HISTAMINA E TIRAMINA EM EMBUTIDOS CÁRNEOS / HISTAMINE AND TYRAMINE IN MEAT SAUSAGES

The consumption of fermented goods has been related with outbreaks of foods intoxications, like these biogenic amines have been associated to the consummations of these products. The aims went to assess the potential of amines formation in dry fermented sausage added or not with starter and fiber, determination of amines formation in cooked salami added or not with organic acids. The dry fermented sausages were fabricated and cased in artificial collagen no eatable gut then, ripening under controlled conditions of temperature, moisture and air stream. The cooked salamis were added with organic acids (lactic and citric), cased in artificial collagen no eatable gut and cooked. The products were according to the microbiologic available legal patterns. The histamine reached amounts of 145,61; 93,38 and 130,10 mg·kg-1 and the tyramine reached amounts of 67,05; 70,28 and 71,87 mg·kg-1 for the dry fermented sausages treatments: control, starter and starter and fiber respectively. The histamine in the cooked salami treatments: control, lactic acid and citric acid reached amounts of 48,06; 55,37 and 49,16 mg·kg-1 and the tyramine reached amounts of 11,00; 11,06 and 11,35 mg·kg-1 respectively. There were no differences at the level tested (p<0,05). In conclude the starter did not avoid the amine formation, attributed to the polluting bacterium found already in the raw materials. The temperature and the organic acids did not influence in the final amounts of amines in the cooked salami. The taste assess was not influenced by the presence of amines. The addition of glucose and sucrose avoided larger amines amounts in the fermented salami and the wheat fiber did not interfere in the metabolism of the microorganisms. / O consumo de alimentos fermentados tem sido relacionado com surtos de intoxicações alimentares e as aminas biogênicas são associadas ao consumo destes produtos. Os objetivos foram avaliar o potencial de formação de aminas em salame tipo Italiano, adicionado ou não, de cultura starter e fibra; determinar a formação de aminas no salame cozido, adicionado ou não de ácidos orgânicos. Os salames foram elaborados e embutidos em tripa artificial de colágeno não comestível. Maturados sob condições de temperatura, umidade e velocidade do ar controlados. Na fabricação do salame cozido, foram adicionados ácidos orgânicos (láctico e cítrico), embutidos em tripa artificial de colágeno não comestível e as peças cozidas. Os produtos se apresentaram conforme os padrões microbiológicos vigentes. A histamina alcançou valores de 145,61; 93,38 e 130,10 mg·kg-1 e a tiramina valores de 67,05; 70,28 e 71,87 mg·kg-1 para os tratamentos do salame controle, starter e starter e fibra respectivamente. A histamina alcançou valores de 48,06; 55,37 e 49,16 mg·kg-1 e a tiramina valores de 11,00; 11,06 e 11,35 mg·kg-1 para os tratamentos do salame cozido controle, ácido láctico e ácido cítrico respectivamente. Não houve diferenças ao nível testado (p<0,05). Conclui-se que o starter comercial não evitou a formação de aminas, atribuída às bactérias contaminantes encontradas já nas matérias-primas. A temperatura e os ácidos orgânicos não influenciaram nos níveis finais de aminas no salame cozido. A análise sensorial não foi influenciada pela presença das aminas. A adição de glicose e sacarose evitou maiores acúmulos de aminas no salame fermentado e a fibra de trigo não interferiu no metabolismo dos microrganismos.

