CSR möter svensk papperstidning : CSR i svensk dagsress 2008.

Syfte: Syftet med denna studie är att skapa ökad förståelse för medias roll i förmedlingen av begreppet CSR, corporate social responsibility. Med media menas i studien svensk papperstidning. Metod: Diskursanalys kring CSR i svensk tryckpress, inspirerad av Michael Foucault och Norman Fairclough, applicerad på nyhetsartiklar. I studien används ordet diskurs som en förklaring till att nyhetstexter påverkar den sociala verkligheten. CSR innebär företags sociala ansvar, kring frågor om bland annat miljö, som går utöver vad lagen kräver. Slutsatser: Analysresultatet visar att CSR i nästintill samtliga artiklar benämns synonymt med samhällsansvar, som alltså varit det mest dominerande området som behandlats i de artiklar som har analyserats. Det framkommer även att det finns skillnader i artiklarnas intertextualitet, beroende på vilken tidning de tillhör. När CSR har omskrivits negativt har det varit kopplat till kategorin affärsnytta.

Introduction Bacterial biofilms have been implicated in the pathogenesis of Chronic Rhinosinusitis (CRS). This thesis consists of a number of separate studies. The results of each study were designed to help provide an evolution of knowledge that could be applied to our subsequent investigations on the topic of bacterial biofilms and chronic rhinosinusitis. In vitro studies were utilized to document the capacity of CRS bacteria to form biofilms as well as to investigate the efficacy of various antimicrobials at high concentrations. Additionally, an in vivo sheep model was developed to examine different biofilm detection techniques. Finally, a study of CRS patients was conducted to investigate the incidence of biofilm related sinus disease. Methods Our in vitro studies used 96 well crystal violet microtiter plate assays to determine the biofilm growth characteristics of S.aureus isolated from patients with CRS. Established biofilms were then subjected various antimicrobial agents, and the degree of biofilm reduction calculated to examine their potential for sinus biofilm treatment. A sheep sinusitis model involved performing endoscopic sinus surgery, occlusion of frontal sinus ostia and the introduction of bacteria. Mucosal specimens were subsequently examined for the presence of bacterial biofilms using transmission electron microscopy (TEM), scanning electron microscopy (SEM) and confocal scanning laser microscopy (CSLM). CSLM was also used in a prospective study to document the presence bacterial biofilms on the mucosa of patients with CRS compared to controls. Results The findings of in vitro experiments revealed that not all isolates were capable of forming biofilms. Of the antibiotics tested, only Mupirocin was capable of reducing biofilm mass by 90% in all isolates. The animal model showed considerable variation in biofilm detection rates. The CSLM biofilm detection rate was 100% in obstructed sinuses with bacteria introduced, whereas TEM detected only 66%. Both these objective measures failed to identify biofilms in control groups. SEM found biofilms in all experimental groups including controls. CSLM analysis of CRS patients found Bacterial biofilms in 44% and no biofilms in controls. Conclusion The demonstration of biofilms in the sheep model for sinusitis and biofilms on the mucosal specimens of patients with CRS, and the ability of bacteria in CRS to form biofilms in vitro, further supports the hypothesis that biofilms play a role in the pathogenesis of CRS. CSLM is the modality of choice in documenting the presence of bacterial biofilms on sinus mucosal surfaces due to the inherent flaws of sampling error and subjectivity of TEM and SEM. Niterói, RJ / A região de Cabo Frio (RJ) é uma área de ressurgência que favorece a produtividade primária e promove elevado acúmulo de matéria orgânica (MO) nos sedimentos, a qual afeta as condições redox do ambiente de sedimentação e na interface água-sedimento, promovendo efeitos sobre a especiação inorgânica do enxofre (S). O presente estudo utiliza a geoquímica inorgânica das espécies reduzidas do S, operacionalmente definidas como acid-volatile sulfufide (AVS) e chromium-reducible sulfur (CRS), a composição isotópica (δ34S) do sulfato da água intersticial e do CRS, e a razão COT/CRS (carbono orgânico total/CRS) para avaliar a diagênese recente do S e a condição redox nos sedimentos. Adicionalmente foi estudada a morfologia da pirita e a distribuição de tamanho dos frambóides através de microscopia eletrônica de varredura (MEV) para inferir o estado de oxidação do ambiente de formação. Foram coletados quatro box-cores na plataforma continental do Sistema de