The aim of the present clinical trial was to evaluate the effects of two isolation techniques on the clinical performance of Class V restorations, as well on the periodontal conditions of restored sites. Patients presenting at least two noncarious cervical lesions (NCLs) were enrolled in this study. The NCLs were randomized into the following groups: (1) isolation performed with rubber dam and gingival retraction clamp and (2) isolation provided with cotton rolls and gingival retraction cord. Both techniques were used with a saliva suction device. All restorative procedures were performed using a selfetching adhesive system and a nanofilled composite resin according to the manufacturer s instructions. The clinical performance of restorations was recorded in terms of fracture and retention of restoration, marginal adaptation, marginal staining, postoperative hypersensitivity, and preservation of tooth vitality at 1 week and 6 months after placement.The periodontal condition of restored sites was evaluated based on the presence of supragingival plaque, gingival marginal bleeding, probing depth, and relative gingival recession. Thirty patients were enrolled in the study, yielding a total of 136 restorations (68 restorations per group). At the 6-month followup, one restoration from the rubber dam group and three restorations from the cotton roll group lost retention. In this period, the highest incidence of small marginal defects was observed in the group isolated with cotton rolls (p= 0.01). Both groups showed a statistically significant increase in supragingival plaque at six months after restoration placement (p< 0.05). However, no significant differences were detected among the two groups tested with respect to any of the criteria used to evaluate the restorations or the periodontal condition (p> 0.05). Within the limits of this study, it can be concluded that both isolation techniques resulted in equally clinically acceptable restorations without producing negative effects on periodontal tissues / Durante os procedimentos restauradores, diferentes métodos de isolamento do campo operatório podem ser utilizados a fim de promover controle de umidade e retração de tecidos gengivais. O objetivo do presente ensaio clinico foi avaliar os efeitos de duas técnicas de isolamento no desempenho clínico de restaurações Classe V, bem como na condição periodontal dos sítios restaurados. Pacientes apresentando, no mínimo, duas lesões cervicais não cariosas (LCNCs) foram selecionados para este estudo. As LCNCs foram randomizadas nos seguintes grupos: (1) isolamento com lençol de borracha e grampo retrator de gengiva e (2) isolamento com rolos de algodão e fio retrator de gengiva. Um sugador de saliva foi utilizado em ambas as técnicas. Todos os procedimentos restauradores foram executados com sistema adesivo autocondicionante e compósito restaurador nanoparticulado, seguindo as instruções dos fabricantes. O desempenho clinico das restaurações foi avaliado em termos de fratura e retenção da restauração, adaptação marginal, manchamento marginal, sensibilidade pós-operatória e preservação da vitalidade pulpar após 1 semana e 6 meses da inserção das restaurações.A condição periodontal dos sítios restaurados foi avaliada com base na presença de placa supragengival, sangramento gengival marginal, profundidade de sondagem e recessão gengival relativa. Trinta pacientes foram incluídos no estudo, resultando num total de 136 restaurações (68 restaurações por grupo). Após seis meses de acompanhamento, uma restauração do grupo isolado com lençol de borracha e três restaurações do grupo isolado com rolos de algodão perderam a retenção. Neste período, uma maior incidência de pequenos defeitos marginais foi observada no grupo que recebeu isolamento com rolos de algodão (p= 0.01). Ambos os grupos apresentaram um aumento significativo do acúmulo de placa supragengival após 6 meses da inserção das restaurações (p< 0.05). Porém, não foram detectadas diferenças estatisticamente significativas entre os dois grupos testados considerando os critérios utilizados para avaliação das restaurações e da condição periodontal (p> 0.05). Efeitos de diferentes protocolos de limpeza de resíduos do cimento endodôntico e a repercussão sobre a interface de adesão na dentina bovina /

