Análise genômicae suscetibilidade de Pseudomonas aeruginosa isoladas de rede de água de consultórios odontológicos da cidade de Barretos-SP /

Orientador: Fernando Antônio de Ávila / Banca: José Moacir Marim / Banca: Patricia Amoroso / Banca: Simone Barone Salgado Marques / Banca: Tammy Priscilla Chioda Delfino / Resumo: Foram estudadas 180 amostras de água de 45 consultórios odontológicos da cidade de Barretos-SP, com o objetivo de isolar, identificar, determinar a contagem de isolados de Pseudomonas aeruginosa UFC/mL, determinar o perfil clonal e avaliar a diversidade genômica dos isolados e a suscetibilidade das mesmas frente a diferentes antibióticos. As amostras de água foram filtradas em filtro Millipore® e a membrana colocada sobre o centro de uma placa de Petri contendo agar cetrimida, As colônias típicas de bactérias do gênero Pseudomonas foram identificadas pelo método de Gram, inoculação em TSI agar (Triple Sugar Iron), crescimento a 42ºC, produção de pigmento, produção de alginato, oxidase, motilidade e alcalinização da acetamida. O teste utilizado para análise genômica foi o ERIC-PCR. Dos 76 (42,2%) isolados de Pseudomonas aeruginosa, 15 eram provenientes das amostras de torneira de lavagem das mãos, 18 de reservatório de garrafa pet, 23 de seringa tríplice e 20 do motor de alta rotação. Todos os isolados de Pseudomonas aeruginosa foram submetidos ao teste de suscetibilidade segundo a técnica de Kirby- Bauer. Dos antibióticos testados o que apresentou melhor resultado quanto à sensibilidade (65,8%) foi a ciprofloxacina. Quanto à similaridade genética dos isolados dos diferentes pontos analisados, foram encontrados nove "clusters" de 100% de similaridade. / Abstract: The biofilm found in water supplies and lines of hospitals and dental units is extremely important because it presents a large number of bacteria, leading to risk of infection in immunocompromised patients vulnerable to opportunistic pathogens such as the Pseudomonas aeruginosa. One hundred eighty water samples from dental units of the city of Barretos-SP were evaluated. The water samples filtered in Millipore® filter were incubated in plates containing Cetrimide ágar. The bacteria colonies were identified through the gram-staining test, inoculation in agar T.S.I (triple sugar Iron), growth at 42 ° C, pigment production, production of alginate, oxidase acetamide alkalization and motility observation. All Pseudomonas aeruginosa bacteria were submitted to the susceptibility test according to the Kirby-Bauer technique. The test used for genomic analysis was the ERIC-PCR. From the total microorganisms studied, 76 (42,2%) were positive for Pseudomonas aeruginosa, isolates from 180 water samples, where 15 strains were from hand washing incoming local water supplies, 18 from PET bottled water, 23 from 3-in-1 syringes and 20 from the high speed handpiece. In relation to the antibiotics tested, the one presenting the best result with regard to sensibility was ciprofloxacin with 65.8%. Byron's Shakespearean Imitations

Though Byron is known for his provocative denials of the importance of Shakespeare, his public derogations of the early modern playwright are in fact a pose that hides the respect he had for the playwright’s powerful poetic vision, a regard which is recorded most comprehensively in the Shakespearean references of Don Juan. Byron imitated Shakespeare by repeating and adapting the older poet’s observations on the imitative nature of desire and the structure of emulous ambition as a source of violence. His appropriations make his work part of the modern shift away from earlier European societies, wherein ritual means of mitigating desire’s potentially inimical impact on human communities were supplemented with an increased reliance on market mechanisms to defer the effects of emulation and resentment. Finding himself among the first modern celebrities, Byron deploys Shakespeare’s representations of desire to trace the processes that produced the arc of his own fame and notoriety. Drawing on his deep knowledge of Shakespeare, Byron’s poetic vision—in its observations on the contagious nature of desire—exhibits elements of Shakespeare’s own vivid depictions of imitation as a key conduit for his characters’ cupidity, ambitions, and violence. Exploring how he plays with and integrates these representations into his letters, journals, poetry, and plays, my dissertation investigates Byron’s intuitions on the nature of human desire by focusing on his engagement with one of literature’s greatest observers of human behaviour, Shakespeare.

