Influência do vírus da artrite encefalite caprina no imunograma sanguíneo e lácteo de cabras naturalmente infectadas / Influence of caprine arthritis encephalitis virus on blood and milk immunogram of naturally infected goats

A Artrite Encefalite Caprina (AEC) é uma lentivirose multisistêmica que tem a glândula mamária caprina como um dos alvos lesionais da doença. Pode causar uma manifestação mamária específica, chamada de mastite endurativa, além da mama representar uma importante via de eliminação do vírus, mesmo em animais que não apresentam a forma clínica. Considerando-se a possibilidade desta virose, cuja célula alvo predominante é o monócito, levar a alterações imunológicas que influenciem a susceptibilidade do animal a outras doenças, objetivou-se avaliar essa interação do vírus e da imunidade do hospedeiro por meio de fenotipagem, fagocitose e produção de Espécies reativas de oxigênio (ERO) e dos mecanismos de morte das células sanguíneas e lácteas de cabras naturalmente infectadas. Para isso 200 cabras foram triadas e, destas selecionadas oito fêmeas não sororreagentes e dez sororreagentes na pesquisa de anticorpos séricos para AEC, dentro dos padrões hematológicos da espécie e com dois exames bacteriológicos do leite negativos. O leite e o sangue colhido destes animais foram submetidos às seguintes provas: fenotipagem dos leucócitos CD4+, CD8+, CD14+, PG68A+ e CD45+, fagocitose de Staphylococcus aureus e de Escherichia coli por células CD14+ e PG68A+, produção de ERO e marcação com Anexina-V e Iodeto de Propídeo (PI) por granulócitos e células mononucleares. A infecção pelo VAEC pode ser associada a um aumento de células CD14+ no leite, mas não no sangue. Os outros perfis celulares não sofreram alterações. Quanto a função fagocítica, o vírus reduziu a porcentagem de fagocitose de Staphylococcus aureus por granulócitos PG68A+ do leite. Esta alteração não ocorreu na fagocitose de Escherichia coli e na produção de ERO dessas células, nem na fagocitose e produção de ERO pelas células CD14+ ressaltando que nesses processos a espécie bacteriana pode sofrer interações com o vírus ou mesmo com a resposta imune do organismo infectado. O VAEC não influenciou significativamente os mecanismos de morte ora investigados, mas a tendência dos resultados sugere que possa haver indução de morte por apoptose e/ou por necrose nos granulócitos do sangue e influenciar no processo de necrose destas células no leite, sem alterar esses processos nas células mononucleares. Ressalta-se a importância da interação monócito-neutrófilo na glândula mamária, principalmente nos animais sororreagentes para AEC, mesmo na ausência de mastite bacteriana. / The caprine arthritis encephalitis (CAE) is a multisystem lentivirose that have the goat mammary gland as one target of the lesional disease. May cause a mammary specific expression, called indurative mastitis, beyond these organ represent a major route of virus elimination, even in animals that do not exhibit this clinical manifestation. Considering the possibility of this virus, whose target cell are predominantly monocyte, lead to immunological changes that influence the animal\'s susceptibility to other diseases, this study aimed to evaluate the interaction of virus and host immunity by phenotyping, phagocytosis and production of reactive oxygen species (ROS) and the mechanisms of cell death, in blood and milk of goats naturally infected. For that, 200 goats were screened and was selected eight females non sero-reagents and ten seroreagents for serum antibodies against CAE, with the haematological analises within the species standards and two negative bacteriological tests of milk. Milk and blood from these animals underwent the following tests: phenotyping of leukocytes CD4+, CD8+, CD14+, PG68A+ and CD45+, phagocytosis of Staphylococcus aureus and Escherichia coli ROS production by CD14+ and PG68A+ cells, and labeling with Annexin-V and propidium iodide (PI) for granulocytes and mononuclear cells. CAEV infection may be associated with an increase in CD14+ cells in milk, but not in blood. The other cellular profile did not change. In the phagocytic function, the virus reduced the percentage of phagocytosis of Staphylococcus aureus by granulocytes PG68A+ milk. This change did not occur in the phagocytosis of Escherichia coli and production of ROS in these cells, nor in phagocytosis and ROS production by CD14+ cells, noting that in those cases the bacterial species may undergo interactions with the virus or even with the immune response of the infected organism. The VAEC not significantly affect the mechanism of death now investigated, but the tendency of the results suggests that can induce apoptosis and/or necrosis in the blood granulocytes and influence the process of necrosis of these cells in milk, without changing these processes mononuclear cells. We emphasize the importance of monocyte-neutrophil interaction in the mammary gland, especially in animals seropositive for CAE, even in the absence of bacterial mastitis.

