Polymeric Hairy Nanoparticles with Helical Hairs: Synthesis and Self-Assembly

Nanoscale particles based on the nature of building blocks often self-assemble into superstructures with distinctive spatial arrangements which can be used as functional materials for different application. Micro-phase separated hairy nanoparticle with helical hair can self-assemble to form supramolecular material which may mimic the properties and functions of the natural polymers such as protein and cellulose. Beside this hairy/core-shell nanoparticles also may find many applications such as in asymmetric catalysis, nano-fillers in tire and rubbers, model systems for biology, lithography and as sensors. In this work, we have successfully synthesized two hairy nanoparticles both of which has cross-linked polystyrene core with helical poly (3- methyl 4- vinyl pyridine) and poly (2- methoxystyrene) brushes respectively by living anionic polymerization via one-pot synthesis. NMR spectroscopy was used to determine that polymerization was successful and compositions of HNPs have the agreement with the targeted HNPs structure. By tailoring the architecture (functionalization of polymer chains, the degree of polymerization, grafting density) of HNPs, it is possible to control the final properties of the system. Differential Scanning Calorimetry was used to demonstrate the thermal properties of the synthesized HNPs which corresponds to polymer composition. Dynamic light scattering, SEM and AFM images were recorded to measure the particle size and morphology of the particles. Circular dichroism spectroscopy was used to determine the induced chirality of helical polymer brushes by complexing it with the small chiral molecule. SEM and AFM imaging were recorded to find out the morphology and hierarchically self-assembly of the hairy nanoparticle system. The synthesized particles may have great potential to successfully generate self-assembled suprastructures which can further solve the chiral resolution problem and can also find different applications.

Heavy metal contamination significantly reduces crop yields, causing serious problems in agriculture and having a major impact on human health if these contaminants enter the food chain. Understanding the mechanisms of plant responses could help to increase their resistance to heavy metals as well as their potential use in phytoremediation. Carbohydrates play an important role in plant growth and development as well as in defense reactions. This work summarizes the results of four publications focused on the effects of arsenic and thorium on antioxidant mechanisms in tobacco plants and horseradish root crops. Attention is paid, among other things, to the dynamics of sugar contents, which are potentially important molecules involved in the fight against oxidative stress. The first publication summarizes arsenic effects on plant physiological parameters, focusing on arsenic tolerance-enhancing mechanisms as well as summarizing the ability of plants to cope with arsenic-induced oxidative and nitrosative stress. Emphasis was placed on, among other things, a topic that was unjustly neglected in previous publications - i.e. carbohydrate metabolism. Stimuli-Responsive Janus Particles

Janus particles, named after the two faced Roman god Janus, possess unique asymmetry and combine two distinct functions at their opposite sides, allowing them to target complex self-assembled architectures and materials inaccessible for homogeneous building blocks. In this study, three areas regarding the topic of Janus particles were explored: the synthesis of Janus particles, their (self-) assembly, and applications. In the first part of the work, we have drawn our attention to the optimization of the synthetic procedures concerning the preparation of Janus particles and to the extending of the current Janus particle library by adding new geometries to the list. In the case of spherical Janus particles, we have developed an easy approach to tailor the Janus ratio of the resulting particles, thus, extending the possibilities of the Pickering emulsion approach for the creation of a variety of Janus particle architectures. Additionally, a new methodology was employed to measure directly and in situ the position/contact angle of the prepared Janus particles with different Janus ratios at a water-oil interface. It was further concluded that having simply two different functionalities on a particle surface does not necessarily imply amphiphilic behavior: only in the case of large wettability contrasts our particles were in a true Janus regime. In the case of platelet-like Janus particles, we have developed a completely new approach for their large-scale synthesis, which involved a reduced number of steps compared to the spherical Janus particles. In the second part of the work, the assembly behavior of various kinds of functional spherical Janus particles was investigated depending on the nature of the Janus particles and the surrounding media conditions. Oppositely charged, uncharged amphiphilic, and charged amphiphilic Janus particles were fabricated comprising different responsive polymers on their surface, and their assembly was investigated depending on the pH value of the dispersion, the ionic strength, or the solvent. It was found that, under specific conditions, the Janus particles formed hierarchical chain-like structures in solutions, which were not observed in the case of the homogeneous particle mixtures. The obtained results indicate that the fundamental understanding of the Janus particle assembly mechanisms is crucial for the programmed formation of desired structures. In the third part of the work, we have focused on the applications of our developed hybrid hairy Janus particles and proposed two main directions that would benefit from the unique properties or architecture of the Janus particles. The first direction is based on the exploitation of the superior interfacial activity of the Janus particles and their use for interfacial catalysis. The second proposed direction for the application of Janus particles is based on their use as building blocks for functional structured surfaces. The prepared surfaces were thoroughly characterized and tested for their performance toward anti-icing as well as anti-fouling applications. Ultimately, the developed functional surfaces based on Janus particles as building blocks are very promising for their future application in the coating technology.

