<p>Coeliac disease is a chronic inflammatory disease treated with a gluten-free diet, excluding barley, rye and wheat. Hence, there is a demand for methods able to detect gluten in foods in order to ensure correct labeling of products. According to the Codex Alimentarius Commission, 20ppm gluten is the maximum amount allowed in food labeled gluten-free.</p><p>PCR can detect DNA from cereals in food. Four real-time PCR-systems,</p><p>using TaqMan®-probes for detection of barley, oat, rye and wheat were optimized and evaluated. Evaluations were carried out using seeds. Primers were targeted to genes coding for prolamines, seed storage proteins. PCR-systems targeted to barley, oat and wheat were shown to be specific for the cereals corresponding to each system. The system targeted to rye showed cross-reactions with durum wheat and spelt wheat. Detection limits were 50pg, corresponding to <10 haploid genome copies for each cereal. All systems were able to detect 250ppm amounts of DNA, most likely even smaller amounts are detectable. All systems showed an amplification efficiency of ≥95%.</p><p>Systems for detection of barley, oat and wheat are ready for further evaluation, using food products as samples. The rye system however, needs to be re-designed before further evaluation can take place.</p>

Ce travail de thèse vise à apporter des connaissances structurales et fonctionnelles sur les protéines du gluten. Pour cela, nous utilisons les concepts et méthodes de la physique des polymères et de la matière molle. Plus précisément, nous optimisons un protocole d’extraction basé sur la séparation de phases liquide-liquide. Ce dernier permet d’obtenir des isolats de protéines à différents rapports massiques gluténines/gliadines que nous étudions ensuite dans un solvant eau/éthanol 50/50 (v/v). Les résultats, montrent que les protéines se comportent comme des chaînes de polymères en solvant θ, en régime dilué et semi-dilué avec des tailles caractéristiques définis par diffusion de rayons X et de neutrons aux petits angles. De plus, 2 tailles d’objets sont distinguées en régime dilué par diffusion dynamique de la lumière: d’une part des protéines monomériques de l’ordre d’une dizaine de nanomètres associées aux et -gliadines et à des polymères de gluténines de faibles masses molaires et d’autre part des assemblages polymériques de l’ordre de 100 nm, principalement composés de ω-gliadines et polymères de gluténines de haute masse molaire. Ces assemblages sont mis en avant par une combinaison de mesures réalisées par chromatographie d’exclusion de taille et par fractionnement par flux de forces asymétrique et permettent de rationaliser les diagrammes de phases de ces mélanges protéiques, en fonction de la température. L’étude de la dynamique de séparation de phases de ces mélanges protéiques, par diffusion de rayons X aux petits angles, montre que celle-ci est pilotée par un phénomène de décomposition spinodale. Cette décomposition peut être arrêtée lors de trempes en température profondes mais également observée à toutes les températures de trempe, pour les échantillons les plus riches en gluténines, formant un gel dès le régime monophasique, d’après leur étude par rhéologie / The aim of this thesis is to provide structural and functional knowledge on wheat gluten proteins. For that, we use the physical methods and the concept of soft matter. We optimize an extraction protocol based on a liquid-liquid phase separation. With this protocol, we obtain protein batches with different glutenin/gliadin mass ratios, which we then study in a 50/50 water/ethanol solvent (v/v). We show that proteins behave like polymer chains in θ solvent in dilute and semi-dilute regime, whose characteristic size are extracted by small angle X-ray and neutron scattering. Moreover, two sizes of objects are evidenced in dilute regime by dynamic light scattering: monomeric proteins with a size around 10 nm which can be associated to α/β, and γ-gliadins and polymeric glutenins with low molecular weight and polymeric assemblies with a size around 100 nm composed of ω-gliadins and glutenins polymers with high molecular weight. These assemblies are revealed by a combination of size exclusion chromatography and asymmetric flow field flow fractionation and allow one to rationalize the phase diagrams of the protein mixtures with temperature. The study of the dynamics of the phase separation of these protein mixtures by small angle X-ray scattering shows that the phase separation proceeds through a spinodal decomposition phenomenon. An arrested phase separation is observed for deep quenches but also at all temperature quenches for the most glutenin rich samples, which are gels in the monophasic regime, as confirmed by rheology.

