Écotoxicité de la deltaméthrine et du malathion sur différentes souches de "Daphnia magna" (Crustacea, Cladocera) : apport de la protéomique dans la recherche de nouvelles cibles cellulaires / Ecotoxicity of deltamethrin and malathion with diverse strains of Daphnia magna (Crustacea, Cladocera) : contribution of proteomics investigation in the search for new cellular targets

L’objectif de ce travail a consisté à évaluer différents effets écotoxiques de la deltaméthrine et du malathion sur trois souches du crustacé cladocère Daphnia magna. Deux de ces 3 souches appartiennent au même clone A (souches 1 et 3), ce qui a permis d’observer des différences de sensibilité inter- et intra-clonale. Les tests d’écotoxicité aiguë (48h) ont révélé, aussi bien pour la deltaméthrine que pour le malathion, que la souche 1 est plus sensible que la souche 2, elle-même plus sensible que la souche 3. Les tests chroniques (21 jours) ont montré que le critère commun majoritairement affecté par les deux pesticides est le nombre de nouveaux nés par adulte. Ces tests ont confirmé la sensibilité de la souche 1. La deltaméthrine et le malathion sont embryotoxiques puisque l’exposition des daphnies aux fortes concentrations de deltaméthrine et de malathion a induit des malformations. Pour ces effets, la souche 1 est également plus sensible que la souche 2, elle-même plus sensible que la souche 3. Nous avons aussi observé l’apparition de mâles, mais uniquement dans la descendance de la souche 1 exposée à la deltaméthrine. Le dosage de l’activité spécifique de l’acétylcholinestérase (AChE), pour les deux insecticides, a montré une inversion de l’ordre de sensibilité des souches. L’AChE, biomarqueur d’exposition et d’effet, peut être donc considérée aussi comme biomarqueur de susceptibilité. L’analyse protéomique a permis de révéler plusieurs cibles cellulaires de la deltaméthrine chez la souche 1 par rapport à la souche 2. Elle est surtout venue confirmer la plus grande sensibilité de la souche 1 par rapport à la souche 2. L’ensemble de ces résultats rappelle que le facteur génétique est une source de variation et souligne les perspectives offertes par l’analyse protéomique / The aim of this work was to evaluate different ecotoxic effects of deltamethrin and malathion on three strains of the cladoceran Daphnia magna. Two of the three strains belong to the same clone A (strains 1 and 3), which allowed to observe differences in inter-and intra-clonal sensitivity. The acute ecotoxicity tests (48h) showed for both deltamethrin and malathion, that one strain is more sensitive than strain 2, itself more sensitive than strain 3. Chronic tests (21days) showed that the common criterion mainly affected by both pesticides is the number of neonates per adult. These tests confirmed the sensitivity of the strain 1. Deltamethrin and malathion are embryotoxic as daphnid exposure to high concentrations of deltamethrin and malathion induced malformations. For these effects, strain 1 is also more sensitive than strain 2, itself more sensitive than strain 3. We also observed the appearance of males, but only in the offspring of the strain 1 exposed to deltamethrin. The determination of the specific activity of acetylcholinesterase (AChE), for both insecticides, showed a reversal of the order of susceptibility testing. Thus, as a biomarker of exposure and effect, AChE can also be considered as a biomarker of susceptibility. Proteomic analysis revealed several cellular targets of deltamethrin in strain 1 compared to strain 2. It confirms the greater sensitivity of strain 1 compared to strain 2. All these results point out that the genetic factor is a source of variation and highlight the opportunities offered by proteomic analysis

Testes ecotoxicológicos para avaliação do potencial impacto ambiental em corpos receptores por efluente de Estação de Tratamento de Esgoto / Ecotoxicological tests to evaluate the potential environmental impact in receiving bodies by effluent from Sewage Treatment Plant