Artrite-encefalite dos caprinos - aspectos clínicos e epidemiológicos / Caprine arthritis encephalitis - Clinical and epidemiological features

Estudou-se a freqüência da ocorrência de anticorpos antivírus da Artrite-encefalite dos Caprinos, em caprinos de 14 plantéis localizados no Estado de São Paulo, por um período de 2 anos, utilizando-se a técnica de imunodifusão em gel de ágar. A prevalência obtida foi de 26,3%, sendo significativamente maior nos caprinos mantidos em regime intensivo (31,8% - 733/2303) de criação do que no sistema semi-extensivo (13,1% - 128- 977). A infecção pelo vírus da Artrite-encefalite dos Caprinos aumentou gradativa e significativamente após os 6 meses de idade, havendo predominância da infecção nos caprinos mais velhos. Não se detectou influência de fatores sexuais sobre a prevalência da enfermidade determinada em caprinos do sexo feminino (27,9% - 663/2375) e masculino (32,3% - 94/291). A prevalência da doença foi significativamente maior nos caprinos das raças Anglo Nubiana (63,8% - 88/138) e Toggenbourg (56,0% - 28/50) do que nas demais raças estudadas: Saanen (27,4% - 673/2458), Alpina (11,9% - 59/497), Bôer (5,9% - 2/34) e caprinos mestiços (10,7% - 11/103). Paralelamente realizou-se estudo clínico dos animais infectados pelo vírus da Artrite-encefalite dos Caprinos, quando pudemos demonstrar que 17,1% (64/374) dos caprinos sororeagentes apresentavam a forma clínica articular da enfermidade e que 6,6% (17/249) das cabras sororeagentes apresentavam a forma mamária. A possibilidade de transmissão vertical transplacentária foi menor do que 3,8%. Verificou-se ser pequena a possibilidade dos cabritos se infectarem pelo colostro de cabras sororeagentes positivas, mas podem se infectar pelo leite oriundos dessas cabras infectadas (18,8% - 3/16). O tempo de duração dos anticorpos séricos adquiridos passivamente pelo colostro variou de 60 a 120 dias. Demonstrou-se, indiretamente, a presença e a viabilidade do vírus no sangue circulante, colostro e leite de caprinos infectados, bem como a possibilidadede infectar animais susceptíveis por inoculação, sendo o período de incubação de 45 a 60 dias. Não se demonstrou diferenças significativas dos teores séricos de proteína total, gamaglobulina e da atividade enzimática da glutamiltransferase - γGT em cabritos que receberam colostro de cabras infectadas ou não infectadas pelo mencionado vírus. / The frequency of occurrence of antibodies anti-caprine arthritis-encephalitis virus was studied in 14 herds in the State of São Paulo, during 2 years, using agar gel immunodiffusion. Prevalence was equal to 26.3%, and was significantly higher in animals kept under an intensive management scheme (31.8% - 733/2303), than in animals kept under a semi-extensive one (13.1% - 128- 977). Infection by the caprine arthritis-encephalitis virus increased gradual and significantly after 6 months of age. Infection was predominant in older animals. There was no gender influence on the prevalence of the disease, both in females (27.9% - 663/2375) and males (32.3% - 94/291). In relation to breed, prevalence of the disease was significantly higher in Anglo-Nubian (63.8% - 88/138) and Toggenbourg animals (56.0% - 28/50), than in the rest of the breeds studied: Saanen (27.4% - 673/2458), Alpine (11.9% - 59/497), Boer (5.9% - 2/34) and mixed breed animals (10.7% - 11/103). A clinical study of the animals infected by caprine arthritis-encephalitis virus was also performed. It was observed that 17.1% (64/374) of seroreagents presented the articular form of the disease and that 6.6% (17/249) of the seroreagent females presented the mammary form of the disease. The possibility of vertical, transplacentary transmission was lower than 3.8%. It was observed that the probability of infection of kids by colostrum of infected females was low (18.8% - 3/16). Antibodies passively acquired by colostrum ingestion lasted from 60 to 120 days. The presence and viability of the virus circulating in blood, colostrum and milk of infected animals, as well as the possibility of infection of susceptible animals by inoculation was indirectly demonstrated. Incubation period ranged from 45 to 60 days. There was no significant difference in serum levels of total protein and gammaglobulin, and in enzymatic activity of glutamiltransferase - γGT in kids that received colostrum from infected and non-infected females.

