Étude multi-isotopique d’une population urbaine de Montréal (Saint-Antoine, 1799-1854) : adaptation et pratiques alimentaires parmi les individus locaux et migrants

Par l’intermédiaire d’analyses isotopiques de l’émail et de la dentine (δ13C, δ15N, δ18O), l’étude vise à reconstituer pour la première fois les schémas de mobilité et les pratiques alimentaires au cours de l’enfance d'une population inhumée au cimetière catholique du faubourg de Saint-Antoine (1799 -1854) (N=24). Ce dernier est un site clé, lié à la transition de la ville de Montréal vers l'industrialisation et reflète une croissance démographique exponentielle reliée au flux migratoire accru en provenance surtout des îles Britanniques (Écosse, Angleterre, Irlande), des États-Unis, du milieu rural canadien et d’autres régions possiblement (Afrique, Caraïbes). De plus, six individus, victimes du naufrage du Carrick of Whitehaven sur la côte gaspésienne (Cap-des-Rosiers, 1847), ont fourni des données sur les migrants irlandais de première génération. Pour tous les individus, un seul échantillon a été prélevé pour l’émail (couronne inférieure) et la dentine (racine/apex) sur une prémolaire ou deuxième molaire permanente pour couvrir une période de vie entre 2 ans et l’adolescence. En plus, pour quatre individus, deux méthodes de micro-échantillonnage en séquence de la dentine (micro-poinçon, micro-tranche) ont été réalisées pour comparer leur efficacité et obtenir des données plus détaillées. Les résultats généraux suggèrent une diversité alimentaire au cours de l’enfance basée sur l’origine des personnes : le régime alimentaire à Montréal était principalement basé sur les ressources en C3, avec l’ajout de ressources en C4 pour les locaux et les gens originaires du Haut-Canada; et l’apport en aliments carnés était plus important chez les immigrants des îles Britanniques ou des États-Unis. Ces faits concordent avec les sources historiques qui mentionnent un afflux de migrants et une alimentation diversifiée selon les groupes culturels à Montréal. Les données isotopiques observées pour Cap-des-Rosiers sont beaucoup plus homogènes qu’à Saint-Antoine, car elles reflètent un événement migratoire ponctuel. Comme trois individus de Saint-Antoine ont des résultats isotopiques très similaires à ceux de Cap-des-Rosiers, il est proposé qu’il s’agisse d’Irlandais voire de personnes ayant un parcours de vie similaire à des migrants venus des îles Britanniques. Finalement, l’un d’entre eux (9M-S2) a révélé grâce aux micro-tranches (technique la plus adaptée pour obtenir des variations temporelles très fines) une augmentation du δ15N vers l’âge de 4 ans, possiblement relié à un stress physiologique (ex. famine). / By using isotopic analyses of enamel and dentin (δ 13C, δ 15N, δ 18O), this study aims to reconstruct for the first time the mobility patterns and dietary practices during infancy of a population buried in the Saint Antoine Catholic cemetery (1799 -1854) (N=24). The latter is a key site linked to the city of Montreal's transition to industrialization and reflects a period of exponential population growth linked to increased migration especially from the British Isles (Scotland, England, Ireland), the United States, rural Canada, and to a few other regions possibly (Africa, Caribbean). Moreover, six individuals, victims of the shipwreck of the Carrick of Whitehaven on the Gaspé Coast (Capdes-Rosiers, 1847), provided data on first-generation Irish migrants. For all individuals, a single sample was taken from enamel (lower crown) and dentin (root/apex) of one permanent premolar or second molar to cover a life span between 2 years and the adolescence. In addition, for four individuals, two methods of sequential micro-sampling of dentin (micro-punch, micro-slice) were carried out to compare their effectiveness and obtain detailed data. Overall results suggest dietary diversity during childhood based on people's origins: the diet in Montreal was mainly based on C3 resources, with the addition of C4 resources for locals and people from Upper Canada (Ontario); and meat intake was higher among immigrants from the British Isles or the United-States. These facts are consistent with historical sources that mention an influx of migrants and a diversified diet among the cultural groups of Montreal. The isotopic data observed for Cap-des-Rosiers are much more homogeneous than for Saint-Antoine, reflecting one tragic migratory event. As three individuals from Saint-Antoine have isotopic results very similar to those from Cap-des-Rosiers, it is proposed that they are Irish or people with a life history similar to migrants from the British Isles. Finally, one of them (9M-S2) revealed an increase in δ 15N around the age of 4 years old, possibly linked to physiological stress (e.g. famine) thanks to micro-slicing (the most suitable technique for obtaining very detailed temporal variations).

