Mythos als Zivilisationskritik: Die Pragmatisierung einer erweiterten negativen Dialektik in Werken Heiner Müllers

This thesis analyzes three works of the GDR dramatist Heiner Müller: his early prose poem Orpheus gepflügt, his learning play Mauser, and his late piece Verkommenes Ufer Medeamaterial Landschaft mit Argonauten. It demonstrates how Müller, throughout different career stages, pragmatizes myth to further critical thinking. Ancient Greek myths and Christian symbolism play a crucial role in Müller’s strategy of calling into question the very systems that lay claim to an absolute truth. Müller both alludes to and openly employs myths to identify their inherent dialectical tension operative in everyday life as well as in secular explanatory models used to legitimize political agendas. He expands Theodor W. Adorno’s concept of negative dialectics through an emphasis on the mythical pole of the dialectical dyad “myth and enlightenment.” By drawing attention to myths inherent in civilization, Müller opens up space for the imagination and the potential of the irrational to initiate change.

It's the Little Luxuries! How Centrum Warenhaus Rebranded the GDR

archives@tulane.edu / This thesis explores the GDR's propangandistic rebranding campaign of Centrum Warenhaus on Alexanderplatz. Such a campaign was exhibited in the remodeling of the Alexanderplatz between 1964 and 1970. The GDR's state department store Centrum Warenhaus on Alexanderplatz offered an alternative to capitalism best described as socialist consumerism. This system emanated state-driven policies like solidarity and brotherhood alongside East Germany's consumerist desires. Yet, this alternative did not last because it offered a figurative window to the west. Ironically, western products were rarely sold at Centrum Warenhaus and were only available for purchase at Intershops. These small-scaled stores created the societal divisions inherent in market capitalism. Access relied upon connection and western currency acquisition. As these two systems tried to exist simultaneously in East Berlin, the allure of the west overpowered each of them. The system of socialist consumerism in East Germany could no longer support itself. Its failure lay not in the creation of a capitalist alternative, but in its inability to provide such material sustenance to all of its citizens. / 1 / Lindsey Marie Harris

Invisible No More?: An Analysis of Disarmament, Demobilization, and Reintegration Programs for Former ‘Girl Soldiers Who Become Mothers’ and Their Children

DDR is meant to bridge the gap between child soldiering and civilian life. The challenges in the delivery of effective DDR programs serve as barriers to adequate support for child soldiers. Increasingly, research has also considered the gender-specific challenges and barriers that former girl soldiers face and the limited (if any) support they receive through DDR initiatives. DDR supports for girl soldiers who have become mothers are even less likely to meet their complex needs. As such, ‘girl soldiers as mothers’ and their children remain outside the dominant DDR discourse and subsequently do not get the attention, they need in DDR programming documents. While the discourse within DDR programs has progressed to include sexual violence and some gender-specific needs, these additions are insufficient for meeting the diverse needs of girl child soldiers and their children. The result is girl soldiers who become mothers and their children remain unprepared for returning to their communities or society, and their communities continue to be unprepared to receive them.

Progress towards a clinically-successful ATR inhibitor for cancer therapy

Yes / The DNA damage response (DDR) is now known to play an important role in both cancer development and its treatment. Targeting proteins such as ATR (Ataxia telangiectasia mutated and Rad3-related) kinase, a major regulator of DDR, has demonstrated significant therapeutic potential in cancer treatment, with ATR inhibitors having shown anti-tumour activity not just monotherapies, but also in potentiating the effects of conventional chemotherapy, radiotherapy, and immunotherapy. This review focuses on the biology of ATR, its functional role in cancer development and treatment, and the rationale behind inhibition of this target as a therapeutic approach, including evaluation of the progress and current status of development of potent and specific ATR inhibitors that have emerged in recent decades. The current applications of these inhibitors both in preclinical and clinical studies either as single agents or in combinations with chemotherapy, radiotherapy and immunotherapy are also extensively discussed. This review concludes with some insights into the various concerns raised or observed with ATR inhibition in both the preclinical and clinical settings, with some suggested solutions.

Caractérisation de MAPKBP1, un nouveau gène non-ciliaire impliqué dans des formes tardives de néphronophtise / Characterisation of MAPKBP1, a novel non-ciliary gene implicated in late onset nephronophthisis