I denna idéanalytiska och jämförande fallstudien kommer fyra samtida högerextrema manifest att analyseras för att utröna vilka bakomliggande idéer respektive författare ger uttryck för. Till grund för denna analys kommer ideologin ekofascism användas för att utröna ifall deras innehåll överensstämmer med denna ideologi. Därutöver kommer eventuella likheter och skillnader mellan manifesten presenteras. Ett analytiskt ramverk för ekofascism kommer att operationaliseras utifrån skrifter från två filosofer, Pentti Linkola och Savitri Devi. Detta ramverk kommer sedermera appliceras tematiskt på de valda manifesten för att besvara de uppställda frågeställningar. Manifesten har valts för att möjliggöra en komparation över tid, då det första manifestet var skrivet 1995, det andra 2007, och de tre sista 2019. Uppsatsens viktigaste slutsats är att det skett ett idémässigt förskjutande över tid från de två första manifesten till de tre sista, där dessa kännetecknas av rasistiska och/eller antisemitiska tankegångar. Slutligen fann uppsatsen att i alla manifest, bortsett från Kaczynskis, var eventuella eko-tankegångar underordnade andra problemområden som dess författare identifierat.

Transactional Analysis in the Elementary Classroom: PAC for Children

The focus of this study is on the development of an original script designed to introduce concepts of structural and transactional analysis to elementary school children. Included in Chapter One are reviews of Transactional Analysis and the PAC communication model. Classroom application of Transactional Analysis principles is examined in Chapter Two. Chapter Three examines needs and characteristics of young children. Qualities of good children's literature are discussed in relationship to the selection and explication of script material. The manuscript appears in Appendix B. This report accepts evidence that Transactional Analysis training can be an additive part of the elementary school curriculum. It further proposes that story material conscientiously designed for young children could prove effective and entertaining training vehicles for Transactional Analysis concepts.

