Estudo do crescimento celular em culturas embriogênicas e não-embriogênicas de cana-de-açúcar. / Study of cell growth in embryogenic and non-embryogenic cultures of sugarcane.

Muitos estudos têm sido direcionados para gerar melhorias na cultura de cana-de-açúcar, sendo uma das alternativas o uso de técnicas biotecnológicas, como a embriogênese somática (ES). O objetivo deste projeto foi estudar a multiplicação celular na ES de cana-de-açúcar da variedade SP80-3280. Culturas embriogênicas (CE) e não-embriogênicas (CNE) foram monitoradas e parâmetros bioquímicos foram analisados. Foi observado que CE e CNE apresentaram crescimento máximo aos 24 dias de cultivo, sendo que as CE apresentaram maior incremento em matéria fresca. Dos carboidratos analisados, a sacarose foi predominante em CE, enquanto a glicose predominou em CNE. Os conteúdos de glicose e frutose variaram simultaneamente em CE. As concentrações de amido, poliaminas (Pas) totais e a razão entre PAs [Razão PA= Put/(Spd+Spm)] apresentaram maiores valores nas CNE, sendo a espermidina a poliamina predominante em CE e putrescina em CNE. O conteúdo de proteínas totais foi significativamente maior em CE, em todas as fases de crescimento. / Many studies have been directed to generate improvements in the sugarcane cultures, one of the alternative use of biotechnology techniques such as somatic embryogenesis (SE). The objective of this project was to study the cell multiplication in SE of sugarcane, variety SP80-3280. Embryogenic cultures (EC) and non-embryogenic (NEC) were monitored and biochemical parameters were analyzed. Was observed that EC and NEC showed maximum growth to 24 days of culture, and the EC showed greater increase in fresh weight. Of the carbohydrates tested, sucrose was predominant in EC, while glucose was predominant in the NEC. The contents of glucose and fructose varied simultaneously in EC. The concentrations of starch, polyamines (PAs) and the ratio of total PAs [Ratio PA = Put / (Spd + Spm)] showed higher values in the NEC, and the spermidine is predominant polyamine to EC and putrescine is predominant in NEC. The content of total protein was significantly higher in CE, at all stages of growth.

Caracterização química e enzimática do processo de adoçamento da manga \'Keitt\' / Chemical and enzymatic characterization of the mango Keitt sweetening process

Dentre as características que definem um fruto maduro, o adoçamento é um dos mais importantes. Porém, no que concerne à manga, os dados existentes são escassos e pouco esclarecedores. Neste trabalho foi estudada a síntese e a degradação do amido da manga \'Keitt\', nos aspectos químicos (teores de amido, de amilose e de açúcares solúveis), nos aspectos bioquímicos (atividade de enzimas relacionadas à degradação do amido, perfil de enzimas ligadas ao grânulo de amido) e aspecto morfológico do grânulo de amido (preliminar), e o amadurecimento do fruto. A manga \'Keitt\' teve um padrão atípico de fruto climatérico, com a produção de pequenas quantidades de etileno e C02, culminando em picos após o processo de amadurecimento ter sido iniciado. Todo o amido acumulado (cerca de 8 %) durante o desenvolvimento até os 3 dias após a colheita (dpc), foi totalmente degradado a partir dos 5 dpc chegando ao final do amadurecimento com apenas traços do seu conteúdo inicial. Ao mesmo tempo acumulou cerca de 10 % de açúcares solúveis, com predominância da sacarose. As enzimas que potencialmente podem degradar o amido, tiveram perfil de atividade compatível com a sua atuação. Houve um aumento bastante significativo de atividade da α-amilase durante a formação do fruto e da β--amilase durante o amadurecimento do fruto. As fosforilases e isoamilases, embora tivessem atividade suficiente para atuar durante a degradação do amido, demonstraram pelo perfil de atividade terem bastante importância durante a síntese do amido. Os grânulos de amido, como observados por microscopia eletrônica de varredura, tem grânulos redondos, lisos e pequenos (até 20 µm), que diminuem de tamanho durante o amadurecimento da manga. As proteínas ligadas ao grânulo, como visto por eletroforese em condições dissociantes, aumentaram em número e quantidade depois da colheita da manga, mostrando que várias proteínas de alto e baixo peso molecular, aderiram ao grânulo antes do início da degradação. / Sweetening is one of the most important characteristics concerning ripe fruit. However, physiological and biochemical changes associated to mango fruit ripening process, including mango sweetening, is still poor understood. In this work the synthesis and breakdown of the starch were studied focusing starch, amylose, and soluble sugars content. Also the activities of some enzymes that participate in starch metabolism were evaluated during development and ripening of mango \'Keitt\'. Thought Scanning Electron Microscopy, granule starch shape and superficial changes related to the degree of mango ripening, were investigated. Results shown that mango fruit (cv. Keitt) has not a typical profile of ethylene and respiration during the ripening process, with very low leveis of both parameters. There was a massive conversion of the starch accumulated (~8 % )during fruit development to soluble sugar (~10 %), with a predominance of sucrose. The activity of α-amylase increased at least 20 times during fruit development while &#946:-amylase showed detectable activity after fruit harvesting. Starchphosphorylases and isoamylases activities can be linked with both starch synthesis and degradation. Granule starch isolated from mango, showed spherical shape and small size (~20 µm) when the mango achieve full starch content (~3 days after harvest), with about 25 % of amylose. During fruit ripening, granule size suffered superficial corrosion and decreased in size (~6 µm). SDS-PAGE gels showed that after harvesting increased the number of starch bounded proteins with low and high molecular weight, inclusive inside the granule. These proteins can be associated with starch degradation process.

