Utveckling av mobil kylförvaringslösning för hormonpennor / Development of a mobile cold storage solution for hormone injection systems

Att leva med diabetes kan vara krångligt då det hela tiden krävs planering av aktiviteter, måltider och medicinering. Sjukdomen behandlas framförallt med insulin som kan förekomma i pump, sprutor och injektionspennor. Det vanligaste sättet att injicera insulin är med pennor, och antalet pennanvändare ökar årligen. Insulin har stränga förvaringskrav och är känsligt mot hög värme, starkt ljus, skakningar och frysning. De personer som lever i länder där temperaturen över- eller understiger säkert förvarings-intervall har ett behov av att skydda sin medicin, men också resande till utsatta platser drabbas. Andra mediciner och hormon har förvaringskrav inom samma ramar som insulinet och intresse och behov finns för en förvaringslösning som kan säkerställa trygg förvaring av läkemedel som används dagligen. För att säkerställa en korrekt temperatur i en framtida produkt undersöks teorier inom värmeöverföring och kunskaper om isolerande material i kombination med Climators ClimSel™-kylelement. Det finns ett flertal konkurrerande lösningar på marknaden, dock har de vissa negativa egenskaper som medför att det skulle kunna finnas utrymme för ytterligare produkter om de får tillräckligt bra prestanda. Enligt förstudien finns behov av att skydda hormonerna mot både kyla och värme. Produkten utvecklas för att tillgodose behovet hos vardagsanvändare av hormoner för att maximera marknadspotentialen. En stor del av arbetet koncentreras på temperaturtester där olika kylelement, isoleringsmaterial och geometrier testas och utvärderas för att utveckla den optimala systemdesignen. Förvaringslösningens detaljdesign utvecklas med olika kreativitetsmetoder med fokus på användbarhet, target cost och optimal prestanda. Resultatet påvisar att förvaringslösningens prestanda blir för låg vid en geometri och volym som anses acceptabel hos användarna, vilket medför att det finns svårigheter i att uppnå en relevant marknadspotential. / Living with diabetes can be difficult due to the required extra planning of activities, meals and medica-tion. The disease is treated primarily with insulin, which can be delivered by pump, syringes or pens. The most common way to inject insulin is by pen, and the number of pen users is increasing annually. Insulin has strict storage requirements and is sensitive to heat, strong light, vibrations and freezing. Users in countries where the temperature goes above or below the safe storage interval have a need to protect their medicine, as well as users travelling to affected places. Other drugs and hormones have storage requirements within the same framework as insulin, which means that there is an additional interest and need for a storage solution that can ensure safe storage of medicine that is used daily. To ensure the correct temperature in a future product, this project will examine theories of heat transfer and knowledge of insulating materials in combination with a range of ClimSel™-cooling elements produced by Climator Sweden AB. There are several competing solutions currently on the market; however, they have some negative char-acteristics which mean that there could be room for additional products if they have good enough per-formance. According to the preliminary study there is a demand of a product that protects the hormones against both excessive cold as well as heat. The product is developed to meet the needs of everyday users of hormones to maximize market potential. Most of the work focused on temperature tests involving different cooling elements, insulation materials as well as geometries. All which are tested and evaluated to develop the optimal system design. Addition-ally detailed design is conducted with different creativity methods with focus on usability, target cost and performance. The results show that the finalized concepts performance is not high enough at an accepted geometry and volume, which means that there are great difficulties in achieving a relevant market pene-tration.

