Teaching English reading comprehension to Grade 2 First Additional Language learners

English reading comprehension is an educational challenge worldwide. This study investigated how Grade 2 teachers teach English reading comprehension to First Additional Language (FAL) learners at three selected independent schools in Gauteng where the language of learning and teaching (LoLT) is English. Three English medium independent schools in Gauteng, South Africa participated in the study. The participants at each research site consisted of Grade 2 teachers (main participants) and Grade 2 learners (secondary participants). Grade 2 teachers were selected purposively on the criteria that they had taught for more than two years and were teaching English reading comprehension to Grade 2 FAL learners. This study followed a qualitative approach, and was situated within the interpretivist paradigm. The research design accommodated a case study. The conceptual framework was based on the revised taxonomy by Krathwohl and Anderson for questioning which integrated the role of the teacher in a reading lesson. The three stages of reading, before reading, during reading and after reading were divided into the different levels of thinking and reasoning as suggested in Vygotsky's Zone of Proximal Development (ZPD) theory. The data collection strategies were semi-structured interviews with Grade 2 teachers; observing reading lessons they gave; and keeping a journal in which personal reflections on the investigation into teaching and learning practices were documented. Once data was collected, the recordings of the semi-structured interviews were transcribed. The observation checklists and the reflection journal entries were analysed. Data was analysed using a coding process and five themes and twelve sub-themes emerged. The findings show that the interactive approach was common in the teaching of English reading comprehension. However, parental involvement, the availability of the trained teachers and appropriate resources were necessary. A learner's individual reading needs and reasoning ability have to be considered through the use of differentiated teaching practices. Questioning throughout the reading process improves a learner's understanding of the text. / Die leesbegrip van Engels is 'n wêreldwye uitdaging. Hierdie studie het ondersoek hoe Graad 2-onderwysers onderrig in leesbegrip in Engels aan leerders van Engels as Eerste Addisionele Taal gee by drie uitgesoekte onafhanklike skole in Gauteng waar die taal van leer en onderrig (TLO) Engels is. Drie onafhanklike Engelsmediumskole in Gauteng, Suid-Afrika het aan die studie deelgeneem. By elke skool was die deelnemers Graad 2-onderwysers (hoofdeelnemers) en Graad 2-leerders (sekondêre deelnemers). Graad 2-onderwysers is doelbewus gekies op grond van die kriterium dat hulle langer as twee jaar onderrig gegee het en ten tye van die studie Graad 2-EAT-leerders onderrig het. Die studie volg 'n kwalitatiewe benadering binne die interpretivistiese paradigma. Die navorsingsontwerp behels 'n gevallestudie. Die konseptuele raamwerk is gebaseer op Krathwohl en Anderson se hersiene taksonomie van vraagstelling wat die onderwyser se rol in 'n leesles integreer. Die drie leesstadia voor, gedurende en na lees word in die verskillende vlakke van lees en redenering verdeel, soos wat in Vygotsky se teorie van die Sone van Naaste Ontwikkeling (ZPD) aan die hand gedoen word. As dataversamelingstrategieë is semigestruktureerde onderhoude met Graad 2-onderwysers gevoer, lesse waargeneem en 'n joernaal gehou waarin persoonlike besinning oor die ondersoek van onderrig- en leerpraktyke gedokumenteer is. Na dataversameling is die opnames van die semigestruktureerde onderhoude getranskribeer. Die waarnemings-kontrolelyste en die inskrywings in die besinningsjoernaal is ontleed. Data is met behulp van 'n koderingsproses ontleed, wat vyf temas en twaalf subtemas na vore gebring het. Daar is bevind dat die interaktiewe benadering algemeen in die onderrig van leesbegrip van Engels gebruik word. Die betrokkenheid van ouers, beskikbaarheid van opgeleide onderwysers en geskikte hulpbronne was egter noodsaaklik. Leerders se individuele leesbehoeftes en redeneringsvermoë moet by wyse van gedifferensieerde onderrigpraktyke in aanmerking geneem word. Phosphoinositide-3-kinase/akt - Dependent Signaling is Required for Maintenance of [Ca<sup>2+</sup>]<sub>I,</sub>I<sub>Ca</sub>, and Ca<sup>2+</sup> Transients in HL-1 Cardiomyocytes