The main aim of this thesis was to prove the hypothesis that the intake of blood with specific antibodies against sand fly saliva affects sand fly fecundity and mortality. Phlebotomus argentipes and Mesocricetus auratus were used for most experiments. ELISA revealed high levels of specific antibodies in repeatedly exposed hosts. However, a five-day study of mortality and fecundity of bloodfed females demonstrated that feeding on repeatedly bitten hamsters has no effect on number of eggs or survival of females. Salivary antigens of P. argentipes recognized by sera of repeatedly bitten hamsters were characterized by immunoblotting and mass spectrometry. Immunoblotting showed that antibodies in the hamster sera recognize salivary proteins with molecular weight from 25 to 60 kDa. Mass spectrometry revealed that the antigens correspond to D7, apyrases, antigen 5-related proteins and yellow-related proteins. In addition, Phlebotomus females were fed through a chicken membrane on rabbit blood with high concentrations of histamine, serotonin or prostaglandin E2 to find out if they influence fecundity or mortality of sandfly females. Etude du métabolisme des amines biogènes chez les bactéries lactiques du vin / Study of biogenic amines metabolism in wine lactic acid bacteria

Les amines biogènes sont des composés allergènes, notamment rencontrés dans les produits fermentés tel que le vin. Les bactéries lactiques du vin, dont Oenococcus oeni, le principal acteur de la fermentation malolactique, sont capables de produire ces molécules, à partir de précurseurs azotés. Afin de réduire la teneur en amines biogènes, il est nécessaire de comprendre le rôle de cette production, chez les souches impliquées dans la synthèse de ces métabolites. Le projet européen BiamFood FP7 (n°211441) a été mis en place pour répondre à cette problématique. Au cours de mon travail de thèse, des outils moléculaires ont été développés afin de muter de manière ciblée les gènes de O. oeni et d’exprimer des gènes d’intérêt. Ainsi, les clusters hdc de Lactobacillus hilgardii et odc de O. oeni, responsables respectivement de la synthèse de l’histamine et de la putrescine, ont été clonés dans le vecteur pGID052 et transférés dans la souche de laboratoire O. oeni ATCC BAA-1163, déficiente pour la production d’amines biogènes. Cette stratégie n’ayant pas donnée lieu à la production d’amine biogène, mon travail de recherche s’est orienté vers la caractérisation biochimique de l’ornithine décarboxylase de O. oeni. L’ornithine décarboxylase de O. oeni BR14/97 a été sur-produite chez E. coli via le système pET. Après purification de l’enzyme, le pH optimal et la température optimale de fonctionnement, 5,5 et 35°C respectivement, ont été caractérisés. Les constantes cinétiques Km = 1 mM et Vmax = 0,57 U. mg-1 ont également été déterminées en mesurant la putrescine produite par HPLC (Gomez-Alonso et al, 2007). De plus, l’ODC de O. oeni BR14/97 est spécifique de la L-ornithine et ne peut pas dégrader la L-lysine en cadavérine, ce qui implique la présence d’une voie métabolique propre à la production de cadavérine chez cette souche. Dans un second axe du projet, l’influence de la source azotée et notamment des peptides versus acides aminés libres, a été étudiée chez une souche de Lactobacillus plantarum, isolée du vin et productrice de tyramine. Les peptides contenant de la tyrosine, sont utilisés par cette souche pour former de la tyramine. La production augmente en phase stationnaire de croissance, et est corrélée à l’expression du transporteur tyrP. L’analyse de la tyrosine présente dans le milieu de culture montre que ces peptides seraient dégradés dans le milieu extracellulaire chez L. plantarum. / Biogenic amines are indesirable compounds found in fermented products like wine. Lactic acid bacteria from wine, including Oenococcus oeni, the main actor of malolactic fermentation, are able to produce these molecules from nitrogenous precursors. In order to limited biogenic amines accumulation, it is necessary to understand the role of this production by strains responsible for the synthesis of these metabolites in food. That is why the European BiamFood project was put in place. Along my thesis, molecular tools were developed in order to mutate O. oeni genes (encoding decarboxylases), and to express genes of interest. Genetic clusters hdc from L. higardii and odc from O. oeni, responsible for histamine and putrescine synthesis respectively, were cloned into the pGID052 vector and transferred into the laboratory strain O. oeni ATCC BAA-1163, which is not able to produce any biogenic amines. However no biogenic amines production was observed. My work thesis was then redirected in the in vitro biochemical characterization of the ODC from O. oeni. The odc from O. oeni BR14/97 was overproduced in E. coli BL21 via the pET system. Optima pH and temperature were determined with the purified enzyme obtained. Kinetic constants Km = 1 mM and Vmax = 0,57 U.mg-1 were also determined by measuring putrescine production by HPLC (Gomez-Alonso et al, 2007). Furthermore, ODC from O. oeni BR14/97 is specific for the L-ornithine and can not decarboxylated the L-lysine into cadaverine. In a second part of the BiamFood project, the impact of nitrogen sources, especially peptides compared to free amino acids, was studied in a Lactobacillus plantarum strain, isolated from wine and able to produce tyramine. Peptides containing tyrosine residues are used by this strain to produce tyramine. Tyramine production increases during the growth and reaches a maximum at stationary phase. The production is correlated with tyrP expression. The tyrosine measured in culture media shows that peptides may be hydrolyzed in the extracellular medium in L. plantarum.