Characterization of the Caprine Arthritis Encephalitis Virus (CAEV) Rev N-Terminal Elements Required for Efficient Interaction With the RRE

The Caprine Arthritis Encephalitis Virus (CAEV) genome encodes three structural (gag, pol, and env) and three accessory (rev, tat, and vif) genes. The Rev-C protein regulates Gag, Pol and Env expression by transporting their mRNAs to the cytoplasm. Rev trans-activation requires binding of Rev to an RNA structure called the Rev Response Element (RRE-C). Previous mutational analyses have shown that two domains of Rev are required for its function. The basic domain mediates RRE binding and multimer formation, and the nuclear export signal (NES) mediates trans-activation. Preliminary experiments demonstrate that Rev-C N-terminal deletion mutants bind the RRE less avidly than does wildtype Rev. As a result, it was hypothesized that an additional domain located in the N-terminal exon of Rev-C was required for optimal RRE binding. To test this hypothesis, Rev-C alanine scanning mutants were generated and in vitro RRE binding assays were performed. Alteration of Rev-C amino acids K13, E14, N15, V19, T20, M21 and R27 dramatically decreased affinity for RRE-C. These data demonstrate that Rev-C N-terminal amino acids are required for optimal RRE-C binding and suggest that a third functional domain exists within the N-terminus of Rev-C.

Analysis of Caprine Arthritis Encephalitis Virus (CAEV) Temporal Gene Expression in Infected Cells

Caprine arthritis encephalitis virus (CAEV) is a lentivirus that is closely related to visna virus and more distantly related to the human lentivirus, Human Immunodeficiency Virus type 1 (HIV-1). The CAEV genome contains several small open reading frames (ORFs) that encode viral regulatory proteins. One of these non-structural proteins, Rev-C, is required for cytoplasmic transport of viral un/incompletely spliced mRNAs and efficient viral replication. In HIV-1 and visna virus, Rev is responsible for the temporal shift from non-structural protein synthesis to synthesis of structural proteins that is observed during the viral infectious cycle. Since it encodes a Rev protein, CAEV would be predicted to exhibit a similar temporal shift in gene expression during its replicative cycle. Immunoprecipitation analysis of 35S-pulse labeled, CAEV-infected goat synovial membrane (GSM) cells indicates that Rev-C is more abundant than is Gag at 12 h post-infection (PI); at later times PI Gag predominates. Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR) experiments using nuclear and cytoplasmic RNA from CAEV-infected GSM cells indicates that the viral unspliced gag mRNA accumulates significantly in the cytoplasm only after Rev is detected. These data indicate that a temporal shift from viral non-structural to structural gene expression occurs in CAEV infected GSM cells.

