"Avaliação da expressão dos receptores de interleucina-8, CXCR1 e CXCR2, e da atividade proliferativa em fibroblastos de quelóide e de pele normal" / Determination of the interleukin-8 receptors CXCR1 and CXCR2, and proliferative activity in keloids and normal skin fibroblasts

O quelóide é um tumor fibroso benigno que ocorre durante a cicatrização da pele em indivíduos geneticamente predispostos. A cicatrização é um processo biológico complexo e depende da interação de diferentes estruturas teciduais e de um grande número de tipos celulares residentes e infiltrativos, que produzem citocinas. A interleucina 8 (IL-8), citocina pró-inflamatória, é super-expressa pelos fibroblastos durante o desenvolvimento do tecido de granulação, acelerando o processo de cicatrização. Como o quelóide resulta de uma reparação tecidual anormal após lesão da pele, o presente estudo teve por objetivo determinar a expressão dos receptores da IL-8, CXCR1 e CXCR2, e a capacidade proliferativa, pelo ciclo celular, dos fibroblastos queloideanos cultivados e extraídos ex vivo, por citometria de fluxo. Fibroblastos de cicatriz queloideana e de pele normal foram obtidos de 21 pacientes da raça negra, com idade variando entre 10 e 40 anos, de lesões com até 2 anos de evolução. Em nosso estudo constatamos expressão reduzida dos receptores para a IL-8, CXCR1(35,7%±11,2) e CXCR2 (27,8%±11,3), em fibroblastos de cicatriz queloideana cultivados, comparando com a pele normal (44,1±16,2 e 46,3±27,1 respectivamente). Entretanto, essa diferença só foi significante para o receptor CXCR2. A baixa expressão desses receptores poderia ser decorrente da atividade de metaloproteinases, que regulam a expressão de proteínas da superfície celular, através de clivagem enzimática, ou a capacidade reduzida de internalização e a reciclagem de receptores, mantida por filamentos de actina do citoesqueleto, que nos fibroblastos do quelóide estão diminuídos. Em relação ao ciclo celular de fibroblastos cultivados do quelóide e da pele normal, verificamos diferenças não significantes da capacidade de replicação (fase S do ciclo celular) e de apoptose. No quelóide observamos significante aumento de células na fase G2/M, indicando aumento da velocidade de divisão celular. Para confirmar esses achados estudamos o ciclo celular de fibroblastos extraídos ex vivo, da porção periférica e central do quelóide e da pele normal. Os fibroblastos da porção periférica apresentaram porcentagem significantemente maior de células com capacidade replicativa, fase S do ciclo (22,9% ± 11,6), em relação à porção central (4,7% ± 2,9) e à pele normal (6,8% ± 4,9), e maior velocidade de divisão celular, fase G2/M (18,6 ± 12,0), em relação à porção central (35,6 ± 7,0) e pele normal (32,3 ± 6,9). Verificamos que a porção central apresentou maior porcentagem de células em apoptose (7,0% ± 2,1), comparado à porção periférica (4,9% ± 1,9) e pele normal (2,0% ± 0,86). Esses dados indicam que as células da porção periférica do quelóide parecem ser responsáveis pela elevada taxa de proliferação, justificando o crescimento expansivo a partir das margens da cicatriz queloideana, com desenvolvimento de lesão semelhante a tumor, bem como a porção central ser responsável pela fibrose, contendo células quiescentes e apoptóticas. Esses resultados sugerem modulação diferencial das reações celulares através das vias de sinalização para proliferação ou morte celular programada. Neste sentido, a baixa expressão dos receptores da IL-8, CXCR1 e principalmente de CXCR2, nos fibroblastos do quelóide sugere capacidade reduzida da IL-8 em promover cicatrização acelerada. A baixa atividade da IL-8 sobre os fibroblastos queloideanos estaria promovendo desregulação da resposta inflamatória e com isso atraindo novas células inflamatórias para o local e produzindo sinais alterados, como grande produção da citocina TGFβ. Essa desregulação do processo de cicatrização, com alteração de citocinas e da matriz extracelular, poderia ser responsável pelas duas populações de fibroblastos, uma proliferativa na periferia e outra quiescente e apoptótica na porção central. Finalizando, podemos concluir que nossos resultados correspondem às alterações histológicas e clínicas do quelóide que se expande nos limites da lesão. / A keloid is a benign fibrous tumor that occurs during wound healing in genetically predisposed individuals. Healing is a complex biological process and depends on the interaction of different tissue structures and a great number of resident and infiltrative cell types. The interleukin-8 (IL-8), a proinflammatory chemokine, showed higher expression in fibroblasts during the development of the granulation tissue, promoting more rapid tissue maturation. Since keloids result from abnormal wound healing, the objective of the present study was to determine the expression of CXCR1 and CXCR2, IL-8 receptors, and the proliferation capacity, throughout the cell cycle, of the keloid fibroblasts extracted ex vivo and those submitted to in vitro cultivation. Normal skin and keloid scar fibroblasts were obtained from 21 African-Brazilian patients, aged from 10 to 40 years, whose lesions had evolved for no longer than 2 years. Expression of receptors and the cell cycle was assessed by flow cytometry. We showed lower expression of the CXCR1 (35,7% ± 11,2) and CXCR2 (27,8%±11,3) in keloid fibroblasts, when compared with normal skin (44,1 ± 16,2 e 46,3 ± 27,1 respectively), but the difference was not significant for the CXCR1 receptor. This lower expression of IL-8 receptors in keloid fibroblasts could be due to the action of metalloproteinases, which regulate the surface protein enzymatically, or fibroblastic cytoskeleton conditions, which influence receptor internalization and recycling. The distribution assessment of cell cycle phases of fibroblasts cultivated from keloid scars and normal skin did not show significant difference in replication capacity and apoptosis. The keloid fibroblasts presented a significantly higher proportion of cells in the G2/M phase, suggesting higher rate of cell division. To confirm these results we studied the cell cycle of fibroblasts extracted ex vivo, now separated by central and