Who is afraid of MT?

Machine translation (MT) is experiencing a renaissance. On one hand, machine translation is becoming more common and used in ever larger scale, on the other hand many translators have an almost hostile attitude towards machine translation programs and those translators who use MT as a tool. Either it is assumed that the MT can never be as good as a human translation or machine translation is viewed as the ultimate enemy of the translator and as a job killer. The article discusses with various examples the limits and possibilities of machine translation. It demonstrates that machine translation can be better than human translations – even if they were made by experienced professional translators. The paper also reports the results of a test that showed that translation customers must expect that even well-known and expensive translation service providers deliver a quality that is on par with poor MT. Overall, it is argued that machine translation programs are no more and no less than an additional tool with which the translation industry can satisfy certain requirements. This abstract was also – as the entire article – automatically translated into English.

Exploring New Horizons in Microwave-Promoted Iminyl Radical Chemistry and Synthesis of Bulky Dehydroamino Acids

The first project in this dissertation presents a simplified and efficient protocol for synthesizing pyrrolines through 5-exo iminyl radical cyclizations. The microwave irradiation of O-Phenyloximes tethered to alkenes causes N-O homolysis resulting in iminyl radical generation, which subsequently undergoes 5-exo-trig cyclizations furnishing pyrrolines. This eliminates the need for toxic radical initiators (AIBN, benzoyl peroxide), propagating agents (Bu3SnH, (Me3Si)3SiH), and expensive catalysts or single-electron transfer (SET) cycles. We explored the scope of diverse traps and substrates for iminyl radical cyclizations. The iminyl radical cyclizations formed versatile pyrrolines with moderate to excellent yields. The diastereoselectivity also ranged from low to high. Moreover, these versatile pyrrolines were further transformed via various reactions, such as hydrogenation, allylation, dihydroxylation, and cross-metathesis. The second part of this project extends the scope of the non-redox iminyl-radical based approach to γ-C(sp3)−H ketone activation. The sequence of N-O homolysis triggered by microwave irradiation of O-phenyloximes, 1,5-hydrogen atom transfer (HAT), trapping of the radical intermediate, and in situ imine hydrolysis, ultimately leads to the formal γ-C–H functionalization of ketones. We achieved both C-O and C-C bond formation by using diverse O-phenyloxime substrates. This work's notable achievement was accomplishing γ-C–H activation of 1o carbon atoms, a feat that has not been attained using SET-based iminyl radical chemistry. The third part of this dissertation focuses on the influence that dehydroamino acids have on secondary structures. This project describes the synthesis of incipient 310 helical tetrapeptides containing dehydroamino acids. A bulky dehydroethylnorvaline-containing tetrapeptide was synthesized. Based on our published data, we speculated that dehydroethylnorvaline might increase peptide proteolytic stability.

Pedofiler begår inte alltid sexuella övergrepp och de som begår sexuella övergrepp är inte alltid pedofiler. Det är viktigt att som professionell kunna skilja på person och handling. The study's main findings is that news media and social medias image of pedophiles as equivalent to perpetrators of child sexual abuse is not consistent with what the literature and the professionals say. Pedophiles do not always commit sexual abuse and those who commit sexual abuse are not always pedophiles. It is important as a professional to distinguish between person and action. Further, the results of the study show that pedophilia is a complex sexually deviant preference that requires multiple perspectives to be explained and understood.

Propriétés biochimiques, enzymatiques et physiologiques de la Human Airway Trypsin-like protease (HAT)

