L'étude des cellules vivantes et la dentine humaine par microscopie confocale Raman / The Study of living cells and human dentin by confocal Raman microscopy

"L'étude des cellules vivantes et la dentine humaine par microscopie confocale Raman" La microscopie confocale Raman est utilisée pour suivre des médicaments et des nanoparticules dans les cellules et dans les tissus durs. La microscopie Raman est non-invasive, ne nécessite aucun marqueur et permet une imagerie à haute résolution. Dans la première partie de l'étude cette méthode est utilisée pour suivre un médicament anticancéreux, le paclitaxel, au sein d'une lignée de cellules cancéreuses vivantes Michigan Cancer Foundation-7 (MCF-7). Les images Raman ont été traitées par un algorithme de partitionnement des données par k-moyennes pour détecter le paclitaxel dans les cellules. La distribution du paclitaxel dans les cellules est vérifiée par le calcul du coefficient de corrélation de Pearson entre le spectre de référence du traitement et les spectres de l'image entière. Le temps progressif de diffusion du paclitaxel dans toute la cellule est observé. Cette observation demande une étude complémentaire sur l'action pharmaceutique du produit, basé sur la liaison rapide de la tubuline libre au paclitaxel cristallisé. L'apoptose dans les cellules a été suivies par partitionnement de données et par corrélation. Le partitionnement de données a été utilisé pour déterminer la position de mitochondries dans les cellules ; le cytochrome C de distribution à l'intérieur des cellules est basé sur l'analyse de corrélation. L'apoptose des cellules est défini par le cytochrome C dans le cytoplasme de diffusion. Le cytochrome C agit comme un déclencheur pour l'activation en cascade des caspases, et sa libération par les mitochondries est un signe d'apoptose. La Co-localisation de cytochrome C est effectuée après incubation de cellules avec une concentration différente de paclitaxel. L'autre produit étudié est le dioxyde de titane. Le titane est largement utilisé pour les matériaux orthopédiques et dentaires implantés dans le corps humain. Il est inévitable que le sang prenne contact avec la surface de l'implant et des nanoparticules. Les nanoparticules de dioxyde de titane ont été suivies en intracellulaire dans les cellules MCF-7 et TERT épithéliales humaines (lignée orale cellulaire de kératinocytes OKF6/TERT-2). La détection des nanoparticules et leur toxicité ont été étudiées en utilisant deux méthodes d'analyse. La microscopie confocale Raman a également été utilisée pour réaliser l'analyse structurale et chimique de la jonction émail-dentine-résine et de la carie dentaire, grâce à une analyse précise des constituants minéraux et organiques. La microscopie Raman associée à des méthodes d'analyse de données ouvre de nouvelles portes pour la recherche en biologie-santé et en particulier en odontologie. / "The Study of living cells and human dentin by confocal Raman microscopy"Confocal Raman microscopy is employed to trace drugs and nanoparticles intracellular and in hard tissues. Raman spectroscopy a non-invasive, label-free and high spatial resolution imaging technique in first part of the study is being used to trace the anticancer drug paclitaxel in living Michigan Cancer Foundation-7 (MCF-7) cells. An analytical method was developed and applied to Raman data acquired. The Raman images were treated by K-mean cluster analysis to detect the drug in cells. Distribution of paclitaxel in cells is verified by calculating the Pearson correlation coefficient between the reference spectrum of the drug and the whole Raman image spectra. A time dependent gradual diffusion of paclitaxel all over the cell is observed suggesting a complementary picture of the pharmaceutical action of this drug based on rapid binding of free tubulin to crystallized paclitaxel. The apoptosis in the cells were followed by post-measurement analysis including K-mean clustering and Pearson correlation coefficient. K-mean clustering was used to determine mitochondria position in cells and cytochrome c distribution inside the cells was based on correlation analysis. Cell apoptosis is defined as cytochrome c diffusion in cytoplasm. Cytochrome c acts as a trigger for the activation of the caspase cascade, and its release from mitochondria is a sign of apoptosis. Co-localization of cytochrome c is done after cell incubation with different concentration of paclitaxel. The other product used was titanium dioxide. Titanium has been widely used for orthopedic and dental implant materials. When biomaterial is implanted into the human body, it is unavoidable that blood will contact the implant surface and nanoparticles. The question is: do these nanoparticles cause toxicity? Titanium dioxide nanoparticles were followed intracellular in MCF-7 cells and TERT epithelial human oral keratinocyte cell line (OKF6/TERT-2). Detection of nanoparticles and their toxicity were studied using two analytical methods. Confocal Raman microscopy were also used to obtain Structural analysis and chemical profile of Enamel – Dentine- Resin and Raman map of decay and sound dentin samples, through accurate analysis of the mineral and organic components. The Raman spectroscopy combined with this novel method developed in this study, will provide accurate finger prints of chemical composition and by post-measurement analysis of the data acquired more information would be obtained, which might open new gates in pharmaceutical and dentistry researches.

