Tea phenols in bulk and nanoparticle form modify DNA damage in human lymphocytes from colon cancer patients and healthy individuals treated in vitro with platinum-based chemotherapeutic drugs

No / Tea catechin epigallocatechin-3-gallate (EGCG) and other polyphenols, such as theaflavins (TFs), are increasingly proving useful as chemopreventives in a number of human cancers. They can also affect normal cells. The polyphenols in tea are known to have antioxidant properties that can quench free radical species, and pro-oxidant activities that appear to be responsible for the induction of apoptosis in tumor cells. The bioavailability of these natural compounds is an important factor that determines their efficacy. Nanoparticle (NP)-mediated delivery techniques of EGCG and TFs have been found to improve their bioavailability to a level that could benefit their effectiveness as chemopreventives. AIM: The present study was conducted to compare the effects of TFs and EGCG, when used in the bulk form and in the polymer (poly[lactic-co-glycolic acid])-based NP form, in oxaliplatin- and satraplatin-treated lymphocytes as surrogate cells from colorectal cancer patients and healthy volunteers. NPs were examined for their size distribution, surface morphology, entrapment efficiency and release profile. Lymphocytes were treated in the Comet assay with oxaliplatin and satraplatin, washed and treated with bulk or NP forms of tea phenols, washed and then treated with hydrogen peroxide to determine single-strand breaks after crosslinking. The results of DNA damage measurements by the Comet assay revealed opposite trends in bulk and NP forms of TFs, as well as EGCG. Both the compounds in the bulk form produced statistically significant concentration-dependent reductions in DNA damage in oxaliplatin- or satraplatin-treated lymphocytes. In contrast, when used in the NP form both TFs and EGCG, although initially causing a reduction, produced a concentration-dependent statistically significant increase in DNA damage in the lymphocytes. These observations support the notion that TFs and EGCG act as both antioxidants and pro-oxidants, depending on the form in which they are administered under the conditions of investigation.

Extrinsic and intrinsic factors that regulate cell fitness in telomerase-inhibited human cells

Les extrémités des chromosomes eucaryotes ressemblent à une cassure double brin et, en tant que telles, peuvent conduire à l'activation indésirable de la réponse aux dommages de l'ADN. Les télomères sont une structure ribonucléoprotéique qui coiffe les extrémités des chromosomes et les protège contre l'activation indésirable de la réparation des dommages à l'ADN. Après chaque division cellulaire, on observe un raccourcissement progressif des télomères, ce qui limite leur potentiel prolifératif. Une enzyme spécialisée, la télomérase, reconstitue les télomères pour contrebalancer leur érosion. La télomérase est régulée à la baisse dans la plupart des cellules somatiques. Cependant, l'activité de la télomérase est détectée dans la plupart des cellules souches adultes, bien qu'à de faibles niveaux. Le déficit en télomérase a été associé à un groupe de "troubles de la biologie des télomères" (ou téloméropathies), englobant des maladies de vieillissement prématuré, des syndromes d'insuffisance de la moelle osseuse, des fibroses pulmonaires et des maladies du foie. À l'inverse, dans le cancer, environ 85 % des types de tumeurs sont positifs à la télomérase. Par conséquent, l'inhibition de la télomérase est depuis longtemps considérée comme une cible attrayante pour le traitement du cancer. Dans la présente étude, nous avons cherché à découvrir les facteurs qui affectent la fonction de la télomérase humaine et d'autres protéines associées aux télomères ou à la télomérase. Tout d'abord, nous nous sommes concentrés sur l'identification de nouveaux inhibiteurs de la télomérase à partir de composés naturels. Une nouvelle catéchine a été identifiée dans les extraits végétaux de Burkea africana. Les catéchines sont une classe de molécules que l'on trouve couramment dans le thé vert. La catéchine isolée a inhibé la télomérase humaine recombinante in vitro avec un IC50 de 16,19 μM. Dans un deuxième chapitre, nous avons utilisé un criblage d'édition de bases CRISPR dans une lignée cellulaire humaine pour étudier des mutations cliniquement pertinentes dans 22 gènes importants pour l'homéostasie des télomères. Nous avons identifié des variantes qui affectent négativement l'aptitude cellulaire, y compris certaines variantes précédemment annotées comme variantes de signification incertaine. Nous avons également détecté pour la première fois des variantes hTERT qui confèrent une résistance à la petite molécule BIBR1532, un inhibiteur de la télomérase. Nous avons montré que ces allèles résistants aux médicaments permettent l'immortalisation cellulaire et ont un potentiel tumorigène accru. L'ensemble de ces études souligne l'importance de la télomérase humaine pour le maintien des télomères et