattari and literary criticism, especially in Linda Hutcheon, Blanchot and Foucault such as autopoietic machine, humor, fabulation, metafiction, parody, space of literature and work. In the plane of composition, the novel of the Brazilian humorist makes its fictional reality visible, parodying literary and historiographical works, mainly the Doylian detective narratives, and work related to the Rio de Janeiro of the late nineteenth century. In this sense, this work develops a writing that remains in itself, displaces significations and sees its fictional reality. In Soares?s heterocosm, the narrative brings together the space of a library. The appeal of the work is in the convening of stereotypes presented in books on the capital of the empire of the tropics at the end of the second reign, as well as on detective narratives. The repetition with difference of books and works call into question deep-rooted images, making room for fabulation ?in flagrante delicto? of worlds that constitute the writing surface and not reduce itself to sedimentation.

Identifizierung und funktionelle Charakterisierung mitochondrialer Proteinkinasen und-phosphatasen in Saccharomyces cerevisiae

Über die Proteinphosphorylierung in den Mitochondrien der Hefe Saccharomyces cerevisiae ist im Gegensatz zu anderen Kompartimenten nur wenig bekannt. Insbesondere hinsichtlich der physiologischen Bedeutung sowie den an der Modifikation beteiligten Enzymen sind kaum Daten verfügbar. Vor diesem Hintergrund stand die Identifizierung und molekularbiologische Charakterisierung mitochondrialer Proteinkinasen (PKasen) und Proteinphosphatasen (PPasen) im Fokus dieser Arbeit. Unter Verwendung komparativer 2D DIGE-Analysen konnten zwei Strategien erfolgreich verfolgt werden: Zum einen wurde die Konsequenz einer Gendeletion von ausgewählten PKasen bzw. PPasen mit putativer mitochondrialer Lokalisation untersucht. Dabei gelang es, die an der in vivo Regulation des Pyruvatdehydrogenase(PDH)-Komplexes beteiligten Enzyme erstmalig zu identifizieren und im Folgenden deren regulatorisches Zusammenspiel umfassend zu analysieren. Zum anderen wurde in einem inversen Ansatz beispielhaft für die PKase Sat4p untersucht, welche Auswirkungen eine Überexpression dieser Kinase auf das mt Proteom hat. Erste Hinweise, welche zur Identifizierung der PDH-Kinasen (Pkp1p und Pkp2p) bzw. PDH Phosphatasen (Ppp1p und Ppp2p) führten, lieferten die signifikanten Spotänderungen von Pda1p (E1α-Untereinheit des PDH-Komplexes) in den 2D-DIGE-Analysen. Im Folgenden wurde die mitochondriale Lokalisation der vier regulatorischen Enzyme unter Verwendung Epitop-getaggter Varianten nachgewiesen sowie Pda1p in unabhängigen phosphospezifischen Analysen als Target der Phosphorylierung verifiziert. Die Phosphorylierungsstelle von Pda1p konnte massenspektrometrisch dem Serin313 zugeordnet werden. PDH-Aktivitätsmessungen zeigten, dass die Phosphorylierung von Pda1p den PDH Komplex inaktiviert, während eine Dephosphorylierung zur Aktivierung führt. Dabei