CHEMOMETRIC ANALYSIS OF COMPREHENSIVE TWO-DIMENSIONAL LIQUID CHROMATOGRAPHIC-DIODE ARRAY DETECTION DATA: PEAK RESOLUTION, QUANTIFICATION AND RAPID SCREENINGBailey, Hope P. 09 October 2012 (has links)
This research project sought to explore, compare and develop chemometric methods with the goal of resolving chromatographically overlapped peaks though the use of spectral information gained from the four-way data sets associated with comprehensive two-dimensional liquid chromatography with diode array detection (LC ´ LC-DAD). A chemometric method combining iterative key set factor analysis (IKSFA) and multivariate curve resolution-alternating least squares (MCR-ALS) was developed. In the section of urine data analyzed, over 50 peaks were found, with 18 visually observable and 32 additional compounds found only after application of the chemometric method. Upon successful chemometric resolution of chromatographically overlapped peaks, accurate and precise quantification was then necessary. Of the compared methods for quantification, the manual baseline method was determined to offer the best precisions. Of the 50 found peaks from the urine analysis, 34 were successfully quantified using the manual baseline method with percent relative standard deviations ranging from 0.09 to 16. The accuracy of quantification was then investigated by the analysis of wastewater treatment plant effluent (WWTPE) samples. The chemometrically determined concentration of the unknown phenytoin sample was found to not exhibit a significant difference from the result obtained by the LC-MS/MS reference method, and the precision of the IKSFA-ALS method was better than that of the precision of the LC-MS/MS analysis. Chromatographic factors (data complexity, large dynamic range, retention time shifting, chromatographic and spectral peak overlap and background removal, were all found to affect the quantification results. The last part of this work focused on rapid screening methods that were capable of locating peaks between samples that exhibited significant differences in concentration. The aim here was to reduce the amount of data required to be resolved and quantified to only those peaks that were of interest. This would then reduce the time required to analyze large, complex samples by eliminating the need to first quantify all peaks in a given sample for many different samples. Both the similarity index (SI) method and the Fisher ratio (FR) method were found to fulfill this requirement in a rapid means of screening fifteen wine samples.

A Tale of Two Silos: Collaborative School Facility Planning in Post-Katrina New OrleansMikulak, Lauren 05 August 2010 (has links)
Cities and schools are traditionally planned in separate silos by local governments and school boards. Collaborative school facility planning (CSP) unites these two silos and integrates decision-making by city and school entities. This research addresses the relationship between urban planning and school facility planning in New Orleans, Louisiana, where CSP is particularly important in light of rebuilding efforts since Hurricane Katrina. The researcher examines the extent, challenges, and opportunities for CSP in New Orleans. Based on interviews and recent planning texts, little collaboration existed in the past; but the decommissioning of 52 New Orleans public school facilities presents an unmistakable opportunity for improved CSP in the near future. The broader implications of this thesis suggest that an alternative model for CSP is needed for low-growth, urban communities whose primary concern is not new school construction but old facility closure and reuse.

Making Plans - Telling Stories : Planning in Karlskrona/Sweden 1980 - 2010Walter, Mareile January 2013 (has links)
The aim of this thesis is to understand how a repertoire of municipal planning narratives evolved and how these were used as a means to explain, legitimise and produce change in a city that went through a process of urban transformation. The focus is set on the role of narratives in municipal plans as a mental preparation for change. In order to reach this aim, a framework for narrative analysis is developed that shall facilitate a critical reading of such municipal planning documents as comprehensive plans. This shall help to understand among other things how place and community are constructed. This framework is used to interpret four documents of the municipality of Karlskrona, one introductory guide for new inhabitants from 1980, and three consecutive comprehensive plans, adopted in 1991, 2002 and 2010. In short, the narrative analysis consists of four different ways of reading each respective document. First, more or less coherent narratives are identified in the texts. Second, they are analysed with respect to their literary and rhetoric form, in a way that is inspired by historian and literary theorist Hayden White. A third reading places the documents’ narratives into their historical context. Finally, they are classified as certain narratives of place identity on the basis of a typology developed by sociologist Manuel Castells. He states that identities can be constructed with help of narratives that legitimise the existing societal structures, that stand in opposition to these structures, or that create a new identity out of available resources. Based on these readings, I find that the four documents use very different literary and rhetorical forms and that they construct the place’s identity in ways clearly distinct from each other. They express various moral and political perspectives and convey clearly distinct social norms regarding the role of inhabitants and the municipality. Over the decades, there has been a clear shift of expressed values from those that support a leading role of the (local) state in fostering local development to those that highlight the importance of market actors and market forces. A similar change has occurred from the pronunciation of state responsibility for the inhabitant’s well-being to a greater focus on individual responsibility. This confirms the notion that municipal planning is increasingly influenced by ideas of neoliberal development. It could also be observed that storytelling and a purposeful narrative construction of place identity have become more prominent as instruments of planning. Planning narratives were clearly used to explain and legitimise shifts or persistence in municipal policymaking. Due to this it can be concluded that in the eyes of local policy makers, the municipality seems to have gone through a complete process of urban transformation from being in a state of decline to one of stabilised growth.