These tumors derived from transformed cells or artificially or naturally. This work describes a cell line of fetal goat cornea (CorFC) and its growth in media supplemented with low FBS aimed at producing virus antigens caprine arthritis-encephalitis for antibodies by agar gel immunodiffusion. The cell line has CorFC fibroblastic appearance and has been cultivated for more than two years, more than 40 passages without noticeable change in the morphology or the rate of cell multiplication. Of the 163 serum samples tested by micro-AGID with antigen (Ag) commercial (Biovetech, Brazil), 29 (17.79%) were positive, of these, 28 were also positive for micro-AGID-MEM with Ag and Ag -DMEM/12 Ag and 29 with RPMI-1640. We observed excellent agreement adjusted kappa (k) between the micro-AGID tests employing the commercial Ag, Ag-MEM and Ag-DMEM/F12 (k = 0.98) between the antigen and perfect commercial and Ag-RPMI 1640 (k = 1,00). Due to their growth characteristics of cells CorFC have behaved as a continuous lineage, which can only be definitively confirmed with continued passages. The studied cell culture media (MEM, DMEM/F12 and RPMI 1640) showed to be adequate to nourish the cell lineage CorFC. However, considering jointly the medium RPMI 1640 was the most recommended for cultivation, in supplementation of 2% FBS for scheduling and 0.1% to manutenção.As CorFC cell line grown in MEM, and DMEM/F12 RPMI 1640 proved to be highly permissive to CAEV replication of the virus in medium with low FBS, with production of higher quality antigens, reducing input costs and simplify the processes of purification of proteins, especially when the RPMI 1640 is used. / As células que crescem em cultivo in vitro estão divididas em três classes: primárias, de linhagem finita ou cultivos secundários e de linhagem contínua, que podem ser multiplicadas indefinidamente. Estas derivam de tumores ou células transformadas artificial ou naturalmente. Neste trabalho é descrita uma linhagem de células de córnea de feto caprino (CorFC) e seu cultivo em meios suplementados com baixo teor de SFB visando à produção de antígenos do vírus da artrite-encefalite caprina para pesquisa de anticorpos pela imunodifusão em gel de Agar. A linhagem celular CorFC apresenta aparência fibroblástica e vem sendo cultivada há mais de 2 anos, por mais de 40 passagens, sem alteração perceptível na morfologia ou na taxa de multiplicação celular. Das 163 amostras de soros testados pela micro-IDGA, com antígeno (Ag) comercial (Biovetech, Brasil), 29 (17,79%) apresentaram resultado positivo; dessas, 28 também foram positivas à micro-IDGA com Ag-MEM e Ag-DMEM/12 e 29 com o Ag-RPMI 1640. Foi observada ótima concordância ajustada de kappa (k) entre os testes de micro-IDGA empregando-se o Ag comercial, Ag-MEM e Ag-DMEM/F12 (k = 0,98) e perfeita entre o antígeno comercial e Ag-RPMI 1640 (k = 1,00). Devido às suas características de crescimento as células CorFC têm se comportado como de linhagem contínua, o que só poderá ser definitivamente comprovado com a continuação das passagens. Os meios de cultivo celular estudados (MEM, DMEM/F12 e RPMI 1640) demonstraram-se adequados para nutrir as células de linhagem CorFC. Entretanto, considerando em conjunto, o meio RPMI 1640 seria o mais recomendado para seu cultivo, nas suplementações de 2% de SFB para escalonamento e de 0,1% para manutenção.As células da linhagem CorFC cultivadas em MEM, DMEM/F12 e RPMI 1640 mostraram-se altamente permissíveis à replicação do vírus CAEV em meio com baixo teor de SFB, com produção de antígenos de melhor qualidade, redução de custos com insumos e simplificação no processos de purificação de proteínas, sobretudo quando o meio RPMI 1640 é usado.