war der Einfluss der Deletion der PDH Kinasen bzw. der PDH-Phosphatasen unterschiedlich stark ausgeprägt. Während Ppp1p und Ppp2p partiell redundante Funktionen besitzen, lassen die Analysen komplementäre Aktivitäten von Pkp1p und Pkp2p vermuten. Eine physikalische Interaktion der beiden Kinasen wurde in vivo nachgewiesen und deutet auf die Bildung funktioneller Heteromere hin. Durch Analysen in der 2D-BN/SDS-PAGE konnte eine Assoziation der PDH-Kinasen sowie PDH-Phosphatasen mit dem hochmolekularen, etwa 8 MDa großen PDH-Komplex sowie mit PDH-Subkomplexen geringeren Molekulargewichts gezeigt werden. Die Erkenntnisse dieser Arbeit ermöglichten in Verbindung mit denen eigener Vorarbeiten die Erstellung eines Modells zur PDH-Regulation in Saccharomyces cerevisiae. Neben der Aktivitätsregulation durch die von Pkp1p/Pkp2p bzw. Ppp1p/Ppp2p katalysierte Phosphorylierung wird eine Funktion der regulatorischen Enzyme – insbesondere der PDH-Kinasen – an der Assemblierung bzw. Stabilisierung des PDH-Komplexes postuliert. Es konnte somit gezeigt werden, dass in der Hefe ein ähnlicher, aber nicht identischer Regulationsmechanismus vorliegt wie in höheren Eukaryoten. Die zweite Strategie, welche in dieser Arbeit exemplarisch für eine PKase verfolgt wurde, führte zur Identifikation einer bislang unbekannten Funktion der Kinase Sat4p in den Mitochondrien. Es konnte gezeigt werden, dass Sat4p dual lokalisiert in der cytoplasmatischen sowie mitochondrialen Fraktion vorliegt und selbst Target der Phosphorylierung ist. Die Überexpression von Sat4p führte nicht nur zu einem verminderten Wachstum auf nicht fermentierbaren Medien, sondern auch zur Beeinflussung spezifischer mitochondrialer Proteingruppen. Während die Spots der Proteine Pil1p und Lsp1p eine höhere Intensität zeigten, wiesen die Fe/S-Proteine Aco1p und Lys4p eine verminderte „steady-state“-Konzentration auf. Darüber hinaus lagen die Proteine, welche Liponsäure als prosthetische Gruppe tragen (Lat1p, Kgd2p und Gcv3p), im Tet-Sat4-Stamm vorwiegend in ihrer nicht-lipoylierten Form vor. Die Lipoylierungsstellen aller drei Proteine konnten im Wildtyp unter Nutzung von nanoLC-MS/MS erstmals experimentell bestimmt und Lys75 (Lat1p), Lys114 (Kgd2p) bzw. Lys102 (Gcv3p) zugeordnet werden. Die fehlende Lipoylierung der Proteine bzw. die verminderte Proteinkonzentration der Aconitase führte zu einer stark verminderten Aktivität der betroffenen Enzymkomplexe. Neben den in der Literatur beschriebenen putativen Funktionen von Sat4p bei der Regulation cytoplasmatischer Proteine wurde basierend auf den Erkenntnissen der Analysen eine spezifische Funktion der Kinase in den Mitochondrien postuliert. Das Modell schlägt eine Rolle von Sat4p in den späten Schritten der Maturation einer spezifischen Gruppe von mitochondrialer Fe/S-Proteinen vor. Die Beeinträchtigung der Lipoatsynthase Lip5p, welche neben Aco1p und Lys4p ebenfalls zu dieser Gruppe gehört, führt vermutlich sekundär zum beobachteten Verlust der Lipoylierung von Lat1p, Kgd2p und Gcv3p.