la santé cellulaire, contribuant ainsi à une meilleure compréhension du rôle de la télomérase dans le cancer et les troubles de la biologie des télomères. / The extremities of eukaryotic chromosomes resemble a double-stranded break and, as such, can lead to the unwanted activation of the DNA damage response. Telomeres are a ribonucleoprotein structure that caps the ends of the chromosomes and protects them from the unwanted activation of DNA damage recognition and repair processes. After each cellular division, progressive telomere shortening is observed, limiting cellular proliferative potential. A specialized enzyme called telomerase replenishes telomeres to counterbalance telomere erosion. Telomerase is downregulated in most somatic cells. However, telomerase activity is detected in most adult stem cells, although at low levels. Telomerase deficiency has been linked to a group of “Telomere Biology Disorders” (or telomeropathies), encompassing premature aging diseases, bone marrow failure syndromes, pulmonary fibrosis and liver diseases. Conversely, in cancer, around 85% of tumour types are telomerase-positive. Therefore, telomerase inhibition has long been considered an attractive target for cancer therapy. In the present study, we aimed to uncover factors that affect the function of human telomerase and other telomere or telomerase-associated proteins. Firstly, we focused on identifying new telomerase inhibitors from natural compounds. A new catechin was identified in the plant extracts from Burkea africana. Catechins are a class of molecules commonly found in green tea. The isolated catechin inhibited recombinant human telomerase in vitro with an IC50 of 16.19 μM. In the second chapter, we employed a CRISPR base editing screen in a human cell line to investigate clinically-relevant mutations in 22 genes important for telomere homeostasis. We identified variants that negatively affected cell fitness, including some variants previously annotated as variants of uncertain significance. Also, we uncovered hTERT variants that confer resistance to the small molecule BIBR1532, a telomerase inhibitor. We showed that these drug-resistant alleles permit cellular immortalization and exhibit tumorigenic potential at levels comparable to wild-type telomerase. Combined, these studies highlight the importance of human telomerase for telomere maintenance and cell fitness, thereby furthering our understanding of the role of telomerase in cancer and telomere biology disorders.

It is composed of a fluid extract about Piptadenia colubrina, Schinus terebinthifolius, Cereus peruvianus and Physalis angulata. Among the plants in their composition, S. terebinthifolius and P. colubrina have in common phenolic compounds which are assigned most of its pharmacological effects. The tannins, gallic acid and catechin were selected as markers for quality control. The aim of this study was the development and validation of analytical method by HPLC/UV/DAD for the separation and simultaneous quantification of gallic acid (GAC) and catechin (CTQ) in Sanativo?. The chromatographic system was to stationary phase, C-18 RP column, 4,6 x 150 mm (5 mm) under a temperature of 35 ? C, detection at 270 and 210 nm. The mobile phase consisted of 0.05% trifluoroacetic acid and methanol in the proportions 88:12 (v/v), a flow rate of 1 ml/min. The analytical method presented a retention factor of 0.30 and 1.36, tail factor of 1.8 and 1.63 for gallic acid and catechin, respectively, resolution of 18.2, and theoretical plates above 2000. The method validation parameters met the requirements of Resolution n ? 899 of May 29, 2003, ANVISA. The correlation coefficient of linear regression analysis for GAC and CTQ from the standard solution was 0.9958 and 0.9973 and when performed from the Sanativo? 0.9973 and 0.9936, the matrix does not interfere in the range 70 to 110 %. The limits of detection and quantification for GAC and CQT were 3.25 and 0.863, and 9.57 and 2.55 mg/mL, respectively. The markers, GAC and CQT, showed repetibility (coefficient of variation of 0.94 % and 2.36 %) and satisfactory recovery (100.02 ? 1.11 % and 101.32 ? 1.36 %). Guarana (Paullinia cupana Kunth.) is used as an invigoranting and challenging due to the presence in the seeds of methylxanthines, especially caffeine and theobromine and theophylline lesser amount, and tannins. The test is an dissolution of the tests used in assessing the quality of these medicines and throught this you can evalueted the release of the active principle from a solid form in an environment that simulates the location of its absorption. This study aimed to evaluete the behavior of dissolution of capsules and tablets containinf guarana. This study investigated initially, spectrophotometric and chromatographic methods for quantification of tannins and methylxanthines in pharmaceutical forms and the means of dissolution. The results obtained by spectophotometry showed lack of specificity of this method under our experimental conditions for quantification of