peripheral portions of keloid and normal skin. The peripheral fibroblasts showed significant high cell proportions in phase S (22,9% ± 11,6), compared with the central portion (4,7% ± 2,9) and normal skin (6,8% ± 4,9), and higher cells in division phase G2/M (18,6% ± 12,0), compared with the central portion (35,6% ± 7,0) and normal skin (32,3% ± 6,9). The central portion showed higher proportion of apoptosis (7,0% ± 2,1), compared with the peripheral portion (4,9% ± 1,9) and normal skin (2,0% ± 0,86). These results suggest that the keloid peripheral cells could be responsible for the proliferation rate, justifying the expansive keloid growth at the borders of the keloid scar, in a similar fashion to tumor development and the central portion being responsible for fibrosis, with quiescent and apoptotic cells. These results suggest a differentiated modulation of cell reactions by signal pathways for programmed cellular proliferation or death. In this sense, the low expression of the IL-8 receptors CXCR1 and CXCR2 in keloid fibroblasts suggests a diminished capacity of IL-8 to promote accelerated healing. This low expression of IL-8 receptors in keloid fibroblasts could promote the dysregulation of the inflammatory response and thus attract more inflammatory cells to the site, producing different signals, such as a high production of the TGFβ cytokine. This dysregulation of the healing process, with changed cytokine and extracellular matrix expression, could be responsible for two different cell populations of fibroblasts, one proliferation at the periphery and the other fibrotic at the center of the lesion, with apoptotic and quiescent cells. Finally, we conclude that our results correspond to the histological and clinical changes of keloids that grow beyond the wound boundaries.

Análise do fator transcricional de meiose grauzone em Culex quinquefasciatus infectado por Wolbachia / Analysis of the transcriptional factor of meiosis grauzone in Culex quinquefasciatus infected by Wolbachia

Wolbachia pipientis é uma alfa-protobactéria intracelular obrigatória, endossimbionte de artrópodes e nematódeos que é herdada por via materna ao longo das gerações. A presença desta bactéria nos tecidos germinativos pode provocar nos hospedeiros alterações fenotípicas reprodutivas, como partenogênese, feminização genética de machos, morte de machos, incompatibilidade citoplasmática (IC) e alterações de fitness. Alguns mecanismos moleculares dessas alterações baseiam-se na modulação da expressão gênica do hospedeiro, ou seja, a bactéria pode suprimir ou estimular genes de forma a produzir ambiência favorável à manutenção da endossimbiose. Devido a esse potencial manipulador, Wolbachia tem sido testada como \"ferramenta\" para controle populacional de insetos vetores de patógenos. O mosquito Culex quinquefasciatus, naturalmente infectado por Wolbachia na região Neotropical, é um importante vetor de diversos patógenos que atingem humanos e animais. A presença da bactéria causa IC e altera o fitness no mosquito, mediante alterações temporais na ovogênese, fecundidade e fertilidade reprodutivas. Sabe-se que a presença da Wolbachia altera a expressão do gene grauzone e há fortes indícios de que esta expressão diferencial induza à IC em Cx. quinquefasciatus. Sabe-se também que este gene possui duas cópias parálogas em Cx. quinquefasciatus, porém estudos observaram a relevância de apenas um parálogo como importante regulador dos ciclos celulares da ovogênese e espermatogênese. No entanto, as bases genéticas dos fenótipos IC e \"fitness alterado\" do modelo Wolbachia-Cx. quinquefasciatus Neotropical ainda permanecem desconhecidas. Objetivamos inicialmente neste modelo quantificar e silenciar a expressão do gene grauzone (ambos parálogos) para suportar a hipótese pré-existente de que a superexpressão deste gene em mosquitos infectados por Wolbachia causa as alterações fenotípicas reprodutivas. Durante o desenvolvimento do projeto houve intercorrências que alteraram o rumo do trabalho: a necessidade de substituição da colônia experimental de mosquitos devido a baixas demográficas e à alta variabilidade intraespecífica do gene grauzone. Frente às intercorrências, formulamos como neo-objetivo a comparação filogenética entre as variantes do gene grauzone. Detectamos também variabilidade em um dos parálogos e concluímos que as cópias parálogas do gene encerram proteínas estruturalmente distintas e talvez funcionalmente distintas no tocante à alteração reprodutiva (IC) causada pela presença da Wolbachia. Foi possível observar que fêmeas infectadas apresentam amplificações de grauzone mais intensas quando comparadas com fêmeas não-infectadas no 4° dia de emergência, corroborando dados da literatura. Em conjunto, esses achados indicam que é promissora a continuidade do estudo do papel de grauzone na IC, mas demonstra também que este gene é mais complexo do que se imaginava, o que demandará maior esforço investigativo. A expectativa de uso futuro de Wolbachia como controlador biológico de mosquitos-vetores poderá se beneficiar de estudos como aqui proposto. / Wolbachia pipientis is an obligate intracellular alpha-protobacterium, endosymbiont of arthropods and nematodes, which is inherited through maternal route over generations. The presence of this bacterium in germinative tissues can cause reproductive phenotypic alterations in the hosts, such as parthenogenesis, genetic feminization of males, death of males, cytoplasmic incompatibility (CI) and fitness changes. Some molecular mechanisms of these alterations are based on the modulation of gene expression of the host, that is, the bacterium can suppress or stimulate genes in order to produce favorable environment for the maintenance of the endosymbiosis. Due to this potential manipulator, Wolbachia has been tested as a \"tool\" for population control of pathogen vector insects. The mosquito Culex quinquefasciatus, naturally infected by Wolbachia in the Neotropical region, is an important vector of several pathogens that affect humans and animals. The presence of the bacteria causes CI and changes the fitness in the mosquito, through temporal changes in ovogenesis, reproductive