TRANS*VERSAL ANIMACIES AND THE MATTERING OF BLACK TRANS* POLITICAL LIFE

This article explores trans*versal connections between transness, blackness, and the animal. Drawn from the conceptual vocabulary of cultural theorist Felix Guattari, this article argues that the central purpose of transversality is to create linkages between previously unexplored singularities in a field, and then to create connections in other conceptual topographies at different levels of discursivity. The article advances an extension of Guattari's transversal into a more capacious concept of the trans*versal, to analyze the #blacklivesmatter and #blacktranslivematter movements that draw on critical animal studies to reveal ways that species hierarchies are always present in processes of racialization that allow some lives to matter more, or less, than others.

La spectroscopie infrarouge permet d’identifier la structure 3D de systèmes moléculaires, par comparaison des spectres mesurés et simulés. Nous travaillons en phase gazeuse, où les molécules et clusters sont libres d’interactions intermoléculaires. Notre travail combine les expériences IR-UV ion-dip et le calcul de spectres IR anharmoniques par la méthode DFT-MD. Le spectre IR est calculé pour les structures 3D de plus basses énergies,le meilleur accord donnant la connaissance de la structure présente dans les conditions expérimentales.Nous démontrons que le domaine de l’IR lointain/THz (<800 cm-1, <24 THz) permet d’identifier sans ambiguïté la structure 3D de molécules et clusters en phase gazeuse, là ou les signatures du domaine 1000-4000 cm-1 peuvent être limitées. Les systèmes considérés sont des dipeptides, un modèle de feuillet β, dérivés du phénol (et complexés à l’eau) des paires de bases de l’ADN, dont les structures sont bâties sur des liaisons hydrogène intra/intermoléculaires. / Infrared spectroscopy allows the assignment of three dimensional structures of molecular systems, by comparing experimental and theoretical spectra. Our investigations take place in the gas phase, where molecules and clusters are free of intermolecular interactions.Our work combines experimental IR-UV ion dip spectroscopy and theoretical DFT-MD anharmonic spectroscopy. The infrared spectrum is calculated for low energy 3Dstructures and the best match between theory and experiment provides the information about the structure present in the experimental conditions.We demonstrate for several systems that far infrared/THz spectroscopy (<800 cm-1, <24THz) allows conformational assignment without ambiguities, contrary to the more traditional 1000-4000 cm-1 range. Systems investigated here are dipeptides, a β-sheet model, phenol derivatives (also complexe dwith water molecules), DNA base pairs, all these structures being built on intra-/intermolecular hydrogen bonds.

Characterization of the Bacteriophage Felix O1 Endolysin and Potential Application for Salmonella Bioremediation

There is an increasing incidence of antimicrobial-resistant organisms isolated from food and food products. Coupled with that rising incidence is increased media scrutiny and coverage of outbreaks of foodborne illnesses. Consequently, consumers increasingly demand safer food, and that the antimicrobial measures used be other than antimicrobial drugs. A possible solution is to use bacteriophages, or the purified holin and endolysin proteins that make them lethal and lytic, as antimicrobial food treatments or additives. The bacteriophage Felix O1 is a promising candidate for development as an anti-Salmonella food treatment. This dissertation describes the work done to determine if these proteins could be of value as bioremedial agents. Endolysin treatments of Gram negative bacteria require two agents: the lytic endolysin, and a second agent to permeabilize the outer membrane of the bacterium. The holin protein was proposed as an outer membrane permeabilization agent. Methods used to locate the holin gene included BLAST analysis, analysis of putative Felix O1 proteins for transmembrane domains, and examination of the lysin sequence for an N-terminal signal sequence. Analyses did not reveal a promising candidate. Cloning of rIIA as a potential holin was attempted without success. Results of various analyses are discussed, as are chemical alternatives to the use of purified holin as a permeabilization agent. The endolysin, LysO1, was successfully cloned and characterized. PHYRE analysis predicted that the enzyme structure is composed of α helices arranged into two lobes, with the active site in a cleft between them. The enzyme lysed all tested strains of Salmonella and a tested strain of the foodborne pathogen Escherichia coli. Campylobacter jejuni susceptibility remains ambiguous, and the enzyme had no effect on Listeria monocytogenes or Micrococcus luteus. LysO1 was most active at alkaline pH and low ionic strength; optimal activity was observed in 25 mM buffer at pH 10. If removed from frozen storage, the enzyme was most thermostable at 30 °C. Lytic activity was adversely affected by the presence of the divalent cations calcium, magnesium, and zinc, and by high ionic strength. Considerable time was devoted to development of the activity assay used to further characterize the enzyme, and details of those experiments are provided. Logical extensions of the research project, such as further characterization and testing needed to obtain government approval for widespread use of the treatment, and possible pursuit of treatment based on an enzyme derivative such as an antimicrobial peptide, are discussed. / Ph. Comoumsótrabalho - sobre os projetos de escala geográfica de Nelson Felix investiga um conjunto específico de obras do artista, precisamente aquelas cuja construção envolve deslocamentos geográficos extremos, uma experiência dilatada de temporalidade e a premissa do entrelaçamento contínuo entre os trabalhos. Marcada pelo rigor construtivo e por uma série de determinações simbólicas, a produção de Felix recorre a formas orgânicas, a ciclos planetários, à música, à poesia escrita, em um jogo de equivalências e rebatimentos que se manifesta em uma forma constelar, simultaneamente atenta e insubordinada aos paradigmas estéticos contemporâneos. Assim, partindo de reflexões que problematizam a relação ambivalente da obra de Nelson Felix com a tradição da escultura a pesquisa encontra, nos processos de construção dos projetos do artista, os conceitos que irão nortear a discussão em pauta. Essas reflexões se atêm, basicamente, à contiguidade entre os trabalhos, à temporalidade supra-humana a que são submetidos e à monumental invisibilidade da obra. / Comoumsótrabalho (Asasinglejob) - about the geographic scale works of Nelson Felix - investigates a specific set of works by the artist, precisely those whose construction involves extreme geographic dislocation, a dilated temporal experience and the premise of continuous interconnection between all of the artist\'s works. Marked by the constructive rigor and by a series of symbolic determinations, Felix\'s production turns to organic forms, planetary cycles, music and written poetry, in a game of equivalences and reverberation that presents itself in a stellar way, simultaneously aware of and unsubordinated to the contemporary aesthetic paradigms. Thus, starting from reflections that problematize the ambivalent relations between the work of Nelson Felix and the tradition of sculpture, the research finds, in the processes of construction of projects by the artist, the concepts that will guide this discussion. These reflexes basically rely on continuation of works, on the super-human temporality to which they are submitted and on the monumental invisibility of the work.