Die Arbeit bietet einen praxisbezogenen Überblick über den rationellen Einsatz von thermischen Kälteanlagen. Dabei wird von der Stoffdatenanlayse, bei der eine unfassende Analyse und Kritik und eine Korrektur der Ansätze durchgeführt wird, der darauf aufbauenden rechentechnischen Simulation der wichtigsten Maschinen über eine Optimierung der Kaltwasserspeicherung durch einen neuartigen Speicherbehälter bis zur Darlegung einer effizienten Einbindung in eine Systemschaltung und der energetischen Bewertung die gesamte Bandbreite der thermisch betriebenen Kälteerzeugung abgedeckt. In diesem umfassenden Ansatz stellt die Arbeit ein Novum dar. Insbesondere im Bereich der Kaltwasserspeicherung wird eine konstruktiv neue Lösung vorgeschlagen, die einen wirtschaftlichen Einsatz der Anlagen gerade im innerstädtischen Bereich, der ein hohes Entwicklungspotenzial besitzt, verbessert oder überhaupt erst ermöglicht. In Verbindung mit den Erkenntnissen zur Anlagensimulation und Einsatzoptimierung trägt die Arbeit zu einem verstärkten Einsatz von thermischen Kälteanlagen und damit zur Erschließung energetischer Einsparpotenziale bei. Des Weiteren wurde die Effizienz zweier momentan nicht genutzter Technologien dargestellt und eine Umsetzung empfohlen. Die Ergebnisse lassen sich auf mehrere Arten kommerziell nutzen. Einerseits wird die Erstellung von effizienten Simulationstools für verschiedene thermisch betriebene Kälteanlagen und Wärmepumpen ermöglicht, andererseits die Umsetzung einer neuen effizienten Speicherlösung beschrieben und die Rahmenbedingungen des Einsatzes innovativer Technologien dargelegt (Resorptions- und Absorptionsanlage mit mechanischer Kompression) und eine sinnvolle Nutzung im energiepolitischen Rahmen der Bundesrepublik anhand von Bewertungsalgorithmen ermöglicht. Gerade im Hinblick auf den von der Bundesregierung angestrebten Ausbau der Fernwärmenutzung in Deutschland lassen sich durch die Nutzung von KWK-Wärme im Sommer neue wirtschaftliche Potenziale erschließen. Durch die kritische Analyse der thermodynamischen Grundlagen der Mischung der Stoffe Lithiumbromid und Wasser erstellt die Arbeit die Basis für eine wissenschaftlich korrekte Berechnung von Simulationswerkzeugen und konstruktive Umsetzungen von thermischen Kälteanlagen / Wärmepumpen und ermöglicht sowohl Planern und Wissenschaftlern als auch Studenten oder interessierten Laien einen umfassenden Blick auf die Technologien und ihre energiepolitisch sinnvollen Einsatzchancen. / The thesis contains a practical overview of the rational use of thermal chillers. Based on an extensive analysis of the data on chemical media, a computer simulation of the most important absorption chillers / heat pumps was realised. Furthermore an optimisation of the cold water storage by a novel storage tank construction and explanations of the efficient integration of absorption chillers into an integrated network are given. Especially the novel horizontal cold water storage advances the general conditions of the application of absorption chillers in municipal areas. Also the efficiencies of two currently neglected technologies are shown.

Enhancement of 'Hass' avocado shelf life using ultra-low temperature shipping or 1-MCP treatment and cold chain management.

Avocados are becoming an increasingly important crop in South Africa, where the main producing areas include Limpopo, Mpumalanga and KwaZulu-Natal provinces. The South African avocado industry faces considerable challenges including increasing competition exporting avocados, particularly to the European market. The processes involved to export avocados has markedly improved over the past two decades, however there is always room for improvement and it is necessary to remain competitive on a global scale. Issues such as fruit being partially soft on arrival, quality defects and cold chain management breakdown are still present. It is necessary to investigate new aspects of cold storage such as extending the storage period and understanding the physiological aspects involved. To improvement such issues, an investigation was conducted on ultra-low temperature shipping (1°C) as well as the use of 1-MCP; the implementation of deliberate cold chain breaks to achieve a better understanding as to the quality influences involved; an extended storage period of 56 days to assess the quality issues and benefits involved; as well as investigating the physiological aspects involved with all above treatments on 'Hass' avocados. An initial study saw early-, mid- and late-season 'Hass' avocados stored at 1°C or 5.5°C for 28 days. Additional treatments included fruit treated and not treated with 1-MCP as well as waxed and unwaxed fruit. Storage at 1°C was comparable with 1-MCP treatment for both fruit softening in storage and extending the ripening period. Storage at 5.5°C resulted in partial in-transit ripening, if 1-MCP was not used. Early-season fruit incurred the most external chilling injury but overall levels were minimal and not concerning. Mid-season fruit were the most sound in terms of quality. It is suggested that 1°C can be used as a viable economic alternative to 1-MCP for long distance shipping of 'Hass' up to 28 days. The cold chain break trial included a 24 hour delay