fertility and fertility. The presence of Wolbachia is known to alter the expression of the grauzone gene and there are strong indications that this differential expression induces the CI in Cx. quinquefasciatus. It is known that this gene occurs with two paralogs copies in Cx. quinquefasciatus, but studies have observed the relevance of only one paralog as important regulator of oogenesis and spermatogenesis cell cycles. However, the genetic basis of the phenotypes \"changed fitness\" and CI of Wolbachia-Cx quinquefasciatus Neotropical model remain unknown. We initially aimed at quantifying and knockdown of grauzone gene (both paralogs) to support the preexisting hypothesis that overexpression of this gene in Wolbachia-infected mosquitoes causes reproductive phenotypic changes. During the development of the project there were intercurrences that altered the course of work: the need to replace mosquito populations due to demographic lows and the high intraspecific variability of grauzone gene. In view of the intercurrences we formulated the neo-objective phylogenetic comparison between the variants of the grauzone gene. We also detected variability in one of the paralogs and concluded that the paralogs copies of the gene contain structurally distinct and perhaps functionally distinct proteins with respect to the reproductive alteration (CI) caused by the presence of Wolbachia. It was possible to observe that infected females show more intense grauzone amplifications when compared to uninfected females on the 4th day of emergence, corroborating data from the literature. Taken together, these findings indicate that the continuity of the study of the role of grauzone in CI is promising, but also demonstrates that this gene is more complex than previously thought, which will require more investigative effort. The expectation of future use of Wolbachia as a biological control of mosquito-vectors may benefit from studies as proposed here.

A crotoxina é a toxina mais abundante e ativa do veneno da cascavel brasileira Crotalus durissus terrificus. É composta por duas sub-unidades ligadas não covalentemente. A sub-unidade ácida é enzimaticamente inativa e não possui toxicidade, é também conhecida como crotapotina e potencializa a ação da fosfolipase A2 (sub-unidade básica). Alguns autores atribuem à crotoxina um efeito citotóxico e sugerem seu uso como um agente terapêutico contra tumores. Neste trabalho, foi investigada a citotoxidade da crotoxina e suas subunidades em células de melanoma murino e fibroblastos. Os ensaios de indicam que a crotoxina é tóxica para as células de melanoma promovendo a morte até em concentrações mais baixas enquanto os fibroblastos foram afetados somente em concentrações mais altas. Ensaios com as subunidades mostraram que a fosfolipase A2 foi mais tóxica para as células de melanoma que para os fibroblastos. / Crotoxin is the most abundant and active toxin from the venom of the Brazilian rattlesnake Crotalus durissus terrificus. It is composed of two subunits non covalently linked. One acidic with no enzymatic or toxic activity, known as crotapotin, which enhances the phospholipase A2 (basic subunit) action. Some authors have also attributed to this toxin a direct cytotoxic effect, and have suggested its use as a therapeutical agent against tumoral cells. In the present work, we investigated the cytotoxic effect of crotoxin and its subunitis on murine melanoma cells and fibroblasts. Our results indicate that crotoxin is highly toxic to the melanoma cells inducing cell death even at the lowest concentration while the fibroblast were only affected with the higher concentrations. The subunits assays showed that the phospholipase A2 had a higher toxicity on melanoma cells than on fibroblasts.

Aparato de importação de proteínas mitocondriais em Aspergillus fumigatus: caracterização fenotípica da deleção da menor subunidade do complexo TIM23 / Proteomic changes of the epithelial-mesenchymal transition (TMS) in cancer of ovary: involvement in cell cycle control and energy metabolism

O câncer de ovário (OvCa) se destaca dentre as neoplasias ginecológicas por ser um dos mais letais e de difícil diagnóstico. O OvCa ocorre devido ao acúmulo de alterações celulares progressivas promovidas por mutações no genoma de uma célula que, consequentemente, alteram as complexas vias de regulação celular que respondem a fatores internos, como reprogramação genética, ou externos, como a resposta a fatores de crescimento, que juntamente com outras alterações moleculares favorecem a progressão e a metástase. Uma importante etapa da cascata metastática é a transição epitélio-mesenquimal (EMT), um processo bem orquestrado que resulta na perda do fenótipo epitelial e aquisição do fenótipo mesenquimal pelas células tumorais, que adquirem um caráter mais invasivo e migratório, além de se tornarem mais resistentes às drogas. A desregulação de fatores de transcrição como ZEB1, TWIST e SNAI1, vias de sinalização, microRNAs e fatores de crescimento incluindo EGF, TGF? e HGF podem desencadear a EMT. Após a eficiente indução da EMT com EGF na linhagem epitelial de adenocarcinoma de ovário humano Caov-3, foi realizada a análise proteômica quantitativa detalhada, baseada na análise de frações subcelulares enriquecidas em proteínas de membrana, citosol e núcleo, obtidas por centrifugação diferencial e subsequente fracionamento de proteínas por SDS-PAGE, a fim de compreender mais profundamente os mecanismos moleculares modulados pela EMT no OvCa. A partir da análise dos dados coletados em um sistema de espectrometria de massas de alta resolução acoplados a cromatografia líquida (LCMS/MS) e com o auxílio da bioinformática foram identificadas redes de interação proteína-proteína diferencialmente expressas, relacionadas principalmente com a regulação do ciclo celular e do metabolismo. A indução da EMT por EGF resultou na ativação de importantes vias de sinalização, tais como PI3K/Akt/mTOR e Ras/MAPK Erk, além da parada do ciclo celular na fase G1 regulada pelo aumento dos níveis de p21Waf1/Cip1, independentemente de p53, e diminuição de proteínas checkpoint. Através da proteômica dirigida, o monitoramento de reações