methylxanthines. In the analysis of tannins, the procedure is not feasible for an routine laboratory to be long and larorious. The method by High Performance Liquid Chromatography allowed the quantification of the five markers simultaneously, with precision, accuracy, selectivity and robustness. The dissolution test using apparatus as man, 37 ºC ± 0,5 ºC, speed of 75 rpm in 900 mL of 0.1 M HCl medium pH 1,2 provided the best results was validated and used in assessing the behaviour of dissolution of the capsules and tablets based on guarana. It was found that all markers were quantified by the method are dissolved between 75% and 80% in time of 10 minutes. An analysis of the capsules did not break in any of the conditions employed. For the tablet was also observed in non-compliance, non-identification of catechin and epicatechin in the finished product, worrying factor, as the phenolic compounds are markers that should be present in the guarana powder. Considering the increasing use of plants is a clear need for definition of quality standards to ensure its therapeutic action and accordingly the study of dissolution is an essential tool in ensuring the quality of plants / Os fitoterápicos são medicamentos obtidos empregando-se exclusivamente matérias-primas ativas vegetais. O guaraná (Paullinia cupana Kunth.) é utilizado como revigorante e estimulante devido à presença, em suas sementes, de metilxantinas, principalmente cafeína e em menor quantidade teofilina e teobromina, além de taninos condensados. O teste de dissolução é um dos ensaios empregados na avaliação da qualidade destes medicamentos e por meio deste pode se avaliar a liberação do princípio ativo a partir de uma forma farmacêutica sólida em um meio que simula o local de sua absorção. Este trabalho visou avaliar o comportamento de dissolução de cápsulas e comprimidos contendo guaraná. Neste estudo foram investigados inicialmente, métodos espectrofotométricos e cromatográficos para a quantificação de metilxantinas e taninos nas formas farmacêuticas e no meio de dissolução. Os resultados obtidos por espectrofotometria demonstraram ausência de especificidade deste método nas condições experimentais empregadas para quantificação de metilxantinas. Na análise dos taninos, o procedimento é inviável para uma rotina laboratorial por ser demorado e trabalhoso. O método por Cromatografia Liquida de Alta Eficiência permitiu a quantificação dos marcadores simultaneamente, com precisão, exatidão, seletividade e robustez. O ensaio de dissolução utilizando pá como aparato, 37 ºC ± 5 ºC, velocidade de 75 rpm em 900 mL de meio HCl 0,1 M pH 1,2 forneceu os melhores resultados, foi validado e utilizado na avaliação do comportamento de dissolução de cápsulas e comprimidos a base de guaraná. Foi verificado que todos os marcadores quantificados pelo método encontraram-se dissolvidos entre 75% e 80%, no tempo de 10 minutos. Uma das cápsulas analisadas não dissolveu em nenhuma das condições empregadas. Para o comprimido também foi observada uma não conformidade, a não identificação de catequina e epicatequina no produto acabado, fator preocupante, visto que os taninos condensados são marcadores que devem estar presentes no pó de guaraná. Considerando o crescente uso dos fitoterápicos fica clara a necessidade de definição de parâmetros de qualidade que assegurem a sua ação terapêutica e nesse sentido o estudo de dissolução constituem uma ferramenta essencial na garantia de qualidade dos fitoterápicos

Efecto de la aplicación de copigmentos vegetales sobre la composición fenólica y aromática de vinos tintos de Monastrell elaborados con diferentes técnicas de vinificación

[ES] El mercado actual del vino tinto demanda vinos de gran potencia visual, elevada concentración aromática y que no resulten excesivamente astringentes ni amargos. Asimismo, el consumidor final demanda cada vez más vinos que se desarrollen en una viticultura más orgánica que garantice la sostenibilidad del cultivo. Para cumplir con estas expectativas, se está limitando la utilización de compuestos químicos en el viñedo, sustituyéndolos por otros naturales con menor impacto medioambiental, y se están utilizando en bodega técnicas como la maceración prefermentativa, que contribuye a la extracción de antocianos y aromas, o la microoxigenación, que permite estabilizar los compuestos polifenólicos en un tiempo reducido. El color de los vinos depende de su concentración en antocianos, pero también la forma y el estado en que éstos se encuentren en el vino, que depende de diferentes factores, siendo uno de ellos el fenómeno de la copigmentación como paso anterior a la polimerización y estabilización. Además, el oxígeno juega un papel crucial en la evolución del color, así como en la oxidación de los polifenoles y en la formación de complejos más estables. El aroma también es muy importante en los vinos, ya que es uno de los primeros aspectos que percibimos. Por ello, es de gran importancia la utilización de técnicas de vinificación que permitan extraer el mayor