Odhad geografické polohy stanic v Internetu / Location estimation of Internet nodes

This paper deals with methods of stations’ IP geolocation. It’s describes the methods of passive and active geolocation and it’s more focused on active searching methods, which usees measuring the latency in network. The factors causing delays in data transfer are discussed first, followed by discussion of the issue of measuring these delays. After that a brief description of PlanetLab experimental network, which nodes were used for delay measurment. Main topic is practical implementation of method Constraint-based Geolocation in Java programming language. Last but not least the measurement results of CBG algorithm are tested.

Rôle de la Transcortine (CBG) dans la variabilité des réponses de stress / Role of transcortin (CBG) in the variability of stress responses

Une grande diversité dans la réponse adaptative au stress est observée entre les individus favorisant une sensibilité variable face aux stresseurs et pouvant conduire à une vulnérabilité à développer divers troubles et pathologies. Cette diversité est sous tendue par les caractéristiques propres de chaque individu, déterminées par le patrimoine génétique en interaction avec les facteurs environnementaux. Des études génétiques menées au laboratoire ont permis de placer le gène de la Cbg comme un candidat important influençant les réponses de stress. L’équipe a alors développé un modèle de souris déficiente pour le gène Cbg (k.o. total). La CBG est une glycoprotéine plasmatique responsable de la biodisponibilité et du transport jusqu’à leur cible des glucocorticoïdes, produits finaux de l’axe corticotrope. A l’aide de ce modèle original, l’objectif de mon travail de thèse a été d’étudier les conséquences fonctionnelles de la déficience en CBG sur les réponses de stress. Nous avons ainsi analysé l’activité et la réactivité de l’axe corticotrope ainsi que les comportements émotionnels des mâles et des femelles k.o. Cbg dans des conditions de repos, de stress aigu et dans un contexte mimant l’effet d’un style de vie occidentale (modélisé par une alimentation enrichie en gras, associée à un stress chronique). Nous présentons ainsi un modèle murin unique d’hypo-réponse des glucocorticoïdes au stress associé à une réponse comportementale adaptative ralentie au niveau émotionnel et cognitif. L’ensemble de ces travaux contribue à placer la CBG et son gène comme acteur majeur de la variabilité individuelle des réponses de stress. / A great diversity in the adaptive response to stress is observed between individuals favoring a variable sensitivity to face stressors and leading to a vulnerability to develop various disorders and diseases. This diversity is due to the characteristics of each individual, as determined by the genetic background in interaction with environmental factors. Genetic studies conducted in the laboratory demonstrated that the Cbg gene is an important candidate influencing stress responses. The team then developed a mouse model deficient for the gene Cbg (total k.o.). CBG is a plasma glycoprotein responsible for the bioavailability and the transport of glucocorticoids, the final products of the HPA axis, to their target.Using this original model, the objective of my thesis was to study the functional consequences of CBG deficiency on responses to stress exposure. We have analyzed the activity and reactivity of the HPA axis and the emotional behaviors of males and females k.o. Cbg in resting conditions, acute stress and in a context that mimics the effect of a Western life style (modeled by a high fat diet, associated with chronic stress). We present an unique mouse model of glucocorticoid hyposignaling in response to stress associated with behavioral responses slowed down at the emotional and cognitive levels. Overall, this work contributes to place CBG and its gene as major actor of individual variability to stress.

Geolokace stanic v síti Internet / Geolocation of Internet hosts

This master thesis deals with methods of stations’ geolocation on the Internet, it means the estimation geographic location of unknown station, which is conected to this network. The introductory part describes the methods for determining position, which don‘t need any measeruments of the network. The next section is devoted to points, where delay is invoked, to the causes of latency in the Internet, to their types and some usual values. The following part features ways to measure delays and route, including several tools for this purpose. The penulminate section describes some chosen geolocation techniques based on RTT measurement. The final chapter is devoted to practical demonstration geolocation using the CBG method, when the delay measurement was carried out in an experimental PlanetLab network.