Várias soluções tecnológicas foram propostas para melhorar a cadeia produtiva, como a combinação de uvas e até mesmo o aproveitamento do resíduo do processo de produção, contribuindo na qualidade funcional, econômica e ambiental. No Brasil, uvas Vitis labrusca e híbridas representam a maioria daquelas destinadas aos sucos. A uva híbrida 'Niagara Rosada' que apresenta alta produtividade e aceitabilidade, com excedente de produção, pode ser uma opção para a produção de suco e utilização do resíduo para suplementação alimentar. Dessa forma, o perfil de compostos fenólicos e nitrogenados, bem como capacidade antioxidante e bioativa, coloração e sensorial foram realizadas com o objetivo de verificar a qualidade dos sucos de uva e seus resíduos produzidos a partir de combinações de 'Niagara Rosada'. Os sucos foram elaborados com 'Bordo', 'BRS Violeta', 'IAC 138-22 Máximo', separadamente e em blends com 'Niagara Rosada'. Os sucos foram produzidos por prensagem a quente, engarrafados (vidro âmbar) e pasteurizados. Um controle comercial (C) de 'Niagara Rosada' também foi analisado junto aos sucos. Os resíduos da produção do suco (bagaços) foram secos, moídos, embalados e armazenados para as análises. Various technological solutions have been proposed to improve the production chain, such as the combination of grapes and even the use of the residue of the production process, contributing to functional, economic and environmental quality. In Brazil, Vitis labrusca and hybrid grapes represent the majority of those destined for juices. The hybrid grape 'Niagara Rosada', which presents high productivity and acceptability, with surplus production, can be an option for the production of juice and use of the residue for food supplementation. Thus, the profile of phenolic and nitrogen compounds, as well as antioxidant and bioactive capacity, coloring and sensory were carried out with the aim of verifying the quality of grape juices and their residues produced from 'Niagara Rosada' combinations. The juices were made with 'Bordo', 'BRS Violeta', 'IAC 138-22 Máximo', separately and in blends with 'Niagara Rosada'. The juices were produced by hot pressing, bottled (amber glass) and pasteurized. Novos dispositivos de microextração e pré-concentração para eletroforese capilar utilizando membranas microporosas ou processos eletroquímicos e sua potencialidade na análise de aminas e metais / New devices to extraction and preconcentration with microporous membranes and electrochemical procedures prior to capillary electrophoresis: some applications to amines and metals analysis

A eletroforese capilar (CE) é uma técnica de separação eficiente, que tem merecido grande atenção nas últimas duas décadas, e que oferece soluções analíticas alternativas ou complementares à cromatografia líquida num número crescente campos de aplicação, incluindo análises de fármacos, compostos biológicos e biogênicos, dentre outros. Entre as vantagens comparativas da técnica figuram separações mais rápidas com pequeno dispêndio de amostras, solução tampão (e solvente orgânico, se usado), baixo custo de operação e manutenção. Todavia, a exigüidade de amostra no capilar tem como reflexo, via de regra, limites de detecção menos favoráveis que os da HPLC, uma limitação significativa na análise de traços, suplantável por técnicas e procedimentos de extração/acumulação prévia dos analitos. Nesta tese concebeu-se e implementou-se a combinação inédita entre a CE e a préconcentração eletroquímica de metais. Novos dispositivos para extração líquido/gás/líquido mediada por membranas microporosas também foram propostos e aplicados à analise de aminas voláteis, sempre utilizando detector de condutância medida sem contato direto com a solução (C4D, contactless capacitively coupled conductivity detection). Primeiramente, sem recorrer à preconcentração, definiu-se condições de análise para 16 aminas (metilamina, dimetilamina, trimetilamina, propilamina, n-butilamina, s-butilamina, t-butilamina, hexilamina, DMAE, etanolamina, dietanolamina, trietanolamina, 1,3-diaminopropano, 1,4-diaminobutano, 1,5-diaminopentano e 1,6-diaminohexano) juntamente com íon amônio, e, para análise de 9 metais (TI++, Cd2+, Pb2+,Cr3+, Zn2+, Ag+, Cu2+, Co2+ e Ni2+). Um método completo para análise de dimetilaminoetanol em formulações cosméticas por CE-C4D foi desenvolvido e validado. Os estudos de extração e pré-concentração líquido/gás/líquido foram realizados utilizando filamento oco de Oxyphan preenchido com solução coletora. Foi construída cela para extração gasosa constituída de recipiente fechado no qual se introduz amostra com analitos voláteis, ou volatilizáveis pela adição de reagente e/ou aquecimento (pervaporação). A bobina coletora de filamento oco preenchido com solução coletora foi montada na parte interna da tampa da cela. Testes realizados com amostras