Ressurgência de Cabo Frio (SRCF) para este estudo. Os conteúdos de COT variam entre 0,8 e 2,3%, as maiores concentrações foram observadas nas estações da porção média da plataforma. Baixos valores de enxofre total (ST) (< 0,2%) e CRS (< 0,05%) foram encontrados nas camadas superficiais, ocorrendo incremento com a profundidade, indicando o aumento da sulfato redução (SR). As estações com o maior conteúdo de ST foram as localizadas na porção média da plataforma, sendo este padrão de acumulação favorecido pela presença dos vórtices sob a plataforma. A razão COT/CRS indica que o fator limitante para a formação da pirita é o enxofre. Até 50% do S pode estar associado a MO, caracterizando o processo de sulfidização da MO. A composição isotópica do sulfato ao longo dos perfis é mantida constante entre +18,7 e +23‰ e sem fracionamento com respeito ao sulfato da água do mar (+ 21‰), indicando baixas taxas de SR. No entanto, a composição isotópica do CRS apresentou sinais altamente empobrecidos em 34S (-25,6 até -40,8 ‰), indicando reações de oxidação no ciclo do S, promovidas pela intensa bioturbaçao na área e/ou pelo regime hidrodinâmico regional. As diferentes características morfológicas, como as superfícies de oxidação nos microcristais, os processos de crescimento secundário e os poliframboides encontrados nas análises morfológicas da pirita, assim como também a variabilidade na distribuição dos diâmetros dos frambóides evidenciaram uma formação sob condição redox altamente dinâmica. Estes resultados confirmam a complexidade hidrodinâmica do SRCF, o qual afeta inúmeros processos sobre a plataforma, dentre os quais a diagênese no ciclo do enxofre / The Cabo Frio region (RJ) is an upwelling area, which favors the high primary productivity and promotes accumulation of organic matter (OM) in sediments, affecting the redox conditions of the sedimentation environment, the sediment-water interface and mainly the speciation of inorganic sulfur (S). Therefore, this study uses the inorganic geochemistry of reduced S species operationally defined as acid-volatile sulfufide (AVS) and chromium-reducible sulfur (CRS), the isotopic composition (δ34S) of pore water sulfate and CRS, and the COT / CRS ratio (total organic carbon / CRS), to evaluate the recent S diagenesis and redox conditions in sediments. Additionally, was study the pyrite morphology and size distribution of framboids by scanning electron microscopy (SEM) to infer the oxidation state of the environment. In this sense, were collected four box-cores on the continental shelf of the upwelling system of Cabo Frio (SRCF). The TOC contents range from 0.8 to 2.3%, the highest concentrations were observed at stations in the middle shelf. Low values of total sulfur (TS) (<0.2%) and CRS (<0.05%) were found in the superficial layers occurring an increase with depth, indicating an increase in sulfate reduction. The stations with the highest content of ST were located in the middle shelf, this pattern of accumulation is favored by the presence of cyclonic eddy under the platform. The COT/CRS ratio indicates that the limiting factor for the pyrite formation is the sulfur. The isotopic composition of sulfate did not vary much with the sediment depths and is kept constant between +18.7 and +23 ‰ and without fractionation of sulfate with respect to sea water (21 + ‰) indicating low net sulfate reductions, but the isotopic composition of the CRS is highly depleted (-25.6 to -40.8 ‰), indicating oxidation reactions in the cycle of S, promoted by the intense bioturbation in the area and the regional hydrodynamic regime. The different morphological characteristics observed, such as surface oxidation in the microcrystals, the processes of secondary growth and poliframboides found, as well as the variability in the framboids diameters distribution, show the formation under highly dynamic redox condition. These results confirm the complexity of SRCF due to the occurrence of different hydrodynamic conditions over the shelf

An Insight in the Mälarbanan Project : A Study of the Hydrological and Geotechnical Aspects of the Mälarbanan Project / En inblick i Mälarbananprojektet : En studie omde hydrologiska och geotekniska aspekternai Mälarbananprojektet