múltiplas (MRM) revelou que, após a indução da EMT por EGF, o metabolismo das células Caov-3 foi alterado de uma maneira bastante peculiar. O estudo proteômico descrito permitiu a correlação entre processo da EMT induzido por EGF com o controle translacional, a regulação do ciclo celular e a alteração do metabolismo energético. / Ovarian cancer (OvCa) stands out among gynecological malignancies for being one of the most lethal and difficult to diagnose. OvCa occurs due to the accumulation of progressive cell changes promoted by mutations in the cell genome which, consequently, alter the complex cellular regulation pathways that respond to internal factors, such as genetic reprogramming, or external, such as response to growth factors, which together with other molecular changes favor the progression and metastasis. An important step of the metastatic cascade is the epithelial-mesenchymal transition (EMT), a well-orchestrated process that results in the loss of epithelial phenotype and acquisition of mesenchymal phenotype by tumor cells that acquire a more invasive and migratory character, and become more resistant to drugs. Deregulation of transcription factors such as ZEB1, TWIST and SNAI1, signaling pathways, microRNAs and growth factors including EGF, TGF? and HGF can trigger EMT. After an efficient EMT induction by EGF in the epithelial cell line of human adenocarcinoma ovarian Caov-3, detailed quantitative proteomic analysis was performed based on analysis of subcellular fractions enriched in proteins from membrane, cytosol and nucleus, obtained by differential centrifugation and subsequent fractionation of proteins by SDS-PAGE, in order to understand deeply the molecular mechanisms modulated by EMT in OvCa. From the analysis of data collected in a highresolution mass spectrometry system coupled to liquid chromatography (LC-MS/MS) and with the aid of bioinformatics were identified protein-protein interaction networks differentially expressed, mainly related to regulation cell cycle and metabolism. EGF induced-EMT resulted in the activation of major signaling pathways such as PI3K/Akt/mTOR and Ras/MAPK Erk, in addition to G1 phase cell cycle arrest regulated by increased levels of p21Waf1/Cip1, regardless of p53, and reduction of checkpoint proteins. Through the targeted proteomics, multiple reaction monitoring (MRM) showed that after EGF induced-EMT, Caov-3 cells metabolism was changed in a very particular way. The proteomic study described allowed the correlation between EMT process induced by EGF with translational control, regulation of cell cycle and the change in the energy metabolism.

Determinação da relação entre proliferação celular, atividade metabólica e estágios da gestação e estabelecimento da ploidia e do ciclo celular em células de placentas bovinas / Relationship among cell proliferation, metabolic activity and days of pregnancy and determination of ploidy and cell cycle in bovine placenta

As regiões organizadoras de nucléolos (Nucleolar Organizer Regions - NORs) são regiões de cromatina levemente corada em volta da qual, no final da telófase, o nucléolo é novamente formado após a mitose. Um grupo peculiar de proteínas ácidas que têm alta afinidade por prata são localizadas nos mesmos locais que as NORs, o que confere as mesmas a propriedade de serem clara e rapidamente visualizadas por colorações que utilizam nitrato de prata. As NORs, quando coradas por prata são chamadas de AgNORs. O número de AgNORs está estritamente relacionado com a atividade transcricional do RNAr e com a agilidade e rapidez da proliferação celular. A homeostase celular utiliza-se de mecanismos através dos quais os tecidos de um organismo mantêm-se ou renovam-se, e para que isso ocorra, as células dispõe de dois programas genéticos principais: o ciclo celular e a apoptose. Considerando o crescimento da placenta e consequentemente das células trofoblásticas mono e binucleadas, esse trabalho teve dois principais objetivos: 1. determinar a relação quantitativa entre a proliferação celular e o estágio da gestação através da quantificação das AgNORs utilizadas como marcadores em placentas bovinas, evidenciando a intensa atividade proliferativa e metabólica das células binucleadas e estabelecendo o padrão de normalidade para gestações bovinas; e 2. determinar a ploidia das células do placentônio bovino e, através da análise dos estágios do ciclo celular, estabelecer a cinética celular envolvida na formação e diferenciação das células trofoblásticas binucleadas. Houve um aumento da quantidade de AgNORs nas células trofoblásticas mononucleadas a medida em que a gestação avançou, principalmente no terceiro trimestre de gestação. Isso indica um aumento na atividade proliferativa e/ou metabólica dessas células. O número de AgNORs expresso pela células binucleadas apenas aumentou do primeiro para o segundo trimestre, permanecendo, após esse estágio, praticamente constante. Isso nos leva a crer que a atividade das células binucleadas atinge seu ponto máximo no segundo trimestre e permanece com o seu metabolismo alto até o final da gestação. Além disso, esse número foi extremamente maior do que o observado nas células trofoblásticas mononucleadas, evidenciando a intensa atividade metabólica dessa célula. Na análise do ciclo celular, dois tipos celulares diferiam quanto ao seu tamanho (área) e quanto a sua granulosidade no placentônio, caracterizando diferenças quanto sua ploidia, sendo uma população diplóide e a outra tetraplóide. Para a fase G0-G1, a porcentagem obtida para o primeiro trimestre foi significativamente maior quanto comparada com o segundo e com o terceiro trimestre. Para a fase G2-M, encontramos um padrão inverso ao observado na fase G0-G1. Assim, podemos concluir que no primeiro trimestre de gestação as células tetraplóides estão formadas, mas não diferenciadas. A medida em que a gestação avança e a demanda metabólica fetal aumenta, essas células entram em processo de diferenciação para sua completa transformação em células capazes de produzir diversas substâncias e suprir as necessidades gestacionais maternas e fetais. / Nucleolar Organizer Regions (NORs) are weakly stained chromatin regions around