Hiram, Roddy January 2016 (has links)
Abstract : Pulmonary hypertension (PH) is rare disease characterized by an important remodelling and proinflammatory cells recruitement into the pulmonary artery wall. Because of the late diagnostic, the patient care is often performed when PH is at its irreversible and most severe stage. Unfortunatly, none of the actual treatments are able to cure the patients for the long term. We hypothesize that inflammation could be a major event at the origin of all the other cellular dysfunctions that characterise PH. Resolvins; metabolites from Oméga-3 could resolve inflammation and potentially prevent or reverse PH. In the present study, two models of PH have been used. The first one is an in vitro model involving cultured human pulmonary arteries (HPA) in which inflammatory or hyperreactive conditions have been induced to evaluate the effects of Resolvin D1 and E1 and their precursors. The second model is a well-known in vivo model of monocrotalineinduced PH in rats, treated with MAG-DHA (monoacylglyceride form of docosahexaenoic acid) to evaluate the curative ability of this compound to resolve the disease at its severe stage. Results show that 24-h pre-treatment with TNFα, IL-6 or ET-1 increased the reactivity and Ca2+ sensitivity of HPA as revealed by agonist challenges with: 80 mM KCl, 1 μM 5- hydroxytryptamine (5-HT), 30 nM U-46619 and 1 μM PDBu. However, 300 nM RvD1 or RvE1, as well as 1 μM MAG-DHA or MAG-EPA (monoacylglyceride form of eicosapentaenoic acid) strongly reversed the over responsiveness induced by proinflammatory and hyperreactive treatments. Moreover, a 7-day treatment with MAG-DHA is able to resolve the inflammatory status in a rat model of monocrotaline-induced pulmonary hypertension. In both models, the inflammatory status enhanced the expression of inflammatory biomarkers (TNF-α, COX-2, STAT-3) as well as the detection of MMP9 and phosphorylated nuclear factors such as P-c-Fos, P-c-Jun and P-NF-κB involved in the activation of wall remodeling. Hence, RvD1, RvE1 and their precursors normalized the expression of these inflammatory biomarkers. In conclusion, Resolvin D1 and E1 and their respective precursors MAG-DHA and MAGEPA could inhibit inflammation status to prevent and potentially cure pulmonary hypertension.

Optimisation des profils lipidiques du cerveau de rats déficients en oméga-3 au sevrage par l'utilisation de matière grasse laitière : conséquences au niveau du lipidome / Optimization of lipid profiles in the brain of rats deficient in omega-3 at weaning by the use of dairy-fat : consequence at the lipidomics level

Du, Qin 17 December 2010 (has links)
The accretion of docosahexaenoic acid (DHA) in brain membranes mainly occurs around delivery and during the first two years of life. One of the main goals of neonatal nutrition is to optimize the incorporation of n-3 long chain polyinsaturate fatty acids (LC-PUFA) into the central nervous system, including DHA. Our goal was to study the impact of several kinds of diets based on dairy-fat to improve the n-3 LC-PUFA bioconversion and DHA accretion into brain. The other endpoint was to assess if beyond the brain fatty acid profiles, the dietary intakes would bring about a wider effect such as the one that can be appreciated through a lipidomic approach. We compared the nutritional effect of dairy-fat based diets to that of palm-oil based diets in the rat reproductive model. Mother rats were made deficient in α-linoleic acid (ALA) (0.4% of fatty acids (FA) for 6 weeks prior to mating and throughout gestation and lactation. After weaning, the resulting deficient 40 rat pups of either gender were split into 4 groups and received 10% fat diets made with either 1.5%ALA palm oil blend (P1), same added with 0.12% DHA and 0.4% arachidonic acid (ARA) (P2) , 1.5% ALA dairy-fat blend (B1) or 2.5% ALA dairy-fat blend (B2). The brain, red blood cell (RBC) and plasma fatty acid profiles were analyzed and treated using multivariate statistics. B1 was superior to both palm-oil based diets to improve the brain DHA contents (14.4% increase, P < 0.05). B2 brought an additional benefit for this parameter. The brain DHA contents in males were significantly lower than for the female because of a diet x gender interaction. This effect was smoothed with the dairy-fat diets or the palm-oil based diet augmented with preformed DHA.We calculated a new fatty acid index to predict the brain DHA contents, based on the fatty acid profiles measured in RBC and plasma, and which demonstrated a better performance than the existing published index. For the first time, we showed a profound and yet unsuspected effect of diets until now on the brain lipidome (lipids molecular species), affecting about 15% of the features detected. These changes were ascribed to the cholesterol and fatty acid metabolism, and to the lipid messengers, among others. In conclusion, our data appear highly relevant to improve infant formulas. The first use of the lipidomic approach in neonatal nutrition open the paths of new researches in the area of infant nutrition