In “Mälarbananprojektet”, Trafikverket (The Swedish Transport Administration in Stockholm) want to expand the railway. Mälarbanan is at the present a railway that extends between Stockholm and Hovsta. The railroad will be expanded to four tracks from two tracks and the aim of the Mälarbanan project is to separate commuter trains from other trains. As there is limited space on the surface, space underneath the surface must be used. This is why it is important to determine how far below the surface the construction will have to take place, which most likely will require a groundwater drawdown. This thesis focuses on an area with two reference points in Solna. A lowering of the pressure levels normally affects the surrounding in the form of subsidences, also known assettlements​, in the ground. As the area has a clay layer believed to be around 7 m thick, and clay is sensitive to groundwater drawdowns, the drawdown must be minimal. The soil engineers have established that the maximal groundwater drawdowns should not exceed 3.0 m, as anything over 3.0 m may cause extreme settlements in the settlement sensitive clays over the next 100 years. The modelling of drawdown and pore pressure conditions is conducted in a software called ​Visual MODFLOW.​ These different groundwater drawdowns, or scenarios, computed in the settlement calculations are 0 m, 0.3 m, 1 m and 3 m. The calculations have been computed for 1, 2, 5, 20, 40 and 100 years from now. Besides settlement occurring from groundwater drawdown, settlements in the form of primary and secondary consolidation (creep) normally occur from constant loading. This requires a software for settlement calculations that takes creep effects into consideration and the software used in this case was ​GS Settlement​. The parameters necessary for the calculations are obtained by conducting CPT, collecting undisturbed soil samples for fall cone tests and CRS-tests in a laboratory. These methods are supposed to decide how and if the project can proceed. / Trafikverket i Stockholm planerar att bygga ut Mälarbanan i samarbete med WSP. I nuläget sträcker sig Mälarbanan från Stockholm till Hovsta. Planen är att bygga ut från två spår till fyra spår och därmed separera pendeltåg från andra tåg. Då det finns begränsat med utrymme på ytan, måste man utnyttja utrymme under ytan. Detta kräver mest troligt en grundvattensänkning. Detta arbete fokuserar på två punkter i Solna Business Park. Grundvattensänkning påverkar omgivningen genom att bilda sättningar, en sjunkande rörelse i markytan. Då lera är extremt känsligt mot grundvattensänkningar, och lermäktigheten under markytan är ca 7 m, bör man sänka grundvattenytan så lite som det går. I detta projekt vill man inte sänka grundvattenytan med mer än 3,0 meter, då sänkningar som överstiger 3,0 m kan leda till extrema sättningar i den sättningskänsliga leran de kommande 100 åren. Detta, i samband med portryckssituationen, modelleras i ​Visual MODFLOW​. Programmet producerar en situation för varje sänkning. Dessa sänkningar är: 0 m, 0,3 m, 1 m och 3 m. Detta implementeras i sättningsberäkningar i ​GS Settlement​, som beräknar sättningarna de kommande 100 åren. Detta program tar med krypsättningar i beräkningarna. Microscopie et spectroscopie de phase. Développements en diffusion Raman cohérente (CRS) et en thermo-plasmonique / Phase microscopy and spectroscopy for Coherent Raman Scattering (CRS) and Thermoplasmonics

La microscopie par diffusion Raman cohérente anti-Stokes (CARS) est une technique de spectro-imagerie qui permet de cartographier les modes vibrationnels intra-moléculaires d'un échantillon biologique, sans nécessité de marquage préalable. La mesure CARS est cependant dégradée par un "fond non-résonant" qui détériore le contraste. Récemment, la microscopie par diffusion Raman stimulée (SRS) fut proposée comme une alternative à la microscopie CARS, permettant d'obtenir une imagerie "sur fond noir". Dans cette thèse, nous décrivons le développement d'un microscope SRS. Nous évaluons le caractère spécifique des contrastes CARS et SRS dans le cadre d'une application biomédicale concrète, à savoir la détection de mélanomes humains. Nous présentons une description exhaustive des phénomènes physiques pouvant conduire à des artéfacts de mesure en SRS. Nous proposons finalement une technique basée sur l'utilisation de trois faisceaux d'excitation à trois couleurs, permettant de supprimer ces artéfacts. Dans une seconde partie, nous nous intéressons à la microscopie CARS en configuration plein champ. Nous proposons une méthode permettant de supprimer le fond non-résonant. Celle-ci est basée sur une analyse de front d'onde du champ anti-Stokes. En guise d'ouverture, nous proposons une technique - toujours basée sur l'analyse de front d'onde - permettant de réaliser la spectroscopie d'absorption quantitative de nano-objets. Nous illustrons le potentiel de cette technique en réalisant des mesures sur des matrices de nanoparticules d'or et sur des nanoparticules uniques. / Coherent Anti-Stokes Raman Scattering (CARS) microscopy is a technique that can map