potencial aromático de los precursores que se encuentran en las uvas. Para conseguir incrementar la calidad de los vinos tintos elaborados con la variedad Monastrell, estableciendo una metodología de elaboración que permita mejorar la concentración polifenólica y aromática de estos vinos, se ha estudiado el efecto de la aplicación en la uva, 10 días antes de la vendimia, de sustancias copigmentantes, tales como, extractos vegetales (romero, trigo sarraceno y té verde) y copigmentos puros (ácido cafeico, rutina y catequina). Asimismo, los copigmentos puros también se aplicaron a la entrada en bodega. Se ensayaron dos técnicas enológicas diferentes: la vinificación tradicional y la maceración prefermentativa en frío (6-8ºC), seguida de vinificación tradicional. Por último, una vez concluida la fermentación maloláctica, los vinos obtenidos fueron sometidos a una adición controlada de acetaldehído durante 45 días, seguido de un proceso de conservación en botella de 12 meses. Los resultados mostraron que los tratamientos con copigmentos, especialmente con extractos vegetales ricos en copigmentos, incrementaron la concentración de antocianos polimerizados y totales, y de compuestos aromáticos favorables para la calidad, destacando el extracto de romero por su acción directa sobre el color de los vinos de Monastrell. Utilizar la maceración prefermentativa en la elaboración de los vinos procedentes de tratamientos de copigmentación, ha permitido incrementar la concentración de polifenoles totales y taninos condensados, así como la de ésteres y acetatos. La microoxigenación, mediante la aplicación de acetaldehído durante la conservación de los vinos tratados previamente con copigmentos, dio lugar a un incremento del porcentaje de antocianos polimerizados, de la concentración de antocianos, de taninos condensados y de polifenoles totales, no afectando a la composición aromática de los vinos. Por todo ello, se puede establecer que la combinación de tratamientos de copigmentación en el campo, especialmente con extractos vegetales, de maceración prefermentativa durante la elaboración, y de microoxigenación durante la conservación, permite elaborar vinos de Monastrell con un color más estable, mayor concentración de compuestos polifenólicos, y mayor concentración de ésteres y acetatos, compuestos relacionados con aromas positivos florales, frutales y especiados. / [CA] El mercat actual del vi negre demana vins de gran potència visual, elevada concentració aromàtica i que no siguen excessivament astringents ni amargs. Així mateix, el consumidor final demana cada vegada més vins que es desenvolupen en una viticultura més orgànica que garanteisca la sostenibilitat del cultiu. Per complir amb aquestes expectatives, s'està limitant la utilització de compostos químics a la vinya, substituint-los per altres naturals amb menor impacte mediambiental, i s'estan utilitzant al celler tècniques com la maceració prefermentativa, que contribueix a l'extracció d'antocians i aromes, o la microoxigenació, que permet estabilitzar els compostos polifenòlics en un temps reduït. El color dels vins depèn de la seua concentració en antocians, però també en la forma i l'estat en que aquests es troben en el vi, que depèn de diferents factors, siguent un d'ells el fenòmen de la copigmentació com pas anterior a la polimerització i l'estabilització. A més a més, l'oxígen juga un paper crucial en l'evolució del color, així com en l'oxidació dels polifenols i en la formació dels complexos més estables. L'aroma també és molt important en els vins, ja que és un dels primers aspectes que percebem. Per això, és de gran importància la utilització de tècniques de vinificació que permeten extraure el major potencial aromàtic dels precursors que es troben al raïm. Per aconseguir augmentar la qualitat dels vins negres elaborats amb la varietat Monastrell, establint una metodologia d'elaboració que permeta millorar la concentració polifenòlica i aromàtica d'aquests vins, s'ha estudiat l'efecte de l'aplicació en el raïm, 10 dies abans de la verema, de substàncies copigmentants, com ara, extractes vegetals (romer, blat negre i te verd) i copigments purs (àcid cafeic, rutina i catequina). Així mateix, els copigments purs també es van aplicar a l'entrada al celler. Es van assajar dos tècniques enològiques diferents: la vinificació tradicional i la maceració prefermentativa en fred (6-8ºC), seguida de vinificació tradicional. Finalment, una vegada conclosa la fermentació malolàctica, els vins obtinguts van ser sotmesos a una addició controlada d'acetaldehid durant 45 dies, seguit d'un procés de conservació en ampolla de 12 mesos. Els resultats van mostrar que els tractaments amb copigments, especialment amb extractes vegetals rics en copigments, van augmentar la concentració d'antocians polimeritzats i totals, i de compostos aromàtics favorables per a la qualitat, destacant l'extracte de romer per la seua acció directa sobre el color dels vins de Monastrell. Utilitzar la maceració prefermentativa en l'elaboració dels