Functional genomics of nodulins in the model legume Lotus japonicus

During this PhD project three technical platforms were either improved or newly established in order to identify interesting genes involved in SNF, validate their expression and functionally characterise them. An existing 5.6K cDNA array (Colebatch et al., 2004) was extended to produce the 9.6K LjNEST array, while a second array, the 11.6K LjKDRI array, was also produced. Furthermore, the protocol for array hybridisation was substantially improved (Ott et al., in press). After functional classification of all clones according to the MIPS database and annotation of their corresponding tentative consensus sequence (TIGR) these cDNA arrays were used by several international collaborators and by our group (Krusell et al., 2005; in press). To confirm results obtained from the cDNA array analysis different sets of cDNA pools were generated that facilitate rapid qRT-PCR analysis of candidate gene expression. As stable transformation of Lotus japonicus takes several months, an Agrobacterium rhizogenes transformation system was established in the lab and growth conditions for screening transformants for symbiotic phenotypes were improved. These platforms enable us to identify genes, validate their expression and functionally characterise them in the minimum of time.<br> The resources that I helped to establish, were used in collaboration with other people to characterise several genes like the potassium transporter LjKup and the sulphate transporter LjSst1, that were transcriptionally induced in nodules compared to uninfected roots, in more detail (Desbrosses et al., 2004; Krusell et al., 2005). Another gene that was studied in detail was LjAox1. This gene was identified during cDNA array experiments and detailed expression analysis revealed a strong and early induction of the gene during nodulation with high expression in young nodules which declines with the age of the nodule. Therefore, LjAox1 is an early nodulin. Promoter:gus fusions revealed an LjAox1 expression around the nodule endodermis. The physiological role of LjAox1 is currently being persued via RNAi.<br> Using RNA interference, the synthesis of all symbiotic leghemoglobins was silenced simultaneously in Lotus japonicus. As a result, growth of LbRNAi lines was severely inhibited compared to wild-type plants when plants were grown under symbiotic conditions in the absence of mineral nitrogen. The nodules of these plants were arrested in growth 14 post inoculation and lacked the characteristic pinkish colour. Growing these transgenic plants in conditions where reduced nitrogen is available for the plant led to normal plant growth and development. This demonstrates that leghemoglobins are not required for plant development per se, and proves for the first time that leghemoglobins are indispensable for symbiotic nitrogen fixation. Absence of leghemoglobins in LbRNAi nodules led to significant increases in free-oxygen concentrations throughout the nodules, a decrease in energy status as reflected by the ATP/ADP ratio, and an absence of the bacterial nitrogenase protein. The bacterial population within nodules of LbRNAi plants was slightly reduced. Alterations of plant nitrogen and carbon metabolism in LbRNAi nodules was reflected in changes in amino acid composition and starch deposition (Ott et al., 2005). These data provide strong evidence that nodule leghemoglobins function as oxygen transporters that facilitate high flux rates of oxygen to the sites of respiration at low free oxygen concentrations within the infected cells. / Pflanzen der Ordnung der Leguminosen sind von weltweiter Bedeutung für Landwirtschaft und die allgemeine Nährstoffzusammensetzung von Böden. Die physiologische Besonderheit der Leguminosen liegt in ihrer Fähigkeit begründet, zusammen mit Bakterien, den sogenannten Rhizobien, eine Symbiose einzugehen, im Zuge derer es möglich wird, molekularen Luftstickstoff zu binden. Dieser biochemische Prozess findet in neu gebildeten Pflanzenorganen, den sogenannten Wurzelknöllchen statt.