Orientador: Milton Carlos kuga / Resumo: O objetivo deste estudo foi avaliar a eficiência do etanol a 95 % (Rinse-N-Dry [RD]) ou da solução de formamida (Endosolv R [ER]), associados com a microabrasão com micropartículas de vidro (Concepsis Scrub [Cs]), na limpeza da dentina impregnada com o cimento à base de resina epóxi (AH Plus) e seus efeitos sobre a interface adesiva. Quarenta espécimes de dentina foram impregnadas com o cimento endodôntico (AH Plus). Em seguida foram limpos com os seguintes protocolos (n = 10): G1(E), somente RD; G2(ER), somente ER; G3(E + Cs), RD e Cs; G4(ER + Cs), ER e Cs. No G5 (controle) nenhum tratamento foi realizado. Os espécimes foram analisados em SEM, para avaliar a presença de resíduos (500x) e contagem de túbulos de dentinários abertos (1000x). Outros 50 espécimes foram preparados esubmetidos a similares procedimentos e protocolos de limpeza. Após 7 dias, tubos contendo resina composta foram fixados na dentina com um sistema adesivo "condiciona-e-lava" (Scotchbond Multi-porpose; 3M ESPE; 3M; St. Paul; MN, USA) e submetidos ao teste de microcisalhamento. G2 e G4 demonstraram maior persistência de resíduos e menor incidência de túbulos dentinários abertos que G1 e G3(P<0.05) e não houve diferença entre G1 e G3 ou G2 e G4 (P>0.05). A resistência de união do sistema adesivo foi similar entre todos os grupos (P>0.05). A microabrasão imediata interferiu negativamente na ação dos protocolos de limpeza, porém todos mantiveram resíduos sobre a superfície dentinária. Entretanto, após 7 dias... G2 and G4 showed higher residues persistence and lower incidence of open dentinal tubules than G1 and G3 (P <0.05). No difference was found between G1 and G3 or G2 and G4 (P> 0.05). Adhesive system bond strength was similar in all groups (P> 0.05). Microabrasion negatively affected the cleaning protocols action, all groups presented residues on the dentin surface. Dois adesivos "universais ou multi-molde" (Scotchbond Universal, 3M ESPE e All-Bond Universal, Bisco) foram testados nos modos autocondicionantes e com condicionamento prévio e comparados aos métodos tradicionais de união ao esmalte e dentina (grupos controles) utilizando os adesivos Optibond FL (Kerr) e Clearfil SE Bond (Kuraray Noritake). Esmalte e dentina de terceiros molares humanos foram utilizados nos procedimentos adesivos, os quais foram realizados com as instruções dos fabricantes. Após a aplicação dos adesivos, um bloco de compósito foi construído em incrementos. Os dentes foram seccionados longitudinalmente para obtenção de espécimes em formato de "palitos" para o ensaio de resistência de união por microtração (n=8) e as análises em microscopia eletrônica de transmissão (MET) (n=2). Os espécimes foram testados após 24 horas ou um ano de armazenamento em saliva artificial. Para a MET, solução traçadora de nitrato de prata foi utilizada para mostrar espaços nanométricos e evidências de degradação na área de união. As superfícies fraturadas dos espécimes foram observadas em microscopia eletrônica de varredura e os dados de resistência de união analisadas pela ANOVA três-fatores, teste de Tukey e Dunnett (5%). O condicionamento prévio do esmalte aumentou a resistência de união dos adesivos universais, mas essa reduziu após o armazenamento por um ano. Os adesivos universais mostraram menor resistência de união em esmalte que o grupo controle quando utilizados no modo autocondicionante no tempo de 24 horas. Nenhuma diferença significativa foi encontrada na resistência de união em dentina nos dois tempos de avaliação quando comparados aos adesivos universais e os controle. O armazenamento por um ano reduziu a resistência de união em dentina apenas para um adesivo universal usando no modo autocondicionante. A análise fractográfica em esmalte mostrou predomínio de fraturas mistas e coesivas na camada de adesivo. Em dentina, o predomínio foram fraturas mistas, adesivas e coesivas em compósito. A observação da ultramorfologia em MET mostrou a formação de hibridização e zonas de interação entre todos os adesivos em esmalte e dentina nos dois modos de aplicação. Sinais de nanoinfiltração foram identificados na área de união dentina-resina para os adesivos Scotchbond Universal e Clearfil SE Bond após armazenamento por um ano. Os resultados sugerem que armazenamento por um ano pode reduzir a resistência de união quando os adesivos universais foram aplicados em esmalte condicionado, mas em dentina condicionada não houve redução da resistência de união, exceto para o adesivo Scotchbond Universal. No esmalte a resistência de união do grupo controle foi maior que a dos adesivos universais no modo autocondicionante. Após o armazenamento por um ano, a resistência de união dos adesivos universais em esmalte tratado com ácido fosfórico não diferiu do grupo controle. Em dentina, não foi encontrada diferença significativa entre adesivos universais e os grupos controles, independente do modo de condicionamento / Abstract: The aims of this study were to analyze the ultramorphological characteristics of tooth¿resin interfaces, the microtensile bond strength of multimode adhesive systems bonded to enamel and dentin, and the failure pattern of tested specimens. Two multimode adhesives (Scotchbond Universal, 3M ESPE; All-Bond Universal, Bisco) were tested in both self-etch and etch-and-rinse modes and compared to control groups using traditional