A patologia desta infecção é caracterizada pela formação de granulomas em torno dos ovos liberados pelos vermes adultos, no interior do sistema portahepático. Sabe-se que, os pacientes portadores da infecção crônica apresentam uma modulação da capacidade proliferativa em resposta ao antígeno solúvel de ovos de S. mansoni (SEA) e, este estado parece ser benéfico para o hospedeiro e tem sido considerada fundamental para o equilíbrio hospedeiro-parasito. Em contrapartida, indivíduos não infectados residentes em área endêmica para esquistossomose apresentam uma elevada reatividade celular em resposta a estes antígenos. Neste trabalho, comparamos a influência do SEA na progressão do ciclo celular de linfócitos T do sangue periférico de pacientes portadores de infecção crônica pelo S. mansoni (XTO), de indivíduos não infectados residentes em área endêmica (NEG) e de indivíduos não infectados, porém não residentes em área endêmica, denominados por DS. Nossos resultados mostraram que pacientes XTO apresentam uma modulação da resposta ao SEA e, sugerem que esta imunossupressão pode ser devido a participação das células T reguladoras, que secretam IL- 10 em resposta ao SEA. A citocina IL-10 pode atuar inibindo a ativação celular de linfócitos T CD4+ e CD8+, por meio do bloqueio destas células nas fases G0/G1 do ciclo celular, promovendo a anergia nestas células. Este bloqueio da progressão do ciclo celular se dá por inibição da expressão da ciclina D1,2,3 nos linfócitos T. Por outro lado, nas culturas celulares dos indivíduos NEG estimuladas com SEA, foram detectados níveis elevados de TNF-α, ativação celular dos linfócitos T CD4+ e CD8+ e um aumento da expressão da ciclina D1,2,3, promovendo a progressão através do ciclo celular em resposta ao SEA. Em áreas endêmicas, existe uma alta prevalência de ancilostomídeos co-infectando pacientes portadores de S. mansoni. Neste contexto, avaliamos também a influência da presença da co-infecção pelo N. americanus na resposta imunológica ao SEA de pacientes infectados por S. mansoni. Nossos resultados mostraram que tanto as PBMC de pacientes mono-infectados (MONO) quanto as de co-infectados (CO) apresentaram anergia celular em resposta ao SEA, sugerindo que a presença da infecção por N. americanus não influencia na resposta imune ao SEA. / S. mansoni infection is frequent in regions where factors such as poor sanitary conditions occur and may lead to high prevalence of the disease mainly in individuals of the age groups of 10 to 20 years. The pathology caused by the infection is characterized by granulomatous reactions around the eggs eliminated by the parasites that are trapped in the tissues and the hepatic-portal system. It is well known that during the chronic phase individuals that harbor the infection modulate their cellular immune response to soluble egg antigens (SEA) and this immune reactivity is highly accepted to be beneficial to the host and fundamental for the maintenance of equilibrium of the host-parasite interaction. In contrast to infected individual, those not infected but living in an endemic area for the disease show high cellular reactivity to the same antigens. In this thesis, we evaluate the effect of that the response to SEA has on the cell cycle progression of peripheral blood T lymphocyte from patients in the chronic phase of the disease (XTO), non-infected individuals living in endemic areas (NEG) and of non-infected individuals that do not reside in endemic areas (DS). Our results show that cells from XTO have a modulated response to SEA and suggest that this immunosuppression may be due to regulatory T cells that secret IL-10 in response to SEA. IL-10 may act via the inhibition of the activation of CD4+ and CD8+ T lymphocytes by blocking these cells in the G0/G1 phase of the cell cycle promoting anergy of these cells. The blockage of cell cycle progression occurs via the inhibition of the expression of cyclin D1,2,3 by the T lymphocytes. On the other hand, cells from NEG stimulated in vitro with SEA, high levels of TNF-α secretion was detected, together with the activation of both CD4+ and CD8+ cells and increase on the expression of cyclin D1,2,3 with consequent progression of these cell through the cell cycle. In endemic areas, there is a high prevalence of hookworms co-infecting individuals with S. mansoni. We therefore, evaluated the putative influence of the co-infection with N. americanus on the immune response of S. mansoni infected patients to in vitro SEA stimulation. Our results show that peripheral blood mononuclear cells (PBMC) from mono-infected patients (MONO) as well as of co-infected (CO) were anergic to SEA, suggesting that the presence of the infection by N. americanus does not influence the response to SEA.