Ce travail de thèse est une approche originale de la compréhension des mécanismes physico-chimiques s'opérant à l'interface des restaurations céramo-céramiques par la microscopie confocale Raman. Il est scindé en trois parties. La première partie expose les caractéristiques d'une chape céramique à noyau zircone (Y-TZP) recouverte d'une céramique feldspathique. Les principaux facteurs responsables de la qualité de l'interface ont été recherchés par une revue de littérature. Le principe de la microscopie confocale Raman a été décrit. La seconde partie est la mise au point des méthodes d'analyse spectrale et d'imagerie Raman, en surface et en profondeur, sur matériaux isolés et à l'interface. La répartition des phases cristallines ou amorphes a été estimée de part et d'autre de cette dernière. La composition chimique élémentaire des composants et une ligne de balayage par traversée de l'interface ont été obtenues par spectroscopie dispersive en énergie (EDS). Le troisième chapitre explore les changements chimiques et morphologiques de l'interface sous différents procédés de fabrication : avec ou sans l'utilisation d'un Liner, avec ou sans utilisation d'une cuisson de régénération de l'Y-TZP. Les images Raman ont été traitées par la fonction d'analyse K-Means Cluster. Une cartographie de la distribution des éléments chimiques a été effectuée par EDS. / This thesis is an original approach to the understanding of the physical and chemical mechanisms operating at the interface of core-veneer all-ceramic restorations by confocal Raman microscopy. It is divided into three sections. The first part describes the characteristics of a zirconia core (Y-TZP) layered with feldspathic ceramic. The main factors affecting the interface have been investigated in the literature review. The principle of confocal Raman microscopy is described. The second part describes the development of spectral analysis and Raman imaging methods, on both the surface and at depth, on isolated materials and at the interface. The allocation of crystalline or amorphous phases was estimated around the interface. The elemental chemical analysis of the components and scanning line through the interface were obtained using energy dispersive X-ray spectroscopy (EDS). The third section explores the chemical and morphological changes of the interface under different manufacturing methods : with or without an optional liner material between the two components and with or without the use of a regeneration firing of the Y-TZP core. Raman images were processed by K-Means Cluster analysis function. The elemental distribution around the interface was estimated using EDS.