Integrated or Comprehensive sharing? : Drivers, enablers and barriers to civilian-military information sharingWaller, Adam January 2016 (has links)
The UN and NATO have implemented separate approaches to civilian-military cooperation. Central in both approaches is a need for information sharing between civilian and military actors. Without shared information, cooperative planning becomes impossible. For military actors secret information makes sharing difficult and for civilian actors, principles hinder close cooperation. Scholars in the field of intelligence study have identified that states and organizations share information if the benefits of such, outweigh costs and risks. This thesis examines institutional differences between the UN mission MINUSMA in Mali and NATO mission ISAF in Afghanistan, in order to better understand how institutional factors, affect sharing of information. With an outset in Rational Choice Institutionalism and by use of Elinor Ostrom’s Institutional Development and Analysis Framework, an analytical instrument is designed. Through inductive review of interviews, first-hand accounts and reports; factors that drive, enable and hinder civilian-military sharing are indicated. The thesis indicates that the institutional framework of MINUSMA forms interdependency between civilian and military actors, while sharing in ISAF was enabled only when common goals were agreed upon. Military and civilian actors, in both MINUSMA and ISAF point to unclear mandates and vague goals as primary barriers to civilian-military sharing.

Faculty of Health Sciences / The purpose of this research was to ascertain and examine the current genetic knowledge of the nurses in their final year of the Diploma in Comprehensive Nursing (General Nursing, Community Nursing, Psychiatric Nursing) and Midwifery.
The research design was an exploratory, descriptive, quantitative survey, using a sample of final year student nurses at two of the three nursing colleges in Gauteng. Data were collected through the use of a self-administered paper-based questionnaire, which was adapted from a questionnaire used for a study to assess the genetic knowledge of general practitioners (Trenton, 2003). The questionnaire was adapted using the broad outline for subject content given in the curriculum for the four-year Diploma in Comprehensive Nursing (General Nursing, Community Health and Psychiatric Nursing) and Midwifery, the South African Nursing Council regulations, and the Department of Health Policy Guidelines. Random sampling was utilised to select two of the three nursing colleges in Gauteng and the total population of final year students was used as the sample. Data were analysed by means of descriptive statistics.
It was concluded through this study that, although nurses showed fairly adequate knowledge in the areas of teratogens and multifactorial inheritance, they showed a lack of knowledge with regard to basic genetic inheritance patterns, genetic conditions, epidemiology and prenatal diagnostic techniques. It can thus be recommended that an expanded component of genetics be included in the basic curriculum of the four year Diploma in Comprehensive Nursing (General, Community, Psychiatric) and Midwifery programme in accordance with programmes initiated in other countries.

Ce qui ne constituait qu’une « sous-culture » pour quelques passionnés, devient un genre de plus en plus légitime. Ce succès, attribué par les journalistes au renouveau créatif des séries américaines, est largement alimenté par la programmation des chaînes françaises. La télévision n’est cependant plus leur seul diffuseur : DVD, streaming et échange de fichiers sur Internet ont largement pris le relais. Ces médias participent désormais à la configuration des séries, incitent à de nouvelles stratégies et invitent les spectateurs à des usages diversifiés.Les chercheurs montrent eux aussi un intérêt croissant pour les séries, les considèrent comme un terrain propice à l’étude des représentations sociales (sociologie), des « objets narratifs » (narratologique) ou encore des « programmes télévisés » (sémio-pragmatique).Alors, qu’est-ce qu’une série ? Dans le but de répondre à cette question, nous avons adopté une conception élargie des séries comme objets culturels médiatiques, configurés par l’ensemble complexe des médiations socio-techniques qui leur donnent forme. What had previously constituted but a small "subculture" was rising to become a legitimate genre in its own right. This success, that journalists attribute to a creative renewal in American series, is largely fed by French television network programming. Television, however, is no longer the sole broadcaster : DVD and streaming and sharing online are now heavily present. Henceforth, these media will now participate in the configuration of series, incite new strategies and invite audiences to diversify their usages.Researchers themselves show an increasing interest in series, considering them a rich field for the study of social representations (sociology), "narrative objects" (narratology) or also "televised programs" (semio-pragmatics).So, what is a series ? In order to answer this question we have adopted a broad conception, regarding them as being objects of media culture configured by a complex ensemble of socio-technical mediations which give them form. To understand series how they construct their culture, we implement a comprehensive study of the practices and points of view of their communication actors (professionals, media and audiences). Through the use of different methods (discourse analysis, comprehensive interviews, portraits and transversal analysis) we outline series, with regard to their mediations, allowing us to discover the complexity of the object and the necessity of reflection upon the methods to employ in their analysis.