vins procedents de tractaments de copigmentació, ha permès augmentar la concentració de polifenols totals i tanins condensats, així com la d'èsters i acetats. Furthermore, the final consumer increasingly demands developed wines in a more organic viticulture in order to ensure the sustainability of the crop. To meet these expectations, the use of chemical compounds in the vineyard is being limited, replacing them by another natural ones with less environmental impact, and techniques such as pre-fermentation maceration are being used in the winery, which contributes to the extraction of anthocyanins and aromas, or micro-oxygenation, that allows polyphenolic compounds to be stabilized in a reduced time. The colour of wines depends on their concentration in anthocyanins, but also the form and state in which they are found in the wine, which depends on several factors, one of them being the phenomenon of copigmentation as a previous step to polymerization and stabilization. In addition, the oxygen has a crucial role in colour's evolution, as well as, in the oxidation of polyphenols and in the formation of more stable complexes. Aroma is also very important in wines, due to the fact that is one of the first aspects that we perceive. For this reason, the use of vinification techniques that allow extracting the greatest aromatic potential of the precursors found in the grapes, is of great importance. In order to increase the quality of red wines made with the Monastrell variety, establishing a production methodology that allows improving the polyphenolic and aromatic concentration of these wines, the effect of the application on the grape, 10 days before harvest, has been studied, of copigmenting substances such as plant extracts (rosemary, buckwheat and green tea) and pure copigments (caffeic acid, rutin and catechin). Likewise, pure copigments were also applied in the cellar entrance. Two different enological techniques were tested: traditional vinification and cold pre-fermentation maceration (6-8ºC), followed by traditional vinification. Finally, once the malolactic fermentation was concluded, the wines obtained were subjected to a controlled addition of acetaldehyde for 45 days, followed by a 12-month bottle preservation process. The results showed that treatments with copigments, especially with plant extracts rich in copigments, increased the concentration of polymerized and total anthocyanins, and aromatic compounds favorable for quality, highlighting rosemary extract for its direct action on the colour of wines of Monastrell. Using pre-fermentation maceration in the production of wines from copigmentation treatments has made it possible to increase the concentration of total polyphenols and condensed tannins, as well as that of esters and acetates. Micro-oxygenation through the application of acetaldehyde during the conservation of wines previously treated with copigments gave rise to an increase in the percentage of polymerized anthocyanins, in the concentration of anthocyanins, in condensed tannins and in total polyphenols, without affecting the aromatic composition of the wines. For all of these reasons, it can be established that the combination of copigmentation treatments in the field, particularly with plant extracts, pre-fermentation maceration during production and micro-oxygenation during storage, allows the production of Monastrell wines with a more stable colour, higher concentration of polyphenolics, and higher concentration of esters and acetates, compounds related to positive floral, fruity and spicy aromas. / Anaya Martínez, JA. Dietary flavonoid (-)epicatechin stimulates phosphatidylinositol 3-kinase-dependent anti-oxidant response element activity and up-regulates glutathione in cortical astrocytes

No / Flavonoids are plant-derived polyphenolic compounds with neuroprotective properties. Recent work suggests that, in addition to acting as hydrogen donors, they activate protective signalling pathways. The anti-oxidant response element (ARE) promotes the expression of protective proteins including those required for glutathione synthesis (xCT cystine antiporter, gamma-glutamylcysteine synthetase and glutathione synthase). The use of a luciferase reporter (ARE-luc) assay showed that the dietary flavan-3-ol (-)epicatechin activates this pathway in primary cortical astrocytes but not neurones. We also examined the distribution of NF-E2-related factor-2 (Nrf2), a key transcription factor in ARE-mediated gene expression. We found, using immunocytochemistry, that Nrf2 accumulated in the nuclei of astrocytes following exposure to tert-butylhydroquinone (100 microM) and (-)epicatechin (100 nM). (-)Epicatechin signalling via Nrf2 was inhibited by wortmannin implicating a phosphatidylinositol 3-kinase-dependent pathway. Finally, (-)epicatechin increased glutathione levels in astrocytes consistent with an up-regulation of ARE-mediated gene expression. Together, this suggests that flavonoids may be cytoprotective by increasing anti-oxidant gene expression.