The CHSE-214 salmon cell line as a model to study molecular regulation of long-chain polyunsaturated fatty acid biosynthesis in salmonids

Rubio Mejia, Olga Liliana January 2015 (has links)
The main source of omega-3 (n-3) long-chain polyunsaturated fatty acids (LC-PUFA) in our diet is supplied by fish, and an ever-increasing proportion of these are being produced by aquaculture. The drive for the growing market demand and production from sustainable sources has led to the use of high-energy (fat) diets and, recently, to the replacement of fishmeal and fish oil with non-marine components, such as plant meals and vegetable oils that are devoid of n-3 LC-PUFA. Both changes impact greatly on lipid and fatty acid metabolism in fish, with health implications for the fish and the human consumer. This impact highlights the need to investigate the basic molecular mechanisms underlying the regulation of lipid and fatty acid metabolism in fish, specifically focussing on the pathways of lipid homeostasis and LC-PUFA synthesis. The aim of this study was to develop and utilise Chinook salmon embryo (CHSE-214) cell line as a model for Atlantic salmon, Salmo salar L., to enable an integrated approach to study the biochemical and molecular regulation of lipid metabolism in fish. In particular, α-linolenic acid (LNA, 18:3n-3) and linoleic acid (LOA, 18:2n-6), which are essential fatty acids abundantly found in vegetable oils, and are precursors of LC-PUFA, were supplemented in combination with other fatty acids, to explore the effect of these on total lipid content, lipid class, FA composition and gene expression of CHSE-214 cell line. Total lipid content was extracted, followed by determination of lipid class and fatty acid analyses. Gene expression analyses of transcription/nuclear factors and various target genes in Atlantic salmon, including those involved in pathways of LC-PUFA synthesis and fatty acid oxidation, were carried out. The results demonstrated that CHSE-214 cell line, under experimental conditions, is able to convert LNA to eicosapentaenoic acid (EPA, 20:5n-3), and LOA to arachidonic acid (ARA, 20:4n-6), but not LNA and/or EPA to docosahexaenoic acid (DHA, 22:6n-3), highlighting the activity of elongase and desaturase enzymes during the conversion process. Changes occurring on the fatty acid profile and also at molecular level were observed. Understanding the role that transcription factors play in the regulation of lipid biosynthesis in fish will allow endogenous LC-PUFA synthesis to be optimised. The results from this study could be used to improve the efficiency of alternative, sustainable diets in aquaculture, while maintaining the nutritional quality of farmed fish for the final consumer. CHSE-214 cell line can therefore be used as a model to study the molecular mechanisms involved in the LC-PUFA biosynthesis, particularly in the conversion of LNA to EPA, which can then be reproduced in vivo, saving time and money.

Supplementation of diets of swine females with polyunsaturated fatty acids (PUFA) containing omega-3, especially the docosahexaenoic acid (DHA) may be an alternative to improve their reproductive performance. The objective of this study was to evaluate reproductive performance and serum levels of metabolic marker in swine females supplemented with omega-3. In this study, 463 swine females were supplemented in the diet with a source of DHA extracted from microalgae, from the 85th day of gestation and during both the lactation and the weaning-to-estrus interval (WEI). The DHA was included in the diet at: 3.5; 7.0; 14.0; and 28.0 g/d. Blood samples were collected at the beginning of the supplementation (during gestation), ten days after farrowing (during lactation) and one day prior to weaning, for determination of serum levels of metabolic markers: triglycerides; cholesterol; insulinlike growth factor type I (IGF-I); and non-esterified fatty acids (NEFA). No effect of the treatments was observed on the stillborn rate, total litter size and WEI at the parity in which supplementation occurred (P > 0.05), and also no effect occurred on total litter size and stillborn rate at the subsequent parity (P > 0.05). During the gestation, serum levels of cholesterol and IGF-I were lower (P < 0.05) and NEFA serum levels were greater than during lactation and the WEI (P < 0.05). Also during gestation, serum triglyceride levels were reduced (P < 0.05) only for females supplemented with 14.0 and 28.0 g of omega-3. The supplementation with DHA did not benefit the evaluated reproductive parameters of swine females, possibly due to the excellent performance observed during the experimental period.