de tecidos de peixe demonstraram o funcionamento do sistema, que poderá ser aperfeiçoado e aplicado, por exemplo, à análise de aminas biogênicas primárias de baixo peso molecular, indicadoras de decomposição de certos alimentos. Criou-se também extrator microvolumétrico em que o filamento de Oxyphan é montado concentricamente no interior de um capilar de sílica fundida, para realização de estudos de extração líquido/gás/líquido em fluxo. Um sistema em fluxo compreendendo válvulas e microbombas foi montado e colocado sob controle de um programa de computador (Labview), que, entre outras funções, controla o deslocamento da solução coletora exposta no filamento oco (13 &#181;L) até o ponto exato da interface em que se dá a injeção no capilar da CE. Pela primeira vez recorreu-se à acumulação eletroquímica (ECPC) de analitos num sistema FIA-CE-C4D - uma configuração baseada inteiramente na eletroquímica -, visando expandir os limites de detecção das determinações e eliminar interferência de matriz por troca de meio. Implementou-se a pré-concentração catódica seguida de redissolução anódica com uma célula eletroquímica especial, em que o capilar da eletroforese é posicionado junto ao eletrodo de trabalho, feito de ouro obtido a partir de CD\'s graváveis. Uma célula microfluídica ECPC-FIA-CE-C4D também foi construída. Tomando íons Cu2+ como sistema modelo, demonstrou-se o aumento do sinal referente aos íons cobre(lI) no eletroferograma para tempos crescentes de acumulação eletroquímica. Para a solução de alto teor salino contendo mistura dos íons Cd(II), Pb(II), Zn(lI) e Cu(II), comprovou-se a eliminação da interferência da matriz por troca de meio antes da redissolução anódica. Os testes iniciais indicam que os novos dispositivos e sistemas propostos nesta tese para uso em conjunto com a CE têm potencialidade para desencadear novas pesquisas, desenvolvimento de métodos e aplicações, sendo passíveis também de miniaturização e automação. / Capillary electrophoresis (CE) is an efficient separation technique that evolved rapidly in the last couple of decades, now offering complementary or alternative analytical solutions to liquid chromatography for an expanding number of application fields including drugs, biological and biogenic compounds. Some comparative advantages of CE are faster separation, very low consumption of buffer solutions and samples (and organic solvents, if any), lower maintenance and operational costs. However, the quantification of a few nanoliters of analyte in a capillary, as a rule, compromises the detection limits attainable by HPLC, a shortcomming in trace analysis, suplantable by extraction/preconcentration of the analytes from the sample. The combination of electrochemical preconcentration (ECPC) with CE is first proposed in this thesis and tested with metal ions. New devices for liquid/gas/liquid extraction based on microporous filaments filled with a collector solution were proposed and applied to the analysis of volatile amines, The detector of choice was the C4D (capacitively coupled contactless conductivity detection) and CE-C4D working conditions were defined for the separation and determination of: i) 16 amines (methylamine, dimethylamine, trimethylamine, propylamine, n-butylamine, s-butylamine, t-butylamine, hexilamine, ethanolamine, diethanolamine, triethanolamine, DMAE, 1,3-diaminopropane, 1,4- diaminobutane, 1,5-diaminopentane and 1,6-diaminohexane) plus ammonium, ii) nine metallic ions (Tl+, Cd2+, Pb2+, Cr3+ Zn2+, Ag+, Cu2+, Co2+ e Ni2+). A complete method of analysis for Dimethyl-amino-ethanol (DMAE) in cosmetic formulations was established and validated. Liquid/gas/liquid extraction studies were based on the use of a polypropylene microporous filment (Oxyphan® ) filled with the collection solution. A closed cell was devised in which an Oxyphan® coil is installed beneath the cap and volatilization of sample components can be promoted by heating (pervaporation) or addition of a reactant. The device was successfully tested with fish samples by the detection of primary low molecular weight biogenic amines, a procedure of special interest in fast checking of fish or other foods for decomposition. A microvolumetric version of the liquid/gas/liquid extractor was created with an Oxyphan® hollow fiber mounted concentrically inside a silica capillary. A flow system comprising the micropumps, solenoid valves and the microextrator was designed, automatically operated under software control (Labview). This FIA-CE system was tested by the extraction/enrichment of volatile amines in the collecting solution (13 &#181;L) followed by accurate displacement to the interface with the CE capillary by the micropumps. Pioneering work with ECPC-FIA-CE-C4D an all-electrochemical system was conducted in this thesis aiming expanded detection limits and matrix effect