This diploma focuses on the selection of gluten-free foods from the perspective of a diagnosed celiac. In the theoretical part is described the history of celiac disease, anatomy and physiology of small intestine, pathogenesis, manifestation and forms of celiac disease, diagnosis, screening and complications from celiac disease. It also describes a treatment of celiac disease which shows inappropriate and appropriate aliments in gluten free diet. One chapter deals with labeling of gluten free foods in the Czech Republic and in Canada. There is also a comparison of the Czech Republic and Canada regarding health care, government and restaurants offering gluten free dishes. The aim of the research is to map the selection of gluten free foods in people with diagnosed celiac disease, the second target is to explore differences in food selection in the Czech Republic and Canada. The third objective is focused on factors influencing the choice of gluten free foods. The research was conducted through a questionnaire survey. Facteurs déterminants du pouvoir de panification de l’amidon de manioc modifié par fermentation et irradiation UV / Determinant factors affecting breadmaking ability of cassava starch modified by fermentation and UV irradiationMaldonado Alvarado, Pedro Gustavo 21 May 2014 (has links)
Compte tenu que la capacité de panification de l'amidon aigre, amidon de manioc modifié par fermentation et irradiation UV, n'a pas encore été complètement élucidée, l'influence du génotype et les traitements post-récolte sur le pouvoir de panification de l'amidon aigre a été l'objectif de ce travail. 13 génotypes de manioc de Colombie, cultivés à 1000 m. et 1700 m.p.n.m (3 de plaine et 10 d'altitude, respectivement) modifiés par fermentation (0 ou30 jours) et séchage (four ou soleil) ont été étudiés. Les analyses de la taille des granules, la viscosité d'empesage RVA et la viscosité intrinsèque révèlent que les traitements post-récolte ont été les facteurs prépondérants en vue de l'amélioration de la capacité de panification tandis que le facteur génotype a présenté une influence moins importante. Parmi les traitements post-récolte, la fermentation a présenté un effet plus prononcé sur la structure des granules d'amidon, que le séchage au soleil. Cependant, la combinaison de ces deux traitements a été nécessaire pour obtenir une forte capacité de panification. L'effet d'altitude de culture a été mis en évidence lors de la fermentation par des sensibilités différentes trouvées entre les granules de plaine et d'altitude (exocorrosion et endocorrosion, respectivement). Les génotypes d'altitude, donc, ont montré une bonne capacité de panification. Néanmoins, d'autres facteurs entrent en jeu, la teneur en amylose mesurée par DSC a influencé négativement l'expansion de la pâte, probablement en raison de la formation de complexes amylose-lipide. Par conséquent, la combinaison de génotypes de manioc (la teneur en amylose principalement) et les traitements post-récolte sont un élément clé de la capacité de panification de l'amidon aigre. Ces contributions peuvent aider à l'établissement de standards de qualité de l'amidon aigre vis à vis de leur potentielle utilisation dans la fabrication des produits sans-gluten. / The breadmaking ability of sour cassava starch, cassava starch modified by fermentation and UV irradiation, have not been yet fully elucidated. The influence of genotype and postharvest treatments on expansion ability of sour cassava was the objective of this work. Thirteen cassava genotypes have been studied. All genotypes proceeded from Colombia. Two altitudes of origin (1000 m. and 1700 m.a.s.l. refered to respectively as lowland and highland), fermentation treatments (0 or 30 days) and drying treatments (oven or sun) were considered. Analyses of the granule size, RVA pasting behavior and intrinsic viscosity were analysed as indicators of breadmaking ability. Results showed that post-harvest treatments were prevailing factors in improving breadmaking ability while the genotype factors had a smaller influence. Among post-harvest treatments, fermentation had a greater influence than sun-drying on these indicators, in particular on starch granular structure. The combination of both treatments was needed to obtain a high capacity bread. Furthermore, the altitude of origin of cassava genotypes was observed to affect the fermentation process: different sensitivities to fermentation were found between granules of lowland and highland (exocorrosion and endocorrosion, respectively). Highland genotypes showed a better breadmaking ability. Other influencing factor related to genotype was the amylose content measured by DSC: it negatively influenced the expansion of the dough, probably due to the formation of amylose-lipid complex. As a conclusion, the combination of cassava genotypes (mainly amylose content) and post-harvest treatment is key for expansion ability of sour starch. This knowledge is a first, essential step to provide quality standards for sour starch, targeted particularly for use in gluten-free products.