Photonic approach for the study of dental hard tissues and carious lesion detection / Approche photonique pour l’étude des tissus durs dentaires et la détection des lésions carieuses

Les propriétés photoniques des tissus durs dentaires nous ont permis d’étudier l’email et la dentine a un niveau moléculaire (in vitro) en utilisant des techniques de microscopie optique non linéaires. La microscopie confocale Raman est technique d’imagine de haute résolution permettant d’analyse d’échantillon sans préparation spécifique ni marquage. Cette méthode nous a permis de reconstituer une cartographie de la réticulation du collagène et de la cristallinité au niveau de la jonction émail-dentine et cela avec une résolution spatiale non atteinte jusque-là. Cette analyse chimique de la jonction émail-dentine a permis de redéfinir la largeur de cette zone de transition. Cette largeur est nettement supérieure à celles proposées par les études précédentes. Par ailleurs, l’étude portant sur les changements de fluorescence intrinsèque entre les tissues dentaires sains et cariés suggèrent l’implication de la protoporphyrin IX et de la pentosidine dans l’expression de la fluorescence rouge des tissus cariés. La microscopie multiphotonique quant à elle nous a permis de détecter la lésion carieuse et de suivre son développement en utilisant la génération de seconde harmonique (SHG) et la fluorescence par excitation à deux photons (2PEF). Nos études ont démontré la validité du ratio SHG/2PEF comme paramètre fiable pour la détection de la lésion carieuse. Les études proposées par cette thèse montrent le potentiel des propriétés photoniques de l’émail et de la dentine en utilisant les microscopies Raman et multiphotoniques dans l’étude de ces tissus au niveau moléculaire. Cela offre de nouvelles perspectives en recherche et en applications cliniques. / Photonic properties of dental hard tissues allowed us to proceed to in vitro analysis of enamel and dentin on a molecular level. Confocal Raman microscopy has been used to produce a mapping of collagen cross-link and crystallinity of human dentin–enamel junction (DEJ) with a spatial resolution not achieved up to now. The method is a non-invasive, label-free and a high spatial resolution imaging technique. This chemical analysis of DEJ led us to redefine a wider width of this transition zone and advance our understanding of dental histology. A study on the intrinsic fluorescence changes of sound and carious tissues using conventional fluorescence microscopy suggests the involvement of protoporphyrin IX and pentosidine in the fluorescence red-shift observed in carious tissues. Multiphoton microscopy allowed to detect nonlinear optical signal changes during caries process using second harmonic generation (SHG) and two-photon excitation fluorescence (2PEF). Our studies led us to propose the ratio SHG/2PEF as valuable parameter to monitor caries lesion. Collectively, advances described in this thesis show the potential of photonic properties of enamel and dentin using Raman and multiphoton microcopies for molecular investigations on sound as much as on carious tissues. It opens new perspective in dental research and clinical applications.

Xerogel de pentóxido de vanádio: novas abordagens na caracterização e aplicações / Vanadium pentoxide: new insights in their characterization and applications

Um estudo refinado dos filmes derivados do gel de pentóxido de vanádio foi realizado por meio de técnicas espectroscópicas e microscópicas visando compreender melhor as características deste material na escala microscópica. Foi demonstrado que o gel de pentóxido de vanádio é uma suspensão de nanopartículas anisotrópicas que se mantêm estáveis mesmo em altas concentrações. As propriedades estruturais e espectroscópicas destas nanopartículas foram revistas e discutidas no estado da arte. Em paralelo foi possível investigar fenômenos como a transição de fase induzida por laser e desenvolver aplicações tecnológicas em áreas como litografia e sensoriamento de gases. Ficou destacada nesta tese o grande potencial de investigação da microscopia Raman confocal na área de nanomateriais. / A refined study of films derived from vanadium pentoxide gel was carried out based on spectroscopic and microscopic techniques, aiming a better understanding of this material on a microscopic scale. It was demonstrated that the vanadium pentoxide gel consists of a suspension of anisotropic nanoparticles which remains stable even at high concentrations. The structural and spectroscopic properties of such nanoparticles were reviewed and discussed, providing new insights at the state of the art. It was also investigated the occurrence of some relevant phenomena, such as the laser induced phase transition, and to develop new technological applications in areas such as gas sensing and lithography. The collection of results described in thesis highlighted a great research potential of confocal Raman microscopy, in the field of nanomaterials.