reduction. An electrochemical cell for the interface with FIA-CE system was built in acrylic using gold electrodes made from gold-sputtered CD-Rs. A microfluidic version of the cell interfaced with the CE capillary was also conceived. The full system was evaluated by using Cu2+ as a model specie; an increase in analytical signal was observed as a function of the cathodic deposition time before stripping. For a mixture of Cd2+, Cu2+, Zn2+ and Pb2+ in a high saline media, elimination of the matrix interference by medium exchange before stripping was demonstrated. Initial favorable evaluations show that the devices and systems proposed in 12 the thesis have the potential to engender further research of methods and applications as well as miniaturization and automation of procedures.

Tvorba biogenních aminů v mase vybraných druhů ryb / The formation of biogenic amines in flesh of selected fish species

The thesis deals with the use and effectiveness of some less common methods of conservation of fish meat. The formation of biogenic amines in meat is observed in connection with the non-traditional preservative methods. Amines can serve as indicators of protein degradation. The quality of fish was considered in connection with the increasing content of selected biogenic amines (putrescine, cadaverine, spermidine, spermine, 2-fenylathylamine, histamine, tyramine and tryptamine). Ultra performance liquid chromatography (UPLC) was used as the method for determination of biogenic amines. Amines were derivatized by dansylchloride before their UPLC separation. The fish flesh was vacuum-packed. Samples were stored for several weeks in a thermostat at the selected storage temperature after the application of selected preservative technique. Beta-irradiation and high hydrostatic pressure were used for the preservation of fish flesh. Control samples were not exposed to the â-irradiation and high pressure. The organoleptic properties were studied for all samples (smell/odor, insight and shape). Beta-irradiation was applied to fish meat of common carp (Cyprinus carpio) and rainbow trout (Oncorhynchus mykiss). Both these species of freshwater fish are economically significant. Carp and trout are the species being mostly consumed in the Czech Republic. Fish meat is considered to be provided the flesh is fresh. A testing series of samples was created with common carp to determine the appropriate dose of â-irradiation. The maximum permissible dose of irradiation for fish meat is 3 kGy. Fish samples were exposed this dose in the first experiment. The dose of irradiation was reduced in following experiments based on the experience from the initial experiment. The doses of 0.25, 0.5, 0.75, 1.0 and 2.0 kGy were applied to rainbow trout. The value of 0.75 kGy of â-irradiation or higher (1.0, 2.0 and 3.0 kGy) prolonged the shelf life of fish meat, which was stored for three months (98 days). Prolonging of the shelf life of fish meat to approximately 98 days at 3.5 °C is redundant from technical point of view. For that reason lower doses 0.25, 0.5 and 0.75 kGy were tested in more detail in the repeated experiment with carp meat. Lower doses of â-irradiation are considered to be more acceptable and-at the same time-sufficiently effective for delaying the beginning of degradation processes. 6 High hydrostatic pressure was applied to meat of common carp (Cyprinus carpio), rainbow trout (Oncorhynchus mykiss) and pike (Exos lucius). Pike is another very popular kind of freshwater fish. Pike flesh is very tasty, but in spite of this, pike is not so much popular among consumers compared to carp and trout. The cause is its high price. Samples of pike were stored at standard temperature 3.5 °C and also at higher temperature 12 °C (unlike experiments with â-irradiation). Lower temperature of storage (3.5 °C) followed the conditions of storing of fish meat in industrial refrigeration facilities and households. The high pressure might not be sufficient for preservation at higher storage temperatures. This assumption was based on available information. Samples were treated by high pressure and stored at both 3.5 °C and 12 °C to verify this assumption. Higher temperature simulated either failure of refrigeration equipment or unsuitable store temperature of meat. In all species selected freshwater fish two levels of high pressure were applied ? 300 and 500 MPa. Both levels had significantly reduced the formation of biogenic amines, especially in samples stored at 3.5 °C. At this temperature, the effect of 300 and 500 MPa delayed start of degradation processes in fish meat by 3?4 weeks. At 12 °C and 500 MPa, high pressure extended the sustainability of meat by no more than one week. 500 MPa is effective treatment at the lower temperature of 3.5 °C. High pressure is not reliable preservative techniques at higher temperature.

