Caratterizzazione di genotipi di sorgo in funzione di tratti legati alla tolleranza alla siccità / CHARACTERIZATION OF SORGHUM GENOTYPES FOR TRAITS RELATED TO DROUGHT TOLERANCE

L’incessante aumento della popolazione mondiale ed il conseguente incremento della richiesta di risorse alimentari ed energetiche, congiuntamente al mutevole scenario climatico, sempre più incline a periodi di siccità prolungata in misura sempre maggiore in alcune zone del pianeta, fa sì che sempre più attenzione sia rivolta allo sviluppo ed all’implementazione di risorse energetiche rinnovabili a bassi input. Il sorgo zuccherino (Sorghum bicolor Moench) è una coltura bioenergetica in grado di fornire cibo, bioetanolo e biogas. Lo studio di tale coltura in risposta al deficit idrico promuove una più approfondita conoscenza dei meccanismi alla base dei processi fotosintetici, e di come, e quanto, questi possano essere influenzati dall’assenza temporanea, o più o meno prolungata, di disponibilità idrica. La produzione di biomassa e la sua composizione chimica sono state valutate per genotipi di nuova costituzione in confronto a quelli già disponibili in commercio, ai fini della produzione di biogas e bioetanolo. Una più approfondita analisi fisio-fenologica e molecolare è stata condotta su sei genotipi di sorgo con lo scopo di combinare in una visione di insieme più integrata la risposta alla siccità in sorgo. Due genotipi (uno sensibile e l’altro tollerante la siccità) sono stati selezionati per l’analisi trascrittomica in risposta allo stress idrico al fine di individuare geni candidati potenzialmente utili ai fini di una selezione assistita da marcatore. / Due to the increasing human population and the consequent surging energy and water demand, it is necessary to implement energy and fuel production from low input renewable sources. Sweet sorghum (Sorghum bicolor Moench) is a low input multipurpose crop that provides food, feed and bioethanol from conversion of sugars accumulated in the stalk and biogas from anaerobic digestion of whole aboveground dry biomass. This multipurpose crop was studied in response to water deficit. In particular, the biomass production and its composition were evaluated in response to drought for new developed and commercial genotypes for biogas and bioethanol production. The physiologic and molecular approaches were combined in order to provide an integrated view on drought tolerance in sorghum enabling to know which are the mechanisms and with which extent they were affected by drought in this bioenergy crop. The transcriptomic analysis was performed on two sorghum genotypes (one sensitive and the other one tolerant to drought) with RNA-Seq technology in order to evaluate the diversity existing in the sorghum transcriptome that could be related to drought tolerance and to identify candidate genes that could be used as potentially marker for the marker assisted selection.

Growing mallee eucalypts as short-rotation tree crops in the semi-arid wheatbelt of Western Australia

[Truncated abstract] Insufficient water use by annual crop and pasture species leading to costly rises in saline watertables has prompted research into potentially profitable deep-rooted perennial species in the Western Australian wheatbelt. Native mallee eucalypts are currently being developed as a short-rotation coppice crop for production of leaf oils, activated carbon and bio-electricity for low rainfall areas (300—450 mm) too dry for many of the traditional timber and forage species. The research in this study was aimed at developing a knowledge base necessary to grow and manage coppiced mallee eucalypts for both high productivity and salinity control. This firstly necessitated identification of suitable species, climatic and site requirements favourable to rapid growth, and understanding of factors likely to affect yield of the desirable leaf oil constituent, 1,8-cineole. This was undertaken using nine mallee taxa at twelve sites with two harvest regimes. E. kochii subsp. plenissima emerged as showing promise in the central and northern wheatbelt, particularly at a deep acid sand site (Gn 2.61; Northcote, 1979), so