Identificación de genotipos y linajes de los cuatro serotipos del virus dengue en el Perú durante los años 1998 - 2012

El dengue es una enfermedad febril aguda viral causada por la infección con uno de los cuatro serotipos del virus del dengue (DENV-1, 2, 3 y 4) que se trasmite al hombre a través del mosquito del género Aedes aegypti. En el presente estudio se identificaron los genotipos y linajes de los cuatro serotipos del virus dengue a partir de los aislamientos obtenidos en muestras procedentes de diferentes regiones geográficas de Perú durante los años 1998 - 2012. El diseño del estudio fue descriptivo - retrospectivo de las características genéticas, 109 cepas que pertenecen a uno de los cuatro serotipos del virus dengue fueron procesadas por la RT-Nested PCR para amplificar la región del gen E/NS1 y se secuenció el genoma completo de una cepa de DENV-2. Las secuencias fueron alineadas con el programa CLUSTAL X y analizadas con el programa MEGA 5.0 con el algoritmo de distancia Neighbor Joining para E/NS1 y el método de Maximum Likelihood para el genoma completo. Se identificó el serotipo DENV-1, genotipo V, con tres linajes; respecto al serotipo DENV-2, se estableció dos genotipos: el genotipo América con dos linajes y genotipo América/Asia con cinco linajes. El serotipo DENV-3, presentó el genotipo III con cinco linajes y finalmente se halló el serotipo DENV-4 con dos linajes. Se concluye que los cuatro serotipos de los virus dengue aislados en diferentes áreas geográficas de Perú durante los años 1998 - 2012 presentaron variabilidad genética a nivel de genotipos y linajes, siendo el DENV-3 más divergente con cinco linajes y el DENV-4 menos divergente con dos linajes, respecto a los otros serotipos estudiados. Was identified serotype DENV-1, genotype V, with three lineages; respect to serotype DENV-2, were described two genotypes: genotype America with two lineages and genotype Americas/Asia with five lineages. DENV-3 genotype III presented the five lineages and ultimately was found DENV-4 with two lineages. We conclude that the four serotypes of dengue viruses isolated in different geographical areas of Peru during 1998 - 2012 showed genetic variation at the level of genotypes and lineages, DENV-3 remains the most divergent with five lineages and least divergent DENV-4 with two lineages, compared to the other serotypes examined. O vírus dengue (DENV) é mantido na natureza através de replicação cíclica em hospedeiros vertebrados e mosquitos Aedes, sendo o Aedes aegypti o principal vetor. O seqüenciamento completo de DENV-3 isolado de Ae. aegypti naturalmente infectado do Rio de Janeiro em 2001 e de um caso humano em 2002, demonstrou uma similaridade de 99% com DENV-3 isolado de um caso fatal humano ocorrido no mesmo período. A análise da região 3´NC do genoma viral demonstrou uma mutação nesta região, sugerindo uma deleção de 8 nucleotídeos (nts) na inserção de 11nts, característica de DENV-3 isolados no Brasil. Neste estudo, avaliamos se as diferentes variantes de DENV-3 na interação vírus-vetor através da determinação da competência vetorial em Ae. aegypti. As cepas de DENV-3 BR74886 #5 (cepa representativa do vírus com inserção de 11nts na região 3\2019NC) e BR73356 #5 (cepa representativa do vírus com a deleção de 8 nts), apresentando títulos de 8 x 107 PFU/mL e 7,3 x 107 PFU/mL, respectivamente, mantiveram suas características na região 3\2019NC do genoma viral após cinco passagens em cultura celular e foram selecionadas para a infecção experimental. A estratégia de infecção consistiu na utilização de 2.925 fêmeas de Ae. aegypti, sendo que 2.340 da geração F1 da população de Tubiacanga (RJ) e 585 da cepa controle Paea A população experimental se mostrou competente para transmitir as duas cepas virais de DENV-3, no entanto a disseminação viral no corpo do mosquito apresentou-se de forma heterogênea, sugerindo haver vantagens para a cepa com inserção de 11 nts, uma vez que disseminou-se mais rapidamente. Quando as fêmeas de Ae. aegypti foram alimentadas com ambas as cepas, a disseminação no vetor comportou-se de maneira semelhante à observada quando alimentadas com a cepa representativa da inserção de 11 nts. A análise das cepas de DENV-3 detectadas nas cabeças das fêmeas após replicação in-vivo por 14 dias, não identificou