This master's thesis project is the first half of a historical novel concerning the involvement of Elvira Field Strang Baker, the first plural wife of James Jesse Strang, with the "Beaver Island Mormons" who followed Strang from Nauvoo shortly after the death of Joseph Smith in 1844. The events portrayed are historical, although fictionalized. This portion of the novel contains a brief introduction to her childhood in Chapter One and follows her involvement with the Strangite movement beginning in April 1847 to the coronation of her husband in. Elvira was the first plural wife of James Jesse Strang who to this day is the only crowned American king. She married Strang in July 1849 and kept her marriage a secret for one year until Strang announced her as his wife during the above-mentioned coronation ceremony. Elvira was a woman ahead of her time. She was educated and had the opportunity to enjoy professional success which was rare for a woman of the mid-eighteenth century. She was a teacher, a trained tailor, an author of articles for her husband's newspapers, and one of his most capable administrators. While this portion of the novel focuses primarily on the early days of Elvira's acquaintance with James, his subsequent courtship, and the early days of their marriage, it also follows Elvira's movement within this unorthodox community that was supposed to be Zion.

The Oppression and Sexism of African-American Women: Then and Now: Substantial Contributions to the History of Musical Theatre

A wise Martin Luther King Jr. once said, "Freedom is never given by the oppressor; it must be demanded by the oppressed (King 1)." For as long as men and women have shared the planet, sexism has been a universal issue in civilization. In a social justice context, American society has found ways to oppress people for centuries. The Oxford Dictionary defines sexism as a "prejudice, stereotyping, or discrimination, typically against women, on the basis of sex ("sexism")." Voting rights in America were established in 1790, but it took years of petitioning at various women's rights conventions before the Nineteenth Amendment to the United States Constitution stating "the right of citizens of the United States to vote shall not be denied or abridged by the United States or by any State on account of sex" was passed in 1920 ("Nineteenth Amendment"). Traditionally, men were supposed to be the strong, decisive, driven, courageous, money-making breed, while women were expected to be the nurturing, affectionate, weak subordinates. Today, we find men and women working in careers previously linked with sexism; men as nurses and teachers, women as CEOs and factory workers. Statistics show that today there are an increasing number of women providing the financial support in their families. As with sexism, people also have been oppressed by racism for centuries. According to The Oxford Dictionary, racism is defined as a "prejudice, discrimination, or antagonism directed against someone of a different race based on the belief that one's own race is superior ("racism")." It has been argued that African Americans have been one of the most oppressed groups in America. Even after they were emancipated in 1865, it was nearly one hundred years later that their rights were protected with the Civil Rights Act of 1964. Before the act's passing, African Americans were denied equal education, employment, housing property, and a political voice. My interest in this topic was peaked right around the same time I became interested in performing on the musical theatre stage. I got my start in local community theatres, and up until college, was the only African American cast in the productions. I started playing multiple ensemble roles per show, and throughout the years advanced myself to "supporting character" but never the lead. Admittedly, there were times when I wasn't as talented as the women who snagged the leading roles, but many a time when I was just as talented or more qualified for the role, it went to another woman - most times of Caucasian descent. What did they have that I didn’t have? When I got accepted into The University of Central Florida as a BFA Musical Theatre student, I auditioned for the plays and musicals every semester, and each season I began to see the same patterns of who was cast for each show. Roles I thought I would get often went to White actors. I felt victimized in this modern-day example of racism. But racism goes beyond black and White. Internal racism between the light-skinned and dark-skinned African American women I was competing with became a factor as well. There were many times when an audition notice called for an African American woman; however, an unsettling trend became very apparent to me; if the casting description was for a maid, or something of that nature, larger, dark-skinned women would get the majority of the callbacks, which would lead to them getting cast. On the flip side, if an audition notice called for an African American ingénue type, more of the slimmer, lighter-skinned women were called back and later cast. Has American society cast a racial stigma for African American beauty?

Starting in November of 2016, a documentary series aired on the original network A&E where it continues to capture the attention of thousands of viewers in America. Scientology: The Aftermath, starring the well-known King of Queens and Dancing with the Stars actress Leah Remini and former senior executive of the church of Scientology International and Sea Organization Mike Rinder, aim to reveal the "truth" that hides behind the church of Scientology. The show interviews ex-practitioners who claim their lives have been deeply impacted by the church and who want to assist in revealing the shocking stories of abuse and harassment the church tries to keep secret. The goal of the show, in short, is to not only share the "real" face of Scientology to the public, but to also reach out and assist people who have been affected personally by the church; some have lost their family and friends while others have been followed or monitored by church members. A simple Google search on Scientology brings up multiple news headlines on the church being labeled as a "cult" and "criminal" while magazines in the lines of grocery stores display the downfall on the marriage of Tom Cruise and Katie Holmes due to the intensity of the Church of Scientology. Yet Scientology isn’t alone in receiving negative attention from the media. Christian Science has been accused of depriving the ill of needed medical attention which has led to some members becoming severely ill and dying in some cases. Most of these cases are children that were deprived of needed medical assistance due to their parents' religious views. Religious Studies scholar, Mary Bednarowski, adds that due to these circumstances, Christian Science has long experienced harsh criticism. With families and anti-cult movements protesting in the streets, ads, books, and magazines displaying their opposition, and the media labeling these movements as "dangerous", why would people join and remain in these New Religious Movements? What is it that makes these religious movements alluring and what are their adherents benefiting from them?