Gil Sánchez, Alfonso 30 June 2011 (has links)
A supply of omega-3 long-chain polyunsaturated fatty acids (LC-PUFA) is essential for fetal development. Objective: We assessed placental transfer of 13C-labelled fatty acids (FA) 12 hours after oral application. Design: 11 pregnant women received 12h before elective caesarean section 0.5mg/kg 13C-palmitic acid (PA, 16:0), 13C-oleic acid (OA, 18:1n-9), 13C-linoleic acid (LA, 18:2n-6) and 0.1mg/kg 13C-docosahexaenoic acid (DHA, 22:6n-3) orally. Maternal blood samples, venous cord blood and placental tissue were collected and FA concentrations and their tracer content determined. Results: Most 13C-PA and 13C-OA in maternal plasma were in triglycerides (TG), while 13C-LA and 13C-DHA were mainly found in both plasma phospholipids and TG. Placenta/maternal plasma ratios and fetal/maternal plasma ratios for 13C-DHA were higher than for any other FA. Conclusions: We found a significantly higher ratio of 13C-DHA concentrations between cord to maternal plasma than for the other FA, in agreement with preferential transfer of DHA across the placenta.

The independent effects of purified EPA and DHA supplementation on cardiovascular risk in treated-hypertensive type 2 diabetic individuals