Aditivos químicos para o tratamento da cana-de-açúcar in natura e ensilada (Saccharum officinarum L.) / Chemical additives for the treatment of green chopped and ensiled sugarcane

O objetivo do presente trabalho foi avaliar o efeito de aditivos químicos sobre as perdas de matéria seca, composição química, valor nutritivo e estabilidade aeróbia da cana-de-açúcar in natura ou ensilada. No primeiro experimento, a cana-de-açúcar foi ensilada em silos laboratoriais. O experimento foi desenvolvido em delineamento experimental inteiramente casualizado com quatro repetições por tratamento. Os tratamentos avaliados foram: L. buchneri, cal virgem e calcário em doses de 1,0 e 1,5% da MV e gesso a 1,0% da MV, diluídos em 40L de água por tonelada de massa verde. Após a abertura dos silos, foram realizadas avaliações de perdas, estabilidade aeróbia, valor nutritivo e composição química. Os tratamentos contendo fontes de CaO ou CaCO3 apresentaram maior recuperação de matéria seca e menores perdas totais e gasosas. Nesses tratamentos também foram observados menor teor de etanol e maior concentração de ácido lático e de carboidratos solúveis. Para as variáveis de valor nutritivo, as silagens tratadas com esses aditivos apresentaram alto teor de cinzas, menor teor de fibra, maior coeficiente de digestibilidade e pequena alteração da fração protéica. As silagens de cana-de-açúcar com gesso ou L. buchneri apresentaram desempenho semelhante a silagem controle para as variáveis acima mencionadas. No ensaio de estabilidade aeróbia, a silagem aditivada com 1,5% de cal virgem apresentou desempenho superior e semelhante as silagens com L. buchneri e gesso. De forma geral, durante a fase anaeróbia as silagens tratadas com fontes de CaO ou CaCO3 apresentaram desempenho superior. Entretanto, na fase aeróbia, somente o tratamento contendo 1,5% de cal virgem manteve desempenho satisfatório. O segundo experimento avaliou o tratamento em montes da cana-de-açúcar picada in natura com doses de cal virgem durante dez dias de exposição aeróbia. O experimento foi realizado em delineamento inteiramente casualizado com quatro repetições por tratamento e em arranjo fatorial 2x4 com medidas repetidas no tempo. O fatorial foi composto por dois modos de aplicação e quatro doses do aditivo (0, 0,5, 1,0 e 1,5% da MV). Durante o ensaio foram realizadas avaliações de estabilidade aeróbia, valor nutritivo e composição química. Para as variáveis de estabilidade aeróbia, a utilização de cal virgem foi efetiva em evitar o aquecimento da massa de forragem e reduzir as perdas de matéria seca. Para a avaliação químico-bromatológica, não foi constatada diferença nos teores de CHO´S entre os tratamentos. O tratamento da cana-de-açúcar com cal virgem elevou o pH e a fração mineral da forragem. Os maiores teores de proteína bruta foram observados para a forragem não aditivada, contudo os valores obtidos para os demais tratamentos estão dentro da amplitude relatada na literatura. Doses de 1,0% e 1,5% do aditivo foram efetivas em evitar a elevação nos teores de FDN e FDA da canade- açúcar. A fração hemicelulose sofreu solubilização parcial imediatamente após a aplicação do aditivo. Os maiores