<br> In den Pflanzenwissenschaften werden Gene, die im Zuge der Infektion von Leguminosen mit Rhizobien reguliert werden und für den Entwicklungsprozess der Knöllchen eine wichtige Rolle zu spielen scheinen, als Noduline bezeichnet. Mit Hilfe von sogenannten Hochdurchsatzverfahren ist es in den letzten Jahren möglich geworden, die differentielle Expression von Tausenden von Genen gleichzeitig zu beobachten. Zu diesen Verfahren gehören sogenannte cDNA Arrays. Im Zuge dieser Doktorarbeit wurden die weltweit zweitgrößten cDNA Arrays für die Modell-Leguminose Hornklee (Lotus japonicus), der in unserer Gruppe als Untersuchungsobjekt verwendet wird, entwickelt. Mit Hilfe dieser Methode ist es uns möglich, die Regulation von etwa 15.000 Genen gleichzeitig zu untersuchen. Im Zuge von Untersuchungen, die sich mit der Entwicklung von Wurzelknöllchen in Lotus japonicus beschäftigten wurde ein Nodulin, dessen Existenz früher schon einmal beschrieben wurde, noch einmal bestätigt und die Funktion dieses Genes genauer untersucht. Es kodiert für das Enzym Vitamin C Oxidase, das unter Verwendung von molekularem Sauerstoff reduziertes Vitamin C zu einer anderen Form, dem Dehydroascorbat, oxidiert. Dabei wird Wasserstoffperoxid gebildet. Es konnte gezeigt werden, dass sich die Transkription dieses Gens in infizierten Wurzeln kontinuierlich im Verlauf der Symbiose erhöht, jedoch ist die Transkription in jungen Wurzelknöllchen höher als in alten. Darüber hinaus ist es in nur einer Zellschicht der Wurzelknöllchen, die sehr wichtig für die Entwicklung und tatsächliche Funktion der Knöllchen ist, aktiv. Aus den Beobachtungen kann geschlossen werden, dass dieses Gen eine wichtige Funktion in der Entwicklung der Knöllchen zu spielen scheint und vermutlich zur Zellstreckung und Zellteilung in dieser speziellen Zellschicht beiträgt. In einem zweiten Teil der Arbeit wurde sich einem zweiten und dem wohl wichtigsten Nodulin der Leguminosen, dem Leghämoglobin, gewidmet. Leghämoglobin ist dem menschlichen Blutbestandteil Hämoglobin sehr ähnlich und erfüllt dieselbe Aufgabe: es bindet Sauerstoff. Dieser Prozess ist für Leguminosen von erheblicher Bedeutung, da die bereits beschriebene Fixierung von molekularem Luftstickstoff durch ein bakterielles Enzym katalysiert wird, das extrem sauerstoffempfindlich ist. Leghämoglobine gelten unbestritten als die am besten charakterisierten Einweiße aus Wurzelknöllchen und Wissenschaftler behaupten seit fast 40 Jahren, dass sie essentiell für die Funktion der Knöllchen sind. Doch dies wurde bis jetzt nie bewiesen.<br> Mit Hilfe einer neuen Methode, die die spezifische Bildung von Eiweißen verhindert, war es uns möglich, die Synthese von Leghämoglobin in Lotus japonicus vollkommen zu unterdrücken. In Folge dessen zeigen die transgenen Pflanzen deutliche Nährstoffmangelerscheinungen, wenn sie ohne zusätzlichen Stickstoff aber zusammen mit Rhizobien angezogen werden. Sie können zwar Wurzelknöllchen bilden, jedoch sind diese kleiner und haben nicht die charakteristische rötliche Farbe, die bei unveränderten Pflanzen gefunden wird. Der Phänotyp dieser transgenen Pflanzen wird ganz eindeutig durch ihre Unfähigkeit hervorgerufen, Luftstickstoff fixieren zu können. Der Grund dafür ist das Fehlen des bakteriellen Enzyms, das für die Fixierung verantwortlich ist. Dieser Verlust wird durch erhöhte Sauerstoffgehalte in den Knöllchen verursacht. Außerdem konnten durch weitere Untersuchungen eine der vermuteten Funktionsmechanismen von Leghämoglobin bestätigt werden. Diese hier präsentierten Untersuchungen beweisen erstmalig die jahrzehnte alte Hypothese, dass Leghämoglobine essentiell für die Stickstofffixierung in Leguminosen sind.