Avaliação da resposta a longo prazo de células de carcinoma colorretal à 5-fluoruracila e oxaliplatina mimetizando o perfil de tratamento clínico

O carcinoma colorretal (CCR) está entre os três tipos mais frequentes e mortíferos de câncer do mundo. O tratamento de primeira escolha depende do estágio tumoral, sendo que o regime quimioterápico mais utilizado é o FOLFOX (5-fluoruracila + oxaliplatina + leucovorina). Apesar do amplo uso, o mecanismo de ação deste protocolo, especialmente considerando o período entre ciclos de tratamento, bem como os mecanismos envolvidos na frequente resistência de células de CCR ao tratamento são pouco conhecidos. Aqui nós avaliamos a resposta de células de CCR expostas ao tratamento com 5-fluoruracila (5FU) e oxaliplatina (OXA) em perfil semelhante ao tratamento clínico, nas linhagens de CCR humano HCT116 e HT29, em prol de entender os mecanismos de ação e de resistência ao tratamento. Para isso, as células foram tratadas com os quimioterápicos de maneira isolada ou combinados por 2 dias, seguido do replaqueamento em meio livre de droga por 15 dias, mimetizando o regime de tratamento clínico. Foram realizadas análises da proliferação celular e ciclo celular, além de análises dos mecanismos celulares que vêm sendo mostrados como envolvidos na sensibilidade e resistência de células tumorais a compostos citotóxicos, tais como apoptose, senescência e autofagia. Não houve efeito aditivo citotóxico para a combinação de 5FU e OXA a curto prazo (48h), porém, a longo prazo houve um efeito aditivo importante para o co-tratamento quando comparado aos tratamentos isolados. Este efeito aditivo foi mediado pela indução de apoptose (principalmente pela 5FU) e senescência (principalmente pela OXA). Apesar deste efeito, as células de todos os tratamentos retomaram o crescimento bem como mantiveram capacidade clonogênica a partir do dia 7 após o tratamento. Quando nós avaliamos a autofagia nas células sobreviventes ao tratamento, encontramos um aumento transitório, com pico entre os dias 3 e 7, sendo que um perfil semelhante foi observado para o aumento de ROS, com pico entre os dias 5 e 7. A inibição racional da autofagia no período de ativação máxima do mecanismo reduziu de maneira intensa o número de células e a clonogenicidade das células sobreviventes. Dessa forma, a inibição da autofagia em combinação com 5FU e OXA pode potencializar o efeito do tratamento e reduzir a resistência das células de CCR ao tratamento. / Colorectal carcinoma (CRC) is among the three most frequent and deadly types of cancer in the world. The first-choice treatment depends on the tumor stage, and the most commonly used chemotherapy regimen is FOLFOX (5-fluorouracil + oxaliplatin + leucovorin). Despite the wide use, the mechanism of action of this protocol, mainly considering the interval between two cycles of chemotherapy, as well as the mechanisms involved in the frequent resistance of CRC cells to treatment are poorly understood. In this project, we evaluated the response of HCT116 and HT29 human CRC cells exposed to 5-fluoruracil (5FU) and oxaliplatin (OXA) in a profile similar to clinical treatment. Cells were treated with the chemotherapeutics alone or in combination for 2 days, followed by the replating in drug-free medium for 15 days, mimicking the clinical treatment regimen. Analysis of cell proliferation and cell cycle were performed, as well as the analysis of the cellular mechanisms that have been shown as being involved in the sensitivity and resistance of tumor cells to cytotoxic compounds, such asapoptosis, senescence and autophagy. There was no cytotoxic additive effect for the combination of 5FU and OXA in the short-time (48h), but in the long term it was observed an important additive effect for co-treatment when compared to the isolated treatments. This additive effect was mediated by the induction of apoptosis (mainly by 5FU) and senescence (mainly by OXA). Despite this effect, cells from all treatments resumed growth as well as maintained the clonogenic capacity from day 7 after treatment. When we evaluated autophagy in treatment-surviving cells, we found a transient increase, with a peak between days 4 and 6; a similar transient increase was observed for reactive species, peaking between days 6 and 8. The rational inhibition of autophagy in the period of maximal activation of the mechanism greatly reduced the number of cells and the clonogenicity of the surviving cells. Thus, the rational inhibition of autophagy in combination with 5FU and OXA may potentiate the effect of the treatment and reduce the resistance of CRC cells to the treatment.