The phosphoinositide 3-kinases (PI3K/Akt) dependent signaling pathway plays an important role in cardiac function, specifically cardiac contractility. We have reported that sepsis decreases myocardial Akt activation, which correlates with cardiac dysfunction in sepsis. We also reported that preventing sepsis induced changes in myocardial Akt activation ameliorates cardiovascular dysfunction. In this study we investigated the role of PI3K/Akt on cardiomyocyte function by examining the role of PI3K/Akt-dependent signaling on [Ca 2+]i, Ca2+ transients and membrane Ca2+ current, ICa, in cultured murine HL-1 cardiomyocytes. LY294002 (120 μM), a specific PI3K inhibitor, dramatically decreased HL-1 [Ca 2+]i, Ca2+ transients and ICa. We also examined the effect of PI3K isoform specific inhibitors, i.e. α (PI3-kinase α inhibitor 2; 28 nM); ? (TGX-221; 100 nM) and γ (AS-252424; 100 nM), to determine the contribution of specific isoforms to HL-1 [Ca 2+]i regulation. Pharmacologic inhibition of each of the individual PI3K isoforms significantly decreased [Ca2+]i, and inhibited Ca 2+ transients. Triciribine (120 μM), which inhibits AKT downstream of the PI3K pathway, also inhibited [Ca2+]i, and Ca 2+ transients and ICa. We conclude that the PI3K/Akt pathway is required for normal maintenance of [Ca2+]i in HL-1 cardiomyocytes. Thus, myocardial PI3K/Akt-PKB signaling sustains [Ca 2+]i required for excitation-contraction coupling in cardiomyoctyes.

Pratima Sadangi in a Ph.D. thesis submitted to Utkal University proved results on degree of approximation of functions by operators associated with their Fourier series. In this dissertation, we consider degree of approximation of functions in Hα,ρ by different operators. In Chapter 1 we mention basic definitions needed for our work. In Chapter 2 we discuss different methods of summation. In Chapter 3 we define the Hα,ρ metric and present the degree of approximation problem relating to Fourier series and conjugate series of functions in the Hα,ρ metric using Karamata (Κλ) means. In Chapter 4 we present the degree of approximation of an integral associated with the conjugate series by the Euler, Borel and (e,c) means of a series analogous to the Hardy-Littlewood series in the Hα,ρ metric. In Chapter 5 we propose problems to be solved in the future.