A ocorrência do comportamento autolesivo logra índices significativos na população adolescente e pré-adolescente, tornando-se queixa frequentemente observada no contexto escolar e clínico, preocupando a todos os envolvidos. Na atualidade, a Organização Mundial da Saúde (OMS) a categoriza como um problema de saúde pública global, e, portanto, foco de atenção de políticas públicas e sociais prementes dada a gravidade e aumento do risco de tentativa de suicídio no futuro. No Brasil, as pesquisas realizadas ainda são incipientes quando comparadas com a produção internacional, ainda mais no que diz respeito à dinâmica psíquica do jovem que se autolesiona e meios de intervenção. Assim, considerando a perspectiva investigativa atribuída à Psicologia Clínica e a indissociabilidade entre prática e pesquisa, este trabalho buscou contribuir por meio da realização de um estudo de caso único com vistas à análise dos traços de personalidade de uma adolescente, estudante de uma escola pública do Estado de São Paulo, sem diagnóstico psiquiátrico e/ou em atendimento psicológico, além de investigar dados do funcionamento familiar e a relação de tal conduta com a sintomatologia depressiva e ansiosa. Para tanto, o método clínico-qualitativo de cariz psicanalítico foi empregado em consonância com o delineamento de estudo de casos único e o processo Psicodiagnóstico Compreensivo e Interventivo. O Psicodiagnóstico previamente esquematizado contemplou duas etapas: 1) primeira aplicação, que contabilizou sete atendimentos, e, 2) fase de follow-up, com três atendimentos. Em ambas as etapas, abarcou a realização de entrevistas com a responsável, entrevistas com a participante, aplicação do Questionário Desiderativo, Teste de Apercepção Temática Infantil - Figuras Humanas (CATH), Inventário de Depressão Infantil (CDI) e Inventário de Ansiedade de Beck (BAI). Os resultados do caso foram examinados individualmente, em cada etapa, e, posteriormente, em conjunto, com a comparação entre essas duas fases por meio da livre inspeção do material à luz das observações oferecidas pelos instrumentos projetivos e objetivos fundamentados na abordagem psicanalítica psicodinâmica. Assim, com base nos aportes winnicottianos, pode-se observar que, no caso analisado, a conduta autolesiva revelou uma dupla função: impedir o aniquilamento total do Eu, e, concomitantemente, comunicar ao ambiente sobre falhas nos cuidados em fases primitivas, reatualizados na adolescência, na tentativa de resgatar algo ofertado, porém percebido como perdido. Adicionalmente, integrando os achados iniciais com os obtidos na reavaliação de follow-up, a significativa remissão de aspectos dissociados da personalidade da participante e sintomas clínicos após o processo Psicodiagnóstico comporta correlações com o conceito winnicottiano de distúrbio psicossomático, recuperável após a presença de condições ambientais favoráveis, ou ainda com o trânsito da posição kleiniana esquizo-paranóide para a depressiva, denotando a retomada do processo de amadurecimento. Por fim, os dados obtidos permitem sintetizar sobre a importância do ambiente para a restauração do sentimento de confiança e de esperança no caso investigado, corroborando a teoria winnicottiana sobre a subordinação da saúde psíquica ao longo da vida aos cuidados iniciais recebidos / The occurrence of self-injurious behavior achieves significant rates in the adolescent and preadolescent population, becoming a frequent complaint in the school context, causing concern to all involved. Currently, the World Health Organization (WHO) categorizes it as a global public health problem, and therefore, focus attention on pressing public and social policies given the seriousness and increased risk of attempted suicide in the future. In Brazil, the research carried out is still incipient when compared to international production, especially with regard to the psychic dynamics of the self-injuring young person and means of intervention. Thus, considering the research perspective attributed to Clinical Psychology and the indissociability between practice and research, this work sought to contribute through the accomplishment of a single case study with a view to the investigation and analysis of the personality traits of a teenager, student of a school of the State of São Paulo and without psychiatric diagnosis, besides the family functioning and the relation of such conduct with the depressive and anxious symptomatology. The clinical-qualitative psychoanalytic method was used in consonance with the design of a single case study and the Comprehensive and Interventive Psychodiagnostic process. The psychodiagnosis previously planned included two stages: 1) the first application, which counted seven meetings, and 2) the follow-up phase, with three sessions. Interviews with the participant, interviews with the participant, application of the Desiderative Questionnaire, Child Thematic Apperception Test - Human Figures (CAT-H), Infant Depression Inventory (CDI) and Anxiety Inventory of Beck (BAI). The results of the case were examined individually at each stage and then together with the comparison