O fio aparente que liga as produções destes dois observadores históricos do século XX é a preocupação de ambos com o homem comum em detrimento aos fatos históricos e aos espaços geográficos demarcados. O presente exame espera demonstrar como a história ocidental no século XX infiltra-se na particular inscrita desse autor brasileiro, seja pelo cosmopolitismo do terror forjado em ―O mau humor de Wotan‖, seja pelo remoto sertão caracterizado como o próprio mundo pela fala do protagonista Riobaldo. Acerca desta obra, afirma-se que esta topografia, diferente da tradição regionalista, se erige tal qual uma metonímia de todos os lugares, portanto, distante de um saudosismo sertanejo. Alguns exemplos dessa ressonância histórica abundam nesse romance, como os grandes fenômenos vivenciados no século passado: a emancipação feminina e a crítica aos modelos liberais, os quais geraram os bandidos sociais em algumas regiões do globo, e com estes um acontecimento específico na passagem do século XIX para o XX: a eclosão dos primeiros Estados-paralelos de procedência rural originada pelas catástrofes ocorridas na Europa, as quais levaram as personagens das narrativas ―A velha‖ e ―A senhora dos segredos‖ a acreditarem na ilusão de liberdade em território brasileiro. Assim, os temas da ―nova mulher‖ e dos movimentos de resistência social nas periféricas zonas do capitalismo são de grande relevância tanto para a obra rosiana, quanto para o trabalho deste intelectual britânico, pois, em ambos, a documentação histórica se constitui em algo circunstancial em que as dimensões míticas dialogam com as composições ideológicas. Tendo a ambiguidade como a tônica das relações humanas e de poder do século XX e da ficção rosiana, o jagunço ficcional ora se assemelha ao paladino robinwoodiano, arquétipo ideal do bom bandido, quanto ao criminoso comum, figura proscrita pelas leis do Estado e pela aceitação do público geral. Este trabalho analisa — com o auxílio da Estética da Recepção e das contribuições críticas de Antonio Candido (1918-2017) — o percurso traçado pelas sociedades ocidentais no breve século XX no intuito de encontrar outras formas de subexistir em meio à desintegração dos valores desenhados pelo iluminismo setecentista. Integrando essas construções estético-científicas, é possível estabelecer uma interpretação mais completa de uma das muitas faces da realidade contemporânea, época em que o globo, perplexo, observou ruir impérios e a civilidade diante da barbárie praticada em regiões, como o sertão (real ou metafórico), esquecidas pelo capitalismo e pelo desgastado poder público. Nesta dialética, por um lado, amplia-se o estudo do tema do banditismo social hobsbawmiano, acrescentando-se à sua tipologia a figura do jagunço, por outro, denota-se em Guimarães Rosa uma atitude excepcional diante da violência e da barbárie observada em um período de exceção como o nosso em que todas as escritas se configuram em cantos de sobrevivência ou réquiens da liberdade. / This thesis proposes a comparative study of the works of João Guimarães Rosa (1908-1967) — especially the novel Grande sertão: veredas (1956) and the German ―cronicontos‖ Ave, Palavra (1970) — and the historiography of Eric Hobsbawm (1917-2012), clustered in Bandits (1969) and Age of extremes (1994). The apparent thread linking the productions of these two historical observers of the twentieth century is the concern of both with the common man over the historical facts and the demarcated geographical spaces. This survey hopes to show how Western history in the twentieth century infiltrates the particular writing of this Brazilian author, either by the cosmopolitanism of the horror forged in ―O Mau Humor de Wotan‖, or by the remote hinterland characterized as the world itself by the speech of the protagonist Riobaldo. About this work, it is stated that this topography, different from the regionalist tradition, is erected just like a metonymy of all places, therefore, far from a backcountry nostalgia. Some examples of this historical resonance abound in this novel, as the great phenomena experienced in the past century: the emancipation of women and the criticism of the liberal models, which generated social bandits in some regions of the globe, and with these a specific event in the turn of the XIX century: the emergence of the first states-parallel from rural areas caused by disasters in Europe, which led the characters of the narrative ―A Velha‖ and ―A Senhora dos Segredos‖ to believe in the illusion of freedom in Brazil. Thus, the themes of the ―new woman‖ and of social resistance movements in peripheral capitalism areas are of great importance both for Rosa‘s work, and for the work of this British intellectual, because in both the historical documentation constitutes something circumstantial in which the mythic dimensions dialogue with the ideological compositions. Having ambiguity as the keynote of human relationships and power of the twentieth century and Rosa‘s fiction, the fictional bandit now resembles the Robin Wood‘s Paladin, ideal archetype of good villain, as the common criminal, figure proscribed by the laws of the State and the acceptance of the general public. This paper analyzes — with the help of Aesthetics of Reception and critical contributions of Antonio Candido (1918-2017) — the route traced by the Western societies in the short XX century in order to find other ways to coexist amid the disintegration of the values designed by the eighteenth-century