Woodman, Richard John January 2003 (has links)
[Formulae and special characters can only be approximated here. Please see the pdf version of the Abtract for an accurate reproduction.] Type 2 diabetes at least doubles the risk of cardiovascular disease. This can partly be explained by the increased prevalence of risk factors such as hypertension, dyslipidaemia and obesity. However, the underlying abnormality of insulin resistance and the presence of more recently identified risk factors including endothelial dysfunction, increased inflammation, and increased oxidative stress might also contribute towards the heightened cardiovascular risk. Fish oil, which contains eicosapentaenoic acid (EPA, 20:5 n-3), has wide-ranging beneficial effects on these and other abnormalities, and has reduced cardiovascular mortality in secondary prevention studies. Animal and human studies have recently established that in addition to EPA, docosahexaenoic acid (DHA, 22:6 n-3) also has beneficial effects, and furthermore, may have less detrimental effects than EPA on glycaemic control which has worsened in some fish and fish oil studies involving Type 2 diabetic subjects. Study 1 : This intervention study aimed to determine the independent effects of EPA and DHA on cardiovascular risk factors and glycaemic control in individuals with Type 2 diabetes receiving treatment for hypertension. In a double-blind placebo-controlled trial of parallel design, 59 subjects in good to moderate glycaemic control (HbA1c < 9%) were recruited from media advertising and randomised to 4 g/day of EPA, DHA or olive oil (placebo) for 6 weeks. Thirty-nine men and 12 post-menopausal women aged 61.2±1.2 yrs completed the study. Relative to placebo, and with Bonferroni adjustments for multiple comparisons, serum triglycerides fell by 19% (p=0.022) and 15% (p=0.022) in the EPA and DHA groups respectively. There were no changes in serum total cholesterol, or LDL- and HDL-cholesterol, although HDL2-cholesterol increased 16% with EPA (p=0.026) and 12% with DHA (p=0.05). HDL3-cholesterol fell by 11% (p=0.026) with EPA supplementation and LDL particle size increased by 0.26±0.10 nm (p=0.02) with DHA. Urinary F2-isoprostanes, an in-vivo marker of oxidative stress was reduced by 19% following EPA (p=0.034) and by 20% following DHA. DHA but not EPA supplementation reduced collagen-stimulated platelet aggregation (16.9%, p=0.05) and thromboxane release (18.8%, p=0.03), but there were no significant changes in PAF-stimulated platelet aggregation. Fasting glucose rose by 1.40±0.29 mmol/l (p=0.002) following EPA and 0.98±0.29 mmol/l (p=0.002) following DHA. Neither EPA nor DHA had any significant effect on HbA1c, fasting serum insulin or C-peptide, insulin sensitivity, stimulated insulin secretion, 24-hr ambulatory blood pressure and heart rate, markers of inflammation, and fibrinolytic or vascular function. Study 2 : This study aimed to examine the influence and causes of increased inflammation on vascular function in subjects recruited for Study 1. Compared with healthy controls (n=17), the diabetic subjects (n=29) had impaired flow-mediated dilatation (FMD) (3.9±3.0% vs 5.5±2.4%, p=0.07) and glyceryl-trinitrate mediated dilatation (GTNMD) (11.4±4.8% vs 15.4±7.1%, p=0.04) of the brachial artery. They also had higher levels of the inflammatory markers C-reactive protein (2.7±2.6 mg/l vs 1.4±1.1 mg/l, p=0.03), fibrinogen (3.4±0.7 g/l vs 2.7±0.3 g/l, p<0.001) and tumor necrosis factor-alpha (20.9±13.4 pg/l vs 2.5±1.7 pg/l, p<0.001). In diabetic subjects, after adjustment for age and gender, leukocyte count was an independent predictor of FMD (p=0.02), accounting for 17% of total variance. Similarly, leukocyte count accounted for 23% (p<0.001) and IL-6 for 12% (p=0.03) of variance in GTNMD. Von Willebrand factor, a marker of endothelial cell activation was correlated with leukocyte count (r=0.38, p=0.04), FMD (r=-0.35, p=0.06) and GTNMD (r=-0.47, p=0.009), whilst P-selectin, a marker of platelet activation was correlated with fibrinogen (r=0.58, p=0.001). Conclusion : EPA and DHA have similar beneficial effects on triglycerides, HDL2 cholesterol and oxidative stress in individuals with Type 2 diabetes and hypertension. However, DHA also increases LDL particle size and reduces collagen-stimulated platelet aggregation and thromboxane release, thus offering more potential than EPA as an anti-thrombotic agent. The beneficial effects of both oils were potentially offset by deterioration in glycaemic control. Neither oil affected blood pressure or vascular function. Longer-term studies with major morbidity and mortality as the primary outcome measures are required to assess the overall benefits and risks of EPA and DHA. The cross-sectional observations from Study 2 are consistent with the hypothesis that impaired vascular function in individuals with Type 2 diabetes and hypertension is at least in part secondary to increased inflammation, with associated endothelial and platelet activation.

Mental ill health in nursing and midwifery education : a critical discourse analysis

Hargan, Janine M. January 2017 (has links)
Students diagnosed with long-term mental health conditions have been the focus of policy development for over a decade. Student mental health is on the increase and universities are legally obliged to make reasonable adjustments for disabled students. Therefore it is crucial that nursing and midwifery education provides an inclusive learning environment, while maintaining fitness to practice standards. The focus of this study was to explore how discourses of mental health, reasonable adjustments and fitness standards influence nursing and midwifery education for students with a mental health condition. Principles of Wodak’s (2001) critical discourse analysis approach, which gives prominence to dominant discourses, their justifications and persuasive nature was utilised. Ten key written texts and 23 semi-structured interviews with students, lecturers and clinical mentors were conducted to acquire the constructions of mental health, reasonable adjustments and fitness requirements. The findings show that the dominant discourses attributed to students experiencing mental ill health were around medicine, difference and blame, all of which reinforced mental health stigma. In addition, mental health discourses within both verbal and written texts were not underpinned by disability discourses, allowing the exclusion of students who disclose mental ill health from accessing reasonable adjustments. In conclusion, students considered to have a mental health label faced discriminatory barriers and legislative and regulatory requirements of equality were not implemented.