which nucleoli reform after mitosis. A group of highly argyrophilic proteins are localized at the same sites as NORs, allowing NORs to be very clearly and rapidly visualized by silver nitrate staining procedures. They are called AgNORs. The number of AgNORs is strictly related to rRNA transcriptional activity and to the rapidity of cell proliferation. Cellular homeostasis uses two mechanisms to maintain or renew all tissues in organisms: the cell cycle and apoptosis. To comprehend the growth and the maturation of several tissues it is important to understand proliferation cellular kinetics and the cell cycle. The aim of this study was to evaluate the functional activity relation between the proliferative and metabolic capacity of trophoblastic mononucleate and binucleate cells from bovine placentomes at different stages of pregnancy. In addition, this study determined the ploidy and established the cellular kinetics involved in cell formation and differentiation through cell cycle analysis. The results demonstrated that: 1. The AgNOR number of mononucleate trophoblastic cells is related to their proliferative activity and stage of pregnancy; 2. The AgNOR number of binucleate trophoblastic cells is related to their level of metabolic activity and stage of pregnancy; 3. The AgNOR number of binucleate cell was just derived metabolic activity, and not proliferative activity, differing to mononucleate cells; 4. O câncer de ovário (OvCa) se destaca dentre as neoplasias ginecológicas por ser um dos mais letais e de difícil diagnóstico. O OvCa ocorre devido ao acúmulo de alterações celulares progressivas promovidas por mutações no genoma de uma célula que, consequentemente, alteram as complexas vias de regulação celular que respondem a fatores internos, como reprogramação genética, ou externos, como a resposta a fatores de crescimento, que juntamente com outras alterações moleculares favorecem a progressão e a metástase. Uma importante etapa da cascata metastática é a transição epitélio-mesenquimal (EMT), um processo bem orquestrado que resulta na perda do fenótipo epitelial e aquisição do fenótipo mesenquimal pelas células tumorais, que adquirem um caráter mais invasivo e migratório, além de se tornarem mais resistentes às drogas. A desregulação de fatores de transcrição como ZEB1, TWIST e SNAI1, vias de sinalização, microRNAs e fatores de crescimento incluindo EGF, TGF? e HGF podem desencadear a EMT. Após a eficiente indução da EMT com EGF na linhagem epitelial de adenocarcinoma de ovário humano Caov-3, foi realizada a análise proteômica quantitativa detalhada, baseada na análise de frações subcelulares enriquecidas em proteínas de membrana, citosol e núcleo, obtidas por centrifugação diferencial e subsequente fracionamento de proteínas por SDS-PAGE, a fim de compreender mais profundamente os mecanismos moleculares modulados pela EMT no OvCa. A partir da análise dos dados coletados em um sistema de espectrometria de massas de alta resolução acoplados a cromatografia líquida (LCMS/MS) e com o auxílio da bioinformática foram identificadas redes de interação proteína-proteína diferencialmente expressas, relacionadas principalmente com a regulação do ciclo celular e do metabolismo. A indução da EMT por EGF resultou na ativação de importantes vias de sinalização, tais como PI3K/Akt/mTOR e Ras/MAPK Erk, além da parada do ciclo celular na fase G1 regulada pelo aumento dos níveis de p21Waf1/Cip1, independentemente de p53, e diminuição de proteínas checkpoint. Através da proteômica dirigida, o monitoramento de reações múltiplas (MRM) revelou que, após a indução da EMT por EGF, o metabolismo das células Caov-3 foi alterado de uma maneira bastante peculiar. O estudo proteômico descrito permitiu a correlação entre processo da EMT induzido por EGF com o controle translacional, a regulação do ciclo celular e a alteração do metabolismo energético. / Ovarian cancer (OvCa) stands out among gynecological malignancies for being one of the most lethal and difficult to diagnose. OvCa occurs due to the accumulation of progressive cell changes promoted by mutations in the cell genome which, consequently, alter the complex cellular regulation pathways that respond to internal factors, such as genetic reprogramming, or external, such as response to growth factors, which together with other molecular changes favor the progression and metastasis. An important step of the metastatic cascade is the epithelial-mesenchymal transition (EMT), a well-orchestrated process that results in the loss of epithelial phenotype and acquisition of mesenchymal phenotype by tumor cells that acquire a more invasive and migratory character, and become more resistant to drugs. Deregulation of transcription factors such as ZEB1, TWIST and SNAI1, signaling pathways, microRNAs and growth factors including EGF, TGF? and HGF can trigger EMT. After an efficient EMT induction by EGF in the epithelial cell line of human adenocarcinoma ovarian Caov-3, detailed quantitative proteomic analysis was performed based on analysis of subcellular fractions enriched in proteins from membrane, cytosol and nucleus, obtained by differential centrifugation and subsequent fractionation of proteins by SDS-PAGE, in order to understand deeply the molecular mechanisms modulated by EMT in OvCa. From the analysis of data collected in a highresolution mass spectrometry system coupled to liquid chromatography (LC-MS/MS) and with the aid of bioinformatics were identified protein-protein interaction networks differentially expressed, mainly related to regulation cell cycle and metabolism. EGF induced-EMT resulted in the activation of major signaling pathways such as PI3K/Akt/mTOR and Ras/MAPK Erk, in addition to G1 phase cell cycle arrest regulated by increased levels of p21Waf1/Cip1, regardless of p53, and reduction of checkpoint proteins. Through the targeted proteomics, multiple reaction monitoring (MRM) showed that after EGF induced-EMT, Caov-3 cells metabolism was changed in a very particular way. The proteomic study described allowed the correlation between EMT process induced by EGF with translational control, regulation of cell cycle and the change in the energy metabolism.