bonding techniques (Optibond FL, Kerr; Clearfil SE Bond, Kuraray Noritake) in enamel and dentin. Enamel and dentin from third human molars were used for bonding and adhesives used according to the manufacturer¿ instructions. A composite resin block was incrementally applied to bonded surfaces, and teeth were sectioned transversely to the tooth-composite interface to obtain specimens for microtensile bond strength (n=8) and transmission electron microscopy (TEM) analysis (n=2). Specimens were tested after storage for either 24 hours or 1 year in artificial saliva. For TEM, a silver tracer solution was used to reveal nanometer-sized spaces and evidence of degradation within resin¿dentin interfaces. Fractured surfaces of tested specimens were analyzed under scanning electron microscopy. Bond strength data were analyzed by three-way ANOVA and by Tukey¿s and Dunnett¿s tests (?=5%). The acid-etching mode increased the enamel bond strength of multimode adhesives; however, bond strength decreased after storage for 1 year. Multimode adhesives showed lower enamel bond strength than the control group adhesive when used in self-etch mode for 24 hours. No significant differences in dentin bond strength were noted between multimode and control adhesives in either evaluation period. Storage for 1 year only reduced the dentin bond strength for a multimode adhesive used in self-etch mode. Mixed and cohesive-within-adhesive-layer failures were predominant in enamel. In dentin most fractures were mixed, adhesive along the dentin or enamel, and cohesive within the composite layer. TEM analysis identified hybridization and interaction zones in dentin and enamel for all adhesives and application modes. Silver impregnation was detected on dentin¿resin interfaces after storage of specimens for 1 year only with the Scotchbond Universal and Clearfil SE Bond samples. Results suggested that storage for 1 year can reduce bond strength when multimode adhesives are applied on an etched surface, but the dentin bond strength was not affected when using the etch-and-rinse mode (except for the Scotchbond Universal sample). The enamel bond strengths of the control group adhesives were higher than those obtained for multimode adhesives in the self-etch mode. After storage for 1 year, the bond strengths of multimode adhesives in etched enamel did not differ from those of the control group adhesive. Effect of halloysite aluminosilicate clay nanotube incorporation into bonding agents on shear bond strength to human dentin

It has been suggested that filler addition to the adhesives would increase the bonding strength of the adhesive layer. Halloysite aluminosilicate nanotubes (HNTs) are biocompatible, hydrophilic, durable, and have high mechanical strength. These advantages make them good candidates to be used as reinforcing agents for improving the properties of dental adhesives. The objective of this study was to evaluate the effect of incorporating HNTs into a commercial two-step etch-and-rinse adhesive system or one-step self-etch adhesive system on dentin shear bond strength. HNTs were incorporated into the two commercial adhesive systems in 0 wt%, 5 wt%, 10 wt%, and 20 wt%. The commercial control adhesives and the experimental adhesives were used to bond occlusal dentin of 120 extracted human molar teeth and then tested for shear bond strength by a universal testing machine with a semi-circular edge at a crosshead speed of 1.0 mm/min. Debonded specimens were examined under light microscopy to evaluate the fracture pattern. Resin-dentin interface were evaluated under scanning electron microscopy (SEM) after bonding dentin slabs using commercial control adhesives and experimental adhesive that showed numerically highest shear bond strength from each adhesive system. Two-way ANOVA was used to evaluate the effects of adhesive system and nanofiller content on shear bond strength. Pair-wise comparisons between groups were made using Fisher's (LSD) (p < 0.05). For the self-etch adhesive system, only incorporation of 5 wt% showed a significant increase in shear bond strength to dentin compared with the commercial control group. For the etch-and-rinse adhesive system, there was no significant difference in shear bond strength between HNTs filled adhesives groups and the commercial control group. Resin-dentin interface SEM evaluation showed nanotubes infiltrated into dentinal tubules. In conclusion, incorporating the self-etch adhesive system with 5 wt% HNTs increased the bond strength to dentin. Incorporation of up to 10 wt% filler concentration into both the self-etch and the etch-and-rinse adhesive systems did not adversely affect the bond strength to dentin or the handling properties. HNTs can penetrate along with resin tags into dentinal tubules, which could expand the use of their unique properties.