further studies focussed on physiology of its resprouting, water use and water-use efficiency at a similar site near Kalannie. Young E. kochii trees were well equipped with large numbers of meristematic foci and adequate root starch reserves to endure repeated shoot removal. The cutting season and interval between cuts were then demonstrated to have a strong influence on productivity, since first-year coppice growth was slow and root systems appeared to cease in secondary growth during the first 1.5—2.5 years after cutting. After decapitation, trees altered their physiology to promote rapid replacement of shoots. Compared to uncut trees, leaves of coppices were formed with a low carbon content per unit area, and showed high stomatal conductance accompanied by high leaf photosynthetic rates. Whole-plant water use efficiency of coppiced trees was unusually high due to their fast relative growth rates associated with preferential investments of photosynthates into regenerating canopies rather than roots. Despite relatively small leaf areas on coppice shoots over the two years following decapitation, high leaf transpiration rates resulted in coppices using water at rates far in excess of that falling as rain on the tree belt area. Water budgets showed that 20 % of the study paddock would have been needed as 0—2 year coppices in 5 m wide twin-row belts in order to maintain hydrological balance over the study period. Maximum water use occurred where uncut trees were accessing a fresh perched aquifer, but where this was not present water budgets still showed transpiration of uncut trees occurring at rates equivalent to 3—4 times rainfall incident on the tree belt canopy. In this scenario, only 10 % of the paddock surface would have been required under 5 m wide tree belts to restore hydrological balance, but competition losses in adjacent pasture would have been greater

The extraction and the lack of wood to meet demand represent the main problems of the sector. Therefore, this study aimed to evaluate the development and physiological responses of native tree species and exotic scrub under two water regimes in Baixo - AcaraÃ, CearÃ. The seedlings were grown in plastic pots containing 288 cm3 as carbonized rice husk substrate, carnauba straw chopped and hydromorphic soil volumetric proportion of 3:2:2. The experiment was conducted in Marco Ceara explored in the postharvest area. The species aroeira (Myracroduon urundeuva AllemÃo), acÃcia australiana (Acacia mangium Willd), frei jorge (Cordia trichotoma (Vellozo) ArrÃb. ex Steud.), nim (Azadirachtha indica A. Juss.), jatobà (Hymenaea courbaril L.) mogno africano (Khaya ivorensis A. Chev) e sobrasil (Colubrina glandulosa var. reizzi) were planted at 2x3 m spacing with 15 plants/row. We conducted a fertilization foundation(120 g of NPK (10-28-20) + 30 g of FTE BR 12) and semiannually and fertilizer training (50 g of NPK (10-28-20). The first 12 months the entire area was irrigated by micro which was suspended after this period until the end of the experiment only one side, dividing into two sub (plots), with and without irrigation scheme. Monthly for 36 months held up measurements of height and diameter at breast height (DBH) in 12 plants of the central row of the plot of each species in each treatment, also led physiological evaluations for individuals marked during the dry and rainy season each treatment. The experiment followed a design in repeated measures, in an arrangement with split plots. Acacia species studied stood out over the others reaching a height at 36 months of age of 15,18 m and 14,69 m under irrigation under rainfed conditions . Regarding the DBH at 36 months the acaciaa was superior to the others, but developing - better under irrigated conditions (16,88 cm) than under dryland (15,78 cm). Generally the species showed decreases in conductance, transpiration and photosynthesis when exposed to water deficit, on the other hand proved to be efficient in water use indicating strong stomatal regulation. There were no severe damage to the photosynthetic apparatus of the species. We conclude that the species have the potential to be explored in