BP) : a whole crown perspective in the Aquitaine Basin, southwest France, and its margins

À partir de la fin du Pléistocène, une réduction de la taille des dents humaines et une simplification morphologique ont été observées et débattues en lien avec des changements culturels et environnementaux. Suite à de nouvelles découvertes et à la révision des contextes archéologiques de certains gisements, une réévaluation de la nature des variations de plus de 1900 couronnes dentaires est proposée pour 176 individus de la fin du Paléolithique, du Mésolithique et du début du Néolithique provenant du Bassin aquitain et de ses marges. Particulièrement, les variations de la structure interne (épaisseur de l’émail, proportions des tissus dentaires, morphologie de la jonction émail-dentine) ont été évaluées de manière non invasive grâce aux méthodes d’imagerie 3D (microtomographie) et de morphométrie géométrique afin de caractériser et d’interpréter l’évolution des couronnes dentaires selon une approche intégrée. Les résultats des analyses morphométriques montrent une discontinuité entre les populations de la fin du Pléistocène et celles du début de l’Holocène. Une réduction des dimensions externes, des épaisseurs de l’émail et des proportions des tissus est mesurée entre la fin du Paléolithique et le Mésolithique, alors que des différences majeures dans les types d’usure et la distribution de l’émail sont observées entre le Mésolithique et le Néolithique. Ces données suggèrent que les modifications induites par les changements environnementaux de l’Holocène ont eu un impact plus important sur la réduction dentaire dans les populations humaines et que les changements culturels néolithiques ont surtout affecté la distribution de l’émail. Enfin, une corrélation entre le type d’usure occlusale et la distribution de l’épaisseur de l’émail a été mise en évidence et associée à des changements de régime alimentaire. En particulier, l’épaisseur de l’émail peut évoluer rapidement comme une réponse sélective aux changements fonctionnels dans la biomécanique de la mastication. / Since the Late Pleistocene, a reduction in size and a morphological simplification of human teeth have been observed and arguably linked to cultural and environmental changes. Following new discoveries along with the revision of key archaeological contexts, a re-assessment of the nature of crown variations on more than 1900 teeth is proposed for 176 Late Paleolithic, Mesolithic and Early Neolithic individuals from the Aquitaine Basin and its margins. In particular, a non-invasive assessment of internal tooth structure variability (enamel thickness, dental tissue proportions, enamel-dentine junction morphology) has been performed using 3D imaging methods (microtomography) and geometric morphometrics in order to characterize and interpret dental evolution from a whole crown perspective. Results from the morphometric analyses show a discontinuity between Late Pleistocene and Early Holocene populations. External dimensions, enamel thicknesses and tissue proportions are reduced in Mesolithic individuals compared to those of the Late Paleolithic, while major differences are observed in occlusal wear patterns and enamel distribution between Mesolithic and Early Neolithic samples. These data suggest that environmentally-driven modifications during the Early Holocene had a major impact on dental reduction in human populations and that Neolithic cultural changes had mostly affected enamel distribution. Finally, a correlation between occlusal wear pattern and enamel thickness distribution is observed and associated with dietary changes. In particular, enamel thickness may have rapidly evolved as a selective response to functional changes in masticatory biomechanics.