Efeitos do resveratrol sobre as células estreladas hepáticas

As células estreladas hepáticas (HSC) são pericitos específicos do fígado, são células armazenadoras de gordura e produzem componentes de matriz extracelular. As HSCs ao serem ativadas, proliferam, perdem as gotas lipídicas e aumentam a secreção de matriz extracelular. O controle da modulação fenotípica e da proliferação das HSCs é de suma importância para entendermos a fibrose hepática. Esta pesquisa foi realizada com a linhagem celular GRX (representativa das HSCs murinas) obtida a partir de reações fibrogranulomatosas inflamatórias. Estudos anteriores, em nosso laboratório, pesquisaram a ação de diferentes drogas sobre a modulação destas células, tais como: retinol, pentoxifilina, indometacina e insulina. Tem sido relatado que o resveratrol (3,4,5 trihidroxiestilbeno) (RSV) tem ação antioxidante, antiinflamatória, antiproliferativa bem como tem capacidade de ativar ou inibir a transcrição de enzimas envolvidas na regulação de vários eventos celulares, por exemplo metabolismo de lipídeos. Por isso, procuramos utilizar o RSV, um polifenol natural presente na casca das uvas e conseqüentemente em grande quantidade de vinhos tintos, neste modelo celular. As células GRX foram cultivadas (12, 24 e 120 h) em meio DMEM com 5% SFB acrescido ou não de RSV (100 nM, 1µM ou 100 µM) e retinol (RHO) (5µM). Nossos estudos demonstraram que tratamento por 120 h com RSV (100nM e 1µM) inibiu a proliferação das células GRX diminuindo em 35% o número de células. Analisando o efeito de 1µM de RSV sobre o ciclo celular e a apoptose, verificamos que após 120 h de tratamento houve um aumento das células na fase S e Sub-G1. Constatou-se condensação e fragmentação nuclear, sugerindo um possível efeito pró-apoptótico do RSV sobre a célula GRX. Observou-se que o tratamento com RSV (100 nM) por 5 dias não induziu a formação de gotas lipídicas nas células GRX. Porém ao acrescentar RSV+RHO, constatou-se que o RSV não interferiu na síntese e acúmulo de lipídios, provocado pelo RHO (5µM). Propondo que a lipogênese neste modelo de tratamento possa estar associada com a ação da sirtuina (deacetilase NAD-dependente) e do PPARy (receptores ativados por proliferadores de peroxissomos), determinou-se a expressão do mRNA destas duas proteínas. Observou-se que o RSV (100 nM) aumenta a expressão do mRNA da sirtuina 1 (SIRT1) e diminui a expressão de PPARy. Porém, o tratamento com RSV+RHO provocou uma redução na expressão da SIRT1, alterando a relação PPARy/SIRT1, promovendo a síntese de lipídios. Nossos dados mostraram que após 24h de tratamento, o RSV não alterou a incorporação de acetato [C14] em triacilglicerídios (TG), enquanto que, o RHO ou RSV+RHO aumentaram a incorporação do marcador nestes lipídios. O depósito de TG permaneceu constante, porém a síntese diminuiu com o tempo de tratamento. Acredita-se que estes compostos atuam por mecanismos moleculares diferentes modulando o metabolismo de lipídios. Sugerimos, de acordo dados da literatura, que o RHO atua via receptor nuclear LXR formando um heterodímero com PPARy, favorecendo a lipogênese. Por outro lado, o RSV ativa a SIRT1, que reprime o PGC1α (coativador do PPARy). Logo, a inibição do PPARy pelo RSV inibe consequentemente a lipogênese. Concluímos que o resveratrol apresenta um efeito anti-proliferativo, pró-apoptótico sem induzir o fenótipo lipocítico nas células GRX. Desta forma, se mantém em equilíbrio, no espaço de Disse, as células mesenquimais quiescentes e ativadas, contribuindo para restabelecer a homeostase hepática. / The hepatic stellate cells (HSC) are liver-specific pericytes, as well as fat-storing cells and they are also responsible for the extracellular matrix components production. When activated, HSCs lose lipid droplets and increase extracellular matrix secretion. The phenotypical modulation and the HSCs proliferation control have paramount importance to understand the hepatic fibrosis. This research has been carried out with the GRX cell line (HSCs murine representative) which was obtained from inflammatory fibrogranulomatous reactions. Previous studies in our laboratory investigated the action of different drugs on the modulation of these cells, such as: retinol, pentoxifilin, indomethacin and insulin. Resveratrol (RSV) has been referred to as having antioxidant, antiinflammatory and antiproliferative action (3,4,5 trihidroxiestilbeno), as well as the capacity to activate or inhibit the transcription of enzymes involved in the regulation of several cell events, as, for example, lipid metabolism Therefore, we used RSV, which is a natural polyphenol found in the grapes skin and, consequently, in large amounts of red wine, in this cell model. The GRX cells were cultivated (12, 24 and 120 h) in medium DMEM with 5% SFB added RSV or not (100 nM, 1µM or 100 µM) and retinol (RHO) (5µM). Our studies demonstrated that a 120 h treatment with RSV (beginning with 100 nM) inhibited GRX cells proliferation, thus decreasing the cell number in 35%. Assessing the effect of 1µM RSV on the cell cycle and the apoptosis, we found that, after this treatment period, there was an increase of cells in phases S and Sub-G1. Nuclear condensation and fragmentation were observed, which suggested a probable pro-apoptotic RSV effect on the GRX cell. The RSV (100 nM) 120 h treatment did not induce lipid droplets formation in the GRX cells. However, RSV did not interfere in the synthesis or accumulation of lipids caused by RHO (5µM), when RSV+RHO were added. Considering that the lipogenesis in this treatment model might be associated with sirtuin action (deacetilase NAD-dependent) and PPARy (receptors activated by peroxisome proliferators), the mRNA expression of these two proteins was determined. RSV (100 nM) proved to increase sirtuin mRNA expression 1 (SIRT1) and decrease PPARy.expression. However, the treatment with the RSV+RHO caused a reduction in the SIRT1 expression thus alterating the relation PPARy/SIRT1, which promoted the lypid synthesis. Our data showed that after the 24-hour treatment RSV did not change acetate incorporation [C14] in triacylglicerides (TG), whilst RHO or RSV+RHO increased the marker incorporation in these lipids. The TG deposit remained constant, but the synthesis diminished during the treatment period. It appears that these compounds act through different molecular mechanisms when modulating lipids metabolism. According to literature data, we suggest that RHO acts via LXR nuclear receptor, thus forming an heterodímer with PPARy, which favors lypogenesis. On the other hand, RSV activates SIRT1, which inhibits PGC1α (PPARy coactivator). Therefore, the PPARy inhibition by RSV consequently inhibits lypogenesis. We concluded that RSV presents pro-apoptotic, anti-proliferative effect, without inducing lypocytic phenotype in the GRX cells. By this means the mesenquimal activated quiescent cells keep their balance at the Disse space, thus contributing with the hepatic homeostasis restoration.