L’altération de la barrière hématoencéphalique (BHE) par les cellules tumorales et les cellules immunes circulantes peut mener à la neuroinflammation. Les cellules leucémiques promyélocytaires HL-60 sont un excellent modèle pour étudier et comprendre les mécanismes de signalisation moléculaires qui caractérisent le développement tumoral et métastatique. La cancérogenèse peut s’accompagner de modulations de l’expression de biomarqueurs tels que la cyclooxygénase-2 et la métalloprotéase-9. Les recherches décrites dans ce mémoire relatent l’analyse des biomarqueurs inflammatoires et invasifs régulés lors de la différenciation induite par le PMA des cellules HL-60 en macrophages. Le statut de différenciation cellulaire pourrait avoir un impact sur les gènes cibles de la voie NF-κB. Nous émettons l’hypothèse que le PMA active la voie NF-κB et que cette signalisation peut être renversée par l’(-)-épigallocatéchine-gallate (EGCG). En effet, une régulation à la hausse de l’expression de plusieurs gènes combinée à la diminution de l’expression d’IκB mettent en évidence l’implication de la voie NF-κB dans l’activation des mécanismes pro-inflammatoires et pro-invasifs. Les mêmes observations sont faites dans les cellules différenciées appelées «macrophages-like». L’EGCG, un polyphénol dérivé du thé vert, a un potentiel chimiopréventif. Il est capable d’inhiber la signalisation moléculaire passant par la voie NF-κB dans les cellules HL-60 traitées simultanément par l’EGCG et le PMA, mais pas dans les cellules «macrophages-like». Cette différence peut s’expliquer par une modulation de l’expression du récepteur de surface cellulaire de l’EGCG, le récepteur à la laminine de 67 kDa, et de son précurseur de 37 kDa. Collectivement, nos résultats montrent que le statut de différenciation des cellules promyélocytaires HL-60 concorde avec l’activation des mécanismes favorisant le développement d’un cancer et des métastases. Cet effet peut être prévenu par l’utilisation d’agents naturels tel l’EGCG. Le ciblage de biomarqueurs liés au statut de différenciation des cellules tumorales impliquées dans la perturbation de la barrière hématoencéphalique qui cause la neuroinflammation permettrait l’avancement des connaissances dans la prévention de la cancérogenèse. / Blood-brain barrier (BBB) disruption by circulating tumor and immune cells leads to secondary inflammatory infections. Promyelocytic HL-60 cells represent an excellent model to study and to get a better understanding of the molecular signaling mechanisms involved in carcinogenesis and metastasis. The research described in this thesis shows the analysis of several inflammatory and invasive biomarkers regulated during PMA-induced differentiation of promyelocytic HL-60 cells into macrophages. Carcinogenesis involves some modifications in the expression of biomarkers such as cyclooxygenase-2 and matrix metalloprotease-9. The differentiation status could have an impact on the NF-κB signaling pathway that regulates the target genes, given that these target genes expression varies during cell differentiation. We hypothesize that the activation of the NF-κB pathway by PMA can be reverse by (-)-epigallocatechin-gallate (EGCG). Indeed, the up-regulation of downstream genes combined with the down-regulation of IκB expression showed the significant implication of the NF-κB signaling pathway to activate pro-inflammatory and pro-invasive mechanisms linked to carcinogenesis. The same evidence exhibits in the differentiated cells called «macrophages-like». Moreover, the green tea polyphenol, EGCG, shows chemopreventive property since it better inhibited NF-κB signaling in cells treated simultaneously with EGCG and PMA compared to the «macrophages-like». This difference could be due, in part, to the down-regulation of the 67 kDa laminin receptor, known to be the non-integrin membrane receptor for EGCG. All together, our results suggest that the differentiation status of promyelocytic cells is linked to the activation of mechanisms involved in carcinogenesis and metastasis. These phenomena can be prevented by using natural agents such as EGCG. Targeting the specific biomarkers linked to the differentiation status of tumor cells and involved in the disruption of the BBB may help reduce secondary neuroinflammation and enable the advancement of knowledge towards carcinogenesis prevention.

The aim of this diploma thesis Transforming of Executive Coalitions in Prague in the Electoral Period 2010 - 2014 is to analyze influences, which have an important role in a process of establishing executive coalitions and also in their stability. The analysis is based on a classical theory of coalitions, discursive analysis, interviews with direct participants and on official documents. One of the aims is to describe the level of possibility in application of theoretical concepts of coalition theory in the area of Prague's political system. There are many points of view, from which we look at stability and forming of coalitions. For example, very important point of view is an influence of making and implementing politics or an influence of politicians from national level. A big importance is focused on negotiating between participants, their statements in media and their real steps and decisions. The intention of this thesis is not an invention of general model which should be applied on similar local system, but to explain and describe the uniqueness of Prague local system and the election period of 2010 - 2014.