alterações nas características de cada cepa. No entanto, a análise desta região demonstrou uma prevalência do vírus com a inserção de 11 nts quando as fêmeas foram alimentadas com as duas cepas simultaneamente. Variações entre os títulos virais foram observados nas salivas de fêmeas infectadas com as diferentes cepas virais, sugerindo que embora ambas as cepas de DENV-3 possam ser transmitidas na natureza, a cepa com a inserção de 11 nts possui maior eficácia. Os resultados indicam que diferentes cepas virais, variantes genéticas ou mutações que ocorram em um mesmo genótipo podem impactar na competência vetorial dos mosquitos, podendo afetar diretamente o potencial epidêmico de uma cepa de vírus em particular / Dengue is considered the most important arthropod - borne viral disease that affects humans. Dengue virus (DENV) is maintained in nature by a cyclic replication in vertebrate hosts and Aedes mosquitoes, with the Aedes aegypti as the main vector. The complete sequencing of a DENV - 3 strain isolated from Ae. aegypti naturally infected in Rio de Janeiro in 2001 and from a h uman case occurred in 2002 demonstrated a similarity of 99% with a DENV - 3 isolated from a human fatal case occurred in the same period. However, the analysis of the 3 Untranslated Region (UTR) of the viral genome showed a mutation in this region, suggestin g a deletion of 8 nucleotides (nts) within the 11 nucleotides insertion, characteristic of DENV - 3 isolated in Brazil. In this study, we evaluated whether the distinct DENV - 3 variants presenting those characteristics showed differences on the virus - vector i nteraction by determining the vector competence of two populations of Ae. aegypti . The DENV - 3 strain BR74886#5 (with the 11nts insert in the region 3' UTR ) and the strain BR73356#5 (with an 8 nts deletion), presented titers of 8 x 10 7 PFU/mL and 7.3 x 10 7 PF U/mL, respectively, maintained its characteristics in the 3' UTR region of the viral genome after five passages in cell culture and were selected for experimental infection. The infection strategy consisted in the use of 2,925 female Ae. a egypti : 2,340 of a F1 generation from the Tubiacanga (RJ) population and 585 Paea control mosquitoes. The experimental population proved to be competent to transmit the two DENV - 3 strains. However, the viral dissemination in the body of the mosquito presented heterogeneousl y, suggesting that there are advantages for the strain with 11 nts insertion in the 3' UTR , once disseminated more rapidly . When Ae. aegypti were fed with the both strains, the viral dissemination in the vector was similar to that observed when fed with 11 nts insertion in the 3' UTR . The analysis of the 3' UTR from the DENV - 3 strains detected in the heads of females after in - vivo replication for 14 days, did not identify changes in the 3’ UTR of each strain. However, the analysis of the females infected with two strains simultaneously detected only the presence of the strain carrying the 11 nts insertion in the 3' UTR . Viral titer differences were observed in the saliva of the experimentally infected Ae. a egypti females suggesting that even tough both variants are transmissible, the variant presenting the 11nts is more efficiently transmitted. The results indicate that different viral strains, genetic variants or mutations that occur in the same genotype may impact on the vector competence of mosquitoes, which c an directly affect the epidemic potential of a particular virus strain

Dez genótipos (A a J) foram identificados e alguns deles foram ainda classificados em subgenótipos. O genótipo D (HBV/D) tem uma distribuição mundial e possui nove subgenótipos (D1 a D9) descritos. A história evolutiva do HBV/D nas Américas não é bem compreendida e poucas sequências de genoma completo HBV/D estão disponíveis. O objetivo deste estudo é analisar a proporção e a distribuição geográfica dos subgenótipos de D nas Américas, determinar as sequências genômicas completas do HBV/D isolados de diferentes regiões geográficas do Brasil e investigar a origem e a propagação dos subgenótipos de D nas Américas. Para identificar os subgenótipos circulantes nas Américas, foram obtidos do GenBank genomas completos e sequências dos genes pré-S/S e S (n = 609). Foram detectados no continente os