Perfil de atividade da proteína tirosina fosfatase de baixa massa molecular relativa e da fosfatase ácida resistente ao tartarato em osteoblastos humanos durante o ciclo e diferenciação celular / Low molecular weight protein tyrosine phosphatase and tartrate resistant acide phosphatase activity in human osteoblasts during cell cycle and differentiation

O objetivo deste trabalho foi determinar o perfil de atividade enzimática da proteína tirosina fosfatase de baixa massa molecular relativa (PTPBMr) e da fosfatase ácida resistente ao tartarato (TRAP) em osteoblastos humanos durante o ciclo e a diferenciação celular, correlacionando com os níveis de estresse oxidativo intracelulares. A atividade enzimática das fosfatases foi determinada nos períodos de 6, 18, 24, 48 e 72 horas (ciclo celular) e 7, 14, 21, 28 e 35 dias (diferenciação) utilizando o p-nitrofenilfosfato e na presença do inibidor específico phidroximercuribenzoato (pHMB) para a PTP-BMr, e na presença de tartarato e pHMB para a TRAP. A caracterização da diferenciação celular foi determinada medindo o nível de atividade da fosfatase alcalina e a coloração de Von Kossa. O estresse oxidativo foi determinado através da quantificação da glutationa reduzida e oxidada através dos ensaios de cromatografia líquida de alta performance eletroquímica e DTNB. Durante o ciclo celular a atividade específica (AE) da fosfatases foi fortemente diminuída, especificamente da TRAP e PTP-BMr, sendo praticamente zero após 18 horas da adição de soro, sugerindo que a diminuição na atividade destas enzimas seja necessária para a entrada na fase S. Durante a diferenciação celular observou-se um aumento progressivo da expressão de FALC, TRAP e PTP-BMr nos períodos de 7 e 14 dias, sendo máxima no 21o dia, declinando a seguir. Durante a proliferação, os estados mais reduzidos foram observados nos períodos de 18 e 48 horas e durante a diferenciação o estado mais reduzido foi observado no 21o dia. Desta forma, o presente trabalho mostra que as células da linhagem hFOB 1.19 representam um adequado modelo de estudo para análise da participação de fosfatase no ciclo e diferenciação celular, bem como que as atividades da PTP-BMr e TRAP são claramente moduladas durante o ciclo celular e a diferenciação de osteoblastos humanos, esta última dependente de adequado nível de glutationa reduzida. / Low molecular weight protein tyrosine phosphatase (LMW-PTP) and tartrate resistant acid phosphatase (TRAP) activity were determined in hFOB 1.19 human osteoblasts cell line during cell proliferation and differentiation. LMW-PTP and TRAP enzymatic activity were determined at 6, 18, 24, 36, 48 and 72 hours after fetal calf serum stimulation of subconfluent cultures and 7, 14, 21, 28 and 35 days after cell confluence and differentiation. The LMW-PTP and TRAP activity were measured using p-nitrophenylphosphate as substrate. The osteogenic potential of hFOB 1.19 cells was studied by measuring alkaline phosphatase activity, and mineralized nodule formation by Von Kossa staining. The oxitative stress was determined by HPLC and DNTB assays. During cell cycle progression, LMW-PTP and TRAP activities were strongly reduced, being almost undetectable after 18h of serum stimulation, while H3-thymidine incorporation progressively increased, suggesting that the decrease in the LMW-PTP and TRAP activities were necessary for entry into the S phase. During osteoblastic differentiation, the activity of LMW-PTP and alkaline phosphatase progressively increased until the 21th day, decreasing thereafter. In conclusion, this work demonstrates that hFOB 1.19 cells constitute a suitable model system for the study of the role played by LMW-PTP and TRAP in cell cycle progression and cell differentiation, and that LMW-PTP and TRAP activities are clearly modulated during osteoblastic proliferation and differentiation in vitro. The activities of these phosphatases during cell differentiation depended on the correct levels of reduced glutathione.