Coleridge’s Revisionary Practice from 1814 to 1818

Die Dissertation untersucht Samuel Taylor Coleridges Praxis der Selbstredaktion in den Jahren von 1814 bis 1818 und beleuchtet dabei die zentrale Rolle, die William Wordsworths The Excursion, 1814 als Teil von The Recluse erschienen, in der Herausbildung von Coleridges Œuvre einnimmt. Die Arbeit entwickelt ihre zentrale These zu Coleridges Überarbeitungspraxis durch eine detaillierte Analyse der Verfahren, über die Coleridge aufhörte, durch Wordsworth zu sprechen. Ich beziehe mich dabei in erster Linie auf die Überarbeitungen von Biographia Literaria (1817), Sibylline Leaves (1817) und dem 1818 erschienenen rifacciamento zu dem Periodikum The Friend, das ursprünglich 1809–1810 publiziert worden war. Vor dem Hintergrund von Coleridges in den 1790er- und 1800er-Jahren neu aufgekommener und sich später weiterentwickelnder Rezeption von Immanuel Kants kritischer Philosophie werde ich argumentieren, dass sich die „radikale Differenz“ zwischen Coleridge und Wordsworth, die seit den Lyrical Ballads (1798) und dem „Preface“ (1800) bestand, weiter verstärkte, nachdem es Wordsworth nicht gelungen war, das groß angelegte Konzept eines „first genuine philosophical poem“ – The Recluse – zu vollenden. Insbesondere nachdem Coleridge 1807 The Prelude gehört hatte, enttäuschte The Excursion nach den Maßstäben seiner „vergleichenden Kritik“ seine lang gehegten Erwartungen. Während sein organisches Weltbild einen aktiven Geist kannte, der nach universaler „Wahrheit“ sowohl durch innere synthetisierende Kräfte als auch empirische Naturgesetze strebt, gründete sich Wordsworths Gedicht auf ein obskur-labiles Fundament zwischen Außenwelt und Selbst. Coleridges aus seinen eigenen Theorien zu Sprache und Einbildungskraft erwachsene Enttäuschung über The Excursion und die nachfolgende Loslösung von Wordsworth und ihrem gemeinsamen Werk verschafften ihm letztendlich die notwendige Autonomie, die einerseits einen nüchternen Blick auf die eigene Vergangenheit und andererseits eine dialogische Freundschaft zu Wordsworth ermöglichten, innerhalb derer Coleridge die Bedeutsamkeit erkannte, zu einem Freund zu sprechen. / This thesis is an examination of Samuel Taylor Coleridge’s revisionary activity from 1814 to 1818, considering the integral role of William Wordsworth’s The Excursion, published as part of The Recluse in 1814, on Coleridge’s conception of his discrete oeuvre. It is via a detailed analysis of the way Coleridge ceased to speak “through” Wordsworth that this thesis unfolds its principal argument on Coleridge’s revisionary activity. I principally consider the revisions at work in the Biographia Literaria (1817), Sibylline Leaves (1817) and the 1818 rifacciamento to The Friend (the periodical originally issued in 1809-1810). Taking into account Coleridge’s newly-emerging and subsequently evolving responses to Immanuel Kant’s critical philosophy in the 1790s and 1800s, I will argue that the already-existing “radical Difference” between Coleridge and Wordsworth ever since the Lyrical Ballads (1798) and the “Preface” (1800) further intensified following Wordsworth’s failure to bring their grand scheme for a “first genuine philosophical poem”, The Recluse, into completion. Especially after The Prelude Coleridge heard in 1807, The Excursion by means of his “comparative censure” fell short of meeting the long-cherished expectations. Whereas Coleridge’s organic view of the world involved the recognition of an active mind seeking universal “Truth” through the inner synthetic faculties as well as the empirical laws in nature, Wordsworth’s poem was founded upon an obscurely precarious ground between the phenomenal world and the inner self. Ultimately, Coleridge’s disappointment with The Excursion on the basis of his theories on language and imagination, and the ensuing detachment from Wordsworth and their joint oeuvre gave him the autonomy to revise his past works in a way that ensured formation of a more sober relationship with his own past and a dialogic friendship with Wordsworth in which Coleridge came to realise the importance of speaking to a friend.