Flexural strength and shear bond strength of self-etching/self-adhesive resin luting agents

Indiana University-Purdue University Indianapolis (IUPUI) / Traditional resin luting agents generally have mechanical properties that are superior to the newer so-called “universal” self-etching/self-adhesive resin luting agents. However, recent reports indicate that some properties of these new luting agents have been improved, approaching those of the traditional etch and rinse resin luting agents. The objective of this study was to test some mechanical properties of four of these self-etching/self-adhesive resin luting agents [Maxcem Elite (ME), Multilink Automix (MA), RelyX Unicem (RU), SmartCem 2 (SC)] and compare them to a traditional etch and rinse resin luting agent [RelyX ARC (RA)] and a resin-modified glass ionomer luting cement [Fuji Plus (FP)], both of which have much longer histories of clinical success. By comparing the properties of the newer cements to the standards, it may be possible to determine how clinically successful the newer cements may be. The mechanical properties tested were flexural strength (FS) and shear bond strength (SBS). The FS test included making beams of each material, storing them in water for periods of time (24 hours and 90 days) and then performing a three-point bending test on a universal testing machine. The 90 day groups were thermocycled. The SBS test involved preparing human molar specimens, making flat dentin surfaces. Composite cylinders were fabricated, luted to the dentin surfaces with each of the materials tested, stored in water for periods of time (24 hours or 90 days), and then a knife edge shear test was performed on a universal testing machine. The 90 day groups were thermocycled. A Weibull-distribution survival analysis was performed. The results revealed significant differences in the FS of all materials tested at 24 hours. After 90 days and thermocycling, only SC and RA were not significantly different. At both time periods, FP had the lowest and MA the highest FS. The SBS results showed MA, RA, and RU to have the highest bond strengths; SC and ME the lowest at 24 hours. After 90 days and thermocycling, RA had significantly higher bond strength than all other groups; ME, FP and SC had the lowest. The self-etching/self-adhesive resin luting agents all performed at least as well as FP, with the exception of SC (SBS 24 hour). They did not all perform as well as RA, with the exception of SC (FS 90 day), MA (SBS 24 hour, FS 24 hour and 90 day), and RU (SBS 24 hour). The newer luting agents should expect to have clinical success, regarding flexural strength and shear bond strength, at least as good as resin-modified glass ionomer luting cements and approach the level of traditional etch and rinse resin luting agents.

Quantitative comparison of nanoleakage among five resin luting agents after aging

Indiana University-Purdue University Indianapolis (IUPUI) / Potential problems of one-step adhesives have been identified, including water uptake and subsequent plasticization, water-and enzyme-induced nanoleakage, and the presence of voids due to phase-separation or osmosis. Clinically, adhesive failures due to marginal degradation present as retention loss, marginal discoloration, and secondary caries. However, the mechanisms of adhesive interface degradation of self-etching and self-bonding resin luting agents are not fully understood. The objective of the study was to investigate adhesive layer degradation by using a nanoleakage technique with five different resin luting agents. Materials and Methods: Five different resin luting systems, Variolink II, Panavia F2.0, RelyX Unicem, RelyX Unicem2, and Maxcem Elite were evaluated in this study. The 25 dentin specimens were randomly divided into five resin luting agent groups. Flat dentin surfaces were created mid-coronally and were luted with luting agents. Then, each tooth was sectioned occluso-gingivally. The first half of each tooth was used as a control group and the other half was used as the experimental group. The control group was immersed in artificial saliva at 37°C and SEM examination with chemical analysis was performed within 48 hours. In the tested group, all specimens were immersed in artificial saliva at 37°C for 10 days and thermocycled. For the SEM examination, the specimens were immersed in a 50-percent ammoniacal silver nitrate solution for 24 hours.22 SEM was used for observation of silver penetration of the specimens. Three scan lines were selected. For elemental analysis, natural apatite, olivine minerals, and pure silver metal were chosen as standards for Ca, Si and Ag. Data were analyzed using ANOVA with a 5-percent significance level. Results: At the bottom of the hybrid layer, there was no significant difference in silver uptake within the adhesive interface between luting agents (p > 0.05) and there was no significant change in silver uptake within the adhesive interface after thermocycling (aging) (p > 0.05). Conclusion: All resin luting agents exhibited nanoleakage after both 24-hour storage and 10-day storage with thermocycling.