Efeitos do resveratrol sobre as células estreladas hepáticas

As células estreladas hepáticas (HSC) são pericitos específicos do fígado, são células armazenadoras de gordura e produzem componentes de matriz extracelular. As HSCs ao serem ativadas, proliferam, perdem as gotas lipídicas e aumentam a secreção de matriz extracelular. O controle da modulação fenotípica e da proliferação das HSCs é de suma importância para entendermos a fibrose hepática. Esta pesquisa foi realizada com a linhagem celular GRX (representativa das HSCs murinas) obtida a partir de reações fibrogranulomatosas inflamatórias. Estudos anteriores, em nosso laboratório, pesquisaram a ação de diferentes drogas sobre a modulação destas células, tais como: retinol, pentoxifilina, indometacina e insulina. Tem sido relatado que o resveratrol (3,4,5 trihidroxiestilbeno) (RSV) tem ação antioxidante, antiinflamatória, antiproliferativa bem como tem capacidade de ativar ou inibir a transcrição de enzimas envolvidas na regulação de vários eventos celulares, por exemplo metabolismo de lipídeos. Por isso, procuramos utilizar o RSV, um polifenol natural presente na casca das uvas e conseqüentemente em grande quantidade de vinhos tintos, neste modelo celular. As células GRX foram cultivadas (12, 24 e 120 h) em meio DMEM com 5% SFB acrescido ou não de RSV (100 nM, 1µM ou 100 µM) e retinol (RHO) (5µM). Nossos estudos demonstraram que tratamento por 120 h com RSV (100nM e 1µM) inibiu a proliferação das células GRX diminuindo em 35% o número de células. Analisando o efeito de 1µM de RSV sobre o ciclo celular e a apoptose, verificamos que após 120 h de tratamento houve um aumento das células na fase S e Sub-G1. Constatou-se condensação e fragmentação nuclear, sugerindo um possível efeito pró-apoptótico do RSV sobre a célula GRX. Observou-se que o tratamento com RSV (100 nM) por 5 dias não induziu a formação de gotas lipídicas nas células GRX. Porém ao acrescentar RSV+RHO, constatou-se que o RSV não interferiu na síntese e acúmulo de lipídios, provocado pelo RHO (5µM). Propondo que a lipogênese neste modelo de tratamento possa estar associada com a ação da sirtuina (deacetilase NAD-dependente) e do PPARy (receptores ativados por proliferadores de peroxissomos), determinou-se a expressão do mRNA destas duas proteínas. Observou-se que o RSV (100 nM) aumenta a expressão do mRNA da sirtuina 1 (SIRT1) e diminui a expressão de PPARy. Porém, o tratamento com RSV+RHO provocou uma redução na expressão da SIRT1, alterando a relação PPARy/SIRT1, promovendo a síntese de lipídios. Nossos dados mostraram que após 24h de tratamento, o RSV não alterou a incorporação de acetato [C14] em triacilglicerídios (TG), enquanto que, o RHO ou RSV+RHO aumentaram a incorporação do marcador nestes lipídios. O depósito de TG permaneceu constante, porém a síntese diminuiu com o tempo de tratamento. Acredita-se que estes compostos atuam por mecanismos moleculares diferentes modulando o metabolismo de lipídios. Sugerimos, de acordo dados da literatura, que o RHO atua via receptor nuclear LXR formando um heterodímero com PPARy, favorecendo a lipogênese. Por outro lado, o RSV ativa a SIRT1, que reprime o PGC1α (coativador do PPARy). Logo, a inibição do PPARy pelo RSV inibe consequentemente a lipogênese. Concluímos que o resveratrol apresenta um efeito anti-proliferativo, pró-apoptótico sem induzir o fenótipo lipocítico nas células GRX. Desta forma, se mantém em equilíbrio, no espaço de Disse, as células mesenquimais quiescentes e ativadas, contribuindo para restabelecer a homeostase hepática. / The hepatic stellate cells (HSC) are liver-specific pericytes, as well as fat-storing cells and they are also responsible for the extracellular matrix components production. When activated, HSCs lose lipid droplets and increase extracellular matrix secretion. The phenotypical modulation and the HSCs proliferation control have paramount importance to understand the hepatic fibrosis. This research has been carried out with the GRX cell line (HSCs murine representative) which was obtained from inflammatory fibrogranulomatous reactions. Previous studies in our laboratory investigated the action of different drugs on the modulation of these cells, such as: retinol, pentoxifilin, indomethacin and insulin. Resveratrol (RSV) has been referred to as having antioxidant, antiinflammatory and antiproliferative action (3,4,5 trihidroxiestilbeno), as well as the capacity to activate or inhibit the transcription of enzymes involved in the regulation of several cell events, as, for example, lipid metabolism Therefore, we used RSV, which is a natural polyphenol found in the grapes skin and, consequently, in large amounts of red wine, in this cell model. The GRX cells were cultivated (12, 24 and 120 h) in medium DMEM with 5% SFB added RSV or not (100 nM, 1µM or 100 µM) and retinol (RHO) (5µM). Our studies demonstrated that a 120 h treatment with RSV (beginning with 100 nM) inhibited GRX cells proliferation, thus decreasing the cell number in 35%. Assessing the effect of 1µM RSV on the cell cycle and the apoptosis, we found that, after this treatment period, there was an increase of cells in phases S and Sub-G1. Nuclear condensation and fragmentation were observed, which suggested a probable pro-apoptotic RSV effect on the GRX cell. The RSV (100 nM) 120 h treatment did not induce lipid droplets formation in the GRX cells. However, RSV did not interfere in the synthesis or accumulation of lipids caused by RHO (5µM), when RSV+RHO were added. Considering that the lipogenesis in this treatment model might be associated with sirtuin action (deacetilase NAD-dependent) and PPARy (receptors activated by peroxisome proliferators), the mRNA expression of these two proteins was determined. RSV (100 nM) proved to increase sirtuin mRNA expression 1 (SIRT1) and decrease PPARy.expression. However, the treatment with the RSV+RHO caused a reduction in the SIRT1 expression thus alterating the relation PPARy/SIRT1, which promoted the lypid synthesis. Our data showed that after the 24-hour treatment RSV did not change acetate incorporation [C14] in triacylglicerides (TG), whilst RHO or RSV+RHO increased the marker incorporation in these lipids. The TG deposit remained constant, but the synthesis diminished during the treatment period. It appears that these compounds act through different molecular mechanisms when modulating lipids metabolism. According to literature data, we suggest that RHO acts via LXR nuclear receptor, thus forming an heterodímer with PPARy, which favors lypogenesis. On the other hand, RSV activates SIRT1, which inhibits PGC1α (PPARy coactivator). Therefore, the PPARy inhibition by RSV consequently inhibits lypogenesis. We concluded that RSV presents pro-apoptotic, anti-proliferative effect, without inducing lypocytic phenotype in the GRX cells. By this means the mesenquimal activated quiescent cells keep their balance at the Disse space, thus contributing with the hepatic homeostasis restoration.