the soil and climatic conditions of the region. / O Cearà possui cerca de 750 indÃstrias moveleiras, entre mÃdias e pequenas. O extrativismo e a falta de madeira para atender a demanda representam os principais problemas do setor. Diante disso, objetivou-se nesta pesquisa avaliar o desenvolvimento e respostas fisiolÃgicas de espÃcies arbÃreas nativas da caatinga e exÃticas sob dois regimes hÃdricos no perÃmetro irrigado Baixo AcaraÃ, CearÃ. AtravÃs de mudas que foram produzidas em tubetes de 288 cm3 contendo como substrato casca de arroz carbonizada, bagana de carnaÃba triturada e solo hidromÃrfico na proporÃÃo volumÃtrica de 3:2:2. O experimento foi conduzido em AcaraÃ, Cearà em Ãrea experimental conduzida pela Embrapa AgroindÃstria Tropical, com as espÃcies aroeira (Myracroduon urundeuva AllemÃo), acÃcia (Acacia mangium Willd), frei jorge (Cordia trichotoma (Vellozo) ArrÃb. ex Steud.), nim (Azadirachtha indica A. Juss.), jatobà (Hymenaea courbaril L.) mogno africano (Khaya ivorensis A. Chev) e sobrasil (Colubrina glandulosa var. reizzi). Plantadas no espaÃamento 2x3 m em trÃs linhas com 15 plantas/linha. Efetuou-se uma adubaÃÃo de fundaÃÃo (120 g de NPK (10-28-20) + 30 g de FTE BR 12) e semestralmente adubaÃÃo de formaÃÃo (50 g de NPK (10-28-20). Nos primeiros 12 meses toda a Ãrea foi irrigada por microaspersÃo, suspensa apÃs este perÃodo atà o final do experimento somente de um lado, dividindo em duas subÃreas (parcelas), regime irrigado e irrigaÃÃo de salvamento. Semestralmente durante 36 meses procederam-se avaliaÃÃes da altura e diÃmetro a altura do peito (DAP) em 12 plantas da fileira central das espÃcies em cada regime hÃdrico. TambÃm foram conduzidas avaliaÃÃes fisiolÃgicas durante a estaÃÃo seca e chuvosa em cada regime hÃdrico. O experimento seguiu um delineamento em medidas repetidas, em um arranjo com parcelas subsubdivididas. AcÃcia destacou-se em relaÃÃo Ãs demais atingindo altura e DAP aos 36 meses de idade de 15,18 m e 16,88 cm sob irrigaÃÃo e 14,69 m e 15,78 cm sob irrigaÃÃo de salvamento. De modo geral as espÃcies apresentaram decrÃscimos na condutÃncia, transpiraÃÃo e fotossÃntese quando submetidas a dÃficit hÃdrico, por outro lado mostraram-se eficiente no uso da Ãgua indicando forte regulaÃÃo estomÃtica. NÃo foi verificado danos severos no aparato fotossintÃtico das espÃcies. Conclui-se que as espÃcies possuem potencial para serem exploradas nas condiÃÃes edafoclimÃticas da regiÃo.

Optimization of postharvest Technology for "Rojo brillante" Persimmon and Introduction of New Varieties in the Mediterranean Area

Tesis por compendio / [ES] En las últimas dos décadas, la producción de caqui en España ha aumentado exponencialmente y la superficie de cultivo se ha multiplicado casi por ocho. En la actualidad, la producción de caqui está centralizada en el cultivar 'Rojo Brillante', que es mundialmente apreciado. Sin embargo, esta centralización supone una importante limitación comercial. En la presente Tesis se abordan dos objetivos principales: 1) Optimización de la tecnología pre y postcosecha con el fin de garantizar la calidad del caqui 'Rojo Brillante' durante la frigoconservación y el transporte a mercados internacionales. 2) Evaluación del comportamiento bajo condiciones mediterráneas de variedades introducidas de otras regiones productoras con el fin de ampliar el rango varietal de caqui. El primer objetivo de esta Tesis se logró a través de los estudios presentados en los Capítulos I al V. El segundo objetivo se abordó en los Capítulos VI y VII. En el Capítulo I se esclarecieron las causas del pardeamiento interno del fruto en los envíos a los mercados internacionales. La temperatura a la que se expone la fruta inmediatamente tras la aplicación del tratamiento de desastringencia con altas concentraciones de CO2 se identificó como el principal factor implicado en esta alteración. Los resultados obtenidos mostraron que esta alteración se puede evitar mediante la implementación de un período de atemperado de 24 h después del tratamiento con CO2, antes de conservar las frutas a bajas temperaturas. En el Capítulo II se evaluó la eficacia de un tratamiento de desastringencia