before cold storage, a deliberate 8 hour break at day 14 of cold storage where fruit were removed from cold storage and a control of 28 days cold storage where no break was involved. Early-, mid- and late-season 'Hass' avocados were stored at 1°C or 5.5°C for 28 days. Additional treatments included fruit treated and not treated with 1-MCP as well as waxed and unwaxed fruit. It was found that cold chain breaks do influence the amount of water loss, fruit softening and days taken to ripen. Storage at 1°C did not entirely negate the effects of cold chain breaks compared with 5.5°C, but did result in fruit which were harder at the end of storage and took longer to ripen. The use of 1-MCP also had advantageous effects with respect to significantly lengthening the ripening period, even when a cold chain break occurred, compared with fruit not treated with 1-MCP. As results of the study differed in some respects to those of previous studies, it is recommended that further work be conducted to determine what fruit or pre-harvest factors affect the fruit physiological changes which take place when cold chain breaks occur. Having the option to make use of an extended storage period would be of benefit to the industry if delays occur and fruit have to be maintained under cold storage. Extended storage of South African avocados, especially at the end of the season would also allow for the option of strategically holding back fruit from the export market in order to extend the supply period. It would not only benefit export options, but would also be highly beneficial to local pre-packers, as it would reduce the need to import fruit from the Northern hemisphere production areas during the South African off-season. Early-, mid- and late-season 'Hass' avocados were stored at 1°C or 5.5°C for 56 days. Additional treatments included fruit treated and not treated with 1-MCP as well as waxed and unwaxed fruit. The combination of 1°C with the use of 1-MCP resulted in a good shelf life as well as maintenance of internal quality and integrity. External chilling injury is of concern for early-season fruit, however, mid- and late-season fruit did not incur extensive damage. It is, therefore, advised that fruit placed in extended storage are marketed through the 'Ready ripe' program to mask any chilling injury on the 'Hass' fruit. Avocados are renown as a "healthy food" due to their nutritional value as well as containing relatively high concentrations of antioxidants. The fruit also contain high amounts of C7 sugars which can act as antioxidants. Additionally, C7 sugars and other antioxidants play important roles in fruit quality. Therefore, it is important to understand how varying storage conditions and treatments affect the levels of these physiological parameters. Treatments of cold chain break/delay included a deliberate 8 hour break at day 14 of cold storage where fruit were removed from cold storage, a 24 hour delay before cold storage and a control of 28 days where no break was involved. A 56 day extended storage period was also used. Early-, mid- and late-season 'Hass' avocados were stored at 1°C or 5.5°C for 28 days. Additional treatments included fruit treated and not treated with 1-MCP as well as waxed and unwaxed fruit. The use of 1-MCP maintained higher levels of antioxidants, ascorbic acid and C7 sugars for both the 28 day and the 56 day storage periods. The 24 hour delay had a tendency to increase consumption of anti-oxidant and sugar reserves. The use of 1°C resulted in antioxidant and ascorbic acid levels decreasing while maintaining higher sugar levels. Overall, high stress imposed on fruit decreased reserves resulting in poor quality fruit. Prediction of post-storage quality in canning apricots and peaches using near infrared spectroscopy (NIRS) and chemometrics

Thesis (MSc Food Sc)--Stellenbosch University, 2003. / ENGLISH ABSTRACT: Post-storage quality of the stone fruit, apricots and peaches, is the major factor determining their suitability for canning after cold storage in South Africa. Short harvesting periods and the limited capacity of the factory to process the large quantities of fruit within two days after delivery, necessitates cold storage until canning. Apricots develop internal breakdown, whereas peaches develop internal breakdown accompanied by loosening of the skin and adhesion of the flesh to the stone. The deterioration takes place within the fruit during a cold storage period of one to two weeks. The tendency of the fruit to develop internal defects can, to date, not be identified prior to storage and are only discovered after destoning during canning. Near infrared spectroscopy (NIRS) combined with chemometrics were investigated as a non-destructive method to predict post-storage quality in Bulida apricots and clingstone peach cultivars. Near infrared (NIR) spectra (645-1201 nm), measured on the intact fruit just after harvesting, were correlated with subjective quality evaluations performed on the cut and destoned fruit after cold storage. The cold storage periods for apricots were four weeks (2002 season) and three and two weeks for peach cultivars for the 2002 and 2003 seasons, respectively. Soft independent modelling by class analogy (SIMCA) and