Corrections of high-order nonlinearities in the LHC and High-Luminosity LHC beam optics

Der Einfluss von Nichtlinearitäten höherer Ordnung der Magnetfelder auf die Leistung des Large Hadron Collider (LHC) und dessen geplante High-Luminosity-Aufrüstung, dem HL-LHC, wurde umfangreich untersucht. Insbesondere hat sich gezeigt, dass das Vorhandensein solcher Fehler in den Insertion Regions (IR) erhebliche Auswirkungen hat, bedingt durch hohe Beta-Funktionen und Feed-Down auf niedrigere Ordnungen aufgrund der Kreuzungsschemata. Augenmerk dieser Arbeit ist auf die Erforschung diverser Methoden zur effektiven Behandlung dieser Nichtlinearitäten höherer Ordnung gerichtet, mit dem Ziel, sie zu identifizieren und korrigieren, um die Strahloptik zu optimieren und die Maschinenleistung zu verbessern. Simulationsstudien werden eingesetzt, in denen mit verschiedenen Fehlerquellen assoziierte Resonanzantreibende Terme (RDTs) gezielt angegangen werden. Besondere Aufmerksamkeit gilt Dekapol- und Dodekapolfehlern, die in früheren Messungen im LHC schädliche Auswirkungen durch Feed-Down auf Amplituden-Detuning gezeigt haben. Die erwartete Erhöhung der Sensitivität der Optik gegenüber Fehlern in den IRs des HL-LHC unterstreicht weiter die Bedeutung der Behandlung dieser Fehler. Des Weitern werden Korrekturoptionen mit Hilfe der nichtlinearen Korrektorpaketen entwickelt. Experimentelle Studien werden durchgeführt, um die Ergebnisse zu validieren. Erhebliche Anstrengungen wurden unternommen, um die Feed-Down Effekte von Dekapol- und Dodekapol-Feldfehlern zu mindern. Um diese Herausforderung anzugehen, wurden neuartige Korrekturalgorithmen eingeführt, die erstmals die Dodekapol-Korrektoren in den IRs im operationellen Betrieb ansteuern. Die Ergebnisse dieser Experimente liefern wertvolle Erkenntnisse zur Minderung von Fehlern höherer Ordnung und tragen zum besseren Verständnis der Strahldynamik in modernen und zukünftigen Teilchenbeschleunigern bei. / The impact of high-order nonlinear magnetic field errors on the performance of the Large Hadron Collider (LHC) and its planned High-Luminosity upgrade, the HL-LHC, has been extensively studied. Particularly, the presence of such errors in the Insertion Regions (IR) has shown significant repercussions due to the high beta-functions and feed-down to lower orders caused by crossing schemes. This thesis aims to explore different methods for effectively addressing these high-order errors, with the ultimate goal of identifying and correcting them to optimize beam optics and enhance machine performance. Simulation studies are employed, using a novel and flexible correction algorithm developed during the course of this PhD research. Various strategies are investigated to improve corrections by targeting Resonance Driving Terms (RDTs) associated with diverse error sources. Special attention is devoted to decapole and dodecapole errors, which have demonstrated detrimental effects on amplitude detuning due to feed-down based on previous measurements in the LHC. The anticipated increase in optics sensitivity to errors in the IRs of the HL-LHC further underscores the importance of addressing these errors. Correction options are evaluated, focusing on the utilization of the nonlinear corrector packages to address errors in the new separation and recombination dipoles in the HL-LHC, where increased decapole errors had been expected. Experimental studies are conducted to validate the findings. Significant efforts are dedicated to mitigating the feed-down effects arising from decapole and dodecapole field errors. To address this challenge, novel corrections involving the operational implementation of dodecapole correctors in the IRs have been introduced for the first time. The results of these experiments provide valuable insights into the mitigation of high-order errors and contribute to the overall understanding of beam dynamics in advanced particle accelerators.

Simulation Studies of Digital Filters for the Phase-II Upgrade of the Liquid-Argon Calorimeters of the ATLAS Detector at the High-Luminosity LHC