subgenótipos HBV/D1-D4 e HBV/D7. O HBV/D1 foi encontrado na Argentina (83%), Brasil (2%), Cuba (3%) e no Canadá (18%), enquanto que HBV/D2 foi detectado na Argentina (4%), Brasil (17%), Canadá (18%), Chile (75%), Cuba (5%) e EUA (90%). O subgenótipo HBV/D3 foi o mais frequente no Brasil (56%) e foi encontrado em todos os países americanos, exceto na Venezuela e Groelândia. O HBV/D4 foi o subgenótipo mais frequente no Canadá (35%), Cuba (76%), Haiti (84%), Martinica (80%) e Venezuela (100%). O subgenótipo HBV/D7 foi observado apenas em Cuba (8%) Ademais, amostras de soro HBsAg positivas, coletadas de todas as cinco regiões geográficas brasileiras e caracterizadas como HBV/D foram selecionadas para o sequenciamento do genoma completo. Quarenta e cinco sequências de genoma completo foram determinadas (D1, n = 1; D2, n = 11; D3, n = 32; D4, n = 1). Para investigar a origem e a propagação do HBV/D nas Américas, foram criados diferentes arquivos contendo genomas completos dos subgenótipos D1 a D4, incluindo as sequências brasileiras sequenciadas neste trabalho, bem como sequências do GenBank de diferentes origens geográficas e data de coleta conhecida. As análises foram realizadas usando o pacote BEAST v.1.8.2. A análise filogeográfica sugeriu que o HBV/D1 foi introduzido no Brasil por isolados da Síria e, na Argentina por isolados da Turquia. As sequências HBV/D2 brasileiras e argentinas ficaram relacionadas a sequências da Europa Oriental e Rússia, de onde parece ter se originado os isolados circulantes nestes países. O HBV/D2 dos EUA parece ter se originado da Índia. Já o HBV/D3 não apresentou uma origem clara nas Américas, mas provavelmente foi introduzido pelos europeus. A análise do HBV/D4 sugeriu que este subgenótipo foi provavelmente introduzido pelos escravos africanos trazidos para trabalhar nas Américas. Nossos resultados sugerem que os subgenótipos de D tiveram diferentes introduções no continente americano e demonstram a utilidade de ferramentas computacionais desenvolvidas recentemente para investigar a história evolutiva do HBV / Hepatitis B virus (HBV) infection is a major global health problem. Ten genotypes (A to J) have been identified and some of them have been further divided into subgenotypes. Genotype D (HBV/D) has a worldwide distribution and nine subgenotypes (D1 to D9) have so far been described. The evolutionary history of HBV/D in the Americas is not well understood and few HBV/D complete genome sequences are available. The aim of this study is to examine the proportion and geographical distribution of D subgenotypes in the Americas, determine the full-length genomic sequences of HBV/D isolates from different Brazilian regions and investigate the origin and spread of D subgenotypes in the Americas. To identify the circulating subgenotypes, we downloaded American HBV/D complete and partial (pré-S/S or S gene) available in GenBank (n=609). It was detected in the Americas the subgenotypes HBV/D1-D4 and HBV/D7. HBV/D1 was found in Argentina (83%), Brazil (2%), Cuba (3%) and Canada (18%), while HBV/D2 was detected in Argentina (4%), Brazil (17%), Canada (18%), Chile (75%), Cuba (5%) and USA (90%).The subgenotype HBV/D3 was the most prevalent subgenotype in Brazil (56%) and was found in all American countries except Venezuela and Greenland. HBV/D4 was the most prevalent subgenotype in Canada (35%), Cuba (76%), Haiti (84%), Martinique (80%) and Venezuela (100%). HBV/D7 was observed only in Cuba (8%). In addition, HBsAg positive serum samples, collected from all five regions of Brazil and characterized as having HBV/D strains were selected for full genome sequencing. 45 full-length sequences were determined (D1, n=1; D2, n=11; D3, n=32; D4, n=1). To investigate the origin and spread of HBV/D in the Americas, we compiled different data sets of complete genomes for subgenotypes D1 to D4, using the Brazilian sequences as well as GenBank sequences from different geographic origins and known collection date The analyses were carried out by using BEAST v.1.8.2 software package. The phylogeoghaphic analysis suggested that HBV/D1 was introduced in Brazil by Syrian strains and in Argentina by Turkish strains. The Brazilian and Argentinian D2 sequences were closely related to Eastern Europe and Russian isolates, where are the most probable locations to be the source of this subgenotype in these countries. The HBV/D2 from from USA seems to have originated from India. HBV/D3 had no clear introduction in the Americas, but probably was brought by the Europeans. The analysis of HBV/D4 suggested that this subgenotype was probable introduced by African slaves brought to work in the Americas. Our results suggest that D subgenotypes had different introductions in the American continent and demonstrate the usefulness of recently developed computational tools for investigating the evolutionary history of HBV