Untersuchungen zur Strömungs-Struktur Interaktion an dynamisch bewegten, flexiblen Oberflächen

Die experimentellen Untersuchungen zur Strömungs-Struktur-Interaktion kommen zu folgenden Ergebnissen. Die als entrainment bezeichnete Verhaltensweise von Fischen kann durch den gezielten Ausgleich von Auftriebs-, Widerstands- und einer erstmals nachgewiesenen Saugkraft und der damit verbundenen Reduzierung der lokomotorischen Energie erklärt werden. Des Weiteren zeigen die Experimente an einer wellenförmig bewegten Oberfläche, dass die Strömung an einem Wellenberg zyklisch zwischen laminarem und turbulentem Regime wechselt und dass diese Oszillation zur Reduzierung des Form- und Gesamtwiderstandes der geschleppten Oberfläche führt. Dünne, flexible Häutchen hairy flaps an der Rückseite eines Zylinders führen zu einer Änderung der Struktur der Wirbelablösung an diesem. Dadurch wird sowohl die auf den Zylinder wirkende Auftriebskraft als auch die Widerstandskraft um bis zu 65% reduziert. Für die Interaktion zwischen der Strömung um zwei hintereinander positionierte elastische Zylinder und ihrer Kinematik konnte die Synchronisierung (lock-in) ihrer Bewegung mit einer verbundenen Zerstörung der Wirbelstraße hinter dem zweiten Zylinder gezeigt werden. / The experimental investigations presented herein explain the behavioural adaptation of fish called entrainment for the first time. The results confirm a balance of lift-, drag- and a suction-force, explaining the reduction of locomotive energy. Furthermore, flow measurements around an undulating membrane affirm an oscillation between laminar and turbulent flow over one period of the motion and that this oscillation decreases the pressure- and drag-force of the towed membrane. Experiments on thin and flexible flaps attached at the lee-side of a cylinder, show that the flaps alter the natural vortex separation cycle in such a way that the vortices do not shed in a staggered side-by-side arrangement but in line in a row with the cylinder wake axis. Thus, flow fluctuations are reduced by 42% in stream-wise - and 35% in transversal direction at best, compared to a reference case without hairy-flaps. Finally, investigations on the flow around and on the kinematics of two flexible cylinders in a tandem arrangement demonstrate a synchronisation of their motion (lock-in), resulting in the destruction of the vortex-street behind the second cylinder.