Avaliação do ciclo celular de células tronco/progenitoras hemopoéticas da medula óssea de camundongos submetidos à desnutrição protéica / Hematopoietic stem/progenitor cell cycle evaluation from bone marrow of malnourished mice

A hemopoese é um processo dinâmico regulado pelo microambiente no qual se situa. O principal tecido hemopoético após o nascimento, a medula óssea, é constituído basicamente por substâncias solúveis, como fatores de crescimento, por uma matriz extracelular (MEC) e por células estromais, além das células hemopoéticas. Esse microambiente indutor íntegro é capaz de regular os processos de sobrevivência, proliferação e diferenciação celular, induzindo a célula a sair de um estado quiescente e entrar em ciclo celular. Contudo, na desnutrição protéica (DP) observa-se redução significativa da celularidade das células hemopoéticas, tanto no compartimento periférico quanto no central, a medula óssea. O comprometimento estrutural do microambinte medular decorrente da desnutrição pode prejudicar a sinalização de indução do ciclo celular, fato este que justificaria o quadro de pancitopenia. Portanto, no presente estudo nos propusemos avaliar o ciclo celular de células tronco/progenitoras hemopoéticas (CTPH) da medula óssea de camundongos desnutridos. Para tanto, utilizamos um modelo murino, sendo a desnutrição induzida a partir de uma ração hipoprotéica. As CTPH foram obtidas por método de depleção imunomagnética e utilizadas para a avaliação do ciclo celular a partir da incorporação de Iodeto de Propídeo (PI) e Laranja de Acridina (AO). Também, foram quantificadas proteínas regulatórias do ciclo celular por western blot e avaliada a expressão de receptores para fibronectina, VLA4 e VLA5. Paralelamente, em modelo ex vivo, avaliou-se a influência de fatores de crescimento e de uma matriz de fibronectina sobre a proliferação das CTPH. Considerando a importância das células estromais na sinalização celular, realizamos o ensaio de CFU-F para a quantificação de células estromais e dos fatores de crescimento secretados. Por fim, avaliamos a eficácia da recuperação nutricional frente às alterações no ciclo celular observadas no modelo de desnutrição. Resumidamente, observou-se um comprometimento no ciclo celular das CTPH de camundongos desnutridos, com um aumento desta população celular nas fases GO/G1. As proteínas indutórias do ciclo celular apresentaram uma expressão reduzida enquanto as proteínas inibitórias apresentaram um aumento de expressão nas CTPH dos animais desnutridos. Ex vivo, porém, as CTPH dos animais desnutridos responderam adequadamente aos estímulos externos fornecidos, uma vez que a proliferação celular foi igual para ambos os grupos. A avaliação da cultura de CFU-F indicou comprometimento quantitativo das células estromais nos animais desnutridos e uma redução da produção de citocinas importantes no controle da hemopoese. Após a renutrição, observamos uma reversão das alterações do ciclo celular, com recuperação da celularidade medular, aumento da população de CTPH nas fases proliferativas (S/G2/M) e a normalização da expressão das proteínas regulatórias do ciclo celular. Podemos concluir que a desnutrição protéica compromete o ciclo celular de CTPH, provavelmente devido às alterações no microambiente medular, comprovadas pela redução de células estromais e de fatores de crescimento. Uma vez que o fornecimento de quantidades ótimas de fatores de crescimento e de uma matriz de fibronectina, mimetizando ex vivo o microambiente íntegro, estimula as células de animais desnutrido a proliferarem, podemos sugerir que todo o mecanismo intracelular de controle do ciclo, aparentemente, não foi comprometido. Em relação à recuperação nutricional, podemos afirmar que esta é eficaz na reversão do quadro de pancitopenia, sendo demonstrado um aumento da proliferação das CTPH. / Hematopoiesis is a dynamic process governed by the microenvironment in witch it is located. Basically, the main hematopoietic tissue, the bone marrow, is composed by soluble factors such growth factors, extracellular matrix (ECM) and stromal cells, besides the hematopoietic cells. This intact inducible microenvironment is capable to control cell survival, proliferation and differentiation, inducing cell to exit a quiescent state and enter the cell cycle. However, in protein malnutrition (PM) is observed a significant reduction of hematopoietic cells, both in peripheral and central compartments. The bone marrow structural impairment due to malnutrition could harm the cell cycle signaling, a fact that could justify the establishment of pancitopenia. Therefore, in this study we set out to assess the cell cycle of hematopoietic stem/progenitors cells (HSPC) from bone marrow of malnourished mice. We used a murine model, and malnutrition