Caractérisation des propriétés structurales et mécaniques des composantes pariétales du bois à l'échelle du tissu / Mechanical and structural properties characterization of wood cell wall components at the tissue level

Les bois sont une combinaison complexe de très fortes hétérogénéités multi-échelles, avec une trame de fond par espèce qui caractérise ses trois plans d'anisotropie. Les propriétés macroscopiques des bois dépendent de l'ensemble des échelles inférieures d'hétérogénéités : les cernes de croissance, les cellules, ainsi que l'organisation et la concentration des composés chimiques des parois cellulaires. Alors que l'effet des microfibrilles de cellulose cristalline de la phase solide sur les propriétés macroscopiques a été étudié, l'un des défis actuels consiste à évaluer le déterminisme des arrangements moléculaires au sein des parois sur les propriétés des cellules au sein d'un tissu. Dans ce contexte, le présent travail de thèse se développe selon deux axes : (i) La collecte d'informations sur les organisations pariétales à travers la diversité des cellules, et (ii) la caractérisation des propriétés micro-mécaniques à l'échelle du tissu, avec la prise en compte des échelles inférieures d'hétérogénéités. L'essentiel des descriptions sub-pariétales a été réalisé à l'aide d'un micro-spectromètre confocal Raman, et d'outils d'analyses dédiés qui ont été développés dans le cadre de cette étude. Les résultats obtenus ont par exemple permis de caractériser la paroi d'une cellule en formation (thylle), l'installation de la paroi secondaire dans des fibres, l'influence de stimuli environnementaux sur l'organisation sub-pariétale, ou encore le caractère continu de l'organisation spatiale des microfibrilles dans l'épaisseur des parois de différents types de cellules matures. La caractérisation mécanique a été réalisée selon la direction longitudinale des cellules, dans des essais de traction sur des éprouvettes de très faibles dimensions (épaisseur inférieure à 100 µm). La mise en évidence des phénomènes complexes en jeu dans le comportement mécanique a fait l'objet d'une étude sur la variation des propriétés structurales et mécaniques à l'intérieur d'un cerne. Les différentes phases de la relation contrainte-déformation ont été discutées, et une modélisation simplifiée par homogénéisation multi-échelle a notamment permis de poser un ensemble de pistes de réflexions sur le déterminisme de l'organisation sub-pariétale sur les propriétés mécaniques des cellules au sein d'un tissu. / Woods are a highly complex combination of multi-scale heterogeneities. Macroscopic properties of wood depend on all smaller scales of heterogeneity: growth rings, cells, as well as organization and concentration of cell wall chemical compounds. While the effect of the crystalline elements (microfibrils) of the solid phase on the macroscopic properties has been studied, one of the challenges is to estimate the determinism of the cell-wall molecular arrangements on the cells' properties inside a tissue. In this context, the present study is developed along two lines: (i) The collection of information on cell wall organization across the diversity of cells, and (ii) the characterization of micro-mechanical properties at the tissue level, with the consideration of smaller scales of heterogeneity. Most sub-cell wall descriptions were performed using a confocal Raman micro-spectrometer, and tools of analysis that were developed in this study. For example, obtained results have highlighted the cell wall of a living cell (thyllose), the cell wall formation in poplar fibers close to the cambial region, the impact of environmental stimuli on sub-cell wall organization, or else the continuous variation of the spatial organization of crystalline compounds in wall of different mature cells. The mechanical characterization was realized in the longitudinal direction on small samples (thickness below 100µm). The understanding of complex phenomena involved in the mechanical behavior was approached through a study on the variation of structural and mechanical properties within a ring. The different parts of the stress-strain curve have been discussed, and a simplified homogenization model has help to highlighted the determinism of sub-cell wall organization on the mechanical properties of cells at the tissue level.