METHODS: Exposed flat dentin of extracted third molars was only ground with 600-grit SiC paper/1 minute (normal dentin - N), or subsequently eroded by a regular-cola soft-drink (eroded dentin - E). N and E were acid-etched, washed, dried and rehydrated with 1.5 μL, respectively, of distillated water (control - NC / EC); of 0.004% CHX (N0.004% / E0.004%); or of 2% CHX (N2% / E2%). Adper Single Bond 2 was applied in all specimens and resin composite buildups were constructed with Filtek Z350. Specimens were sectioned in beams, which were tested (μTBS) immediately or after 6 months of aging. RESULTS: Microtensile bond strength to eroded dentin was always significantly lower than that to normal dentin. Application of tested CHX solutions did not exert a significant effect immediately; however, after aging, the 2% CHX prevented abrupt bond strength loss both to eroded and normal dentin. / Revisión por pares

Buscou-se minimizar ou prevenir a degradação das fibrilas colágenas, tanto no controle da progressão do desgaste erosivo em dentina, quanto na preservação das interfaces adesivas aí estabelecidas, por meio do uso de agentes antiproteolíticos (clorexidina/CHX e epigalocatequina-3-galato/EGCG) ou, ao mesmo tempo, promotores de ligações cruzadas entre elas (proantocianidina/PAC). O papel de algumas dessas estratégias no estabelecimento, e conservação, de interfaces adesivas em dentina erodida, substrato adverso à interação com materiais adesivos, tem sido, porém, pouquíssimas vezes reportado. Este estudo in vitro propôs-se a avaliar, pois, o efeito de tais agentes, usados como estratégias para prevenção/controle do desgaste, na resistência de união (RU) do conjunto sistema adesivo condicione e lave simplificado-resina composta à dentina erodida, comparada à normal. A dentina superficial oclusal de terceiros molares foi apenas submetida à ação de uma lixa de SiC (granulação 600; 1 min; N: substrato normal) ou sequencialmente a desafio erosivo inicial (Coca-Cola®; 5 min). Recebeu, então, ou não (C: controle/sem aplicação), a aplicação de um dos géis com os seguintes princípios ativos - P: placebo/sem princípio ativo; CHX: digluconato de clorexidina a 0,12%; EGCG: epigalocatequina-3-galato a 400 ?m; PAC: proantocianidina a 10%. Aquela de início desmineralizada ainda foi submetida a ciclagem de pH (Coca-Cola®; imersões de 5 min, 3x/dia, 5 dias; E: substrato erodido). Após condicionamento (H3PO4 a 37%; 15 s; lavagem 30 s; secagem com papel absorvente), o adesivo AdperTM Single Bond 2® foi aplicado em todos os espécimes e a porção coronária, reconstruída com a resina FiltekTM Z350®. Transcorridas 24 h (água destilada/37?C), os espécimes foram seccionados em palitos e testados (?TBS; 0,5 mm/min). As superfícies fraturadas de cada palito foram avaliadas utilizando-se um microscópio digital (50x de aumento). Os valores de RU obtidos foram organizados considerando- se cada dente como unidade experimental e os testes de Análise de Variância a 2 critérios e de Tukey aplicados (?=0,05). Um dente extra para cada grupo foi tratado exatamente como os outros, mas o corante fluorescente rodamina B foi previamente adicionado (0,16 mg/mL) ao sistema adesivo para permitir a avaliação qualitativa da interface adesiva por meio de Microscopia Confocal de Varredura a Laser. Analisando-se os dados obtidos pôde-se observar que, diferentemente da variável aplicação de géis para prevenção/controle do desgaste erosivo (p=0,076), a variável condição do substrato dentinário exerceu influência significante sobre os resultados (p<0,001). Ademais, não houve interação entre elas (p=0,979). Os valores imediatos de RU ao substrato erodido foram, pois, sempre inferiores que aqueles ao substrato normal, independentemente da aplicação, ou não, de qualquer um dos géis para prevenção/controle do desgaste erosivo. Quanto ao padrão de fratura dos palitos testados, as falhas adesivas e mistas foram predominantes em relação às coesivas, independentemente se em dentina ou em resina. Menores