Studies of viral and cellular proteins involved in herpes simplex virus type-1 egress

Ahmed, Md Firoz January 2019 (has links)
The egress pathway of herpes simplex virus-1 (HSV-1) is a complicated process mediated by co-ordinated activity of several virus glycoproteins. The virions are first assembled and enveloped at trans-Golgi-network (TGN) or endosome membranes and then travel through a guided pathway that is directed towards the cell adherent points for secretion. Once secreted the vast majority of virions remain associated with the extracellular membrane of cells and very few free virions are released into the culture medium (< 1%). The mechanisms that mediate both the targeted secretion of newly assembled virions at cell contact points and post-secretion attachment of virions with the extracellular surface of cells are poorly understood, and were the topics of this research. In this thesis, an HSV-1 passage mutant of increased virion secretion phenotype had been studied. Genome sequencing of the mutant virus identified mutations in three viral envelope proteins. Study of recombinant viruses that were constructed based on those three mutations revealed that a single amino acid change in glycoprotein I (gI) of glycine to arginine at residue 39 is responsible for the increased release of virus. The result suggests the principal effect of this mutation is to modify the secretory pathway used by virions during their release from infected cells. Data also suggests a role of gC in the attachment of virions to the extracellular surface of cells after egress. In the context of HSV-1 envelopment and egress glycoprotein E (gE), which forms a heterodimeric complex with gI (gE/gI), is known to be important. The gE/gI complex has been shown to interact with many tegument proteins and have a redundant role in secondary envelopment. The gE/gI complex has been also proposed to colocalise with various cellular components and sort the nascent virions to cell contact points. However, there is little understanding of the cellular proteins that gE/gI interact with, or the mechanisms that mediate targeted secretion of virions. This research has identified a novel interactome of gE/gI by mass-spectrometric analysis utilising stable isotope labelling with amino acids in cell culture (SILAC) medium. Among the cellular interactome obtained, Nipsnap1 was validated by co-precipitation assays from both infected and transfected cells, and furthermore using cell free systems, suggesting gE and Nipsnap1 directly interact. Nipsnap1 and its homologue Nipsnap2 have been proposed to contribute in vesicle transport and membrane fusion in cells. Using CRISPR-Cas9 technology these proteins were knocked out in a keratinocyte cell line (HaCaT) to investigate their role in HSV-1 egress. However, little or no effect on HSV-1 egress could be observed upon loss of either or both of these proteins suggesting the biological significance of gE-Nipsnap1 interaction may not be directly linked to any egress function of gE/gI. Two further interesting 'hits' from the gE/gI interactome were interferon-induced transmembrane protein type-2 (IFITM2), a virus restriction factor, and Myoferlin that has a putative role in endocytic vesicle recycling. This study could validate gE-Myoferlin interaction and co-localisation in infected or transfected cells however, functional significance of this interaction remains to be determined. Overall, the research of this thesis has provided a better understanding of the role of the gE/gI complex in HSV-1 egress and investigated the role of some interesting cellular proteins in the context of virion egress.

Foamy Virus Budding and Release

Hütter, Sylvia, Zurnic, Irena, Lindemann, Dirk 28 November 2013 (has links) (PDF)
Like all other viruses, a successful egress of functional particles from infected cells is a prerequisite for foamy virus (FV) spread within the host. The budding process of FVs involves steps, which are shared by other retroviruses, such as interaction of the capsid protein with components of cellular vacuolar protein sorting (Vps) machinery via late domains identified in some FV capsid proteins. Additionally, there are features of the FV budding strategy quite unique to the spumaretroviruses. This includes secretion of non-infectious subviral particles and a strict dependence on capsid-glycoprotein interaction for release of infectious virions from the cells. Virus-like particle release is not possible since FV capsid proteins lack a membrane-targeting signal. It is noteworthy that in experimental systems, the important capsid-glycoprotein interaction could be bypassed by fusing heterologous membrane-targeting signals to the capsid protein, thus enabling glycoprotein-independent egress. Aside from that, other systems have been developed to enable envelopment of FV capsids by heterologous Env proteins. In this review article, we will summarize the current knowledge on FV budding, the viral components and their domains involved as well as alternative and artificial ways to promote budding of FV particle structures, a feature important for alteration of target tissue tropism of FV-based gene transfer systems.

The paper aims to evaluate, by hearing egress students from the program of Post-Graduation strictosensu in Education of PUC-Campinas, from 1993 to 2004, the contributions brought by the course to their trajectory of personal and professional lives and to their institutional insertion. As a methodology, the qualitative research was used, through the application of a questionnaire sent via e-mail to 135 students of that period. Only 56 students replied. First of all, the Project of the Program of Post Graduation strictosensu was analyzed; then, other voices were heard such as the ones from External Evaluation Organs (CAPES) and the ones from the egress students. From that, a significance net was built, justifying the importance degree of an evaluatory, dynamic and shared process. Finally, the present research aims to give subsidies to the Program, suggesting changes that may be necessary in order to maintain its pedagogical propositions according to the transforming challenges of contemporary society. The studied information was debated to suggest strategies that may lead to a shared, dynamic internal evaluation, opened to the academic community and that can show a formative character to the evaluation process. Thus, the strengthening and improvement of the Program and its members can be reached. The trajectory of the egress student after leaving the course and the development of his/her qualification process were focused. It was noticed that values built during the course allowed most of the egresses of the given period to excel in subjects concerned to Education, mainly in higher education. Mestrado profissional em saúde: interações estabelecidas entre centro de pesquisa e unidades de trabalho / Professional Master in Health: interactions established between research center and work units