induced from a low protein diet. The HPSC were obtained by immunomagnetic depletion method and used for the evaluation of cell cycle from the incorporation of propidium iodide (PI) and Acridine Orange (AO). Also, we quantified the cell cycle regulatory proteins by western blot and evaluated the expression of receptors for fibronectin, VLA4 and VLA5. Meanwhile, in ex vivo model, we evaluated the influence of growth factors and a matrix of fibronectin on the proliferation of HSPC. Considering the importance of stromal cells in cell signaling, we performed the CFU-F assay for the quantification of stromal cells and growth factors secreted. Finally, we evaluated the efficacy of nutritional recovery in the face of cell cycle alterations observed in the model of malnutrition. Briefly, we observed an impairment in the cell cycle of HSPC of undernourished mice, with an increase in this cell population in G0/G1 phase. The proteins that induce cell cycle showed a reduced expression whereas the inhibitory proteins showed increased expression in HPSC of malnourished animals. Ex vivo, however, HSPC of malnourished animals responded appropriately to external stimuli provided, since cell proliferation was similar for both groups. Assessing the culture of CFU-F showed a quantitative impairment of stromal cells in malnourished animals and reducing production of cytokines important in controlling hematopoiesis. After refeeding, we observed a normalization of the cell cycle, with recovery of cellularity, an increased population of proliferative phases HSPC (S/G2/M) and normalization of the expression of cell cycle regulatory proteins. We can conclude that malnutrition compromises the HPSC cell cycle, probably due to changes in the bone marrow microenvironment, as proven by the reduction of stromal cells and growth factors. Since the supply of great quantities of growth factors and a matrix of fibronectin, mimicking an intact microenvironment, stimulates the cells of malnourished animals to proliferate, we suggest that the whole mechanism of intracellular cycle control is apparently not been compromised. Regarding the nutritional recovery, we can say that this is effective in reversal of the pancytopenia, and demonstrated an increased proliferation of HSPC.

Micronúcleos são estruturas constituídas por material cromatínico contido por um envoltório nuclear e menores que o núcleo principal. Apesar do amplo uso do teste do micronúcleo na avaliação do potencial genotóxico de diversas substâncias, os trabalhos cujas preocupações sejam elucidar o mecanismo de formação dessas estruturas, seu conteúdo e a fase do ciclo celular em que surgem, são raros. O objetivo do presente trabalho foi estudar a cinética de surgimento de micronúcleos em células A549, provenientes de carcinoma de pulmão humano, submetidas à sincronização. Após duplo bloqueio por ausência de soro fetal bovino e adição de vincristina ao meio, o surgimento de micronúcleos foi acompanhado com o auxílio de um marcador para fase S. Os resultados obtidos indicam que micronúcleos podem surgir tanto durante a mitose quanto na intérfase e possuem capacidade de replicação de DNA, independentemente do núcleo principal. A capacidade de escapar ao duplo bloqueio permite sugerir que o ponto de checagem de ligação ao fuso mitótico em A549 não é funcional. Também foram observadas seqüências amplificadas de EGFR no interior dos micronúcleos. A interpretação do significado dos micronúcleos é importante para a definição de sua relação com a expulsão de oncogenes em células tumorais ou de outras seqüências amplificadas. / Micronuclei are structures composed by chromatin material contained in the nuclear envelope and smaller than the main nucleus. Micronucleus test has been widely used in the genotoxic potential evaluation of different compounds. Although a few report concerning on the mechanism of micronucleus genesis, its DNA sequences content and the cell cycle phase when they arise. The aim of this work was to analyze the kinetic of micronuclei in A549 cells from human lung carcinoma submitted to cell cycle synchronization. After double blocking by fetal bovine serum deprivation and vincristine treatment, micronucleus formation was monitored with a S-phase marker. The results have showed that both in the mitosis and in the interphasic phase, micronuclei may arise and they were able to replicate its DNA. This process seemed to be independent of main nucleus. Cellular ability to escape from double blocking suggests that mitotic spindle checkpoint in A549 is not functional. Moreover, EGFR sequences were detected into the micronucleus. It is important to elucidate the micronucleus meaning to describe precisely its association with elimination of oncogene or other amplified sequences from the tumor cells.