A molecular approach to taxol biosynthesis

Secondary metabolism in plants produces numerous compounds with wide-ranging activities, including the antineoplastic compound taxol and related taxanes. The biotechnological production of taxol has so far been based on empirical studies. The aim of the present work has been to study how the factors that enhance taxane production affect the metabolic profiles and gene expression in productive cells. As a consequence of this work, new potential candidates have been obtained for unknown taxane biosynthetic genes, some bottle-neck steps of taxane biosynthesis (in vitro and in silico) have been identified and a master regulator, not only for taxane biosynthesis, but also for other secondary metabolism routes, has been characterized. Coronatine, a powerful and less harmful elicitor than methyl jasmonate, has been successfully assayed and found to increase taxane production. In the different studies of this work, the expression level of genes that participate in taxol biosynthesis has been determined, clarifying their involvement in the production of this anti-cancer agent. / El metabolismo secundario de las plantas produce numerosos compuestos con un amplio rango de actividades, entre los que se encuentra el compuesto antineoplásico taxol y los taxanos relacionados, la producción biotecnológica del cual se basa en estudios empíricos. El objetivo de este trabajo ha sido estudiar como los factores que incrementan la producción de taxanos afectan los perfiles metabólicos y la expresión génica en los cultivos. De esta manera se han identificado nuevos genes candidatos que codifican para los genes desconocidos de la biosíntesis, algunos pasos limitantes de ésta y se ha caracterizado un regulador relacionado con el metabolismo secundario. Se ha ensayado la coronatina, un elicitor más eficiente para mejorar la producción de taxanos y menos dañino que el jasmonato de metilo. En los diferentes ensayos de este trabajo han sido determinados los niveles de expresión de genes que participan en la biosíntesis de taxol, ayudando a comprender su papel en la producción de este anticancerígeno.

Sur le problème à deux corps et le rayonnement gravitationnel en théories scalaire-tenseur et Einstein-Maxwell-dilaton / On the motion and gravitational radiation of binary systems in scalar-tensor and Einstein-Maxwell-dilaton theories

Avec la naissance de l’"astronomie gravitationnelle", vient l’opportunité inédite de tester la relativité générale et ses alternatives dans un régime de champ fort jamais observé jusqu’alors : celui de la coalescence d’un système binaire d’objets compacts. Cette thèse propose d’étudier le problème du mouvement ainsi que du rayonnement gravitationnel d’un tel système en gravités modifiées, en y adaptant et en généralisant certains développements analytiques clés de la relativité générale. On montre d’abord comment étendre le formalisme "effective-one-body" (EOB) à une large classe de gravités modifiées, parmi lesquelles les théories scalaire-tenseur. Dans ces dernières, l’interaction gravitationnelle est modifiée par l’ajout d’un degré de liberté scalaire (sans masse) à la relativité générale. Le lagrangien à deux corps correspondant étant connu à l’ordre post-post-keplerien, nous construisons un hamiltonien EOB associé, décrivant le mouvement d’une particule test dans des champs effectifs. Ceci permet de simplifier la dynamique à deux corps et d’en définir une resommation ; et ainsi, d’en explorer le régime de champ fort, près de la coalescence du système. On "s’attaque" ensuite, et pour la première fois, à la description analytique d’un système binaire de trous noirs "chevelus", afin d’obtenir les formes d’ondes gravitationnelles (EOB) associées ; et ce, sur l’exemple simple des théories Einstein-Maxwell-dilaton, qui généralisent les théories scalaire-tenseur par l’ajout d’un champ vectoriel (sans masse). Pour ce faire, on calcule le lagrangien à deux corps à l’ordre post-keplerien ainsi que le flux d’énergie rayonnée à l’infini à l’ordre quadrupolaire. Tout comme en relativité générale, ces développements reposent sur la description de la trajectoire des trous noirs par les lignes d’univers de particules ponctuelles, décrites par une action "skeleton" généralisant celle, géodésique, de la relativité générale. Enfin, à l’aide des "superpotentiels" de Katz, que l’on généralise pour définir la masse (nœtherienne) d’un trou noir à "cheveux" vectoriel et scalaire, on montre que la première loi de la thermodynamique qui en découle est particulièrement adaptée, lorsqu’un trou noir est membre d’un système binaire, pour en décrire les réajustements éventuels sous l’influence d’un compagnon lointain. La thermodynamique des trous noirs est alors utilisée pour interpréter et discuter du domaine de validité de leur "skeletonisation". / With the birth of "gravitational wave astronomy" comes the opportunity to test general relativity and its alternatives in a strong field regime that had never been observed so far: that of the coalescence of a compact binary sytem. This thesis studies the problem of motion and gravitational radiation from such systems in modified gravities, by adapting some of the key analytical tools that were first developed in the context of general relativity. First, we show how to widen the "effective-one-body" (EOB) formalism to a large class of modified gravities, including, e.g., scalar-tensor theories. In the latter, the gravitational interaction is described by supplementing general relativity with a (massless) scalar degree of freedom. The corresponding two-body lagrangian being known at post-post-keplerian order, we build an associated EOB hamiltonian, which describes the motion of a test particle orbiting in effective external fields. This enables to simplify and resum the two-body dynamics; and hence, to explore the strong-field regime near merger. We then "tackle", for the first time, the analytical description of "hairy" binary black hole systems, and obtain their (EOB) gravitational waveform counterparts in Einstein-Maxwell-dilaton theories, which generalize scalar-tensor theories by means of a (massless) vector field. To that end, we derive the two-body lagrangian at post-keplerian order as well as the energy flux radiated at infinity at quadrupolar order. As in general relativity, our developments rely on the phenomenological description of the black hole’s trajectories as worldlines of point particles that are, in turn, described by a "skeleton" action generalizing that of general relativity. Finally, we develop a formalism based on Katz’ "superpotentials" to define the mass (as a nœther charge) of a black hole that is endowed with vector and scalar "hair". We then deduce the first law of thermodynamics, which is particularly suitable to describe its readjustments when interacting with a faraway companion. Black hole thermodynamics is lastly shown to be a powerful tool to interpret and discuss the scope of their "skeletonization".