Determinación de la variabilidad genética de aislados chilenos del virus diarrea viral bovina (VDVB) por filogenia molecular de la región 5 no codificante del genoma viral

Memoria para optar al Título Profesional de Médico Veterinario / El virus diarrea viral bovina (VDVB) es el agente causal del complejo Diarrea Viral Bovina/ Enfermedad de las Mucosas y otros cuadros clínicos, que generan importantes pérdidas económicas en la industria del bovino. El VDVB presenta una gran variabilidad genómica, que ha llevado a clasificarlo en dos genotipos (1 y 2). En el genotipo VDVB-1 se identifican 15 subgrupos virales (1a-1o) y en el genotipo VDVB-2 sólo dos (2a y 2b). En Chile, el virus presenta una amplia diseminación, con prevalencias serológicas de un 69,2% en la Región de la Araucanía, De los Ríos y De los Lagos y de un 59,7% y 86% en bovinos de leche y de carne de la Región Metropolitana respectivamente. El objetivo de esta memoria fue determinar la composición genética de los virus del VDVB que infectan a los bovinos en Chile. Para ello, a cuarenta y cinco virus obtenidos desde distintas regiones de Chile se procedió a establecer su parentesco genético por filogenia molecular de la región 5´ no codificante (5´NCR) del genoma viral. Los resultados indican que cuarenta virus pertenecen al genotipo VDVB-1 y cinco al genotipo VDVB-2. En el genotipo VDVB-1, 2 virus pertenecen al subgrupo VDVB-1a, quince al VDVB-1b, veinte al VDVB-1j y tres al VDVB-1i, siendo este último subgrupo detectado por primera vez en Chile. Todos los virus del genotipo VDVB-2 pertenecen al subgrupo VDVB-2a. Los resultados permiten concluir que los virus del VDVB que circulan actualmente en el ganado bovino de Chile presentan una alta variabilidad genómica y su composición genética es muy similar a la de los virus presentes en Argentina. / Proyecto FONDECYT 1060581

Detección molecular del gen M del virus distemper canino / molecular detection of canine distemper virus M gene

Memoria para optar al Título Profesional de Médico Veterinario. / La importancia que reviste el Distemper Canino -una de las principales enfermedades infecciosas que afectan tanto a los caninos domésticos y silvestres como también a varios otros mamíferos terrestres y marinos- junto a la dificultad de su diagnóstico -basado en los signos clínicos- ha motivado que a la fecha se hayan realizado diversos estudios en la Facultad de Ciencias Veterinarias y Pecuarias de la Universidad de Chile para la detección óptima del agente etiológico: el Virus Distemper Canino. La técnica molecular utilizada es la Reacción en Cadena de la Polimerasa asociada a transcripción reversa, utilizando como blanco de detección los diferentes genes que componen su genoma: N, P, F, M, H y L, con el fin de encontrar el método que permita el diagnóstico de la enfermedad ante-mortem de manera precoz. Así, en esta Memoria de Título, se utiliza el gen M que codifica la proteína de la Matriz viral y se implementó un protocolo de detección molecular que completa la información respecto a cuál sería el gen óptimo a utilizar en el diagnóstico de la enfermedad. Para ello se utilizaron 20 muestras de RNA total, obtenido desde sangre periférica de perros de distintas razas, edades y estado de vacunación contra Virus Distemper Canino, que presentaron signos clínicos y que ya eran positivos a la RT-PCR que detecta el gen N del virus. La técnica detectó el genoma viral en el 70% de aquellas analizadas, observándose una banda cercana a los 500 pb, cuyos productos amplificados presentaron intensidad y nitidez evidentes a la visualización. El porcentaje de identidad nucleotídica (PIN) respecto de las dos secuencias consenso obtenidas del total de muestras (mediante el programa online