coeficientes de DVIVMS e DVIVMO foram observados para as doses altas do aditivo, no modo de aplicação a seco, principalmente nas primeiras horas após o tratamento. / The objective of this trial was to evaluate the effects of chemical additives over the dry matter losses, chemical composition, nutritive value and aerobic stability of green chopped and ensiled sugarcane. In the first trial, the sugar cane was ensiled in experimental silos. The trial was carried out in a completely randomized experimental design with four replicates per treatment. The treatments evaluated were: L. buchneri, CaO and CaCO3, 1.0 e 1.5% and Ca(SO4)2, 1.0% (wet basis), all of them diluted to 40L of water per ton of fresh forage. After the silos were opened, the following variables were analyzed: losses, aerobic stability, nutritive value and chemical composition. Higher recovery rates and lower dry matter losses and gaseous production were observed for the silages containing CaO or CaCO3. In these treatments were also observed lower ethanol production and higher lactic acid and water soluble carbohydrates contents. The analysis of nutritive value showed that the utilization of CaO and CaCO3 increased the ash content, diminished the fibrous portion, improved the organic and dry matter digestibility and resulted in small changes of the crude protein content. The utilization of L. buchneri and Ca(SO4)2 resulted in silages similar to the control treatment for these variables. In the aerobic stability assay, only the silage containing 1.5% of CaO maintained the best performance. The silages treated with L. buchneri or Ca(SO4)2 showed similar performance. In summary, the silages treated with CaO or CaCO3 showed the best performance in the anaerobic phase and in the aerobic assay, only the treatment containing 1.5% of CaO maintained the positive performance across both assay. In the second trial, the green chopped sugar cane was treated with doses of CaO in an aerobic assay during ten days. The trial was carried out as a completely randomized experimental design with four replicates in a factorial design with repeated measures. The factorial was composed by two application methods and by increasing doses of CaO (0, 0.5, 1.0 e 1.5% in FF). The following variables were analyzed during the assay: aerobic stability, nutritive value and chemical composition. During the aerobic assay, the treatment with CaO reduced the heating and the dry matter losses in fresh sugar cane. For the chemical composition and nutritive value, there were no differences between treatments for the water soluble carbohydrates concentration. The utilization of CaO increased the pH and ash content of forages. Higher levels of crude protein were observed for the control forage, however the utilization of CaO did not reduced significantly the crude protein value. Doses of 1.0 and 1.5% of CaO inhibited the NDF and ADF uprising contents in the fresh sugar cane. The hemicellulose portion was solubilized immediately after the addition of CaO. Higher IVDMD and IVOMD coefficients were observed for the forage treated with 1.0% of CaO applied dried, mainly during the period immediately after treatment.