This study analyzed the behavior of human skin fragment fibroblasts in a simulated microgravity environment. Methods: Human fibroblast cells in the 8th and 17th passage, cultured under standard incubator conditions at 37? C with 5% CO2, were submitted to simulated microgravity in a 3D-clinostat for a period of 24h and 40h. After exposure, both passage cells were analyzed and compared with the control group (1G) in population doubling assays, tests of passage and microscopic analysis, as well as PCR analysis for detection of variations in the gene expression related to the cell cycle (p21, p16). Results: Before microgravity exposure, cells belonging to the 17th passage presented characteristics of cells in an apoptotic state. After 24h and 40h of microgravity, the cells of both groups showed themselves to be more confluent and elongated. PCR analysis demonstrated that p21 expression was decreased while p16 increased. In addition, PCR analysis showed a difference in expression of p21 and p16 genes between the 24h and 40h samples. Discussion: The present research showed cells to be more confluent and elongated after microgravity exposure, a characteristic of cells with fewer passages, suggesting alterations in their cytoskeleton. This result was confirmed by PCR analysis where a decrease in p21 expression was demonstrated. This result corroborates previous findings that among 588 genes tested, the p21 gene presented a negative expression. Conversely, the p16 gene showed a positive expression. Since both the p21 and p16 genes are related to the cell cycle, these results suggest the hypothesis of important changes having occurred in the cellular cytoskeleton and, consequently, a probable alteration in the production of cell cycle regulatory proteins (cyclins). Furthermore, RT-PCR analysis demonstrated a difference in p21 and p16 gene expression between the 24h and 40h samples, indicating the need for a more detailed comparison between the exposure times. Atividade antiproliferativa dos alcalóides piperidínicos (-)- cassina e (-)-espectalina sobre células de carcinoma hepatocelular humano - HepG2

O câncer tem sido apontado como um dos problemas mais críticos encontrados no sistema público de saúde em todo o mundo, além de ser um dos maiores desafios para a medicina deste século. O carcinoma hepatocelular é a segunda maior causa de morte por câncer em todo o mundo e o quinto mais incidente. Infelizmente, a maioria dos pacientes são diagnosticados em estágio avançado, quando as opções de tratamento são apenas paliativas. Dessa forma, torna-se relevante a busca por novas opções terapêuticas mais efetivas para o carcinoma hepatocelular. Neste contexto, as plantas representam uma importante fonte de descoberta de novos compostos com propriedades farmacológicas, incluindo agentes antineoplásicos. O presente trabalho objetivou investigar o potencial antitumoral in vitro dos alcaloides (-)-cassina e (-)-espectalina, os quais são derivados de Senna spectabilis. A viabilidade celular foi determinada por diferentes métodos colorimétricos (MTS e rezasurina) e pelo ensaio de exclusão em azul de tripano. Ensaio clonogênico foi realizado para avaliar a formação de colônias, enquanto a cinética de progressão do ciclo celular foi determinada por citometria de fluxo. O padrão de organização de elementos do citoesqueleto bem como a frequência de mitoses foram analisadas por imunofluorescência. O perfil de expressão foi determinado por qRT-PCR (ciclina D1) e Western blotting (Histona H3 fosforilada, ERK fosforilada e ERK total). Os resultados mostraram que a mistura de alcaloides reduz significativamente a viabilidade celular, contudo o efeito foi dependente da concentração utilizada. A redução na proliferação celular foi decorrente do bloqueio do ciclo celular preferencialmente na transição G1/S. Os eventos moleculares responsáveis pela regulação negativa do ciclo celular, pelo menos em parte, estão relacionados à capacidade dos alcaloides de inibir expressão de ciclina D1 e ativação de ERK, uma proteína da via de MAPKs. Além disso, os alcaloides também foram efetivos em alterar o padrão de organização normal dos filamentos de actina e microtubulos. Portanto, conclui-se que os alcaloides (-)-cassina e (-)-espectalina apresentam atividade antitumoral promissora contra células de carcinoma hepatocelular e sua estrutura química deve ser considerada para futuros estudos anticâncer in vivo. / Cancer is one of the most critical problems of public health in the world and one of the main challenges for medicine in this century. The hepatocellular carcinoma is the second cause of cancer death and the fifth most incident. Unfortunately, most patients are diagnosed at advanced stage, when the treatment options are palliative. Consequently, the search for novel therapeutic options for hepatocellular carcinoma becomes imperative. In the context, the plants represent an important source for discovery of novel compounds with pharmacological potential including antineoplastic agents. Herein, we aimed to investigate in vitro antitumor potential of an alkaloid mixture derived from Senna spectabilis, (–)-cassine and (–)-spectaline. Cell viability was measured by different colorimetric methods (MTS and rezasurin assay) and trypan blue exclusion. Clonogenic formation assay were promoted in order to evaluate colony formation inspection, while cell cycle progression was determined by flow cytometry. Cytoskeleton elements and mitosis frequency were analyzed by immunofluorescence. Gene expression was determined by immunoblot (ERK1, p-ERK and p-histone H3) and qRT-PCR (Cyclin D1). Alkaloid mixture significantly reduced cell viability of cancer cells in a concentration-dependent manner. Cell proliferation reduction occurred by cell cycle arrest mainly in G1/S transition. We demonstrated that antiproliferative potential of the alkaloids is due to, at least in part, down-regulation of cyclin D1 and ERK inactivation, which is a MAPK pathway protein. Alkaloids also drastically altered F-actin and microtubules distribution pattern.