O estado nutricional influencia o crescimento gástrico, e enquanto a proliferação celular é estimulada pelo jejum em filhotes, ela é inibida em ratos adultos. A corticosterona também age no desenvolvimento, e seus efeitos são regulados pela corticosterone binding globulin (CBG) e receptores de glicocorticóides (GR). Para investigar se a atividade da corticosterona responde ao jejum e como possíveis mudanças poderiam controlar o ciclo celular epitelial gástrico, nós avaliamos diferentes parâmetros durante a progressão do jejum em ratos de 18 e 40 dias de vida pós-natal. A restrição alimentar induziu um aumento de corticosterona no plasma em ambas as idades, mas apenas em filhotes o binding da CBG se elevou após o jejum curto, permanecendo alto até o final do tratamento. O jejum aumentou a atividade transcricional do GR na mucosa gástrica e os níveis protéicos, porém o efeito foi mais pronunciado em adultos. Além disso, observamos que nos filhotes, o GR é principalmente citoplasmático, enquanto em animais adultos, o receptor é acumulado no núcleo durante o jejum. As proteínas HSP 70 e HSP 90 foram diferencialmente reguladas, e podem contribuir para a estabilidade do GR no citoplasma em filhotes, e para o trânsito de GR para o núcleo em animais adultos. Quanto ao ciclo celular epitelial, observamos que em filhotes, ciclina D1 e p21 aumentaram durante o jejum, enquanto em ratos adultos, a ciclina E diminuiu e a p27 aumentou muito. Assim, nós demonstramos que a atividade da corticosterona é diferencialmente regulada pelo jejum em filhotes e ratos adultos, e que as variaçõe observadas poderiam atenuar um possível efeito supressor durante o desenvolvimento pós-natal. Sugerimos que este mecanismo pode estimular a proliferação celular e possibilitar o crescimento da mucosa gástrica durante condições nutricionais adversas. / The nutritional status influences gastric growth, and interestingly, whereas cell proliferation is stimulated by fasting in suckling rats, it is inhibited in adult animals. Corticosterone takes part in the mechanisms that govern development, and its effects are regulated in particular by corticosterone binding globulin (CBG) and glucocorticoid receptor (GR). To investigate whether corticosterone activity responds to fasting and how possible changes might control gastric epithelial cell cycle, we evaluated different parameters during the progression of fasting in 18- and 40-d-old rats. Food restriction induced higher corticosterone plasma concentration at both ages, but only in pups did CBG binding increase after short and long-term treatments. Fasting also increased gastric GR at transcriptional and protein levels, but the effect was more pronounced in 40-d-old animals. Moreover, in pups, GR was observed in the cytoplasm, whereas in adults, it accumulated in the nucleus after the onset of fasting. HSP 70 and HSP 90 were differentially regulated, and might contribute to the stability of GR and to the high cytoplasmic levels in pups and elevated shuttling in adult rats. As for gastric epithelial cell cycle, whereas cyclin D1 and p21 increased during fasting in pups, in adults, cyclin E slowly decreased concomitant with higher p27. In summary, we demonstrated that corticosterone function is differentially regulated by fasting in 18- and 40-d-old rats, and such variation might attenuate any possible suppressive effects during postnatal development. We suggest that this mechanism could ultimately increase cell proliferation and allow regular gastric growth during adverse nutritional conditions.

Impact of CBG deficiency on emotional and cognitive processes / L’impact de la déficience en CBG sur les processus émotionnels et cognitifs