Clustal Omega), arrojó un 99% para ambas secuencias, comparadas con las secuencias de Virus Distemper Canino que entrega el programa informático de alineamiento de secuencias online de libre acceso BLAST. Los resultados permiten concluir que, si bien la elección del gen M como blanco de detección permite detectar VDC, no resulta de elección para la detección certera del virus, y por ende, para ser utilizada en el diagnóstico de la enfermedad causada por este / The importance of the Canine Distemper -one of the main infectious diseases affecting domestic and wild canines as well as several other mammals both terrestrial and marine- and the difficulty of its diagnosis -based on clinical signs- has motivated to the date several studies that have been carried out in the Faculty of Veterinary and Animal Sciences of the University of Chile to detect the etiological agent: the Canine Distemper Virus. The molecular technique used is the Polymerase Chain Reaction associated with reverse transcription and the different genes that make up its genome have been used as a detection target: N, P, F, M, H and L, in order to find an method that allows the early diagnosis of ante-mortem disease. Thus, in this Title Memory, the M gene that encodes the protein of the virus Matrix is used and a molecular detection protocol was implemented that completes the information regarding which would be the optimal gene to be used in the diagnosis of the disease. For this, 20 samples of total RNA were used, obtained from peripheral blood of dogs of different breeds, ages and vaccination status against Canine Distemper Virus, which showed clinical signs and which were already positive to the RT-PCR that detects the N gene of virus. The technique detected the viral genome in 70% of those analyzed, generating a band close to 500 bp, whose amplified products presented clear intensity and sharpness to the visualization. The percentage of nucleotide identity (PIN) with respect to the two consensus sequences obtained from the total samples (using the Clustal Omega online program) yielded 99% for both sequences compared to the Canine Distemper Virus sequences delivered by the alignment software of free access online sequences BLAST. The results allow us to conclude that although the choice of the M gene as a detection target allows to detect VDC, it is not of choice for the accurate detection of the virus, and therefore, to be used in the diagnosis of the disease caused by it

Análise do viroma de soro de matrizes suínas com partos normais e com natimortalidade / Virome analysis on sera of sows with and without CASES of natimortality

Falhas reprodutivas são importante causa de prejuízos econômicos na suinocultura. Elas implicam na diminuição do número de leitões nascidos vivos e aumentam o descarte de animais e as taxas de reposição de matrizes, levando à redução da produtividade do rebanho. Embora a maioria dos casos de natimortalidade sejam associados a fatores não infecciosos, os agentes infecciosos possuem um papel importante e ainda pouco conhecido na etiologia deste quadro. Até o presente, nenhum trabalho foi realizado visando o estudo do conjunto de vírus que possam estar presentes em matrizes com eventos de natimortalidade por ocasião do parto. Em função disso, o presente trabalho teve por objetivo examinar o viroma do soro de matrizes suínas com e sem casos de natimortalidade. Foram coletadas 94 amostras de soro de matrizes de seis granjas distribuídas em cinco municípios do Rio Grande do Sul. Em cada granja foram formados dois pools de soros: um composto por matrizes que pariram (um ou mais) natimortos e outro por matrizes que pariram leitegadas sem natimortos. Os pools foram submetidos à extração de ácido nucleico viral, enriquecimento e sequenciamento de alto desempenho, buscando a identificação de agentes que possam representar um fator de risco à natimortalidade em suínos Não foi possível identificar diferenças significativas nos viromas de matrizes correlacionadas à ocorrência de natimortalidade. Não obstante, foi possível identificar uma ampla variedade de genomas virais, a maioria deles correspondendo a vírus das famílias Anelloviridae. Este estudo permitiu ainda identificar 20 genomas completos de três espécies de vírus: torque teno vírus suíno 1a e 1b, circovírus