recientemente patentado basado en la aplicación de una nueva cera que contiene etanol en su formulación. Los resultados mostraron que esta cera podría ser una alternativa al tratamiento convencional de desastringencia con altas concentraciones de CO2 cuando la fruta se envía a mercados lejanos a bajas temperaturas. La aplicación de la nueva cera antes de conservación en frío provocó la pérdida de astringencia de la fruta tras 30 días a 0 °C, preservando la firmeza y la calidad interna. Los resultados del Capítulo III demostraron que la realización de dos aplicaciones de ácido giberélico (AG) en precosecha permite retrasar el proceso de maduración de la fruta en el árbol con respecto a una sola aplicación. La combinación de AG en precosecha y 1-metilciclopropeno (1-MCP) en postcosecha resultó en una mayor calidad del fruto durante la frigoconservación en comparación con la aplicación únicamente de 1-MCP. Además, la fruta tratada con múltiples aplicaciones de AG mostró una firmeza ligeramente mayor tras de conservación a baja temperatura. En el Capítulo IV se demostró que, paralelamente al retraso de la maduración, el tratamiento con AG también retrasa la senescencia del cáliz, incrementando la calidad externa del fruto. La senescencia del cáliz durante la maduración del fruto se relacionó con una disminución de los parámetros de fluorescencia de clorofila (Fo, Fm y Fv/Fm). En el Capítulo V se estudiaron las condiciones óptimas de almacenamiento del caqui producido bajo cultivo ecológico. Según los resultados, no se deben almacenar los frutos cosechados con firmeza inferior a 30 N. La calidad del fruto durante la conservación dependió de las condiciones de almacenamiento, momento de aplicación del tratamiento de deastringencia y estado de madurez inicial. Por último, en los Capítulos VI y VII se abordó la evaluación bajo condiciones mediterráneas de 14 variedades de caquis no-astringentes procedentes de otros países. Se identificaron las variedades tempranas y tardías que pueden ser potencialmente interesantes para ampliar el período de cosecha en nuestra región. Los estudios postcosecha mostraron que todas las variedades estudiadas presentan una buena aptitud para ser comercializadas en los mercados nacionales y europeos. Sin embargo, la sensibilidad a los daños por frío que presentaron algunas de las variedades estudiadas podría limitar su comercialización a países de ultramar. Además, se observó que la respuesta al tratamiento con 1-MCP, para el control de los daños por frío, dependió en gran medida de la variedad. / [CA] En les últimes dues dècades, la producció de caqui a Espanya ha augmentat exponencialment i la superfície de cultiu s'ha multiplicat quasi per huit vegades. En l'actualitat, la producció de caqui està centralitzada en el la cultivar 'Rojo Brillante', que és mundialment apreciat. Malgrat això, aquesta centralització suposa d'una banda una important limitació comercial, ja que la producció es concentra en un període de collita relativament curt i, d'altra banda implica un alt risc fitosanitari. En la present Tesi s'aborden dos objectius principals: 1) Optimització de la tecnologia pre i postcollita amb la finalitat de garantir la qualitat del caqui 'Rojo brillante' durant la frigoconservació i el transport a mercats internacionals. 2) Avaluació del comportament de varietats introduïdes d'altres regions productores sota condicions mediterrànies amb la finalitat d'ampliar el rang varietal de caqui. En el Capítol I es van esclarir les causes de l l'enfosquiment intern. La temperatura a la qual s'exposa la fruita immediatament després de l'aplicació del tractament de desastringència amb altes concentracions de CO¿ es va identificar com el principal factor implicat en aquesta alteració. Els resultats obtinguts van mostrar que aquesta alteració es pot evitar mitjançant la implementació d'un període de temperat de 24 h després del tractament amb CO¿, abans de transferir la fruita a baixes temperatures. En el Capítol II es va avaluar l'eficàcia d'un tractament de desastringència recentment patentat basat en l'aplicació d'una nova cera que conté etanol en la seua formulació en caquis 'Rojo