Enlightenment. By integrating these aesthetic and scientific constructions, it is possible to establish a more complete interpretation of one of the many faces of contemporary reality, time in which the perplexed globe watched crumbling empires and civility in the face of barbarism practiced in regions such as the hinterland (real or metaphorical) forgotten by the capitalism and worn government. This dialectic, on one hand, broadens the study of the theme of Hobsbawm‘s social banditry, adding to his typology the gangster figure. On the other hand, it denotes in Guimarães Rosa an exceptional attitude against the violence and barbarism seen in a period of exception, like ours, in which all the writings are configured in survival corners or requiems of freedom. / En esta Tesis es propuesto un estudio comparativo entre las obras de João Guimarães Rosa (1908-1967) — con destaque para el romance Grande sertão: veredas (1956) y los ―cronicontos‖ alemanes de Ave, palavra (1970) — y la historiografía de Eric Hobsbawm (1917-2012), alzada en Bandidos (1969) y Era dos Extremos (1994). El hilo aparente que liga las producciones de estos dos observadores históricos del siglo XX es la preocupación de ambos con el hombre común en detrimento a los hechos históricos y a los espacios geográficos demarcados. Cette thèse interroge le destin de Judas chez cinq écrivains : Paul Claudel (Mort deJudas), Marcel Pagnol (Judas), Jean Ferniot (Saint Judas), Armel Job (Judas le bien-aimé)et Eric-Emmanuel Schmitt (L’Évangile selon Pilate). Passant du cadre général à celuiparticulier, la recherche se propose d’envisager la figure de Judas dans les transpositionsbibliques contemporaines et d’évaluer les nouvelles visions que le siècle a inventées.L’analyse comprend des approches s’appuyant sur les contributions du domaine du mythe,de l’exégèse biblique, historique, de la sociologie, de la psychanalyse. La plupart des récitscontemporains font de Judas un sujet moderne très complexe : il devient figure de l’artiste,du philosophe et de l’acteur.Les problématiques d’ordre social et politique se révèlent primordiales : dans uncontexte succédant à l’Holocauste, Judas renoue avec l’univers judaïque. Retranscrivant lemythe du Juif errant, Judas devient figure de la scission, du départ et de l’errance ; lesrelations au sein de la famille sont vivement remises en cause. La figure du traître devientaussi le support d’une parole restitutive. L’idiotie de filiation dostoïevskienne attribuée àJudas fait bon ménage avec le contexte littéraire contemporain ainsi qu’avec celuilégendaire réévalué.Constamment hésitant entre la réhabilitation de Judas proposée par Pagnol, Ferniot,Job, Schmitt et la réprobation du personnage par Claudel, le parcours propose un richeréseau construit autour de thématiques communes mais différemment évaluées. / This doctoral dissertation questions Juda's destiny in five authors: Paul Claudel(The Death of Juda), Marcel Pagnol (Juda), Jean Ferniot (Saint Juda), Armel Job (TheBeloved Juda) and Eric-Emmanuel Schmitt (The Gospel according to Pilate). Fromgeneral to specific, this research aims at outlining the image of Judas within thecontemporary biblical renditions and to evaluate new perspectives on it. This analysistouches upon myth, biblical and historical exegesis, sociology and psychoanalysis. Mostcontemporary stories make Juda a very complex and modern subject: he embodies theartist, the philosopher, the actor.Social and political issues prove to be crucial: in a context which follows theHolocaust, Juda reconnects with the Judaic universe. By rewriting the myth of thewandering Jew, Juda becomes the figure of the scission, of the departure and of the roving;family relationships are strongly questioned. Furthermore, the figure of the traitor becomesthe repository for a restitutive speech. The idiocy, in the Dostoievskian sense, associatedwith Judas, blends well both with the contemporary literary context as well as with thereconsidered mythical one.Constantly hesitating between Judas' redemption that Pagnol, Ferniot, Job andSchmitt adopt and the condemnation of Claudel's character, this thesis focuses on aconvoluted network built around common themes that are tackled differently.

Ny betygsreform-Lpo94- En implementeringsstudie om två kommuners erfarenheter av den nya betygsreformen

Riksdagen antog 1990 regeringens proposition om ansvaret för skolan. Beslutet innebar en klarare ansvarsfördelning mellan staten och kommunerna när det gäller verksamheten inom exempelvis grundskolan. Denna nya mål- och resultatstyrning innebär att riksdag och regeringen anger nationella mål och riktlinjer för arbetet i skolan. Målen skall gälla för alla skolor och skall garantera att utbildningen i landet blir likvärdig. Inom dessa ramar som staten anger är det kommunerna som har huvudansvaret för verksamheten. Kommunerna har stor frihet att avgöra hur verksamheten skall organiseras för att de nationella målen skall uppnås. En av de centrala delarna är att svara för att skolan får de resurser och de förutsättningar i övrigt som behövs. Denna uppsats belyser hur två kommuner har implementerat samt bearbetat den nya betygsreformen vidare diskuteras vad för orsaker som ligger bakom det stora antalet elever med icke godkänt i ett eller flera ämnen. Kan implementeringen av det nya betygssystemet ses som en förklaring till detta och hur skall problemet bearbetas. För att belysa detta har intervjuer med lärare och politiker genomförts i de två kommunerna