porcentagens de falhas coesivas ainda puderam ser verificadas para o substrato erodido, com relação ao normal. No que se refere à análise qualitativa, observou-se, para o erodido, comparado ao normal, independetemente do tratamento para controle do desgaste, camada escura subjacente à de adesivo propriamente, representação da menor concentração de material marcado por rodamina B, bem como tags resinosos com maior comprimento e em maior quantidade. Assim sendo, conclui-se, por ora, que as estratégias estudadas não foram capazes de favorecer, tampouco de prejudicar, o estabelecimento de interface adesiva em dentina erodida. / In order to minimize the degradation of collagen fibrils, both to control the progression of the dentin erosive wear and to preserve adhesive interfaces established there, antiproteolytic (chlorhexidine/CHX and epigallocatechin-3-gallate/EGCG) or, at the same time, collagen cross-linking agents (grape seed proanthocyanidin extract/PAC) started to be successfully used. Some of these strategies in the establishment and preservation of adhesive interfaces on eroded dentin, a substrate adverse to the interaction with adhesive materials, has been seldom reported yet. This in vitro study aimed to evaluate, thus, the effect of such agents, used as strategies for wear preventing/controlling, on bond strength (BS) of a simplified etch-and-rinse adhesive system adhesive plus a resin composite to eroded, and normal (parameter for comparisons) dentin. Superficial occlusal dentin of third molars was only ground with a SiC paper (600-grit; 1 min; N: normal substrate), or subsequently submitted to an initial erosive challenge (Coca-Cola®; 5 min). It, then, received or not (C: control/without application), application of one of the gels with the following active principles - P: placebo/without active principle; CHX: 0.012% chlorhexidine digluconate; EGCG: epigallocatechin-3-gallate at 400 ?m; PAC: 10% proanthocyanidin. Initial demineralized dentin was still submitted to a pH cycling (Coca-Cola®, 5 min immersions, 3x/day, 5 days, E: eroded substrate). After acid-etched (37% H3PO4; 15 sec; 30 sec washing; drying with absorbent paper), the adhesive Adper(TM) Single Bond 2® was applied in all specimens and resin composite buildups were constructed with Filtek(TM)Z350®. After 24 h (distilled water/37?C), specimens were sectioned in beams and tested (?TBS; 0.5 mm/min). Obtained BS values were organized considering each tooth as an experimental unit and two-way ANOVA and Tukey tests were applied (?=0.05). An extra tooth for each group was treated just like the others, but the adhesive system was marked by the addition of rhodamine B (0.16 mg/mL) to allow qualitative evaluation of the adhesive interface by means of Confocal Laser Scanning Microscopy. As opposed to the variable application of gels for wear preventing/controlling (p=0.076), the condition of the dentin substrate had a significant influence on the results (p<0.001). In addition, there was no interaction between them (p=0.979). Immediate BS values to the eroded substrate were always lower than that to the normal substrate, regardless of the application or not of any of the gels for wear preventing/controlling. As for the fracture pattern of the tested beams, adhesive and mixed failures were predominant in relation to the cohesive failures, regardless of whether in dentin or in resin. Lower percentages of cohesive failures could still be verified for the eroded substrate, relative to the normal one. Concerning the qualitative analysis, it was observed, for the eroded substrate, compared to the normal, independently of the treatment for wear preventing/controlling, a dark layer underlying that of the adhesive itself, which represents a lower concentration of rhodamine B-labeled material, as well as resinous tags with greater length and in greater quantity. Therefore, studied strategies were not able either to improve, neither to impair, the establishment of adhesive interface on eroded dentin.