Brillante' i 'Triumph'. Els resultats van mostrar que aquesta cera podria ser una alternativa al tractament convencional de desastringència amb altes concentracions de CO¿ quan la fruita s'envia a mercats llunyans a baixes temperatures. L'aplicació de la nova cera abans de conservació en fred va provocar la pèrdua d'astringència de la fruita després de 30 dies a 0 °C, preservant la fermesa i la qualitat interna. Els resultats del Capítol III van demostrar que l'aplicació de dos tractaments d'àcid giberèlic (AG) en precollita permet retardar el procés de maduració de la fruita en l'arbre respecte a una sola aplicació. La combinació de AG en precollita i 1-metilciclopropeno (1-MCP) en postcollita va incrementar la qualitat del fruit durant la frigoconservació en comparació amb l'aplicació únicament de 1-MCP. A més, la fruita tractada amb múltiples aplicacions de AG va mostrar una fermesa lleugerament major després de conservació a baixa temperatura; no obstant això, una sola aplicació de AG també va ser eficaç per a previndre la manifestació de danys per fred en la fruita. En el Capítol IV es va demostrar que, paral·lelament al retard de la maduració, el tractament amb AG també retarda la senescència del calze, incrementant la qualitat externa del fruit. La senescència del calze durant la maduració del fruit es va relacionar amb una disminució dels paràmetres de fluorescència de clorofil·la (Fo, Fm i Fv/Fm). En el Capítol V es van estudiar les condicions òptimes d'emmagatzematge del caqui produït baix cultiu ecològic. D'acord amb els resultats obtinguts, no s'han d'emmagatzemar els fruits collits amb fermesa inferior a 30 N. La conservació del fruit va dependre de les condicions d'emmagatzematge, moment d'aplicació del tractament de desastringència i estat de maduresa de la fruita. Finalment, en els Capítols VI i VII es va abordar l'avaluació de 14 varietats de caquis no-astringents. Es van identificar les varietats primerenques i tardanes que poden ser potencialment interessants per a ampliar el període de collita. D'altra banda, els estudis postcollita van mostrar que totes les varietats estudiades presenten una bona aptitud per a ser comercialitzades en els mercats nacionals i europeus. No obstant això, la sensibilitat als danys per fred que van presentar algunes de les varietats estudiades podria limitar la seua comercialització a països d'ultramar. A més, es va observar que la resposta al tractament amb 1-MCP, per al control dels danys per fred, va dependre en gran manera de la varietat. / [EN] In the last two decades, persimmon production in Spain has increased exponentially and the cultivation area has expanded almost eight-fold. The current production of persimmon is focused on the cultivar 'Rojo Brillante', which is appreciated worldwide due to the high quality of the fruit. Nevertheless, centralized production of this single cultivar implies a major commercial limitation as its high volume of production is concentrated in a relative short harvesting period. Furthermore, it also implies a high phytosanitary risk. In this context, this Thesis addresses two main objectives: 1) Optimization of pre- and postharvest technology in order to guarantee the quality of 'Rojo Brillante' persimmon after cold storage and transportation to overseas markets. 2) Evaluation of the behavior of foreign varieties of persimmon under Mediterranean conditions in order to extend the varietal range. The first objective of this Thesis was tackled through the studies presented in Chapters I to V. The second objective was covered in Chapters VI and VII. Chapter I dealt with the causes of internal browning disorder manifested after cold shipment of 'Rojo Brillante' persimmons to overseas markets. The temperature to which fruit is exposed immediately after CO2 deastringency treatment was identified as the main factor involved in this alteration. Our results showed that this disorder can be avoided by implementing an attemperation period of 24 h after the CO2 treatment, before transferring fruit to cold storage. In Chapter II, the effectiveness of a recently patented deastringency treatment based on applying a new wax