Conceitos supramoleculares e morfologia interfacial em células solares de TiO2 / Supramolecular concepts and interfacial morphology in TiO2 solar cells

Esta tese tem como objetivo contribuir para o conhecimento e desenvolvimento das células solares sensibilizadas por corantes, através da elaboração de novas espécies supramoleculares e de estudos fundamentais de caracterização do filme de TiO2-P25 e da interface TiO2-Corante pela técnica de microscopia Raman confocal. Os estudos de microcopia Raman confocal da distribuição das fases cristalinas rutilo e anatase em filmes de TiO2-P25 (Degussa) mostraram que a fase rutilo está presente em agregados de diferentes tamanhos (de 250 nm a 3 µm) distribuídos na fase anatase e que estes agregados permaneceram mesmo após os processos de preparação da suspensão coloidal. Na presença do corante N3, verificou-se que a irradiação com laser em 532 nm, usando potência acima de 25 mW cm-2, provoca a absorção e posterior combustão do filme interfacial, com drásticas mudanças na morfologia visualizadas por microscopia Raman confocal, revelando a ocorrência da transformação de anatase em rutilo na região de incidência do feixe de laser. Outro ponto investigado nesta tese, foi a síntese e aplicação de novos sensibilizadores supramoleculares em células solares sensibilizadas por corante (DSC), visando diminuir os processos de recombinação e melhor aproveitamento do espectro solar através da estabilização do corante foto-oxidado e da transferência vetorial de elétron/energia. Além dos trabalhos com novas porfirinas supramoleculares, reproduzidas no apêndice, esta tese apresenta um estudo detalhado de uma série, formada por três espécies diméricas, constituídas pelas unidades [Ru(dcbH2)Cl]+ e [Ru(dmb)2Cl]+, unidas por ligantes ponte com dimensões lineares crescentes, isto é: bpy, bpe e bpeb (dcbH2= ácido-2,2\'-bipiridina-4,4\'-dicarboxílico, dmb= 4,4\'-dimetil-2,2\'- bipiridina, bpy= 4,4\'-bipiridina, bpe= trans-1,2-bis(4-piridil)-etileno e bpeb= trans-1,4- bis[2-(piridil)etenil]-benzeno). O rendimento global das DSCs correspondentes cresceram com o aumento do tamanho da ponte, passando de 2,78 % para o corante dim-Ru-bpy, para 2,89 e 3,25 % nos corantes dim-Ru-bpe e dim-Rbpeb, respectivamente. Foi observado que os valores de Jsc e η têm correlação linear com a distância de separação do buraco e da superfície do eletrodo de TiO2, ratificando que o processo de retardação da recombinação de carga, causado pelo aumento da ponte, é um fator muito importante para esta classe de compostos. Os resultados de IPCE mostraram que a unidade [Ru(dmb)2Cl(P)]+ também é responsável pela fotoinjeção de elétrons no TiO2. Assim, a excitação de qualquer uma das unidades, sempre acaba formando a espécie TiO2(e-)-Ru-Ru(h+) no filme interfacial. Os resultados confirmam as expectativas esperadas no planejamento energético vetorial, no qual o HOMO foi centrado na unidade distante da superfície e o LUMO ficou sobre a dcbH2 diretamente ancorada no superfície do TiO2. / This thesis encompasses our efforts to improve the knowledge and contribute to the development of dye-sensitized solar cells, by focusing on supramolecular design of new dyes and on critical aspects of the morphology of the TiO2-P25 and TiO2-Dye interfaces, as probed by confocal Raman microscopy. According to Raman imaging of the crystalline phase distribution of TiO2-P25 (Degussa) rutile was present as aggregates of different sizes (from 250 nm to 3 µm) dispersed in the 25 nm anatase powder, persisting even after applying criterious procedures for generating uniform colloidal suspensions. In addition, the irradiation of TiO2-P25 films containing adsorbed N3 dye, using a 532 nm laser (power over to 25 mW cm-2) led to the absorption and instantaneous combustion of the dye, promoting drastic local changes associated with the anatase to rutile conversion. The design and application of new supramolecular sensitizers in DSC has also been pursued, aiming an improvement of efficiency by slowing down the electron-hole recombination process, while enhancing the light harvesting effects in the visible range by means of vectorial electron/energy transfer. Studies