La fibrose kystique (FK) est caractérisée par une inflammation pulmonaire chronique. Cette dernière est définie entre autres par une augmentation importante de la production de mucus. Celui-ci s'accumule dans les bronches empêchant ainsi l'éventuel passage de l'air. De plus, le mucus forme une barrière protectrice pour les bactéries qui va causer l'inflammation des poumons. La protéase human airway trypsin-like (HAT) a été retrouvée dans les expectorations de patients atteints d'asthme et bronchite chronique, pathologies similaires à la FK. Elle semble jouer un rôle dans l'inflammation car elle augmente entre autres la production de MUC5AC, protéine composant majoritairement le mucus. Le but de cette étude était de synthétiser la HAT dans une conformation active dans un système eucaryote pour étudier ses propriétés biochimiques, enzymatiques ainsi que son potentiel pro-inflammatoire dans la lignée cellulaire pulmonaire Calu-3. L'ADNc codant pour la partie soluble de la HAT a été inséré dans le vecteur pMT-BiP V5-His. Cet ADN recombinant fut ensuite transfecté dans les cellules de Drosophile Schneider 2 (S2). La HAT recombinante a par la suite été purifiée et son activité protéolytique testée avec un substrat fluorogénique. Ses propriétés biochimiques et enzymatiques ont été étudiées ainsi que son effet sur la production d'IL-8 et sa détection dans les expectorations de patients. La HAT purifiée était «enzymatiquement» pure, donc l'effet protéolytique observé avec le substrat fluorogénique est dû à la protéase. Les analyses de ses propriétés biochimiques montrent que l'activité protéolytique de la HAT est inhibée par un pH acide (<6.5), certains inhibiteurs synthétiques (l'aprotinine, la leupeptine et le PEFABLOC) et endogène (i.e: alpha-2 anti-plasmine (a2-AP)) de protéases à sérine. D'un autre côté, les essais enzymatiques révèlent que la HAT a une préférence pour les substrats ayant une arginine en P4, P3 et Pl plutôt que ceux ayant un glutamate en P4, P2 et P1'. Finalement, l'étude sur l'implication physiologique de l'enzyme démontre que la protéase ne semble pas avoir d'effet ou au plus peu d'effet sur la production d' IL-8. [Symboles non conformes]

Therefore the different homonyms and polysemous senses of lexical items and the underlying semantic relations between lexical items are accounted for. A description should be given of the lexical items which form the macrostructure of HAT. These lexical items include sublexical, multilexical and free lexical items. The treatment of multilexical items in HAT is not satisfactory at present and suggestions are made for the improvement of the situation. Possible solutions for distinguishing between homonymy and polysemy are given. Suggestions to determine the possible polysemous senses of a lexical item are also made. The ordering of polysemous senses and different homonyms are discussed. It is essential to make use of a synchronic criterion to distinguish between homonymy and polysemy. The HAT's treatment of semantic relations between lexical items is not always very succesfull. The semantic relation which receives the most substantial treatment is that of semantic inclusion between lexical items. The HAT's treatment of synonyms is not without problems. More attention should be given to semantic linking and semantic opposition between lexical items. An evaluation of the system of cross-references in HAT is given, with special reference to the function of cross-references. Sometimes indistinct typographical conventions are used and they confuse the user of the dictionary. The indistinct use of typographical conventions has a detrimental influence on the transfer of information. The cross-reference system of HAT can be improved by an informative explanation in the general introduction, focusing on ways and means to use it more specifically, unambiguously and consistently. A distinction is made between various types of definitions used to define lemmas. Well known lexicographic definitions such as definitions per genus et differentia, synonymdefinitions, example definitions, circular definitions and descriptive definitions are discussed. The usage definition and two definitions which explicate a semantic plus value, the contrast and diminutive definitions, are identified. Attention is also given to the role of the general introduction in a standard synchronic dictionary. Suggestions are made to improve the general introduction of HAT. / AFRIKAANSE OPSOMMING: Wanneer semantiese inligting in 'n woordeboek ondersoek word, word alle aspekte van leksikale items se betekenis in ag geneem. Planet-induced Stellar Pulsations in HAT-P-2's Eccentric System

Extrasolar planets on eccentric short-period orbits provide a laboratory in which to study radiative and tidal interactions between a planet and its host star under extreme forcing conditions. Studying such systems probes how the planet's atmosphere redistributes the time-varying heat flux from its host and how the host star responds to transient tidal distortion. Here, we report the insights into the planet-star interactions in HAT-P-2's eccentric planetary system gained from the analysis of similar to 350 hr of 4.5 mu m observations with the Spitzer Space Telescope. The observations show no sign of orbit-to-orbit variability nor of orbital evolution of the eccentric planetary companion, HAT-P-2b. The extensive coverage allows us to better differentiate instrumental systematics from the transient heating of HAT-P-2b's 4.5 mu m photosphere and yields the detection of stellar pulsations with an amplitude of approximately 40 ppm. These pulsation modes correspond to exact harmonics of the planet's orbital frequency, indicative of a tidal origin. Transient tidal effects can excite pulsation modes in the envelope of a star, but, to date, such pulsations had only been detected in highly eccentric stellar binaries. Current stellar models are unable to reproduce HAT-P-2's pulsations, suggesting that our understanding of the interactions at play in this system is incomplete.