O crescimento das cidades sem planejamento adequado tem como consequências a ampla geração e incorreta destinação de resíduos sólidos e líquidos e a falta de esgotamento sanitário adequado, resultando em impactos negativos ao meio ambiente e aos corpos hídricos. O esgoto doméstico é composto predominantemente por matéria orgânica, além de nitrogênio, fósforo, organismos patogênicos e elementos potencialmente tóxicos (e.g.: fármacos), que podem ou não ser removidos nas estações de tratamento de esgoto. Esse efluente final (esgoto tratado), é encaminhado para o corpo receptor, normalmente um rio ou córrego. Quando não removidos durante os processos de tratamento, os compostos remanescentes no esgoto podem causar alterações físicas, químicas e principalmente biológicas nos corpos receptores. Entre as principais alterações biológicas destaca-se a perda de biodiversidade aquática presente no corpo hídrico receptor. O presente trabalho avaliou o possível impacto ambiental causado pelo efluente final de Estação de Tratamento de Esgoto (E.T.E.) em corpos hídricos receptores por meio de testes ecotoxicológicos. Foram selecionadas três estações de tratamento de esgoto, as quais foram avaliadas. Foram coletadas amostras do afluente bruto (P0), efluente final (P1), um ponto a montante (P2) e um ponto a jusante (P3) da mistura do esgoto com o corpo receptor de cada estação. Foram realizados ensaios de toxicidade aguda utilizando as espécies de invertebrados aquáticos Daphnia magna, Chironomus sancticaroli, Allonais inaequalis. Os organismos foram expostos às amostras coletadas para avaliação da sobrevivência. Para a E.T.E. 1, foi constatado adequada eficiência de remoção de toxicidade do esgoto quando comparado o afluente bruto ao efluente tratado, e não foram observados impactos do lançamento do efluente no corpo receptor. A E.T.E. 2, mostrou baixa eficiência de remoção de toxicidade do afluente bruto para o efluente tratado, refletindo em potencial tóxico a longo prazo ao corpo receptor. Da mesma forma, a E.T.E. 3 se mostrou ineficiente para remoção de toxicidade do afluente bruto para o efluente tratado, que refletiu em potencial tóxico ao corpo receptor em longo prazo. De acordo com os resultados obtidos no presente estudo, ressalta-se a importância da utilização de testes ecotoxicológicos como complementação aos parâmetros físicos e químicos exigidos pela legislação para avaliação da eficiência de remoção de contaminantes presentes no esgoto e que possuem potencial para geração de impacto a curto ou longo prazo à biota aquática presente no corpo hídrico receptor. / The increase of cities, without adequate planning, results in the wide generation and incorrect destination of solid and liquid wastes and the lack of adequate sanitary structure, resulting in negative impacts on the environment. Domestic sewage is composed predominantly of organic matter, in addition to nitrogen, phosphorus, pathogenic organisms and potentially toxic elements (e.g., drugs), which may or may not be removed at sewage treatment plants. This final effluent (treated sewage) is conveyed to water bodies, usually a river or stream. When not removed during the treatment processes, the remaining compounds in the sewage can cause physical, chemical and especially biological changes in the water bodies. Among the main biological changes is the loss of aquatic biodiversity present in the receiving water body. Thus, the present study evaluated the possible environmental impact caused by the final effluent from Sewage Treatment Plant (S.T.P.) in receiving water bodies by means of ecotoxicological tests. Three sewage treatment stations were selected. Samples of the raw tributary (P0), final effluent (P1) and the upstream (P2) and downstream (P3) points of the sewage mixture with the receiving body of each station were collected. Acute tests were carried out using the aquatic species Daphnia magna, Chironomus sancticaroli, Allonais inaequalis. The organisms were exposed to the samples collected for evaluation of survival in acute tests. For E.T.E. 1, it was found good toxicity removal efficiency of the station when comparing the raw affluent to the treated effluent, and no impacts of effluent release on the receiving body were observed. E.T.E. 2, showed low toxicity removal efficiency from the raw affluent to the treated effluent, reflecting long term toxic potential to the water body. Likewise, E.T.E. 3 was shown to be inefficient to remove toxicity from the raw affluent to the treated effluent, which reflected toxic potential to the water body in long term. According to the results obtained in the present study, the importance of the use of ecotoxicological tests as complementation of the physical and chemical parameters required by the legislation to evaluate the efficiency of removal of pollutants present in the sewage that have potential to generate impact in the short or long term to aquatic biota present in the body water recipient.

Approche moléculaire et mécaniste de la réponse transgénérationnelle lors d'une irradiation gamma chronique chez le cladocère Daphnia magna / Molecular and mechanistic studies of the transgenerational response during a chronic gamma irradiation in the cladoceran Daphnia magna