concerning another efficient supramolecular porphyrin dye, have also been performed and published (Appendix). As the main subject, a series of dimeric species have been reported. They were based on the [Ru(dcbH2)Cl]+ and [Ru(dmb)2Cl]+ units connected by linear bridging ligands of increasing lengths, such as: bpy, bpe and bpeb (dcbH2= 2,2\'-bipyridine-4,4\'-dicarboxylic acid, dmb= 4,4\'-dimethyl-2,2\'-bipyridine, bpy= 4,4\'-bipyridine, bpe= trans-1,2-bis(4- pyridyl)-ethylene e bpeb= trans-1,4-bis[2-(4-pyridyl)ethenyl]-benzene). The overall yield of the corresponding DSCs increased with the bridging ligand length, from 2.78 % for dim-Ru-bpy, to 2.89 % and 3.25 % for dim-Ru-bpe and dim-Ru-bpeb, respectively. The electrochemical parameters associated with the short circuit current exhibited a linear correlation with the hole separation distance between the TiO2 electrode and the binuclear dye terminal, confirming that the retardation of charge recombination through the increasing distance is indeed a relevant factor for this series of compounds. The IPCE results indicated that the [Ru(dmb)2Cl(P)]+ unit is also involved in electron transfer, such that the dye excitation always leads the TiO2(e-)-Ru-Ru(h+) species. This conclusion confirmed the success of the supramolecular design and vectorial transfer strategy, in which the HOMO center was placed far away from the surface, but in communication with the LUMO center located at the dcbH2 group anchored on TiO2.

Além dos trabalhos com novas porfirinas supramoleculares, reproduzidas no apêndice, esta tese apresenta um estudo detalhado de uma série, formada por três espécies diméricas, constituídas pelas unidades [Ru(dcbH2)Cl]+ e [Ru(dmb)2Cl]+, unidas por ligantes ponte com dimensões lineares crescentes, isto é: bpy, bpe e bpeb (dcbH2= ácido-2,2\'-bipiridina-4,4\'-dicarboxílico, dmb= 4,4\'-dimetil-2,2\'- bipiridina, bpy= 4,4\'-bipiridina, bpe= trans-1,2-bis(4-piridil)-etileno e bpeb= trans-1,4- bis[2-(piridil)etenil]-benzeno). O rendimento global das DSCs correspondentes cresceram com o aumento do tamanho da ponte, passando de 2,78 % para o corante dim-Ru-bpy, para 2,89 e 3,25 % nos corantes dim-Ru-bpe e dim-Rbpeb, respectivamente. Foi observado que os valores de Jsc e η têm correlação linear com a distância de separação do buraco e da superfície do eletrodo de TiO2, ratificando que o processo de retardação da recombinação de carga, causado pelo aumento da ponte, é um fator muito importante para esta classe de compostos. Os resultados de IPCE mostraram que a unidade [Ru(dmb)2Cl(P)]+ também é responsável pela fotoinjeção de elétrons no TiO2. Assim, a excitação de qualquer uma das unidades, sempre acaba formando a espécie TiO2(e-)-Ru-Ru(h+) no filme interfacial. Os resultados confirmam as expectativas esperadas no planejamento energético vetorial, no qual o HOMO foi centrado na unidade distante da superfície e o LUMO ficou sobre a dcbH2 diretamente ancorada no superfície do TiO2. / This thesis encompasses our efforts to improve the knowledge and contribute to the development of dye-sensitized solar cells, by focusing on supramolecular design of new dyes and on critical aspects of the morphology of the TiO2-P25 and TiO2-Dye interfaces, as probed by confocal Raman microscopy. According to Raman imaging of the crystalline phase distribution of TiO2-P25 (Degussa) rutile was present as aggregates of different sizes (from 250 nm to 3 µm) dispersed in the 25 nm anatase powder, persisting even after applying criterious procedures for generating uniform colloidal suspensions. In addition, the irradiation of TiO2-P25 films containing adsorbed N3 dye, using a 532 nm laser (power over to 25 mW cm-2) led to the absorption and instantaneous combustion of the dye, promoting drastic local changes associated with the anatase to rutile conversion. The design and application of new supramolecular sensitizers in DSC has also been pursued, aiming an improvement of efficiency by slowing down the electron-hole recombination process, while enhancing the light harvesting effects in the visible range by means of vectorial electron/energy transfer. Studies concerning another efficient supramolecular porphyrin