Todavia, a exigüidade de amostra no capilar tem como reflexo, via de regra, limites de detecção menos favoráveis que os da HPLC, uma limitação significativa na análise de traços, suplantável por técnicas e procedimentos de extração/acumulação prévia dos analitos. Nesta tese concebeu-se e implementou-se a combinação inédita entre a CE e a préconcentração eletroquímica de metais. Novos dispositivos para extração líquido/gás/líquido mediada por membranas microporosas também foram propostos e aplicados à analise de aminas voláteis, sempre utilizando detector de condutância medida sem contato direto com a solução (C4D, contactless capacitively coupled conductivity detection). Primeiramente, sem recorrer à preconcentração, definiu-se condições de análise para 16 aminas (metilamina, dimetilamina, trimetilamina, propilamina, n-butilamina, s-butilamina, t-butilamina, hexilamina, DMAE, etanolamina, dietanolamina, trietanolamina, 1,3-diaminopropano, 1,4-diaminobutano, 1,5-diaminopentano e 1,6-diaminohexano) juntamente com íon amônio, e, para análise de 9 metais (TI++, Cd2+, Pb2+,Cr3+, Zn2+, Ag+, Cu2+, Co2+ e Ni2+). Um método completo para análise de dimetilaminoetanol em formulações cosméticas por CE-C4D foi desenvolvido e validado. Os estudos de extração e pré-concentração líquido/gás/líquido foram realizados utilizando filamento oco de Oxyphan preenchido com solução coletora. Foi construída cela para extração gasosa constituída de recipiente fechado no qual se introduz amostra com analitos voláteis, ou volatilizáveis pela adição de reagente e/ou aquecimento (pervaporação). A bobina coletora de filamento oco preenchido com solução coletora foi montada na parte interna da tampa da cela. Testes realizados com amostras de tecidos de peixe demonstraram o funcionamento do sistema, que poderá ser aperfeiçoado e aplicado, por exemplo, à análise de aminas biogênicas primárias de baixo peso molecular, indicadoras de decomposição de certos alimentos. Criou-se também extrator microvolumétrico em que o filamento de Oxyphan é montado concentricamente no interior de um capilar de sílica fundida, para realização de estudos de extração líquido/gás/líquido em fluxo. Um sistema em fluxo compreendendo válvulas e microbombas foi montado e colocado sob controle de um programa de computador (Labview), que, entre outras funções, controla o deslocamento da solução coletora exposta no filamento oco (13 &#181;L) até o ponto exato da interface em que se dá a injeção no capilar da CE. Pela primeira vez recorreu-se à acumulação eletroquímica (ECPC) de analitos num sistema FIA-CE-C4D - uma configuração baseada inteiramente na eletroquímica -, visando expandir os limites de detecção das determinações e eliminar interferência de matriz por troca de meio. Implementou-se a pré-concentração catódica seguida de redissolução anódica com uma célula eletroquímica especial, em que o capilar da eletroforese é posicionado junto ao eletrodo de trabalho, feito de ouro obtido a partir de CD\'s graváveis. Uma célula microfluídica ECPC-FIA-CE-C4D também foi construída. Tomando íons Cu2+ como sistema modelo, demonstrou-se o aumento do sinal referente aos íons cobre(lI) no eletroferograma para tempos crescentes de acumulação eletroquímica. Para a solução de alto teor salino contendo mistura dos íons Cd(II), Pb(II), Zn(lI) e Cu(II), comprovou-se a eliminação da interferência da matriz por troca de meio antes da redissolução anódica. Os testes iniciais indicam que os novos dispositivos e sistemas propostos nesta tese para uso em conjunto com a CE têm potencialidade para desencadear novas pesquisas, desenvolvimento de métodos e aplicações, sendo passíveis também de miniaturização e