Da mesma forma, em geral todas as observações sobre o ciclo nucleolar foram feitas a partir da análise de células mitóticas. Assim, o intuito do presente estudo foi analisar características reprodutivas e do ciclo nucleolar meiótico de espécies de morcegos Neotropicais. No epitélio seminífero, três tipos de espermatogônias foram reconhecidos: Ad, Ap e B. Essas após entrarem em meiose, passam por duas divisões celulares e formam espermátides haplóides, que sofrem diferenciação, compactando seu DNA, alongando seu núcleo e formando o flagelo e acrossomo, num processo que, em microscopia eletrônica de transmissão, pode ser dividido em doze fases. Observamos que o processo de espermiogênese ocorre de forma semelhante em Platyrrhinus lineatus, Molossus molossus e Myotis nigricans e que os espermatozóides de P. lineatus e M. molossus apresentam semelhanças, como o formato mais alongado da cabeça, a presença do perforatorium e espessamentos nas fibras densas externas 1, 5 e 6. Por outro lado, M. nigricans apresenta a ausência de perforatorium e espessamentos nas fibras axilares 1, 5, 6 e 9. Comparando os padrões reprodutivos de Artibeus planirostris e M. nigricans, observamos que, apesar de ambos serem espécies Neotropicais, eles diferem acentuadamente em sua reprodução. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: Although the process of reproduction has been studied in many mammals, relatively little attention has been given to this study in bats, this large and diverse order of organisms, which presents different reproductive strategies to adjust the animals to various environmental conditions and behaviors that they are inserted. Likewise, in general all studies on the nucleolar cycle were made based on the analysis of mitotic cells. Thus, the purpose of this study was to analyze the reproductive characteristics and the nucleolar meiotic cycle of Neotropical bat species. In the seminiferous epithelium, three types of spermatogonia were recognized: Ad, Ap and B. These, after enter in meiosis, undergo two cell divisions to form haploid spermatids, which undergo differentiation, compressing their DNA, stretching its nucleus and forming the acrosome and flagellum, a process that, in transmission electron microscopy, can be divided into twelve stages. We note that the process of spermiogenesis occured similarly in Platyrrhinus lineatus, Molossus molossus and Myotis nigricans and that the spermatozoon of P. lineatus and M. molossus have similarities, as the more elongated shape of the head, the presence of perforatorium and thickening in the outer dense fibers 1, 5 and 6. On the other hand, M. nigricans did not present the perforatorium and the thickening was presented in the fibers 1, 5, 6 and 9. Comparing the reproductive patterns of Artibeus planirostris and M. nigricans, we observed that, although both are Neotropical species, they differ markedly in their reproduction. Avaliação do ciclo celular de células hematopoiéticas da medula óssea de camundongos submetidos à desnutrição protéica / Evaluation of cell cycle by hematopoietic cells of malnourished mice

As diferentes formas da desnutrição acometem milhões de pessoas em todo mundo, afetando geralmente crianças, recém-nascidos abaixo do peso para sua idade gestacional, idosos e pacientes sob nutrição parenteral. A desnutrição protéica (DP) induz atrofia em órgãos hemopoéticos, induzindo pancitopenia de sangue periférico, modifica a resposta imune específica e inespecífica, e desta forma, a associação entre doença e DP é freqüente. Alguns estudos indicam que a desnutrição produz vários efeitos celulares como redução da capacidade de proliferação, em vários órgãos. Como conseqüência da constante e elevada demanda de proteína pelo tecido hemopoético, este pode apresentar alterações qualitativas e quantitativas em condições de desnutrição protéica. Neste trabalho avaliamos os efeitos da DP sobre o ciclo celular de células de medula óssea (MO) de camundongos. Camundongos Swiss machos, adultos, foram alimentados ou com ração contendo 2% de proteína (grupo desnutrido, D) ou com ração com 20% de proteína (grupo controle, C). A fonte protéica utilizada foi caseína. Após perda de 25% de peso corpóreo do grupo D, colheu-se sangue e células da MO e do baço. As células do sangue foram quantificadas bem como as do baço e da MO. As células da medula óssea foram posteriormente coradas com laranja de acridina (AO) e Iodeto de propídeo (PI), avaliando-se, por citometria de fluxo, respectivamente DNA/RNA e DNA. Estes mesmos procedimentos foram executados após 10 e 15 dias da administração, em ambos os grupos, do quimioterápico 5-Fluoracil (que leva as células a estado de repouso -G0). Os animais do grupo desnutrido, assim como aqueles após 10 e 15 dias da administração do 5-Fluoracil, apresentaram anemia, além de redução significativa de número de hemácias, hematócrito e reticulócitos, e alterações quantitativas nas linhagens medulares e do baço. Os dados da citometria de fluxo (expressos em % de células da população analisada) mostram que em DP, uma maior quantidade de células da MO encontra-se fora do ciclo celular (G0), e uma menor população de células nas fases proliferativas do ciclo celular, S/G2/M, em populações de todas linhagens medulares, exceto eritróide e linfóide B, e todo os estágios maturativos, assim como células Sca-1, células tronco murina. Esta maior população celular em G0 também é observada após o efeito do 5-Fluoracil, o que corrobora a suposição de incapacidade de retorno ao ciclo celular por parte das células de medula dos animais desnutridos. Estes resultados podem explicar, em parte, a hipoplasia medular e pancitopenia sanguínea. / The different forms of the malnutrition attacks millions of people in everybody generally, affecting children, just-born below of the weight for its gestacional age, aged and patient under parenteral nutrition. The protein malnutrition induces atrophy in haematopoietic organs, inducing pancitopenia of peripheral blood, modifies the specific and inespecífica immune reply, and of this form, the association