multivariate adaptive regression splines (MARS) were applied to the spectral and reference data to develop models for good and poor post-storage quality. The ability of these models to predict post-storage quality was evaluated in terms of recognition (sensitivity) and rejection (specificity) of the samples in independent validation sets. Total correct classification rates of 50.00% and 69.00% were obtained with Bulida apricots, using SIMCA and MARS, respectively. Classification results with apricots showed that MARS performed better than SIMCA and is thus recommended for this application. Total correct classification rates of 53.00% to 60.00% (SIMCA) and 57.65% to 65.12% (MARS) were obtained for data sets of combined peach cultivars within seasons and over both seasons. Additional aspects of fruit quality were investigated to identify possible indices of post-storage quality. Classification trees were used to find correlations between the post-storage quality and the fruit mass, diameter, firmness and soluble solids content (SSC). Among these, fruit diameter and firmness were the major indices of post-storage quality. Accurate predictions of firmness could not be achieved by near infrared spectroscopy (NlRS), making the combination of NIRS and classification trees not yet suitable for predicting post-storage quality. NIRS was further used to predict poststorage SSC within seasons in Bulida apricots and intact peach cultivars. This confirmed sufficient NIR light penetration into the intact fruit and also provided a further application of NIRS for ripeness evaluation in the canning industry. Validations on peach samples obtained correlation coefficients (r) of 0.77-0.85 and SEP-values of 1.35-1.60 °Brix using partial least squares (PLS) regression. MARS obtained r = 0.77-0.82 and SEP = 1.42-1.55 °Brix. Predictions of sse in apricots were less accurate, with r = 0.39-0.88, SEP = 1.24-2.21 °Brix (PLS) and r = 0.51-0.82, SEP = 1.54-2.19 °Brix (MARS). It is suggested that the accuracy of sse measurements, and the subsequent predictions, were affected by the cold storage periods as well as internal variation within the fruit. This study showed that a combination of NIRS and chemometrics can be used to predict post-storage quality in intact peaches and apricots. A small scale feasibility study showed that 4% (R117 720) (apricot industry) and 3% (R610 740) (peach industry) of production losses can be saved if this method is implemented in the South African canning industry. Although it was difficult to assign specific chemical components or quality attributes to the formulation of the storage potential models, important hidden information in the spectra could be revealed by chemometric classification methods. NIRS promises to be a useful and unique quality evaluation tool for the South African fruit canning industry. Several recommendations are made for the canning practices to reduce losses and for future research to improve the current prediction models. / AFRIKAANSE OPSOMMING: Die kwaliteit van die steenvrugte, appelkose en perskes, is die hoof bepalende faktor vir hul geskiktheid vir inmaakdoeleindes na koelopberging in Suid-Afrika. Soroprevalência da brucelose bovina em frigoríficos com diferentes níveis de serviço de inspeção no estado de São Paulo /

Orientador: Jane Megid / Banca: Luis Antonio Mathias / Banca: José Paes de Almeida Nogueira Pinto / Resumo: Foi estimada a prevalência estadual da brucelose bovina em abatedouros nos diferentes níveis de inspeção sanitária no Estado de São Paulo após a implantação do Programa Nacional de Controle e Erradicação da Brucelose e Tuberculose (PNCEBT), instituído em 2001. Para tanto, foram amostrados 1.490 bovinos em nove frigoríficos distribuídos em diferentes regiões do estado, dos quais três com inspeção federal (SIF), três com inspeção estadual (SISP) e três com inspeção municipal (SIM). Os soros foram analisados pelas provas do antígeno acidificado tamponado (AAT), e os reagentes foram confirmados pela reação de fixação de complemento (RFC). As prevalências obtidas por meio da RFC e seus respectivos intervalos de confiança, nos diferentes serviços de fiscalização, foram 0,40% [0,00% - 0,95%] (SIF), 2,00% [0,77% - 3,23%] (SISP) e 4,29% [2,67% - 6,48%] IC95% (SIM) e no estado foi estimada em 2,21% [1,47% - 2,96%]. Com os resultados obtidos, foi possível discutir a situação da brucelose bovina no Estado de São Paulo bem como o risco à saúde a que os trabalhadores de abatedouros estão expostos e a diferença entre os sistemas de inspeção vigentes no país. Concluiu-se que a soroprevalência da brucelose bovina no Estado de São Paulo permaneceu estável. Sugere-se que a adoção de novas medidas para complementar o PNCEBT poderiam acelerar o controle e a posterior erradicação da enfermidade; e que o aumento da fiscalização nos abatedouros de inspeção estadual e municipal acrescido de vigilância ativa nesses pontos poderia auxiliar no combate à doença / Abstract: This study estimated the prevalence of bovine brucellosis in state of São Paulo as well as the prevalence in slaughterhouses with different levels of sanitary inspection in the same state after the implementation of the National Program for the Control and Eradication of Brucellosis