automação. / Capillary electrophoresis (CE) is an efficient separation technique that evolved rapidly in the last couple of decades, now offering complementary or alternative analytical solutions to liquid chromatography for an expanding number of application fields including drugs, biological and biogenic compounds. Some comparative advantages of CE are faster separation, very low consumption of buffer solutions and samples (and organic solvents, if any), lower maintenance and operational costs. However, the quantification of a few nanoliters of analyte in a capillary, as a rule, compromises the detection limits attainable by HPLC, a shortcomming in trace analysis, suplantable by extraction/preconcentration of the analytes from the sample. The combination of electrochemical preconcentration (ECPC) with CE is first proposed in this thesis and tested with metal ions. New devices for liquid/gas/liquid extraction based on microporous filaments filled with a collector solution were proposed and applied to the analysis of volatile amines, The detector of choice was the C4D (capacitively coupled contactless conductivity detection) and CE-C4D working conditions were defined for the separation and determination of: i) 16 amines (methylamine, dimethylamine, trimethylamine, propylamine, n-butylamine, s-butylamine, t-butylamine, hexilamine, ethanolamine, diethanolamine, triethanolamine, DMAE, 1,3-diaminopropane, 1,4- diaminobutane, 1,5-diaminopentane and 1,6-diaminohexane) plus ammonium, ii) nine metallic ions (Tl+, Cd2+, Pb2+, Cr3+ Zn2+, Ag+, Cu2+, Co2+ e Ni2+). A complete method of analysis for Dimethyl-amino-ethanol (DMAE) in cosmetic formulations was established and validated. Liquid/gas/liquid extraction studies were based on the use of a polypropylene microporous filment (Oxyphan® ) filled with the collection solution. A closed cell was devised in which an Oxyphan® coil is installed beneath the cap and volatilization of sample components can be promoted by heating (pervaporation) or addition of a reactant. The device was successfully tested with fish samples by the detection of primary low molecular weight biogenic amines, a procedure of special interest in fast checking of fish or other foods for decomposition. A microvolumetric version of the liquid/gas/liquid extractor was created with an Oxyphan® hollow fiber mounted concentrically inside a silica capillary. A flow system comprising the micropumps, solenoid valves and the microextrator was designed, automatically operated under software control (Labview). This FIA-CE system was tested by the extraction/enrichment of volatile amines in the collecting solution (13 &#181;L) followed by accurate displacement to the interface with the CE capillary by the micropumps. Pioneering work with ECPC-FIA-CE-C4D an all-electrochemical system was conducted in this thesis aiming expanded detection limits and matrix effect reduction. An electrochemical cell for the interface with FIA-CE system was built in acrylic using gold electrodes made from gold-sputtered CD-Rs. A microfluidic version of the cell interfaced with the CE capillary was also conceived. The full system was evaluated by using Cu2+ as a model specie; an increase in analytical signal was observed as a function of the cathodic deposition time before stripping. For a mixture of Cd2+, Cu2+, Zn2+ and Pb2+ in a high saline media, elimination of the matrix interference by medium exchange before stripping was demonstrated. Initial favorable evaluations show that the devices and systems proposed in 12 the thesis have the potential to engender further research of methods and applications as well as miniaturization and automation of procedures.