between these two phases through the free inspection of the material in the light of the observations offered by the projective and objective instruments based on the psychoanalytic psychoanalytic approach. Thus, on the basis of Winnicott\'s contributions, it can be observed that, in the case analyzed, self-defeating behavior revealed a double function: to prevent the total annihilation of the ego, and, at the same time, to communicate to the environment about the failures in care, in an attempt to rescue something offered, but perceived as lost. Additionally, integrating the findings with those obtained in the follow-up reassessment, the significant remission of aspects dissociated from the participant\'s personality and clinical symptoms after the Psychodiagnostic process correlates with the winnicottian concept of psychosomatic disorder, recoverable after the presence of favorable environmental conditions , or with the transit from the kleinian position schizopanoid to the depressive, denoting the resumption of the maturation process. Finally, the data obtained allow us to synthesize the importance of the environment for the restoration of the feeling of trust and hope in the case investigated, corroborating the Winnicottian theory about the subordination of the psychic health throughout the life to the initial care received

The Need for Comprehensive LiberalismGoncalves, Eduardo January 2017 (has links)
Thesis advisor: Stephen Hudson / There has been a growing consensus within political philosophy that liberalism is the most rational form of political organization. The arguments in favor of liberalism are theoretical and also based on historical observations. The view of liberalism as a moral conception of the good life, however, seems to betray its original historical purpose, namely, to provide a peaceful political forum despite competing comprehensive doctrines. How can liberalism be a thick moral conviction of its own if it was meant to temper such zero-sum convictions? To pose this question more concretely: If historic wars between strong religious convictions were tempered by provisional liberal ceasefires, could we accept the evolution of liberalism into a strong conviction of its own? This paradoxical development of liberalism in history runs parallel with contemporary philosophical debates. Whether the most proper conception of liberalism is comprehensive, and whether it is legitimized upon such comprehensiveness are both hotly debated. Exploring these historical and philosophical avenues uncover what I think is a need for a conception of comprehensive liberalism. It is beyond my scope here to formulate a new conception of liberalism that decisively settles the debate. I do, however, point to reasons why comprehensive liberalism should be the focus of contemporary efforts. First, I find that history shows a trend of liberalism growing into a positive doctrine of its own. Second, I invoke two famous traditional conceptions, one successful and one failed, which fuel our baseline intuitions of liberalism with comprehensive, not political justifications. Finally, I show that while contemporary philosophy surrounding liberalism developed a political conception, it cannot avoid slipping towards comprehensiveness. A Dor Crônica por Afecções Musculoesqueléticas (DCAME) é uma doença que, além de atingir o sistema osteomuscular, principalmente de mulheres da faixa etária entre 45 a 64 anos, frequentemente acarreta desordens psicológicas, sociais e espirituais e é influenciada por elas, podendo ainda desenvolver grande carga de desgosto para os doentes quando não diagnosticada e tratada adequadamente. Assim, dentre os aspectos citados, destaca-se o Bem-Estar Espiritual (BEE) como uma expressão da espiritualidade prática que, embora recentemente abordada em estudos científicos foi, desde o início da humanidade, apontada como uma dimensão importante no processo saúde-doença, por possibilitar, além de outros benefícios, o restabelecimento mais rápido da saúde. Foi neste contexto que se tornou necessário compreender e refletir sobre os significados do BEE que permearam a vivência de mulheres portadoras de DCAME e as formas pelas quais o BEE foi manifestado por esta população, para facilitar o esclarecimento tanto das causas da dor atribuídas por elas quanto das formas de manejo e dos impactos ocasionados pela dor. Desta forma, este estudo, que foi realizado na Clínica de Dor do Hospital das Clínicas de Ribeirão Preto (CDHCRP) e conduzido de acordo com as normatizações da Comissão Nacional de Ética em Pesquisa (CONEP), utilizou-se de abordagem qualitativa. Neste trabalho, após seleção por amostragem por conveniência, 11 sujeitos foram selecionados e entrevistados individualmente numa sala reservada a este fim, sendo um guia à entrevista um roteiro norteador de entrevista semiestruturada com questões fechadas para identificação e delineamento sociodemográfico e questões abertas para a apreensão dos significados de BEE bem como dos conhecimentos acerca das causas, dos impactos e das formas de alívio da dor. Os dados coletados foram gravados em um aparelho de MP3 e transcritos