dye, have also been performed and published (Appendix). As the main subject, a series of dimeric species have been reported. They were based on the [Ru(dcbH2)Cl]+ and [Ru(dmb)2Cl]+ units connected by linear bridging ligands of increasing lengths, such as: bpy, bpe and bpeb (dcbH2= 2,2\'-bipyridine-4,4\'-dicarboxylic acid, dmb= 4,4\'-dimethyl-2,2\'-bipyridine, bpy= 4,4\'-bipyridine, bpe= trans-1,2-bis(4- pyridyl)-ethylene e bpeb= trans-1,4-bis[2-(4-pyridyl)ethenyl]-benzene). The overall yield of the corresponding DSCs increased with the bridging ligand length, from 2.78 % for dim-Ru-bpy, to 2.89 % and 3.25 % for dim-Ru-bpe and dim-Ru-bpeb, respectively. The electrochemical parameters associated with the short circuit current exhibited a linear correlation with the hole separation distance between the TiO2 electrode and the binuclear dye terminal, confirming that the retardation of charge recombination through the increasing distance is indeed a relevant factor for this series of compounds. The IPCE results indicated that the [Ru(dmb)2Cl(P)]+ unit is also involved in electron transfer, such that the dye excitation always leads the TiO2(e-)-Ru-Ru(h+) species. This conclusion confirmed the success of the supramolecular design and vectorial transfer strategy, in which the HOMO center was placed far away from the surface, but in communication with the LUMO center located at the dcbH2 group anchored on TiO2.

L'objet de cette thèse est l'étude de la pénétration des actifs cosmétiques dans la peau. Les axes d'investigation principaux ont concerné l'influence des propriétés physicochimiques des actifs et des ingrédients de la formule sur les mécanismes de pénétration. Les actifs cosmétiques choisis sont le rétinol, actif lipophile, et la caféine, actif hydrophile. Les formulations investiguées sont des émulsions de type huile dans eau, comparées aux solutions de tensioactifs correspondantes. Trois huiles cosmétiques ont été utilisées: Butylène glycol de cocoate, Octyldodecyl myristate et la Paraffine liquide, stabilisées en émulsion avec des tensioactifs ester de polyéthylène glycol (PEG20 et PEG6) possédant des longueurs de chaîne carbonées variables (C8, C12, C18 et C18:1). La pénétration percutanée a été mesurée quantitativement en utilisant la méthode des cellules de diffusion de Franz en fonctionnement statique et dynamique et qualitativement par la microscopie confocale Raman. Avec cette combinaison de techniques analytiques, il est possible, de mesurer la pénétration et d’évaluer l'impact de chaque composant de la formulation sur la pénétration cutanée d'un actif. Une corrélation a pu être établie entre l’effet fluidifiant d’une huile et l’augmentation de la pénétration du rétinol. Par ailleurs les tensioactifs, même s’ils ont montré un effet moindre en terme de fluidification conduisent également à une augmentation de la pénétration en raison d’une variation du coefficient de partage de l’actif entre la formule et la peau. Concernant la caféine, l’influence de la structure des tensioactifs et en particulier de la longueur de chaîne carbonée a été mise en évidence / The purpose of this thesis is to study the penetration of cosmetics actives into the skin. The main lines of investigation concerned the influence of actives and formulation components physicochemical properties on the penetration mechanisms. The selected cosmetic actives are retinol, lipophilic, and caffeine, hydrophilic. The investigated formulations are oil in water emulsions, compared to their corresponding surfactant solutions. Three cosmetic oils were used: Butylene glycol cocoate, Octyldodecyl myristate and liquid paraffin. Emulsions are stabilized with polyethylene glycol ester surfactants (PEG20 and PEG6) having variable carbon chain lengths (C8, C12, C18 and C18: 1). Percutaneous penetration was measured quantitatively using Franz diffusion cells in a static and dynamic way and qualitatively by confocal Raman microscopy. With this combination of analytical techniques, it is possible to measure the penetration and evaluate the impact of each formulation component on skin penetration of an active. A correlation could be established between the fluidizing effect of an oil and the increase in retinol penetration. Moreover, the surfactants, although they showed less effect in terms of fluidizing also lead to an increase in penetration due to a variation of the active partition coefficient between the formula and the skin. In vitro enamel subsurface lesions : characterization and treatment / Lésions carieuses de l'émail in vitro : caractérisation et traitement