the spatial distribution of intra-molecular vibrational modes of a biological sample. This method thus provides molecular specificity, without staining the sample. However, CARS signal is hampered by a "non-resonant background" which reduces the contrast. Recently, Stimulated Raman Scattering (SRS) microscopy has been proposed as an alternative to CARS microscopy because it is a background free method. In this thesis, we describe the development of a SRS microscope. We evaluate the specificity of CARS and SRS contrasts in a concrete biomedical application, the detection of human melanomas. We present a comprehensive description of the physical phenomena that can lead to artifacts in SRS microscopy. We show that the scattering properties of the sample can lead to artifacts. We propose a technique based on three excitation beams of different color which suppresses these artifacts. In the second part, we focus on CARS microscopy in a wide-field configuration. We implement a technique to remove the non-resonant background. This method is based on wavefront sensing of the anti-Stokes Field by quadriwave lateral sharing interferometry (QWLSI). We demonstrate that the measurement of the intensity and phase of the complex CARS field allows to retrieve the spontaneous Raman scattering spectrum. As a perspective, we propose a technique, still based on wavefront sensing by QWLSI, to achieve quantitative absorption spectroscopy of nano-objects. We illustrate the potential of this technique by performing measurements on arrays of gold nanoparticles and on single nanoparticles.

Jämförelse mellan CPT-sondering och CRS-försök med avseende på deformationsegenskaper / Comparison Between CPT-probing and CRS-Testing with Respect to Deformation Properties

Deformationsmoduler förklarar hur ett material eller ett område beter sig och deformeras vid olika laster. CRS-försök, komprimerar prover under en konstant deformationshastighet med erhållna moduler såsom: M0 och ML, med hjälp av rutinundersökning. Vid CPT-sondering penetrerar en konisk spets kontinuerligt genom en jordlagerföljd, med erhållen modul: Elasticitets-modul. Metoderna är väldigt olika vad gäller erhållna parametrar, moduler, deformationsgrupper och användning. CRS-försök genomförs laborativt, med prover tagna på vissa specifika djup, medan CPT-sondering genomförs in-situ med information om hela jordlagerföljden som ges. Att jämföra de olika metodernas moduler mellan varandra är inte möjlig, eftersom CONRAD som utvärderar och presenterar CPT resultat, inte utvärderar elasticitets-moduler för lera, utan för friktionsjord. Dem kompletterar varandra väldigt bra, då i områden som i Uppsala som har en med stor mäktighet av lera, visar med CRS-försök att det är lera som är närvarande för att sedan när CPT- sondering börjar registrera Elasticitets-moduler visar på att friktionsjord har påträffats. På så sätt kan deformationsegenskaper för en hel jordlagerföljd erhållas för en borrpunkt. Borrpunkter från Kronåsen och Kungsängen i Uppsala kommun, och Fyrklövern i Upplands Väsby kommun har undersökts med avseende på deformationsegenskaper och resultatet visar på stora skillnader i deras deformationsmoduler sinsemellan. / Deformation modules explains how a material or an area behave or deform when different amount of stresses act on it. CRS-testing, which compresses samples with a constant deformation rate, gives us modules like M0 and ML, with the help of routine examinations. CPT-probing, on the other hand, penetrate a conical tip through stratigraphy and gives us the elasticity module. These methods are very different in many ways, like what parameters, modules, deformation groups and use, are given. CRS-testing is conducted in a laboratory, with samples taken from specific depths, while CPT-probing is conducted in the field, with information about the whole stratigraphy is given. To directly compare these two methods modules are not possible. Due to CONRAD that evaluates and present CPT-probing results, does not evaluate elasticity modules for clay, but only for frictional soils. They complement each other very well due to in areas like Uppsala, which has a great depth of clay, shows with CRS-testing that clay is present but when CPT starts to register elasticity modules, shows that we moved in to frictional soil. With this, deformation properties can be shown for the whole stratigraphy in a drill point. Drill points from Kronåsen and Kungsängen in Uppsala municipality, and in Fyrklövern from Upplands Väsby municipality has been evaluated with respect to deformation properties and the results shows great differences in the deformation modules between themselves.