containing ethanol was assayed in 'Rojo Brillante' and 'Triumph' persimmons. The use of this new ethanol-based wax could be a potential alternative to the conventional CO2 deastringency treatment when the fruit are sent to distant overseas markets at low temperatures. Application of the new wax before cold storage led to loss of fruit astringency after 30 days at 0 °C while preserving fruit firmness and internal quality. The results of Chapter III demonstrated that two applications of gibberellic acid (GA) made it possible to delay the fruit maturity process on the tree more than with just a single application. The combination of GA and 1-methylciclopropene (1-MCP) preserved the fruit quality better than with the application of 1-MCP alone during cold storage. Moreover, the fruit treated by multiple GA applications showed a slightly higher firmness after cold storage. However, a single GA application was also effective in preventing the manifestation of chilling injury (CI) in fruit. Chapter IV demonstrated that in parallel to delaying maturity, the GA treatment also delayed calyx senescence, which meant that the fruit preserved a good appearance. The calyx lobe senescence during fruit ripening was linked to a decrease in all Chlorophyll Fluorescence Imaging (CFI) parameters (Fo, Fm, and Fv/Fm). Chapter V addressed the optimization of storage conditions for organic persimmons. According to our results, fruit harvested with a firmness lower than 30 N must not be stored. Fruit harvested with a higher level of firmness could be stored for up to three weeks depending on the storage conditions, the moment of application of the CO2 deastringency treatment, and the stage of fruit maturity. The longest storage period (3 weeks) was achieved when fruit were harvested with a firmness of around 45 N, submitted to CO2 treatment and then stored at 15 °C. Finally, 14 foreign varieties grown under Mediterranean conditions were evaluated to identify early- and late-season varieties with potential to extend the harvesting period. This is presented in Chapters VI and VII. In general, all the evaluated varieties fulfilled the quality criteria to be commercialized in domestic and European markets. Periodic Fluctuation of Tidal Volumes Further Improves Variable Ventilation in Experimental Acute Respiratory Distress Syndrome

In experimental acute respiratory distress syndrome (ARDS), random variation of tidal volumes (VT ) during volume controlled ventilation improves gas exchange and respiratory system mechanics (so-called stochastic resonance hypothesis). It is unknown whether those positive effects may be further enhanced by periodic VT fluctuation at distinct frequencies, also known as deterministic frequency resonance.We hypothesized that the positive effects of variable ventilation on lung functionmay be further amplified by periodic VT fluctuation at specific frequencies. In anesthetized and mechanically ventilated pigs, severe ARDS was induced by saline lung lavage and injurious VT (double-hit model). Animals were then randomly assigned to 6 h of protective ventilation with one of four VT patterns: (1) random variation of VT (WN); (2) P04, main VT frequency of 0.13Hz; (3) P10, main VT frequency of 0.05Hz; (4) VCV, conventional non-variable volume controlled ventilation. In groups with variable VT , the coefficient of variation was identical (30%). We assessed lung mechanics and gas exchange, and determined lung histology and inflammation. Compared to VCV, WN, P04, and P10 resulted in lower respiratory system elastance (63 ± 13 cm H2O/L vs. 50 ± 14 cm H2O/L, 48.4 ± 21 cm H2O/L, and 45.1 ± 5.9 cm H2O/L respectively, P < 0.05 all), but only P10 improved PaO2/FIO2 after 6 h of ventilation (318 ± 96 vs. 445 ± 110mm Hg, P < 0.05). Cycle-by-cycle analysis of lung mechanics suggested intertidal recruitment/de-recruitment in P10. Lung histologic damage and inflammation did not differ among groups. In this experimental model of severe ARDS, periodic VT fluctuation at a frequency of 0.05Hz improved oxygenation during variable ventilation, suggesting that deterministic resonance adds further benefit to variable ventilation.