Samarium(II) mediated radical cascades of keto esters for the generation of molecular complexity

A highly regio- and diastereoselective approach towards complex 6-membered lactones was developed using allyl/propargyl benzyl ethers and delta keto esters. Crucially, the classical ET reagent SmI2 gave unsatisfactory results and it was necessary to develop and screen new Sm(II) cyclopentadienyl complexes to deliver high selectivity in the transformation. The methodology was extended to a one-pot approach to complex cycloheptanols using SmI2-H2O in a second stage of the process. Samarium(II) folding cascades were developed where simple, linear starting materials are converted to complex polycyclic architectures bearing multiple stereocentres. It was found that, depending on the sidechain in the starting material, it was possible to achieve four different pathways from the common radical intermediate. Crucially, transannular 1,5-HAT from tertiary and benzylic positions was observed to give diverse products. A proposed 1,5-HAT facilitated SmI2-mediated 6-membered lactone radical cyclisations for the first time without an activating proton donor additive. Enantioselective samarium(II) mediated cyclisation cascades were achieved, where simple beta keto esters are converted to complex polycyclic architectures bearing up to five contiguous stereocentres with high diastereo- and enantiocontrol. In the process, a simple and easy to prepare chiral aminodiol was employed which could be recycled after the reaction. Unprecedented, enantioselective transannular radical cascades allowed access to unique 3- dimensional scaffolds inaccessible by other synthetic methods.

Antibody and Antigen in Heparin-Induced Thrombocytopenia

Immune heparin-induced thrombocytopenia (HIT) is a potentially serious complication of heparin therapy and is associated with antibodies directed against a complex of platelet factor 4 (PF4) and heparin. Early diagnosis of HIT is important to reduce morbidity and mortality. I developed an enzyme immunoassay that detects the binding of HIT IgG to PF4-heparin in the fluid phase. This required techniques to purify and biotinylate PF4. The fluid phase assay produces consistently low background and can detect low levels of anti-PF4-heparin. It is suited to testing alternative anticoagulants because, unlike in an ELISA, a clearly defined amount of antigen is available for antibody binding. I was able to detect anti-PF4-heparin IgG in 93% of HIT patients. I also investigated cross-reactivity of anti-PF4-heparin antibodies with PF4 complexed to alternative heparin-like anticoagulants. Low molecular weight heparins cross-reacted with 88% of the sera from HIT patients while half of the HIT sera weakly cross-reacted with PF4-danaparoid (Orgaran). The thrombocytopenia and thrombosis of most of these patients resolved during danaparoid therapy, indicating that detection of low affinity antibodies to PF4-danaparoid by immunoassay may not be an absolute contraindication for danaparoid administration. While HIT patients possess antibodies to PF4-heparin, I observed that HIT antibodies will also bind to PF4 alone adsorbed on polystyrene ELISA wells but not to soluble PF4 in the absence of heparin. Having developed a technique to affinity-purify anti-PF4-heparin HIT IgG, I provide the first estimates of the avidity of HIT IgG. HIT IgG displayed relatively high functional affinity for both PF4-heparin (Kd=7-30nM) and polystyrene adsorbed PF4 alone (Kd=20-70nM). Furthermore, agarose beads coated with PF4 alone were almost as effective as beads coated with PF4 plus heparin in depleting HIT plasmas of anti-PF4-heparin antibodies. I conclude that the HIT antibodies which bind to polystyrene adsorbed PF4 without heparin are largely the same IgG molecules that bind PF4-heparin and thus most HIT antibodies bind epitope(s) on PF4 and not epitope(s) formed by part of a PF4 molecule and part of a heparin molecule. Binding of PF4 to heparin (optimal) or polystyrene/agarose (sub-optimal) promotes recognition of this epitope. Under conditions that are more physiological and sensitive than previous studies, I observed that affinity-purified HIT IgG will cause platelet aggregation upon the addition of heparin. Platelets activated with HIT IgG increased their release and surface expression of PF4. I quantitated the binding of affinity-purified HIT 125I-IgG to platelets as they activate in a plasma milieu. Binding of the HIT IgG was dependent upon heparin and some degree of platelet activation. Blocking the platelet Fc??? receptor-II with the monoclonal antibody IV.3 did not prevent HIT IgG binding to activated platelets. I conclude that anti-PF4-heparin IgG is the only component specific to HIT plasma that is required to induce platelet aggregation. The Fab region of HIT IgG binds to PF4-heparin that is on the surface of activated platelets. I propose that only then does the Fc portion of the bound IgG activate other platelets via the Fc receptor. My data support a dynamic model of platelet activation where released PF4 enhances further antibody binding and more release.