and Tuberculosis (PNCEBT) established in 2001. Were sampled 1,490 animals in nine slaughterhouses distributed in different regions of the state, three with federal inspection (FI), three with state inspection (SI), and three with municipal inspection (MI). Sera were analyzed by Rose Bengal Test (RBT) and reaction of complement fixation test (CFT). The prevalence obtained through CFT in different inspection services were, 0.40% [0.00% - 0.95%] (FI), 2.00% [0.77% - 3.23%] (SI) e 4.29% [2.67% - 6.48%] IC95% (MI) and the state prevalence was estimated at 2.21% [1.47% - 2.96%]. With the results obtained, it was possible to evaluate the situation of the bovine brucellosis in state of São Paulo as well as the risk to public health that workers are exposed in slaughterhouses and the difference between the current inspection systems in the country . It was concluded that the seroprevalence of brucellosis in the state of São Paulo remained stable, and the adoption of new measures to enhance the PNCEBT could acelerate the control and eventual eradication of the disease. Os abatedouros e frigoríficos caracterizam-se por agroindústrias potencialmente poluidoras devido a carga orgânica componente dos efluentes líquidos e resíduos sólidos gerados em seu processo. Para a minimização do impacto ambiental gerado por estes empreendimentos faz-se necessária a implementação de processos de tratamento que possibilitem a redução da carga orgânica, a transformação de resíduos em produtos de valor agregado e o aproveitamento energético. A implementação de biodigestores para o tratamento dos resíduos sólidos e efluentes líquidos gerados nos frigoríficos de suínos é uma opção interessante, haja visto o processo de digestão anaeróbia que compõe o sistema e os benefícios possíveis com sua aplicação. Neste intuito, o presente trabalho teve como objetivo a avaliação de um digestor anaeróbio de fluxo contínuo, em escala real, utilizado para o tratamento dos resíduos sólidos e efluentes líquidos oriundos do processo. Para isto procedeu-se a caracterização físico-química dos substratos de alimentação do biodigestor bem como do afluente e efluente do sistema, além da realização de análises com o intuito de avaliar as características do biofertilizante e do biogás originados no processo. Os resultados demonstraram que o substrato de alimentação do biodigestor é composto por diversas origens e, cada qual, possui características físico- químicas específicas e diferenciadas porém, quando unificadas, propiciam o desenvolvimento de um ambiente suscetível ao desempenho da digestão anaeróbia. Avaliando a eficiência do sistema em geral, considerando os valores obtidos nas análises do afluente e efluente do mesmo, observou-se que alguns parâmetros apresentam eficiência enquanto, outros, resultaram em aumento de concentração. Este fato pode ter sido desencadeado por fatores externos ao sistema. Quanto ao biofertilizante oriundo do biodigestor, foi caracterizado como resíduo sólido não inerte sendo possível, com algumas restrições, a sua aplicação em solo. O biogás, por sua vez, apresentou concentrações interessantes de metano, sendo, portanto, passível de utilização em diversos processos industriais. / Slaughterhouses are potentially polluting agricultural industries due to the organic load present in the wastewater and solid waste generated in the process. To minimize the environmental impact generated by this type of activity, it is necessary to provide a treatment processes that enables the reduction of the organic matter present in slaughterhouse waste, transforms waste into value-added products and exploits energy use efficiently. The implementation of biodigesters to treat solid waste and wastewater generated in slaughterhouses is an interesting option, given the anaerobic digestion process that makes up the system and the possible benefits it provides. Therefore, the present study evaluated the efficiency of an anaerobic continuous flow digester, in real scale, used for the treatment of solid waste and liquid effluents from the process. In order to do so, physicochemical characterization of the digester feed substrates, as well as, characterization of the influent and effluent of the system were performed. In addition, analyzes were executed to evaluate the characteristics of the bio-fertilizer and biogas originating in the process. The results showed that the digester feed substrate is composed of various origins and, each has a specific and distinct physicochemical characteristic, but when these different characteristics are combined they favor the development of an environment prone to the performance of the anaerobic digestion. By assessing the overall system efficiency, while considering the values obtained for the influent and effluent analysis, some parameters were considered efficient, whereas others result in the increase of concentration. This may have been triggered by factors which are external to the system. The bio-fertilizer originating from the digester was characterized as non-inert solid waste, which might require certain restrictions in its application in soil. Biogas, in turn, presented interesting methane concentration and is therefore likely to be used in various industrial processes.