After 0, 7, 14, 35 and 56 days of maturation were made the determination of bioactive amines (putrescine, cadaverine, histamine, tiramine, serotonin, agmatine, tryptamine, feniletilamine, espermine and espermidine), standard count in plaque of aerobes microorganisms and total count of lactic bacteria. The count of microorganisms of the samples has correlated positively during the storage time (r>0,91 e p<0,001), achieving maximum scores of 6,67 and 7,6 LogUFC.g-1 for viable microorganisms aerobes and lactic bacteria, respectively, after 56 days. The breed variation of the animals and the electric stimulation of the carcass have not demonstrated to cause significant effects in the microbiotics and tenor of bioactive amines during the period observed, except as for the tenors of espermine, after 14 days, when a bigger concentration of such polyamine was detected in the mixed breed meat. Significant positive correlations (p<0,001), were also observed between the lactic bacteria count and the total of bioactive amines in the meat of carcass electrically stimulated, in both Red Angus and zebuines mixed breeds, which it did not occur in the meat of carcass without post-mortem electric stimulation. . / Aminas bioativas são bases orgânicas de baixo peso molecular encontradas em uma gama de alimentos de origem animal e vegetal e estão frequentemente envolvidas no desenvolvimento e potencialização de patologias humanas importantes, como desordens neurológicas, doenças gastrointestinais, hipo e hipertensão, respostas imunes anormais e câncer. O processo de maturação da carne bovina favorece o crescimento de bactérias anaeróbias facultativas, sobretudo bactérias láticas, as quais contribuem para a produção e aumento da concentração de aminas em alimentos. Amostras de contra-filé (músculo Longissimus Dorsi) de 12 carcaças de bovinos Red Angus e mestiços zebuínos, abatidos com aproximadamente 350kg e 3 anos de idade, com e sem estimulação elétrica da carcaça (estimulação de baixa voltagem), foram acondicionadas a vácuo e maturadas sob refrigeração por até 56 dias ( 1,0±0,5ºC). Aos 0, 7, 14, 35 e 56 dias de maturação foram efetuadas as determinações de aminas bioativas (putrescina, cadaverina, histamina, tiramina, serotonina, agmatina, triptamina, feniletilamina, espermina e espermidina), contagem total de bactérias láticas e contagem padrão em placa de microrganismos aeróbios. As contagens de bactérias láticas e microrganismos aeróbios correlacionaram-se positivamente com o tempo de maturação, atingindo ao final do período 6,67 e 7,6 LogUFC.g-1 para microrganismos aeróbios viáveis e bactérias láticas, respectivamente. Entre os diferentes tratamentos estudados, não foram observadas diferenças estatisticamente significativas nos teores de aminas bioativas e contagens de microrganismos (aeróbios e bactérias láticas), exceto aos 14 dias, quando os teores de espermina em amostras de carne de animais mestiços foram significativamente maiores (p<0,05) do que em carnes de Red Angus, com e sem estimulação elétrica. Em tratamentos iguais e ao longo do período de armazenamento houve aumento significativo tanto nas contagens de microrganismos aeróbios e bactérias lácticas quanto nos teores totais de aminas bioativas. Esse aumento é particularmente significativo a partir do 35º dia de maturação. Correlações positivas significativas (p<0,001) foram observadas entre contagem de bactérias láticas, total de aminas bioativas e tempo de maturação em amostras de carne de carcaças estimuladas eletricamente, tanto de animais Red Angus quanto mestiços zebuínos, o que não ocorreu nas carnes de carcaças que não sofreram estimulação elétrica post-mortem. .

Neuroendocrine Modulation of Complex Behavior and Physiology in C. elegans

To survive, animals must adapt to a complex and challenging world in a way that is flexible and responsive, while maintaining internal homeostasis. Neuromodulators provide a means to systemically alter behavioral or physiological state based on intrinsic or extrinsic cues, however dysregulated neuroendocrine signaling has negative consequences for fitness and survival. Here I examine neuroendocrine function and dysfunction using the escape response in Caenorhabditis elegans. The RFamide neuropeptide FLP-18 is a co-transmitter with the monoamine tyramine and functions both synergistically and antagonistically to tyramine in coordinating escape behavior. Using behavioral analysis and calcium imaging, I show that FLP-18 functions primarily through the G-protein coupled receptor (GPCR) NPR-5 to increase calcium levels in muscle, enhancing locomotion rate, bending and reversal behavior during the escape response. Furthermore, I examine the relationship between persistent acute stress and resilience using repeated activation of the escape response as a model of neuroendocrine dysregulation. Repeated activation of the escape response shortens lifespan and renders animals more susceptible to thermal, oxidative, and nutritional stress. Tyramine release is necessary and sufficient for this effect and activity of the tyraminergic RIM neurons is differentially regulated by acute versus long-term stressors. Impaired stress resistance requires both the GPCR TYRA-3 in the intestine and intestinal neuropeptide release. Activation of the insulin receptor DAF-2 is downstream of TYRA-3 and inhibits the transcription factors DAF-16/FOXO, SKN-1/Nrf2 and HSF-1, linking monoamine signaling in acute stress to the insulin signaling pathway and impaired resilience to long-term stressors.