A exposição a baixos níveis de chumbo é associada a desordens cognitivas e neurológicas em crianças. Foi descrito um aumento dos níveis de chumbo no sangue de crianças que vivem em comunidades que recebem água fluoretada. Este estudo testou se flúor a 100 ppm na forma de ácido fluorsilícico na água induziria aumento na concentração de chumbo no sangue e tecidos calcificados de ratos Wistar expostos a baixos níveis de chumbo desde a idade gestacional. Ratas foram separadas em quatro grupos: controle e grupos que receberam água que continham 100 mg/L de fluoreto (F), 30 mg/L de chumbo (Pb), ou 100 mg/L de F e 30 mg/L de Pb (F+Pb), desde uma semana antes do acasalamento até que os filhotes completassem 81 dias. Todos os animais foram pesados ao longo do experimento. Sangue e tecidos calcificados foram coletados com 81 dias para análise de chumbo, flúor e fósforo em esmalte, dentina, osso total, osso superficial e sangue. Chumbo foi determinado por ICP-MS (Inductively Coupled Plasma Mass Spectrometry). Flúor foi medido através de elétrodo íon-específico e fósforo foi determinado por reação colorimétrica. As concentrações de chumbo triplicaram no grupo F+Pb (76.7±11.0 µg/dL) em comparação ao grupo Pb (22.6±8.5 µg/dL)(p <0.001), com a mesma tendência observada em todos os tecidos calcificados analisados (p <0.001 para todas as comparações). No esmalte, as concentrações de chumbo analisadas foram 2.5 vezes mais altas no grupo F+Pb em comparação ao grupo Pb (4,369±1,353 µg/g versus 1,768±1,892 µg/g). Na dentina, a concentração de chumbo encontrada no grupo F+Pb era 8.5±2.0 µg/g versus 4.9±1.7 µg/g no grupo Pb. Na superfície óssea, a concentração de chumbo encontrada no grupo F+Pb era 3.1 vezes maior do que as determinadas no grupo Pb, com 28.0±10.6 e 9.0±3.7 µg/g nos grupos F+Pb e Pb, respectivamente. No osso total, os valores de chumbo dobraram no grupo F+Pb (14.2±2.6 µg/g) em comparação com o grupo Pb (6.8±1.7 µg/g). Os valores de chumbo estavam abaixo do limite de detecção na maioria das amostras dos grupos Controle e F. As concentrações de flúor aumentaram em ambos os grupos expostos a flúor (F e F+Pb), com diferenças estatisticamente significantes dos grupos controle e Pb, mas nenhuma diferença foi encontrada nas concentrações de F entre os grupos F e F+Pb em quaisquer dos tecidos calcificados testados. Em conclusão, este estudo mostra um aumento nas concentrações de chumbo no sangue total, esmalte, dentina, superfície óssea e osso total em ratas com 81 dias expostas ao flúor e chumbo desde o período pré-natal. / Low-level lead exposure is linked to cognitive and neurological disorders in children. An increased risk for higher blood lead levels was described for children living in communities that receive fluoridated-water. This study tested whether water fluoride would induce increases in the blood and calcified tissue lead concentrations in Wistar rats exposed to low lead levels since gestational age. Female rats were allocated in four groups: control, and 3 groups that received water containing 100 mg/L of fluoride (F), 30 mg/L of lead (Pb), or 100 mg/L of F and 30 mg/L of Pb (F+Pb). Females mated and delivered their pups receiving the same water treatment. Female pups were maintained on the same water regimen until day 81. Lead was determined by ICP-MS (Inductively Coupled Plasma Mass Spectrometry), and fluoride was measured by ion-specific electrode in the whole blood, superficial enamel, dentine, surface bane, and whole bane. Mean whole blood lead concentrations triplicated in the F+Pb group (76.7±11.0 µg/dL) in comparison to the Pb group (22.6±8.5 µg/dL)(p < 0.001), with the same trend observed in all calcified tissues analyzed (p < 0.001 for all comparisons). In the enamel, mean lead concentrations were 2.5 times higher in the F+Pb group compared with the Pb group (4,369±1,353 µg/g versus 1,768±892 µg/g). Fluoride concentrations were increased in both groups exposed to fluoride (F and F+Pb), with statistically significant differences from the control and Pb groups, but no differences in the F concentrations were found between the F and F+Pb group in any of the calcified tissues tested. In conclusion, this study shows a fluoride-induced increase in the concentrations of lead of whole blood, enamel, dentine, surface bane and whole bane of 81-day female rats exposed to lead since gestational age, suggesting that a biological effect not recognized so far may underlie the epidemiological association between increased blood lead levels in children living in water-fluoridated communities.