O Clostridium perfringens está envolvido com prejuízos na avicultura, pois a doença (Enterite Necrotica) na maioria das vezes é subclínica, e o produtor só observa na hora do abate que a ave não ganhou o peso esperado. O controle dessa e de outras enfermidades e agentes patogênicos na avicultura de corte tem grande importância uma vez que o Brasil está entre os principais países exportadores de carne. O objetivo deste trabalho primeiramente foi pesquisar Clostridium perfringens em frangos de corte sadios, provenientes de dois grandes frigoríficos com Inspeção federal, um no Estado de São Paulo e outro no Estado de Minas Gerais. Foram obtidos 300 raspados intestinais, os quais foram semeados em Agar SPS e incubados em anaerobiose. A identificação dos tipos A, B, C, D e E de C. perfringens foi realizada mediante o emprego da PCR multiplex por amplificação dos genes codificadores das toxinas alfa, beta, épsilon e iota do mesmo. A partir das amostras positivas na PCR (37), procedeu-se o isolamento do agente. Depois de muitas tentativas, verificou-se por meio do sequenciamento da região 16S rDNA, que as colônias sulfito redutoras que estavam sendo isoladas eram Enterococcus spp, microrganismo coexistente na microbiota intestinal de frango de corte, que compete com o C. perfringens pelo crescimento das colônias no mesmo meio de cultura, ainda que este seja seletivo e diferencial para clostrídios sulfito redutores. Dessa forma, notou-se que o crescimento de colônias de enterococos interferia muito na identificação e enumeração do C. perfringens e ainda apresentavam colônias muito semelhantes às de C. perfringens nos oito meios de cultura testados. Clostridium perfringens is involved with losses in the poultry industry, because the disease is most often subclinical, and producer only observed at the time of slaughter the bird has not gained the expected weight. The control of this and other diseases and pathogens in poultry production is very important since Brazil is among the major meat exporting countries. The aim of this study was to evaluate the presence of Clostridium perfringens in poultry coming from two large slaughter-house, one from São Paulo state and another from Minas Gerais. Were obtained 300 intestinal scraped and cecal contents, which were sown onto SPS agar and incubated anaerobically. The identification of C. perfringens types A, B, C, D and E was performed by the use of multiplex PCR. After identification of the presence of C. perfringens in 37 of 300 samples by amplification of the cpa gene, alpha toxin encoder, the isolation of pure colonies was proceeded. However, through the sequencing of 16S rDNA region, it was also found the presence of Enterococcus spp. that lives in the intestinal microbiota of poultry and disputes in growth on the same culture medium, even been selective and differential for clostridia. Thus, it was observed that colony morphology of enterococci is very similar to C. perfringens in this medium what make isolation of pure colonies difficult. Soroprevalência da brucelose bovina em frigoríficos com diferentes níveis de serviço de inspeção no estado de São Paulo / Seroprevalence of bivine brucellosis in slaugtherhouses with inspection service from the state of São Paulo

Para tanto, foram amostrados 1.490 bovinos em nove frigoríficos distribuídos em diferentes regiões do estado, dos quais três com inspeção federal (SIF), três com inspeção estadual (SISP) e três com inspeção municipal (SIM). Os soros foram analisados pelas provas do antígeno acidificado tamponado (AAT), e os reagentes foram confirmados pela reação de fixação de complemento (RFC). As prevalências obtidas por meio da RFC e seus respectivos intervalos de confiança, nos diferentes serviços de fiscalização, foram 0,40% [0,00% - 0,95%] (SIF), 2,00% [0,77% - 3,23%] (SISP) e 4,29% [2,67% - 6,48%] IC95% (SIM) e no estado foi estimada em 2,21% [1,47% - 2,96%]. Com os resultados obtidos, foi possível discutir a situação da brucelose bovina no Estado de São Paulo bem como o risco à saúde a que os trabalhadores de abatedouros estão expostos e a diferença entre os sistemas de inspeção vigentes no país. Concluiu-se que a soroprevalência da brucelose bovina no Estado de São Paulo permaneceu estável. Sugere-se que a adoção