na íntegra para o início da análise dos dados que foi feita por meio da Análise Temática (AT), uma das técnicas da Análise de Conteúdo (AC) e possibilitaram o surgimento de quatro temas: Significados e Manifestações do BEE; Modelos explicativos para a dor; Formas de manejo da doença e Impactos da dor crônica. A partir destes temas, algumas interpretações acerca do BEE puderam ser destacadas. Relatado como um aspecto humano de difícil definição, o BEE pôde ser manifestado de diferentes maneiras. As formas apresentadas foram o bom relacionamento consigo mesmo, com o outro e com Deus, o desenvolvimento de esperança, da confiança, da fé em si e no tratamento que realizaram, o conforto físico, a ocupação, o lazer, o aprendizado e a empatia no contato com o outro. Neste trabalho, algumas causas manifestadas foram as de origem psicossocial, as sobrecargas físicas e comportamentos que perpetuam o problema, além da dor como resposta a um pecado. Destacadas algumas causas, o trabalho apontou para a utilização das preces como coadjuvante do tratamento médico convencional por todas as entrevistadas e da técnica Reiki. Tendo em vista o exposto, a DCAME mesmo impactando de forma negativa na vida das entrevistadas, desenvolvendo inclusive o medo da perda da autonomia e independência, possibilitou a modificação de suas visões de mundo, sendo fonte de descoberta de valores humanos tais como a coragem, a resiliência, a valorização da vida bem como da empatia pelo sofrimento alheio. Em suma, este estudo destacou a importância da DCAME enquanto doença multidimensional que leva o indivíduo a experimentar necessidades espirituais importantes, sendo o incentivo a busca da integralidade no contexto hospitalar para o correto manejo do problema mais do que uma emergência para a saúde pública. / Chronic Musculoskeletal Pain (CMP) is a disease that, in addition to achieving the musculoskeletal system, especially the women aged 45 to 64 years, often entails psychological, social and spiritual disorders and is influenced by them and may develop big load of grief for patients when not diagnosed and treated properly. Thus, among the aspects mentioned, Spiritual Well-Being (SWB) as an expression of practical spirituality which, although recently has been addressed in scientific studies, since the beginning of mankind, considered as an important dimension in the health-disease process, by allowing, among other benefits, the most rapid restoration of health. In this context, it has become necessary to understand and reflect on the meanings of SWB that permeated the experience of women with CMP and the ways in which the SWB was expressed by this population, both to facilitate the clarification of the causes of pain attributed by them as forms of management and the impacts caused by pain. Thus, this study, which was conducted at the Pain Clinic of the University Hospital of Ribeirão Preto (PCUHRP) and conducted according to the regulations of the National Committee of Ethics in Research (NCER), used a qualitative approach. In this work, after being selected by convenience sampling, 11 subjects were selected and individually interviewed in a room reserved for this purpose, with a guide to interview a guiding semi- structured interview with closed questions and to identify sociodemographic and open design issues for the seizure of SWB meanings and knowledge about the causes, impacts and forms of pain relief. Data were recorded on an MP3 player and transcribed to the top of the data analysis that was done by means of the Thematic Analysis (TA), one of the techniques of Content Analysis (CA) and made possible the emergence of four themes: Meanings and Manifestations of SWB; Explanatory models for pain; Forms of disease management and Impacts of chronic pain. From these themes, some interpretations of the SWB could be highlighted. Reported as a human aspect difficult to define, SWB could be manifested in different ways. The forms submitted were good relationship with oneself, with others and with God, the development of hope, trust, and faith in themselves and who performed the treatment, physical comfort, occupation, leisure, learning and empathy contact with each other. In this work, some causes were expressed psychosocial origin, physical burdens and behaviors that perpetuate the problem, in addition to pain in response to a sin. Highlighted some causes, the paper points out the use of prayer as an adjunct to conventional medical treatment for all interviewees and Reiki technique. In view of the above, the same CMP negatively impacting on the lives of the interviewees, including developing the fear of loss of autonomy and independence, enabled the modification of their worldviews, a source of discovery of human values such as courage, resilience, valuing life and empathy for others\' suffering. In summary, this study highlighted the importance of CMP as multidimensional disease that causes the individual to experience significant spiritual needs, and encouraging the pursuit of comprehensive health care in the hospital setting to the correct handling of the problem more than a public health emergency.