Analyse d'une interface adhérente dentine/titane en fonction de différents matériaux d'assemblage / Study of a dentin adhesive / titanium interface mediated by different resin luting cement

Le fil directeur durant cette période de recherche a été l’étude d’une interface dentine/titane assemblée par collage. L’objectif était d’optimiser et de simplifier des étapes du protocole d’assemblage, d’améliorer et d’étudier les performances de l’interface adhérente. Dans ce travail, les interfaces ont été testées in vitro par cisaillement.Cette étude a comporté trois volets :Le but de la première étude a été d’évaluer un assemblage par ciment verre ionomère (CVI), le Fuji Plus®, appliqué en association avec des adhésifs auto-mordançants sur la dentine. Trois des combinaisons « système auto-mordançant/Fuji Plus® » ont été appliquées et les résultats obtenus montrent des améliorations significatives entre 9% et 44% par rapport aux valeurs obtenues sur des éprouvettes de témoin.Le second volet expérimental a eu pour objectif de tester la compatibilité entre cinq systèmes adhésifs auto-mordançants (SAM) et deux composites d’assemblage (CA). Les SAM1 se sont révélés totalement incompatibles avec les CA chémopolymérisés ou duals. Inversement, les SAM2 étaient compatibles avec des CA chémopolymérisants ou duals. Une corrélation positive a été trouvée entre le pH des SAM ou les résines testées et les données de cisaillement.La dernière étude a été dédiée à l’évaluation de l'effet du stockage dans l’eau sur la résistance à la flexion (σf) et sur la résistance au cisaillement (RC) d’une interface dentine/titane assemblée par quatre résines adhésives auto-mordançantes (MRAM). Cette étude a montré que le stockage dans l'eau durant 60 jours a affecté négativement la résistance à la flexion de certains matériaux. L'adhésion dentine/titane offerte par Rely X® et G-Cem® était significativement plus élevée que celle de Maxcem® et SmartCem2®. / The aim of this work was to study a bonding dentin/titanium interface. The objective was to optimize and simplify the steps of the bonding protocol, improve and know well the performance of the adhesive interface. In this work, all interfaces have been tested in vitro by shearing.This study included three parts :The purpose of the first study was to evaluate an assemblage by glass ionomer cement (GIC), the Fuji plus®, applied in combination with self-etch bonding system on dentin. Three combinations "system self-etching / Fuji Plus®" were applied and the results obtained by test showed significant improvements from 9% to 44% higher than the values obtained on control specimens.The second experimental part was designed to test the compatibility between five self-etch bonding systems (SEBS) and two resin luting cements (RLC). Significant differences of bond strength in shear were observed according to both the curing mode of the RLC and the adhesive. The SEBS1 proved totally incompatible with chemical or dual cured RLC. Conversely, two-steps SEBS were compatible with RLC chemical/dual cured RLC. A positive correlation was found between the pH of the SEBS or the tested resins and the shear data.The latest study was devoted to evaluate the effect of the storage condition on flexural strength (σf) and shear bond strength (SBS) of a dentin/titanium interface assembled by four self-etch adhesive resin (SEARC). This study showed that the storage in water for 60 days has negatively affected the bending resistance of certain materials. The dentin/titanium adherence offered by Rely X® and G-Cem® was significantly higher than that of Maxcem® and SmartCem2®.