suíno tipo 3 (PCV3) e vírus circulares DNA fita simples codificantes de replicase (CRESS), seis dos quais até o presente ainda não reportados em suínos. Em duas granjas, em matrizes que apresentaram natimortalidade, foram identificados genomas de PCV3, cuja participação como potencial causador de problemas reprodutivos precisa ser futuramente investigada. Não foram identificados vírus com genoma de RNA. It leads to a decrease in the number of piglets reared per sow and may imply in the need for replacement of sows, reducing the productivity in a herd. Although the majority of cases of stillbirths have been attributed to non-infectious causes, several infectious agents have been implicated in the etiology of such condition. Nevertheless, other as yet unknown agents may be involved in the pathogenesis of stillbirths. The aim of this work was to investigate the virome in sera of sows without and with one or more cases of stillbirth in the litter. Sera were collected from 94 sows of six commercial farms in five municipalities in the state of Rio Grande do Sul, Brazil. Two pools of sera were collected from each farm: one representative of sows that had at least on stillbirth in the last litter and another composed by sera of sows that had litters with no stillbirths. The pools were subjected to nucleic acid extraction, enrichment and high throughput sequencing. No significant differences were detected in the serum viromes of sows with or without stillbirth Nevertheless, it was possible to identify a wide variety of viral genomes, most of these representing viruses of Anelloviridae family. In addition, the present work allowed the identification of 20 complete genome sequences including torque teno sus virus 1a and 1b, porcine circovirus 3 (PCV3) and circular rep-encoding ssDNA viruses (CRESS), including six species not previously reported in swine. In two farms, PCV3 genomes were identified in the serum pools of sows which had cases of stillbirth. The role for this virus as a potential cause of reproductive failure needs additional investigations. No genomes of viruses with RNA genomes were identified. This study provides a contribution to the knowledge on the serum virome of pregnant sows. In addition, it is expected to aid in the identification of possible causes of stillbirth in swine.

Embora a maioria dos casos de natimortalidade sejam associados a fatores não infecciosos, os agentes infecciosos possuem um papel importante e ainda pouco conhecido na etiologia deste quadro. Até o presente, nenhum trabalho foi realizado visando o estudo do conjunto de vírus que possam estar presentes em matrizes com eventos de natimortalidade por ocasião do parto. Em função disso, o presente trabalho teve por objetivo examinar o viroma do soro de matrizes suínas com e sem casos de natimortalidade. Foram coletadas 94 amostras de soro de matrizes de seis granjas distribuídas em cinco municípios do Rio Grande do Sul. Em cada granja foram formados dois pools de soros: um composto por matrizes que pariram (um ou mais) natimortos e outro por matrizes que pariram leitegadas sem natimortos. Os pools foram submetidos à extração de ácido nucleico viral, enriquecimento e sequenciamento de alto desempenho, buscando a identificação de agentes que possam representar um fator de risco à natimortalidade em suínos Não foi possível identificar diferenças significativas nos viromas de matrizes correlacionadas à ocorrência de natimortalidade. Não obstante, foi possível identificar uma ampla variedade de genomas virais, a maioria deles correspondendo a vírus das famílias Anelloviridae. Este estudo permitiu ainda identificar 20 genomas completos de três espécies de vírus: torque teno vírus suíno 1a e 1b, circovírus suíno tipo 3 (PCV3) e vírus circulares DNA fita simples codificantes de replicase (CRESS), seis dos quais até o presente ainda não reportados em suínos. Em duas granjas, em matrizes que apresentaram natimortalidade, foram identificados genomas de PCV3, cuja participação como potencial causador de problemas reprodutivos precisa ser futuramente investigada. Não foram identificados vírus com genoma de RNA. In women these infections can progress to cervical cancer, which is estimated incidence for the Northern region in 2010 was the largest in Brazil. The nature of