de novas medidas para complementar o PNCEBT poderiam acelerar o controle e a posterior erradicação da enfermidade; e que o aumento da fiscalização nos abatedouros de inspeção estadual e municipal acrescido de vigilância ativa nesses pontos poderia auxiliar no combate à doença / This study estimated the prevalence of bovine brucellosis in state of São Paulo as well as the prevalence in slaughterhouses with different levels of sanitary inspection in the same state after the implementation of the National Program for the Control and Eradication of Brucellosis and Tuberculosis (PNCEBT) established in 2001. Were sampled 1,490 animals in nine slaughterhouses distributed in different regions of the state, three with federal inspection (FI), three with state inspection (SI), and three with municipal inspection (MI). Sera were analyzed by Rose Bengal Test (RBT) and reaction of complement fixation test (CFT). The prevalence obtained through CFT in different inspection services were, 0.40% [0.00% - 0.95%] (FI), 2.00% [0.77% - 3.23%] (SI) e 4.29% [2.67% - 6.48%] IC95% (MI) and the state prevalence was estimated at 2.21% [1.47% - 2.96%]. With the results obtained, it was possible to evaluate the situation of the bovine brucellosis in state of São Paulo as well as the risk to public health that workers are exposed in slaughterhouses and the difference between the current inspection systems in the country . It was concluded that the seroprevalence of brucellosis in the state of São Paulo remained stable, and the adoption of new measures to enhance the PNCEBT could acelerate the control and eventual eradication of the disease. Os frutos foram adquiridos em pomar comercial localizado no município de Taiúva-SP nos meses de outubro e novembro de 2007 e 2008. O trabalho foi dividido em cinco experimentos: no primeiro foram estudados estádios de maturação “de vez” e “maduro”. No segundo testou-se o efeito da aplicação de ácido ascórbico ou cloreto de cálcio. No terceiro verificou-se o efeito de embalagens como atmosfera modificada passiva. No quarto, foi verificado o efeito de temperaturas de refrigeração e tipos de cortes. No quinto testou-se o efeito da aplicação de ácido ascórbico, l-cisteína, ácido ascórbico e cloreto de cálcio, ácido cítrico e isoascorbato de sódio na prevenção do escurecimento enzimático. As análises de aparência, massa fresca, firmeza, coloração, composição gasosa, sólidos solúveis, acidez titulável, razão sólidos/acidez, pH, açúcares solúveis e redutores, pectina total e solúvel, solubilização, ácido ascórbico, atividade das enzimas peroxidase e polifenoloxidase foram efetuadas a cada três dias para os experimentos 1, 2, 3 e 5 e, a cada quatro dias para o quarto experimento. Frutos no estádio de maturação “de vez” levaram a produtos com melhor aparência e maiores teores de açúcares e de ácido ascórbico. Produtos tratados com ácido ascórbico a 2% apresentaram melhor aparência, maior conteúdo de açúcares e de ácido ascórbico, manutenção da cor e menor atividade enzimática. O filme de PVC 14 μm proporcionou boa conservação da aparência e prevenção do escurecimento dos produtos. A interação do armazenamento a 3 ºC e corte do fruto em oito fatias proporcionaram... / This study aimed to evaluate the maturation stage, packaging types, natural additives use and storage temperatures of minimally processed „Aurora-1‟ peaches in order to maintain their quality and shelf life. The fruits were purchased from a commercial orchard in the area of Taiúva-SP in October and November, 2007 and 2008. The work was divided in five experiments: in the first one, maturated green and maturated stages were studied. In the second one, it was tested the effect of ascorbic acid or calcium chloride application. In the third one, it was observed the packaging effect as passive modified atmosphere. In the fourth one, it was showed the effect of cooling temperatures and cutting types. In the fifth one, it was tested the effect of ascorbic acid, l-cysteine, ascorbic acid and calcium chloride, citric acid and sodium isoascorbate to prevent enzymatic browning. Analyses of appearance, fresh weight, firmness, color, gas composition, soluble solids, acidity, ratio, pH, soluble and reducing sugars, total and soluble pectin, solubilization, ascorbic acid, peroxidase and polyphenoloxidase activity were done each three days for the first, second, third and fifth experiments, and each four days for the fourth one. Matured green fruits led to products with better appearance and higher levels of sugars and ascorbic acid. Products treated with 2% ascorbic acid showed a better appearance, higher content of sugars and ascorbic acid, color maintenance, and lower enzymatic activity. 