The spider mite occurrence, on soybean (Glycine max (L.) Merril), is sporadic and associated whit dry periods. Recently, in continuous years, was reported mite outbreaks in soybean, in some sites of Rio Grande do Sul State. In this context two research was conducted, one in summer crop 2004/05 and other in 2005/06. The first was carried for study the mite occurrence and geographic distribution, on soybean, from some place of Rio Grande do Sul State. The second was conducted for study the effect of the two soybean cultivars and of some weed management systems on mite density and study their population dynamic on soybean. In summer crop 2004/05 was carried some mite samplings in soybean field at State. The mite species found, all beloung to the Tetranychidae family, are Mononychellus planki (McGregor), Tetranychus desertorum Banks, T. gigas Pritchard & Baker, T. ludeni Zacher and T. urticae Koch. Most of this species are well distributed in the diferents sampling regions of this study. From the results was made a species distribution map and a taxonomic key. In summer crop 2005/06, in Santa Maria, RS, 11 treatments was study. Its was constitute in two soybean cultivars, one transgenic glyphosato-tolerant and other non-transgenic, every one whit five weed management systems: (1) without management; (2) hand management; (3) herbicide bentazone + sethoxydim; (4) bentazone only; (5) sethoxydim only. The 11th treatment was the herbicide glyphosate sprayed on the transgenic soybean. The mite species found in this study are M. planki and T. gigas. The weed management systems is more determinant, on the spider mite population on soybean, than the soybean cultivar. The mite population presenteded inverse correlation whit weed infestation and weed density. The climatic fators determined the mite population flutuation and the population peak occured in the initial period of soybean pods development. / A ocorrência de ácaros-praga em soja (Glycine max (L.) Merril) é esporádica e está, comumente, associada à períodos de estiagem. Recentemente, em safras seguidas, foram observados ataques severos de ácaros à soja em algumas regiões do estado do Rio Grande do Sul. Neste contexto foram realizados dois experimentos, um na safra agrícola 2004/05 e outro em 2005/06. O primeiro teve objetivo de estudar a ocorrência e a distribuição geográfica de ácaros fitófagos, associados à soja, em municípios de seis regiões produtoras do estado do Rio Grande do Sul. O segundo objetivou avaliar o efeito de duas cultivares de soja e de diferentes sistemas de manejo de plantas daninhas sobre a densidade populacional de ácaros em soja e estudar a dinâmica populacional destes ácaros na cultura. Em 2004/05 foram realizadas amostragens de ácaros em lavouras de soja no Estado. As espécies de ácaros encontradas são Mononychellus planki (McGregor), Tetranychus desertorum Banks, T. gigas Pritchard & Baker, T. ludeni Zacher e T. urticae Koch, todos são tetraniquídeos. A maior parte destas espécies estiveram bem distribuídas nas diferentes regiões amostradas neste levantamento. Com base nestes resultados foi elaborado um mapa com a distribuição das espécies e uma chave de identificação. Em 2005/06 foram estudados, em Santa Maria, RS, 11 tratamentos. Estes consistiram em duas cultivares de soja, uma transgênica glifosato-tolerante e a outra não-transgênica, cada uma sob cinco sistemas de manejo de plantas daninhas: (1) sem controle; (2) arranquio manual; (3) manejo tradicional com os herbicidas bentazona e setoxidim; (4) apenas com bentazona; (5) apenas com setoxidim. E adicionalmente um tratamento com o herbicida glifosato sobre a cultivar de soja transgênica. Os ácaros-praga ocorrentes foram M. planki e T. gigas. A densidade populacional destes ácaros em soja foi mais afetada pelos sistemas de manejo de plantas daninhas do que pela cultivar, e esteve inversamente correlacionada com a quantidade e diversidade de plantas daninhas. A flutuação populacional destes ácaros foi influenciada por fatores climáticos e o pico populacional ocorreu na fase de início da formação de legumes da soja.