Trigo, Fernando Rossi 21 December 2017 (has links)
The postgraduate system brought contributions to universities, mainly for the improvement in the quality of education and knowledge production. Established in the decade of 1960s, with the purpose of qualifying professionals, mostly professors of the university itself, it aimed at expanding their training in research and, as a result, encouraging knowledge production. In this sense, teaching and research have been strengthened. By the end of the decade of 1990, CAPES recognized the Professional Master - stricto sensu - as a postgraduate modality. Focusing on the professionals in business, it proposes an approach to research, with the commitment of reaching and modifying work environment. In parallel, in 1990, the Blood Center of the Clinical Hospital of the Ribeirão Preto Medical School of the University of São Paulo was founded. With the initial proposal of providing assistance and care to the population, this health center has lived with the research carried out in the academy, what enabled to reconcile knowledge production with the practice developed in its daily routine. In 2012, it launched the professional master in Hemotherapy and Biotechnology, with the aim of training and qualifying professionals in this field. In this work, the objective was to study this program, proposing to investigate the mechanisms of interaction established between the research center and partner work units, from the perspective of egress students (1st and 2nd classes). Because it has an exploratory and initial character, we planned to work with indicators exclusive to the professional modality, allowing to strengthen the follow-up and evaluation of the program offered, mainly those indicators related to the applicability of projects and to the impact of the course done on the professional act of the egress student. Therefore, the present work is an exploratory-descriptive study using quantitative and qualitative approaches, with six major investigation topics: the program offered; the research project developed; intellectual production; the unfolding of the experience of groups of professionals from different states of Brazil; the contribution of the supervisor for the development of research projects; and the contribution resulting from the approach of students to research. With the features of specific training and possibility of gathering professionals of the area of Hemotherapy from the nine regions of Brazil, the program has enabled to accomplish the objectives expected in the professional modality (stricto sensu) and established in the project creation in 2011. Part of these results is owed primarily to three mechanisms of interaction identified: the research project, experience of living as a group, and supervisor, each one with specific contributions to the process of students\' training and learning, enabling to achieve changes in work units and in professional conduct and competences. This first characterization of the proposal allows to create a comparison point for the recently implemented program, which will go through its first four-yearly assessment in 2017. Difficulties and limitations have also been diagnosed and must to be reviewed, restructured and managed, such as the practical content of the course, the chart of technical administrative professionals and the time for titling, with an average of 35.4 months, what demands some thoughts. The regular follow-up of egress students and creation of a continuous platform for data construction will allow to intervene and adjust the program, making possible to notice maturation and growth phases, with special attention to indicators related to the professional modality.

This kind of graduation has been widespread in the country since the mid-1990s, after the enactment of the law nº 9.394/96 - LDB, and by a series of decrees, orders and advices of the MEC / CNE. Among the outstanding main goals and objectives of the PNE for higher education, it may highlighted: diversified higher education system to meet the specific demands of clienteles training; linkage between curriculum and the labor market; financing and management; creating policies for access to higher education to facilitate the admission of students from vulnerable educational groups; increasing the of offer for higher education for the aged group 18-24 years. This study aims to reflect and discuss the importance of the CSTs in professional education of Brazilian workers through the expectation of PNE. The study is based on qualitative method, an exploratory and descriptive research, characterized as a single case study, through field research conducted with students and graduates of the course of Industrial Electronics, Faculty of Technology Senai Anchieta, in the city of Sao Paulo. To achieve its objectives, the study was divided into two stages: the first analyzes the variables that characterize the profile of the student in training in order to identify their perceptions regarding the choice by CST, knowledge about this type of education and future expectations of the profession. In the second part, the study analyzes the perception of graduate about the skills and competencies acquired during training, their degree of satisfaction with salary and career path, and the Technologist professional acceptance by the labor market. In a study conducted with students in training it was found that the dropout rates of the first classes were high, being identified as the main causes difficulties in reconciling study schedule with work and family economic hardship. The profile of the student training in IES under study is mostly the age group between 18 and 24 years; males; egress of secondary education done in public schools; resident in peripheral and natural neighborhood of the city RMSP; the student himself is responsible for payment of tuition; no third party interference in their choice of CST; have favorable perceptions regarding future employability as technologist. A survey of graduates showed that they have a favorable perception of the training received, except the fact that they had not attended courses that develop managerial skills and business; regarding the employability and career prospects were concerned in professional development technologist by the labor market; reported being dissatisfied about salary received; regarding continuing education be recognized that the process that will keep updated professionally. As a conclusion of this study, it is recommended to probing other possible causes of truancy, through a research with students from other courses with different thematic axes and public IES. It is pointed-out also the need to offer in the curriculum proposal of the IES under study, apart from technical disciplines, others in order to develop skills in people management and business management.

The research aimed to investigate how the graduated students from Tourism Course with emphasis in hotel, realized the role of their Course in their professional lives. The quantity-qualitative research involved 48 graduated students. The data were collected from a questionnaire with opened and closed questions. Concerning to the results, the search brought important information to a reflexive auto-evaluation process as: while students, the majority of them had expectations about the insertion of this professional in the tourism area, and nowadays, half of them are out of their professional area; only 33,3% declared absolutely satisfied with the course; in suggesting changes to improve the Course, the focus was in practical activities, apparently trying to translate a technician view of the Pedagogical Politic Project of the Course. Concluding, the dialogue with the graduated students was essential to renew the thought of the Course staff bringing challenge questions.