An isotopic and historical study of diet and migration during the great Irish Potato Famine (1845-1852) : high-resolution carbon and nitrogen isotope profiling of teeth to investigate migration and short-term dietary change at the Union workhouse, Kilkenny and Lukin street, London

Historical evidence from contemporary documents established that Irish migrants to London during the Great Irish Famine (1845-1852) were likely to come from low socio-economic groups in south-west Ireland, and has characterised mid-19th-century health status and living conditions in both locations. Using samples from 119 individuals from the Catholic cemetery at Lukin Street, London (1843-1854) and 20 from the Union Workhouse Famine cemetery, Kilkenny, Ireland (1847-51), mean bone collagen isotope values were established for the well-documented Irish pre-Famine potato-based diet (δ¹⁵N 10.6‰, δ¹³C -19.1‰), and the diet of contemporaneous Londoners (δ¹⁵N 12.6‰, δ¹³C -19.1‰). The introduction of maize as a short-term Famine relief food was identified in three Kilkenny juveniles with bone collagen δ¹³C above -17‰, and incremental dentine collagen demonstrating temporal changes in δ¹³C consistent with dietary change from C3 to C4 plants. Bone collagen values for two Lukin Street individuals were consistent with high marine protein consumption. Techniques developed in this study to sample increments of dentine representing nine months or less of life have improved temporal resolution not only for migration events but also short-term dietary changes and physiological status during childhood. Combining epigraphic, osteological and archaeological evidence, individual 'lifeways' have been constructed using isotope data and provide insights into the connection between health, diet and skeletal manifestations of deprivation during childhood and adolescence. New models are investigated for examining maternal and infant health using dentine collagen increments formed in utero and combining dentine and bone collagen values to explore the effects of nutritional stress on bone turnover.

In conclusion, this study shows a fluoride-induced increase in the concentrations of lead of whole blood, enamel, dentine, surface bane and whole bane of 81-day female rats exposed to lead since gestational age, suggesting that a biological effect not recognized so far may underlie the epidemiological association between increased blood lead levels in children living in water-fluoridated communities.

No. of bitstreams: 1 FELIPE GUTIERREZ REYES.pdf: 2973646 bytes, checksum: 0e8c702449c97c861e9f0314e3b69e2f (MD5) Previous issue date: 2016-02-23 / Objectives: This study evaluated the effect of addition of copper nanoparticles at different concentrations into a simplified etch-and-rinse (ER) adhesive system (Ambar [AM]) on antimicrobial activity (AMA), degree of conversion of adhesives discs (DC-d) and dentin-resin interface (DC-i), the ultimate tensile strength (UTS), 28-day cumulative water sorption (WS), solubility (So) and copper release (CR) as well as immediate (IM) and 1-year (1Y) resin–dentine bond strength (μTBS) and nanoleakage (NL). Methods: Seven experimental adhesive systems were formulated according to the addition of copper nanoparticles (0 [control], 0.0075, 0.015, 0.06, 0.1, 0.5 and 1%) in AM adhesive system. We tested the antimicrobial activity of synthesized adhesives against Streptococcus mutans using agar diffusion assays. For DC-d, specimens were constructed and tested after 24 h for FTIR and micro-Raman spectroscopy. For UTS, specimens were tested after 24 h and 28 days. For WS and So, after specimens build-up, they were stored in water and the properties measured for 28 days. For CR, specimens were stored in 2% nitric acid solution and the properties were measured for 28 days. The occlusal enamel of thirty-five molars was removed and adhesives were applied to dentine surface after 37% phosphoric acid etching. After composite resin build-ups, specimens were longitudinally sectioned to obtain resin–dentine bonded sticks (0.8 mm2). Specimens were tested in tension at 0.5 mm/min in IM or 1Y (μTBS). For NL, 2 bonded sticks from each tooth were prepared and analyzed under SEM in IM or 1Y. For DC-i, 2 bonded sticks were prepared and analyzed under micro-Raman spectroscopy. The μTBS and NL data of each adhesive were subjected to