La grande diversité des réponses de stress observée entre individus a pour origine des facteurs génétiques en interaction avec des facteurs environnementaux. Certaines réponses peuvent être moins adaptées et accroitre la vulnérabilité de l’individu aux divers troubles et pathologies liées au stress. La CBG est une glycoprotéine plasmatique impliquée dans la biodisponibilité des glucocorticoïdes, un des principaux médiateurs de la réponse au stress. Des études génétiques ont montré que des polymorphismes du gène codant la CBG ont un impact significatif sur la réponse des glucocorticoïdes au stress. Pour comprendre les mécanismes de l’impact de la CBG sur l’action des glucocorticoïdes et les conséquences sur les réponses endocriniennes et comportementales de stress, notre équipe a développé un modèle de souris déficiente pour le gène Cbg. Ces souris présentent une réponse diminuée des glucocorticoïdes au stress, associée à un niveau élevé de comportement émotionnel de type dépressif. Cette thèse a pour but d’explorer plus en profondeur les altérations physiologiques et comportementales des souris Cbg ko. Nous avons montré que le niveau plus faible de glucocorticoïdes observé chez la souris Cbg ko provient d’une élimination plasmatique plus importante. Une étude chez la souris Cbg ko femelles a montré que les estrogènes se surimposent à la déficience en CBG pour induire des comportements de type dépressif. Nous avons également démontré que la déficience en CBG conduit a une atténuation de la sensibilité comportementale et endocrinienne au stress chronique. Enfin, nous avons observé une détérioration de la mémoire long terme de ces souris. Par ailleurs, nous montrons que dans des conditions de stress chronique associé à un régime alimentaire déséquilibré le métabolisme du glucose était altéré chez les animaux déficients en CBG. Ces résultats renforcent l’importance du rôle de la CBG influençant l’ensemble des mécanismes d’actions des glucocorticoïdes par la modulation de leurs niveaux et de leur disponibilité. / The great diversity in the response to stress observed among individuals originates from their genetic background in interaction with environmental factors. Some responses can be less adaptive and increase the vulnerability to develop stress-associated disorders. CBG is a plasma glycoprotein that regulates the bioavailability of glucocorticoids, one of the main mediators of the stress response. Genetic studies pointed out variations in the gene coding for CBG as a major factor influencing the glucocorticoid response to stress. To better understand the mechanisms involved and the consequences on endocrine and behavioral responses to stress, our team has developed a mouse model of CBG deficiency. These mice present blunted glucocorticoid response to stress associated with increased despair-like behaviors. This thesis aimed at further exploring the physiological and behavioral alterations presented by the Cbg ko mice. We showed that the lower glucocorticoid levels observed in Cbg ko mice stems from their higher clearance from plasma. A study performed on Cbg ko female mice revealed that estrogens outpass CBG deficiency in inducing despair-like behavior. Additionally, we evidenced that CBG deficiency leads to lower behavioral and endocrine sensitivity to chronic stress, and we observed impairment of hippocampal-dependent long-term memory in these mice. Finally, we found that chronic stress combined to high-fat diet leads to alteration in glucose metabolism in CBG deficient animals. These findings reinforce the important role of CBG influencing the broad range of actions of glucocorticoids by modulating their levels and availability.

Využití znalosti topologie páteřních sítí pro určování fyzické polohy stanic v síti Internet / Geolocation in Internet using network topologies

The thesis discusses about modern geolocation methods and it describes the basic principles of their work. The work is divided into 2 parts - the theoretical one and the practical one. The first part of the thesis is focused on the description of these methods and on the explanation of its basic concepts which are used for determining of the physical position of the station according to its IP address. The second more extensive part of the work focuses on the description of the realization of algorithm in the Octant method in the programming language of Java and its use in the experimental net of PlanetLab. One important thing is to create a set of reference points and targets, which are necessary for the testing of the whole algorithm of the Octant Method. The results of estimated accuracy of target location obtained by the Octant method and their comparison with the results obtained by active methods of CBG, SOI and with the passive method of GeoIP are listed at the end of this work.

Určení polohy stanic v síti Internet pomocí přenosového zpoždění / Geolocation of Internet nodes based on communication latency

This thesis covers the topic of determination of geographical location of a host in internet network while utilizing measurement of the end to end delay and implementation of Constraint-Based Geolocation. Gradually I go through issue of the delay in computer networks and ways how to measure it. Next chapter describes a few ways to geolocate host in internet network with emphasis on the CBG method. Another chapter is dedicated to describing a way to project spherical coordinates to the two dimensional space, which has been used in implementation of geolocation method. Chapter about implementation builds upon the facts given in previous chapters while functions of the program written in the JAVA programing language are being explained. Two similar geolocation methods were implemented. By comparing the results gained by implementation, new method of geolocation is proposed and devised. It combines properities of both previous methods. There are results of the implemented methods and their comparation to the one of the source documents used in creation of this thesis in summary section.