Am Large Hadron Collider und am ATLAS-Detektor werden umfangreiche Aufrüstungsarbeiten vorgenommen. Diese Arbeiten sind in mehrere Phasen gegliedert und umfassen unter Anderem Änderungen an der Ausleseelektronik der Flüssigargonkalorimeter; insbesondere ist es geplant, während der letzten Phase ihren Primärpfad vollständig auszutauschen. Die Elektronik besteht aus einem analogen und einem digitalen Teil: während ersterer die Signalpulse verstärkt und sie zur leichteren Abtastung verformt, führt letzterer einen Algorithmus zur Energierekonstruktion aus. Beide Teile müssen während der Aufrüstung verbessert werden, damit der Detektor interessante Kollisionsereignisse präzise rekonstruieren und uninteressante effizient verwerfen kann. In dieser Dissertation werden Simulationsstudien präsentiert, die sowohl die analoge als auch die digitale Auslese der Flüssigargonkalorimeter optimieren. Die Korrektheit der Simulation wird mithilfe von Kalibrationsdaten geprüft, die im sog. Run 2 des ATLAS-Detektors aufgenommen worden sind. Der Einfluss verschiedener Parameter der Signalverformung auf die Energieauflösung wird analysiert und die Nützlichkeit einer erhöhten Abtastrate von 80 MHz untersucht. Des Weiteren gibt diese Arbeit eine Übersicht über lineare und nichtlineare Energierekonstruktionsalgorithmen. Schließlich wird eine Auswahl von ihnen hinsichtlich ihrer Leistungsfähigkeit miteinander verglichen. Es wird gezeigt, dass ein Erhöhen der Ordnung des Optimalfilters, der gegenwärtig verwendete Algorithmus, die Energieauflösung um 2 bis 3 % verbessern kann, und zwar in allen Regionen des Detektors. Der Wiener Filter mit Vorwärtskorrektur, ein nichtlinearer Algorithmus, verbessert sie um bis zu 10 % in einigen Regionen, verschlechtert sie aber in anderen. This effort also affects the liquid-argon calorimeters, whose main readout electronics will be replaced completely during the final phase. The electronics consist of an analog and a digital portion: the former amplifies the signal and shapes it to facilitate sampling, the latter executes an energy reconstruction algorithm. Both must be improved during the upgrade so that the detector may accurately reconstruct interesting collision events and efficiently suppress uninteresting ones. In this thesis, simulation studies are presented that optimize both the analog and the digital readout of the liquid-argon calorimeters. The simulation is verified using calibration data that has been measured during Run 2 of the ATLAS detector. The influence of several parameters of the analog shaping stage on the energy resolution is analyzed and the utility of an increased signal sampling rate of 80 MHz is investigated. Furthermore, a number of linear and non-linear energy reconstruction algorithms is reviewed and the performance of a selection of them is compared. It is demonstrated that increasing the order of the Optimal Filter, the algorithm currently in use, improves energy resolution by 2 to 3 % in all detector regions. The Wiener filter with forward correction, a non-linear algorithm, gives an improvement of up to 10 % in some regions, but degrades the resolution in others. Detector Development for the High Luminosity Large Hadron Collider

Um das Entdeckungspotential des Large Hadron Colliders auszunutzen, wird er beginnend 2024 zum High Luminosity Large Hadron Collider ausgebaut. Neue Detektorherausforderungen entstehen durch die höhere instantane Luminosität und den höheren Teilchenfluss. Der neue ATLAS Inner Tracker wird den aktuellen Spurdetektor ersetzen, um mit diesen Herausforderungen umzugehen. Es gibt viele Pixeldetektortechnologien zur Teilchenspurerkennung, jedoch muss ihre Eignung für den ATLAS Inner Tracker untersucht werden. Aktive Hochspannungs-CMOS-Sensoren, die in industriellen Prozessen produziert werden, bieten eine schnelle Auslese und Strahlenhärte. In dieser Arbeit wird der HV2FEI4v2-Sensor, der kapazitiv mit dem ATLAS-FE-I4-Auslesechip gekoppelt ist, dahingehend charakterisiert, ob er für eine Verwendung in einer der äußeren Lagen des ATLAS Inner Tracker geeignet ist. Schlüsselgrößen des Prototypens, wie die Treffereffizienz und die Subpixelentschlüsselung, werden untersucht. Der frühe HV2FEI4v2-Prototyp zeigt vielversprechende Ergebnisse, die als Ausgangspunkt für weitere Entwicklungen dienen. Aktive CMOS-Sensoren stellen einen möglichen Kandidaten für einen kosteneffizienten Detektor für den High Luminosity Large Hadron Collider.