Detta examensarbete undersöker möjligheten att tillämpa tidiga och ej så beräkningstunga CFD-simuleringar (’Computational Fluid Dynamics’) för optimering av gasväxlingskomponenter tidigt i produktutvecklingsprocessen. Examensarbetet har innefattat framtagande av generellt applicerbara inställningar och riktlinjer vid modellering av gasväxlingskomponenter med programvaran OpenFOAM, optimerade för att nyttja så låg beräkningskraft som möjligt men fortfarande erhålla tillräckligt noggranna och pålitliga resultat. Rapporten ger en teoretiskt genomlysning med motivering av simuleringsrelaterade val i OpenFOAM i syfte att validera och efter behov justera de inställningar och parameterval ansatta i det befintliga Excel skriptet – framtaget av avdelningen med ansvar för avgasefterbehandlingssystem – som automatiserar uppställningen av OpenFOAM:s simuleringsrelaterade filstruktur. Detta i syfte att erhålla och säkerställa pålitliga resultat vid användning av skriptet anpassat för simulering av gasväxlingskomponenter. Rapporten redovisar även bakomliggande analyser för olika inställningar och val i OpenFOAM i syfte att tjäna som välgrundade riktlinjer genom hela CFD-processen; såsom för skapande av geometriska modeller, generering av volymsnät, åtgärder för att säkerställa konvergens samt uppställning och tolkning av resultatet. En av examensarbetets leveranser är en metodbeskrivning utformad med praktiska riktlinjer för genomförandet av hela CFD-processen med avsedda programvaror samt med tillhörande praxis avseende simuleringsrelaterade val genom processen. Ett antal genomförda fallstudier i examensarbetet visar att OpenFOAM har potential att utföra initiala och indikativa simuleringar för att mer tidseffektivt möjliggöra en bredare idégenerering i konceptutvecklingsfasen. / This thesis investigates the possibility of applying early and not so computational heavy simulations using Computational Fluid Dynamics (CFD) for optimization of gas exchange components early in the product development process. The scope of the thesis includes the development of generally applicable settings and guidelines – documented in a methodology – when modelling gas exchange components with the software OpenFOAM, optimized to use as low computational power as possible but still obtaining sufficiently accurate and reliable data. The report consists of a theoretical study and motives for simulation-related choices in OpenFOAM in order to adjust and validate the settings and parameter choices applied in the existing Excel script – developed by the department responsible for exhaust aftertreatment systems – that automates the set-up of OpenFOAM's simulation-related file structure. This in order to obtain and ensure reliable results using the script adapted to the environment for gas exchange components. The report also presents underlying analyses for different settings and choices in order to serve as well-founded guidelines throughout the CFD-process; such as the creation of geometric models, the generation of mesh, measures to ensure convergence, and the set-up and interpretation of the results. One of the deliverables of the thesis is a method description designed with practical guidelines for the entire CFD-process with intended software’s and with associated best practices in regards of simulation-related choices throughout the process. A number of case studies – that has been simulated in this thesis – show that OpenFOAM has the potential to perform initial and indicative simulations enabling a more time-efficiently and broader ideation in the concept development phase.

Stable Isotopes and Metabolite Profiles as Physiological Markers for the Drought Stress Sensitivity in Douglas-Fir Provenances (Pseudotsuga menziesii (MIRB.) FRANCO)

In Mitteleuropa werden zukünftig häufigere Trocken- und Hitzeperioden mit wirtschaftlichen Einbußen in der Waldwirtschaft erwartet. Die Douglasie (Pseudotsuga menziesii (Mirb.) Franco) wird als Alternative für die wirtschaftlich bedeutsame, jedoch trockenheitsempfindliche Fichte diskutiert (Picea abies (L.) H.Karst.). Zwei Unterarten, die Küsten- (FDC) und die Inlandsdouglasie (FDI), sind im ausgedehnten natürlichen Verbreitungsgebiet in Nordamerika beheimatet, welches ein großes Potenzial für die Auswahl produktiver und trockenresistenter Herkünfte bietet. Unser Ziel war, die Trockenreaktion verschiedener Douglasienherkünfte unter Verknüpfung morphologischer und physiologischer Parameter und die der Trockenheitsresistenz bzw. -empfindlichkeit zugrundeliegenden Mechanismen zu erforschen. Ein Herkunftsversuch in Südwestdeutschland ermöglichte die Untersuchung 50-jähriger Douglasien verschiedener Herkünfte entlang eines Höhengradienten. Unter kontrollierten Bedingungen simulierten wir die Effekte einer Hitzewelle auf Jungbäume zweier Provenienzen. Wir analysierten die Kohlenstoff- und