two-way repeated measures ANOVA. For UTS, WS, So, copper release, DC-d and DC-i data of each adhesive were subjected to a one-way ANOVA. Tukey’s post hoc test was used for pair-wise comparisons (α = 0.05). Results: The addition of copper nanoparticles provided antimicrobial properties to the adhesives at all concentrations (p < 0,05), and did not influence UTS, WS and SO (p > 0.05). Higher CR was observed in adhesives with higher concentration of copper nanoparticles (p < 0.05). The addition of 1% of copper nanoparticles decreased the DC-d and DC-i significantly. After 1Y, significant reductions of μTBS and increases of NL were observed in the control group (p < 0.05). Reductions of μTBS and increase of NL over time were not observed for copper-containing adhesives.Conclusions: The addition of copper nanoparticles in concentrations up to 0.5% in the simplified ER Ambar adhesive system may be an alternative to provide antimicrobial properties, increase the long-term stability of resin–dentine interfaces, 12 without reducing adhesives’ mechanical properties evaluated. / Objetivos: Este estudo avaliou o efeito da adição de nanopartículas de cobre, em diferentes concentrações, em um sistema adesivo convencional (CON) simplificado Ambar [FGM] sobre a atividade antimicrobiana (AAM), grau de conversão de corpos de prova de adesivos (GC-c) e na interface de união a dentina (GC-i), resistência máxima à tração (RT), sorção cumulativa de água (SO), solubilidade (SB) e liberação de cobre (LC) durante 28 dias, bem como a resistência de união (RU) e nanoinfiltração (NI) na interface de união à dentina nos tempos imediatos (IM) e após 1 ano (1A). Material e métodos: Sete sistemas adesivos experimentais foram formulados de acordo com a adição de nanopartículas de cobre (0 [controle], 0,0075, 0,015, 0,06, 0,1, 0,5 e 1%) no sistema adesivo Ambar. Foi testada a atividade antimicrobiana dos adesivos formulados contra Streptococcus mutans por meio de ensaios de difusão em ágar. Para GC-c, as amostras foram construídas e testadas após 24 h por meio de espectrofotometria no infravermelho com Transformação de Fourier (FTIR) e espectroscopia de micro-Raman. Para RT, as amostras foram testadas depois de 24 horas e 28 dias. Para SO e SB, depois de confeccionados os espécimes, eles foram armazenados em água e as propriedades medidas por 28 dias. Para LC, as amostras foram armazenadas em solução de ácido nítrico a 2% e as propriedades medidas durante 28 dias. O esmalte oclusal de trinta e cinco molares foi removido e os adesivos foram aplicados na superfície da dentina depois de condicionamento com ácido fosfórico a 37%. Depois de restaurações de resina composta, os espécimes foram seccionados longitudinalmente para se obter espécimes (palitos) de resina-dentina (0,8 mm2). Os espécimes foram testados em tensão a 0,5 mm/min no IM ou 1A (RU). Para NI, 2 espécimes de cada dente foram preparadas e analisadas em MEV e testados no IM ou 1A. Para GC-i, 2 espécimes foram preparados e analisados sob espectroscopia de micro-Raman. Os dados do RT foram analisados por uma análise de variância de 2 fatores. Para AAM, SO, SB, e liberação de cobre, os dados foram analisados por análise de variância de fator único, e para RU e NI os dados foram submetidos a análise de variância de 2 fatores para medidas repetidas. Também foi feito o teste post hoc de Tukey para múltiplas comparações. A significância estatística foi predefinida em α = 0,05.Resultados: A adição de nanopartículas de cobre adicionou propriedades antimicrobianas aos adesivos em todas as concentrações (p < 0,05), e não influenciou a RT, SO e SB (p > 0,05). LC elevadas foram observadas em adesivos com maior concentração de nanopartículas de cobre (p < 0,05). A adição de 1% de 10 nanopartículas de cobre diminuiu significativamente o GC-c e GC-i. Após 1 ano, decréscimos significativos de RU e aumentos de NI foram observados no grupo controle (p < 0,05). Decréscimos significativos de RU e aumentos de NI não foram observados nos adesivos contendo cobre (p > 0,05). Conclusões: A adição de nanopartículas de cobre em concentrações de até 0,5% no sistema adesivo convencional simplificado Ambar pode ser uma alternativa para adicionar atividade antimicrobiana e aumentar a estabilidade a longo prazo das interfaces resina-dentina, sem comprometer as propriedades mecânicas dos adesivos.