O estabelecimento de interfaces adesivas no substrato dentinário artificialmente erodido, conceitualmente pobre em minerais, revestido por matriz orgânica exposta, é pouco promissor. Por ora, tornam-no similar àquele verificado para a dentina normal/hígida apenas a asperização ou, quando do uso de adesivos universais, a desproteinização com hipoclorito de sódio. Manter a camada de fibrilas colágenas, por meio do uso de inibidores de proteinases, talvez mesmo que compactada por abrasão, pode, porém, ser conveniente, já que desempenha importante papel nos processos de des e remineralização, inclusive contendo a progressão do desgaste erosivo e erosivo-abrasivo, e no estabelecimento de uma camada híbrida propriamente dita. Avaliou-se, pois, de imediato, a resistência de união do conjunto sistema adesivo condicione e lave simplificado-resina composta à dentina, considerando-se como fatores experimentais a condição desse substrato, em três níveis (N: normal; E: erodido; EA: erodido-abrasionado), e a aplicação de um gel para prevenção/controle do desgaste erosivo em três níveis (C: controle/sem aplicação; P: placebo/sem princípio ativo; CHX: digluconato de clorexidina a 0,12%). Para determinação do substrato normal, a dentina superficial oclusal de terceiros molares foi apenas submetida à ação de uma lixa de SiC (#600; 1 min; N); para a do erodido e do erodidoabrasionado, sequencialmente, de pronto, a desafio erosivo inicial (Coca-Cola®; 5 min). Aplicou-se sobre ela, então, ou não (controle/sem aplicação), um dos géis: placebo/sem princípio ativo; ou à base de CHX a 0,12%. Aquela de início desmineralizada ainda foi submetida à ciclagem de pH (Coca-Cola®; imersões de 5 min, 3x/dia, 5 dias), determinando-se, fatidicamente, o substrato erodido (E), ou a ciclagem de pH associada a escovação (escova elétrica/dentifrício com flúor diluído em água, 2,5 N, 30 s, 2x/dia, após 1º e último desafios ácidos), determinando-se o substrato erodido-abrasionado (EA). Quanto à análise qualitativa, nota-se que tags resinosos se manifestam em maior número e comprimento nos substratos submetidos a desafio erosivo inicial, ou seja, o erodido e o erodido-abrasionado, independentemente da aplicação de qualquer estratégia para controle da progressão do desgaste. Dentre eles, para o substrato erodido, há uma identificável camada escura subjacente à de adesivo propriamente dito, entre os tags resinosos, representação da menor concentração de material marcado por rodamina B. Para o substrato erodido-abrasionado, verifica-se camada híbrida devidamente expressa por anuviado vermelho subjacente à camada de adesivo, mais e menos espessa, respectivamente, que a estabelecida no substrato normal e erodido. Destarte, conclui-se que o gel de CHX não foi capaz de determinar, por meio da preservação/controle do desgaste erosivo, ou erosivo-abrasivo, substrato tão favorável à adesão com os materiais resinosos quanto o normal, mesmo que não o tenha desajudado ainda mais. / The establishment of adhesive interfaces on artificially eroded dentin substrate, conceptually poor in minerals, coated with an exposed organic matrix, is little promising. For the time being, only diamond bur surface roughening and deproteinization with sodium hypochlorite, when universal adhesives are used, make it similar to the obtained for normal/sound dentin. Maintaining the layer of collagen fibrils, using protease inhibitors, perhaps even when compressed by abrasion, may, however, be convenient, since it plays an important role in the demineralization and remineralization processes, including the erosive and erosive-abrasive wear progression, and in establishing a hybrid layer itself. The aim of this study was to evaluate, immediately, bond strength (BS) of an etch-and-rinse adhesive system plus a composite to dentin, considering the condition of its substrate in three levels (N: normal; E: eroded; EA: eroded-abraded), and the application of a gel for the prevention/control of the erosive wear in three levels (C: control / without application, P: placebo / without an active principle, and CHX: 0.12% chlorhexidine digluconate). To determine the normal substrate, occlusal dentin from third molars was subjected solely to the action of a SiC sandpaper (# 600; 1 min; N); to determine the eroded and eroded-abraded, it was sequentially, and readily, subjected to an initial erosive challenge (Coca-Cola®, 5 min). One of the gels were applied, or not (control / without application), on it: placebo / without active principle; or containing 0.12% CHX. The initial demineralized one was still submitted to a pH cycling (Coca-Cola®, immersions of 5 min, 3x /day, 5 days), determining the eroded substrate (E), or to the pH cycling associated to brushing (electric toothbrush/fluoride toothpaste diluted in water, 2.5 N, 30 s, 2x/day, after 1st and last acid challenges), determining the eroded-abraded substrate (EA). After acid-etching (37% H3PO4, 15 sec, 30 sec washing, drying with absorbent paper), Adper(TM) Single Bond 2® adhesive was applied to all specimens and resin composite buildups constructed with Filtek(TM)Z350®. As for the qualitative analysis, it is noticed that resinous tags are manifested in greater number and length in the substrates submitted to erosive initial challenge, that is, eroded and erodedabraded substrates, regardless of the application of any strategy to control progression of wear. Among them, for the eroded substrate, there is an identifiable dark layer underlying that of the adhesive itself, among the resin tags, representing the lowest concentration of material marked by rhodamine B. For the eroded-abraded substrate, hybrid layer is duly expressed by a red blur underlying the layer of adhesive, more and less thick than that established in the normal and eroded substrates, respectively. Therefore, it is concluded that the CHX gel was not able to determine, while preserving/controlling the erosive or erosive-abrasive wear, a substrate as favorable to bonding with resinous materials as a sound one, even if it has not impaired it even more.