Sauerstoff-Stabilisotopenzusammensetzung, den Gaswechsel der Blätter, Veränderungen im Stoffwechsel und das Baumwachstum. Unsere Ergebnisse zeigen bei FDC aus humiden Regionen hohe Wachstumseinbußen unter Trockenheit und moderat bis stark verringerte stomatäre Leitfähigkeit, unterstützt durch Photoprotektion. FDC aus Regionen mit starker Sommertrockenheit reagierten kaum mit Stomataschluss und Wachstumseinbußen auf Trockenheit, jedoch mit starker Osmoregulation und Monoterpen-Emissionen, welche zur Trockenresistenz beitragen könnten. FDI aus einer ariden Region zeigten hohe An, geringes Wachstum und stark antioxidative und photoprotektive Mechanismen. Die Herkünfte unterscheiden sich stark in ihrer Trockenreaktion und ihren Schutzmechanismen. Der Anbau trockenresistenter Herkünfte wird an Standorten von Vorteil sein, für die eine Häufung von ariden Sommerperioden vorhergesagt wird. / In Central Europe, more frequent periods of dry and hot weather are expected in the future with economic losses in the forestry sector. Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) is discussed as a timber species alternative to the economically important but drought-sensitive spruce (Picea abies (L.) H. Karst.). Two subspecies, the coastal (FDC) and the interior Douglas-fir (FDI), are native to an extensive natural range in North America, offering a great potential for the selection of productive and drought tolerant provenances. Our goal was to investigate the drought response of different Douglas-fir provenances on the morphological and physiological level, as well as the mechanisms underlying drought resistance or susceptibility. A provenance trial in southwestern Germany established in 1958 allowed the study of 50-year-old Douglas-fir trees of diverse provenances along a height gradient. Under controlled conditions, we simulated the effects of a heat wave on young trees of two provenances. We analyzed carbon and oxygen stable isotopic composition, leaf gas exchange, changes in metabolism and tree growth. FDC from humid regions responded to drought with strong growth decline and a medium to strong stomatal closure, supported by enhanced instantaneous photoprotection. FDC from regions with very dry summer conditions showed a small growth decline and anisohydric regulation of stomatal conductance under drought, supported by high levels of osmotic adjustment. High monoterpene emissions might contribute to the drought resistance. FDI from an arid region showed high assimilation rates, low growth potential and a high antioxidant, photoprotective, drought and heat protective potential. The provenances differ greatly in their dry reaction and their protective mechanisms. The cultivation of drought resistant crops will be beneficial at sites predicted to accumulate arid summer periods.

Spark ignited engines are still important for conventional as well as for hybrid power trains and are thus objective to optimization. Today a lot of functionalities arise from software solutions, which have to be calibrated. Modern engine technologies provide an extensive variability considering their valve train, fuel injection and load control. Thus, calibration efforts are really high and shall be reduced by introduction of virtual methods. In this work a physical 0D combustion model is set up, which can cope with a new generation of spark ignition engines. Therefore, at first cylinder thermodynamics are modeled and validated in the whole engine map with the help of a real-time capable approach. Afterwards an up to date turbulence model is introduced, which is based on a quasi-dimensional k-epsilon-approach and can cope with turbulence production from large scale shearing. A simplified model for ignition delay is implemented which emphasizes the transfer from laminar to turbulent flame propagation after ignition. The modeling is completed with the calculation of overall heat release rates in a 0D entrainment approach with the help of turbulent flame velocities. After validation of all sub-models, the 0D combustion prediction is used in combination with a 1D gas exchange analysis to virtually calibrate the modern engine torque structure and the ECU function for exhaust gas temperature with extensive simulations. / Moderne Ottomotoren spielen heute sowohl in konventionellen als auch hybriden Fahrzeugantrieben eine große Rolle. Aktuelle Konzepte sind hochvariabel bezüglich Ventilsteuerung, Kraftstoffeinspritzung und Laststeuerung und ihre Optimierungspotentiale erwachsen zumeist aus neuen Softwarefunktionen. Deren Applikation ist zeit- und kostenintensiv und soll durch virtuelle Methoden unterstützt werden. In der vorliegenden Arbeit wird ein physikalisches 0D Verbrennungsmodell für Ottomotoren aufgebaut und bis zur praktischen Anwendung geführt. Dafür wurde zuerst die Thermodynamik echtzeitfähig modelliert und im gesamten Motorenkennfeld abgeglichen.