the infection depends on the degree of integration of viral DNA with host DNA linked primarily to genes of oncoproteins E6 and E7 of HPV. The determination of the viral types can be held from differences in the viral capsid L1 gene and the variants of a particular type of HPV can be identified through the study of viral non-coding region. Currently the development of prophylactic vaccines against HPV particles using "pseudo-viral" formed by the L1 protein of different subtypes of high risk, while a growing number of studies that use the oncoproteins E6 and E7 in the development of therapeutic vaccines. However, it is necessary for the development of such antiviral vaccines also consider the great diversity of variants of HPV types exist, since differences between the genomic regions of these variants may influence the degree of their infections. This paper describes the complete genome sequence of a variant of HPV 16, detected in Amazonian region, using techniques of genetic engineering and the analysis of this genome by bioinformatics tools. It was observed by analysis of genetic distance that the genome of this variant has a genetic proximity of those identified in the literature as "African variants, and phylogenetic analysis, performed from the non-coding region, support this hypothesis. In addition, several mutations were detected in the genome and obtained, resulting in changes in the positions and number of restriction sites in its sequence. The major differences between the genetic regions of the genome sequenced and the corresponding variants in Africa have been observed over E7. It is expected, with that work, look for future research projects involving protein expression and genomic analysis of HPV in the Amazon region to the regional peculiarities in variants and provide a concise and complete reference on the genome of HPV 16 in the region. / O Papillomavirus Humano é responsável por lesões na mucosa oral, anal e do trato urogenital masculino e feminino, transmitidas por contato direto ou indireto com a pele infectada ou através de relações sexuais. Na mulher essas infecções podem evoluir para um câncer de colo do útero, cuja estimativa de incidência para a região Norte no ano de 2010 foi a maior do Brasil. A natureza das infecções depende do grau de integração do DNA viral com o DNA do hospedeiro associada, principalmente, aos genes das oncoproteínas E6 e E7 do HPV. A determinação dos tipos virais pode ser realizada a partir de diferenças no gene L1 do capsídeo viral e as variantes de um determinado tipo de HPV podem ser identificadas por meio do estudo da Região Não Codificadora viral. Atualmente o desenvolvimento de vacinas profiláticas contra o HPV utiliza partículas pseudo-virais formadas pela proteína L1 de tipos virais de alto risco, enquanto cresce o número de estudos que utilizam as oncoproteínas E6 e E7 no desenvolvimento de vacinas terapêuticas. Contudo, é necessário que o desenvolvimento de tais vacinas antivirais também considere a grande diversidade das variantes dos tipos de HPV existentes, uma vez que diferenças entre as regiões genômicas dessas variantes podem influenciar o grau de suas infecções. Este trabalho descreve o sequenciamento completo do genoma de uma variante do HPV 16, detectado no Estado do Amazonas, utilizando técnicas de Engenharia Genética, bem como a análise desse genoma por ferramentas de Bioinformática. Observou-se, pela análise de distâncias genéticas, que o genoma dessa variante apresenta grande proximidade genética dos exemplares identificados na literatura como variantes africanas , e as análises filogenéticas, realizadas a partir da Região Não Codificadora, reforçam essa hipótese. Além disso, também foram detectadas várias mutações ao longo do genoma obtido, resultando em alterações nas posições e na quantidade de sítios de restrição de sua sequência. As maiores diferenças entre as regiões gênicas do genoma sequenciado e as correspondentes nas variantes africanas foram observadas ao longo de E7. Espera-se, com esse trabalho, atentar os futuros projetos de pesquisa que envolvam expressão de proteínas e análises genômicas de HPV na região amazônica para as peculiaridades existentes nas variantes regionais e fornecer uma referência concisa e completa sobre o genoma do HPV 16 na região.