Impact of Whole Grain Consumption Compliance on Gluten Sensitivity and Bowel HealthRoberts, Sarah Anne 01 January 2015 (has links)
While many health benefits have been associated with increased whole grain consumption, current researchers have not considered if the consumption of whole grains in currently recommended or higher amounts actually leads to health problems, specifically to a correlated increase in gluten sensitivity. The purpose of this study was to determine if diets high in whole grains or those that met the recommended daily intake of whole grains help minimize or increase gluten sensitivity, and when whole grains are consumed as recommended if they cause more harm than good. The theoretical basis for this quantitative, cross sectional design was the precaution adoption process model, allowing for the examination of preventive behaviors as a series of cognitive steps over time. Individuals (N = 5,746) from the National Health and Nutrition Examination Survey 2007 to 2012 were assessed for daily intake of whole grains before and after the release of the 2010 Dietary Guidelines for Americans, and from 2007 to 2010 for bowel health and sensitivity to whole grains. SAS correlations and regression analysis at p < .05 were analyzed. There was an increase in whole grain intake by 7.4% and in bowel sensitivity with 50% reporting increases in gas, but more data are needed to determine exact amounts that caused these increases in symptoms. Understanding the complete picture, policy makers and others will be more informed about current recommendations and the way that Americans eat, as well as if changes are needed for the future.

Exploring Dietary Sacrifice in Intimate Relationships for Couples with Celiac DiseaseAlley, Lindsey Marie 30 March 2015 (has links)
Prior research on eating behaviors has shown that romantic partners actively merge their dietary preferences throughout the course of a relationship and find significant value in cooking and eating the same foods together at the same times. Yet, little is known regarding the impacts of specific dietary support processes involved in maintaining said communal diet when one partner drastically alters his or her eating patterns. The current study defined dietary sacrifice as a phenomenon within the context of Celiac Disease (CD): a chronic illness that requires strict adherence to the gluten-free diet (GFD). Drawing from existing research on sacrifice within romantic relationships (e.g., Impett & Gordon, 2008), this project examined whether non-Celiac partners' adherence to the GFD during shared mealtimes impacted relationship satisfaction for both couple members. Female Celiacs and their non-Celiac cohabitating partners (N=152 couples) were recruited for an online survey through various support organizations. Given the dyadic design of this study, the Actor-Partner Interdependence Mediation Model (APIMeM; Ledermann, Macho, & Kenny, 2011) was used to examine the mediating influence of Dietary Approach and Avoidance Motives. Results indicated that partner support in the form of shared GFD adherence bolstered couple happiness to the extent that it was performed for positive gains (e.g., promoting health and well-being) by the non-Celiac. While dietary sacrifice was positively associated with Celiacs' relationship satisfaction above and beyond non-Celiacs' endorsement of Dietary Avoidance Motives, both dyad members experienced significantly lower relationship satisfaction when non-Celiac partners adhered to the diet to deflect negative outcomes (e.g., rejection, fighting). This study serves as the first application of relationship sacrifice research to a specific health issue, and the first psychological exploration into intimate partners' dietary support processes within the Celiac population.