Proteinograma da secreção láctea de cabras / Goat milk proteinogram

O objetivo do presente estudo foi estabelecer os valores de referência do proteinograma de soro lácteo por meio da técnica de eletroforese SDS-PAGE para a lactação plena e avaliar os efeitos do processo de secagem da glândula mamária, fase colostral e primeiro mês de lactação, fase de lactação, número de lactações, isolamento bacteriano e infecção pelo VCAE nas proteínas da secreção láctea de cabras da raça Saanen. Foram analisadas, entre 2007 e 2010, 545 amostras de leite provenientes de 185 cabras em diversas fases da lactação. Durante a lactação plena, baseado nos resultados dos intervalos de confiança, foram determinados os seguintes valores de referência: proteína total entre 2.940,0 e 3.050 mg/dL; proteína do soro lácteo entre 903,0 e 973,0 mg/dL; lactoferrina entre 68,0 e 77,0 mg/dL; albumina entre 88,0 e 97,0 mg/dL; imunoglobulina cadeia pesada entre 93,3 e 103,0 mg/dL; imunoglobulina cadeia leve entre 132,7 e 146,0 mg/dL; β-lactoglobulina entre 299,0 e 329,0 mg/dL e α-lactoalbumina entre 213,0 e 229,5 mg/dL. Os valores absolutos de proteína total, proteína do soro e frações protéicas aumentam durante a secagem da glândula. Antes da secagem predominavam as frações de β-Lg e α-La, a partir do 3º dia, ocorre o surgimento das novas frações e a alteração do perfil protéico sem que haja o predomínio de nenhuma fração. A fase colostral, primeiras 24 horas de lactação, determinam as maiores concentrações de proteína total, proteína do soro e frações protéicas que diminuem após as primeiras 12 horas de lactação estabilizando após o 5º dia. No colostro as imunoglobulinas são predominantes, e após o período de transição do colostro para o leite as frações β-Lg e α-La são predominantes. Nos primeiros 15 dias de lactação, devido à influência da fase colostral, observa-se que as concentrações de proteína total e proteína do soro lácteo são maiores. A partir desse momento permanecem estáveis voltando a aumentar no final da lactação. As frações protéicas do soro de leite (lactoferrina, albumina sérica, imunoglobulina de cadeia pesada, imunoglobulina de cadeia leve, β-Lg e α-La) também são máximas nos primeiros 15 dias de lactação e diminuem ao longo do período. A concentração de proteína do soro e suas frações em cabras primíparas foi menor quando comparadas com cabras pluríparas. O isolamento bacteriano não influencia as concentrações de proteína total do leite e proteína do soro lácteo de cabras, contudo a concentração de lactoferrina é maior e as concentrações de β-Lg e α-La são menores em amostras com isolamento bacteriano. O CAEV não influencia as concentrações de proteína total do leite e proteína do soro lácteo de cabras, contudo a concentração de lactoferrina é maior e a concentração de e α-La é menor em cabras sororeagentes positivas ao VCAE. / The aim of this study was to establish reference values of the whey protein through the technique of SDS-PAGE for the full lactation and to evaluate the effects of the dry period of the mammary gland, colostral phase and first month of lactation, lactation, lactation number, bacterial isolation and VCAE infection in proteins of milk secretion in Saanen goats. Were analyzed between 2007 and 2010, 545 milk samples from 185 goats at different stages of lactation. Prevailed prior to dry period the fractions of β-Lg and α-La from the 3rd day, occurs the emergence of new fractions and protein profile changes without the predominance of any fraction The colostral phase, the first 24 hours of lactation, determine the highest concentrations of total protein, whey protein and protein fractions that decrease after the first 12 hours of lactation stabilized after the 5th day. Immunoglobulin in colostrum is prevalent, and after the period of transition from colostrum to milk fractions β-Lg and α-La are predominant. In the first 15 days of lactation, due to the influence of colostral phase, it is observed that the concentrations of total protein and whey protein are higher. From then remain stable before rising again in late lactation. The protein fractions of whey (lactoferrin, serum albumin, immunoglobulin heavy chain, immunoglobulin light chain, β-Lg and α-La) are also maximal in the first 15 days of lactation and decrease during the period. The concentration of whey protein and protein fractions in heifers are smaller when compared with multiparous goats. Bacteria isolation does not influence the concentrations of total protein from milk and whey protein of goats, but the concentration of lactoferrin is increased and the concentrations of β-Lg and α-La is smaller in samples with bacterial isolation. The CAEV does not influence the concentrations of total protein and whey protein in goat, but the concentration of lactoferrin is higher and concentration of α-La is less in goat positive by the CAEV.