Die experimentellen Untersuchungen zur Strömungs-Struktur-Interaktion kommen zu folgenden Ergebnissen. Die als entrainment bezeichnete Verhaltensweise von Fischen kann durch den gezielten Ausgleich von Auftriebs-, Widerstands- und einer erstmals nachgewiesenen Saugkraft und der damit verbundenen Reduzierung der lokomotorischen Energie erklärt werden. Des Weiteren zeigen die Experimente an einer wellenförmig bewegten Oberfläche, dass die Strömung an einem Wellenberg zyklisch zwischen laminarem und turbulentem Regime wechselt und dass diese Oszillation zur Reduzierung des Form- und Gesamtwiderstandes der geschleppten Oberfläche führt. Dünne, flexible Häutchen hairy flaps an der Rückseite eines Zylinders führen zu einer Änderung der Struktur der Wirbelablösung an diesem. Dadurch wird sowohl die auf den Zylinder wirkende Auftriebskraft als auch die Widerstandskraft um bis zu 65% reduziert. Für die Interaktion zwischen der Strömung um zwei hintereinander positionierte elastische Zylinder und ihrer Kinematik konnte die Synchronisierung (lock-in) ihrer Bewegung mit einer verbundenen Zerstörung der Wirbelstraße hinter dem zweiten Zylinder gezeigt werden.:Zusammenfassung 1 Conclusions 7 1. Anhang 155 / The experimental investigations presented herein explain the behavioural adaptation of fish called entrainment for the first time. The results confirm a balance of lift-, drag- and a suction-force, explaining the reduction of locomotive energy. Furthermore, flow measurements around an undulating membrane affirm an oscillation between laminar and turbulent flow over one period of the motion and that this oscillation decreases the pressure- and drag-force of the towed membrane. Experiments on thin and flexible flaps attached at the lee-side of a cylinder, show that the flaps alter the natural vortex separation cycle in such a way that the vortices do not shed in a staggered side-by-side arrangement but in line in a row with the cylinder wake axis. Thus, flow fluctuations are reduced by 42% in stream-wise - and 35% in transversal direction at best, compared to a reference case without hairy-flaps. Finally, investigations on the flow around and on the kinematics of two flexible cylinders in a tandem arrangement demonstrate a synchronisation of their motion (lock-in), resulting in the destruction of the vortex-street behind the second cylinder.:Zusammenfassung 1 Conclusions 7 1.