Schott como materia prima en forma de almidón o harina es una alternativa sostenible y nutritiva al trigo y otros granos. Esta alternativa permite a los agricultores minimizar las pérdidas después de su cosecha y garantizar la seguridad alimentaria ya que sus propiedades nutricionales, digestivas y saludables son reconocidas por la comunidad científica. Sin embargo, la información existente sobre este rizoma dirigida a su aplicación es bastante limitada. Esta tesis expone la caracterización funcional y tecnológica de los almidones obtenidos a partir de cormos y cormelos de Xanthosoma sagittifolium (L.) Schott. Asimismo, se evaluó tecnológicamente el efecto de la combinación de enzimas, hidrocoloides, almidón de patata, harina pregelatinizada sobre la harina de los cormelos de la Colocasia esculenta (L.) Schott en el desarrollo de un pan sin gluten. Además, se evaluaron las propiedades tecnológicas y digestivas de una fórmula básica para puré desarrollada a partir de harina de cormelos de Xanthosoma spp. y Colocasia spp. Además, la revisión bibliográfica realizada permitió poner en contexto los efectos saludables demostrados clínicamente de esta materia prima y sus componentes. Se demostró que existen diferencias significativas entre el almidón de cormos y cormelos de la misma especie. Se concluyó que la harina de Colocasia esculenta (L.) Schott es una buena opción para incrementar el valor nutricional de los panes sin gluten. Entre las estrategias probadas, el pan elaborado a partir de la mezcla con almidón de patata resultó la menos aconsejable. Además, todas las estrategias aplicadas originaron panes con menor índice glucémico que sus homólogos sin gluten reportados en otros estudios. Por primera vez, este trabajo recomienda el uso de harina de cormelos de Xanthosoma sagittifolium (L.) Schott y Colocasia esculenta (L.) Schott y para desarrollar purés con un valor nutricional agregado. La revisión bibliográfica realizada permitió recopilar los efectos demostrados clínicamente, concretamente antihiperglucémicos, antihepatotóxicos, antihipertensivos, hipoglucemiantes, anticancerosos, hipolipidémicos y prebióticos, entre otros, de los compuestos bioactivos presentes en esta planta. / [CA] L'ús de Colocasia esculenta (L.) Schott and Xanthosoma sagittifolium (L.) Schott as a raw material in form of starch or flour is a nutritious and sustainable alternative to wheat and other grains. This alternative allows farmers to minimize losses after harvest and guarantee food safety since its nutritional, digestive and healthy properties are recognized by the scientific community. Nevertheless, there is still scarce information about these rhizomes that limit their application. This thesis illuminates the functional and technological characterization of the starches obtained from corms and cormels of Xanthosoma sagittifolium (L.) Schott. Likewise, the effect of the combination of enzymes, hydrocolloids, potato starch, pregelatinized flour on Colocasia esculenta (L.) Schott cormels flour toward the development of a gluten-free bread was technologically evaluated. Furthermore, the technological and digestive properties of a basic formula for pastes developed from cormels flour of Xanthosoma sagittifolium (L.) Schott and Colocasia esculenta (L.) Schott was evaluated. In addition, a review was conducted and the health effects clinically demonstrated are exposed. It was shown that there are significant differences between the starch of corms and cormels of the same species. It was concluded that the flour from Colocasia esculenta (L.) Schott cormels is a good option to increase the nutritional value of gluten-free breads. Among the strategies tested, the bread made from mixtures with potato starch was the least desirable, but all strategies tested gave breads with lower glycemic index than gluten-free counterparts reported in other studies. For the first time, this works recommends the use of cormels flour from Xanthosoma sagittifolium (L.) Schott and Colocasia esculenta (L.) Schott was recommended to develop pastes to nutritional value to gluten-free products. Influence du procédé de congélation sur les levures et les propriétés techno-fonctionnelles des pâtes sucrées (type Kougelhopf) / Effect of freezing tretment on yests cells and techno-fonctionnel properties of sweet Doughs (Kougelhopf)Meziani, Smaïl 15 November 2011 (has links)