The role of biogas in a more sustainable energy system in Sweden

There are numerous problems in the world that need to be dealt with in order to achieve sustainable development. The energy system has significant negative impacts on many of these problems, and there is a need for a transition towards more sustainable energy. Sweden has already started this transition and is using large amounts of renewable energy. However, within the transport sector and the manufacturing sector in particular, large amounts of fossil fuels are still used. Biogas is one alternative that can help solve several sustainability problems and that could be part of a future more sustainable energy system. However, it is not certain what biogas is most suitable to be used for. The aim of this thesis is to investigate how biogas should be used in a future more sustainable energy system, by answering three research questions: 1) In what ways can biogas be used in a more sustainable energy system? 2) How can we assess whether biogas is suitable in a specific context? and 3) What determines whether it is easy or difficult for a user to start using biogas? These questions are explored in a Swedish context using four appended articles, which are based on two collaborative projects using a combination of workshops, literature reviews and interviews. Biogas can be used for heat, electricity or fuel in the manufacturing or transport sector. In Sweden, heat and electricity are mainly of interest for smaller production scales, while production on larger scales will likely be dominated by upgrading mostly to CBG but also to LBG. CBG can be used for less energy-intensive purposes, such as cars or buses, while the growing interest in LBG in Sweden may open up new market segments for biogas which are more energy-intensive, such as heavy trucks or shipping, or in geographical locations that are further away from the site of production. Several sustainability assessment methods exist that can be used to evaluate whether biogas is suitable in a specific context, such as multi-criteria assessments or scenario analyses. These methods can include a number of different aspects that are relevant to biogas use, such as GHG emissions, safety issues, and the vitality of the surrounding region. In order to introduce biogas, six main factors were identified that can make this easier or more difficult: technical maturity, tank volume, distance between the producer and the user, scale of energy use, policies and costs, and strategies of individual organizations. Overall, the rise in LBG production creates new opportunities for biogas use in both geographical and usage areas that did not previously use biogas. There is no simple answer to what biogas should be used for in the future – rather, this depends on the circumstances. It is also possible that the usage areas that are most suitable now for biogas might not be the most suitable areas in the future, depending on developments within, for example, the electricity system and hydrogen. However, CBG and LBG are likely to dominate biogas production in Sweden until then.

Tankstationsdrift genom egenanvändning av solel : En systemanalys av samlokalisering av solkraft och en tankstation för fordonsgas / Filling station operation through self-consumption of solar energy

This thesis evaluates the energy system effects off co-localizing solar photovoltaics (PV)  production with a filling station for compressed vehicle gas. A MATLAB model was constructed based on gas and electricity consumption data from three actual filling stations for vehicle gas in Sweden. The model was used to simulate how the introduction of a PV  and PVB (photovoltaic battery) system would affect the amount of electricity bought from  and sold to the grid as well as the economic implications connected to this. The results show that a self-consumption of 42 % can be reach already without available energy  storage for an installed power of PV panels matching the power of the gas compressor. A battery storage with capacity corresponding to one hour of electricity generation doubles the self-consumption. The increased self-sufficiency has the potential to decrease the strain on the electricity grid, depending on spatial and temporal factors. The simplistic economic model in this project shows that the return of investment is just over 15 years for a PV system, and less than 21 years for a small battery storage with capacity to store less than one hour of electricity production, however, this should be examined in greater detail if the system is to be implemented.  Since transport has an important societal function, the access to fuel is key from an energy  security point of view. Filling stations require access to electricity to operate, meaning fuel can’t be obtained in case of an interruption. Therefore, the possibility of self-sufficiency during the summer months (May through September) through the added PVB system was evaluated, resulting in enough electric energy to supply 31 % of the current fuel volume supplied yearly at normal conditions. This corresponds to 145 tons of vehicle gas, which could power a private car around 3 220 000 kilometers. The connection between the electricity and transport sectors could be furthered integrated by installing the proposed system which turns the filling station into a prosumer, ideally with some opportunity of self-sufficiency if needed. The modelled filling station handles gaseous fuel, however the conceptual design applies also to liquid fuels. To cover an even larger portion of the fleet, charging of electric vehicles at the filling station could also be examined.