between illness and DPE are frequent. Some studies indicate that the malnutrition produces some effect cellular as reduction of the proliferation capacity, in some organs. As consequence of the constant and raised protein demand for the bone marrow, that can present qualitative and quantitative alterations in conditions of protein malnutrition. In this work we evaluate the effect of the DP on the cellular cycle of cells of bone marrow of mice. Mice Swiss males, adults, had been fed or with ration I contend 2% of protein (unfed group, D) or with ration with 20% of protein (group has controlled, C). The used protein source was casein. After loss of 25% of corporeal weight of group D, harvested blood and cells of bone marrow and spleen. The cells of the blood had been quantified as well as the ones of spleen and bone marrow. The cells of the bone marrow later had been coradas with orange of acridine (AO) and propidium iodide of (PI), evaluating, for citometria of flow, respectively DNA/RNA and DNA. These same procedures had been executed after 10 and 15 days of the administration, in both the groups, of the 5-Fluoracil (that it takes the cells the state of rest - G0). The animals of the malnutrition group, as well as those after 10 and 15 days of the administration of the 5-Fluoracil, had presented anemia, beyond significant reduction of number of erythrocytis, hematocrit and reticulocytes, and quantitative alterations in the progenitors by bone marrow and of spleen. The data of the flow cytometric (express in % of cells of the analyzed population) show that in DP, a bigger amount of cells of outside meets bone marrow of the cellular cycle (G0), and a lesser population of cells in the proliferative phases of the cellular cycle, S/G2/M, in populations of all progenitors by bone marrow, except erythroids and lymphoids B, and all the maturative periods of training, as well as cells Sca-1, stem cells murina. This bigger cellular population in G0 also is observed after the effect of the 5-Fluoracil, what it corroborates the assumption of incapacity of return to the cellular cycle on the part of the cells of marrow of the malnutrition animals. These results can explain, in part, the hipoplasia to medular and sanguineous pancitopenia.

Efeitos anti-neoplásicos da raiz de Pfaffia paniculata (Ginseng brasileiro) no modelo de hepatocarcinogênese murina e em cultura de células de hepatocarcinoma humano / Antineoplastic effects of the roots of Pfaffia paniculata (Brazilian ginseng) in a murine hepatocarcinogenesis model and in human hepatocarcinoma cells

A raiz pulverizada de Pfaffia paniculata e seu extrato butanólico apresentam propriedades antineoplásicas, quimiopreventivas e antiangiogênicas, onde muitos indícios conferem às saponinas triterpenóides presentes nestas raízes as propriedades antitumorais observadas. Sabe-se que saponinas de diversos tipos de plantas possuem capacidade de interferir diretamente no ciclo celular de células tumorais. Assim, considerando os efeitos inibitórios das raízes e extratos de P. paniculata sobre a proliferação celular, o objetivo deste trabalho foi compreender os mecanismos envolvidos na ação quimiopreventiva desta raiz, tanto na fase de iniciação da carcinogênese hepática, quanto sobre células tumorais já estabelecidas. Inicialmente, foram avaliados os efeitos de diferentes concentrações da raiz em pó de P. paniculata adicionada à ração em camundongos submetidos ao modelo de hepatocarcinogênese, pela análise da proliferação celular, indução da apoptose e a comunicação intercelular hepática. Seqüencialmente, foram avaliados os efeitos das frações purificadas do extrato butanólico destas raízes sobre linhagem de células de carcinoma hepatocelular humano. Neste experimento foram analisadas a influência do tratamento sobre a viabilidade celular, fases e proteínas do ciclo celular, proliferação e presença de morte celular. No modelo de hepatocarcinogênese o tratamento com a raiz reduziu a proliferação celular, aumentou a apoptose e desencadeou processo inflamatório crônico em intensidades dependentes da concentração testada e não afetou a comunicação intercelular. Estes resultados indicam que os efeitos quimiopreventivos da P. Paniculata são decorrentes do controle da proliferação celular e apoptose, e são diretamente influenciados pela concentração da raiz. No experimento in vitro o tratamento reduziu a concentração das células vivas sem aumentar a concentração de células mortas, diminuiu a porcentagem de células na fase G2 do ciclo celular, reduziu a expressão dos genes das proteínas ciclinas D1, E e CDK6 e aumentou a expressão de p27, e não induziu apoptose. Estes resultados mostraram que a redução na concentração de células observadas após o tratamento com a fração do extrato butanólico de P. paniculata não foi decorrente de indução de apoptose. O tratamento parou o ciclo celular das células tumorais HepG2 em G1, inibindo proteínas importantes para a progressão do ciclo e estimulando a expressão de p27 um conhecido gene inibidor de CDKs. Os efeitos antiproliferativos observados no experimento in vivo em camundongos, reproduziram-se in vitro em células tumorais humanas, o que pode indicar que as propriedades antineoplásicas anteriormente observadas não são espécie específica. Em linhas gerais, as raízes e/ou as frações do extrato butanólico obtido a partir das raízes de P. paniculata apresentam propriedades antineoplásicas por inibir a proliferação celular e induzir apoptose in vivo e por parar o ciclo celular in vitro. Estes resultados consagram as propriedades antineoplásicas de Pfaffia paniculata e motivam o desenvolvimento de mais estudos sobre as suas potencialidades. / The powdered roots of Pfaffia paniculata and their butanolic extract present antineoplastic, chemopreventive and antiangiogenic properties, and many evidences suggest that the triterpenoid saponins are the responsible for these properties. It is well known that saponins from several types of plants have the capacity to directly interfere on the tumor cell cycle. Therefore, considering the inhibitory effects