Régulation de la fonction vasculaire pendant le vieillissement : rôles de l’environnement post-natal et du gène suppresseur de tumeurs p53

La dysfonction endothéliale vasculaire constitue un marqueur précoce des maladies cardiovasculaires car l’endothélium est l’une des premières cibles des facteurs de risque cardiovasculaire. La présence d'un stress chronique engendré par les facteurs de risque cardiovasculaire sollicite les mécanismes de défense endogènes, tels que les enzymes antioxydantes, qui servent au maintien de la fonction endothéliale. L’environnement vasculaire auquel l’endothélium est exposé a un effet direct sur son fonctionnement à long terme. Certaines habitudes de vie sont ainsi associées à une bonne santé cardiovasculaire. Par exemple, la diète méditerranéenne et/ou la pratique régulière de l’exercice physique aident à maintenir une fonction endothéliale adéquate et à réduire l’incidence des maladies cardiovasculaires. D'autre part, certains gènes clés, comme le gène suppresseur de tumeurs p53, régulent plusieurs voies métaboliques importantes pour préserver l’intégrité des cellules endothéliales. Nous posons l’hypothèse que l’environnement vasculaire post-natal influence la mise en place de mécanismes de défenses endogènes tels que les enzymes antioxydantes afin de faire face à des stress plus tard dans la vie. Notre objectif global était d’évaluer les impacts d’interventions post-natales bénéfiques et d’une diminution endogène du gène suppresseur de tumeurs p53, sur la fonction endothéliale vasculaire et sur sa capacité à faire face à un stress métabolique. Dans une première étude, nous avons soumis des souris saines C57Bl/6 dès leur sevrage et jusqu’à l’âge de 9 mois, à un programme d’exercice physique volontaire (course dans une roue) ou à un antioxydant (catéchine), comparé à un groupe de souris sédentaires et sans antioxydant. Puis les interventions ont été stoppées et une diète riche en gras a été introduite, ou non, pour une période de 3 mois; les souris ont été sacrifiées à l'âge de 9 ou 12 mois. Nous avons observé que l’exercice a protégé les cellules endothéliales des effets délétères induits par la diète riche en gras en préservant la fonction endothéliale par le maintien d’un profil rédox sain et en évitant la hausse de l’inflammation. La catéchine a maintenu la fonction endothéliale aortique, mais n’a pas prévenu le profil inflammatoire en présence de la diète riche en gras. Finalement, chez les souris sédentaires, la fonction endothéliale a été détériorée en présence de la diète riche en gras, sans indice d’inflammation vasculaire. Dans une seconde étude, des souris partiellement déficientes en p53 (p53+/-) et contrôles C57Bl/6 ont été exposées à la même diète riche en gras à partir de 3 mois et ce jusqu’à l’âge de 6 mois. Notre raisonnement était basé sur la démonstration que p53 est un régulateur de l’expression des enzymes antioxydantes in vitro. Chez les souris p53+/-, les cellules endothéliales ont été protégées du stress induit par l’hypercholestérolémie engendrée par la diète riche en gras. Cependant, chez les souris p53+/- cette protection pourrait être secondaire à un métabolisme accru des acides biliaires, qui en prévenant la hausse de cholestérol, protègerait indirectement l'endothélium. Nous avons donc pu démontrer l’importance de l’environnement vasculaire sur la fonction endothéliale. La diète riche en gras a stimulé certains mécanismes de défense vasculaires tels que la voie des EDHF et la superoxyde dismutase afin de maintenir la fonction endothéliale malgré les conditions pro-athérosclérotiques. Nous avons observé que l’exercice et la catéchine influencent différemment l’endothélium malgré leurs capacités antioxydantes. Ces études soulignent la sensibilité de l’endothélium aux changements