Para avaliar a fagocitose de células da série monócitos-macrófagos de cabras naturalmente infectadas pelo vírus da artrite encefalite caprina a vírus (VAEC), 30 cabras da raça Saanen foram utilizadas e alocadas em dois grupos distintos, sendo um grupo formado por 15 animais soropositivos à imunodifusão em gel de ágar para detecção de anticorpos séricos antivírus da AEC e o outro formado por 15 animais soronegativos ao teste. Células mononucleares de sangue periférico foram isoladas por gradiente de concentração e plaqueadas em placas de poliestireno para isolamento de células da série monócito-macrófago. Após adesão das mesmas, adicionou-se a bactéria Corynebacterium pseudotuberculosis como desafio antigênico. Dois tipos de fagocitose foram observados em relação ao número de bactérias internalizadas nos vacúolos citoplasmáticos dos macrófagos, isto é, verificou-se a presença de células fagocitando em média 12 bactérias e outro grupo fagocitando um número incontável de bactérias. Dessa forma, classificou-se a fagocitose em fagocitose + (até 12 bactérias) e fagocitose ++ (mais de 12 bactérias). Em relação à fagocitose total (fagocitose + e fagocitose ++) não foram verificadas diferenças estatística significativas entre os grupos experimentais (p < 0,41). Todavia, ao discriminar a fagocitose conforme a quantificação de bactérias fagocitadas, encontrou-se diferença estatística entre os grupos, sendo que o grupo positivo apresentou maior porcentagem de fagocitose ++ (p < 0,001). O grupo negativo apresentou maior porcentagem de fagocitose + (p < 0,012). Correlação positiva entre monócitos fagocitando e fagocitose ++ foi observada no grupo de animais soropositivos (p < 0,006), porém, não foi observada nenhuma correlação no grupo negativo (p < 0,49). Esses resultados demonstram uma possível alteração na intensidade da fagocitose de macrófagos de animais infectados com o VAEC, sugerindo que os animais com artrite encefalite caprina estejam mais susceptíveis à linfadenite caseosa. / To evaluate the phagocytosis from monocyte-macrophage line cells 30 naturally infected Saanen goats with caprine arthritis-encephalitis vírus (VCAE) were used, and divided uniformly in different groups according to agar gel immunodiffusion test (AGID). Peripheral Blood Mononuclear Cell (PBMC) was isolated by density gradient centrifugation. The monocyte-macrophage cells were separated from PBMC by their adherence properties. Afterwards, the phagocytosis function was assessed by phagocytosis assay using Corynebacterium psudotuberculosis as a source of antigen. Therefore, two distinct types of phagocytosis were observed and were set according to the number of bacteria within the cytoplasmatic vacuoles. Thus, the phagocytosis rates were also divided in two groups, on the first was observed up to 12 bacteria in the vacuoles; on the other hand in the second group an uncountable number of bacteria were usually seen. Consequently, the phagocytosis rates were also divided in phagocytosis + (up to 12 bacteria) and phagocytosis ++ (more than 12 bacteria). The results from the phagocytosis rates show any difference, however when the phagocytosis rates were separated in the order of the number of Corynebacterium pseudotuberculosis phagocyted, the phagocytosis ++ from positive animals in the sera test were higher than the negative one (p < 0.001). Nevertheless, the negative group presents higher pahgocytosis + (p < 0.012). Furthermore a positive and significant correlation between phagocytosis ++ and monocyte phagocytosis (p < 0.006) were also encountered in the positive animals, however the same were not observed in the negative group (p < 0.49). In face of, the results from the present trial point out to higher susceptibility to caseous linphadenitis in goats infected with VCAE due to the alteration on the phagocytosis strength in these animals.