of the roots and extracts of P. paniculata on cell proliferation, the aim of this study was to search for the mechanisms involved in the chemopreventive effects of this root, both in the initiation phase of the hepatocarcinogenesis and on tumor cell lineage. Initially, the effects of different concentrations of the powdered root of P. paniculata added to the mouse food were evaluated in mice submitted to hepatocarcinogenesis model. Cell proliferation, induction of apoptosis, and hepatic intercellular communication were evaluated. The effects of the purified fractions of the butanolic extract of these roots were then evaluated in human hepatocarcinoma cells. In this experiment, the influence of the treatment on cell viability, phases and proteins of cell cycle, cell proliferation and cell death were evaluated. In the hepatocarcinogenesis model, the treatment with the root decreased cell proliferation, increased apoptosis and induced a chronic inflammatory process dependent on the concentration tested, and did not affect cell communication. These results indicate that the chemopreventive effects of P. Paniculata are apparently dependent on the control of cell proliferation and apoptosis and are directly influenced by the concentration of the root. In the in vitro treatment, it has been observed a reduction in the concentration of live cells without increasing the concentration of dead cells, decreased the level of G2 phase cells, reduced the expression of proteins cyclins D1, E and CDK6, increased the expression of p27, and did not induce apoptosis. These results showed that the reduction in the concentration of cells after the treatment with the butanolic extract of P. paniculata was not due to induction of apoptosis. The treatment inhibited the progression of the cell cycle of HepG2 cells in phase G1, by the inhibition of the expression of proteins that are important to the progression of the cycle, and stimulating the expression of p27, a known inhibitor of CDKs. The antiproliferative effects observed in the in vivo experiments were repeated in the in vitro study with human tumor cells. This may indicate that the antineoplastic properties previously observed are not species-specific. In conclusion, the roots and/or butanolic extract obtained from P. paniculata present antineoplastic properties due to inhibition of cell proliferation and induction of apoptosis in vivo, and due to the cell cycle arrest in vitro. These results reinforce the antineoplastic properties of Pfaffia paniculata and motivate the development of more studies focusing on its antineoplastic potentials.

Uma dieta adequada pode contribuir para a prevenção de diversas doenças crônicas, dentre elas, o câncer. Estudos epidemiológicos apontam que as populações de países asiáticos sofrem menos com estas doenças, e o alto consumo de soja (e suas isoflavonas daidzeína e genisteína) está associado a este fato. As isoflavonas da soja daidzeína e genisteína possuem grande potencial de prevenção do câncer, uma vez que muitos estudos já demonstraram que elas interferem em diversos estágios do processo de carcinogênese. A modulação da proliferação e morte celular é um dos efeitos quimiopreventivos das isoflavonas mais investigados, porém a maioria dos estudos utiliza-se de concentrações fisiologicamente irrelevantes, ou seja, concentrações que não são encontradas no organismo humano. Portanto, o objetivo do nosso estudo foi analisar os efeitos da máxima concentração fisiologicamente relevante dessas isoflavonas em células humanas in vitro. Três linhagens celulares foram utilizadas: MCF-7 (tumoral de mama), HB4a (não tumoral de mama) e OVCAR-3 (tumoral de ovário). O efeito citotóxico, a expressão gênica (CASP-3, CASP-7, BAX, BCL-xL e COX-2) e a indução de apoptose foram investigadas nas três linhagens celulares. Além disso, foi estudado o efeito das isoflavonas sobre o ciclo celular das linhagens MCF-7 e HB4a. Somente uma concentração alta de genisteína (100 μM) após 72h de tratamento induziu citotoxicidade na linhagem OVCAR-3. Ambas as isoflavonas em concentração de 10 μM não induziram apoptose nesta linhagem, e a expressão dos genes analisados não mudou significativamente frente ao tratamento com daidzeína ou genisteína. O tratamento das... / Nowadays, mainly because of Nutrigenomics advances, it is known that diet has a major influence on health and welfare of people. A proper diet can contribute to the prevention of several chronic diseases, including cancer. Epidemiological studies indicate that Asian populations suffer less from these diseases, and high intake of soy (and its isoflavones daidzein and genistein) is associated with this fact. The soy isoflavones daidzein and genistein have great potential for cancer prevention, since many studies have shown that they interfere with several stages of carcinogenesis. Modulation of cellular proliferation and death is one of the most investigated chemopreventive effects of isoflavones, but in most of the studies, physiological irrelevant concentrations are used, i.e., concentrations that are not found in human body. The objective of our study was to analyze the effects of maximal physiological relevant concentration of these isoflavones in in vitro human cell lines. Three cell lineages were used: MCF-7 (from breast carcinoma), HB4a (breast non-tumoral cell) and OVCAR-3 (from ovarian carcinoma). Cytotoxic effect, gene expression (CASP-3, CASP-7, BAX, BCL-xL and COX-2) and induction of apoptosis were investigated in all cell lines. In addition, it was studied the effect of the isoflavones on the cell cycle of MCF-7 and HB4a. Only the highest concentration of genistein (100 μM) was cytotoxic to OVCAR-3 after 72 hours of treatment of the cells. Both isoflavones at 10 μM did not induce apoptosis in this cell line and expression of the studied genes did not change in front of daidzein or genistein treatment. Daidzein treatment of HB4a cells was not cytotoxic in none of the experimental conditions. Daidzein 10 or 25 μM also was not able to induce apoptosis or to arrest cell cycle of nontumoral HB4a.