Avaliação de fontes de carboidrato e proteína e sua utilização na dieta pré-inicial de frangos de corte. / Carbohydrate and protein sources evaluation and its utilization in the diets of newly hatched chicks.

Existe uma série de evidências que os pintos de corte na fase pré-inicial têm seu sistema digestório ainda imaturo, implicando em menor capacidade de aproveitamento dos alimentos. Em função desse fato, ingredientes de maior digestibilidade seriam recomendados para proporcionar um desenvolvimento adequado do organismo. Com isso, o objetivo do presente trabalho foi determinar a energia metabolizável aparente corrigida pelo balanço de nitrogênio (EMAn) de ingredientes como fontes de carboidrato e proteína para frangos de corte na fase pré- inicial, bem como avaliar a utilização destes ingredientes sobre o desenvolvimento inicial de órgãos do trato gastrintestinal, desempenho e características da carcaça das aves. Os ingredientes avaliados foram: milho (MI), amido de milho (AMI), amido de mandioca (AMA), glicose (GLI), lactose (LAC) e sacarose (SAC), como fontes de carboidrato; e farelo de soja (FS), soja integral micronizada (SIM), isolado protéico de soja (IS), ovo em pó (OP), plasma sanguíneo (PS), farelo de glúten de milho (GM) e levedura seca (LS), como fontes de proteína. Para a determinação da EMAn foram desenvolvidos vários ensaios de metabolismo utilizando-se o método de coleta total de excretas. Para a avaliação do desempenho, desenvolvimento do trato gastrintestinal e características da carcaça foram conduzidos diversos experimentos, em gaiolas e em piso de galpão experimental, com a inclusão dos ingredientes nas dietas. Os valores de EMAn para a fase pré- inicial das fontes de carboidrato foram: 3.213; 3.269; 3.690; 3.427; 1.225 e 3.524 kcal/kg para MI, AMI, AMA, GLI, LAC e SAC, respectivamente. Para as fontes protéicas a EMAn foi: 2.085; 4.068; 2.110; 5.095; 3.831; 3.374 e 2.037 kcal/kg para FS, SIM, IS, OP, PS, GM e LS, respectivamente. Para as aves criadas em gaiolas pode-se dizer que o desenvolvimento do trato gastrintestinal, de maneira geral, não foi influenciado pelas diferentes fontes de carboidrato, mas o consumo de ração das aves foi aumentado pelo fornecimento de carboidrato de maior disponibilidade na dieta préinicial, contribuindo para um aumento no peso vivo das aves. As diferentes fontes de proteína afetaram o desenvolvimento dos órgãos, mas esse efeito não foi suficiente para explicar as variações no desempenho das aves. A adição de diferentes fontes de proteína e carboidrato, bem como suas misturas, em dietas pré- iniciais de frangos de corte criados sob piso de galpão experimental, promoveram alterações no desempenho e no desenvolvimento do intestino delgado das aves até o sétimo dia de idade, entretanto as diferenças de desempenho não foram mantidas nas fases subseqüentes, contribuindo para que as características da carcaça e composição da carne de perna não apresentassem diferenças no momento do abate. / There are some evidences in the literature that newly hatched chicks have the digestive system still immature, what result in a small capacity to metabolize feed ingredients. So, more digestible ingredients could be recommended to allow an adequate development of the body. Herewith, the objective of this work was to determine the Ncorrected apparent metabolizable energy (EMAn) of carbohydrate and protein sources for newly hatched chicks, and to evaluate the utilization of these ingredients on the early development of gastrointestinal tract organs, performance and carcass characteristics of birds. The ingredients evaluated were: corn (MI), corn starch (AMI), cassava starch (AMA), glucose (GLI), lactose (LAC) and sucrose (SAC), as carbohydrates sources, and soybean meal (FS), micronized full fat soybean (SIM), isolated soy protein (IS), dried whole eggs (OP), blood plasma (PS), corn gluten meal (GM) and dried sugar cane yeast (LS), as protein sources. Several metabolism trials were conducted to determine the EMAn, using the total excreta collection method. To evaluate the performance, gastrointestinal tract development and carcass characteristics, experiments were conducted, in cages and floor pens, using diets containing those ingredients. The EMAn of the carbohydrate sources determined for chicks in the first week were: 3,213; 3,269; 3,690; 3,427; 1,225 and 3,524 kcal/kg for MI, AMI, AMA, GLI, LAC and SAC, respectively. The EMAn for protein sources were: 2,085; 4,068; 2,110; 5,095; 3,831; 3,374 and 2,037 kcal/kg for FS, SIM, IS, OP, PS, GM and LS, respectively. For the birds reared in cages, the results showed, in general, that the different carbohydrate sources did not affect the gastrointestinal tract development, but the feed consumption of the birds was improved by the more available carbohydrate sources in the newly hatched chicks diets, contributing to a higher liveweight of the birds. Otherwise, the different protein sources affected the gastrointestinal tract development, but these effects did not explain the changes on broiler performance. The addition of different protein and carbohydrate sources, and their mixtures, in the diets of newly hatched chicks reared in the floor pens resulted in differences on performance and on small intestine development of the birds until seven days of age, but these differences were not maintained on the following phases, contributing to a similar carcass characteristics and similar chemical composition of the whole leg meat at slaughter time.

Influence de la disponibilité en glucides sur la performance : nouvelles stratégies d’apports glucidiques en fonction des besoins de la programmation d’entraînement / Impact of muscle glycogen availability on performance : new carbohydrate intake strategies according to the needs of training program