Afin de protéger durablement les écosystèmes face aux rejets planifiés ou accidentels de radionucléides dans l’environnement, il est essentiel d’évaluer l’impact de l’exposition des organismes aux radiations ionisantes sur le long terme, à l’échelle de plusieurs générations. Dans ce contexte, ce travail de doctorat vise à améliorer la caractérisation des processus moléculaires et la prédiction des effets transgénérationnels lors d’une exposition aux radiations gamma. Une approche expérimentale concerne l’étude des modifications épigénétiques radio-induites, c’est-à-dire des modifications des mécanismes régulant l’activité des gènes sans modification de la séquence d’ADN elle-même et de leur transmission au fil des générations. Cette étude a permis de mettre en évidence que certaines modifications de la méthylation de l’ADN, l’un des mécanismes épigénétiques les plus étudiés, peuvent être transmises par la lignée germinale aux les générations non-exposées (génération F3) suite à une exposition parentale (génération F0) externe aux radiations gamma (6,5 µGy.h-1 et 41,3 mGy.h-1) pendant 25 jours. Dans une seconde approche, un modèle mécaniste DEBtox (Budget Energétique Dynamique appliqué à la toxicologie) est modifié pour permettre l’analyse des effets des radiations gamma sur la croissance et la reproduction de D. magna à l’échelle de plusieurs générations. Pour ce faire, on utilise des compartiments de dommage, dont le niveau peut être hérité d’une génération à la suivante. Le modèle est ajusté aux données avec des méthodes d’inférence bayésienne afin d’estimer les paramètres tout en tenant compte des incertitudes qui leur sont associées. / In order to durably protect ecosystems facing planned or accidental releases of radionuclides, the long-term impact of organism exposure to ionizing radiation must be studied on a multigenerational scale. The aim of this PhD is to improve the characterization of molecular processes and the prediction of transgenerational effects during a gamma irradiation. First, an experimental approach investigated on radio-induced modifications of epigenetic processes, i.e. changes in mechanisms that regulate gene expression without changing DNA sequence itself and on the transmission of these modifications to subsequent generations. Significant changes in DNA methylation, a well-studied epigenetic mechanism, detected in generation F3 clearly showed that epigenetic modifications could be transmitted to unexposed generations, in response to the exposure of a parental generation (F0) to external gamma radiation (6.5 µGy.h-1 et 41.3 mGy.h-1) for 25 days. Second, a mechanistic modelling approach used a modified version of the DEBtox model (Dynamic Energy Budget model applied to toxicology) in order to analyze effects of gamma radiation on D. magna growth and reproduction over several generations. To that end, damage compartments, with damage levels that were transmitted from one generation to the next, were included. The model was fitted to data using Bayesian inference methods, in order to estimate the parameters while considering their associated uncertainty.

É crescente a utilização de fármacos mundialmente, principalmente devido às inúmeras doenças existentes, tanto humanas quanto para a criação de animais. Contudo, estes compostos podem causar um grande impacto no ambiente, pois são descarregados nos cursos d\'água através dos efluentes. Com isto, os fármacos podem contaminar também o solo e o sedimento, devido ao fato de serem desenvolvidos para apresentarem propriedades persistentes, como é o caso dos antimicrobianos e hormônios. Estes poluentes foram encontrados em estudos com água superficial, subterrânea, solo e sedimento em concentrações que variaram de ng.L-1 a \'mü\'g.L-1, e são capazes de provocar alterações no sistema endócrino de muitos organismos aquáticos, resistência microbiana, além de prejuízos à saúde humana. Assim, este trabalho teve como objetivos realizar um levantamento bibliográfico sobre os aspectos relacionados à presença de fármacos no ambiente, e as metodologias de detecção dos mesmos em amostras ambientais. Assim como determinar a presença de hormônios (estriol, estrona, 17\'beta\' -estradiol, 17\'alfa\'- etinilestradiol, progesterona e testosterona) e antimicrobianos (ciprofloxacina, norfloxacina, eritromicina e enrofloxacina) em amostras de água superficial e tratada; e também analisar a toxicidade destes antibióticos e hormônios em Daphnia magna. E, de acordo com o levantamento bibliográfico, concluiu-se que é necessário um aumento no volume de estudos destes compostos no ambiente, e principalmente, uma avaliação dos efeitos sinérgicos destes compostos nos organismos que têm contato com as matrizes ambientais como água, solo e sedimento. Para a detecção dos fármacos em amostras de água superficial e tratada, amostras foram coletadas ao longo do Rio Piracicaba (6 pontos) e em uma residência na cidade de Piracicaba, de março de 2011 a dezembro de 2012. Para as análises foi utilizado um cromatógrafo líquido acoplado a um espectrômetro de massas com fonte de ionização electrospray (LC-ESI-MS/MS). E, de acordo com os resultados obtidos algumas amostras apresentaram contaminação com os hormônios estriol, estrona, progesterona, 17\'beta\'-estradiol, e o 17\'alfa\'-etinilestradiol foram detectados em algumas das amostras durante o período de coleta, e suas maiores concentrações foram de 90, 28, 26, 137 e 194 ng.L-1, respectivamente, e com o antimicrobiano norfloxacina (variando de 8 a 18 ng.L-1), indicando aporte de esgoto sanitário em alguns pontos no Rio Piracicaba. Os resultados indicaram pouca toxicidade dos antimicrobianos norfloxacina, sulfadiazina e eritromicina, e do hormônio estriol, para a Daphnia magna / There is a growing use of drugs worldwide due to the numerous diseases both human and in animal husbandry. However, these compounds can have a big impact on the environment as they are discharged into waterways via wastewater. With this, drugs can also contaminate soil and sediment due to the fact that they are developed for presenting persistent properties, as is the case of antibiotics and hormones. These pollutants were found in studies with surface water, groundwater, soil and sediment concentrations from ng.L-1 to \'mü\'g.L-1, being able to cause changes in the endocrine system of many aquatic organisms, microbial resistance, and damage to human health. Thus, this study aimed to conduct a literature survey on the aspects related to the presence of pharmaceuticals in environment, and methods of detection thereof in environmental samples. As determine the presence of hormones (estriol, estrone, 17\'beta\'-estradiol, 17\'alfa\'-ethinylestradiol, progesterone and testosterone) and antimicrobials (ciprofloxacin, norfloxacin, enrofloxacin and erythromycin) on surface samples and treated water, and also assess toxicity of antimicrobials and hormones in Daphnia magna. And, according to literature review, it was concluded that an increase in studies volume of these compounds in environment, and especially, a review of synergistic effects of these compounds in organisms that have contact with environmental matrices such as water, soil and sediment. A comparison between the effects of polylactic acid and polystyrene microplastics on Daphnia magna