No. of bitstreams: 0 Previous issue date: 2009-01-30 / The aim of this study was to evaluate the effect of different concentrations of 2-hydroxyethyl methacrylate (HEMA) triethyleneglycol dimethacrylate (TEGDMA) on the inhibition of matrix metalloproteinase 2 (MMP-2). Mouse gingival explants were cultured overnight in DMEM and the expression of secreted enzymes was analyzed by gelatin zymography in buffers containing 5 mM CaCl2 (Tris-CaCl2) in 50 mM Tris-HCl buffer with the addition of HEMA and TEGDMA at different concentrations (0.62, 1.25, 2.5 or 5.0% (v/v)). The gelatinolytic proteinase present in the conditioned media was characterized as matrix metalloproteinase by means of specific chemical inhibition with 0.5 mM of EDTA and 0.5 mM of NEM. The matrix metalloproteinases present in the conditioned media were characterized as MMP-2 by immunoprecipitation.The eletrophoretic bands were scanned and the transmittance values were analyzed with ImageJ software. Data was plotted and submitted to linear regression to investigate MMP-2 inhibition as a function of HEMA and TEGDMA concentration. Three major bands were detected in the zymographic assays. Those bands were characterized as MMP-2. Zymogene (72 kDa), intermediate (66 kDa) and active forms of MMP-2 (62 kDa) were inhibited by HEMA and TEGDMA in a dose-dependent way. These findings suggest that HEMA and TEGDMA could inhibit MMP-2 expression even at small concentrations. / O objetivo do presente estudo foi avaliar o efeito de diferentes concentrações de 2-hidroxietil metacrilato (HEMA) e trietilenoglicol dimetacrilato (TEGDMA) na inibição da metaloproteinase da matriz 2 (MMP-2). Tecidos gengivais de ratos foram incubados em DMEM e a expressão das enzimas secretadas foi analisada por zimografia em tampões de incubação contendo 5 mM de CaCl2 (Tris-CaCl2) em 50 mM de Tris-HCl com a adição de diferentes concentrações de HEMA e TEGDMA (0,62; 1,25; 2,5 e 5%) em volume. Para caracterizar as enzimas como MMPs foi realizado ensaio de inibição química específica com 0,5 mM de EDTA (um conhecido inibidor de MMPs) e 0,5 de NEM (um conhecido inibidor de proteinases serinas).Para caracterizar as enzimas como MMP-2 foi realizado o ensaio de imunoprecipitação. As bandas produzidas no gel foram escaneadas e os valores de transmitância foram analizados com o auxílio do programa ImageJ. Os resultados foram submetidos a regressão linear em função da concentração de HEMA e TEGDMA. Três bandas foram identificadas após a zimografia. Essas bandas foram caracterizadas como MMP-2. O zimogênio (72 kDa), a forma intermediária (66 kDa) e a forma ativa da MMP-2 (62 kDa) foram inibidas pelo HEMA e pelo TEGDMA de forma dose-dependente. Nossos resultados sugerem que o HEMA e o TEGDMA podem inibir a expressão da MMP-2 mesmo em pequenas concentrações.