Ces travaux ont visé à étudier l’impact de la disponibilité glucidique sur la performance et les adaptations de l’entraînement. L’étude n°1 a permis de caractériser la stratégie de récupération optimale entre deux entraînements chez des athlètes élites en BMX. Les résultats ont permis d’identifier la stratégie nutritionnelle comme étant la stratégie de récupération induisant la plus faible diminution de puissance lors de tests de performance. La deuxième partie a investigué l’impact de la périodisation chronique de l’apport glucidique, par la stratégie « Sleep-Low », sur la performance en endurance et la fonction immunitaire. La stratégie « Sleep-Low» consiste à réaliser des entraînements ciblés en condition de faible disponibilité en glucides, notamment une période récupération faible en glucides et un entraînement à faible intensité réalisé à jeun. Les résultats révèlent que tous les sujets de la stratégie « Sleep-Low » améliorent leur performance en endurance lors d’un 10km en course à pied ou d’un contre-la-montre de 20km. Cette amélioration de la performance est associée à une modification de la stratégie d’allure dans le cas d’un contre-la-montre vers une augmentation des niveaux de puissance développés. Les résultats ont suggéré une modification de la perception d’effort suite à l’implémentation de la stratégie. Ces résultats sont associés à une diminution de la masse grasse chez les participants. L’étude n°3 confirme que la stratégie « Sleep-Low » n’entraîne pas de dégradation de la fonction immunitaire ou de l’efficacité de sommeil. Ces travaux vont dans le sens d’une manipulation de la disponibilité en glucides en fonction de l’objectif des périodes d’entraînement / The aim of the present work is to study the impact of the availability of carbohydrates on performance and on the development of training adaptations. The first section of this thesis aimed at defining the optimal recovery strategies between training sessions for elite BMX pilots. The results revealed that the nutritional strategy is one of the most efficient recovery strategies leading to a lesser decrease in power output. The second section of this thesis investigated the impact of a chronic periodization of carbohydrate intake, through the “Sleep-Low” strategy, on endurance performance and immune function. The “Sleep-Low” strategy consists in specific training sessions performed under low carbohydrate availability, notably during recovery period and low intensity prolonged sessions. All subjects training under the “Sleep-Low” strategy improved their performance on a 10km running trial and on a 20km cycling time trial. This improved performance is associated with a modification of pacing strategy during the 20km cycling time trial toward higher power output. The results suggested a modification of perception of effort after training under the “Sleep-Low” strategy: despite an improvement of performance, participants did not perceive the effort as more difficult. These results are associated to a decrease in fat mass. The study #3 confirmed that the periodization of carbohydrate intake did not impair immune function or sleep efficacy. All the results of the four studies bring new elements in the evolution of carbohydrate recommendations. These works are in the line of a manipulation of carbohydrate availability according to the work required

The anthropometric status and dietary intake of adults engaging in bodybuilding programme around Polowane Municipality in Limpopo Province

(Dietectics)) -- University of Limpopo, 2017 / Introduction: Bodybuilders around Polokwane municipality are adhering to different dietary practices some of which are not evidence based. These practices may predispose these athletes to negative health outcomes sooner or later in life. The aim of this study was to investigate the anthropometric status and dietary intake of bodybuilders around Polokwane Municipality. Methodology: This was a descriptive analytical study where purposive sampling method was used to obtain 51 out of 65 registered bodybuilders within the training gyms around Polokwane. Weight and Height were measured to determine BMI. Four skinfold sites (triceps, biceps, subscapular and suprailliac) were measured using caliper and non-stretchable tape to estimate total body fat percentage. Dietary information was collected using the 24 hour-recall collected for three different days which was validated by the Food Frequency Questionnaire. Results: of the 51 body builders, 94.1% were men and 5.8% women. Few athletes (15.7%) were using ergogenic agents such as powders (13.7%) and tablets (2%). Also, 64.7% of the athletes had normal body weight while 33.3% and 2.0% were overweight and obese respectively. Majority of the athletes (78.4%) had an acceptable body fat percentage. Dietary results showed that majority of the athletes consumed energy (98%) and carbohydrates (100%) below the ISSN bodybuilding recommendations. Conclusion: The dietary intake of the bodybuilding athletes around Polokwane municipality is below the ISSN (2010) bodybuilding recommendations; however, their anthropometric status and body composition is normal.

Analýza vybraných odrůd českého česneku / Analysis of czech garlic varieties

This master thesis deals with analysis of Czech garlic varieties in comparison with two varieties from abroad. Alliin and carbohydrates were determined by high performance liquid chromatography with diode-array detector or evaporative light scattering detector. Crude proteins were calculated from total content of nitrogen that was determinated by Kjeldahl method. Elementary analysis was performed by inductively coupled plasma optical emission spectrometry. In theoretical part of this thesis is describes botanical characterization and cultivation of garlic, chemical composition of garlic, especially groups of substances which were analysed in this thesis. Principle of used methods is described too. The experimental part of this work deals with sample and calibration solution preparation and process of analysis. Obtained results from analysed substances are summarized in the tables, discussed and compared with results of analysis of other authors. Between Czech and Spanish/Chinese garlic are slight differences. Garlics contain 24,66 g/kg dry weight of alliin. Content of crude proteins in Czech garlic is comparable to Chinese garlic but the Spanish garlic has just half of amount crude proteins. Elementary analysis also includes significant differences. Czech varieties contain higher amount of phosphorus, iron and zinc. On the contrary, content of potassium in foreign garlic is higher than in Czech Republic and the analysed amount is 15 g/kg dry weight.