It was designed and constructed four apparatus at the geotechnical laboratory of the Federal University of Viçosa. The cells have an interchangeable high air entry value ceramic disc on its base. It was also designed a software for data acquisition and an apparatus for measurement of the volume of water dislocated from the sample. It was used two residual soils from Minas Gerais. Samples prepared by compaction were used in order to assure homogeneity and to avoid deviations in test results and interpretation. The tests were carried out with different levels of applied suction. Part of the samples was prepared with the addition of different amounts of Portland cement to simulate soil matrix. After cure, these samples added with cement were saturated and subjected to CRS compression tests, using the consolidation test apparatus developed. It was noticed a structure failure at low-tension levels because of the sample elaboration method. The equipment was testified as functional. / Este trabalho apresenta o desenvolvimento de equipamentos para ensaios CRS com sucção controlada e ensaios em solos não saturados cimentados artificialmente. Foram projetadas e construídas quatro células no Laboratório de Geotecnia da Universidade Federal de Viçosa. As células têm em sua base uma pedra porosa, de alto valor de entrada de ar, intercambiável. Também foram desenvolvidos um aparato de medição de volume de água deslocado do corpo-de-prova e um programa para aquisição de dados. Utilizaram-se dois solos residuais jovens de Minas Gerais. Foram usadas amostras remoldadas por compactação com o intuito de assegurar a homogeneidade e evitar desvios nos resultados dos testes e na interpretação dos dados. Os ensaios foram conduzidos com diferentes níveis de sucção. Parte das amostras foi preparada com a adição de teores de cimento Portland pra gerar uma estruturação no solo. Após o período de cura, as amostras com adição de cimento foram saturadas e submetidas ao ensaio CRS, usando os aparatos desenvolvidos. Observou-se uma quebra da estrutura com baixos níveis de tensão devido ao método adotado na elaboração das amostras. Constatou-se um bom funcionamento dos equipamentos desenvolvidos.

Akademiska sjukhuset : Befintliga geotekniska och hydrogeologiska förhållanden och dess markförutsättningar för framtida byggnationer / Uppsala University Hospital : Current geotechnical and hydrogeological conditions and their potential for future construction

Akademiska sjukhuset är en stor, samhällsviktig verksamhet som har under de senaste tio åren genomgått flertalet ny- och ombyggnationer. Sjukhusområdet är beläget på Uppsalaåsen och befinns inom en primär skyddszon, vilket innebär en komplex geologi och krav på aktsamhet av grundvattnet som nyttjas för Uppsalas vattenförsörjning. Marken har visats innehålla stora lerkörtlar och drastiska skiftningar av jordarter, vilket har lett till en sättningsproblematik vid sjukhusområdet. Region Uppsala planerar flertalet ny- och ombyggnationer vid Akademiska sjukhuset och på grund av den komplicerade markbilden önskas en utredning av geotekniken och hydrogeologin vid området. Fokuset i detta examensarbete har legat på sättningsbenägenhet och grundvattnets trycknivåer. Fyra delområden av sjukhusområdet har pekats ut av Region Uppsala för eventuella framtida byggprojekt. Dessa områden är vid parkeringshus T3, produktionsköket, NOP-komplexet samt ny cyklotronbyggnad. Genomförandet av detta examensarbete har delats in i tre delar; kartläggning av jordlagerföljd, sättningsberäkningar utifrån CRS-försök, samt utredning av grundvattnets trycknivå. Jordlagerföljd har tolkats i GeoSuite med hjälp av 474 sonderingar genomförda av konsultföretaget Bjerking. Sonderingarnas samtliga jordskikt interpolerades och illustrerades som nivåkurvor med hjälp av Topocad. För att utreda jordlagerföljden vid de utvalda områdena skapades åtta tvärsnitt utifrån en 3D-modell konstruerad i Civil 3D. De jordlager som studerats är fyllnadsmaterial, lera, friktionsjord, berg samt markyta. Sättningskapacitet och dess tidsförlopp har beräknats utifrån två CRS-försök och max-, min- och medelvärden av grundvattnets trycknivå under sjukhusområdet har beräknats utifrån 20 års dagliga data från en av Uppsala Vattens mätbrunnar i Stadsträdgården.   