Kvalita lipidů našich hospodářsky významných druhů ryb / The lipid quality of our economically significant fish species

KREJSA, Jakub January 2015 (has links)
The level of fish consumption in the Czech Republic is very low. One of the possible factors influencing this situation is low awareness of the customer or consumer. The thesis "The Lipid Quality of our economically significant fish species" works with the list of nineteen economically most significant fish species not only including fish from the semi-intensive and intensive breeding, but also such species that are interesting for the consumption from the sport fishing point of view. The samples taken from particular fish were primarily used for the analysis of the fat content and fatty acid composition. The analysis established very interesting results in this field and created basis for further analysis for establishing atherogenic and thrombogenic index or for formation of nutritive values mentioned as "Big 7" (fat content and the ratio of fatty acid out of it, sacharide content and the sugar ratio in it, protein content, ash content and energy value) by particular fish species according to the regulation ES 1169/2011. The results of this work confirmed that the fat content and its composition are highly influenced by the type of ingested nourishment. The fish bred in semi-intensive way with the highest fat content are Common carp (7,62 ? 2,9 %) and Silver carp (6,87 ? 1,37 %). On the contrary the predatory fish evinced the fat content usually within 1%. The fish from extensive breeding or the types caught in sport fishing are in the fat con-tent more balanced. The fattest fish in this group is Silver Bream (5,52 ? 0,44 %) and the lowest content evinced Goldfish (1,14 ? 0,38 %). Also the species living in the aquaculture and living in the free waters as well were compared (Common carp, Rainbow trout, Peled, Tench, Wels catfish). The fatty acid composition significantly depends on the fat content. The predatory fish contain 195 - 219 mg×100 g-1, the most (p0,05) was found by the peled from the extensive breed (1048 mg×100 g-1 EPA + DHA). The participation of SFA by the most of the fish was under 30%, the content of MUFA was the highest by the Common carp (52,71 ? 7,75 %), the lowest by the European perch (18,83 ? 1,12 %). Pike, Zander and European perch contain relatively the most of PUFA, this is over 50%. All the fish species (Nile tilapia excluded) show in this study the values of atherogenic index below 0,5 and thrombogenic index below 0,4. The total energetic value was counted from the results of nutritional values presented in the "Big 7" list. The higher energetic value is directly proportional to the increasing fat content. The fish species included in this thesis show the total energetic value in the range of 500 - 900 kJ×100 g-1. The results of this study bring complete, laboratory proven information about the nutrition composition of our fish. This data can be appli-cable not only to increase the knowledge of the customers but mainly to be used by breeders and manufacturers of fish as an implement of fulfilling the duty of stating the nutrition facts on their products according to ES 1169/2011 on the providing infor-mation about the eatables to the consumers.

This research aims at finding how BBC and AJE media represented the Israeli-Palestinian conflict. The study examines the news website, which reported the recent escalation of Jerusalem conflict in December 2017, where US President Trump have recognized Jerusalem as Israeli capital, and declared to move the US embassy from Tel-Aviv to Jerusalem. The study uses qualitative research, where it investigated the impact of ideology on media discourse by means of critical discourse analysis (DHA, Topoi, and Social actor’s representation). CDA was carried out for a sample of 8 news articles published on the websites of two networks: the British Broadcasting Corporation World news "BBCWN" and the Middle Eastern Qatari owned "Al Jazeera English". The articles were chosen within the month of December 2017. Articles were analyzed by means of the two-level analysis method, including the thematic and in-depth analysis. On the first, entry-level analysis, I focus on contents of texts and outline the discourse topics. While in the second phase I analyze the means of discursive strategies and the representation of social actors utilized throughout the text. The thematic analysis revealed that, both BBC and AJE have covered the incident similarly in general look. However, in-depth analysis showed that each network had portrayed the images of Israelis and Palestinians differently. On contrast of AJE, BBC tends to perceive Palestinians as a threat and the Israeli one as victims of the Palestinian violation. Moreover, the analysis revealed that each network had different ideologies and aims. The study concludes that AJE articles was biased pro-Palestinians, while BBC articles was biased pro-Israelis.