dans l’environnement vasculaire. En accord avec le vieillissement de la population et la progression des maladies cardiovasculaires, la proportion de personnes ayant une dysfonction endothéliale augmente. Ainsi, une meilleure compréhension des mécanismes ou d’interventions qui permettent le maintien de la fonction endothéliale à long terme s’avère utile. / Endothelial dysfunction is an early marker of atherosclerosis and cardiovascular diseases. Cardiovascular risk factors generate a chronic stress, challenging endogenous defense mechanisms that are critical to maintain endothelial function, such as antioxidant enzymes. The vascular environment impacts the integrity and long-term function of endothelial cells. Thus, a healthy lifestyle is beneficial for cardiovascular health: regular physical training and/or a Mediterranean diet are associated with the maintenance of endothelial function and a lower incidence of cardiovascular diseases. On the other hand, some key genes such as tumor suppressor gene p53, are known to regulate numerous cellular functions that are necessary to maintain endothelial cells integrity. We hypothesized that the post-natal vascular environment impacts the development of endogenous cellular defense mechanisms such as antioxidant enzymes, in order to protect against vascular stress that will occur later in life. Our major goal was to evaluate the impact of post-natal interventions and endogenous reduction of p53 expression on vascular endothelial function and its capacity to resist against a metabolic stress. In our first study, healthy C57Bl/6 mice were exposed from weaning to the age of 9 months to physical voluntary training (running wheel) or to the antioxidant catechin, and were compared to physically inactive mice that did not receive catechin. Then, exercise and catechin were stopped and mice were subjected to a regular or a high fat diet for 3 months; mice were sacrificed either at the age of 9 or 12 months. In trained mice, we observed that exercise prevented endothelial dysfunction and inflammation induced by the high fat diet. In catechin-treated mice, aortic endothelial function was maintained despite exposure to the high fat diet, but an inflammatory profile was observed. In physically inactive mice, the high fat diet induced endothelial dysfunction without inflammation. In our second study, mice partially deficient in p53 (p53+/-) were exposed to the same high fat diet from 3 to 6 months of age. Our rationale was based on the discovery that in vitro, p53 regulates antioxidant enzymes gene expression. In p53+/- mice, endothelial cells were protected from the stress of hypercholesterolemia induced by the high fat diet. However, this endothelial protection could be linked with an unexpected enhanced bile acid metabolism in p53+/- mice: low endogenous expression of p53 prevents the rise in plasma total cholesterol when fed a high fat diet, indirectly protecting the endothelium. In summary, we were able to demonstrate the importance of the vascular environment on endothelial function. The pro-atherosclerotic environment induced by the high fat diet stimulated vascular defense mechanisms, as observed by the activation of the compensatory EDHF pathway and superoxide dismutase activity, to maintain an adequate endothelial function. We observed that exercise and catechin had a different impact on the endothelium despite their antioxidant properties. These studies demonstrate the sensitivity of the endothelium to changes within the vascular environment. As the population is aging and the incidence of cardiovascular diseases is increasing, endothelial dysfunction will be more frequent. Therefore, a better comprehension of mechanisms or interventions that can protect endothelial function can only be beneficial.