The effects of microplastics on zooplankton are an increasing environmental concern. Both primary microplastics that are produced as constituents of cosmetic products, as well as secondary microplastics that are formed by degradation of larger plastic debris, are ubiquitous in aquatic environments. Today, primary microplastics are being phased out and replaced by plant-derived bioplastics. Whether these new materials have similar effects as oil-based microplastics on animals is currently unknown. Here, we compare the effects of secondary microplastic exposure to Daphnia magna, using polylactic acid (PLA) as a representative for bioplastics and polystyrene (PS) for oil-based plastics. To increase the ecological relevance of our tests, we also provided treatments where the particles were coated with bovine serum albumin (BSA) as a means to simulate the coating of biofilms which readily form on particles under natural conditions. Furthermore, to be able to differentiate the effects of general particles from those specific to microplastics, kaolin clay was used as a control treatments, as well as one treatment containing only algae. The objectives were to test the influence of particles on feeding rates, reproduction and growth. PS caused a higher mortality, decreased feeding rate and reproductive output, while PLA and kaolin did not produce any negative effects. BSA did not have a significant effect on reproduction or growth. However, a decrease in reproduction was observed in the plastic treatments. Degradation of PS into styrene monomers is suggested as a possible explanation for the observed toxicity and effects on life history parameters.

Capturer les interactions écologiques en microcosme sous pression chimique à travers le prisme de la modélisation / Capturing ecological interactions within a microcosm under chemical pressure through the prism of modelling

L'évaluation du risque lié aux contaminants est généralement basée sur des données collectées au cours d'essais monospécifiques (à une seule espèce). Par conséquent, les interactions entre espèces, bien qu'elles structurent les écosystèmes, ne sont pas prises en compte. Pour explorer les effets des contaminants sur la dynamique des espèces en interaction, cette thèse vise à modéliser le fonctionnement d'un microcosme de laboratoire de deux litres incluant trois espèces, la daphnie Daphnia magna, la lentille d'eau Lemna minor et la microalgue Pseudokirchneriella subcapitata, exposées à une contamination par le cadmium. La dynamique des trois espèces ainsi que leurs interactions et les effets du cadmium ont été décrits par un modèle mécaniste basé sur des équations différentielles ordinaires couplées. Les principaux processus intervenant dans ce microcosme de trois espèces ont donc été formalisés, notamment la croissance et la survie des daphnies, la croissance et la sédimentation des algues, la croissance des lentilles, le broutage des algues par les daphnies, la compétition interspécifique entre algues et lentilles et les effets du cadmium sur ces différents processus. Les paramètres du modèle ont été estimés par inférence bayésienne, en utilisant simultanément toutes les données issues de différentes expérimentations en laboratoire, réalisées au cours de la thèse spécialement pour cette étude / Contaminant risk assessment is generally based on data collected during single species bioassays (including only one species). As a consequence, interactions between species that occur in ecosystems are not taken into account. To investigate the effects of contaminants on interacting species dynamics, this thesis aims at modelling the functioning of a 2-L laboratory microcosm with three species, the daphnid Daphnia magna, the duckweed Lemna minor and the microalgae Pseudokirchneriella subcapitata, exposed to cadmium contamination. We modelled the dynamics of the three species, their interactions and the effects of cadmium using a mechanistic model based on coupled ordinary differential equations. The main processes occurring in this three-species microcosm were thus formalized, including growth and survival of daphnids, growth and settling of algae, growth of duckweeds, grazing of algae by daphnids, interspecific competition between the algae and duckweeds, and the effects of cadmium on these processes. We estimated model parameters by Bayesian inference, using simultaneously all the data issued from multiple laboratory experiments specifically conducted for this study during the thesis