Resultaten bekräftar åsens komplexa geologi av varierande lermäktigheter under sjukhusområdet. Det är sannolikt att flertalet byggnationer kräver djupgrundläggning, exempelvis vid parkeringshuset T3 och produktionskökets norra del. De erhållna resultaten visar även knappa lerlager vid andra delområden, vilket innebär mindre sättningsbenägenhet och kräver troligtvis endast ytgrundläggning. De sättningsberäkningar som utförts indikerar att parkeringshuset T3 kommer sjunka 20 till 70 cm efter 158 till 213 år utan grundläggning, något som liknar den pågående sättningen vid byggnaden. Grundvattnets trycknivå har under de senaste 20 åren haft ett medelvärde på + 2,3 m (RH2000) och kommer sannolikt inte fluktuera framåt på grund av den rådande vattendomen. Detta innebär att byggnationer inom vissa delar av sjukhusområdet, exempelvis vid produktionsköket, kommer behöva tillstånd inför byggnation inom den primära skyddszonen, vilket Akademiska sjukhuset är beläget i. Resultaten ger en god överblick över hydrogeologin vid Akademiska sjukhuset och indikerar hur de utpekade områdena bör projekteras med avseende på grundläggning för hållbart och ekonomiskt bygge. Det krävs dock mer underlag, bland annat fler sonderingar och CRS-försök, för att få en ökad komplexitet av sjukhusområdet i avseende på jordlagerföljd och sättningsbenägenhet. / Uppsala University Hospital is a large institution of vital societal function and many new buildings have been constructed and others rebuild during the past ten years. The hospital area is located on Uppsalaåsen and within a primary protection zone, i.e. there is a complex geology and requirements for the protection of the groundwater used for Uppsala’s water supply. The ground contains large sections of clay and has drastic changes in soil types. This has resulted in a subsidence problem within the hospital area. Region Uppsala has several plans regarding upcoming constructions at Uppsala University Hospital and due to the complicated soil situation is an investigation of the geotechnics and hydrogeology within the area sought for. The focus of this master thesis is on subsidence capacity and the ground water’s pressure levels. Four zones within the hospital area have been pointed out by Region Uppsala for potential future construction projects. The zones are at the parking garage T3, production kitchen, NOP-complex and new cyclotron building. The master thesis was divided into three parts; locating the soil layer sequence, calculation of subsidence based on CRS tests and study of the ground water’s pressure level. The soil layer sequence was construed in GeoSuite based on 474 probes conducted by the consultant company Bjerking. The probs respective soil layer was interpolated and illustrated as level curves using Topocad. Eight cross-sections were made within the zones of extra interest and the sections were made from a 3D-model created in Civil 3D. The soil layers examined throughout the master thesis were filling material, clay, granular soil, rock and ground surface. The subsidence capacity and its time were calculated based on two CRS trials and max-, min- and average values of the ground water’s pressure levels under the hospital area was computed based on 20 years of daily data retrieved from a measuring well in Stadsträdgården facilitated by Uppsala Vatten. The results confirm the ridge complex geology consisting of varying depths of clay under the hospital area. Several buildings will probably require deep foundations, e.g. the parking garage T3. The results also show areas of shallow layers of clay indicating lower risk of subsidence and hence will require a shallow foundation. The conducted subsidence calculations indicate that the parking garage T3 will sink 20 to 70 cm after 158 to 213 years if no foundation is implemented, which is in line with the current subsidence occurring at the building. The ground water’s pressure level has had an average value of + 2.3 m (RH2000) during the last 20 years and will probably not fluctuate in the future due to the existing decision on water management. Hence, some buildings within certain parts of the hospital area will require permission prior to construction within the primary zone.  The results establish a valuable overview of the hydrogeology at Uppsala University Hospital and give indications regarding how the foundations in the zones of extra interest should be dimensioned sustainable and economically. However, more data is needed, e.g. additional probes and CRS trials, to gain an increased complexity of the hospital area in respect to soil layer sequences and subsidence tendency.