La Néphronophtise (NPH) est une maladie autosomique récessive, qui se caractérise par des lésions tubulo-interstitielles conduisant à une insuffisance rénale terminale avant l'âge adulte. A ce jour des mutations dans 20 gènes (NPHP1-20) ont été identifiées comme responsables de la maladie. Les produits de ces gènes, les néphrocystines ou NPHP, se localisent et ont un rôle au cil primaire, définissant ainsi la NPH comme une ciliopathie. Nous avons identifié des mutations dans un nouveau gène candidat. Ces mutations ont été détectées dans six familles indépendantes, présentant une NPH tardive avec une fibrose rénale importante. Ce gène code MAPKBP1, une protéine très peu étudiée, ayant été décrite comme une protéine d'échafaudage de la voie JNK. MAPKBP1 interagit également avec WDR62, le produit d'un paralogue qui est le second gène le plus fréquemment muté dans les microcéphalies primaire récessives. La protéine WDR62 localise aux pôles du fuseau mitotiques (MSP) d'où elle régule l'orientation de l'axe des mitoses via la voie JNK dans le système nerveux central. Au cours de mon travail de thèse au sein du laboratoire des Maladies Rénales Héréditaires de l'Institut Imagine, j'ai étudié les fonctions cellulaires de la protéine MAPKBP1. J'ai ainsi pu mettre en évidence dans les lignées cellulaires ainsi que dans les tissus, que la protéine MAPKBP1 n'est pas présente au cil primaire et que les fibroblastes de patients ne présentent pas de défauts de ciliogenèse, indiquant que MAPKBP1 pourrait être le représentant d'une nouvelle famille de NPHP ne possédant pas de fonctions ciliaires. De manière intéressante, j'ai pu observer que MAPKBP1 est recruté aux MSP au cours des phases précoces de la mitose. Ainsi, en démontrant que certaines des mutations retrouvées chez les patients affectent le recrutement de MAPKBP1 aux MSP et perturbent également l'interaction de MAPKBP1 avec JNK et/ou WDR62, j'ai pu valider l'effet pathogène de ces mutations. De plus, j'ai montré des défauts dans la voie de réponse aux dommages à l'ADN dans les différents fibroblastes de patients comme récemment observé dans de nombreux modèles NPHP. / Nephronophthisis (NPH) is an autosomal recessive disease, characterised by tubulointerstitial lesions leading to an end-stage renal disease before adulthood. Causative mutations in 20 genes (NPHP1-20) have been identified so far. The products of those genes, the nephrocystins or NPHP, localise and function at the primary cilium, defining NPH as a ciliopathy. We identified mutations in a new candidate gene. Those mutations have been detected in six independent families, displaying late onset NPH with massive fibrosis. This gene encode MAPKBP1, a poorly studied protein that has been described as JNK pathway scaffold protein. MAPKBP1 also interacts with WDR62, the product of a paralogue which is the second most frequently mutated gene in recessive primary microcephalies. The protein WDR62 localises at the mitotic spindle poles (MSP) from where it regulates the orientation of the axis of division, through activation of JNK pathway in the central nervous system. During my work in the laboratory of Hereditary Kidney Diseases in the Imagine Institute, I studied MAPKBP1 cellular functions. I could show that in the various cell lines that have been tested, as well as in human kidney tissues, MAPKBP1 is not present at the primary cilia. And I also could show in patient fibroblasts that there is no ciliary defects, indicating that MAPKBP1 could be a novel NPHP protein that do not display ciliary functions. Interestingly, I could observe that MAPKBP1 is recruited at the MSPs during mitosis. Demonstrating that some patient mutations affect the recruitment of MAPKBP1 at the MSPs and also disturb interaction with JNK and/or WDR62, I could validate the pathogenic effect of those mutations. In addition, I could show defects in the DNA damage response in patient fibroblasts, as observed recently in various NPHP models.

Die Entwicklung der landwirtschaftlichen Produktionsgenossenschaften in den sächsischen Bezirken zwischen 1960 und 1969

Mit der Dissertation wird eine Periodisierung der Landwirtschaft der SBZ/DDR von 1945 bis 1990 vorgestellt und der LPG-Entwicklung speziell von 1960 bis 1969 vorgenommen. Dabei werden Etappen und Phasen abhängig von Zäsuren unterschieden. Es wird der Weg der drei LPG-Typen von der anfangs z. T. noch großbäuerlichen Wirtschaftsweise bis zur „sozialistischen Kooperation“ aufgezeigt. Dabei kam es zu Formen der Konzentration und Spezialisierung der Produktion. In der Dissertation wird auf sächsische Besonderheiten und Abweichungen von der Staatspolitik eingegangen. Bei der Darstellung der Ergebnisse der LPG werden Vergleiche zwischen den sächsischen Bezirken und der DDR gezogen. Die Arbeit enthält umfangreiche Ausführungen zu den beiden Wegen zur Betriebsvergrößerung der LPG. Als wichtig wurde angesehen, die Entwicklung aller drei LPG-Typen herauszuarbeiten. Die LPG als „sozialistische“ Genossenschaften werden als nicht identisch mit dem deutschen Genossenschaftsgesetz und auf dem „Leninschen Genossenschaftsplan“ fußend charakterisiert. Die Arbeit geht auf „sozialistische Experimente“ in der Agrarpolitik der DDR ein. Der Untersuchungszeitraum für das LPG-Modell in Sachsen fällt mit dem Ergebnis der gescheiterten Wirtschaftsreform der DDR, des NÖSPL, zusammen. Das LPG-Modell, auch in den sächsischen Bezirken, zwischen 1960 und 1970 war der westdeutschen Landwirtschaft hinsichtlich der Versorgungssicherheit nicht überlegen.
