Biochemical and Functional Characterization of Novel RNA-binding Proteins Interacting with SMN in Motor Neuron-derived Cells

Spinal muscular atrophy is an autosomal recessive genetic disease that results from the loss and/or degeneration of alpha motor neurons in the lower part of the spinal cord. With ~ 1 in 6000 live births per year being affected, this disease is the second leading cause of infant death and is caused by the loss or decrease of the Survival of Motor Neuron protein (SMN). While a lot is known about the role that SMN plays in the cytoplasmic assembly of spliceosomal small nuclear ribonucleoproteins (snRNPs), it remains a crucial question in the field to gain a better understanding of what specific/distinct function(s) SMN might have in motor neurons. We have identified novel interactions between SMN and two RNA-binding proteins (RBPs) known to be components of axonal RNA granules. More specifically, we demonstrated that SMN interacts with HuD and SERBP1 in a direct fashion in foci-like structures along neurites of motor neuron-derived cells. We have also demonstrated that the SMN/HuD interaction is required for the localization of HuD into RNA granules in neurites of motor neuron-derived cells. Furthermore, I have shown that SERBP1 is down-regulated in the absence of normal levels of SMN and, most importantly, that over-expression of SERBP1 can rescue SMA-like neuronal defects using a cell culture model of the disease. These findings may help shed light on the non-canonical molecular pathway(s) involving SMN and RBPs in motor neurons and underscores the possible therapeutic benefits of targeting these RBPs in the treatment of SMA.

Public Housing in the United States: Using Sustainable Urbanism to Combat Social Exclusion

The United States government has taken steps to assure underprivileged citizens housing in the form of public housing through the Department of Housing and Urban Development, as well as various humanitarian programs in the form of homeless shelters. Yet, all housing is not equal. Our freedom to choose where we live and what type of house we live in is one revered aspect of life as a United States citizen. We can express our individuality, creativity, and personality through the architectural style of our homes. In this sense it is hard to ask for equal housing. I am suggesting that equality comes from adequate access to social participation, social integration, and power, in other words social inclusion. (Room, 7) Investigating and correcting the causes of social exclusion in order to create social inclusion is necessary in order to ascertain equality of housing in America. Sir Winston Churchill once said, “We shape our buildings and afterward our buildings shape us.” (Gieryn, 35) But what happens when you do not have power over the shaping of your own building? Is your building still a reflection of yourself? And if we allow building to include the environment in which your home is located, what if previous use of the land left it degraded? Will the degradation shape you? My thesis considers the question: does the environmental quality and architectural style of public housing in the United States facilitate the social exclusion of these communities? If so, what best practices can we take away from models that have been successful at combatting social exclusion? By answering these questions I strive to develop a proposal to right this currently unjust situation.

Trycksår : Omvårdnadsåtgärder vid trycksårsprevention och sårläkning

Syftet med denna systematiska litteraturstudie var att ta reda på vilka mätinstrument som används för att identifiera patienter med risk att utveckla trycksår samt beskriva vilka omvårdnadsåtgärder som har betydelse för trycksårsprevention och sårläkning. De vetenskapliga artiklarna (n=16) som ingick i denna studie söktes i databasen Elin@dalarna. Sökorden som användes var pressure ulcer i olika kombinationer med nutrition, activity, skin, Norton, treatment och hydrocolloid. Inklusionskriterierna var att artiklarna skulle vara publicerade mellan åren 1999-2006 och vara i fulltext samt svensk och/eller engelskspråkig. Resultatet visade att patienter borde riskbedömas för trycksår så snart som möjligt efter att de anlänt till vårdinrättningen. Bedömningen kunde göras med ett flertal olika evidensbaserade mätinstrument såsom Norton, modifierad Norton, Braden, modifierad Braden, och RAPS skalorna samt bedömning av BMI, serum albuminvärde och hudtemperatur. Sjuksköterskans omvårdnadsåtgärder gällande prevention och sårläkning var att snabbt mobilisera patienter. Hos immobiliserade patienter var det viktigt att dessa vändes varannan timme samt att sjuksköterskan använde kuddar, fårskinn eller anti- decubitus madrasser för att tryckavlasta huden i denna patientgrupp. Resultatet visade även att kosttillägg hade stor betydelse för trycksårsprevention och sårläkning. Det var viktigt att sjuksköterskan rengjorde såret med tvål/vatten eller natriumklorid och använde hydrocolloida förband för att få en optimal sårläkning.