Les taches blanches sont liées à l'hypominéralisation sous la surface de l'émail et sont la 1ère étape du développement de la carie dentaire. La détection précoce des caries dentaires naissantes avant qu'elles n'atteignent le stade de la cavitation offre une opportunité pour des soins dentaires efficaces. Pour réaliser les objectifs de cette étude qui sont de renforcer l'idiome des soins minimalement invasifs, nous avons identifié un modèle de cycle de pH modifié qui permet d’imiter les conditions intra-buccales qui conduisent à la formation de lésions de type tache blanche, dans un court laps de temps. Des techniques optiques non invasives, comme la microscopie Raman confocale et la microscopie multiphotonique, ont été utilisées dans cette étude pour détecter de petits changements dans la composition chimique de l'émail in vitro.De plus, la technique de nano-indentation a été appliquée pour étudier les changements dans les propriétés mécaniques de l'émail et les relier à ceux affectant sa composition chimique après l'induction des caries, afin d‘ajouter de la spécificité chimique-mécanique aux informations sur les lésions sous-surface de l'émail. Les résultats obtenus démontrent un grand potentiel pour les techniques examinées, fournissant une base pour des applications intéressantes dans le diagnostic clinique de différentes pathologies dentaires. Pour résoudre le problème du traitement des lésions primitives sans intervention chirurgicale, l'efficacité de la crème GC Tooth Mousse et du dentifrice contenant de la nano-hydroxyapatite «KAREX» dans l'amélioration de la reminéralisation de l'émail déminéralisé par la localisation du phosphate de calcium amorphe à la surface de la dent a été inspectée. L'étude a indiqué le manque de preuves fiables soutenant l'efficacité des agents reminéralisants dans le traitement des taches blanches. Ce travail doit être poursuivi par d'autres études in-vitro et par des études cliniques. / White spot lesion is the subsurface hypomineralization of enamel indicating the 1st stage of dental caries development. Early detection of incipient dental caries before it reaches the stage of cavitation offers an opportunity for effective dental care. The objectives of this study were to strengthen the idiom of minimally invasive treatment. In order to achieve the specified goals; we have identified a modified pH cycling model that can mimic the intraoral conditions leading to white spot lesions formation in a short time. In addition, non-invasive optical techniques, such as confocal Raman microscopy and multiphoton microscopy were used in this study to detect small changes in the enamel chemical composition in vitro.Furthermore, nano-indentation technique was used to detect the changes in the mechanical properties of enamel and relate them to those affecting its chemical composition after caries induction in order to add chemico-mechanical specificity in providing important information about subsurface lesions in enamel. The obtained results demonstrate a great potential for the examined techniques, providing a basis for interesting applications in the clinical diagnosis of various pathological conditions in dentistry. To treatment the incipient carious lesions non-invasively, the effectiveness of GC Tooth Mousse cream and nHA containing-dentifrice "KAREX" in the remineralization of demineralized enamel through localizing amorphous calcium phosphate at tooth surface has been inspected. The study indicated a lack of reliable evidence supporting the efficacy of remineralizing agents in the treatment of white spot lesions. Within the limitations of this study, further laboratory studies together with clinical research are therefore required to increase the available knowledge on this prevalent subject.