Les pâtes surgelées sont relativement stables et peuvent être fabriquées à l’échelle industrielle, distribuées et cuites à la demande au moment de la vente ou de la consommation (point chaud). La congélation des pâtes sucrées induit une baisse de volume et une augmentation du temps de fermentation, ces conséquences sont dues à deux facteurs : la baisse de la production de CO2 (viabilité des levures) et la faible capacité de rétention de gaz du réseau gluténique. La perte de la qualité des pâtes congelées est accélérée durant le stockage. Cette thèse porte sur l’étude de l’effet de la congélation et de la conservation sur les levures et les propriétés techno-fonctionnelles des pâtes sucrées type Kougelhopf. Ce travail vise à l’étude de l’impact de la vitesse de congélation sur les propriétés microbiologiques, rhéologiques, structurales et sensorielles de ces pâtes. Elles ont été congelées à différentes températures (-20 °C, -30 °C, -40 °C et une immersion dans l'azote liquide) puis conservées à -40 °C pendant 9 semaines. Les principaux résultats de cette étude ont permis de mettre en évidence le rôle de la vitesse de congélation et de la durée de conservation sur les propriétés intrinsèques des pâtes sucrées surgelées. Il en découle que l’activité fermentaire et l’intégrité du réseau du gluten sont tributaire de la vitesse de congélation. En effet, cette dernière contrôle la taille et la localisation des cristaux de glace d’où la recherche d’un compromis entre une vitesse de congélation ni trop rapide pour diminuer la viabilité des levures ni trop lente pour former de gros cristaux pouvant perforer le réseau de gluten de la pâte. Ce travail a démontré que le surdosage de levure reste valable uniquement pour les pâtes sucrées surgelées destinées à être conservées au-delà de 4 semaines. Ce surdosage améliore ainsi la qualité globale du Kougelhopf en compensant la perte de l'activité des levures pendant la congélation et le stockage / The frozen doughs are relatively stable and can be manufactured on an industrial scale, distributed and baked on demand at the point of sale or consumption (Bake-off). Freezing sweet dough induces a decrease in specific volume and an increase in fermentation time, these effects are due to two factors: lower production of CO2 (yeast viability) and losing capacity to retain gas (gluten network integrity). The loss of quality of frozen dough is accelerated during storage. This study focuses on the freezing and frozen storage effects on Kougelhopf sweet doughs. The aim of this work is to study the impact of freezing rate on microbiological, rheological, structural, and sensory properties of sweet doughs. The sweet doughs were frozen at different temperatures (-20°C, -30°C, -40°C and an immersion in liquid nitrogen) and stored at -40°C for 9 weeks. The main results obtained showed an impact of freezing rate and frozen storage duration on the frozen doughs intrinsic properties. This study shown the dependence of fermentation activity and integrity of the gluten network with freezing rate, which controls size and location of ice crystals resulting in research of a compromise between freezing rate nor too fast to reduce yeast viability, nor too slow to form large ice crystals that could perforate gluten network. Added the yeast amount is necessary only for frozen sweet doughs to be stored beyond 4 weeks, which improves the overall quality of Kougelhopf by compensating for yeast activity decrease during freezing and frozen storage

Name: Life of coeliac and his family in these days Aim: The aim of my diploma thesis is to map life difficulties of people with celiac disease. First of all, it is a problem with gluten-free diet which is essential for celiac disease. Problems with eating out, shopping for ingredients for meal preparation at home, expensiveness of this eating, and last but not least, support from insurance companies and the state. Methods: The theoretical part includes findings about celiac disease as an illness, it means the characteristics of its origin, diagnostics and treatment. Furthermore, findings about gluten- free diet and problems related to this kind of eating were worked. Last but not least, diet and availability of food for gluten-free eating are mentioned. I applied a survey method in the practical part, in which case an anonymous survey was created electronically by using Google forms, and then sent out via internet. The survey was filled out by 296 respondents. Outcome: We can see from the outcomes that most of the respondents sometimes break the diet due to social integration.