Plants are continuously confronted with a multitude attack either abiotic but also biotic in nature. Interestingly, despite the abundance of bacteria that plant has to face, only few are able to induce death or disease in the host plant. It is therefore likely that, in addition to secondary metabolites with antimicrobial properties, plants also synthesize secondary metabolites which are able to inhibit the expression of virulence genes in bacteria without affecting either growth or viability, which allows plants to host willingly or not bacterial populations. This work focuses on the identification of such metabolites in Malagasy plants (genera Dalbergia and Combretum) and the demonstration of their inhibitory effect on the expression of virulence genes in two different pathosystems: Rhodococcus fascians (a phytopathogen) and Pseudomonas aeruginosa (an opportunistic pathogen). Thus, two metabolites were isolated using a combination of chromatographic techniques coupled with tests that evaluate the expression of certain genes involved in the virulence mechanisms of these bacteria. The first is a new prenylated isoflavanone, named perbergin, isolated from the bark extract of D. pervillei. It was shown that the perbergin target attR gene expression, encoding a LysR-type transcriptional regulator that plays a key role in regulating the expression of virulence genes of R. fascians and the transition from an epiphytic to a pathogenic lifestyle. Therefore, we have also shown that the expression of all virulence genes known to date in R. fascians is also affected while the expression of genes involved in epiphytic fitness of the bacteria is not altered. In addition, the application of perbergin at the time of infection of plants susceptible to R. fascians shows that this molecule reduces in vivo the virulence of R. fascians, highlighting the potential of perbergin as an anti-infective agent. The second is a flavonoid known as catechin, isolated from the bark extract of C. albiflorum. Catechin significantly inhibits the expression of genes that regulate the mechanism of quorum sensing in P. aeruginosa such as lasI, LasR, rhlI and rhlR but also lasB and rhlA which expression depends on quorum sensing. Therefore, the production of virulence factors such as pyocyanin and elastase is significantly affected. Because of the limited number of our arsenal of antibiotics and their increasing ineffectiveness, the identification of these compounds create a path to an alternative in the fight against pathogenic bacteria and multidrug resistance of pathogenic bacteria to antibiotics. Our results also demonstrate the richness of Malagasy plants as (re)sources of new therapeutic molecules and the importance of widening the range of bacterial targets to be investigated to develop new strategies to fight within the endless war that we are waging against bacteria pathogens.<p><p>Les plantes sont continuellement confrontées à une multitude d’attaques qu’elles soient de nature abiotique ou surtout biotique. Il est intéressant de noter que malgré la multitude de bactéries auxquelles les plantes doivent faire face, seules quelques unes sont capables d’induire la mort ou une maladie chez la plante hôte. Il est dès lors fort probable que, outre les métabolites secondaires ayant des propriétés antimicrobiennes, les plantes synthétisent également des métabolites secondaires capables d’inhiber l’expression des gènes de virulence chez les bactéries sans toutefois affecter ni leur croissance ni leur viabilité, ce qui permet aux plantes de contenir les populations bactériennes qu’elles hébergent de gré ou de force. Ce travail porte sur l’identification de ce type de métabolites dans des plantes malgaches (genres Dalbergia et Combretum) et la démonstration de leurs effets inhibiteurs sur l’expression de gènes de virulence chez deux pathosystèmes différents: Rhodococcus fascians (un phytopathogène) et Pseudomonas aeruginosa (un pathogène opportuniste). Ainsi, deux métabolites ont été isolés en utilisant une combinaison de techniques chromatographiques couplées avec des tests qui évaluent l’expression de certains gènes impliqués dans les mécanismes de virulence de ces bactéries. Le premier est un nouvel isoflavanone prénylé, nommé perbergine, isolé à partir de l’extrait d’écorces de D. pervillei. Il a été montré que la perbergine cible l’expression du gène attR, codant un régulateur transcriptionnel de type LysR qui joue un rôle clé dans la régulation de l’expression des gènes de virulence de R. fascians et qui assure la transition entre un mode de vie épiphyte et le mode pathogène. En conséquence, nous avons également montré que l’expression de l’ensemble des gènes de virulence connu à ce jour chez R. fascians est également affectée alors que l’expression de gènes impliqués dans l’aptitude épiphyte de la bactérie n’est pas altérée. Par ailleurs, l’application de perbergine au moment de l’infection de plantes sensibles à R. fascians montre que cette molécule atténue la virulence de R. fascians in vivo, mettant en exergue le potentiel de la perbergine comme agent anti-infectieux. Le deuxième est un flavonoïde, connu sous le nom de catéchine, isolé de l’extrait d’écorces de C. albiflorum. La catéchine inhibe significativement l’expression des gènes régulateurs du mécanisme du quorum sensing chez P. aeruginosa tels que lasI, lasR, rhlI et rhlR et également lasB et rhlA dont l’expression dépend du quorum sensing. En conséquence, la production des facteurs de virulence tels que la pyocyanine et l’élastase est significativement affectée. Compte tenu de l’appauvrissement de notre arsenal d’antibiotiques et de leur inefficacité croissante, l’identification de ces composés ouvre une voie alternative de lutte contre les bactéries pathogènes et la multirésistance des bactéries pathogènes aux antibiotiques. Nos résultats démontrent également la richesse des plantes malgaches comme (res)sources de nouvelles molécules thérapeutiques et l’importance d’élargir le champ des cibles bactériennes à investiguer pour développer de nouvelles stratégies de lutte dans la guerre sans fin que nous menons contre les bactéries pathogènes. / Doctorat en Sciences / info:eu-repo/semantics/nonPublished