2D vs 3D in a touch-free hand gesture based interface : An exploration of how 2D and 3D visual aids affect a user’s ability to learn a new interface

3D is a popular topic as an increasing amount of media and technology begin to support 3D interaction.  With the rise of interest in 3D interaction, the question of why there is a demand and desire for 3D over 2D interaction becomes relevant. This thesis compares the differences between a 3D heads up display and a 2D heads up display for a touch free gesture based virtual keyboard.  The gesture interface used in the tests is a way of communicating with a system using gestures of the hands tracked by a motion sensor.  This thesis tested 16 users where half of the users used a 2D version of a heads up display and the other half used a 3D version of a heads up display.  Both user groups were tested with identical conditions and in an identical environment.  Raw statistical data was gathered from a logging mechanism in the interface and qualitative data was gathered from questionnaires and observation notes.  The results from the experiment showed that the 2D and 3D heads up display gave very similar results. However, results also showed slightly better qualitative results from the 3D heads up display observation and questionnaire data.  The conclusion indicated no clear advantage for the 2D version or the 3D version.  The discussion shows that many other factors in the design process and selection of users, play a large role in the comparison of 2D vs 3D visualizations.  Factors such as age and familiarity with different levels of technology are indicated to be contributing factors when comparing 2D vs 3D.  The results and discussion hope to provide a starting point for future comparison research in the field of 2D compared to 3D visualization.

A utilização de motores elétricos diretamente nos cubos de roda é uma alternativa de propulsão para um veiculo elétrico ou híbrido muito interessante, pois não necessita do uso de sistemas complexos de transmissão, tornando o conjunto mecânico muito mais simples e, consequentemente, reduzindo a sua massa, atritos e custos. Entretanto, a adição dos motores nas rodas causa o incremento da massa não suspensa, e isto irá afetar o comportamento de dirigibilidade do veículo. Dessa forma, com ajuda da ferramenta de multicorpos, será identificado o que ocorrerá devido ao acréscimo de massa compatível com os motores elétricos de última geração. Para isto, uma série de análises comparativas será realizada, com modelos de veículos baseados na técnica de multicorpos, para o caso de um carro de passeio compacto. Primeiramente, uma análise modal comparando um carro convencional aos carros com a utilização dos motores, no eixo traseiro ou no dianteiro, e uma posterior avaliação das frequências obtidas. Em sequência, também foram realizadas manobras padrão com o modelo de veículo completo e foram observadas maiores influências nos resultados das análises em regime transiente, como a manobra de troca de faixa e a manobra do anzol (\"fishhook\"). Com a ajuda destes resultados, foi possível identificar que as instalações dos motores elétricos nos cubos traseiros causam uma menor influência negativa no desempenho de dirigibilidade, quando comparado com o caso instalado no eixo dianteiro. Através de uma otimização realizada com a ajuda de uma análise de sensibilidade das variáveis do sistema (D.O.E.), baseada na manobra do anzol, foi possível identificar que alterações nas molas, amortecedores e barra estabilizadora são capazes de mitigar os efeitos indesejáveis causados pelo incremento de massa não suspensa. / The electric motors directly in wheel hubs usage is an alternative of propulsion for a electric or hybrid vehicle, since it does not need the use of complex systems of transmission, making the mechanical assembly simpler and, consequently, reducing its mass , frictions and costs. However, the addition of the motors in the wheels causes an increase unsprung mass, and this will affect the vehicle handlings behavior. In that way, with the assistance of the multibody tool, it will be identified the consequences of mass addition, compatible with the moderns electric motors of. In this work, a series of comparative analyzes will be carried out, with vehicle models based on multibody techniques, in the situation of a compact car. First, a modal analysis comparing a conventional car to with the vehicle using hub driven motors, in rear or front axle, and an evaluation of the obtained frequencies. In the sequence, standard maneuvers were also performed with the complete vehicle model, and greater influences were observed in the transient analysis results, such as lane change and fishhook maneuver. With help of these results, it was possible to identify; that the installation of the electric motors in the rear hubs causes a smaller negative influence on the handling performance when compared to the case installed on the front axle. Through an optimization performed with the aid of a system variable sensitivity analysis (D.O.E.) based on the fish hook maneuver, it was possible to identify thtat changes on springs, shock absorbers and stabilizer bar are able to mitigate the undesirable effects caused by the increase of unsprung mass.