Cieľom tejto diplomovej práce je stanoviť kvalitu a zloženie medu, taktiež stanoviť jeho botanický pôvod na základe profilu sacharidov v mede. Med je prírodný produkt vysokej kvality a vďaka značnej spotrebe a nemalej trhovej hodnote je tiež terčom na falšovanie. S cieľom identifikovať podvody a cudzorodé látky, pochádzajúce z cukrových sirupov, boli analyzované oligosacharidy a polysacharidy na báze maltodextrínov. Teoretická časť popisuje chemické zloženie medu a hlavné princípy aplikovanej inštrumentálnej techniky. Poskytuje literárny prehľad existujúcich analytických metód na stanovenie sacharidov prítomných v mede a na odhalenie jeho falšovania. Boli definované hlavné body falšovania a autenticity, vrátane legislatívnych aspektov a opisu bežných druhov falšovania. Experimentálna časť obsahuje postupy, ktoré vedú k vývoju a optimalizácii chromatografických podmienok a parametrov elektrochemického detektora na stanovenie sacharidov a maltodextrínov (oligosacharidov a polysacharidov) vo vzorkách medu. Použitá analytická technika bola vysoko účinná aniónovo-výmenná chromatografia (HPAEC) spojená s pulzným amperometrickým detektorom (PAD). Vyvinutá a optimalizovaná chromatografická metóda bola taktiež validovaná z hľadiska linearity, dynamického rozsahu, analytických limitov, presnosti a správnosti. This diploma thesis examines influence of content of leaf nitrogen and non-structural carbohydrates on Rubisco activity and its content under impact of ambient (A = 385 µmolCO2 mol-1) and elevated (E = 700 µmolCO2 mol-1) concentrations of carbon dioxide in needles of Norway spruce (Picea abies L.). The Rubisco activity was determined spectrophotometrically, the Rubisco content by using SDS-PAGE, the content of non-structural carbohydrates by using the anthron method and total leaf nitrogen was determined by using the Kjeldahl method. The Rubisco activity in needles of Norway spruce did not show acclimation on elevated concentration of carbon dioxide, but the activity was elevated concentration of carbon dioxide stimulated during the growing season. The acclimation was proved by Rubisco content, its content was significantly lower in needles cultivated under elevated carbon dioxide. Content of total leaf nitrogen was statistically significantly lower in needles of Norway spruce cultivated under elevated carbon dioxide then in needles cultivated under ambient concentration of carbon dioxide. On the contrary, the content of non-structural carbohydrates was significantly higher in needles cultivated under elevated concentration of carbon dioxide then in variant A. Seasonal changes of Rubisco content depended on content of non-structural carbohydrates, the Rubisco content decreased with increasing content of non-structural carbohydrates. No dependence was detected between seasonal changes of the total leaf content and the Rubisco content.

Posouzení vlivu pasterizace a filtrace na obsah vybraných chemických složek piva / Evaluation of the influence of the pasterization and filtration on selected chemical components of beer

The aim of this work was to study the influence of the pasteurization and filtration on selected chemical compounds in beer. Specifically, the influence on elemental composition, content of organic acids, carbohydrates and phenolic substances was studied. Optical emission spectrometry with inductively coupled plasma (ICP-OES) was used for elemental analysis of beer. Analysis of organic acids was evaluated by Ion chromatography. For analysis of carbohydrates was used High performance liquid chromatography with ELSD detector (HPLC-ELSD), phenolic compounds was evaluated by HPLC with diode array detector (HPLC-DAD). Data from analysis of beer was performed by usage of statistical methods (analysis of variance and principal component analysis). Parameters that showed the most significant differences between individual groups of beers were obtained. Correlations between the variables and the technological beer processing operations were found. 12 parameters of total 23 were affected. The effect of filtration and pasteurization was observed in content of the iron, silicon, potassium and magnesium, malate, monosaccharides glucose and fructose and phenolic compounds.