14μm PVC film use showed good appearance preservation and browning prevention of the products. Interaction between storage at 3°C and fruit cutting into eight slices provided the best product appearance, with a higher content of soluble solids. 1% Cysteine gave non-characteristic product odor, which limited its use. Qualidade do manejo no frigorífico: efeitos no bem-estar animal e na qualidade da carcaça e da carne

Foram avaliados 42.360 animais em três plantas frigoríficas sob o controle do Serviço de Inspeção Federal (SIF). Foram registrados indicadores de bem-estar animal (frequências de quedas sofridas pelos bovinos), da qualidade de manejo (frequências de porteiradas sofridas pelos animais e utilização do bastão elétrico pelos manejadores), durante o manejo de condução dos bovinos nos frigoríficos, que foram usados para compor o Índice de Qualidade de Manejo (IQM). Foram registrados os indicadores de qualidade de carcaça (pH e hematomas observados nas carcaças). Registrou-se também o número de carcaças com valor de pH acima de 5,9 e com presença de múltiplos hematomas (pelo menos três); além das ocorrências de abates de emergência realizados e atos de abuso (animais pisoteados e fraturados sendo manejados). Para efeito de controle foram registradas também as seguintes informações do manejo: horário do dia, categorias de animais manejados e se houve mistura de categorias nos lotes de manejo. Para análise dos dados foram utilizados os procedimentos GLM e CORR do SAS, além de teste de Qui-quadrado e Análise de Componentes Principais (PCA). Houve efeito significativo (P<0,01) dos indicadores de bemestar animal e de manejo sobre o IQM nas três plantas frigoríficas, sendo que o maior e menor valor foram observados no frigorífico 3 e 1, respectivamente. Os valores de IQM sofreram aumento em todas as plantas quando ocorreram atos de abuso durante o manejo (P<0,01). Houve efeito significativo (P<0,01) de período do dia em IQM, quedas e porteiradas, com aumento nesses indicadores ao longo do dia de trabalho nos três ... / The aim of this study was to evaluate the association between beef cattle, pre-slaughter handling, animal welfare and carcass quality besides testing the applicability of an index that assess the handling quality in slaughterhouses. The study was carried out in three plants licensed to export meat located in the southeast of Brazil, assessing 42,360 cattle. One indicator of animal welfare (falls) and two indicators of handling quality (animals hit by gates and electric prod use) were recorded. These indicators were used to calculate the Handling Quality Scores (HQS). The occurence of bruises and meat pH were also recorded as indicators of carcass and meat quality. The occurence of carcasses with multiple bruises (at least three), meat with high pH (≥ 5,9), and the occurrence of emergency slaughter and acts of abuse (trampled animals and non-ambulatory animals being drove inappropriately) were also recorded. For control purposes the following handling information were also recorded: periods of day, cattle category and different cattle categories being handled together. Data was analyzed by Generalized Linear Models and Pearson Correlation by using the GLM and CORR procedures of SAS program. It was also performed Chi-squared test and Principal Components Analysis (PCA). There was a significant effect (P<0.01) of animal welfare indicators and handling indicators on the HQS in the three plants. The highest and lowest HQS values were observed in plants 3 and 1, respectively. HQS values increased in all plants when acts of abuse occurred during handling (P<0.01). It was also observed an effect (P<0.01) of the periods of day with increasing values of HQS and frequency of falls, electric prod use and animals hit by gates throughout the working day in all plants.