The Evolution of HUD in The Elder Scrolls and Deus Ex

This thesis explains the process of two researchers conducting a formal analysis of the graphical user interfaces (GUIs), specifically the heads-up displays (HUDs), of the main entries of the two game series The Elder Scrolls (1994 - 2011) and Deus Ex (2000 - 2011). The goal: to see how the evolution of their HUDs over time compare and to analyse them. After playing the games and taking notes about their HUDs, the researchers find that there is a common trend between the two series. The HUDs become more simplified over time, with some non-critical elements of the HUD being removed. They also become more precise at instructing and informing players about how to play the games. While similar additions and changes are made, the timing is different and the researchers conclude that further research is necessary in order to decide if the observed pattern of HUD evolution is unique to the two series or can hold up as a framework for a larger selection of games. As is, this thesis presents the patterns and trends within the heads-up display design of the observed games as well as a library of the observed games’ GUI elements, that future GUI designers and researchers can use to be made aware of past design decisions and revisions that has been common throughout several other games within the genre. / Denna kandidatuppsats går igenom undersökningen utav heads-up displays (Förkortas till HUD, uttalas “hadd”) som en del av de grafiska användargränssnitten tillhörande spelserierna The Elder Scrolls (1994-2011) och Deus Ex (2000-2011). Målet var att se till hur dessa utvecklades med tidens gång. Efter att data hade samlats in från spelen kom författarna fram till att det fanns en gemensam trend för hur HUDar i de två spelserierna utvecklades. Dessa HUDar hade blivit simplifierade med tiden, där icke-kritiska element försvann. De hade också blivit bättre på att instruera och informera spelare i hur de spelar spelen. Samtidigt som liknande tillägg och förändringar skedde för båda spelserierna så skiljde sig tidpunkten för när detta skedde. Författarna kom till slutsatsen att mer forskning är nödvändigt för att det ska kunna fastslås huruvida det observerade mönstret är unikt till de observerade spelserierna eller kan appliceras på andra spelserier också. Denna kandidatuppsats presenterar de mönster, designtrender, samt ett bibliotek av de komponenter, som funnits i datorspelens HUDar och kan användas av gränssnittsdesigners och forskare för att få insikt om tidigare designval och förändringar som har blivit vanligt förekommande genom flera andra spel av samma genre.

Bedömning av tryckskador hos individer med mörk hudpigmentering / Assessment of pressure injuries in individuals with dark skin pigmentation

Introduktion: Trycksår räknas som en vårdskada, som drabbar mest individer som är sängbundna och rullstolsburna.Trycksår är associerade med många olika medicinska tillstånd. Vårdskadan kan leda till förlängd vårdtid hos individer samt krävar mer kostnad. Personer med mörkt hudpigment löper större risk att utveckla trycksår. Detta pga. att det är svårt att identifiera kategori 1 trycksår hos de personerna. Syfte: Syftet med studien är att undersöka hur sjukvårdspersonal kan identifiera tryckskador i hudkostymen hos individer med mörk hudpigmentering samt att ta reda på orsaken till brist på vägledningen.  Metod: Deskriptiv litteraturstudie med utgångspunkt från kvantitativa studier. Tio kvantitativa artiklar har använts, varav fem artiklar hittades vid primär sökning medan resten hittades vid sekundär och tredje sökning. Databaser pubmed och Cinahl använts vid artikelsökningar. Resultat: Det finns några metoder som är relevanta för kliniska bedömningar av trycksår hos individer med mörk hudpigmentering. Trycksår kan identifieras hos personen med mörk hudpigment med hjälp av ultraljud, munsell färgtabeller och bildtekniker. Brist på vägledning om hur trycksår identifieras hos individer med mörk hudfärg syns redan under sjuksköterskeutbildningen, vilket tydliggör att högskolor och universitetet inte bidrar mer undervisning om trycksår riktad mot personer med mörkt hudpigment. Slutsats: Det går inte att dra slutsatser pga att studier med låg kvalitet inkuluderades till studien. Det finns metoder som kan användas vid kliniska bedömningar av trycksår hos personer med mörk hudpigmentering bland annat med hjälp av ultraljud och bildtekniker. Dock syns fortfarande brist på vägledning och därför skulle vidare forskning inom området vara nödvändigt. / Background : Pressure ulcer is considered a medical injury, which affects mostly people who are bedridden and wheelchair bound. Pressure ulcer is associated with many different medical conditions. The care injury can lead to extended care time for individuals and is more costly. People with dark skin pigments are at greater risk of developing pressure ulcers. This is due to that it is difficult to identify category 1 pressure ulcer in those individuals. Aim: The aim of the study is to investigate how healthcare professionals can identify pressure injuries in the skin suit of individuals with dark skin pigmentation and to find out the reason for the lack of guidance. Metod: Descriptive literature study based on quantitative studies. Ten quantitative articles have been used. Of which five of them were found in primary search while the rest were found in secondary and third search. Databases pubmed and Cinahl used in article searches. Resultat: There are some methods that are relevant for clinical assessment of pressure ulcers in individuals with dark skin pigmentation. Pressure ulcers can be identified in the person with dark skin pigmentation using ultrasound, munsell color charts and imaging techniques. Lack of guidance on how to identify pressure ulcers in individuals with dark skin tones can already be seen during nursing education, that colleges and universities do not contribute more teaching about pressure ulcers aimed at people with dark skin pigments. Slutsats: It is not possible to draw conclusions due to the fact that low-quality studies were included in the study. There are methods that can be used in clinical assessments of pressure ulcers in people with dark skin pigmentation, among other things with the help of ultrasound and imaging techniques. However, there is still a lack of guidance and therefore further research in this area would be necessary.