In this study, we tested the long-term and negative effects of microcystin-producing cyanobacterium Microcystis aeruginosa from Vietnam on Daphnia magna under the laboratory conditions. The test organisms were fed with mixtures of green alga Scenedesmus armatus. and toxic M. aeruginosa at different ratios (10% Microcystis + 90% Scenedesmus, 50% Microcystis + 50% Scenedesmus, 100% Microcystis, and 100% Scenedesmus) for over a period of 21 days. The life history traits of the organisms such as, survival, maturation, fecundity were daily recorded. Besides, the intrinsic population rate of D. magna in each treatment was also calculated based on the survivorship, the reproductive age and the clutch size of the animals. The results showed that survival, maturation and reproduction of the D. magna fed with 10, 50 and 100% M. aeruginosa was impaired. Additionally, the intrinsic population rate of the exposed D. magna was lower than that of the control. This study evidenced the adverse effects of toxic M. aeruginosa on both the individual and intrinsic population levels of D. magna. To our knowledge, this is the first report on the chronically detrimental impacts of toxic M. aeruginosa isolated from Vietnam on D. magna and contributed the scientific information on the severe influences of toxic cyanobacteria world wide. / Trong bài viết này, chúng tôi nghiên cứu ảnh hưởng xấu mãn tính của loài vi khuẩn lam Microcystis aeruginosa có khả năng sản sinh độc tố microcysin từ Việt Nam lên Daphnia magna trong điều kiện phòng thí nghiệm. Sinh vật thí nghiệm được cho ăn với hỗn hợp tảo lục Scenedesmus armatus và M. aeruginosa có độc ở các tỷ lệ khác nhau (10% Microcystis + 90% Scenedesmus, 50% Microcystis + 50% Scenedesmus, 100% Microcystis, và 100% Scenedesmus) trong thời gian 21 ngày. Les activités anthropiques liées à l'industrie nucléaire contribuent à des rejets continus de radionucléides dans les écosystèmes terrestres et aquatiques. Les travaux réalisés au cours de ce doctorat visent à apporter de nouvelles connaissances relatives aux effets des radiations ionisantes au cours d’une exposition multigénérationnelle de l’invertébré aquatique, Daphnia magna. Une irradiation gamma externe à des débits de doses pertinents du point de vue environnemental a été réalisée sur D. magna pendant trois générations successives. Les résultats mettent en évidence une accumulation et une transmission d’altérations de l’ADN au fil des générations, en parallèle d’une augmentation de la sensibilité des daphnies. Les données d’irradiation gamma et celles d’une étude antérieure de contamination alpha ont été analysées à l’aide du modèle mathématique DEBtox. Le modèle montre que les deux types de rayonnements agissent différemment sur les daphnies au cours des générations. Ce projet de recherche indique clairement qu’à l’avenir il est important d’étudier et de comprendre les effets transgénérationnels des radiations ionisantes à faibles doses. / Anthropogenic activities related to the nuclear industry contribute to continuous discharges of radionuclides into terrestrial and aquatic ecosystems. The aim of this PhD was to bring new knowledge on the effects of ionizing radiation during a multigenerational expose of the aquatic invertebrate, Daphnia magna. An external gamma radiation at environmentally relevant dose rates was performed on D. magna over three successive generations. Results show an accumulation and a transmission of DNA alterations over generations, in parallel of an increase in sensitivity of organisms. Gamma radiation data and those of a previous study of alpha contamination were analyzed using the mathematical model DEBtox. The model shows that the two types of radiation act differently on daphnia over generations. This research clearly indicates the importance of further studying and understanding transgenerational effects induced by low doses radiation in the future.
